The morphology and morphogenesis of a new oxytrichid ciliate, Notohymena limus n. sp. were studied in vivo and after protargol impregnation. The new ciliate was isolated from the sewage sludge at Delhi Jal Board Sewage Treatment Plant located at Rithala, Delhi, India, using the non-flooded Petri dish method. N. limus n. sp. is characterized as follows: flexible dorsoventrally flattened ellipsoidal body; Notohymena-pattern undulating membranes; adoral zone of membranelles (AZM) occupied about 39% of the body length, and consists of around 26 membranelles; large and deep buccal cavity; colorless subpellicular granules present in groups and arranged around the bases of dorsal bristles; 4 macronuclear nodules; 2 micronuclei; 18 fronto-ventral-transverse (FVT) cirri in typical Oxytricha-pattern; 6 dorsal rows of bristles; 3 caudal cirri; about 16 right and 15 left marginal cirri; N. limus n. sp. is a new species on the basis of the combination of morphological, morphometric and morphogenetic characteristic features.  相似文献

SYNOPSIS. Kinetosomal changes, as indicative of cytoplasmic reorganization in binary fission and during and after conjugation, were followed in a zoochlorellae-bearing species of Euplotes. The preconjugant peristome, designated the first generation peristome, breaks down partially after the conjugants have paired; the basal section, comprising the shorter adoral membranelles and the undulating, membrane, is resorbed. A new peristome, the second generation peristome, arises as a small pit near the left ventral margin, in midline, at the time when the micronuclei are in the first meiotic prophase. By the time of the second meiotic division a single set of new cirri, the second generation cirri, has formed in each conjugant. This second set is not perfect, lacking one of the frontals. Neither the second generation peristome nor cirri develop very far, or migrate, until after separation of the conjugants. Then the new peristome replaces the old one and the new cirri become functional. However, the new peristome lacks an undulating membrane and does not complete its development, bearing only a fraction of the normal number of membranelles. At its posterior termination, at the time of condensation of the macronuclear anlage, another peristome, the third generation peristome, is formed and develops as a granular, and later striated, invagination extending posteriorly. It appears to integrate with its predecessor and, as its constituent membranelles develop, a third generation single set of new cirri arises. These replace the imperfect previous set, all of the cirri being represented. In anticipation of the first postconjugant fission, all of the cirral apparatus is discarded again and two new sets (fourth generation cirri) originate; the old (combination second and third generation) peristome is retained by the proter while a new one is provided for the opisthe. It is evident, therefore, that a rather far-reaching cytoplasmic reorganization accompanies the nuclear changes of conjugation, seeming, for the most part, to follow the nuclear changes. The old macronuclear fragments have been found not to fuse with the macronuclear anlage.  相似文献

The morphology and infraciliature of a new ciliate, Kiitricha minuta n. sp., isolated from the Yellow Sea, were investigated using live observation and protargol impregnation. Kiitricha minuta represents a third member of the rarely known order Kiitrichida. It is unique in the subclass Hypotrichia in having many rows of small uniform cirri along the right side of the body and the dorsal kineties composed of dikinetids, most of which bear two cilia. Kiitricha minuta n. sp. is ovoid and measures about 60 x 45 microm in vivo. It has a huge buccal cavity occupying about 80% of the body length, numerous body extrusomes, one macronucleus and two micronuclei, 27-27 adoral membranelles, 9-12 frontoventral cirral rows, a submarginal row of 7-9 cirri, 6 or 7 transverse cirri, and roughly 7-9 dorsal kineties. This new species differs distinctly from its only congener Kiitricha marina by its smaller size (60 microm vs. 80-150 microm), the presence of body extrusomes (vs. absent), the different macronuclear pattern (one vs. two nodules), and the lower number of frontoventral cirral rows (9-12 vs. 21-26), which terminate at the anterior two-thirds of body (vs. extend to the posterior). The new term "submarginal cirral row" is introduced to distinguish from the marginal cirral row in typical hypotrichs sensu lato. Based on our new observations and the literature, an improved diagnosis for the genus Kiitricha is provided and its phylogenetic importance is discussed.  相似文献

Soil samples were taken from a temporary pond located in Buenos Aires province, Argentina, during the dry phase in the summer of 2005. The ciliates were studied alive and after staining with protargol. Deviata rositae n. sp. measures 112-154 microm in length and 21-28 microm in width in vivo and has a vermiform body. The contractile vacuole is located in the mid-body on the left. The macronucleus is moniliform and there are 1-3 micronuclei. The oral apparatus is composed of 14-18 adoral membranelles and straight paroral and endoral membranes that never intersect each other. The somatic ciliature is arranged in four frontal cirri, one buccal cirrus, six long and slightly spiraled rows of cirri with the first right row extending up to the equatorial or sub-equatorial region, and two dorsal rows of dikinetids. This new species of Deviata primarily differs from its congeners by the number of macronuclear nodules and the number and disposition of the dorsal rows of dikinetids.  相似文献

Leptoamphisiella vermisGruber, 1888 n. g., n. comb. (basionym Epiclintes vermisGruber, 1888) is an extraordinarily large and worm-like marine stichotrichous ciliate. Based on a population isolated recently from coastal waters of Qingdao, China, the living morphology and infraciliature are redescribed and its taxonomic position is defined. Accordingly, a new diagnosis for this species is suggested: large, marine Leptoamphisiella with a conspicuous layer of pellicular alveoli; 400-1000 microm x 40-70 microm in vivo; body band-like, highly flexible; about 40 membranelles; always three frontal and two buccal cirri; 52-80 transverse cirri (TC) extending to the posterior end of the buccal field; 57-79 left midventral and 44-62 right midventral cirri; 62-102 cirri in left and 63-91 cirri in right marginal rows (MR); 9-13 dorsal kineties that extend the full body length; about 100 macronuclear nodules and 5-13 micronuclei. The diagnosis for the new genus is as follows: vermiform Pseudoamphisiellidae with strongly contractile body, differentiated frontal, buccal, and highly developed TC; two remarkably separated midventral rows; one MR on each side of the body; frontoterminal and caudal cirri absent. Leptoamphisiella vermis n. comb. is fixed as the type species of the new genus.  相似文献

A new hypotrichous ciliate, Apoterritricha lutea n. g., n. sp., was discovered in a sample of a terrestrial liverwort from Korea. Its morphology was studied using detailed in vivo observation and protargol impregnation. Its phylogenetic relationships were revealed by analyses of the 18S rRNA gene. This new taxon is characterized by a combination of the following traits: (i) ellipsoidal to narrowly ellipsoidal body with an average size of 230 × 85 μm; (ii) two macronuclear nodules and two to five micronuclei; (iii) golden yellow cortical granules, forming small groups along the microtubular appendages of cirri, adoral membranelles, and dorsal kineties; (iv) typically three frontal cirri, one buccal cirrus, four frontoventral cirri, seven midventral cirri, two pretransverse cirri, seven transverse cirri, ca. 38 left, and ca. 36 right marginal cirri; and (v) on average six dorsal kineties, three dorsomarginal kineties, and three caudal cirri. In molecular phylogenies, A. lutea clusters with strong support within a clade containing Afrokeronopsis aurea and several “typical” oxytrichids having golden yellow to brown cortical granules. In this light we propose a hypothesis that is not unambiguously rejected by the present phylogenetic analyses, which shows how the Afrokeronopsis‐like pattern could have evolved from a Rubrioxytricha‐like ancestor via an Apoterritricha‐like stage by cirri‐multiplication.  相似文献

The micronuclei of the ciliated protozoan Pseudourostyla cristata were eliminated by amputation shortly before binary fusion. The amicronucleate cell lines derived from regenerants were maintained for more than a year. They exhibited a lower viability and reduced vigor in asexual propagation. There was some improvement in the growth of the cell lines 1 mo after operation, but the growth rate remained subnormal even up to 1 yr of culture. The exact cause of the poor growth and survival in the first 3 wk after operation, whether the loss of the micronucleus or operational damage, remains to be determined. It is nevertheless clear that the micronucleus is important for subsequent asexual propagation. The amicronucleate cell lines were permanently crippled in morphogenesis, unlike the situation in Paramecium amicronucleates in which stomatogenesis returned to near-normal during asexual propagation. They always included some cells with a characteristically defective adoral zone of membranelles, reduced number of frontal-ventral-transverse cirri, and reduced body length. They were also reluctant to encyst. It is evident that the micronucleus is important for maintaining normality of the oral apparatus. It is postulated that the permanent stomatogenic crippling of amicronucleates might be related to genomic reduction in the developing macronucleus in sexual reproduction, as exhibited by other hypotrichs. The morphological defects associated with the adoral zone of membranelles may be rationalized as arising from the spreading of a zone of degeneration in the cortex affecting the left edge of the membranelles.  相似文献

Morphogenetic events during the division of the marine hypotrichous ciliate, Hemigastrostyla enigmatica (Dragesco & Dragesco-Kernéis, 1986) Song & Wilbert, 1997 are described. The morphogenesis is characterized by:(1) 5 frontoventral-transverse cirral anlagen develop into 8 frontal, 5 ventral and 5 transverse cirri after Oxytricha-pattern;(2) there may be 6 FVT-anlagen in some individuals giving rise to more cirri which, however, will be resorbed after division;(3) anlage of the right marginal row at least in the opisthe occurs de novo right to the parental structures instead of within them;(4) according to the origin, the two extra ventral cirri right to transverse ones are not ventral or transverse cirri, which are from the retained old structure;(5) dorsal kineties originate from one group of DK-anlagen in both dividers with, very uniquely, an additional fragmentation of DK1, and(6) oral primordia will be formed in both dividing parts, from which the newly-built membranelles in the proter replace the posterior part of the parental AZM with a particular 'piecing together mode. Some features during the morphogenesis (e.g. variable number of cirral anlagen, presence of primary primordia, the mode of rebuilding of the proter's adoral zone of membranelles, origin of dorsal kineties and caudal cirri etc.) indicate that the genus Hemigastrostyla might present a intermediate form between oxytrichids and other related higher taxa. Based on our new observations, an improved diagnosis for genus Hemigastrostyla is given: marine or brackish water Oxytrichidae with slightly to conspicuously cephalized body shape; mostly 8–10 frontal, 5 ventral, 5 transverse and two to several extra ventral cirri to the right of the transverse ones, which are from the retained parental structure; caudal cirri present.  相似文献

In a study on ciliate diversity, we discovered the new hypotrich species, Gonostomum jangbogoensis n. sp., in freshwater from Terra Nova Bay, Victoria Land, southeast Antarctica. We describe its morphology and morphogenesis using standard methods, and the SSU rRNA gene phylogeny is provided as well. Morphology of Gonostomum jangbogoensis n. sp. is characterized as follows: slender to elongated body shape; grayish under low magnification; cortical granules present; 32–41 adoral membranelles; 3 enlarged frontal cirri; 1 buccal cirrus; 2 frontoterminal cirri; 3 or 4 frontoventral cirral pairs, 2 pretransverse cirri, 6–7 transverse cirri; 13–19 left and 18–26 right marginal cirri; 17–23 paroral kinetids; 3 dorsal kineties; 3 caudal cirri; 2 macronuclear nodules with 1–3 micronuclei. The morphogenesis of the new species confirms that it has at least seven frontal-ventral-transverse cirral anlagen, which is also reported in Gonostomum sp. 1 sensu Shin from Korea. Even though these two populations occur very far from each other, the morphometric data prove that this character state, the seven cirral anlagen, is a stable feature across these populations and might be an apomorphy. The phylogenetic analyses show that the genus Gonostomum is non-monophyletic and that the new species is a sister to G. bromelicola.  相似文献

Morphogenesis during the binary fission of the stichotrich ciliate Pseudoamphisiella alveolata, isolated from Jiaozhou Bay near Qingdao, China, was investigated using protargol silver impregnation. The process is characterized as follows: (1) in the proter, only the posterior part of the parental adoral zone of membranelles is renewed, where the membranelles dedifferentiate and then rebuild the UM-anlage and the missing membranelles, (2) the oral primordium in the opisthe and the FVT-anlagen in both dividers are formed de novo on the cell surface, (3) an "extra" anlage, which is generated on the right of the right marginal anlage, develops into three or four "extra" marginal cirri that connect the caudal cirri with the marginal rows, (4) the right marginal anlage is formed within the old structure, (5) the FVT-cirri develop in a primary mode, and (6) unlike most stichotrichs, the right marginal anlagen in both dividers generate closely together. As an additional contribution, the diversity of morphogenetic patterns within the genus Pseudoamphisiella is discussed. Based on both morphogenetic and SS rRNA gene sequencing data, the systematic position of the genus Pseudoamphisiella as well as the family Pseudoamphisiellidae Song et al. 1997 is briefly analyzed. The results indicate that they should very possibly represent a higher evolved group in the order Urostylida.  相似文献

Summary The morphological modification of ventral cilia associated with cell paralysis of Euplotes when attacked by the predator Litonotus are analyzed by electron microscopy (both SEM and TEM). The locomotory organelles of Euplotes (membranelles and cirri) lose their typical compact morphology. Single cilia of both organelles undergo a modification at their terminal portion. At this level, the axoneme becomes folded, and the ciliary membrane enlarges into a conspicuous rounded bulbus. The possible nature of this bulbus and the different behaviour of membranelles, cirri, and dorsal bristles are discussed.  相似文献

The morphology and morphogenesis of a new soil hypotrich ciliate, Sterkiella multicirrata sp. nov., was investigated using live observation and protargol staining. The new species is characterised by: body elliptical, 110–180 × 45–75 μm in vivo, an average of 35 adoral membranelles; usually 19 frontoventral‐transverse cirri, consisting of three frontal, five frontoventral, one buccal, four postoral ventral, two pretransverse, and four transverse cirri; four macronuclear nodules, and 2–5 micronuclei. S. multicirrata sp. nov. differs from its congeners mainly in the number of frontoventral‐transverse cirri and macronuclear nodules. Morphogenesis of the new species is similar to its congeners; the primary difference exists in the segmentation of the frontoventral‐transverse cirral anlagen, which is usually generated in a 1:2:3:3:5:5 pattern. Based on the small subunit ribosomal DNA sequence, the phylogenetic position of the new species is discussed.  相似文献

In Pleurotricha lanceolata, the ventral somatic infraciliature presents 13 frontoventral cirri, 5 transverse cirri, one row with 18–19 left marginal cirri and two rows of right marginal cirri of different length. On the dorsal side there are six longitudinal rows of dorsal bristles, four of them bipolar and the other two less than half body length. The oral infraciliature includes the adoral zone of membranelles, with 45–55 membranelles of three or four rows of kinetosomes each, and two undulating membranes (paroral and endoral membranes), each with two rows of kinetosomes. Some structures of the oral and somatic fibrillar systems have also been examined and are similar to those described in other species of hypotrichous ciliates.  相似文献
