When Mallory's trichrome stain is used, acinar nuclei of the Harderian gland of Rana esculenta display different affinities for the dye. Some of the orangiophilic nuclei show affinity for aniline blue (blue nuclei). In the Harderian gland of Rana esculenta their number and the intensity of staining with aniline blue may vary during the year. The affinity for aniline blue disappears following digestion of paraffin sections with RNAase, but not with DNAase or trypsin. Furthermore, in vitro incubation with [5, 6-3H]-Uridine shows a selective incorporation by the majority of blue nuclei. Therefore, the affinity for aniline blue is likely due to increased RNA synthesis. The increment of nuclear RNA shown by these methods is supported by the quantitative determination of total RNAs during the resumption (October) and enhancement (May) of secretory activity, when the percentage of blue nuclei of the acinar cells is at its highest levels of the year. The affinity of RNA-rich nuclei for aniline blue, while others are strictly orangiophil, is discussed on the basis of molecular structure of the dyes used in the staining mixture. Mallory's trichrome stain appears to be an useful tool for detecting changes in cell nuclear status.  相似文献

The Harderian gland in Rana esculenta has been studied during the annual cycle at the histological, histochemical and ultrastructural levels. The Harderian gland has an acinar structure and is the only orbital gland in anuran amphibia. It develops at the medial corner of the orbit from the conjunctival epithelium at the premetamorphic stage. In the adult the glandular secretion reaches a maximum during the months of July and August, drops in September and resumes slowly from October onwards. The secretion is seromucoid and the secretory granules are released into the acinar lumen, mainly by exocytosis. Porphyrins were not detected. No sexual dimorphism was observed in the glandular cells. The resumption of secretory activity in October and the enhancement of secretion in May are marked by the appearance of "blue nuclei" (Mallory stain) in a relatively high percentage of glandular cells. This unusual blue colour, using the Mallory stain (by which nuclei stain red), disappears after digestion of paraffin sections with RNAase, but not with DNAase and trypsin. The blue staining may, therefore, indicate an increased amount of nuclear RNA. The Harderian gland in the frog most probably serves to lubricate and moisten the eye in the absence of the lacrimal gland. However, the gland may also represent an immunoactive organ owing to the presence of numerous mast cells and plasma cells in the interacinar spaces.  相似文献

The Harderian gland of Pelophylax esculentus (previously: Rana esculenta) shows seasonal secretory activity changes. Specifically, the secretory activity reaches a maximum during the hottest months, i.e., July and August, drops in September and slowly increases from October onwards. Expressions of P-CaMKII, P-ERK1 and P-Akt1 correlate well with gland secretory activity; i.e., they peak immediately before the hottest part of the year (maximum secretory activity). When the gland activity declines, kinase expressions drop and remain low until February. Experiments of thermal manipulation indicate that high temperature induces the activation of CaMKII, ERK1 and Akt1, and at low temperatures, Akt1 expression decreases. Experiments of chemical castration indicate that the Harderian gland of Cyproterone acetate-treated frogs shows lower Akt1 activity as compared to controls, but the CaMKII and ERK1 activities remain unchanged. Furthermore, in a period of resumed gland activity (October–December) we observed the highest expression of PCNA, a mitotic marker. Immediately after the proliferative phase, we found the highest expression of caspase 3, an enzyme that plays a key role in apoptosis. In combination, the results suggest the following: 1) CaMKII, ERK1, and Akt1 modulate the annual secretory activity of the frog Harderian gland; 2) CaMKII and ERK1 activities are regulated by temperature, whereas both temperature and testosterone likely play a central role in Akt1 regulation; and 3) proliferation and apoptosis occur to restore and balance, respectively, an adequate cell number, which is essential to gland function.  相似文献

1. Indole metabolism and porphyrin content of the Harderian glands of the male Syrian hamster were measured as functions of drug-induced hypothyroidism and exposure to cold conditions. 2. Harderian gland N-acetyltransferase (NAT) activity was reduced from control levels by hypothyroidism induced by methimazole; exposure to cold had no effect on NAT activity. 3. Immunoreactive melatonin in the Harderian glands was unaffected by the state of thyroid secretion. However, immunoreactive melatonin content declined after 180 and 270 min, at 4 degrees C, suggesting that Harderian gland melatonin may be involved in thermoregulation. 4. Porphyrin content of the Harderian glands was not affected by either thyroid secretion or cold.  相似文献

The effects of gonadectomy and testosterone treatment on the fine structure of the Harderian gland in male and female green frogs were investigated in different periods of the year. Gonadectomy, carried out when the glands are in the lowest secretory phase (September), causes degenerative changes consisting of a reduction of the rough endoplasmic reticulum, the appearance of autolysosomes, and an increase of nuclear heterochromatin. These effects can be prevented by testosterone treatment. No castration effects are found during the recovery (November) and enhancement (April-May) phases of secretory activity. The results suggest that the frog Harderian gland's sensitivity to testosterone changes during the annual cycle. The androgen dependence of the Harderian gland is correlated with the presence of androgen receptors in both male and female forgs.  相似文献

Four combinations of photoperiods and temperatures have been tested on the reproduction and on the activity of the genital apparatus of the snail Helix aspersa. The results show some interaction between photoperiod and temperature and reveal a predominant effect of photoperiod that compensates for the negative effect of low temperatures. A combination of long day (LD, 18L:6D) and a temperature of 20 degrees C is the most favorable condition for egg-laying as well as for the effective functioning of the ovotestis and albumen gland. In contrast, during a short day (SD, 8L:16D), egg-laying is completely inhibited at 15 degrees C and partly inhibited at 20 degrees C; the differentiation of gametes can be observed in the ovotestis, but the mature oocytes are not released and thus they degenerate. In the albumen gland, an important synthetic activity is noticed, but cellular multiplication and the release of the secretory products decrease. These results suggest that short photoperiods induce inhibition of the (neuro)endocrine centers responsible for the control of both ovulation and egg-laying.  相似文献

This study of the morphology, histology, histochemistry, and ultrastructure of the Harderian gland in Geckos (Squamata, Gekkota) revealed previously unreported variation. The gecko Harderian gland is unlike that of other squamates in that each cell of the secretory epithelium has both lipid and protein secretory granules. Lipid secretion has not been reported previously for the squamate Harderian gland. The structure of the protein granules resembles that described for a scincomorph lizard (Podarcis, Lacertidae). Differences between representatives of the subfamilies Gekkoninae and Diplodactylinae suggest possible phylogenetic constraints in the structure or function of Harderian glands within gekkotan lineages. The structural relationship between the Harderian gland and the lacrimal duct supports previous suggestions of a possible functional link between the Harderian gland and the vomeronasal organ. Summary The presence and distribution of gastrin-, gastrin-releasing peptide-, neurotensin-and vasoactive intestinal polypeptide-like immunoreactivity in the Harderian gland ofRana esculenta were studied at different times of the annual cycle. Gastrin-releasing peptide, neurotensin and vasoactive intestinal polypeptide-like substances were found either in the glandular cells, or in the nerve fibers surrounding the glandular acini. Gastrin-like immunoreactivity was confined to the glandular cells. The immunoreactivity varied during the annual cycle, with the greatest concentration being noted during the recovery phase of glandular secretory activity.  相似文献

Analyses of the histology, histochemistry, and ultrastructre of the Harderian gland of Coluber viridiflavus prove the gland to be compound acinar and to produce a seromucous secretion. Acinar cells (type I) contain secretory granules that are composite, consisting ultrastructurally of three distinct parts that are sharply separated. They are similar to the “special secretory granules” described in the cells of the Harderian gland of the lizard Podarcis s. sicula. Some acini of the most anterior and posterior parts of the gland are mucous. Acinar cells (type II) of this type contain secretory granules that are Alcian blue/PAS positve. At the ultrastructural level, they appear homogeneous and of low density, characteristic of mucous secretions. These mucus-secreting anterior and posterior parts of the Harderian gland may by considered as regions of intial differentiation of the anterior and posterior lacrimal galnds.  相似文献

To analyze the role of environmental temperature and photoperiod in the regulation of the annual testicular cycle in Rana perezi, we performed experiments combining high (25 +/- 1 degrees C) or low (6 +/- 1 degrees C) temperature and different photoperiod regimens (18L:6D, 12L:12D, and 6L:18D (hours light:hours dark)) during three phases of the reproductive cycle: winter stage (December) and prebreeding (February) and postbreeding (May, June) periods. Low temperature and short photoperiod in winter induced the arrest of the maturation phase of spermatogenesis and the activation of primary spermatogonia proliferation and spermiohistogenesis. Rana perezi testis responded to long days stimulus in winter, even at low temperature, with induction of the maturation phase of the cycle. Exposure of male frogs to either high temperature or long photoperiod induced a decrease in testosterone levels in winter. During the prebreeding period, an increase in environmental temperature caused a reduction in testosterone, and a lengthening in photoperiod produced the opposite effect. Photoperiod had no effect on testosterone levels during the postbreeding period, but low temperature increased testosterone plasma levels. These results suggest that both temperature and photoperiod effects can vary seasonally, depending on the phase of the annual reproductive cycle in R. perezi.  相似文献

The aim of the present work was to study the sexual differences in secretory mechanisms and intracellular calcium ion dynamics in the Harderian gland of the golden hamster. In both sexes the Harderian gland consisted of small and large lobes. In the intact control male glands the secretory portions of both lobes showed wide lumina that contained secretory material and cytoplasmic fragments, suggestive of the occurrence of exocytosis and apocrine secretion. After perfusion with HEPES-buffered Ringer's solution containing 10 microM carbamylcholine (CCh), the glandular cells showed features of enhanced secretion and a rise in intracellular calcium concentration ([Ca2+]i). In the intact control female gland the lumina of most secretory portions in the large lobe contained porphyrin accretions, and exocytosis was the sole secretory mechanism. Stimulation of the large lobe with 10 microM CCh did not raise [Ca2+]i or cause enhanced secretion. The small lobe in females resembled the male gland in secretory functions, and CCh administration caused enhanced secretion and a rise in [Ca2+]i. Castration in males abolished apocrine secretion; exocytosis became the sole secretory mechanism, and stimulation of the glandular cells with CCh did not cause enhanced secretion or induce a rise in [Ca2+]i. To the contrary, in females, castration restored apocrine secretion and CCh administration caused enhanced secretion and a rise in [Ca2+]i. Castration did not affect the secretory mechanisms and the effect of CCh on the glandular cells in the small lobes of both male and female glands. The present study points to the possibility that sex hormones may control the functioning or expression of muscarinic receptors in the Harderian gland of the golden hamster.  相似文献

The authors performed a study of the mitotic activity and the nuclear/cytoplasmic (N/C) ratio during postnatal life of the lachrymal and Harderian glands of the rat. Based on the results, they concluded: (1) during the first days of postnatal life the development of lachrymal and Harderian glands was characterized by an intense mitotic activity and a low N/C ratio; (2) the period prior to eyelid disjunction was characterized by a diminished mitotic activity and a progressive and slow increase of the N/C ratio; (3) after eyelid disjunction, mitotic activity was reduced and an abrupt increase of the N/C ratio occurred, more evident in the Harderian gland; (4) during the final period of postnatal life studied mitotic activity was absent and the N/C ratio presented a higher, more constant level, which was always higher for the Harderian gland.  相似文献

The Harderian gland of the gecko, Tarentola mauritanica, was studied at the histological, histochemical, and ultrastructural levels. It is a nonlobate compound acinar gland surrounded by a thin capsule of connective tissue. Numerous connective tissue-type mast cells, ultrastructurally similar to those described in other higher vertebrates, were identified in the interstitial tissue between the acini. Pyramidal or columnar-shaped secretory glandular cells were observed in the acini. In the glandular cells, two types of structures could be distinguished on the basis of their high or low electron density. Lipid droplets were found in the cytoplasm of the Harderian gland of both sexes. Histochemical tests showed that the Harderian gland of the gecko is a seromucous gland. The secretion is essentially merocrine, although an apocrine type of secretion is sometimes observed.  相似文献

The effects of paraquat, the active ingredient of Gramoxone, were studied on frogs (Rana esculenta) kept at 4 degrees C or 20 degrees C, to establish its effects on the survival, the antioxidant enzyme system and the lipid peroxidation of the tissues (liver and lung). The lower temperature was found to increase the survival. The activities of the antioxidant enzymes were decreased or not influenced by the LD50 of paraquat at the lower temperature, whereas the LD100 often resulted in a significant enzymatic activity increase. It was an important finding that the lipid peroxidation of the lipid decreased in response to paraquat at 4 degrees C, but increased at 20 degrees C. The lipid peroxidation of the lung increased at both temperatures.  相似文献

The effects of pinealectomy on testicular activity and secretory activity of seminal vesicles were examined in the catfish Heteropneustes fossilis under various combinations of photoperiod and temperature during different periods of the annual reproductive cycle. Pinealectomy had no effect on gonadal activity during the preparatory, prespawning and spawning periods of the reproductive cycle. However, during the postspawning period, under long (LD 14:10) or short (LD 9:15) photoperiod at 25° C or at gradually increasing ambient temperature, pinealectomy accelerated testicular recrudescence and secretory activity of the seminal vesicles. Nevertheless. during this period the presence of the pineal facilitated the recrudescence of testes and seminal vesicles in catfish exposed to continuous light (LL), continuous darkness (DD) and 12 hL:l2 hD (LD) at 25° C. These findings suggest that the role of the pineal in catfish reproduction is variable and depends upon the photoperiod and temperature regimes to which the fish are exposed, as well as on the time of the year and the state of the reproductive cycle. The results also suggest that the effects of pinealectomy in catfish are mediated through an influence on the hypothalamo-hypophyseal gonadal axis.  相似文献

The ontogeny and regulation by isoproterenol of type II thyroxine 5'-deiodinase, N-acetyltransferase, and hydroxyindole-O-methyltransferase activities were studied in the rat Harderian gland. Both 5'-deiodinase and N-acetyltransferase activities exhibited maximal values at the first week of age. These activities gradually decreased till the fourth week. However, hydroxyindole-O-methyltransferase activity did not change during the period of time studied. Neither the different killing times (1600 or 0200 h) nor the photoperiod regimens (darkness or light exposure at night) modified the enzyme activities. On the other hand, isoproterenol, a beta-adrenergic agonist, could not to stimulate 5'-deiodinase at first week of life. Nevertheless, while basal 5'-deiodinase activity was diminishing during development, the enzyme was becoming sensitive to the action of isoproterenol. Thus, isoproterenol elicited increases in Harderian gland 5'-deiodinase activity in rats older than two weeks. However, both N-acetyltransferase and hydroxyindole-O-methyltransferase activities were not affected by isoproterenol treatment in either week studied.  相似文献

Exposure of female catfish, Heteropneustes fossilis to 30-day regimes of long photoperiod (16L), elevated temperature (28 +/- 2 degrees C), or a combination of both stimulated brain tyrosine hydroxylase (TH) activity significantly over that of control fish held in natural conditions in gonad resting (10.5L:13.5D, 10 +/- 2 degrees C) and preparatory (12.5L:11.5D,18 +/- 2 degrees C) phases. The response was high in the combination group in both phases. The increase in TH activity was higher in forebrain regions (telencephalon and hypothalamus) than medulla oblongata. Exposure of the fish to short photoperiod (8L:16D) and total darkness decreased the enzyme activity significantly in both resting and preparatory phases regardless of the temperature. The inhibition was high in fish held under total darkness. Gonadosomatic index (GSI) was significantly elevated in long photoperiod and high temperature groups, alone or in combination, and decreased significantly in short photoperiod (only in preparatory phase) and total darkness groups. Administration of the serotonin precursor 5-hydroxytryptophan (5-HTP; 5mg/100g body weight [BW], three daily intraperitoneal [i.p.] injections prior to sacrificing) stimulated TH activity in fish held under long and normal photoperiods in both phases. Three daily injections of the serotonin blocker parachlorophenylalanine (p-CPA; 10mg/100g BW) and melatonin (75 microg/100g BW) prior to sacrificing inhibited brain TH activity significantly in both phases. GSI was significantly stimulated by 5-HTP, and inhibited by both p-CPA and melatonin injections. Changes in TH activity and GSI can be correlated and explained on the basis of previous reports on changes in catecholamine activity that modulates gonadotropin secretion in the catfish. Further, the photoperiod and temperature-induced changes in TH activity may be modulated by alterations in serotonergic activity.  相似文献

Pulmonary surfactant is synthesised in alveolar type II cells and secreted into the lining of the lung in response to ventilation, temperature changes and autonomic neurotransmitters. Type II cells were isolated from the heterothermic marsupial, Sminthopsis crassicaudata. The neurotransmitters, isoproterenol and carbamylcholine chloride significantly increased phosphatidylcholine secretion at 37 degrees C (basal: 14.2%, isoproterenol: 20.1%, carbamylcholine: 17.0%). Temperature reduced the rate of secretion from dunnart type II cells (e.g. basal: 14.2% at 37 degrees C; 7.2% at 18 degrees C). However, the change in secretory rate between 37 degrees C and 18 degrees C was less than expected if due to temperature alone (Q10= 1.4). The surfactant secretory pathway is therefore modulated by factors other than and in addition to, temperature. The response of dunnart type II cells to the agonists remained the same at both temperatures. Basal secretion was higher in dunnart type II cells (14.2% in 4 h) than has been reported in rat type II cells (1.9% in 3 h) and consequently, the agonist-stimulated increases in secretion from dunnart type II cells (41% above basal in 4 h) were much lower than observed for rat type II cells (200% above basal in 1.5 h).  相似文献
