Horizontal starch gel electrophoresis has been applied to thestudy of esterase, peroxidase and acid phosphatase patternsin seven taxa, namely Hordeum diploids (2n=14) (H. marinum,H. marinum I and H. hystrix), tetraploids (2n=28) (H. bulbosumand H. murinum subsp. leporinum) and Taeniatherum (2n=14) (T.caput-medusae and T. caput-medusae I) in order to elucidatetheir phylogenetic relationships. On the basis of our experimentalresults the seven taxa may be placed in the following threegroups; (1) diploid Hordeum (H. marinum, H. marinum I, H. hystrix);(2) tetraploid Hordeum (H. bulbosum, H. murinum subsp. leporinum);(3) Taeniatherum (T. caput-medusae, T. caput-medusae I). Esterase, peroxidase and acid phosphatase patterns of the twoHordeum diploid taxa (H. marinum and H. marinum I) are verysimilar suggesting their close phylogenetic relationship; thesame is true for both the taxa of the genus Taeniatherum (T.caput-medusae and T. caput-medusae I). The taxa of the Taeniatherumgroup compared with the diploid Hordeum (H. marinum, H. marinumI, H. hystrix) and the tetraploid Hordeum (H. bulbosum, H. murinumsubsp. leporinum) show a lower degree of phylogenetic relationshipand seem to be equally distant from them. The tetraploid Hordeumgroup shows a higher phylogenetic relationship with diploidHordeum group than with the Taeniatherum group. These results confirm that the genus Taeniatherum, previouslyconsidered as part of the genus Hordeum, should be regardedas a separate genus. The neogastropod family Fasciolariidae contains a complex ofgenera related to Latirus Montfort, 1810, many of which havetraits unusual for the family. In a taxonomic revision of someof these genera, based on shell characters, we restrict Latirusto a mainly Indo-West Pacific group of Pliocene to Recent species,which closely resemble the middle Miocene to Recent pantropicalgenus Hemipolygona Rovereto, 1899. Hemipolygona stenomphalus(Habe & Kosuge, 1966) is synonymized with H. recurvirostris(Schubert & Wagner, 1829). Lathyropsis Oostingh, 1939, basedon a small Pliocene species from Indonesia, is here tentativelysubsumed under Polygona Schumacher, 1817. The latter genus,ranging from the late Oligocene to Recent, occurs mainly inthe New World and eastern Atlantic, and contains at least twospecies groups centered on P. infundibulum Schumacher, 1817(type of genus) and P. angulatus (Röding, 1798). Taxa assignedby many authors to Latirulus Cossmann, 1889, are here reassignedto the new genus Turrilatirus (type species: Voluta turritaGmelin, 1791), from the Pliocene to Recent of the Indo-WestPacific. Latirulus is restricted to Eocene species. We assignvarious early Miocene to Recent species from the western Atlanticand eastern Pacific to the new genus Pustulatirus (type species:Latirus mediamericanus, Hertlein & Strong, 1951a). Taxaformerly assigned to Latirus but here removed from Fasciolariidaeinclude Latirus ewekoroensis Adegoke, 1977; the Eocene RusculaCasey, 1904; Lathyrus granifer and L. compactilis, both of Martin,1931; Latirus kirbyi Clark, 1938; Latirus tortilis var. nanafaliusHarris, 1899; and Latirus quercotillaensis Olsson, 1931. The epidermal structure of the five species of ferns, Arthromeriswallichiana (Spr.) Ching., Drymoglossum piloselloides (Prest.),Drynaria quercifolia (L.) J. Smith, Lepisorus nudus (Hook.)Ching. and Pyrrosia nuda (Gies.) Ching., has been investigated.Fifteen types of stomatal structures have been identified ofwhich copolo-desmocytic and coperi-desmocytic are new types.Four more possible stomatal structures: ccpolo-peri-, codesmo-polo-,codesmo-peri- and duplodesmocytic, are suggested. Localizationof starch, insoluble polysaccharides, protein and lipids hasbeen examined histochemically in the guard cells, subsidiarycells and epidermal cells. In Drynaria starch plastids and plastidscontaining both starch and protein are present in guard cells.Starch plastids are present in the subsidiary cells of all speciesexcept in Arthromeris, whereas, they are present in epidermalcells of only Drymoglossum and Lepisorus. Granular or amorphousinsoluble polysaccharides (other than starch) are present inguard cells of all the species, in the subsidiary cells of Arthromeris,Drynaria and Pyrrosia, and in the epidermal cells of Pyrrosia.Except in Pyrrosia lipids are present in the guard cells. Subsidiarycells of Drynaria and the epidermal cells of Arthromeris andDrynaria show lipid bodies. The presence of plasmodesmata andectodesmata is demonstrated in the epidermal cells of Drymoglossum.  相似文献

The stay-green mutations cytG and Gd1d2 prevent the normal yellowingduring senescence of soybean (Glycine max) leaves and cotyledons.Because light plays such an important role in regulating morphogenesisand it promotes the formation of chlorophyll (Chl), we determinedthe effect of cytG and Gd1d2 (in a cv. Clark background) onthe development and some light responses of seedlings. AlthoughcytG and Gd1d2 seeds, particularly the cotyledons, are greenwhen mature, 44 and 71 % respectively of this Chl broke downwhen the seeds were germinated in darkness. Chlorophyllidesand phaeophytins were not present in the seeds in significantamounts. cytG and Gd1d2 as well as wild type (cv. Clark) seedlingsdeveloped a full etiolation syndrome (morphology and lack ofChl) in darkness. Light induced rapid Chl accumulation in thedark-grown seedlings with no apparent difference among the threeisolines. A short (8 h) exposure to light induced some Chl inthe cotyledons of dark-grown plants, and 22 h of light producedfour times more. Following return to darkness, the 8-h groupshowed very little breakdown over the next 12 d. Studies on the changes in leaf number of 12 species of Betulaceaewere carried out by repeated observations during several years.The inner structure of the winter buds of these species wasalso examined. The periods of leaf emergence were long in Alnusand Betula, intermediate in Corylus and short in Ostrya andCarpinus. A heavy summer leaf fall is characteristic only ofthe Gymnothyrsus subgenus of Alnus. One lamina and two stipulesis the basic unit constituting the winter buds. Only the budsof Gymnothyrsus are composed of several of these units. In theAlnaster subgenus of Alnus, Betula and Corylus, there are oneor two scales that seem to have originated from the two stipulesremaining after reduction of the lamina. Eight and 24 scalesof similar origin were found in Ostrya and Carpinus respectively.A common ancestral species having the following primitive charactersis proposed: shoot constituted only of the equal-sized units,period of leaf emergence long, and leaf fall usually occurs.Alnus (Gymnothyrsus) is assumed to be the most primitive typeas it has many characters similar to those of the proposed ancestralspecies. Carpinus is assumed to be the most advanced group.The leaf survivorship curve is assumed to have changed fromthe primitive bell-shaped to an advanced trapezoid concurrentwith an increase in the number of bud scales. We show that, when the three-way association level among thethree binary variables, X, U1 and U2 is fixed, DP = pr(X = 1¦U1= 1) – pr(X = 1¦U1 = 0) increases as the cross-productratio of U1 and U2 increases under the assumption that X ispositively associated with U1 and U2. This study reports the ability of Fusarium to induce orchidseed colouration and germination. The in vitro bioassay germinationtest, using a Fusarium isolate from the protocorm of Cypripediumreginae, was compared with standard chemical procedures of triphenyltetrazolium chloride (TTC) and acid fuchsin (AC) for testingseed viability. With Cypripedium reginae, Cypripedium parviflorumand Platanthera grandiflora, the efficiency of the bioassaywas similar to that of the TTC and AC procedures. However, thebioassay was more appropriate for estimating embryo viabilityafter a prolonged seed pretreatment (more than 2 h) in 10% sodiumhypochlorite, a surface sterilant often used to enhance germinationof terrestrial species. We also obtained in vitro Cypripediumreginae seed germination induction and protocorm formation bythe same Fusarium isolate. This is the first confirmation ofBernard's early reports that orchid fusaria could stimulateseed germination (Bernard N. 1990.Révue Généralede Botanique12 : 108–120). However, the importance ofthe non-mycorrhizal Fusarium fungus in promoting germinationseems to be relatively minor compared to that of specificRhizoctoniaorchid mycorrhizas. Since amplified fragment length polymorphism (AFLP) analysishas proved useful in distinguishing cultivars of Caladium, itwas used to assess the status of species of Caladium vs. Xanthosoma,both in tribe the Caladieae, and to reassess the position ofHapaline in the same tribe. AFLP analysis using three primercombinations was carried out on four species of Caladium(C.bicolor, C. humboldtii, C. lindenii and C. schomburgkii). Resultsshowed that AFLP can distinguish between the different speciesby their unique and different banding patterns. AFLP analysisconfirmed that C. humboldtii is a species distinct from C. bicolorand that C. lindenii is a true Caladium species and does notbelong to Xanthosoma. UPGMA cluster analysis showed that C.bicolor and C. schomburgkii are most similar and that C. humboldtiiis closer to the C. bicolor / C. schomburgkii cluster comparedwith C. lindenii. Genetic relationships between Caladium, Xanthosoma,Hapaline, Alocasia and Protarum were also examined by AFLP analysisusing eight primer combinations. Several useful molecular markerswere specific either to Caladium orXanthosoma , so that AFLPcan be used to distinguish species of these two genera. Geneticanalysis of the genera examined confirms that the Caladieaeand Colocasieae tribes are distinct and that Hapaline fallswithin the tribe Caladieae and that Protarum is most distantfrom all the genera examined. Daphnia can suppress ciliates and rotifers through predationand interference competition, but it is not known whether thisproduces any direct benefit to Daphnia. We conducted survivorshipand cohort lifetable experiments to determine whether Daphniacan utilize ciliates and rotifers as food. Three species ofoligotrich ciliates (Halteria grandinella, Strobilidium gyransand Strobilidiumvelox) and one rotifer (Keratella cochlearis)were used. Lifetable experiments were conducted with a basallevel of algae (Cryptomonas sp.), plus either ciliates or rotifers,while survivorship experiments had only the rotifers or ciliates.Densities of 30 H.grandinella ml–1, 50 S.gyrans ml–1and 15 S.velox ml–1 enhanced Daphnia pulex's populationgrowth rate 35–50% over controls with only algae. TenS.gyrans ml–1 did not produce a significant change inDaphnia's growth rate. Densities of 100 and 300 K.cochlearis–1 increased Daphnia population growth rates by II and10%, respectively. Both 10 and 50 S.gyrans ml–1 enhancedDaphnia's survivorship compared to starved controls, but neither100 nor 300 K.cochlearis l–1 enhanced its survivorship.The amount of enhancement of Daphnia growth rates by rotifersand ciliates is roughly proportional to the death rates imposedby Daphnia. The death rate imposed by Daphnia on rotifers isa function of both algal density and Daphnia size. The relative abilities of freshwater calanoid copepods to grazealgae and filamentous cyanobacteria were examined in three speciesof Boeckella (B.triarticulata, B.hamata, B.dilatata) fed Cydotella,Cryptomonas and two species of Nostoc and Anabaena. Althoughrates of ingestion were highest on Cryptomonas, all the Boeckellaspecies consumed cyanobacteria, some species at similar ratesto Cyclolella. Faecal production on the foods was highest onCydotella and, in B.triarticulata, higher on A.flos-aquae andNostoc sp. 2 than on Cryptomonas. At 1 mg l–1 of thesecyanobacteria, female B.triarticulata voided 70% by volume ofthe material they ingested as compact faecal pellets. By theirgrazing they also decreased dramatically the trichome lengthsof A.flos-aquae, A.oscillarioides and Nostoc sp. 2, therebypossibly altering the potential susceptibility of these filamentouscyanobacteria to grazing by other zooplankton.  相似文献

Ethylene biosynthesis in higher plants is regulated developmentallyand environmentally. To investigate the regulation of ACC synthasegene expression, the promoters of Arabidopsis ACS genes, AtACS4,AtACS5, and AtACS7, were fused to a GUS reporter gene, and therecombinant transgenes were introduced into Arabidopsis to producethree groups of AtACS::GUS transgenic plants. Histochemic andfluorometric study of these transgenic plants revealed thatpromoters of AtACS4, AtACS, and AtACS7 are all active in dark-germinatedseedlings. AtACS5 has the highest promoter activity in leavesof 2-week-old light-grown seedlings among the three AtACS genesstudied. In the mature leaves, AtACS4 and AtACS7 genes are expressedin both veins and areoles, whereas AtACS5 is expressed at ahigher level in the areoles and epidermal cells surroundingtrichomes. The promoter activities of all these AtACS genesare found in the reproductive organs. AtACS5 and AtACS7 arehighly expressed in petals, sepals, carpels, stamens, caulineleaves, inflorescence stems, and siliques, while AtACS4 expressionis undetectable in the petals of open flowers. All three AtACSgenes are expressed in root tissue. In the 2-week-old light-grownArabidopsis, the AtACS4 promoter is responsive to the planthormones IAA, ethylene, and ABA, and to darkness and wounding;the AtACS5 promoter to IAA, ABA, salt, high temperature, andwounding; and the AtACS7 promoter to GA3, ethylene, and ABA,and to darkness and salt. Low-temperature treatment abolishesthe darkness-induced AtACS7 gene expression, but not that ofAtACS4. Each AtACS gene has a unique expression profile duringgrowth and development. It appears that at any developmentalstage or any growth period of Arabidopsis, there is always amember of AtACS multigene family that is actively expressed. Stomatal size and frequency as well as structure and distributionof leaf waxes were compared in seedlings of two varieties ofTheobroma cacao and three families of Hevea brasiliensis. Stomataof both species were located on the abaxial leaf surface only.The stomata of Theobroma averaged 45% shorter and 29% narrowerthan those of Hevea, but stomatal frequency was 44% higher inTheobroma. Stomatal size and frequency differed more among Heveafamilies than between Theobroma varieties. The structure ofthe leaf waxes of Theobroma and Hevea differed appreciably.It also varied greatly between the abaxial and adaxial leafsurfaces of Hevea, but only slightly in Theobroma. The structureof leaf waxes varied little between Theobroma varieties or amongHevea families. Leaf conductances and transpiration rates weremuch higher in Hevea than in Theobroma seedlings. The frxC gene, found in liverwort chloroplast DNA, encodes aprotein of unknown function. The deduced amino acid sequenceof the protein shows significant homology to that of ni-trogenaseFe-protein encoded by the nifH gene. We have cloned the frxCand nifH genes from the nitrogen-fixing cyanobacterium Plectonemaboryanum, using frxC- and nifH-specific probes, and have determinedtheir nucleotide sequences. The amino acid sequence deducedfrom the frxC gene of P. boryanum exhibits 83% homology to thatof the protein encoded by the/rxCgene from liverwort, whereasit exhibits only 34% homology to that encoded by the nifH genefrom the same organism, namely, P. boryanum. Northern blot analysisshowed that the frxC gene was transcribed more actively undernitrogenase-repressed conditions than under nitrogenase-inducedconditions, suggesting that the FrxC protein has a functiondistinct from nitrogen fixation. These results, together withthe phylogenetic relationship between the nifH and frxC genes,indicate that the frxC and nifH genes are derived from a commonancestral gene but have evolved independently to encode proteinswith different functions. Effects of annual variation in rainfall, temperature and humidityon flowering abundance of eight temperate woodland plants (Anemonenemorosa, Cardamine bulbifera, Lamiastrum galeobdolon,Oxalisacetosella , Ranunculus ficaria, Stellaria holostea, Viola reichenbachianaand Viola riviniana) were studied during 12 consecutive years(1989–2000) in a hornbeam (Carpinus betulus) forest insoutheast Sweden. Above-average rainfall/humidity in late summerto early autumn of the preceding year increased flowering abundancein L. galeobdolon, O. acetosella, V. reichenbachiana, V. rivinianaand, especially, in R. ficaria, but not in S. holostea and A.nemorosa. Moreover, flowering of R. ficaria and O. acetosellawas positively related to rainfall/humidity during several partsof, or the entire, preceding year. On the contrary, floweringof S. holostea and A. nemorosa was closely related to low valuesof rainfall/humidity in autumn and/or winter of the precedingyear and also to low humidity in the current year in A. nemorosa.Two long periods (3–4 years) of increasing rainfall deficitcoincided with decreasing flowering abundance in most of thespecies, but not with decreasing vegetative development. Temperaturevariability was less consistently related to flowering. A coolperiod during the preceding summer or autumn seemed importantfor flowering in L. galeobdolon, O. acetosella and the Violaspecies, although these relations were, at least partly, causedby interactions with rainfall/humidity. No significant (P <0.05) correlations were found between flowering and the conditionsprevailing in April to May—the main flowering season—ofthe current year. Pectin degrading enzymes, hemicellulose degrading enzyme andcellulose degrading enzymes were studied in Cuscuta reflexaRoxb., its susceptible hosts, Brassica campestris L., Cocciniaindica W. & A. Datura innoxia Mill, Helianthus annuus L.,Holoptelea indica Planch, Lantana camara L., Medicago sativaL., Manihot utilissima Pohl, Petunia hybrida X Hort exvilm,Pisum sativum L., Phaseolus vulgaris L. and Solanum nigrum L.and non-susceptible plants Ipomoea batata Lam. and Solanum tuberosumL. Pectin esterase and polygalacturonase were present in higheramounts in Cuscuta parasitic on P. vulgaris and S. nigrum, whichneeded more time for haustorial establishment. Exo-l, 4-ß-D-glucosidaseactivity was found in Cuscuta but could not be detected in itshosts. Xylanase and cellulase activity of host plants increasedwhile cellobiase activity decreased as a result of infectionby the parasite. Higher pectin esterase, polygalacturonase,xylanase and exo-l, 4-ß-D-glucosidase activities inthe haustorial region of the parasite is likely to bring aboutthe lysis of the cell wall of the host plant and thus facilitatethe penetration of the parasite haustoria into the host sieveelement, which is necessary for the transport of nutrients betweenthe host and the parasite. We investigated the factors controlling the relative abundancesof two Daphnia species, D.pulex and D.laevis, in a small Wisconsinpond. D.pulex was the dominant Daphnia species in fall 1977and summer-fall 1978; D.laevis was the only Daphnia speciespresent in summer 1979. The abundance of D.laevis was positivelycorrelated with the abundance of the notonectid, Buenoa confusa.In predation trials, notonectides exhibited a distinct preferencefor D.pulex over similarly-sized D.laevis, but Chaoborus larvaefed at similar rates on both Daphnia species. Behavioral observationsrevealed that Buenoa adults were much less efficient at capturingD.laevis than D.pulex. Quantitative results of these predationtrials were combined with estimates of predator and prey densityand distribution to evaluate the effect of predation on thedaphnid populations. The effect of predation varied throughtime and microhabitat, and only infrequently could predationaccount for total prey mortality. D.laevis was most abundantat times and in places where Buenoa predation was most intense.Competition experiments illustrated the competitive superiorityof D.pulex over D.laevis. D.pulex was able to competitivelyexclude D.laevis in long term experiments, and D.pulex's fecunditywas higher than that of D.laevis in shorter experiments. Inlong-term experiments, Chaoborus larvae at natural densitieswere able to keep both Daphnia species at low, constant levelsand neither species clearly dominated when Chaoborus was present.The relative abundances of D.pulex and D.laevis were controlledby a complex of biotic and abiotic factors. Pond depth and predatordensity determined the intensity of predation on daphnid populations.When notonectid predation was intense, D.laevis dominated; whenthe intensity of predation by notonectids was low, D.pulex dominateddue to its superior competitive abilities. Plants of four isolines of soyabean [Glycine max(L.) Merrill]‘Clark’, viz‘L71-920’ (maturity genecomplemente1e2e3 ), ‘L80-5914’ (E1e2e3), ‘Clark’(e1E2E3), and ‘L65-3366’ (E1E2E3), were grown inshort (12.25 h d - 1natural light) and long days (12.25 h d- 1natural light supplemented with 2.75 h d - 1low-irradianceartificial light) from first flowering to maturity in a polythenetunnel maintained at 30/24°C (day/night). Whereas therewere few differences among the isolines grown in short days,in long days the dominant alleles increased crop duration, biomassand seed yield substantially. Increases in biological and economicyield were not solely a consequence of longer crop duration:the dominant alleles also increased crop growth rate and radiationuse efficiency in long days (from 1.3 g MJ - 1total radiationine1e2e3 to 2.8 g MJ - 1inE1E2E3 ). Greater radiation use efficiencyresulted from a relatively longer leaf area duration, betterdistribution and orientation of a larger mass of leaves withinthe canopy, and smaller partitioning of assimilates to reproductivestructures. The work reveals the substantial effects of thethree lociE1 / e1, E2/ e2and E3/e3 on the response of plantgrowth, as well as development, to environment. The effect of nutrient amendment on growth of nine indigenoustree species planted on coal mine spoil was studied. Greatergrowth in fertilized plots was accompanied by greater foliarN and P concentrations in all species. The response to fertilizationvaried among species and was greater in non-leguminous thanin leguminous species. Furthermore, leguminous species exhibitedhigher growth rates compared to non-leguminous species. Thelog-transformed height-diameter relationships were significantfor all tree species and treatments. Acacia catechu, Dalbergiasissoo, Gmelina arborea and Azadirachta indica fitted the elasticsimilarity model of tree growth; whereas Pongamia pinnata andPhyllanthus emblica followed the constant stress model. Tectonagrandis was the only species which fitted the geometric similaritymodel. In Albizia lebbeck and Terminalia bellirica, the b -values(the gradients of the log-transformed height to diameter relationships)were considerably lower (<0.5), and these two species didnot fit any model of tree growth. In several cases, the b -valueswere considerably influenced by nutrient amendment. The log-transformedcrown mass and trunk mass relationships were significant forall treatments and species. The slope of the crown mass:trunkmass relationship was near unity in A. indica, D. sissoo, G.arborea, P. emblica, P. pinnata, T. grandis, and T. bellirica.However, in A. catechu and A. lebbeck, this slope was well belowunity suggesting a greater allocation to non-photosynthetictissue. Fertilizer amendment resulted in a heavier crown relativeto trunk in A. indica, T. grandis and T. bellirica. Four upland and two lowland varieties were grown on floodedand dry soil in pots in a glasshouse. Photosynthetic rate (P),transpiration rate (T), and water content (W) of the secondexpanded leaf from the top of the main stem were measured undercontrolled aeration and illumination in a leaf chamber in thelaboratory, together with leaf area (La), dry matter content(DM), nitrogen content (N), stomatal frequency (Sf), and totalvessel cross-sectional area at the base of the leaf blade (Va).P/La was positively related to T/La and Sf/La among six varietieswhen they were grown on flooded soil. IR 8, a semidwarf indicalowland variety, showed the highest P/La with the highest Sf/Laand T/La. When grown on dry soil P/La was positively correlatedwith W/DM, the latter being negatively related to T/Va. Twoupland varieties, African Moroberekan and Brazilian IAC 1246,showed the highest P/La on dry soil, keeping a higher W/DM witha lower T/La and a lower T/Va. Daytime leaf diffusive conductance(l/rL) and leaf water potential (L) measured on the same orthe same stage leaf in the glasshouse were positively correlatedwith the W/DM measured in the laboratory among varieties grownon dry soil. Simultaneous observation of P, T and W in the laboratoryindicated nonstomatal reduction in P/La due to leaf water deficitin sensitive varieties, although these varieties also showeda markedly lower daytime l/rL in the glasshouse as comparedwith resistant varieties.
