水培大豆和田间生长的大豆,接种根瘤菌 Rhizobium B16-11C 后植株全氮含量、叶片叶绿素含量和净光合速率及种子产量都明显增加。比较 Clark 大豆的结瘤品系和不结瘤品系获类似结果。摘除根瘤后3天内叶片净光合速率无明显变化。大豆植株遮阴、去叶或切掉地上部导致根瘤活性明显下降。但去豆荚不能提高根瘤固氮的比活性。根瘤活性的日变化不能用根瘤蔗糖、淀粉含量或周围温度的变化来解释,其控制因子尚待深入研究。  相似文献

Nodule nitrogen fixation rates are regulated by a mechanism which is responsive to the rhizosphere oxygen concentration. In some legumes, this oxygen-sensitive mechanism appears to involve changes in the gas permeability of a diffusion barrier in the nodule cortex. In soybean evidence for such a mechanism has not been found. The purpose of this research was to make quantitative measurements of soybean nodule gas permeability to test the hypothesis that soybean nodule gas permeability is under physiological control and responsive to the rhizosphere oxygen concentration. Intact hydroponically grown soybean plants were exposed to altered rhizosphere oxygen concentrations, and the nodule gas permeability, acetylene reduction and nodule respiration rates were repeatedly assayed. After a change in the external oxygen concentration, nitrogenase activity and nodule respiration rates displayed a short-term transient response after which the values returned to rates similar to those observed under ambient oxygen conditions. In contrast to steady-state nitrogenase activity and nodule respiration, nodule gas permeability was dramatically affected by the change in oxygen concentration. Decreasing the external oxygen concentration to 0.1 cubic millimeter per cubic millimeter resulted in a mean increase in nodule gas permeability of 63%. Increasing the rhizosphere oxygen concentration resulted in decreased nodule gas permeability. These data are consistent with the hypothesis that soybean nodules are capable of regulating nitrogen fixation and nodule respiration rates in response to changes in the rhizosphere oxygen concentration and indicate that the regulatory mechanism involves physiological control of the nodule gas permeability.  相似文献

The relationship between photosynthesis of soybean and nitrogen fixation of the nodules by symbiotic Rhizobium was studied. The contents of total nitrogen and chlorophyll, the net photosynthetic rate and seed yield of soybean were much higher in either hydroponically cultivated or field-grown plants inoculated with Rhizobium B16–11C (or Clark nodulating strain) than in control without inoculation (or Clark non-nodulating strain). These results show that the symbiotic nitrogen fixation has a beneficial effect on photosynthesis. However, the effect was indirect and slow so that there was no change in the net photosynthetic rate of the soybean leaves until three clays after removing nodules from the soybean roots. On the other hand, decreasing the photosynthate supply to nodule by shade, defoliation or shoot removal of the soybean, the nodule activity declined significantly. It seems that the supply of photosynthate to root nodule is a limiting factor for symbiotic nitrogen fixation. However, the diurnal variation of the nodule activity could not be explained by change neither in the contents of sucrose and starch of the root nodules nor in the ambient temperature. The factor controlling the diurnal variation deserves further study.  相似文献

Nitrogen fixation (acetylene reduction) rates of nodules on intact field-grown soybean (Glycine max) subjected to altered oxygen concentration (0.06-0.4 cubic millimeter per cubic millimeter) returned to initial rates during an 8-hour transitory period. Hydroponically grown soybean plants also displayed a transitory (1-4 hours) response to changes in the rhizosphere oxygen concentration after which the fixation rates returned to those observed under ambient oxygen concentrations. It was hypothesized that soybean nodules contain a regulatory mechanism which maintains a stable oxygen concentration inside nodules at a sufficiently low concentration to allow nitrogenase to function. A possible physiological mechanism which could account for this regulation is adjustment in nodule respiration activity such that nodule oxygen concentration and nitrogen fixation are maintained at stable levels. Experiments designed to characterize the non-steady-state oxygen response and to test for the presence of nodule respiratory control are presented. Non-steady-state acetylene reduction and nodule respiration (oxygen uptake) rates measured after alterations in the external oxygen concentration indicated that the regulatory mechanism required 1 to 4 hours to completely adjust to changes in the external oxygen concentration. Steady-state nodule respiration, however, did not respond to alterations in the rhizosphere oxygen concentration. It was concluded that soybean nodules can adjust to a wide range of rhizosphere oxygen concentrations, but the mechanism which controls nitrogen fixation rates does not involve changes in the nodule respiration rate.  相似文献

Nitrogen fixation activity by soybean (Glycine max (L.) Merr.) nodules has been shown to be especially sensitive to soil dehydration. Specifically, nitrogen fixation rates have been found to decrease in response to soil dehydration preceding alterations in plant gas exchange rates. The objective of this research was to investigate possible genetic variation in the sensitivity of soybean cultivars for nitrogen fixation rates in response to soil drying. Field tests showed substantial variation among cultivars with Jackson and CNS showing the least sensitivity in nitrogen accumulation to soil drying. Glasshouse experiments confirmed a large divergence among cultivars in the nitrogen fixation response to drought. Nitrogen fixation in Jackson was again found to be tolerant of soil drying, but the other five cultivars tested, including CNS, were found to be intolerant. Experiments with CNS which induced localized soil drying around the nodules did not result in decreases in nitrogen fixation rates, but rather nitrogen fixation responded to drying of the entire rooting volume. The osmotic potential of nodules was found to decrease markedly upon soil drying. However, the decrease in nodule osmotic potential occurred after significant decreases in nitrogen fixation rates had already been observed. Overall, the results of this study indicate that important genetic variations for sensitivity of nitrogen fixation to soil drying exist in soybean, and that the variation may be useful in physiology and breeding studies.  相似文献

The low gas permeability of a diffusion barrier in the cortex of soybean nodules plays a significant role in the protection of nitrogenase from oxygen inactivation. It may also set an upper limit on nodule respiration and nitrogen fixation rates. Two methods which have been used to quantify the gas permeability of leguminous nodules are reviewed and found to be unreliable. A new assay technique for determining both the nodule activity and gas permeability is developed and tested. This ‘lag-phase’ assay is based on the time nodules require to reach steady-state ethylene production after being exposed to acetylene. The technique is rapid, insensitive to errors in biochemical parameters associated with nitrogenase, and is non-destructive. The method was tested with intact aeroponically grown soybean plants for which the mean nodule gas permeability was found to be 13.3×10−3 mms−1. This corresponds to a layer of cells approximately 35 um thick and is consistent with previously reported values.  相似文献

Aerial photographs, maps and optically stimulated luminescence dates were combined with existing soil data to construct high resolution chronosequences of soil development over 140 years at a temperate Atlantic UK dune system. Since soil formation had progressed for varying duration under different climate and nitrogen deposition regimes, it was possible to infer their relative influence on soil development compared with location-specific variables such as soil pH, slope and distance to the sea. Results suggest that soil development followed a sigmoid curve. Soil development was faster in wet than in dry dune habitats. In dry dunes, rates were greater than in the literature: they increased with increasing temperature and nitrogen deposition and decreased with increasing summer gales. The combination explained 62% of the variation. Co-correlation meant that effects of nitrogen deposition could not be differentiated from temperature. In wet dune habitats rates increased with temperature and decreased with gales. The combination explained only 23% of the variation; surprisingly, rainfall was not significant. Effects of location-specific variables were not significant in either habitat type. Nitrogen accumulation was faster in wet than dry dune habitats, averaging 43 kg N ha?1 per year overall. Nitrogen accumulation greatly exceeded inputs from atmospheric deposition, suggesting rates of input for biological N fixation are 10–60 kg N ha?1 per year. Recent climate and/or nitrogen deposition regimes may have accelerated soil development compared with past rates. These data suggest the importance of changing climate on soil development rates and highlight the contribution of biological N fixation in early successional systems.  相似文献

We used an acetylene reduction assay to measure rates of nitrogen fixation on a 38-year-oldAlnus hirsuta plantation in central Korea. The diurnal pattern of acetylene reduction changed significantly during May, August, and October, typically varying by 3-fold throughout the course of the day. Maximum rates occurred at 3 p.m. in May and October, but at 6 p.m. in August. Increasing trends were evident during the early growing season, with sustained high rates from mid-May through late September; July had the highest rates, averaging 7.2 μmole g-1 dry nodule h-1. The average nodule biomass for this plantation was 220 kg ha ’. Rates of acetylene reduction were related to soil temperature, but not to soil moisture content. Combining these nodule biomass calculations with seasonal average acetylene reduction rates yielded an estimate of current annual nitrogen fixation of 60 kg N ha-1 for the plantation. This rate of annual nitrogen addition was very large in relation to the yearly nitrogen requirements of coniferous and deciduous forests in central Korea.  相似文献

Both nodulation and nitrogen fixation in soybean [Glycine max (L.) Merr.] are sensitive to soil drying, which can have important negative effects on yield. An exception to this general response has been the identification of the cultivar Jackson as being drought tolerant for N2 fixation. The objectives of this research were to examine nodule formation and growth in Jackson among other soybean cultivars in response to soil drying under field conditions. Two field experiments were conducted to examine the genetic variation in the sensitivity of nodule numbers and dry weights to soil drying. Substantial variation among soybean lines was found, and the drought-tolerance trait was demonstrated again in Jackson. Greenhouse experiments were conducted to further analyze the variation of nodulation response to soil water content. The differences among cultivars observed in the field were confirmed in the greenhouse. Importantly, the relative drought insensitivity of N2 fixation in cultivar Jackson was associated with high individual nodule dry weight under drought conditions, relative to well-watered plants. It was concluded that large variation in nodulation sensitivity to water deficit exists among soybean cultivars and that the response of N2 fixation rates to drought is related in part to nodule formation and growth.  相似文献

In an investigation of the ability of alfalfa to fix nitrogen under field conditions in Scandinavia, N2 fixation during the establishment year ranged between 7.85 and 10.37 g of N m−2, depending on the method used. The methods used were an in situ acetylene reduction method, a 15N isotope dilution method using two reference crops, and a total-N difference method. The dynamics of nitrogenase activity in relation to plant development was studied by using the acetylene reduction method. Also, the diurnal variation in N2 fixation at the field site was studied with the acetylene reduction method; no diurnal change was detected, which is explained by the fact that the nodules within the soil were protected against short-term fluctuations in temperature. The significant amount of nitrogen fixed by alfalfa during its first year even at northern latitudes suggests that this crop offers an alternative to conventional field management of heavily fertilized nonlegume leys.  相似文献

Drought stress is one of the major factors affecting nitrogen fixation by legume-rhizobium symbiosis. Several mechanisms have been previously reported to be involved in the physiological response of symbiotic nitrogen fixation to drought stress, i.e. carbon shortage and nodule carbon metabolism, oxygen limitation, and feedback regulation by the accumulation of N fixation products. The carbon shortage hypothesis was previously investigated by studying the combined effects of CO2 enrichment and water deficits on nodulation and N2 fixation in soybean. Under drought, in a genotype with drought tolerant N2 fixation, approximately four times the amount of 14C was allocated to nodules compared to a drought sensitive genotype. It was found that an important effect of CO2 enrichment of soybean under drought was an enhancement of photo assimilation, an increased partitioning of carbon to nodules, whose main effect was to sustain nodule growth, which helped sustain N2 rates under soil water deficits. The interaction of nodule permeability to O2 and drought stress with N2 fixation was examined in soybean nodules and led to the overall conclusion that O2 limitation seems to be involved only in the initial stages of water deficit stresses in decreasing nodule activity. The involvement of ureides in the drought response of N2 fixation was initially suspected by an increased ureide concentration in shoots and nodules under drought leading to a negative feedback response between ureides and nodule activity. Direct evidence for inhibition of nitrogenase activity by its products, ureides and amides, supported this hypothesis. The overall conclusion was that all three physiological mechanisms are important in understanding the regulation of N2 fixation and its response of to soil drying.  相似文献

2012年6月至2012年10月, 对黄河三角洲芦苇(Phragmites australis)湿地进行了模拟氮沉降试验, 氮沉降水平分别为对照(CK, 0 kg N·hm-2·a-1)、低氮(LN, 50 kg N·hm-2·a-1)和高氮(HN, 100 kg N·hm-2·a-1)。利用LI-8100土壤碳通量测量系统测定土壤呼吸速率。结果表明, 氮沉降促进了芦苇湿地土壤呼吸作用, LN和HN处理使芦苇生长季(6-10月)平均土壤呼吸速率比CK分别提高19%和58%。积水改变了芦苇湿地土壤呼吸日动态。地面无积水时, 各处理土壤呼吸日动态均呈单峰型曲线; 地面有积水时, 土壤呼吸日动态峰值推后或无单峰型波动规律。积水影响土壤呼吸作用对温度的响应。地面无积水时, 各处理土壤呼吸速率均与气温呈极显著的正指数相关关系, 气温分别解释了CK、LN和HN处理下土壤呼吸季节变化的69.9%、64.5%和59.9%; 地面有积水时, 各处理土壤呼吸与气温相关性不显著。CK、LN和HN处理下土壤呼吸温度敏感性系数Q10值分别为1.68、1.75和1.68, 表明LN处理增强了土壤呼吸温度敏感性, HN处理对其影响不显著。  相似文献

通过田间试验采用基于热平衡法的茎流计测定玉米/大豆条带间作群体内作物的蒸腾规律.结果表明:间作群体内,玉米和大豆植株的茎流速率在晴天呈单峰曲线,在阴天则呈多峰曲线.植株的茎流受多个环境因子的影响,其中太阳辐射是影响植株茎流最主要的气象因子.玉米和大豆的单株日茎流量与多个气象因子间存在较好的相关关系,达到极显著水平.茎流观测期内(2008年6月1-30日),间作群体内玉米植株的日均蒸腾量(1.44 mm·d-1)为大豆(0.79 mm·d-1)的1.8倍,玉米和大豆植株的蒸腾量分别占间作群体总蒸腾量的64%和36%.考虑到作物的茎直径和叶面积的空间变异,安装一定数量的茎流探头对于准确测定植株茎流是十分必要的.  相似文献

树干呼吸(E_s)是森林生态系统碳循环过程的重要组成部分,深入理解树干呼吸过程对未来气候变暖的响应及反馈机制有助于更加精确地估算森林生态系统碳储量。为揭示毛白杨树干呼吸及其温度敏感性的昼夜变化和季节动态规律,利用Li-Cor6400便携式光合作用测定系统及其配套使用的土壤呼吸测量气室(LI-6400-09)对冀南平原区毛白杨的树干呼吸和树干温度实施为期1年的连续监测。结果表明:(1)在生长季,毛白杨树干呼吸与树干温度之间在晚上呈现正相关的关系(R~2=0.88);相反,两者在白天为负相关的关系(R~2=0.96)。(2)整个观测期内,毛白杨树干呼吸和树干温度均呈现"钟形"的变化曲线,树干呼吸与树干温度之间存在着较好的指数函数关系(R~2=0.93),且树干呼吸的温度敏感性系数(Q_(10))为2.62;不同季节毛白杨树干呼吸的Q_(10)存在差异,生长季的Q_(10)(1.95)明显低于非生长季(3.00),表明生长呼吸和维持呼吸对温度的响应也并不相同。(3)温度矫正后的毛白杨树干呼吸(R_(15))在昼夜和季节尺度上均存在明显的变异,即夜晚的R_(15)显著高于白天(P0.01),生长季的R_(15)明显高于非生长季(P0.05);树干可溶性糖含量与生长季的R_(15)存在较好的相关性(R~2=0.52),而非生长季的R_(15)却主要受到树干淀粉含量的影响。研究结果表明,在生长季,毛白杨树干呼吸的在日变化主要受到温度的影响,而在季节尺度上Q_(10)的变异则与树干呼吸中维持呼吸所占比例及树干中非结构性碳水化合物(可溶性糖和淀粉)的含量及类型紧密相关。  相似文献

Nitrogenase (EC 1.7.99.2) activity of nodules of cowpea (Vigna unguiculata [L.] Walp), maintained under conditions of a 12-hour day at 30°C and 800 to 1,000 microeinsteins per square meter per second (photosynthetically active radiation) and a 12-hour night at 20°C, showed a marked diurnal variation with the total electron flux through the enzyme at night being 60% of that in the photoperiod. This diurnal pattern was, however, due to changes in hydrogen evolution. The rate of nitrogen fixation, measured by short-term 15N2 assimilation or estimated from the difference in hydrogen evolution in air or Ar:O2 (80:20; v/v), showed no diurnal variation. Carbon dioxide released from nodules showed a diurnal variation synchronized with that of nitrogenase functioning and, as a consequence, the apparent `respiratory cost' of nitrogen fixation in the photoperiod was almost double that at night (9.74 ± 0.38 versus 5.70 ± 0.90 moles CO2 evolved per mole N2 fixed). Separate carbon and nitrogen balances constructed for nodules during the photoperiod and dark period showed that, at night, nodule functioning required up to 40% less carbohydrate to achieve the same level of nitrogen fixation as during the photoperiod (2.4 versus 1.4 moles hexose per mole N2 fixed).

Stored reserves of nonstructural carbohydrate of the nodule only partly satisfied the requirement for carbon at night, and fixation was dependent on continued import of translocated assimilates at all times. Measurements of the soluble nitrogen pools of the nodule together with 15N studies indicated that, both during the day and night, nitrogenous products of fixation were effectively translocated to all organs of the host plant despite low rates of transpiration at night. Reduced fluxes of water through the plant at night were apparently counteracted by increased concentration of nitrogen, especially as ureides, in the xylem stream.

Nutrient biogeochemistry associated with the early stages of soil development in deltaic floodplains has not been well defined. Such a model should follow classic patterns of soil nutrient pools described for alluvial ecosystems that are dominated by mineral matter high in phosphorus and low in carbon and nitrogen. A contrast with classic models of soil development is the anthropogenically enriched high nitrate conditions due to agricultural fertilization in upstream watersheds. Here we determine if short-term patterns of soil chemistry and dissolved inorganic nutrient fluxes along the emerging Wax Lake delta (WLD) chronosequence are consistent with conceptual models of long-term nutrient availability described for other ecosystems. We add a low nitrate treatment more typical of historic delta development to evaluate the role of nitrate enrichment in determining the net dinitrogen (N2) flux. Throughout the 35-year chronosequence, soil nitrogen and organic matter content significantly increased by an order of magnitude, whereas phosphorus exhibited a less pronounced increase. Under ambient nitrate concentrations (>60 μM), mean net N2 fluxes (157.5 μmol N m?2 h?1) indicated greater rates of gross denitrification than gross nitrogen fixation; however, under low nitrate concentrations (<2 μM), soils switched from net denitrification to net nitrogen fixation (?74.5 μmol N m?2 h?1). As soils in the WLD aged, the subsequent increase in organic matter stimulated net N2, oxygen, nitrate, and nitrite fluxes producing greater fluxes in more mature soils. In conclusion, soil nitrogen and carbon accumulation along an emerging delta chronosequence largely coincide with classic patterns of soil development described for alluvial floodplains, and substrate age together with ambient nitrogen availability can be used to predict net N2 fluxes during early delta evolution.  相似文献

We quantified rates of soil respiration among sites within an agricultural landscape in central Iowa, USA. The study was conducted in riparian cool-season grass buffers, in re-established multispecies (switchgrass + poplar) riparian buffers and in adjacent crop (maize and soybean) fields. The objectives were to determine the variability in soil respiration among buffer types and crop fields within a riparian landscape, and to identify those factors correlating with the observed differences. Soil respiration was measured approximately monthly over a two-year period using the soda-lime technique. Mean daily soil respiration across all treatments ranged from 0.14 to 8.3 g C m–2 d–1. There were no significant differences between cool-season grass buffers and re-established forest buffers, but respiration rates beneath switchgrass were significantly lower than those beneath cool-season grass. Soil respiration was significantly greater in both buffer systems than in the cropped fields. Seasonal changes in soil respiration were strongly related to temperature changes. Over all sites, soil temperature and soil moisture together accounted for 69% of the seasonal variability in soil respiration. Annual soil respiration rates correlated strongly with soil organic carbon (R = 0.75, P < 0.001) and fine root (<2 mm) biomass (R = 0.85, P < 0.001). Annual soil respiration rates averaged 1140 g C m–2 for poplar, 1185 g C m–2 for cool-season grass, 1020 g C m–2 for switchgrass, 750 g C m–2 for soybean and 740 g C m–2 for corn. Overall, vegetated buffers had significantly higher soil respiration rates than did adjacent crop fields, indicating greater soil biological activity within the buffers.  相似文献

In short-season soybean production areas, low soil temperature is the major factor limiting soybean establishment, nodulation and nitrogen fixation. Gibberellic acid (GA) pretreatment of crop seeds can overcome low soil temperature inhibition of seed germination and seedling development. However, previous studies have found that the application of GAs decreased legume nodulation and nitrogen fixation under optimal growth conditions. A field experiment was conducted under short season conditions in eastern Canada to determine whether the application of GA3 to soybean seed could accelerate germination, and increase plant nodulation and nitrogen fixation. The results indicated that GA3 application accelerated seedling emergence but decreased plant nodulation and nitrogen accumulation at early plant growth stages. However, these initial negative effects were overcome as the plants developed. Gibberellic acid applied to soybean seed at the time of planting did not influence final grain and protein yield.  相似文献

The effect of the Bradyrhizobium japonicum hydrogenase on nitrogen fixation was evaluated by comparing the growth of Vigna and Glycine species inoculated with a Hup mutant and its Hup+ revertant. In all experiments, the growth of plants inoculated with the strain without hydrogenase was at least equal to the growth of the strain with hydrogenase. For Glycine usuriensis and Glycine max cv. Hodgson in liquid culture, the growth was higher with the Hup strain. It is possible that reduced rates of nitrogen fixation in the presence of hydrogenase are due to O2 depletion caused by the hydrogen oxidizing, since the oxygen pressure in the air appears to be a limiting factor of symbiotic nitrogen fixation in the soybean.  相似文献

This study tested the hypothesis that inoculation of soybean (Glycine max Merr.) with a Bradyrhizobium japonicum strain (USDA110) with greater N2 fixation rates would enhance soybean response to elevated [CO2]. In field experiments at the Soybean Free Air CO2 Enrichment facility, inoculation of soybean with USDA110 increased nodule occupancy from 5% in native soil to 54% in elevated [CO2] and 34% at ambient [CO2]. Despite this success, inoculation with USDA110 did not result in greater photosynthesis, growth or seed yield at ambient or elevated [CO2] in the field, presumably due to competition from native rhizobia. In a growth chamber experiment designed to study the effects of inoculation in the absence of competition, inoculation with USDA110 in sterilized soil resulted in nodule occupation of >90%, significantly greater 15N2 fixation, photosynthetic capacity, leaf N and total plant biomass compared with plants grown with native soil bacteria. However, there was no interaction of rhizobium fertilization with elevated [CO2]; inoculation with USDA110 was equally beneficial at ambient and elevated [CO2]. These results suggest that selected rhizobia could potentially stimulate soybean yield in soils with little or no history of prior soybean production, but that better quality rhizobia do not enhance soybean responses to elevated [CO2].  相似文献
