Abstract: Black spruce (Picea mariana), white spruce (Picea glauca), and jack pine (Pinus banksiana) seedlings were inoculated with Hebeloma crustuliniforme or Laccaria bicolor and subjected to NaCl and Na2SO4 treatments. The effects of ectomycorrhizas on salt uptake, growth, gas exchange, and needle necrosis varied depending on the tree and fungal species. In jack pine seedlings, ectomycorrhizal (ECM) fungi reduced shoot and root dry weights and in the ECM white spruce, there was a small increase in dry weights. Sodium chloride treatment reduced net photosynthesis and transpiration rates in the three studied tree species. However, NaCl-treated black spruce and jack pine colonized by H. crustuliniforme maintained relatively high photosynthetic and transpiration rates and needle necrosis of NaCl-treated black spruce seedlings was reduced by the ECM fungi. Higher concentrations of Na+ were found in shoots compared with roots of the three examined conifer species. ECM fungi reduced the concentrations of Na+ mainly in the shoots and this reduction was greater in plants treated with NaCl compared with Na2SO4. Shoots contained generally higher concentrations of Cl- compared with roots. In the NaCl-treated black spruce and white spruce, both ECM species significantly reduced Cl- concentrations. Our results point to overall greater phytotoxicity of NaCl compared with Na2SO4 and support our earlier findings which demonstrated beneficial effects of ECM fungi for woody plants exposed to NaCl stress.  相似文献

To examine the mechanisms of earlier reported alleviation of fluoride injury in ectomycorrhizal plants by NaCl, jack pine (Pinus banksiana) and white spruce (Picea glauca) seedlings were subjected to 1 mM and 5 mM KF in the presence of either 60 mM NaCl or 10% polyethylene glycol 3350 (PEG) for 2 weeks. Before the treatments, seedlings had either been inoculated with the ectomycorrhizal fungus Suillus tomentosus or remained non-inoculated. The inoculation with S. tomentosus reduced Na uptake by shoots and roots of jack pine seedling and by roots of white spruce that were treated with 60 mM NaCl. Mycorrhizal associations also drastically decreased fluoride uptake by jack pine seedlings, but did not affect shoot fluoride concentrations in white spruce. When NaCl was replaced by PEG in the 5 mM KF treatment solution, shoot fluoride concentrations were reduced by more than twofold without corresponding reductions in transpiration rates in mycorrhizal and non-mycorrhizal white spruce seedlings. When fluoride was present in the treatment solution, Na concentrations were lower in shoots and roots of both jack pine and white spruce mycorrhizal and non-mycorrhizal seedlings. The results suggest that Suillus tomentosus may help alleviate the effects of soil fluoride and salinity in jack pine and that fluoride uptake in white spruce is sensitive to osmotic stress.  相似文献

The effectiveness of ectomycorrhizal inoculation at the tree nursery seedling production stage on growth and survival was examined in jack pine (Pinus banksiana) and white spruce (Picea glauca) planted in oil sands reclamation sites. The seedlings were inoculated with Hebeloma crustuliniforme strain # UAMH 5247, Suillus tomentosus strain # UAMH 6252, and Laccaria bicolor strain # UAMH 8232, as individual pure cultures and in combinations. These treatments were demonstrated to improve salinity resistance and water uptake in conifer seedlings. The field responses of seedlings to ectomycorrhizal inoculation varied between plant species, inoculation treatments, and measured parameters. Seedling inoculation resulted in higher ectomycorrhizal colonization rates compared with non-inoculated control, which had also a relatively small proportion of roots colonized by the nursery contaminant fungi identified as Amphinema byssoides and Thelephora americana. Seedling inoculation had overall a greater effect on relative height growth rates, dry biomass, and stem volumes in jack pine compared with white spruce. However, when examined after two growing seasons, inoculated white spruce seedlings showed up to 75 % higher survival rates than non-inoculated controls. The persistence of inoculated fungi in roots of planted seedlings was examined at the end of the second growing season. Although the inoculation with H. crustuliniforme triggered growth responses, the fungus was not found in the roots of seedlings at the end of the second growing season suggesting a possibility that the observed growth-promoting effect of H. crustuliniforme may be transient. The results suggest that the inoculation of conifer seedlings with ectomycorrhizal fungi could potentially be carried out on a large scale in tree nurseries to benefit postplanting performance in oil sands reclamation sites. However, these practices should take into consideration the differences in responses between the different plant species and fungal strains.  相似文献

We investigated the thermal acclimation of photosynthesis and respiration in black spruce seedlings [ Picea mariana (Mill.) B.S.P.] grown at 22/14 °C [low temperature (LT)] or 30/22 °C [high temperature (HT)] day/night temperatures. Net CO2 assimilation rates ( A net) were greater in LT than in HT seedlings below 30 °C, but were greater in HT seedlings above 30 °C. Dark and day respiration rates were similar between treatments at the respective growth temperatures. When respiration was factored out of the photosynthesis response to temperature, the resulting gross CO2 assimilation rates ( A gross) was lower in HT than in LT seedlings below 30 °C, but was similar above 30 °C. The reduced A gross of HT seedlings was associated with lower needle nitrogen content, lower ribulose 1·5-bisphosphate carboxylase/oxygenase (Rubisco) maximum carboxylation rates ( V cmax) and lower maximum electron transport rates ( J max). Growth treatment did not affect V cmax :  J max. Modelling of the CO2 response of photosynthesis indicated that LT seedlings at 40 °C might have been limited by heat lability of Rubisco activase, but that in HT seedlings, Rubisco capacity was limiting. In sum, thermal acclimation of A net was largely caused by reduced respiration and lower nitrogen investments in needles from HT seedlings. At 40 °C, photosynthesis in LT seedlings might be limited by Rubisco activase capacity, while in HT seedlings, acclimation removed this limitation.  相似文献

Drought adjustments were compared in black spruce ( Picea mariana [Mill] B.S.P), and jack pine ( Pinus banksiana [Lamb.]) by subjecting seedlings to five cycles of dehydration and rehydration. A computer-controlled root misting chamber system, supplied low (−1.5 MPa), moderate (−2.0 MPa), and severe (−2.5 MPa) dehydration, respectively, in cycles 1, 3 and 5. Although cell water relations failed to adjust to chronic dehydration, there was limited osmotic adjustment in black spruce (cycle 3), and water was re-allocated from the apoplast to the symplast in jack pine (cycles 1 and 3). Dehydration postponement was more important than dehydration tolerance. Jack pine was better able to postpone dehydration than black spruce. Specific conductivity, the hydraulic conductivity per unit stem cross-sectional area, was lower in jack pine and slower to decline during chronic dehydration. When specific conductivity was corrected for the greater leaf area in black spruce, the leaf-specific conductivity did not differ in the two species. There was no increase in needle leakage in jack pine and stomata in jack pine seedlings reopened fully after rehydration. Black spruce was more of a 'water spender', and less water stress (−2.0 MPa, cycle 3) was required to lower specific conductivity, compared to jack pine (−2.5 MPa, cycle 5). Leakage from needle membranes increased in black spruce, and stomata failed to reopen after rewatering (cycles 3 and 5). A greater needle area, smaller root system, and a higher specific conductivity lowered the water stress threshold for cavitation in black spruce, which is confined to moister sites in the boreal forest. Jack pine had a larger root system, smaller needle area and lower specific conductivity than black spruce. Because of these static features, jack pine is more drought tolerant and it is often found on sites that are too hot and dry for black spruce.  相似文献

 Plant responses to saturation vapour pressure deficit (SVPD) were studied by subjecting black spruce [Picea mariana (Mill) B.S.P.] and jack pine seedlings (Pinus banksiana Lamb.) to humid (0.3 – 0.8 kPa) or dry (2.0 – 2.5 kPa SVPD) regimes for 4 weeks using a computer-controlled environmental system to control diurnal variation in SVPD. Dry matter accumulation in needles was not altered by increasing SVPD. However, root growth declined by 60% which increased shoot to root ratio and reduced total seedling dry weight in both black spruce and jack pine. Relative growth rate of jack pine also declined to about half the rate of plants grown under humid conditions. In situ root marking studies showed that the decline in root growth of jack pine under the high SVPD was the result of reduced lateral root initiation, whereas root elongation was unaffected by humidity. A 4-week exposure to dry air increased abscisic acid (ABA) levels in needles, but not roots, of jack pine whereas ABA levels in black spruce were not altered. A short (3-day) exposure failed to increase needle ABA levels in either species. These results suggest that the responses of conifers to dry air were not the result of ABA accumulation. Six-week-old, mycorrhiza-free, bareroot jack pine and black spruce seedlings were outplanted in ten reforestation sites, situated between 45–48° latitude N and 69–74° longitude W, within the province of Quebec, representing diverse operational forestry disturbances and ecological conditions. Two months after outplanting, root systems of black spruce seedlings had fewer mycorrhizae than those of jack pine seedlings. Ectomycorrhizal colonization on black spruce seedlings did not vary significantly with the reforestation site. Percent mycorrhizal colonization for these seedlings was positively correlated with seedling dry weight while with the jack pine seedlings, mycorrhizal colonization varied significantly with the outplanting site and there was no correlation between mycorrhizal formation and seedling dry weight. Multiple linear regressions showed pH to be a determinant soil factor for mycorrhizal colonization for the two species. Drainage was the other influential factor affecting colonization of black spruce while organic matter accumulation was more important for jack pine. Inoculation with selected ectomycorrhizal fungi could be more important for black spruce than for jack pine seedlings.  相似文献

To study the role of low UV‐B radiation in modulating the response of antioxidants to ozone, 4‐year‐old pine ( Pinus sylvestris L.) and spruce ( Picea abies L.) seedlings potted in natural soil, were exposed in phytochambers to fluctuating ozone concentrations between 9 and 113 nl 1−1 according to field data recorded at Mt Wank (1175 m above sea level, Bavaria, Germany) and two‐times ambient O3 levels. UV‐B radiation was either added at a biologically effective level of ca 1.2 kJ m−2 day−1 , which is close to that found in March at Mt Wank, or was excluded by filters (<0.08 kJ m−2 day−1). After one growth phase current‐year needles were collected and analysed for antioxidative enzyme activities (superoxide dismutase, SOD, EC 1.15.1.1; catalase, CAT, EC 1.11.1.6; guaiacol peroxidase, POD, EC 1.11.1.7) and soluble antioxidants (ascorbate, glutathione). CAT, POD, ascorbate and glutathione, but not SOD, were increased in needles of both species in response to twice ambient O3 levels. UV‐B radiation in the presence of ambient O3 caused an increase in total SOD activity in spruce but had no effects on antioxidants in pine. Twice ambient O3 levels together with low UV‐B radiation counteracted the O3‐induced increases in ascorbate and CAT in pine but not in spruce. Under these conditions spruce needles showed the highest antioxidative protection and revealed no indication of lipid peroxidation. Pine needles exposed to UV‐B and elevated O3 levels showed elevated lipid peroxidation and a 5‐fold increase in dehydroascorbate, suggesting that this species was less protected and suffered higher oxidative stress than spruce.  相似文献

Within its wide range across Canada, jack pine is exposed to salinity from both natural and anthropogenic sources. To compare the effects of Cl and SO4 on salt injury, sand and solution-culture grown jack pine ( Pinus banksiana Lamb.) seedlings were treated with nutrient solutions containing 60 or 120 m M NaCl, 60 m M Na2SO4, or a mixture of 60 m M NaCl and 30 m M Na2SO4. After 5 weeks of salt treatments, concentrations of Cl, K, Na, and SO4 were determined in roots, stem and needles of the current and previous years growth, and in necrotic needles. To determine the role of water uptake in the absorption and translocation of salts in plants, total transpiration was measured as the loss of water from a sealed system and related to total plant uptake of Cl, Na, and SO4. Sodium uptake and root-to-shoot transport rates were greater in treatments containing Cl. A delay in root-to-shoot transport of both Na and Cl indicates retention of these ions in the roots. Electrolyte leakage of needles was more closely related to treatment Cl concentrations than treatment Na concentrations. The transport of Na ions to the shoot was related to the presence of Cl, but was not related to transpiration rate.  相似文献

Summary Methods were developed for transient gene expression in protoplasts of black spruce (Picea mariana) and jack pine (Pinus banksiana). Protoplasts were isolated from embryogenic suspension cultures of black spruce and from non-embryogenic suspensions of jack pine. Using electroporation, transient expression of the chloramphenicol acetyltransferase (CAT) gene was assayed and shown to be affected by the cell line used, by voltage, temperature, and by the plasmid concentration and conformation. Increasing the plasmid DNA concentration (0–150g ml–1) resulted in higher levels of transient CAT expression. In jack pine, linearized plasmid gave 2.5 times higher levels of CAT enzyme activity than circular. Optimal voltage varied for each cell line of the two species within the range 200–350 V cm–1 (960 F). A heat shock treatment of protoplasts for 5 min at 45 °C resulted in enhanced CAT gene expression for both species.NRCC No. 30491  相似文献

 Japanese larch (Larix kaempferi), white spruce (Picea glauca), black spruce (Picea mariana), red spruce (Picea rubens), jack pine (Pinus banksiana), mugo pine (Pinus mugo), red pine (Pinus resinosa), Japanese black pine (Pinus thunbergii) and Douglas-fir (Pseudotsuga menziesii var. menziesii), were inoculated to test the effective host range of the ectomycorrhizal fungus Laccaria proxima and the possibility of utilizing pulp waste as a potting medium for containerized seedling production. Laccaria proxima tended to improve the container growth of Japanese black pine and white spruce, and significantly improved that of jack pine, mugo pine, black spruce, red spruce and Douglasfir. The growth of red pine and Japanese larch were only slightly improved with L. proxima. Pulp waste (33% by volume) had negative effects on tree seedling growth, except for Douglasfir (no significant effect). The interactions of Laccaria proxima and pulp waste varied; the hosts were significantly positive (P<0.01) in the case of jack pine and black spruce, but there was no significant effect for the rest. Negative effects were found with Japanese black pine. Use of pulp waste in seedling production of jack pine, black spruce, mugo pine, red spruce and Douglasfir inoculated with L. proxima and of Japanese black pine both with and without L. proxima is feasible, but further research is necessary to determine the optimal percentage of pulp waste that can be utilized in seedling production of tree species and the field performance of these seedlings. Ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco; EC 4.1.1.39) from fir ( Abies alba Mill.) and spruce ( Picea abies [L.] Karst.) needles was purified to homogeneity. The enzyme was isolated from crude extracts through quantitative precipitation in 40-55% and 40-60% (NH4)2SO4 for fir and spruce. respectively, followed by linear sucrose gradient centrifugation. Using two dimensional gel electrophoresis, the isoelectric points were determined. For the large subunit (LSU) it was 6.7 for both species, and for the small subunit (SSU) it was 7.1 and 7.7 for fir and spruce, respectively. Very few differences in tryptic peptides and amino acid composition of Rubisco LSU were observed between fir and spruce. By contrast, marked differences characterized the same analyses for the Rubisco SSU of the two species. Moreover, substitution of residues was observed in the sequenced N-terminal region when comparing fir and spruce SSU. The Ouchterlony technique showed no immu-nochemical difference between Rubisco of fir and spruce when a rabbit antiserum to spinach Rubisco was used. The Eadie-Hofstee plots of carboxylase activity indicated that the apparent Km(CO2) were 31 and 36 μ M for the fir and spruce enzymes, respectively.  相似文献

Short photoperiod induces growth cessation in seedlings of Norway spruce ( Picea abies (L.] Karst.). Application of different gibberellins (GAS) to seedlings growing under a short photoperiod show that GA9 and GA20 can not induce growth. In contrast application of GA, and GA4 induced shoot elongation. The results indicate that 3β-hydroxylation of GA9 to GA4 and of GA20 to GA1 is under photoperiodic control. To confirm that conclusion, both qualitative and quantitative analyses of endogenous GAs were performed. GA1, GA3, GA4, GA7, GA9, GA12, GA15, GA15, GA20, GA29, GA34 and GA51 were identified by combined gas chromatography-mass spectrometry in shoots of Norway spruce seedlings. The effect of photoperiod on GA levels was determined by using deuterated and 14C-labelled GAs as intermal standards. In short days, the amounts of GA9, GA4 and GA1 are less than in plants grown in continuous light. There is no significant difference in the amounts of GA3, GA12, and GA20 between the different photoperiods. The lack of accumulation of GA9 and GA20 under short days is discussed.  相似文献

The seasonal changes in photosynthetic properties in 1-year-old needles of Sakhalin spruce ( Picea glehnii ) were measured using the chlorophyll fluorescence technique at various temperatures (5, 10, 20, 25 and 30°C). In the course of seasonal change, a temporary decrease in the quantum yield of PSII electron transport (ΦPSII) was observed just before budbreak. A decline in photochemical quenching ( q P) was observed at the same time as that of ΦPSII but only at the two lowest temperatures (5 and 10°C). Photochemical efficiency of open PSII ( F v'/ F m') also declined just before budbreak at 25 and 30°C. An increase in thermal energy dissipation as indicated by a decrease in F v'/ F m' before budbreak was not significant at lower temperatures (5 and 10°C) in spite of the declines in q P. This implies that thermal energy dissipation necessitated by the decline in ΦPSII might not be sufficiently strong to prevent a decline in q P at lower temperatures. On the other hand, at higher temperatures no decline was observed in q P because ΦPSII decreased to a relatively small extent, therefore thermal energy dissipation is sufficient in coping with the excessive energy accumulation in PSII. Seedlings of Sakhalin spruce exposed to ambient air temperature below 10°C before budbreak exhibited photoinhibition indicated by a decrease in the maximal photochemical efficiency of PSII ( F v/ F m) after an overnight dark adaptation. The present study suggests that 1-year-old shoots of Sakhalin spruce have an increased susceptibility to photoinhibition at low temperature just before budbreak.  相似文献

The effects of sodium fluoride (0.3, 5 and 10 m M NaF) on root hydraulic conductivity, and gas exchange processes were examined in aspen ( Populus tremuloides Michx.) seedlings grown in solution culture. A long-term exposure of roots to NaF significantly decreased root hydraulic conductivity ( L p) and stomatal conductance ( g s). Root absorbed NaF significantly affected electrolyte leakage in leaf tissues and substantially restricted leaf expansion. NaF did not significantly affect leaf chlorophyll contents but decreased net photosynthesis ( P n). A short-term exposure of excised roots to 5 m M NaF and KF significantly decreased root water flow ( Q v) with a concomitant decline in root respiration and reduced g s when applied through intact roots or excised stems. The same molar concentration of NaCl also decreased Q v and g s in intact seedlings, but to a lesser extent than NaF or KF, and did not significantly affect root respiration. The results suggest that fluoride metabolically inhibited Q v or L p, probably by affecting water channel activity. We suggest that the metabolic inhibition of L p by root-absorbed fluoride affected gas exchange and leaf expansion in aspen seedlings.  相似文献

Summary The organization of microtubules during interphase and prophase in embryogenic cultures of black spruce (Picea mariana) was investigated by indirect immunofluorescence. Somatic embryos of black spruce possessed an extensively branched and interconnecting network of fine interphase cortical microtubules. The development of pre-prophase bands (PPBs) in embryogenic black spruce cultures was compared with that in non-embryogenic cell cultures of jack pine (Pinus banksiana). PPBs in both species were initially arranged as a very broad array of microtubules, later (early to mid-prophase) becoming narrower and more intensely fluorescent. The occurrence of pre-prophase bands in relation to the number of phragmoplasts (i.e. PPB index) of black spruce somatic embryos was significantly higher (p<0.01) than that found for jack pine cells.  相似文献

Spruce seedlings [ Picea abies (L.) Karst.] were exposed to a range of concentrations of Zn, Cd, Hg and methyl-Hg for 5 weeks. The chlorophyll and water content of the needles were then estimated. The rates of photosynthesis, transpiration and dark respiration of the intact plant were determined using a Li-cor portable photosynthesis measuring system. Chlorophyll and water contents of needles decreased in response to all metal treatments, as did CO2 uptake. At 1 μ M Cd, 0.1 μ M Hg and 30 and 60 μ M Zn, the decrease in CO2 uptake could be accounted for by decreased chlorophyll concentrations. Decreased transpiration was only found at 5 μ M Cd and 0.01 μ M methyl-Hg. At 5 μ M Cd most of the decrease in CO2 uptake could be explained by decreased chlorophyll levels and stomatal closure induced by water stress. At 0.01 μ M methyl-Hg, besides a decrease in chlorophyll concentration and partial stomatal closure, photosynthetic reactions may have been directly affected. Respiration rates were not influenced by exposure to heavy metals.  相似文献

Interior spruce (Picea glauca engelmannii complex) and black spruce (Picea mariana Mill.) cotyledonary somatic embryos were encapsulated in sodium alginate. Somatic embryo viability was retained, but germination occurred at a reduced frequency compared with the equivalent zygotic embryos. The addition of 0.5% (w/v) activated charcoal to the alginate capsule significantly enhanced root development and germination for somatic embryos but not for zygotic embryos. The possibility of developing an artiflcal endosperm was also investigated, by addition of Litvay (Litvay et al. 1981) nutrients with or without 90 mM sucrose to the alginate-charcoal capsule. This treatment significantly enhanced root development for all embryo categories with the exception of black spruce somatic embryos. The effects of soil temperature on the shoot phenology, carbohydrate dynamics, chlorophyll fluorescence and cold hardiness of 4-year-old Norway spruce seedlings ( Picea abies L. Karst.) were studied. The experiment was carried out under controlled conditions in the Joensuu dasotrons. Air conditions were similar but soil temperatures differed by treatments (9, 13, 18 and 21°C) during the second growing period in the dasotrons. The after-effects of the treatments were investigated during the third growing period following the treatments. Low soil temperature increased the starch content of needles and delayed the loss of starch at the end of the growing season. The photochemical efficiency ( F v/ F m) of the PSII of the current-year needles was reduced at the lowest soil temperature. The cold hardiness of needles correlated with the soluble sugar content. The differences in soil temperature had no effect on the timing of bud burst. No after-effects from the treatments were observed during the third growing period in the dasotrons.  相似文献
