Developments in soil ecotoxicology started with observations on pesticide effects on soil invertebrates in the 1960s. To support the risk assessment of chemicals, in the 1980s and 1990s development of toxicity tests was the main issue, including single species tests and also more realistic test systems like model ecosystems and field tests focusing on structural and functional endpoints. In the mean time, awareness grew about issues like bioavailability and routes of exposure, while biochemical endpoints (biomarkers) were proposed as sensitive and potential early-warning tools. In recent years, interactions between different chemicals (mixture toxicity) and between chemical and other stressors attracted scientific interest. With the development of molecular biology, omics tools are gaining increasing interest, while the ecological relevance of exposure and effects is translating into concepts like (chemical) stress ecology, ecological vulnerability and trait-based approaches. This contribution addresses historical developments and focuses on current issues in soil ecotoxicology. It is concluded that soil ecotoxicological risk assessment would benefit from extending the available battery of toxicity tests by including e.g. isopods, by paying more attention to exposure, bioavailability and toxicokinetics, and by developing more insight into the ecology of soil organisms to support better understanding of exposure and long-term consequences of chemical exposure at the individual, population and community level. Ecotoxicogenomics tools may also be helpful in this, but will require considerable further research before they can be applied in the practice of soil ecotoxicological risk assessment.  相似文献

The European Water Framework Directive (WFD) represents a transformation of the guidelines for water quality assessment and monitoring across all EU Member States. At present, it is widely accepted that the WFD requires holistic and multidisciplinary ecological approaches by integrating multiple lines of evidence. Within the scope of the WFD, the scientific community identified clear opportunities to take advantage of an ecotoxicological line of evidence. In this context, ecotoxicological tools, namely biomarkers and bioassays, were proposed to contribute to the integration of the chemical and biological indicators, and thus to provide an overall insight into the quality of a water body. More than one decade after the publication of the WFD, we reviewed the studies that have attempted to integrate ecotoxicological tools in the assessment of surface water bodies. For this purpose, we reviewed studies providing an ecological water status assessment through more conventional community based approaches, in which biomarkers and/or bioassays were also applied to complement the evaluation. Overall, from our review emerges that studies at community level appear suitable for assessing the ecological quality of water bodies, whereas the bioassays/biomarkers are especially useful as early warning systems and to investigate the causes of ecological impairment, allowing a better understanding of the cause–effect-relationships. In this sense, community level responses and biomarkers/bioassays seem to be clearly complementary, reinforcing the need of combining the approaches of different disciplines to achieve the best evaluation of ecosystem communities’ health.  相似文献

环状RNA(circular RNAs,circRNAs)是一类共价闭合环状非编码RNA,具有进化上保守、结构上稳定、组织特异性表达等特点。CircRNAs可作为miRNAs海绵影响其对基因的调控,还可与RNA结合蛋白(RNA binding proteins,RBPs)相互作用,也有研究表明某些circRNAs还具有被翻译成蛋白质的潜能。CircRNAs已被证实对某些疾病具有特异性、稳定性和调节功能,如癌症、糖尿病、心血管疾病、神经退行性疾病等,其可作为潜在的诊断、预后生物标志物和治疗靶点。最近,有研究发现circRNAs参与了环境化学污染物诱导的毒性效应发生及发展的过程。目前,生态毒理学研究中评价环境化学污染物和毒效应之间关系的毒性终点通常会受遗传多态性和表观遗传学影响,考虑到经环境化学污染物暴露后生物体内circRNAs差异性表达的现象,或许在生态毒理学研究中circRNAs也有作为生物标志物的可能性。基于此,对circRNAs的生物合成与降解、生物学功能、分析方法及其目前在生态毒理学研究中的应用展开综述,并对其作为分子生物标志物在环境污染物暴露早期诊断和生态风险评价中的应用进行了展望,以期为生态毒理学研究和环境风险评价提供参考。  相似文献

This study presents data on the detailed evaluation (tier 2) of a site-specific ecological risk assessment (ssERA) in a former smelter area contaminated with metals (Santo Amaro, Bahia, Brazil). Combining information from three lines of evidence (LoE), chemical (ChemLoE), ecotoxicological (EcotoxLoE) and ecological (EcoLoE), in the Triad approach, integrated risk values were calculated to rank sites and confirm the potential risk disclosed with tier 1. Risk values were calculated for the habitat and for the retention functions in each sampling point. Habitat function included the ChemLoE calculated from total metal concentrations. The EcotoxLoE was based on reproduction tests with terrestrial invertebrates (Folsomia candida, Enchytraeus crypticus, Eisenia andrei), shoot length and plant biomass (Avena sativa, Brassica rapa). For the EcoLoE, ecological parameters (microbial parameters, soil invertebrate community, litter breakdown) were used to derive risk values. Retention function included the ChemLoE, calculated from extractable metal concentrations, and the EcotoxLoE based on eluate tests with aquatic organisms (Daphnia magna reproduction and Pseudokirchneriella subcapitata growth). Results related to the habitat function indicated that the metal residues are sufficient to cause risk to biota, while the low metal levels in extracts and the general lack of toxicity in aquatic tests indicated a high soil retention capacity in most sampling points. Integrated risk of tier 2 showed the same trend of tier 1, suggesting the need to proceed with remediation actions. The high risk levels were related to direct toxicity to organisms and indirect effects, such as failure in the establishment of vegetation and the consequent loss of habitat quality for microorganisms and soil fauna. This study shed some light on the selection of tools for the tier 2 of an ssERA in tropical metal-contaminated sites, focusing on ecological receptors at risk and using available chemical methods, ecological surveys and ecotoxicity tests.  相似文献

Ecological models are useful tools for evaluating the ecological significance of observed or predicted effects of toxic chemicals on individual organisms. Current risk estimation approaches using hazard quotients for individual-level endpoints have limited utility for assessing risks at the population, ecosystem, and landscape levels, which are the most relevant indicators for environmental management. In this paper, we define different types of ecological models, summarize their input and output variables, and present examples of the role of some recommended models in chemical risk assessments. A variety of population and ecosystem models have been applied successfully to evaluate ecological risks, including population viability of endangered species, habitat fragmentation, and toxic chemical issues. In particular, population models are widely available, and their value in predicting dynamics of natural populations has been demonstrated. Although data are often limited on vital rates and doseresponse functions needed for ecological modeling, accurate prediction of ecological effects may not be needed for all assessments. Often, a comparative assessment of risk (e.g., relative to baseline or reference) is of primary interest. Ecological modeling is currently a valuable approach for addressing many chemical risk assessment issues, including screening-level evaluations.  相似文献

Recent increases in international trade have increased the cost to control and eradicate exotic species. Although many species are under quarantine control for agriculture, forestry, and public health, most species invisible to the naked eye are ignored because of the lack of both specialized assessors and risk assessments. We developed a species risk assessment particularly adapted to fungi, nematodes, and mites, that might be unintentionally introduced with exotic forest products and become threats to terrestrial ecosystems. We developed our assessment with reference to existing risk assessments for exotic organisms, including their ecological features such as phoresy and parasitism. We then tested our assessment with well-known organisms and assessed the risks of organisms unintentionally introduced into Japan. The assessment demonstrated scientifically acceptable scores for each organism. We suggest quarantine control of risk pathways as a practical approach for controlling unintentionally introduced organisms that are invisible to the naked eye.  相似文献

Epigenetic inheritance occurs due to different mechanisms such as chromatin and histone modifications, DNA methylation and processes mediated by non-coding RNAs. It leads to changes in gene expressions and the emergence of new traits in different organisms in many diseases such as cancer. Recent advances in experimental methods led to the identification of epigenetic target sites in various organisms. Computational approaches have enabled us to analyze mass data produced by these methods. Next-generation sequencing (NGS) methods have been broadly used to identify these target sites and their patterns. By using these patterns, the emergence of diseases could be prognosticated. In this study, target site prediction tools for two major epigenetic mechanisms comprising histone modification and DNA methylation are reviewed. Publicly accessible databases are reviewed as well. Some suggestions regarding the state-of-the-art methods and databases have been made, including examining patterns of epigenetic changes that are important in epigenotypes detection.  相似文献

Nematodes – there is more to it … Nematodes play a central role – not only in a negative way as dreaded parasites of humans, animals and plants, where they may be harmful in terms of health and economy. The majority of nematodes turn out to be highly beneficial: Firstly, they are a vital part of the food web and by this indispensably involved in nutrient cycles. Ecotoxicological tests (or bioassays) are controlled, reproducible tests where ecological responses are determined quantitatively. Due to numerous difficulties arising when airborne emissions are sampled, relatively few ecotoxicological assays have been applied. Aerosol particles are generally collected on a filter, which limits the quantity of the sample, thus also limiting the range of available test organisms. Bacterial bioassays require low sample quantity, and make a good choice when eco‐ or genotoxicity of the sample are to be determined. Of bacterial assays, the bioluminescence inhibition test has been proven applicable for assessing toxicity of airborne contaminants. A modern concept for the development of novel antiparasitic drugs is the combination of bioinformatics and chemoinformatics approaches. This covers, for example, the identification of target proteins serving as molecular points of attack for parasiticides--the idea is that, owing to some essential role, inhibition of a target protein should eradicate the parasite. To prevent toxicity problems for vertebrate host organisms, it is advantageous that these proteins show significant differences from their vertebrate counterparts. In the present work, we identified potential target proteins in parasitic nematodes (Ascaris suum, Brugia malayi, and Haemonchus contortus) and arthropods (Boophilus microplus and Rhipicephalus appendiculatus) using bioinformatic sequence comparison methods on expressed sequence tags. Interesting target proteins (e.g., S-adenosyl-l-methionine synthetase) were characterized in detail by subjecting them to in-depth bioinformatic analysis. S-Adenosyl-l-methionine synthetase was also used to elucidate chemoinformatics approaches like homology modeling and docking, which represent appropriate methods for generating valuable data for the development of new drug candidates.  相似文献

随着我国快速城市化以及产业结构的调整,遗留下了大量的污染场地,发展和实施污染场地土壤生态风险评估是进行大规模污染场地修复行动的必要条件。本文围绕污染场地土壤生态风险评估的科学原理、框架构建及技术方法等方面的关键问题: 1)评估框架的场地实际针对性;2)概念模型的不确定性;3)土壤复合污染毒性机制;4)评估终点筛选;5)评估方法和框架构建等展开讨论,指出土壤复合污染的制毒机制,即污染物生物有效性和联合效应是污染场地土壤生态风险评估的关键科学问题。耦合美国环保局四步法和欧盟层级法的“证据-权重法”评估框架适用于野外复杂环境条件下的土壤污染生态风险评估。建议今后重点开展以下5个方面的工作: 1)污染场地土壤生态风险评估技术框架与风险管控技术框架之间的联合;2)概念模型研究;3)基于过程的场地土壤污染物反应运移模型研究;4)场地土壤复合污染生态毒理学机制研究;5)生态系统高水平生态风险评估终点研究。旨在为形成我国本土污染场地土壤生态风险评估技术指南提供理论基础和构架。  相似文献

参考国际经济合作与发展组织(OECD)化学品生态毒性测试标准方法,以绿藻(Scenedesmus oblignus)和大型蚤(Daphnia magna)为受试生物,研究了3种金属氧化物纳米颗粒(纳米氧化锌nZnO、纳米二氧化钛nTiO2、纳米氧化铝nAl2O3)水悬浮液的水生生态毒性.结果发现,不同的纳米颗粒具有不同的毒性:nZnO、nTiO2和nAl2O3对斜生栅藻生长的 96 h半效应浓度(EC50)值分别为1.049、15.262、>1000 mg · L-1,而对大型蚤活动抑制的 48 h EC50 值则分别为0.622、35.306 mg · L-1和114.357 mg · L-1.据此可得到3 种金属氧化物纳米颗粒水悬浮液的毒性大小顺序为:nZnO >nTiO2 > nAl2O3.此外,不同的生物对金属氧化物纳米颗粒的敏感性也不同,除nTiO2以外,大型蚤对另外2种纳米颗粒的敏感性强于斜生栅藻.实验结果表明,人工纳米材料的生态毒性和环境效应不容忽视,应重视并深入研究此类纳米材料的毒性作用机制和影响因子,以便能对其进行更好的风险管理.  相似文献

Reflections on Plant and Soil Nematode Ecology: Past,Present and Future   总被引:1,自引:0,他引:1  
The purpose of this review is to highlight key developments in nematode ecology from its beginnings to where it stands today as a discipline within nematology. Emerging areas of research appear to be driven by crop production constraints, environmental health concerns, and advances in technology. In contrast to past ecological studies which mainly focused on management of plant-parasitic nematodes, current studies reflect differential sensitivity of nematode faunae. These differences, identified in both aquatic and terrestrial environments include response to stressors, environmental conditions, and management practices. Methodological advances will continue to influence the role nematodes have in addressing the nature of interactions between organisms, and of organisms with their environments. In particular, the C. elegans genetic model, nematode faunal analysis and nematode metagenetic analysis can be used by ecologists generally and not restricted to nematologists.  相似文献

随着纳米技术产业的高速发展,大量工程纳米颗粒物(Engineering nano-particles,ENPs)被排放到自然水环境中,因此对其进行生态毒性及环境风险的研究尤为迫切。综述了ENPs在水环境中的毒理学机理及理想模式生物筛选的研究进展。目前的研究表明ENPs的毒性作用机制主要包括两方面:一是影响细胞信号通路,二是氧化应激造成基因表达的变化。此外,光催化活性、细胞表面附着、溶解特性、表面特征、赋存形态、溶剂效应及与其他环境污染物的协同作用也是可能的毒性作用机理。模式生物的筛选与确定在纳米生态毒理学研究中极为重要。鱼类作为水环境中普遍存在的脊椎动物,群落庞大,其具有行为端点敏感性高、且在生物毒性实验中存在明显的量效关系等特征,被认为是研究ENPs生态毒理学最适合的水生模式生物。研究表明针对在ENPs影响下的未成年鱼类的行为特征研究比传统的胚胎发育及致死率研究更为有效。无脊椎动物和浮游植物同样在各种水环境中普遍存在,对环境污染物极为敏感,且对有害物质具有显著的富集放大效应,因此作为模式生物也具有一定的优势。  相似文献

While nematodes are most commonly known for their negative impact on plants, animals, and humans, there are a number of species which are commercially explored. This review highlights some of the most important success stories for the application of nematodes. They are used as bioindicators in ecological and toxicity studies, as model organisms for elucidating fundamental biological questions and for high throughput screening of drugs. Besides these indirect uses, direct applications include the use of Beddingia siricidicola against a major forest pest and the commercialization of Steinernema, Heterorhabditis, and Phasmarhabditis as biological pest control products. New directions for the commercialization of nematodes are the use as living food, specifically loaded with essential nutrients for various fish and shrimp larvae. Even human parasites or closely related species have been successfully used for curing autoimmune disorders and are currently in the process of being developed as drugs. With the striving development of life sciences, we are likely to see more applications for nematodes in the future. A prerequisite is that we continue to explore the vast number of yet undiscovered nematode species.  相似文献

环境中的化学品往往以低剂量混合形式存在.对单一化学品高剂量暴露下的生态毒性研究成果,难以适用于环境中低剂量混合物的生态毒理效应诊断及风险评价.文中概述了低剂量化学品混合污染生态毒理及风险评价方面的研究进展,主要包括低剂量化学品混合污染诊断的分子毒理研究方法、风险评价方法,并介绍了简单和复杂混合物的风险评价方案.对低剂量混合污染生态毒理与风险评价研究的发展动向提出了见解,指出低剂量化学混合物的研究需要寻找敏感终点,引入多学科手段,积累更多的数据,建立完善、统一的评价体系.  相似文献

Macromolecules such as DNA, RNA, and proteins are widely used to quantify diversity in natural communities, to monitor the dispersal of organisms and their genes, and to trace phylogenetic relationships among organisms. With such widespread use of molecules as markers, it is easy to forget that they perform functions that are integral to the survival of organisms. The structural and functional properties of macromolecules have been intensively studied in genetics, biochemistry, and physiology. These fields, however, have not generally focused on the ecological significance of molecular variation, but instead have used defective variants as tools for identifying structure and function. Understanding the significance of molecular variation for organismal success or failure is a central problem in ecology, one whose solution will require the integration of diverse approaches and perspectives from ecology, molecular biology, genetics, physiology, and evolutionary biology. I review several studies of bacterial populations evolving in simple environments that have begun to integrate these approaches and perspectives in order to examine the consequences of molecular variation for ecological performance.  相似文献
