Three species of the reef coral genus Madracis display skeletal isotopic characteristics that relate to depth, colony topography, and consequently to coral physiology. The joint interpretation of skeletal 13C and 18O provides information on the ecological plasticity and adaptation to depth of a coral species. Isotopic results are most easily understood in terms of kinetic effects, which reduce both 18O and 13C below isotopic equilibrium values, and metabolic effects, which only influence the skeletal 13C. Madracis mirabilis is adapted to depths shallower than 20 m, and shows the greatest range in kinetic effects and the strongest metabolic 13C enrichments caused by symbiont photosynthesis. Madracis formosa lives deeper than 40 m, and shows a reduced range of kinetic effects and relatively weak metabolic 13C enrichments. Madracis pharensis inhabits depths from 5 to >60 m, and does not attain the strength of kinetic effects of either of the other two species, apparently because it is not quite as well adapted to rapid growth at either extreme.  相似文献

Summary Analysis of deltorphin A position 4 analogues included: backbone constrained N MeHis, spinacine (Spi), N MePhe and the tetrahydroisoquinoline-3-carboxylic acid (Tic); spatially confined side-chain (Phg); and imidazole alkylation ofl- andd-His4 enantiomers. High selectivity was lost with the following replacements: N MeHis4, N MePhe4 and Phg4 reduced binding and the constrained residues also increasedµ binding; ring closure between the side-chain and amino group to yield Spi4 or Tic4 increasedµ affinity. Imidazole methylation of His4 marginally affected opioid binding and doubled selectivity; alkylatedd-His4-derivatives generally maintained selectivity in spite of decreased affinities. Thus, His4 imidazole preserves selectivity by facilitating high binding and by repulsion at theµ receptor. Several low energy conformers of deltorphin A indicated that the His4 imidazole preferred a spatial orientation parallel to the phenolic side-chain of Tyr1 suggestive that this conformation might contribute to high affinity and selectivity.  相似文献

Novel linear hydroxamate/hydroxycarboxylate siderophores from strains of Pseudomonas cepacia were isolated and named ornibactins. The ornibactins represent modified tetrapeptide siderophores, possessing the sequence l-Orn1(N -OH, N -acyl)-d-threo-Asp(-OH)-l-Ser-l-Orn4(N -OH, N -formyl)-1,4-diaminobutane. The N -acyl groups of Orn1(N -OH, N -acyl) may vary and represent the three acids 3-hydroxybutanoic acid, 3-hydroxyhexanoic acid and 3-hydroxyoctanoic acid, leading to a mixture of three different ornibactins, designated according to their acyl chain length as ornibactin-C4, ornibactin-C6 and ornibactin-C8. Each of the siderophores is accompanied by a small amount of a more hydrophilic component with a 16 a.m.u. higher mass. The structure elucidation was based on results from gas chromatography amino acid analysis, electrospray mass spectrometry, and one- and two-dimensional nuclear magnetic resonance techniques.  相似文献

The effect of interspecific competition and element additions (N and P) on four grassland species (Poa pratensis, Lolium perenne, Festuca valida, Taraxacum officinale) grown under field conditions was studied. Two grasses (L. perenne, F. valida) grown in monoculture (absence of competition) showed lower carbon isotope discrimination (13C) and enriched 15N values. Nitrogen addition (as urea) had inconsistent effects on species 13C while caused enrichment of 15N of P. pratensis and F. valida but strong depletion of 15N of T. officinale. Phosphorous had no significant effect on 13C but depleted 15N of all species.  相似文献

Summary The ratio of deuterium to hydrogen (expressed as D) in hydrogen released as water during the combustion of dried plant material was examined. The D value (metabolic hydrogen) determined on plant materials grown under controlled conditions is correlated with pathways of photosynthetic carbon metabolism. C3 plants show mean D values of-132 for shoots and -117 for roots; C4 plants show mean D values of -91 for shoots and-77 for roots and CAM plants a D value of-75 for roots and shoots. The difference between the D value of shoot material from C3 and C4 plants was confirmed in species growing under a range of glasshouse conditions. This difference in D value between C3 and C4 species does not appear to be due to differences in the D value (tissue water) in the plants as a result of physical fractionation of hydrogen isotopes during transpiration. In C3 and C4 plants the hydrogen isotope discrimination is in the same direction as the carbon isotope discrimination and factors contributing to the difference in D values are discussed. In CAM plants grown in the laboratory or collected from the field D values range from-75 to +50 and are correlated with 13C values. When deprived of water, the D value (metabolic hydrogen) in both soluble and insoluble material in leaves of Kalanchoe daigremontiana Hamet et Perr., becomes less negative. These changes may reflect the deuterium enrichment of tissue water during transpiration, or in field conditions, may reflect the different D value of available water in areas of increasing aridity. Whatever the origin of the variable D value in CAM plants, this parameter may be a useful index of the water relations of these plants under natural conditions.  相似文献

Time-series 18O and 13C records from cohabiting massive coralPorites australiensis and giant clamTridacna gigas from the Great Barrier Reed of Australia, and from calcareous green algae in a core through modernHalimeda bioherm accreting in the eastern Java Sea, provide insights into the complex links between environmental factors and stable isotopes imprinted in these reef skeletal materials. The aragonitic coral and giant clam offer 20 years and 15 years of growth history, respectively. The giant clam yields mean 18O and 13C values of-0.5±0.5 and 2.2±0.2 (n=67), which agree well with the predicted equilibrium values. The coral yields mean 18O and 13C values of-5.6±0.5 and-1.8±0.7 (n=84), offering a striking example of kinetic and metabolic fractionation effects. Although both the coral and giant clam harbor symbionts and were exposed to a uniform ambient environment during their growth histories, their distinct isotopic compositions demonstrate dissimilar calcification pathways. The 18O records contain periodicities corresponding to the alternating annual density bands revealed by X-radiography and optical transmitted light. Attenuation of the 18O seasonal amplitudes occurring in the giant clam record 8 years after skeletal growth commenced is attributed to a changeover from fast to slow growth rates. Extreme seasonal 18O amplitudes of up to 2.2 discerned in both the coral and giant clam records exceed the equivalent seasonal temperature contrast in the reef environment, and are caused by the combined effect of rainfall and evaporation during the monsoon and dry seasons, respectively. Thus in addition of being useful temperature recorders, reef skeletal material of sufficient longevity, such asPorites andTridacna, may also indicate rainfall variations. Changing growth rates, determined from the annual growth bands, may exert a primary control on the coral 13C record which shows a remarkable negative shift of 1.7 over its growth history, by comparison with only 0.15 negative shift in the contemporaneous giant clam record. Use of coral 13C records as proxies of fossil fuel CO2 uptake by the ocean must be regarded with caution. The 18O and 13C records fromHalimeda are remarkably uniform over 1000 years of bioherm accretion history (18O=-1.7±0.2; 13C=3.9±0.1,n=28), in spite of variable Mg-calcite cements present in the utricles. Most of the cement infilling is probably syndepositional, and both theHalimeda aragonite and the Mg-calcite cements containign 12.3 mole % MgCO3 are deposited in isotopic equilibrium. Therefore, in favorable circumstances these algal skeletal remains may act as the shallow water analogs of benthic foraminifera in deep sea sediments in recording ambient sea water isotopic composition and temperature.  相似文献

We have isolated and characterized the cDNA that encodes IgD of fugu (Takifugu rubripes). Though the splicing of 1 with the 1 domain was similar to those reported for teleost IgDs, highly unusual and unprecedented domain duplications were found in the constant region of the fugu IgD. The structure of the fugu IgD is like VDJ-1-(1-2-3-4-5-6)2-7-m1-m2. Genomic sequence analysis of the fugu IgD gene supported the results of cDNA sequencing that the first six domains in the constant region are duplicated. Such a novel duplication pattern has not been reported in any other vertebrates. However, IgD secretory domains could not be identified in this study. The deduced amino acid sequence of the fugu IgD constant region showed high identity (35–55%) to the sequences of previously reported teleost IgDs. Gene expression analyses based on RT-PCR demonstrated that the IgD gene is preferentially expressed in presumptive lymphoid tissues; moreover, in situ hybridization showed that IgD-positive cells are distributed throughout the spleen and head kidney. The expression pattern is similar to that of IgM, corroborating the hypothesis that IgD plays an important role in the humoral immune system of this species.The nucleotide sequence data reported in this paper will appear in the DDBJ/EMBL/GenBank nucleotide sequence databases with the accession numbers AB159481 and AB159482.  相似文献

Summary Carbon isotope composition, photosynthetic gas exchange, and nitrogen content were measured in leaves of three varieties of Metrosideros polymorpha growing in sites presenting a variety of precipitation, temperature and edaphic regimes. The eight populations studied could be divided into two groups on the basis of their mean foliar 13C values, one group consisting of three populations with mean 13C values ca.-26 and another group with 13C values ca.-28. Less negative 13C values appeared to be associated with reduced physiological availability of soil moisture resulting from hypoxic conditions at a poorly drained high elevation bog site and from low precipitation at a welldrained, low elevation leeward site. Gas exchange measurements indicated that foliar 13C and intrinsic wateruse efficiency were positively correlated. Maximum photosynthetic rates were nearly constant while maximum stomatal conductance varied substantially in individuals with foliar 13C ranging from-29 to-24. In contrast with the patterns of 13C observed, leaf nitrogen content appeared to be genetically determined and independent of site characteristics. Photosynthetic nitrogenuse efficiency was nearly constant over the range of 13C observed, suggesting that a compromise between intrinsic water- and N-use efficiency did not occur. In one population variations in foliar 13C and gas exchange with leaf cohort age, caused the ratio of intercellular to atmospheric partial pressure of CO2 predicted from gas exchange and that calculated from 13C to be in close agreement only in the two youngest cohorts of fully expanded leaves. The results indicated that with suitable precautions concerning measurement protocol, foliar 13C and gas exchange measurements were reliable indicators of potential resource use efficiency by M. polymorpha along environmental gradients.  相似文献

Severe coral bleaching occurred throughout the tropics in 1997/98. We report high-resolution skeletal oxygen isotope (18O) and carbon isotope (13C) microprofiles for bleached corals from Pandora Reef, Great Barrier Reef, and Ishigaki Island, Japan, in order to examine the ability of Porites corals to record clear signals of bleaching. Analysis of the annual cycle in 18O revealed abrupt reductions in skeletal extension immediately after the 1997–98 summer temperature maximum, indicating that bleaching inhibits coral calcification. Skeletal 13C in the Ishigaki corals showed lower values during bleaching, indicating depressed coral metabolism associated with a reduction in calcification. In contrast, microprofiles of skeletal 13C from the shaded sides of Pandora Reef corals exhibited little change, possibly because algal photosynthesis was already slow prior to bleaching, thus subduing the 13C-response to bleaching. Comparison of 18O microprofiles from bleached corals with instrumental temperature records showed that Porites corals can recover following 5 months with little skeletogenesis. The results indicate that isotopic microprofiling may be the key to identifying gaps in coral growth that are diagnostic of past bleaching events. We have tested this hypothesis using blue UV fluorescent bands to guide us to coral skeleton where isotope microprofiling identifies bleaching events in 1986, 1989, and 1990. These events, detected by proxy, suggest that coral bleaching may have occurred more commonly on Ishigaki Island than previously recorded.  相似文献

Given a uniform N source, the 15N of barley shoots provided a genotypic range within treatments and a separation between control and salt-stress treatments as great as did 13C*. Plant 15N has been represented in the literature as a bioassay of external source 15N and used to infer soil N sources, thus precluding consideration of the plant as a major cause in determining its own 815N. We believe this to be the first report of plant 15N as a genetic trait. We have previously shown that mitochondrial membrane potential () drop promoted by prooxidants and Ca2+ can be reversed but not sustained by ethylene glycol-bis(-aminoethylether)-N,N,N,N-tetraacetic acid (EGTA) unless dithiothreitol (DTT), a disulfide reductant, is also added [Valle, V. G. R., Fagian, M. M., Parentoni, L. S., Meinicke, A. R., and Vercesi, A. E. (1993).Arch. Biochem. Biophys. 307, 1–7]. In this study we show that catalase or ADP are also able to potentiate this EGTA effect. When EGTA is added long after (12 min) the completion of swelling or elimination, no membrane resealing occurs unless the EGTA addition was preceded by the inclusion of DTT, ADP, or catalase soon after was collapsed. Total recovery by EGTA is obtained only in the presence of ADP. The sensitivity of the ADP effect to carboxyatractyloside strongly supports the involvement of the ADP/ATP carrier in this mechanism. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis of solubilized membrane proteins shows that protein aggregation due to thiol cross-linkage formed during drop continues even after is already eliminated. Titration with 5,5-dithio-bis(2-nitrobenzoic acid) supports the data indicating that the formation of protein aggregates is paralleled by a decrease in the content of membrane protein thiols. Since the presence of ADP and EGTA prevents the progress of protein aggregation, we conclude that this process is responsible for both increased permeability to larger molecules and the irreversibility of drop. The protective effect of catalase suggests that the continuous production of protein thiol cross-linking is mediated by mitochondrial generated reactive oxygen species.  相似文献

The study deals with a comparative analysis of the relative abundances of the carbon isotopes 12C and 13C in the metabolites and biomass of the Burkholderia sp. BS3702 and Pseudomonas putida BS202-p strains capable of utilizing aliphatic (n-hexadecane) and aromatic (naphthalene) hydrocarbons as sources of carbon and energy. The isotope compositions of the carbon dioxide, biomass, and exometabolites produced during the growth of Burkholderia sp. BS3702 on n-hexadecane (13C = –44.6 ± 0.2) were characterized by the values of 13CCO 2 = –50.2 ± 0.4, 13Cbiom = –46.6 ± 0.4, and 13Cexo = –41.5 ± 0.4, respectively. The isotope compositions of the carbon dioxide, biomass, and exometabolites produced during the growth of the same bacterial strain on naphthalene (13C = –21 ± 0.4) were characterized by the isotope effects 13CCO 2 = –24.1 ± 0.4, 13Cbiom = –19.2 ± 0.4, and 13Cexo = –19.1 ± 0.4, respectively. The possibility of using the isotope composition of metabolic carbon dioxide for the rapid monitoring of the microbial degradation of petroleum hydrocarbons in the environment is discussed.  相似文献

Estimates of nitrogen fixation by trees on an aridity gradient in Namibia   总被引:5,自引:0,他引:5  
Summary Nitrogen (N2) fixation was estimated along an aridity gradient in Namibia from the natural abundance of 15N (15N value) in 11 woody species of the Mimosacease which were compared with the 15N values in 11 woody non-Mimosaceae. Averaging all species and habitats the calculated contribution of N2 fixation (N f ) to leaf nitrogen (N) concentration of Mimosaceae averaged about 30%, with large variation between and within species. While in Acacia albida N f was only 2%, it was 49% in Acacia hereroensis and Dichrostachys cinerea, and reached 71% in Acacia melifera. In the majority of species N f was 10–30%. There was a marked variation in background 15N values along the aridity gradient, with the highest 15N values in the lowland savanna. The difference between 15N values of Mimosaceae and non-Mimosaceae, which is assumed to result mainly from N2 fixation, was also largest in the lowland savanna. Variations in 15N of Mimosaceae did not affect N concentrations, but higher 15N-values of Mimosaeae are associated with lower carbon isotope ratios (13C value). N2 fixation was associated with reduced intrinsic water use efficiency. The opposite trends were found in non-Mimosaceae, in which N-concentration increased with 15N, but 13C was unaffected. Summary Expression of -crystallin, a lens-specific protein, in 6-day-old chick embryonic brain cells was examined in situ and in vitro. The presence of minute amounts of -crystallin and its mRNA (-mRNA) in brain cells in situ was demonstrated by immunoblot and Northern blot analysis. In spreading cultures of the brain cells, -crystallin and -mRNA showed a significant increase from their in situ level. Immunohistological staining (peroxidase antiperoxidase) with monospecific anti-serum against -crystallin revealed that -producers were both epithelial cells and dendritic cells. Neither lentoidogenesis nor -crystallin expression was observed. Stimulation of -crystallin synthesis in cultured brain cells differed when compared with transdifferentiating cultures of neural retina cells. In the latter, -crystallin synthesis occurred concomitantly with differentiation of morphologically distinct lens cells containing -crystallin.  相似文献

Natural abundance of 15N in tropical plants with emphasis on tree legumes   总被引:6,自引:0,他引:6  
Natural abundance of 15N ( 15N) of leaves harvested from tropical plants in Brazil and Thailand was analyzed. The 15N values of non-N2-fixing trees in Brazil were +4.5±1.9, which is lower than those of soil nitrogen (+8.0±2.2). In contrast, mimosa and kudzu had very low 15N values (–1.4+0.5). The 15N values of Panicum maximum and leguminous trees, except Leucaena leucocephala, were similar to those of non-N2-fixing trees, suggesting that the contribution of fixed N in these plants is negligible. The 15N values of non-N2-fixing trees in Thailand were +4.9±2.0. Leucaena leucocephala, Sesbania grandiflora, Casuarina spp. and Cycas spp. had low 15N values, close to the value of atmospheric N2 (0), pointing to a major contribution of N2 fixation in these plants. Cassia spp. and Tamarindus indica had high 15N values, which confirms that these species are non-nodulating legumes. The 15N values of Acacia spp. and Gliricidia sepium and other potentially nodulating tree legumes were, on average, slightly lower than those of non-N2-fixing trees, indicating a small contribution of N2 fixation in these legumes.  相似文献

Stable isotope analyses are often used to calculate relative contributions of multiple food sources in an animals diet. One prerequisite for a precise calculation is the determination of the diet-tissue fractionation factor. Isotopic ratios in animals are not only affected by the composition of the diet, but also by the amount of food consumed. Previous findings regarding the latter point are controversial. As stable isotope analyses have often been used to investigate aquatic food webs, an experiment with carp (Cyprinus carpio L.) was carried out to test the influence of the feeding level and individual metabolic rate on 13C and 15N values of the whole body. After an initial phase, 49 carp were assigned randomly to four groups and fed the same diet at different levels for 8 weeks. For 15 fish, the energy budget was determined by indirect calorimetry. Feed and individual fish were analysed for their proximate composition, gross energy content and 13C and 15N values. 13C and 15N values differed significantly at different feeding levels. While 13C values of the lipids and 15N values decreased with increasing feeding rate, 13C values of the lipid-free matter showed a non-linear pattern. Data obtained from fish held in the respirometric system revealed a relationship between 13C values and the percentage retention of metabolizable energy. Our results show that reconstructing the diets of fish from the isotopic ratios when the feeding level and individual metabolic rates are unknown would introduce an error into the data used for back-calculation of up to 1 for both 13C and 15N values and may have substantial effects on the results of calculated diets. As other workers have pointed out, the development and application of stable isotopes to nutritional ecology studies is a field in its infancy and gives rise to erroneous, misleading results without nutritional, physiological and ecological knowledge.  相似文献

In this paper we analysed autotrophic sources of the carbon ( 13C) and the trophic position ( 15N) of Leporinus friderici in the influence area of Corumbá Reservoir, Brazil. We collected samples of muscles of fish from different sizes riparian vegetation, C4 grasses, zooplankton, periphyton and particulate organic carbon (POC). There were significant differences for the carbon isotope proportion found in muscles of L.friderici in the different size groups analysed. The highest values of 13C recorded for middle sized individuals is attributed to the large contribution of C4 plants in their diet. Small individuals sampled upstream also receive similar contribution from C4 plants. In contrast the same size group sampled downstream from the reservoir, has a much smaller of C4 plants. The 13C negative character of small individuals from downstream is due to the larger contribution of C3 plants (except periphyton). At larger sizes we found intermediate 13C values. The 15N proportions we found for each size group were not significantly different, however we found decreasing mean values with increasing size. The trophic level calculated from the dietary data was higher than that found with the 13C concentration in the muscle, except for small individuals, when the values were equal.  相似文献

Summary The 13C values of submerged aquatic plants from contrasting but relatively defined habitats, and the 13C values of emergent, floating and submerged leaves of dimorphic aquatic plants, were measured. In many instances the 13C values of dissolved inorganic carbon in the water were also measured. Plant 13C values in the vicinity of-40 to-50 were found in rapidly flowing spring waters with carbonate 13C values of-16 to-21, consistent with the notion that species such as Fontinalis antipyretica almost exclusively assimilate free CO2 via RuP2 carboxylase. Plant 13C values in the vicinity of-10 to-15 in sluggish water with carbonate 13C values of about-5 were observed, consistent with the notion that boundary layer diffusion and/or HCO3 - uptake may determine the 13C value of submerged aquatic plants in these circumstances. Comparisons of 13C values of the same or related species growing in waters of similar carbonate 13C value but different flow rates confirmed this view; more negative 13C values were frequently associated with plants in fast moving water. In Britain, but not in Finland, the 13C values of submerged leaves of dimorphic plants were almost invariably more negative than in aerial leaves. The 13C value of carbonate from chalk streams and in acid springs indicate substantial inputs of respiratory CO2, as opposed to atmospheric carbon. The contributions of these variations in 13C of the carbon source, and of isotope fractionation in diffusion, to the 13C value of submerged parts of dimorphic plants is discussed.  相似文献

We have determined the 18O and 13C values of azooxanthellate (Lophelia pertusa) and zooxanthellate (Porites lutea) corals at a micrometer scale using an ion microprobe (SIMS—secondary ion mass spectrometry). In P. lutea, centers of calcification are small (10 to 15 m) and difficult to locate during measurements. In L. pertusa, they are large (50 m) and arranged in lines of centers of calcification. Our results show that centers of calcification in L. pertusa have a restricted range of variation in 18O [-2.8±0.3 (PDB)], and a larger range in 13C [14.3 to 10.9 (PDB)]. Surrounding skeletal fibers exhibit large isotopic variation both for C and O (up to 12), and 13C and 18O are positively correlated. The C and O isotopic compositions of the center of calcification deviate from this linear trend at the lightest 18O values of the surrounding fibers. Ion microprobe results on P. lutea demonstrate also a large range of variation for the 18O values (up to 10). No correlation is found with C isotopes that exhibit, in comparison with L. pertusa, a small range of variation (2). This variation of 18O at a micrometer scale is probably the result of two processes: (1) an isotopic equilibrium calcification with 1 pH unit variation in the calcification fluid as indicated by direct measurements of coelenteron pH in the coral Galaxea fascicularis (Al-Horani et al. 2003) and (2) a kinetic fractionation. The 13C apparent disequilibrium in P. lutea may be the result of mixing between metabolic CO2 (respiration) and dissolved inorganic carbon (DIC) coming directly from seawater.  相似文献
