Cryogenic storage is considered to be the most convenient method to maintain phenotypic and genetic stability of organisms. A cryopreservation technique based on encapsulation-drying of in vitro-produced arbuscular mycorrhizal fungi has been developed at the Glomeromycota In Vitro Collection. In this study, we investigated fungal morphology (i.e., number and size of spores, number of branched absorbing structures (BAS), hyphal length, and number of anastomosis per hyphal length), activity of acid phosphatase and alkaline phosphatase in extraradical hyphae, and variation in amplified fragment length polymorphism (AFLP) profiles of in vitro-produced isolates of five Rhizophagus species maintained by cryopreservation for 6 months at ?130 °C and compared to the same isolates preserved at 27 °C. Isolates were stable after 6 months cryopreservation. Comparing isolates, the number of BAS increased significantly in one isolate, and hyphal length decreased significantly in another isolate. No other morphological variable was impacted by the mode of preservation. Phosphatase activities in extraradical hyphae and AFLP profiles were not influenced by cryopreservation. These findings indicate that cryopreservation at ?130 °C of encapsulated-dried and in vitro-produced Rhizophagus isolates (i.e., Rhizophagus irregularis, Rhizophagus fasciculatus, Rhizophagus diaphanous, and two undefined isolates) is a suitable alternative for their long-term preservation.  相似文献

The morphology and the mycotrophic habit of a new species ofRhizophagus in mycorrhizal association with the roots ofLitchi chinensis Sonn a tropical fruit tree, has been described. It belongs to the vesicular-arbuscular group of phycomycetous endophytes and has been namedRhizophagus litchii sp. nov.The endophyte could not be brought into culture in artificial media, the presence of living litchi roots was necessary for its growth and development.Root penetration of the endophyte was through the epidermal cells. A taxonomic review of the genus Copelatus Erichson (Coleoptera: Dytiscidae) in Korea is presented. In the present study, five species of Copelatus are recognized from Korea, including one unrecorded species, C. kammuriensis. We provide a key to the Korean species of Copelatus, diagnosis of one unrecorded species, habitus photographs, scanning electron micrographs of the elytron, and illustrations of the aedeagus.  相似文献

South Korean species of the genus Eusterinx Förster are reviewed here. Nine species of the genus Eusterinx are found to occur in South Korea, and five species, Eusterinx (E.) elongata sp. nov. , E. (Holomeristus) hirtis sp. nov. , E. (Ischyracis) fulvipes sp. nov. , E. (I.) petiolata sp. nov. and E. (I.) rufithorax sp. nov. , are described. Four more species of Eusterinx from subgenera Divinatrix, Eusterinx and Holomeristus are recorded from South Korea for the first time. Hitherto, unknown male of the Eastern Palaearctic species E. (Divinatrix) kurilensis Humala is described. A key to all known species of the genus Eusterinx occurring in South Korea is provided.  相似文献

A taxonomic study of the genera Porocallus Sharp and Rhomphocallus Assing in Korea is presented. The genus Rhomphocallus and two species, Rhomphocallus princeps (Sharp) and Rhomphocallus maruyamai Assing, are reported for the first time in Korea. Porocallus insignis Sharp is recorded for the first time in South Korea. Diagnoses, illustrations of habitus and line drawings of diagnostic characters are provided.  相似文献

Three species of the genus ScathophagaS. mellipes (Coquillet), S. scybalaria (Linnaeus) and S. stercoraria (Linnaeus) – are treated in this study. A taxonomic review of the genus Gonioctena in the Korean Peninsula is presented. Ten species belonging to three subgenera are recognized. Spores of two supposedly arbuscular mycorrhizal fungal species, new to the United Kingdom and recently described as Acaulospora alpina and Ambispora brasiliensis (Glomeromycota), were discovered in soil samples from moorland in upland Scotland. Soil and plant trap pot cultures were established, but attempts to establish these fungi in single-species pot cultures with Plantago lanceolata as host were unsuccessful. Nevertheless, based on a 1.5-kb DNA fragment spanning part of the small subunit rRNA gene, the internal transcribed spacer region and part of the large subunit rRNA gene, both these species could be detected directly in field-sampled roots, together with one uncultured species each of Scutellospora, Rhizophagus (former Glomus group Ab, or ‘Glomus intraradices clade’) and Acaulospora. Whereas A. alpina has characteristic morphological similarities to other species in its genus, A. brasiliensis morphologically has little in common with any other species in Ambispora. The molecular phylogeny, DNA barcoding and morphological evidence clearly place A. brasiliensis in the genus Acaulospora. We therefore rename the species, reported from Brazil and Scotland, as Acaulospora brasiliensis comb. nov., and discuss ecological aspects of the very different environments from which A. brasiliensis and A. alpina have been reported.  相似文献
