Background and aims

Strawberry (Fragaria x ananassa) is a high-value crop worldwide. Fusarium oxysporum f. sp. fragariae causes rapid wilting and death of strawberry plants and severe economic losses worldwide. To date, no studies have been conducted to determine colonisation of either susceptible or resistant strawberry plants by F. oxysporum f. sp. fragariae, or whether plant colonisation by F. oxysporum f. sp. fragariae differs between susceptible and resistant cultivars.

Methods

Colonisation of strawberry plants by a pathogenic isolate of F. oxysporum f. sp. fragariae was examined both on the root surface and within root tissue of one resistant cv. Festival and one susceptible cv. Camarosa using light and scanning electron microscopy from 4?h to 7?d post inoculation (pi).

Results

Resistant cv. Festival significantly impeded the spore germination and penetration from 4 to 12 hpi and subsequent growth and colonisation by this pathogen until 7 dpi compared with susceptible cv. Camarosa. At 7 dpi, fungal colonisation in resistant cv. Festival remained mainly confined to the epidermal layer of the root, while in susceptible cv. Camarosa, hyphae not only had heavily colonised the cortical tissue throughout but had also colonised vascular tissues.

Conclusions

This study demonstrates for the first time that resistance of a strawberry cultivar to F. oxysporum f. sp. fragariae is a result of impedance of pathogen growth and colonisation both on the plant surface and within host tissues. Resistance mechanisms identified in this study will be of high value for breeding programmes in developing new disease-resistant cultivars to manage this serious strawberry disorder.  相似文献

In the Philippines, strawberry is grown only in Benguet Province because of its unique climatic conditions. It has been a lucrative source of income for Benguet farmers and adds to the revenue of Benguet Province. The root lesion nematode, Pratylenchus penetrans is an economically important pest of strawberry in the area. It can cause substantial losses to strawberry growers, both by reducing vegetative plant growth and by reducing strawberry yields. The nematode has a very wide host range and hence, is not readily controlled by crop rotation. An alternative approach which growers may wish to consider trying is planting of strawberry varieties which are either resistant or tolerant to this nematode. The relative susceptibility/tolerance of six strawberry cultivars to the root lesion nematode, P. penetrans was evaluated under greenhouse and field conditions. Inoculation of 500 nematodes/pot did not significantly affect the fresh top weight, fresh root weight, and yield of strawberry cultivars Festival, Whitney, Winterdawn, Earlibrite, and Camarosa. The said cultivars had significantly higher number of nematodes recovered from the roots. On the other hand, the highest strawberry yield was recorded in cv Sweet Charlie, however, this was significantly reduced by nematode inoculation .Surprisingly, the number of nematodes recovered from the roots of this cultivar was significantly the lowest among the cultivars tested. Results of the field experiment showed that strawberry cv Sweet Charlie gave the highest marketable yield which was significantly different from the rest of the cultivars tested. This was followed by Festival, and Earlibrite. On the other hand, Camarosa and Whitney gave significantly lower yield than the above cultivars but significantly higher than Winterdawn. In terms of nematodes recovered from the roots, the highest was noted in Whitney, followed by Sweet Charlie and Earlibrite. The lowest was obtained from Camarosa, followed by Festival and Winterdawn. Based on the results of the greenhouse experiment, Festival, Whitney, Winterdawn, Earlibrite and Camarosa can be considered tolerant while Sweet Charlie was slightly susceptible to P. penetrans. However, based on the field trial, Sweet Charlie, Festival and Earlibrite were tolerant while Whitney, Camarosa and Winterdawn were slightly susceptible.  相似文献

Anthracnose (Colletotrichum spp.) is an important disease causing major yield losses and poor oil quality in olives. The objectives were to determine the diversity and distribution pattern of Colletotrichum spp. populations prevalent in olives and their relatedness to anthracnose pathogens in other hosts, assess their pathogenic variability and host preference, and develop diagnostic tools. A total of 128 Colletotrichum spp. isolates representing all olive-growing areas in Portugal and a few isolates from other countries were characterized by molecular and phenotypic assays and compared with reference isolates. Arbitrarily primed PCR data, internal transcribed spacer of rRNA gene and beta-tubulin 2 nucleotide sequences, colony characteristics, and benomyl sensitivity showed Colletotrichum acutatum to be dominant (>97%) with limited occurrence of Colletotrichum gloeosporioides (<3%). Among C. acutatum populations, five molecular groups, A2 to A6, were identified. A2 was widely prevalent (89%), coinciding with a high incidence of anthracnose and environmental conditions suitable to disease spread. A4 was dominant in a particular region, while other C. acutatum groups and C. gloeosporioides were sporadic in their occurrence, mostly related to marginal areas of olive cultivation. C. gloeosporioides, isolated from olive fruits with symptoms indistinguishable from those of C. acutatum, showed same virulence rating as the most virulent C. acutatum isolate from group A2. C. acutatum and C. gloeosporioides isolates tested in infected strawberry fruits and strawberry and lupin plants revealed their cross-infection potential. Diagnostic tools were developed from beta-tubulin 2 sequences to enable rapid and reliable pathogen detection and differentiation of C. acutatum groups.  相似文献

Colletotrichum acutatum causes anthracnose on a wide range of hosts including woody and herbaceous crops, ornamentals, fruits and conifers. Almond, citrus, lupin, olive and strawberry are some of the crops in which C. acutatum diseases are economically important. With the application of molecular markers and diagnostic PCR over the last 10–15 years, C. acutatum was identified as a major pathogen on a number of hosts instead of or along with C. gloeosporioides . C. acutatum displays high levels of genotypic and phenotypic diversity. The global populations of this cosmopolitan pathogen fit into at least eight distinct molecular groups, A1–A8, which show some degree of correlation with the morphological characteristics and varying patterns of host association and geographical distribution. The pathogen has complex epidemiology, exhibiting pathogenic and non-pathogenic lifestyles on target hosts, non-target crops and weeds. C. acutatum populations also show pathogenic variability and cross-infection potential in relation to a number of hosts. Molecular genetic tools are being developed to investigate the pathogenicity mechanisms of this key pathogen. This article mainly focuses on the global population diversity in C. acutatum , pathogen epidemiology and diagnosis, host colonization processes, and the development of tools for the identification and analysis of genes associated with pathogenicity. Background information on the pathogen origin, host range, disease symptoms and disease management strategies is also provided.  相似文献

Actin depolymerizing factors (ADFs) have been recently implicated in plant defense against pathogenic fungi, associated with the cytoskeletal rearrangements that contribute to establish an effective barrier against fungal ingress. In this work, we identified a DNA fragment corresponding to a part of a gene predicted to encode an ADF-like protein in genotypes of Fragaria ananassa resistant to the fungus Colletotrichum acutatum. Bulked segregant analysis combined with AFLP was used to identify polymorphisms linked to resistance in hybrids derived from the cross between the resistant cultivar 'Sweet Charlie' and the susceptible cultivar 'Pájaro'. The sequence of one out of three polymorphic bands detected showed significant BLASTX hits to ADF proteins from other plants. Two possible exons were identified and bioinformatic analysis revealed the presence of the ADF homology domain with two actin-binding sites, an N-terminal phosphorylation site, and a nuclear localization signal. In addition to its possible application in strawberry breeding programs, these finding may contribute to investigate the role of ADFs in plant resistance against fungi.  相似文献

Infection of strawberry plants with binucleate Rhizoctonia spp. results in an increase in peroxidase activity and the appearance of new isoforms of the enzyme. In healthy and diseased roots of two different strawberry genotypes seven peroxidase isoenzymes were found. In healthy strawberry cv. Senga Sengana, which was moderately resistant to infection, four isoenzymes (1, 2, 5, and 6) were found. Moreover the activity of these isoenzymes was increased and three new isoenzymes (3, 4, and 7) were found in infected roots. In the strawberry hybrid 3/2/86/88/R, which is very susceptible to infection, only isoenzyme 2 was present in the roots of healthy plants. Following infection, the activity of isoenzyme 2 was increased and five new isoenzymes (1, 4, 5, 6, and 7) were detectable. The results obtained indicate that strawberry resistance to binucleate Rhizoctonia may be correlated with peroxidase isoenzyme profile with particular reference to isoform 3, which is only present in infected roots of the moderately resistant cv., Senga Sengana.  相似文献

Abstract

Pseudomonas spp. strains capable of inducing systemic resistance were applied to sugarcane by sett treatment followed by soil applications in the field. Later the fungal pathogen Colletotrichum falcatum causing red rot disease of sugarcane was inoculated in the treated canes and its colonization was assessed by ELISA at different nodal positions above the point of inoculation. Studies with three cvs showed a significant variation in pathogen colonization only in disease susceptible cv CoC 671 and not with cvs Co 8021 and BO 91, moderately susceptible and moderately resistant to the disease, respectively. In further studies when pathogen colonization was assessed on the entire stalks of cv CoC 671, the pathogen titre was significantly reduced from three nodes upwards in the treated canes. In the upper nodes no pathogen colonization was noticed in bacteria-treated canes, whereas in the control all the nodes recorded higher titre for pathogen infection. Incorporation of chitin in the talc formulation caused further reduction in fungal colonization in the stalks.  相似文献

Anthracnose, or leaf-curl disease of anemone, caused by Colletotrichum sp., has been reported to occur in Australia, western Europe, and Japan. Symptoms include tissue necrosis, corm rot, leaf crinkles, and characteristic spiral twisting of floral peduncles. Three epidemics of the disease have been recorded in Israel: in 1978, in 1990 to 1993, and in 1996 to 1998. We characterized 92 Colletotrichum isolates associated with anthracnose of anemone (Anemone coronaria L.) for vegetative compatibility (72 isolates) and for molecular genotype (92 isolates) and virulence (4 isolates). Eighty-six of the isolates represented the three epidemics in Israel, one isolate was from Australia, and five isolates originated from western Europe. We divided these isolates into three vegetative-compatibility groups (VCGs). One VCG (ANE-A) included all 10 isolates from the first and second epidemics, and 13 of 62 examined isolates from the third epidemic in Israel, along with the isolate from Australia and 4 of 5 isolates from Europe. Another VCG (ANE-F) included most of the examined isolates (49 of the 62) from the third epidemic, as well as Colletotrichum acutatum from strawberry, in Israel. Based on PCR amplification with species-specific primers, all of the anemone isolates were identified as C. acutatum. Anemone and strawberry isolates of the two VCGs were genotypically similar and indistinguishable when compared by arbitrarily primed PCR of genomic DNA. Only isolate NL-12 from The Netherlands, confirmed as C. acutatum but not compatible with either VCG, had a distinct genotype; this isolate represents a third VCG of C. acutatum. Isolates from anemone and strawberry could infect both plant species in artificial inoculations. VCG ANE-F was recovered from natural infections of both anemone and strawberry, but VCG ANE-A was recovered only from anemone. This study of C. acutatum from anemone illustrates the potential of VCG analysis to reveal distinct subspecific groups within a pathogen population which appears to be genotypically homogeneous by molecular assays.  相似文献

炭疽病是柑橘的主要真菌性病害之一。2007年春,广东省德庆县名优柑橘品种贡柑炭疽病暴发流行。为了明确该县及广东省其他地区柑橘炭疽病菌的种类,为防治提供依据,对采集自广东省6个地区柑橘属10个栽培品种上的炭疽病样本进行病原菌分离,共获得柑橘炭疽病菌单孢菌株75株,对其中10株代表性的菌株进行了种类鉴定。通过培养性状和形态学特征观测、核糖体DNA(rDNA)内转录间区(ITS)序列分析、ITS区特异性引物PCR检测和系统发育关系比较等方面的研究,结果表明:10个柑橘炭疽病菌菌株均为盘长孢状刺盘孢Colletotrichum gloeosporioides,未发现国际上其他国家报道的严重危害柑橘花器和幼果部位的柑橘花后落果病病原菌——尖刺盘孢C.acutatum。  相似文献

苹果炭疽叶枯病是由胶孢炭疽菌(Colletotrichum gloeosporioides)引起的一种真菌病害,现已上升为世界苹果生产中的主要病害之一。了解不同来源的苹果炭疽叶枯病菌致病力差异及明确苹果种质资源对苹果炭疽叶枯病的抗性,对品种选育、品种合理布局以及控制病害的流行具有重要的参考价值。本研究对不同来源的79株病原菌进行了室内致病力测定,获知该菌致病力差异明显,其中强致病力菌株所占比例大。同时,本研究也对327份苹果种质资源进行了室内抗病性鉴定,其中高抗资源160份,中抗资源6份,中感资源22份,高感资源139份。表明我国现保存的苹果种质资源中存在丰富抗病种质。进一步按苹果分类系统分析发现,抗病资源在当前栽培的主要品种群中均有分布,特别是红玉品种群、富士品种群抗病资源最为丰富。  相似文献

Differential gene expression was analyzed after infection with Phytophthora infestans in six potato cultivars with different levels of resistance to late blight. To verify the infection of the potato leaflets, the amount of phytopathogen mRNA within the plant material was quantified by real-time quantitative PCR. The expression of 182 genes selected from two subtracted cDNA libraries was studied with cDNA array hybridization using RNA from non-infected and infected potato leaflets. Gene up- and down-regulation were clearly detectable in all cultivars 72 h post inoculation. Gene expression patterns in susceptible cultivars differed from those in potato varieties with a higher level of resistance. In general, a stronger gene induction was observed in the susceptible cultivars compared to the moderately to highly resistant potato varieties. Five genes with the highest homology to stress and/or defence-related genes were induced specifically in the susceptible cultivars. Four genes responded to pathogen attack independently of the level of resistance of the cultivar used, and three genes were repressed in infected tissue of most cultivars. Even in the absence of P. infestans infection, six genes showed higher expression levels in the somewhat resistant cultivars Bettina and Matilda. Possible reasons for the different levels of gene expression are discussed.  相似文献

Abstract

Macroscopic symptoms were observed in two strawberry cultivars, with the degree of symptom intensity varying depending on the susceptibility of the cultivars, i.e. resistant or susceptible. The symptoms presented as red spots and were observed 30 d following leaf tissue inoculation with the Mycosphaerella fragariae pathogen. A comparison of the superoxide dismutase isoform profiles obtained by gel electrophoresis in all samples extracted from both resistant and susceptible cultivars indicated one constant sharp band, identified as Mn[sbnd]SOD with a molecular mass of 19 kDa. The intensity of this band was higher in all samples derived from the resistant cultivar than in those from the susceptible cultivar. Another superoxide dismutase (SOD) isoform, identified as CuZn[sbnd]SOD with a molecular mass of 16 kDa, was detected in all soluble proteins derived from the resistant cultivar. This isoform was not observed in the susceptible cultivar; however, following an incremental increase in the amount of loaded protein, it was illuminated as a faint band in a sample collected 3 d after inoculation, indicating insufficient production of the CuZn[sbnd]SOD isoform in the susceptible cultivar during an oxidative burst induced by the M. fragaria pathogen. Several bands were also characterized in both cultivars containing Fe and Mn as their co-factors (Fe, Mn[sbnd]SOD). Unlike in the resistant cultivar, where the activity of Fe, Mn[sbnd]SOD isoforms gradually and regularly increased and reached its highest level on the third day after inoculation, the activity of the isoforms changed irregularly over 20 days of study in the susceptible cultivar.  相似文献

The effects of the ring rot causal agent Clavibacter michiganensis subsp. sepedonicus (a virulent strain 5369) on the peroxidase activity of various tissues of potato plants grown under axenic conditions were studied. Root infection enhanced peroxidase activity in all plant tissues (roots, leaves, and stems). In the resistant cv. Lugovskoi, peroxidase activity was much higher than in the susceptible cv. Luk'yanovskii. Co-culturing of the suspension cells of these potato cultivars with the bacterial pathogen also activated peroxidase in the cells of the resistant cultivar; in the cells of the susceptible cultivar, peroxidase activation was less pronounced. Treating suspension cell with exopolysaccharides secreted by the pathogen enhanced the activity of extra- and intracellular peroxidases, and the degree of this enhancement differed in the two potato cultivars.  相似文献

【背景】寄生帚梗柱孢霉是花生黑腐病的病原菌,被我国列为重要的进境植物检疫性有害生物。该病菌2009年已入侵我国广东,造成花生植株基部腐烂而死亡,严重威胁花生生产安全。筛选与种植抗病品种是防控该病害的重要措施。【方法】收集广东推广种植的15个主要花生品种,通过人工接种方法,鉴定这些品种对花生黑腐病菌的抗性水平。【结果】15个供试花生品种中,湛红2号、湛油62等2个品种表现为抗病;湛油75、湛油82、粤油390、粤油410、仲恺花44、仲恺花99、汕油诱1号等7个品种表现为中抗;花育33号、汕油523、汕油辐1号、粤油18、湛油53等5个品种表现为感病;仲恺花332表现为高感。【结论与意义】目前广东生产上推广种植的花生品种多数对黑腐病菌表现为抗病或中抗水平,部分品种表现为感病或高感。该结果可为我省花生品种的推广与布局提供依据。  相似文献

Plants of strawberry (cultivar Pájaro) were transformed with three defense related genes: ch5B, gln2 and ap24 using Agrobacterium tumefaciens. The ch5B gene encodes for a chitinase from Phaseolus vulgaris, while gln2 and ap24 encode for a glucanase and a thaumatin-like protein, respectively, both from Nicotiana tabacum. Sixteen transgenic lines expressing one or a combination of two defense genes were obtained. Phytopathological tests showed that two transgenic lines expressing only the ch5B gene displayed high levels of resistance to gray mold disease (Botrytis cinerea). The resistance was correlated with the presence of the foreign protein CH5B and the increase of chitinolytic activity in leaves. However, resistance toward Colletotrichum acutatum, the etiological agent of the anthracnose disease, was not enhanced in the transgenic plants. These results suggest that the ch5B gene can be used to introduce transgene-mediated resistance to gray mold in strawberry, due to the lack of natural resistance to this disease in the crop.  相似文献

The accumulation of anthocyanin pigments is one of the most important traits that turn strawberry fruit attractive to consumers. During ripening, strawberry fruit color development is associated to anthocyanin synthesis through the phenylpropanoid pathway. Phenylalanine ammonia-lyase (PAL) is a key enzyme in this pathway, having a determining role in strawberry fruit quality. In this work, we studied the level of anthocyanins during fruit ripening of two cultivars that differ in color development (Camarosa and Toyonoka). Toyonoka showed a lower anthocyanin accumulation that was limited to external fruit tissue, while Camarosa accumulated higher amount of anthocyanins in both internal and external sections. In addition, we cloned a full-length gene (FaPAL6) and analyzed its expression in different strawberry plant tissues. The expression of this gene is fruit specific, and increases during fruit ripening in both cultivars along with anthocyanin accumulation. The mRNA level of FaPAL6 was higher in Camarosa. PAL enzyme activity increased at similar rates in both cultivars at early ripening stages, but at the end of ripening PAL activity diminished in Toyonoka while it rose markedly in Camarosa. PAL activity was higher in internal fruit tissue, showing no correlation with anthocyanin level of the same section in both cultivars. The higher FaPAL6 expression and activity detected in Camarosa could be associated to the enhanced anthocyanin accumulation found in this cultivar.  相似文献

In addition to cultural practices, the application of the fungicide mefenoxam is an important disease management tactic used to control both pink rot and leak on potato tubers grown in the USA. Mefenoxam resistance has been identified in many of the potato growing regions, and therefore resistance management strategies are very important for retaining this fungicide as a tool to manage these storage rot diseases. The relationship between mefenoxam efficacy and cultivar susceptibility to pink rot and leak was assessed in post‐harvest inoculation studies. Mefenoxam was applied to potato (Solanum tuberosum) cultivars known to express varying levels of susceptibility to pink rot and leak caused by Phytophthora erythroseptica and Pythium ultimum, respectfully. Tubers harvested from plants treated with in‐furrow and foliar applications of mefenoxam were inoculated with isolates sensitive to the fungicide. Incidence and severity of both diseases ranged widely among cultivars. Russet Norkotah was the most susceptible to infection by P. erythroseptica, while cvs Pike and Atlantic were the most resistant. Cultivars Dark Red Norland, Russet Norkotah, Goldrush and Russet Burbank were most susceptible to infection by P. ultimum whereas Snowden was most resistant. Control of pink rot differed significantly among cultivars following mefenoxam treatment, ranging from 28% (cv. Goldrush) to 67% (cv. Snowden) and generally provided the greatest level of disease control on susceptible and moderately susceptible cultivars such as Russet Norkotah and Snowden, respectively. In contrast, the impact of mefenoxam on leak development was minimal and disease control did not differ significantly among the cultivars. The fungicide failed to control leak in the susceptible cvs Atlantic and Pike and control ranged from 1.7% to 5.2% in cvs Goldrush, Russet Norkotah, Dark Red Norland, Russet Burbank and Kennebec. The greatest level of leak control was achieved with the moderately resistant cv., Snowden, at 12.7%. Cultivars most likely to benefit from mefenoxam treatments should be targeted as part of a pink rot management programme. Judicious use of the fungicide, when matched with the level of cultivar susceptibility, may prove to be an efficient and effective approach to reduce infection rates and possibly manage mefenoxam resistance thereby maintaining longevity of the compound.  相似文献

The induction of pathogenesis-related (PR) proteins in sugarcane (Saccharum officinarum L.) leaves and suspension-cultured cells in response to treatment with a glycoprotein elicitor isolated from Colletotrichum falcatum (the red rot pathogen) was investigated. Treatment of leaves and cells with the elicitor resulted in a much marked increase in the activities of chitinase and β-1,3-glucanase in red rot resistant (BO 91) than susceptible (CoC 671) sugarcane cultivar. SDS-PAGE analysis revealed that C. falcatum elicitor induced the accumulation of several proteins in suspension-cultured cells of resistant cultivar (BO 91); among them the 35 kDa protein was predominant. Whereas, a 27 kDa protein was induced predominantly in the cells of susceptible cultivar upon treatment with the elicitor. When sugarcane leaves were treated with C. falcatum elicitor, two proteins with apparent molecular masses of 25 and 27 kDa were induced both in the resistant and susceptible cultivars. However, the induction was stronger in the resistant than the susceptible cultivar. Immunoblot analysis for chitinase indicated that a protein with an apparent molecular mass of 37 kDa cross-reacting with barley chitinase antiserum was strongly induced in the suspension cultured cells of both the cultivars. The induction of 37 kDa chitinase was more in the cells of resistant cultivar than in the susceptible cultivar. Western blot analysis revealed that a 25 kDa thaumatin-like protein (TLP) cross-reacting with bean TLP antiserum was strongly induced in leaves and cultured cells of both resistant and susceptible cultivars due to elicitor treatment.  相似文献

Investigations of the effects of elevated ozone (O(3)) on the virus-plant system were conducted to inform virus pathogen management strategies better. One susceptible cultivar of tobacco (Nicotiana tabacum L. cv. Yongding) and a resistant cultivar (Nicotiana tabacum L. cv. Vam) to Potato virus Y petiole necrosis strain (PVY(N)) infection were grown in open-top chambers under ambient and elevated O(3) concentrations. Above-ground biomass, foliage chlorophyll, nitrogen and total non-structural carbohydrate (TNCs), soluble protein, total amino acid (TAA) and nicotine content, and peroxidase (POD) activity were measured to estimate the effects of elevated O(3) on the impact of PVY(N) in the two cultivars. Results showed that under ambient O(3), the resistant cultivar possessed greater biomass and a lower C/N ratio after infection than the susceptible cultivar; however, under elevated O(3), the resistant cultivar lost its biomass advantage but maintained a lower C/N ratio. Variation of foliar POD activity could be explained as a resistance cost which was significantly correlated with biomass and C/N ratio of the tobacco cultivar. Chlorophyll content remained steady in the resistant cultivar but decreased significantly in the susceptible cultivar when stressors were applied. Foliar soluble protein and free amino acid content, which were related to resistance cost changes, are also discussed. This study indicated that a virus-resistant tobacco cultivar showed increased sensitivity to elevated O(3) compared to a virus-sensitive cultivar.  相似文献
