The patch-clamptechnique was used to determine the ionic conductances activated by ATPin murine colonic smooth muscle cells. Extracellular ATP, UTP, and2-methylthioadenosine 5'-triphosphate (2-MeS-ATP) increasedoutward currents in cells with amphotericin B-perforated patches. ATP(0.5-1 mM) did not affect whole cell currents of cells dialyzedwith solutions containing ethylene glycol-bis(-aminoethylether)-N,N,N',N'-tetraaceticacid. Apamin (3 × 107M) reduced the outward current activated by ATP by 32 ± 5%. Single channel recordings from cell-attached patches showed that ATP, UTP, and2-MeS-ATP increased the open probability of small-conductance, Ca2+-dependentK+ channels with a slopeconductance of 5.3 ± 0.02 pS. Caffeine (500 µM) enhanced the openprobability of the small-conductance K+ channels, and ATP had no effectafter caffeine. Pyridoxal phosphate 6-azophenyl-2',4'-disulfonic acid tetrasodium (PPADS,104 M), a nonselectiveP2 receptor antagonist, preventedthe increase in open probability caused by ATP and 2-MeS-ATP. PPADS hadno effect on the response to caffeine. ATP-induced hyperpolarization inthe murine colon may be mediated byP2y-induced release of Ca2+ from intracellular stores andactivation of the 5.3-pSCa2+-activatedK+ channels.

The effects ofintracellular nucleotide triphosphates on time-dependent changes inmuscarinic receptor cation currents (Icat) wereinvestigated using the whole cell patch-clamp technique in guinea pigileal muscle. In the absence of nucleotide phosphates in the patchpipette, Icat evoked every 10 min decayedprogressively. This decay was slowed dose dependently by inclusion ofmillimolar concentrations of ATP in the pipette. This required acomparable concentration of Mg2+, was mimicked by UTP andCTP, and was attenuated by simultaneous application of alkalinephosphatase or inhibitors of tyrosine kinase. In contrast, a suddenphotolytic release of millimolar ATP (probably in the free form) causeda marked suppression of Icat. Submillimolarconcentrations of GTP dose dependently increased the amplitude ofIcat as long as ATP and Mg2+ were inthe pipette, but, in their absence, GTP was ineffective at preventingIcat decay. The decay ofIcat was paralleled by altered voltage-dependentgating, i.e., a positive shift in the activation curve and reduction inthe maximal conductance. It is thus likely that ATP exerts tworeciprocal actions on Icat, throughMg2+-dependent and -independent mechanisms, and that theenhancing effect of GTP on Icat is essentiallydifferent from that of ATP.

The electrical conductance of the plasmalemma of cells of Charainflata, due to the diffusion of ions, consists predominantlyof K+, Cl and leak components. When the membrane electricalpotential difference is stepped in a negative direction witha voltage-clamp, the resulting inward current has componentsIK, ICl and IL (leak). During such voltage-clamp steps IK isinactivated, and Ic activated with voltage-dependent half-times.Increases in the external NaCl concentration reduce the magnitudeof IK and increase the magnitude of Ic, but reduce the half-timeof inactivation or activation. The NaCl-induced changes in Ikand ICl and their kinetics were more pronounced at pH0 =6.5than at pH0 =9.5. When the concentration of external CaCl2 wasincreased, Ik, ICl and the half-time of inactivation, (T1/2),of Ik were all reduced. The half-time of activation of ICl wasincreased. The NaCI-induced changes could result from increases in bothexternal ion concentration and osmotic pressure. Previous experimentshave shown that an increase in external osmotic pressure alonealters the properties of the conductances. The effect of oxidants on voltage-dependent K+ currents was examined in mouse colonic smooth muscle cells. Exposure to either chloramine-T (Ch-T), an agent known to oxidize both cysteine and methionine residues, or the colon-specific oxidant monochloramine (NH2Cl) completely suppressed the transient outward K+ current (Ito) while simultaneously enhancing the sustained delayed rectifier K+ current (Idr). In contrast, the cysteine-specific oxidants hydrogen peroxide (H2O2) and 5,5'-dithiobis(2-nitrobenzoic acid) (DTNB) exhibited partial and slow suppression of Ito by inducing a shift in channel availability of -18 mV without affecting Idr. After enhancement by NH2Cl or Ch-T, Idr was sensitive to 10 mM tetraethylammonium but not to other K+ channel blockers, suggesting that it represented activation of the resting Idr and not a separate K+ conductance. Extracellular dithiothreitol (DTT) partially reversed the effect of H2O2 and DTNB on Ito but not the actions of NH2Cl and Ch-T on either Idr or Ito. Dialysis of myocytes with GSH (5 mM) or DTT (5 mM) prevented suppression of Ito by H2O2 and DTNB but did not alter the effects of NH2Cl or Ch-T on either Idr or Ito. Ch-T and NH2Cl completely blocked Ito generated by murine Kv4.1, 4.2, and 4.3 in Xenopus oocytes, an effect not reversible by intracellular DTT. In contrast, intracellular DTT reversed the effect of H2O2 and DTNB on the cloned channels. It has been suggested that L-type Ca2+ channels play an important role in cell swelling-induced vasoconstriction. However, there is no direct evidence that Ca2+ channels in vascular smooth muscle are modulated by cell swelling. We tested the hypothesis that L-type Ca2+ channels in rabbit portal vein myocytes are modulated by hypotonic cell swelling via protein kinase activation. Ba2+ currents (IBa) through L-type Ca2+ channels were recorded in smooth muscle cells freshly isolated from rabbit portal vein with the conventional whole cell patch-clamp technique. Superfusion of cells with hypotonic solution reversibly enhanced Ca2+ channel activity but did not alter the voltage-dependent characteristics of Ca2+ channels. Bath application of selective inhibitors of protein kinase C (PKC), Ro-31–8425 or Go-6983, prevented IBa enhancement by hypotonic swelling, whereas the specific protein kinase A (PKA) inhibitor KT-5720 had no effect. Bath application of phorbol 12,13-dibutyrate (PDBu) significantly increased IBa under isotonic conditions and prevented current stimulation by hypotonic swelling. However, PDBu did not have any effect on IBa when cells were first exposed to hypotonic solution. Furthermore, downregulation of endogenous PKC by overnight treatment of cells with PDBu prevented current enhancement by hypotonic swelling. Mice are useful animal models to study pathogenic mechanisms involved in pulmonary vascular disease. Altered expression and function of voltage-gated K+ (KV) channels in pulmonary artery smooth muscle cells (PASMCs) have been implicated in the development of pulmonary arterial hypertension. KV currents (IK(V)) in mouse PASMCs have not been comprehensively characterized. The main focus of this study was to determine the biophysical and pharmacological properties of IK(V) in freshly dissociated mouse PASMCs with the patch-clamp technique. Three distinct whole cell IK(V) were identified based on the kinetics of activation and inactivation: rapidly activating and noninactivating currents (in 58% of the cells tested), rapidly activating and slowly inactivating currents (23%), and slowly activating and noninactivating currents (17%). Of the cells that demonstrated the rapidly activating noninactivating current, 69% showed IK(V) inhibition with 4-aminopyridine (4-AP), while 31% were unaffected. Whole cell IK(V) were very sensitive to tetraethylammonium (TEA), as 1 mM TEA decreased the current amplitude by 32% while it took 10 mM 4-AP to decrease IK(V) by a similar amount (37%). Contribution of Ca2+-activated K+ (KCa) channels to whole cell IK(V) was minimal, as neither pharmacological inhibition with charybdotoxin or iberiotoxin nor perfusion with Ca2+-free solution had an effect on the whole cell IK(V). Steady-state activation and inactivation curves revealed a window K+ current between –40 and –10 mV with a peak at –31.5 mV. Single-channel recordings revealed large-, intermediate-, and small-amplitude currents, with an averaged slope conductance of 119.4 ± 2.7, 79.8 ± 2.8, 46.0 ± 2.2, and 23.6 ± 0.6 pS, respectively. Ca2+-mediated agonists,including UTP, are being developed for therapeutic use in cysticfibrosis (CF) based on their ability to modulate alternativeCl conductances. As CF isalso characterized by hyperabsorption ofNa+, we determined the effect ofmucosal UTP on transepithelial Na+transport in primary cultures of human bronchial epithelia (HBE). Insymmetrical NaCl, UTP induced an initial increase in short-circuit current (Isc)followed by a sustained inhibition. To differentiate between effects onNa+ absorption andCl secretion,Isc was measuredin the absence of mucosal and serosal Cl(INa). Again,mucosal UTP induced an initial increase and then a sustained decreasethat reduced amiloride-sensitiveINa by 73%. TheCa2+-dependent agonists histamine,bradykinin, serosal UTP, and thapsigargin similarly induced sustainedinhibition (62-84%) ofINa. Mucosal UTPinduced similar sustained inhibition (half-maximal inhibitory concentration 296 nM) ofINa in primarycultures of human CF airway homozygous for the F508 mutation.BAPTA-AM blunted UTP-dependent inhibition ofINa, butinhibitors of protein kinase C (PKC) and phospholipaseA2 had no effect. Indeed, directactivation of PKC by phorbol 12-myristate 13-acetate failed to inhibitNa+ absorption. Apyrase, a tri-and diphosphatase, did not reverse inhibitory effects of UTP onINa, suggesting along-term inhibitory effect of UTP that is independent of receptoroccupancy. After establishment of a mucosa-to-serosaK+ concentration gradient andpermeabilization of the mucosal membrane with nystatin, mucosal UTPinduced an initial increase in K+current followed by a sustained inhibition. We conclude that increasingcellular Ca2+ induces a long-terminhibition of transepithelial Na+transport across normal and CF HBE at least partly due todownregulation of a basolateral membraneK+ conductance. Background

Uridine 5''-triphosphate (UTP) and uridine 5''-diphosphate (UDP) act via P2Y receptors to evoke contraction of rat pulmonary arteries, whilst adenosine 5''-triphosphate (ATP) acts via P2X and P2Y receptors. Pharmacological characterisation of these receptors in intact arteries is complicated by release and extracellular metabolism of nucleotides, so the aim of this study was to characterise the P2Y receptors under conditions that minimise these problems.

Methods

The perforated-patch clamp technique was used to record the Ca2+-dependent, Cl- current (ICl,Ca) activated by P2Y receptor agonists in acutely dissociated smooth muscle cells of rat small (SPA) and large (LPA) intrapulmonary arteries, held at -50 mV. Contractions to ATP were measured in isolated muscle rings. Data were compared by Student''s t test or one way ANOVA.

Results

ATP, UTP and UDP (10-4M) evoked oscillating, inward currents (peak = 13–727 pA) in 71–93% of cells. The first current was usually the largest and in the SPA the response to ATP was significantly greater than those to UTP or UDP (P < 0.05). Subsequent currents tended to decrease in amplitude, with a variable time-course, to a level that was significantly smaller for ATP (P < 0.05), UTP (P < 0.001) and UDP (P < 0.05) in the SPA. The frequency of oscillations was similar for each agonist (mean≈6–11.min-1) and changed little during agonist application. The non-selective P2 receptor antagonist suramin (10-4M) abolished currents evoked by ATP in SPA (n = 4) and LPA (n = 4), but pyridoxalphosphate-6-azophenyl-2'',4''-disulphonic acid (PPADS) (10-4M), also a non-selective P2 antagonist, had no effect (n = 4, 5 respectively). Currents elicited by UTP (n = 37) or UDP (n = 14) were unaffected by either antagonist. Contractions of SPA evoked by ATP were partially inhibited by PPADS (n = 4) and abolished by suramin (n = 5). Both antagonists abolished the contractions in LPA.

Conclusion

At least two P2Y subtypes couple to ICl,Ca in smooth muscle cells of rat SPA and LPA, with no apparent regional variation in their distribution. The suramin-sensitive, PPADS-resistant site activated by ATP most resembles the P2Y11 receptor. However, the suramin- and PPADS-insensitive receptor activated by UTP and UDP does not correspond to any of the known P2Y subtypes. These receptors likely play a significant role in nucleotide-induced vasoconstriction.  相似文献

Bovine adrenocortical zona fasciculata (AZF)cells express a novel ATP-dependent K+-permeable channel(IAC). Whole cell and single-channel recordings were used to characterize IAC channels withrespect to ionic selectivity, conductance, and modulation bynucleotides, inorganic phosphates, and angiotensin II (ANG II). Inoutside-out patch recordings, the activity of unitaryIAC channels is enhanced by ATP in the patchpipette. These channels were K+ selective with nomeasurable Na+ or Ca2+ conductance. Insymmetrical K+ solutions with physiological concentrationsof divalent cations (M2+), IACchannels were outwardly rectifying with outward and inward chordconductances of 94.5 and 27.0 pS, respectively. In the absence ofM2+, conductance was nearly ohmic. Hydrolysis-resistantnucleotides including AMP-PNP and NaUTP were more potent than MgATP asactivators of whole cell IAC currents. Inorganicpolytriphosphate (PPPi) dramatically enhancedIAC activity. In current-clamp recordings, nucleotides and PPPi produced resting potentials in AZFcells that correlated with their effectiveness in activatingIAC. ANG II (10 nM) inhibited whole cellIAC currents when patch pipettes contained 5 mMMgATP but was ineffective in the presence of 5 mM NaUTP and 1 mM MgATP.Inhibition by ANG II was not reduced by selective kinase antagonists.These results demonstrate that IAC is adistinctive K+-selective channel whose activity isincreased by nucleotide triphosphates and PPPi.Furthermore, they suggest a model for IAC gatingthat is controlled through a cycle of ATP binding and hydrolysis.

The role of nitric oxide (NO) in the occurrence of intracellular Ca2+ concentration ([Ca2+]i) oscillations in pituitary GH3 cells was evaluated by studying the effect of increasing or decreasing endogenous NO synthesis with L-arginine and nitro-L-arginine methyl ester (L-NAME), respectively. When NO synthesis was blocked with L-NAME (1 mM) [Ca2+]i, oscillations disappeared in 68% of spontaneously active cells, whereas 41% of the quiescent cells showed [Ca2+]i oscillations in response to the NO synthase (NOS) substrate L-arginine (10 mM). This effect was reproduced by the NO donors NOC-18 and S-nitroso-N-acetylpenicillamine (SNAP). NOC-18 was ineffective in the presence of the L-type voltage-dependent Ca2+ channels (VDCC) blocker nimodipine (1 µM) or in Ca2+-free medium. Conversely, its effect was preserved when Ca2+ release from intracellular Ca2+ stores was inhibited either with the ryanodine-receptor blocker ryanodine (500 µM) or with the inositol 1,4,5-trisphosphate receptor blocker xestospongin C (3 µM). These results suggest that NO induces the appearance of [Ca2+]i oscillations by determining Ca2+ influx. Patch-clamp experiments excluded that NO acted directly on VDCC but suggested that NO determined membrane depolarization because of the inhibition of voltage-gated K+ channels. NOC-18 and SNAP caused a decrease in the amplitude of slow-inactivating (IDR) and ether-à-go-go-related gene (ERG) hyperpolarization-evoked, deactivating K+ currents. Similar results were obtained when GH3 cells were treated with L-arginine. A Cl current activated by extracellular acidification, ICl(pHac), has been characterized in various mammalian cell types. Many of the properties of ICl(pHac) are similar to those of the cell swelling-activated Cl current ICl(swell): ion selectivity (I > Br > Cl > F), pharmacology [ICl(pHac) is inhibited by 4,4'-diisothiocyanostilbene-2,2'-disulfonic acid (DIDS), 1,9-dideoxyforskolin (DDFSK), diphenylamine-2-carboxylic acid (DPC), and niflumic acid], lack of dependence on intra- or extracellular Ca2+, and presence in all cell types tested. ICl(pHac) differs from ICl(swell) in three aspects: 1) its rate of activation and inactivation is very much more rapid, currents reaching a maximum in seconds rather than minutes; 2) it exhibits a slow voltage-dependent activation in contrast to the fast voltage-dependent activation and time- and voltage-dependent inactivation observed for ICl(swell); and 3) it shows a more pronounced outward rectification. Despite these differences, study of the transition between the two currents strongly suggests that ICl(swell) and ICl(pHac) are related and that extracellular acidification reflects a novel stimulus for activating ICl(swell) that, additionally, alters the biophysical properties of the channel. cell swelling-activated chloride current; patch clamp; pH  相似文献

We examined the effectsof metabolic inhibition on intracellular Ca2+ release insingle pulmonary arterial smooth muscle cells (PASMCs). Severemetabolic inhibition with cyanide (CN, 10 mM) increased intracellularcalcium concentration ([Ca2+]i) and activatedCa2+-activated Cl currents[ICl(Ca)] in PASMCs, responses that were greatlyinhibited by BAPTA-AM or caffeine. Mild metabolic inhibition with CN (1 mM) increased spontaneous transient inward currents andCa2+ sparks in PASMCs. In Xenopus oocytes, CNalso induced Ca2+ release and activatedICl(Ca), and these responses were inhibited by thapsigarginand cyclopiazonic acid to deplete sarcoplasmic reticulum (SR)Ca2+, whereas neither heparin nor anti-inositol1,4,5-trisphosphate receptor (IP3R) antibodies affected CNresponses. In both PASMCs and oocytes, CN-evoked Ca2+release was inhibited by carbonyl cyanidem-chlorophenylhydrazone (CCCP) and oligomycin or CCCP andthapsigargin. Whereas hypoxic stimuli resulted in Ca2+release in pulmonary but not mesenteric artery myocytes, CN induced release in both cell types. We conclude that metabolic inhibition withCN increases [Ca2+]i in both pulmonary andsystemic artery myocytes by stimulating Ca2+ release fromthe SR and mitochondria.

Cell shrinkageis an early prerequisite in programmed cell death, and cytoplasmicK+ is a dominant cation that controls intracellular ionhomeostasis and cell volume. Blockade of K+ channelsinhibits apoptotic cell shrinkage and attenuates apoptosis. We examined whether apoptotic repressor with caspase recruitment domain (ARC), an antiapoptotic protein, inhibits cardiomyocyte apoptosis by reducing K+ efflux throughvoltage-gated K+ (Kv) channels. In heart-derived H9c2cells, whole cell Kv currents (IK(V)) wereisolated by using Ca2+-free extracellular (bath) solutionand including 5 mM ATP and 10 mM EGTA in the intracellular (pipette)solution. Extracellular application of 5 mM 4-aminopyridine (4-AP), ablocker of Kv channels, reversibly reduced IK(V)by 50-60% in H9c2 cells. The remaining currents during 4-APtreatment may be generated by K+ efflux through4-AP-insensitive K+ channels. Overexpression of ARC inheart-derived H9c2 cells significantly decreasedIK(V), whereas treatment with staurosporine, apotent apoptosis inducer, enhanced IK(V)in wild-type cells. The staurosporine-induced increase inIK(V) was significantly suppressed and thestaurosporine-mediated apoptosis was markedly inhibited incells overexpressing ARC compared with cells transfected with thecontrol neomycin vector. These results suggest that theantiapoptotic effect of ARC is, in part, due to inhibition of Kvchannels in cardiomyocytes.

To examine effects of cytosolicNa+, K+, and Cs+ on the voltagedependence of the Na+-K+ pump, we measuredNa+-K+ pump current (Ip)of ventricular myocytes voltage-clamped at potentials(Vm) from 100 to +60 mV. Superfusates weredesigned to eliminate voltage dependence at extracellular pump sites.The cytosolic compartment of myocytes was perfused with patch pipette solutions with a Na+ concentration ([Na]pip)of 80 mM and a K+ concentration from 0 to 80 mM or withsolutions containing Na+ in concentrations from 0.1 to 100 mM and K+ in a concentration of either 0 or 80 mM. When[Na]pip was 80 mM, K+ in pipette solutionshad a voltage-dependent inhibitory effect on Ipand induced a negative slope of theIp-Vm relationship. Cs+ in pipette solutions had an effect onIp qualitatively similar to that ofK+. Increases in Ip with increasesin [Na]pip were voltage dependent. The dielectriccoefficient derived from[Na]pip-Ip relationships at thedifferent test potentials was 0.15 when pipette solutions included 80 mM K+ and 0.06 when pipette solutions were K+ free.

The hyperpolarization-activated, cyclic nucleotide-gated (HCN) channels, or cardiac (If)/neuronal (Ih) time- and voltage-dependent inward cation current channels, are conventionally considered as monovalent-selective channels. Recently we discovered that calcium ions can permeate through HCN4 and Ih channels in neurons. This raises the possibility of Ca2+ permeation in If, the Ih counterpart in cardiac myocytes, because of their structural homology. We performed simultaneous measurement of fura-2 Ca2+ signals and whole cell currents produced by HCN2 and HCN4 channels (the 2 cardiac isoforms present in ventricles) expressed in HEK293 cells and by If in rat ventricular myocytes. We observed Ca2+ influx when HCN/If channels were activated. Ca2+ influx was increased with stronger hyperpolarization or longer pulse duration. Cesium, an If channel blocker, inhibited If and Ca2+ influx at the same time. Quantitative analysis revealed that Ca2+ flux contributed to 0.5% of current produced by the HCN2 channel or If. The associated increase in Ca2+ influx was also observed in spontaneously hypertensive rat (SHR) myocytes in which If current density is higher than that of normotensive rat ventricle. In the absence of EGTA (a Ca2+ chelator), preactivation of If channels significantly reduced the action potential duration, and the effect was blocked by another selective If channel blocker, ZD-7288. In the presence of EGTA, however, preactivation of If channels had no effects on action potential duration. Increased extracellular osmolarity ([Os]e) suppresses stimulated hormone secretion from anterior pituitary cells. Ca2+ influx may mediate this effect. We show that increase in [Os]e (by 18–125%) differentially suppresses L-type and T-type Ca2+ channel currents (IL and IT, respectively); IL was more sensitive than IT. Hyperosmotic suppression of IL depended on the magnitude of increase in [Os]e and was correlated with the percent decrease in pituitary cell volume, suggesting that pituitary cell shrinkage can modulate L-type currents. The hyperosmotic suppression of IL and IT persisted after incubation of pituitary cells either with the actin-disrupter cytochalasin D or with the actin stabilizer phalloidin, suggesting that the actin cytoskeleton is not involved in this modulation. The hyperosmotic suppression of Ca2+ influx was not correlated with changes in reversal potential, membrane capacitance, and access resistance. Together, these results suggest that the hyperosmotic suppression of Ca2+ influx involves Ca2+ channel proteins. We therefore recorded the activity of L-type Ca2+ channels from cell-attached patches while exposing the cell outside the patch pipette to hyperosmotic media. Increased [Os]e reduced the activity of Ca2+ channels but did not change single-channel conductance. This hyperosmotic suppression of Ca2+ currents may therefore contribute to the previously reported hyperosmotic suppression of hormone secretion. Inwardlyrectifying K+ current(IKir) infreshly isolated bovine retinal pigment epithelial (RPE) cells wasstudied in the whole cell recording configuration of the patch-clamptechnique. When cells were dialyzed with pipette solution containing noATP, IKir randown completely in <10 min [half time(t1/2) = 1.9 min]. In contrast, dialysis with 2 mM ATP sustainedIKir for 10 min or more. Rundown was also prevented with 4 mM GTP or ADP. When 0.5 mMATP was used,IKir ran down by~71%. Mg2+ was a criticalcofactor because rundown occurred when the pipette solution contained 4 mM ATP but no Mg2+(t1/2 = 1.8 min).IKir also randown when the pipette solution contained 4 mMMg2+ + 4 mM5'-adenylylimidodiphosphate(t1/2 = 2.7 min)or 4 mM adenosine 5'-O-(3-thiotriphosphate)(t1/2 = 1.9 min),nonhydrolyzable and poorly hydrolyzable ATP analogs, respectively. Weconclude that the sustained activity ofIKirin bovine RPE requires intracellular MgATP and that the underlyingmechanism may involve ATP hydrolysis.

Ionic currents elicited via purinergic receptors located in themembrane of Xenopus follicles werestudied using electrophysiological techniques. Follicles responded toATP-activating inward currents with a fast time course(Fin). InRinger solution, reversal potential (Erev) ofFin was 22mV, which did not change with external substitutions ofNa+ orK+, whereas solutions containing50 or 5% of normal Clconcentration shiftedErev to about +4and +60 mV, respectively, and decreasedFin amplitude,indicating thatFin was carriedby Cl.Fin had an onsetdelay of ~400 ms, measured by application of a brief jet of ATP froma micropipette positioned near the follicle (50 µm).Fin was inhibitedby 50% in follicles pretreated with pertussis toxin. This suggests a Gprotein-mediated receptor channel pathway.Fin was mimickedby 2-MeSATP and UTP, the potency order (half-maximal effectiveconcentration) was 2-MeSATP (194 nM) > UTP (454 nM) > ATP(1,086 nM). All agonists generatedCl currents and displayedcross-inhibition on the others.Fin activation byacetylcholine also cross-inhibitedFin-ATPresponses, suggesting that all act on a common channel-activationpathway.

Bone marrow mesenchymal stem cells (MSCs) are used as a cell source for cardiomyoplasty; however, the cellular electrophysiological properties are not fully understood. The present study was to investigate the functional ionic channels in undifferentiated mouse bone marrow MSCs using whole cell patch-voltage clamp technique, RT-PCR, and Western immunoblotting analysis. We found that three types of ionic currents were present in mouse MSCs, including a Ca2+-activated K+ current (IKCa), an inwardly rectifying K+ current (IKir), and a chloride current (ICl). IKir was inhibited by Ba2+, and IKCa was activated by the Ca2+ ionophore A-23187 and inhibited by the intermediate-conductance IKCa channel blocker clotrimazole. ICl was activated by hyposmotic (0.8 T) conditions and inhibited by the chloride channel blockers DIDS and NPPB. The corresponding ion channel genes and proteins, KCa3.1 for IKCa, Kir2.1 for IKir, and Clcn3 for ICl, were confirmed by RT-PCR and Western immunoblotting analysis in mouse MSCs. Pulmonary vasoconstriction and vascularmedial hypertrophy greatly contribute to the elevated pulmonaryvascular resistance in patients with pulmonary hypertension. A rise incytosolic free Ca2+ ([Ca2+]cyt)in pulmonary artery smooth muscle cells (PASMC) triggers vasoconstriction and stimulates cell growth. Membrane potential (Em) regulates[Ca2+]cyt by governing Ca2+influx through voltage-dependent Ca2+ channels. Thusintracellular Ca2+ may serve as a shared signaltransduction element that leads to pulmonary vasoconstriction andvascular remodeling. In PASMC, activity of voltage-gated K+(Kv) channels regulates resting Em. In thisstudy, we investigated whether changes of Kv currents[IK(V)], Em, and[Ca2+]cyt affect cell growth by comparingthese parameters in proliferating and growth-arrested PASMC. Serumdeprivation induced growth arrest of PASMC, whereas chelation ofextracellular Ca2+ abolished PASMC growth. Resting[Ca2+]cyt was significantly higher, andresting Em was more depolarized, inproliferating PASMC than in growth-arrested cells. Consistently, wholecell IK(V) was significantly attenuated in PASMCduring proliferation. Furthermore, Emdepolarization significantly increased resting[Ca2+]cyt and augmented agonist-mediatedrises in [Ca2+]cyt in the absence ofextracellular Ca2+. These results demonstrate that reducedIK(V), depolarized Em, and elevated [Ca2+]cyt may play a criticalrole in stimulating PASMC proliferation. Pulmonary vascular medialhypertrophy in patients with pulmonary hypertension may be partlycaused by a membrane depolarization-mediated increase in[Ca2+]cyt in PASMC.
