Soybean plants (Glycine max [L.] Merr. cv Williams), which were symbiotic with Bradyrhizobium japonicum, and which grew well upon reduced nitrogen supplied solely through N2 fixation processes, often exhibited excess accumulation of starch and sucrose and diminished soluble protein in their source leaves. Nitrate and ammonia, when supplied to the nodulated roots of N2-fixing plants, mediated a reduction of foliar starch accumulation and a corresponding increase in soluble protein in the source leaves. This provided an opportunity to examine the potential metabolic adjustments by which NO3 and NH4+ (N) sufficiency or deficiency exerted an influence upon soybean leaf starch synthesis. When compared with soybean plants supplied with N, elevated starch accumulation was focused in leaf palisade parenchyma tissue of N2-fixing plants. Foliar activities of starch synthesis pathway enzymes including fructose-1,6-bisphosphate phosphatase, phosphohexoisomerase, phosphoglucomutase (PGM), as well as adenosine diphosphate glucose pyrophosphorylase (in some leaves) exhibited highest activities in leaf extracts of N2-fixing plants when expressed on a leaf protein basis. This was interpreted to mean that there was an adaptation of these enzyme activities in the leaves of N2-fixing plants, and this contributed to an increase in starch accumulation. Another major causal factor associated with increased starch accumulation was the elevation in foliar levels of fructose-6-phosphate, glucose-6-phosphate, and glucose-1-phosphate (G1P), which had risen to chloroplast concentrations considerably in excess of the Km values for their respective target enzymes associated with starch synthesis, e.g. elevated G1P with respect to adenosine diphosphate glucose pyrophosphorylase (ADPG-PPiase) binding sites. The cofactor glucose-1,6-bisphosphate (G1,6BP) was found to be obligate for maximal PGM activity in soybean leaf extracts of N2-fixing as well as N-supplemented plants, and G1,6BP levels in N2-fixing plant leaves was twice that of levels in N-supplied treatments. However the concentration of chloroplastic G1,6BP in illuminated leaves was computed to be saturating with respect to PGM in both N2-fixing and N-supplemented plants. This suggested that the higher level of this cofactor in N2-fixing plant leaves did not confer any higher PGM activation and was not a factor in higher starch synthesis rates. Relative to plants supplied with NO3 and NH4+, the source leaf glycerate-3-phosphate (3-PGA) and orthophosphate (Pi) concentrations in leaves of N2-fixing plants were two to four times higher. Although Pi is a physiological competitive inhibitor of leaf chloroplast ADPG-PPiase, and hence, starch synthesis, elevated chloroplast 3-PGA levels in N2-fixing plant leaves apparently prevented interference of Pi with ADPG-PPiase catalysis and starch synthesis.  相似文献

The relative effects of water stress on growth parameters of subterranean clover (Trifolium subterraneum L. cv. Woogenellup) dependent on either N2 or 8 millimolar NH4NO3 for N were examined. Whole-plant carbon exchange rate (CER), acetylene reduction (AR), dry matter production, and Kjeldahl N accumulation were measured on uniform, intact swards of clover that were maintained under adequately watered conditions or were subjected to three cycles of water stress (leaf water potential ≤−30 bar) over an 18-day period. In the absence or presence of water stress, growth rate, net N accumulation rate, and total N concentration of plants dependent on N2 were 25 to 26, 45 to 50, and 20 to 21% less, respectively, than plants supplied with 8 millimolar NH4NO3. The water stress treatment produced less than a 50% decrease in CER regardless of plant N source, a 90% inhibition of AR in plants dependent on N2, and a 41% decline in dry matter production on both N sources. Water stress decreased reduced N accumulation 55% in N2-dependent plants and 50% in NH4NO3-dependent plants. Changes in growth and N accumulation caused a 10 to 11% decrease in total plant N concentration of water-stressed plants compared to adequately irrigated controls, but water stress decreased the N concentration of tissue synthesized during the 18-day treatment period in N2-grown plants more than in plants supplied 8 millimolar NH4NO3. Thus, the relative effect of water stress on growth under the two N regimes was similar, but N accumulation by N2-dependent clover was inhibited to a slightly greater extent (P ≤ 0.001) than in NH4NO3-dependent plants.  相似文献

The control of photosynthetic starch/sucrose formation in leaves of soybean (Glycine max L. Merr.) cultivars was studied in relation to stage of plant development, photosynthetic photoperiod, and nitrogen source. At each sampling, leaf tissue was analyzed for starch content, activities of sucrose-metabolizing enzymes, and labeling of starch and sucrose (by 14CO2 assimilation) in isolated cells. In three of the four varieties tested, nodulated plants had lower leaf starch levels and higher activities of sucrose phosphate synthetase (SPS), and isolated mesophyll cells incorporated more carbon (percentage of total 14CO2 fixed) into sucrose and less into starch as compared to nonnodulated (nitrate-dependent) plants. The variation among cultivars and nitrogen treatments observed in the activity of SPS in leaf extracts was positively correlated with labeling of sucrose in isolated cells (r = 0.81) and negatively correlated with whole leaf starch content (r = −0.66). The results suggested that increased demand for assimilates by nodulated roots may be accommodated by greater partitioning of carbon into sucrose in the mesophyll cells. We have also confirmed the earlier report (Chatterton, Silvius 1979 Plant Physiol 64: 749-753) that photoperiod affects partitioning of fixed carbon into starch. Within two days of transfer of nodulated soybean Ransom plants from a 14-hour to a 7-hour photoperiod, leaf starch accumulation rates doubled, and this effect was associated with increased labeling of starch and decreased labeling of sucrose in isolated cells. Concurrently, activities of SPS, sucrose synthase, and uridine diphosphatase in leaves were decreased.  相似文献

The effect of two N-forms (NH4 + and NO3 ) and NaCl on pattern of accumulation of some essential inorganic nutrients was examined in sunflower (Helianthus annuus L.) cv. Hisun 33. Eight-day-old plants of were subjected for 21 d to Hoagland's nutrient solution containing 8 mM N as NH4 + or NO3 ·, and salinized with and addition of NaCl to the growth medium had no significant effect on total leaf N. However, root N of NH4-supplied plants decreased significantly with increase in NaCl concentration, whereas that of NO3-supplied plants remained unaffected. There was no significant effect of NaCl on leaf or root P, but the NO3-supplied plants had significa concentration of leaf P than that of NH4-supplied plants at varying salt treatments. Salinity of the rooting med did not show any significant effect on Na+ concentrations of leaves or roots of plants subjected to two differen N. NH4-treated plants generally had greater concentrations of Cl in leaves and roots and lower K+ content in leaves than NO3-supplied plants. Ca2+ concentrations of leaves and roots and Mg2+ concentrations of leaves decreased in NH4-supplied plants due to NaCl, but they remained unaffected in NO3-treated plants.  相似文献

Soybeans grown with 2 millimolar NO3, which optimized apparent N2 fixation by Rhizobium symbionts, showed significantly different rates of apparent photosynthesis and C2H2 reduction during seedling development at two irradiances. Those physiological processes were lower for several weeks in plants grown at 1,500 microeinsteins per meter2 per second than in those exposed to 700 microeinsteins per meter2 per second. The irradiance-induced retardation was evident in short-term rates of apparent photosynthesis and N2 fixation, as well as in measures of dry matter and total N accumulation. In spite of their previously inhibited development, plants grown at 1,500 microeinsteins per meter2 per second were indistinguishable by day 28 from those exposed to 700 microeinsteins per meter2 per second in terms of whole-shoot CO2-exchange rate; by day 35 they were identical in terms of whole-plant C2H2-reduction rate. On day 38 there was no significant difference in dry weight or N content between treatments. Shifting plants between irradiance treatments on day 21 showed that the higher irradiance also had a short-term inhibitory effect on C2H2 reduction. The fact that 16 millimolar NO3 prevented the continuous exposure to 1,500 microeinsteins per meter2 per second from inhibiting apparent photosynthesis suggested that seedlings grown on 2 millimolar NO3 with Rhizobium were N-limited. Although rates of apparent photosynthesis were similar by day 28, the additional week required to produce equal rates of apparent N2 fixation between irradiance treatments showed that physiological adaptations of shoots, as well as photosynthesis per se, can affect root nodule activity.  相似文献

Prolonged inorganic nitrogen (NO3 +NH4 +) limitation of non-N2-fixing soybean plants affected leaflet photosynthesis rates, photosynthate accumulation rates and levels, and anaplerotic carbon metabolite levels. Leaflets of nitrogen-limited (N-Lim), 27–31-day-old plants displayed 15 to 23% lower photosynthesis rates than leaflets of nitrogen-sufficient (N-Suff) plants. In contrast, N-Lim plant leaflets displayed higher sucrose and starch levels and rates of accumulation, as well as higher levels of carbon metabolites associated with sucrose and starch synthesis, e. g., glycerate-3-phosphate and glucose phosphates, than N-Suff plant leaflets. Concurrently, levels of soluble protein, chlorophyll, and anaplerotic metabolites, e.g., malate and phosphoenolpyruvate, were lower in leaflets of N-Lim plants than N-Suff plants, suggesting that the enzymes of the anaplerotic carbon metabolite pathway were lower in activity in N-Lim plant leaflets. Malate net accumulation rates in the earliest part of the illumination period were lower in N-Lim than in N-Suff plant leaflets; however, by the midday period, malate accumulation rate in N-Lim plant leaflets exceeded that in leaflets of N-Suff plants. Further, soluble protein accumulation rates in leaflets of N-Suff and N-Lim plants were similar, and the rate of dark respiration, measured in the early part of the dark period, was higher in N-Lim plant leaflets than in N-Suff plant leaflets. It was concluded that during prolonged N-limitation, foliar metabolite conditions favored the channelling of a large proportion of the carbon assimilate into sucrose and starch, while assimilate flow through the anaplerotic pathway was diminished. However, in some daytime periods, there was a normal level of carbon assimilate channelled through the anaplerotic pathway for ultimate use in amino acid and protein synthesis.Abbreviations ADPG-PPiase ADPglucose pyrophosphorylase - Ce CO2 in the leaf photosynthesis measuring cuvette - Ci leaf internal CO2 during photosynthesis measurement - Chl chlorophyll - DHAP dihydroxyacetone phosphate - GAP glyceraldehyde-3-phosphate - Gsw stomatal conductance with units as mmol H2O m–2 s–1 - G1P glucose-1-phosphate - G6P glucose-6-phosphate - F6P fructose-6-phosphate - FBP fructose-1,6-bisphosphate - FBPase-pH 8.1 chloroplastic fructose-1,6-bisP (C-1) phosphatase (pH 8.1) - MAL malate - N inorganic nitrogen, i.e. NO3 +NH4 + (at levels and molar ratios indicated) - PE post-emergence - PEP phosphoenolpyruvate - PEPCase phosphoenolpyruvate carboxylase - PGA 3-phosphoglycerate - PYR pyruvate - PYR kinase pyruvate kinase - Pn net CO2 photoassimilation in leaves - PPFD photosynthetic photon flux density - PPRC pentose phosphate reductive cycle - RuBP ribulose-1,5-bisphosphate; rubisco-ribulose-1,5-bisphosphate carboxylase/oxygenase - SLW specific leaf mass - SPS sucrose-6-phosphate synthase - TCA cycle tricarboxylic acid cycle; triose-P-DAP+GAP  相似文献

An experiment was conducted to test the hypothesis that, when nitrogenase and nitrate reductase both contribute to the nitrogen nutrition of a nodulated legume, nitrogenase activity is inversely proportional to the rate of accumulation of organic nitrogen derived from the reduction of nitrate. Trifolium subterraneum L. plants, inoculated with Rhizobium trifolii and sown as small swards, were allowed to establish a closed canopy and steady rates of growth, dinitrogen fixation, and nitrogen accumulation. Swards were then supplied with nutrient solutions of 0, 0.5, 1.0, or 2.5 mm NO3 with a 29.69% enrichment of 15N and allowed to grow for a further 33 days. Harvests were made to measure dry weight, nitrogen accumulation, 15N accumulation, NO3 content and nitrogenase activity by acetylene reduction assay. Since the 15N of the plant organic matter could have been derived only from the NO3 of the nutrient solution, its rate of accumulation provided a measure of the rate of NO3 reduction. It was found that as this rate increased in response to external NO3 concentration the rate of nitrogenase activity decreased proportionately. It is concluded that the reduction of nitrate and the reduction of dinitrogen act in a complementary manner to supply a plant with organic nitrogen for growth.  相似文献

The comparative induction of nitrate reductase (NR) by ambient NO3 and NO2 as a function of influx, reduction (as NR was induced) and accumulation in detached leaves of 8-day-old barley (Hordeum valgare L.) seedlings was determined. The dynamic interaction of NO3 influx, reduction and accumulation on NR induction was shown. The activity of NR, as it was induced, influenced its further induction by affecting the internal concentration of NO3. As the ambient concentration of NO3 increased, the relative influences imposed by influx and reduction on NO3 accumulation changed with influx becoming a more predominant regulant. Significant levels of NO3 accumulated in NO2-fed leaves. When the leaves were supplied cycloheximide or tungstate along with NO2, about 60% more NO3 accumulated in the leaves than in the absence of the inhibitors. In NO3-supplied leaves NR induction was observed at an ambient concentration of as low as 0.02 mm. No NR induction occurred in leaves supplied with NO2 until the ambient NO2 concentration was 0.5 mm. In fact, NR induction from NO2 solutions was not seen until NO3 was detected in the leaves. The amount of NO3 accumulating in NO2-fed leaves induced similar levels of NR as did equivalent amounts of NO3 accumulating from NO3-fed leaves. In all cases the internal concentration of NO3, but not NO2, was highly correlated with the amount of NR induced. The evidence indicated that NO3 was a more likely inducer of NR than was NO2.  相似文献

The capacity for dissimilatory reduction of NO3 to N2 (N2O) and NH4+ was measured in 15NO3-amended marine sediment. Incubation with acetylene (7 × 10−3 atmospheres [normal]) caused accumulation of N2O in the sediment. The rate of N2O production equaled the rate of N2 production in samples without acetylene. Complete inhibition of the reduction of N2O to N2 suggests that the “acetylene blockage technique” is applicable to assays for denitrification in marine sediments. The capacity for reduction of NO3 by denitrification decreased rapidly with depth in the sediment, whereas the capacity for reduction of NO3 to NH4+ was significant also in deeper layers. The data suggested that the latter process may be equally as significant as denitrification in the turnover of NO3 in marine sediments.  相似文献

Ricinus communis L. plants were grown in nutrient solutions in which N was supplied as NO3 or NH4+, the solutions being maintained at pH 5.5. In NO3-fed plants excess nutrient anion over cation uptake was equivalent to net OH efflux, and the total charge from NO3 and SO42− reduction equated to the sum of organic anion accumulation plus net OH efflux. In NH4+-fed plants a large H+ efflux was recorded in close agreement with excess cation over anion uptake. This H+ efflux equated to the sum of net cation (NH4+ minus SO42−) assimilation plus organic anion accumulation. In vivo nitrate reductase assays revealed that the roots may have the capacity to reduce just under half of the total NO3 that is taken up and reduced in NO3-fed plants. Organic anion concentration in these plants was much higher in the shoots than in the roots. In NH4+-fed plants absorbed NH4+ was almost exclusively assimilated in the roots. These plants were considerably lower in organic anions than NO3-fed plants, but had equal concentrations in shoots and roots. Xylem and phloem saps were collected from plants exposed to both N sources and analyzed for all major contributing ionic and nitrogenous compounds. The results obtained were used to assist in interpreting the ion uptake, assimilation, and accumulation data in terms of shoot/root pH regulation and cycling of nutrients.  相似文献

Phosphorus stress effects on assimilation of nitrate   总被引:13,自引:3,他引:10       下载免费PDF全文
An experiment was conducted to investigate alterations in uptake and assimilation of NO3 by phosphorus-stressed plants. Young tobacco plants (Nicotiana tabacum [L.], cv NC 2326) growing in solution culture were deprived of an external phosphorus (P) supply for 12 days. On selected days, plants were exposed to 15NO3 during the 12 hour light period to determine changes in NO3 assimilation as the P deficiency progressed. Decreased whole-plant growth was evident after 3 days of P deprivation and became more pronounced with time, but root growth was unaffected until after day 6. Uptake of 15NO3 per gram root dry weight and translocation of absorbed 15NO3 out of the root were noticeably restricted in −P plants by day 3, and effects on both increased in severity with time. Whole-plant reduction of 15NO3 and 15N incorporation into insoluble reduced-N in the shoot decreased after day 3. Although the P limitation was associated with a substantial accumulation of amino acids in the shoot, there was no indication of excessive accumulation of soluble reduced-15N in the shoot during the 12 hour 15NO3 exposure periods. The results indicate that alterations in NO3 transport processes in the root system are the primary initial responses limiting synthesis of shoot protein in P-stressed plants. Elevated amino acid levels evidently are associated with enhanced degradation of protein rather than inhibition of concurrent protein synthesis.  相似文献

The effects of the ammonium (NH4+) and nitrate (NO3-) forms of nitrogen and NaCl on the growth, water relations and photosynthesis performance of sunflower (Helianthus annuus L.) were examined under glasshouse conditions. Eight-day-old plants of cv. Hisun 33 were subjected for 21 days to Hoagland's nutrient solution containing 8 mol m-3N as NH4+or NO3-, and salinised with 0, 60, or 120 mol m-3NaCl. Fresh weights of shoots and roots, and leaf area of NO3-supplied non-salinised plants were significantly greater than those of NH4+-supplied non-salinised plants. But addition of NaCl to the rooting medium of these plants had more inhibitory effect on the growth of NO3--supplied plants than on NH4+-supplied plants. Both leaf water and osmotic potentials of plants grown with NH4+were lower than those of plants given NO3-under both non-saline and saline conditions. Chlorophylls a and b concentrations were higher in plants grown with NH4+than N03--supplied plants at the lower two levels of salinisation. The rate of photosynthesis in plants was considerably higher in non-salinised plants grown with NO3-than with NH4+, but with increase in salinisation the photosynthesis rate decreased in NO3--supplied plants, but not in those given NH4+. The rate of transpiration was increased significantly by salinisation in NO3--supplied plants, but not consistently so in NH4+-supplied plants. The stomatal conductances were much higher in plants given NO3-than with NH4+when grown under non-saline conditions, but not when salinised. As a consequence, water-use efficiency in NO3--supplied control plants was better than in NH4+-supplied under non-saline conditions, but worse under saline conditions. The different forms of nitrogen and the addition of NaCl to the growing medium did not affect the relative intercellular concentrations of CO2 (Ci/Ca). Overall, the NH4+form of nitrogen inhibited the growth of sunflowers under non-saline conditions, but NO3-and NaCl interacted to inhibit growth more than did NH4+under saline conditions.  相似文献

The role of NO3 and NO2 in the induction of nitrite reductase (NiR) activity in detached leaves of 8-day-old barley (Hordeum vulgare L.) seedlings was investigated. Barley leaves contained 6 to 8 micromoles NO2/gram fresh weight × hour of endogenous NiR activity when grown in N-free solutions. Supply of both NO2 and NO3 induced the enzyme activity above the endogenous levels (5 and 10 times, respectively at 10 millimolar NO2 and NO3 over a 24 hour period). In NO3-supplied leaves, NiR induction occurred at an ambient NO3 concentration of as low as 0.05 millimolar; however, no NiR induction was found in leaves supplied with NO2 until the ambient NO2 concentration was 0.5 millimolar. Nitrate accumulated in NO2-fed leaves. The amount of NO3 accumulating in NO2-fed leaves induced similar levels of NiR as did equivalent amounts of NO3 accumulating in NO3-fed leaves. Induction of NiR in NO2-fed leaves was not seen until NO3 was detectable (30 nanomoles/gram fresh weight) in the leaves. The internal concentrations of NO3, irrespective of N source, were highly correlated with the levels of NiR induced. When the reduction of NO3 to NO2 was inhibited by WO42−, the induction of NiR was inhibited only partially. The results indicate that in barley leaves NiR is induced by NO3 directly, i.e. without being reduced to NO2, and that absorbed NO2 induces the enzyme activity indirectly after being oxidized to NO3 within the leaf.  相似文献

Leaf area, chlorophyll content, net CO2 photoassimilation, and the partitioning of fixed carbon between leaf sucrose and starch and soluble protein were examined in Glycine max (L) Merr. cv Williams grown under three different nitrogen regimes. One group (Nod+/+) was inoculated with Bradyrhizobium and watered daily with a nutrient solution containing 6 millimolar NH4NO3. A second set (Nod+/−) was inoculated and had N2 fixation as its sole source of nitrogen. A third group (Nod) was not inoculated and was watered daily with a nutrient solution containing 6 millimolar NH4NO3. The mean net micromole CO2 uptake per square decimeter per hour of the most recently matured source leaves was similar among the three groups of plants, being about 310. Mean leaf area of the source leaves, monitored for net photosynthesis was also similar. However, the mean milligram of chlorophyll per square decimeter of Nod+/− test leaves was about 50% lower than the other groups' leaves and indicated nitrogen deficiency. Thus, Nod+/− utilized their chlorophyll more efficiently for photosynthetic CO2 uptake than the plants of the other treatments. The ratio of foliar carbohydrate:protein content was high in Nod+/− but low in the plants from the other two treatments. This inverse relationship between foliar protein and carbohydrate content suggests that more fixed carbon is diverted to the synthesis of protein when nitrogen availability is high. It was also found that Nod+/− sequestered more storage protein in their paraveinal mesophyll than plants of the other treatments. This study indicates that when inorganic nitrogen regimes are used to control photosynthate partitioning, then both leaf carbohydrate and leaf protein must be considered as end products of carbon assimilate allocation.  相似文献

Tomato plants (Lycopersicon esculentum L. var. Ailsa Craig) were grown in water culture in nutrient solution in a series of 10 increasing levels of nitrate nutrition. Using whole plant data derived from analytical and yield data of individual plant parts, the fate of anion charge arising from increased NO3 assimilation was followed in its distribution between organic anion accumulation in the plant and OH efflux into the nutrient solution as calculated by excess anion over cation uptake. With increasing NO3 nutrition the bulk of the anion charge appeared as organic anion accumulation in the plants. OH efflux at a maximum accounted for only 20% of the anion charge shift. The major organic anion accumulated in response to nitrate assimilation was malate. The increase in organic anion accumulation was paralleled by an increase in cation concentration (K+, Ca2+, Mg2+, Na+). Total inorganic anion levels (NO3, SO42−, H2PO4, Cl) were relatively constant. The effect of increasing NO3 nutrition in stimulating organic anion accumulation was much more pronounced in the tops than in the roots.  相似文献

The effects of N source (6 mm nitrogen as NO3 or urea) and tungstate (0, 100, 200, 300, and 400 μm Na2 WO4) on nitrate metabolism, nodulation, and growth of soybean (Glycine max [L.] Merr.) plants were evaluated. Nitrate reductase activity and, to a lesser extent, NO3 content of leaf tissue decreased with the addition of tungstate to the nutrient growth medium. Concomitantly, nodule mass and acetylene reduction activity of NO3-grown plants increased with addition of tungstate to the nutrient solution. In contrast, nodule mass and acetylene reduction activity of urea-grown plants decreased with increased nutrient tungstate levels. The acetylene reduction activity of nodulated roots of NO3-grown plants was less than 10% of the activity of nodulated roots of urea-grown plants when no tungstate was added. At 300 and 400 μm tungstate levels, acetylene reduction activity of nodulated roots of NO3-grown plants exceeded the activity of comparable urea-grown plants.  相似文献

Five rices (Oryza sativa L.) differing in final grain size were studied at the midmilky stage to determine if any factor could be identified which might limit rate of starch accumulation. Only UDP glucose pyrophosphorylase activity increased with increasing grain size. Detached rice panicles incubated in liquid medium containing 1% sucrose and 0.1% glutamine, in addition to minerals and vitamins, produced grains similar to those on intact plants. Sucrose level (0–1.5%) in the medium determined the extent of dry matter and starch accumulation and influenced physiological development of the ripening grains. Chemical and enzymic composition of the grain were similar to previously reported levels in grains of intact panicles analysed at regular intervals after anthesis. Addition of 3-P glycerate or K+ to the medium did not improve dry matter accumulation in the developing grain.  相似文献

Until recently, denitrification was thought to be the only significant pathway for N2 formation and, in turn, the removal of nitrogen in aquatic sediments. The discovery of anaerobic ammonium oxidation in the laboratory suggested that alternative metabolisms might be present in the environment. By using a combination of 15N-labeled NH4+, NO3, and NO2 (and 14N analogues), production of 29N2 and 30N2 was measured in anaerobic sediment slurries from six sites along the Thames estuary. The production of 29N2 in the presence of 15NH4+ and either 14NO3 or 14NO2 confirmed the presence of anaerobic ammonium oxidation, with the stoichiometry of the reaction indicating that the oxidation was coupled to the reduction of NO2. Anaerobic ammonium oxidation proceeded at equal rates via either the direct reduction of NO2 or indirect reduction, following the initial reduction of NO3. Whether NO2 was directly present at 800 μM or it accumulated at 3 to 20 μM (from the reduction of NO3), the rate of 29N2 formation was not affected, which suggested that anaerobic ammonium oxidation was saturated at low concentrations of NO2. We observed a shift in the significance of anaerobic ammonium oxidation to N2 formation relative to denitrification, from 8% near the head of the estuary to less than 1% at the coast. The relative importance of anaerobic ammonium oxidation was positively correlated (P < 0.05) with sediment organic content. This report of anaerobic ammonium oxidation in organically enriched estuarine sediments, though in contrast to a recent report on continental shelf sediments, confirms the presence of this novel metabolism in another aquatic sediment system.  相似文献

Since NO3 availability in the rooting medium seriously limits symbiotic N2 fixation by soybean (Glycine max [L.] Merr.), studies were initiated to select nodulation mutants which were more tolerant to NO3 and were adapted to the Midwest area of the United States. Three independent mutants were selected in the M2 generation from ethyl methanesulfonate or N-nitroso-N-methylurea mutagenized Williams seed. All three mutants (designated NOD1-3, NOD2-4, and NOD3-7) were more extensively nodulated (427 to 770 nodules plant−1) than the Williams parent (187 nodules plant−1) under zero-N growth conditions. This provided evidence that the mutational event(s) affected autoregulatory control of nodulation. Moreover, all three mutants were partially tolerant to NO3; each retained greater acetylene reduction activity when grown hydroponically with 15 millimolar NO3 than did Williams at 1.5 millimolar NO3. The NO3 tolerance did not appear to be related to an altered ability to take up or metabolize NO3, based on solution NO3 depletion and on in vivo nitrate reductase assays. Enhanced nodulation appeared to be controlled by the host plant, being consistent across four Bradyrhizobium japonicum strains tested. In general, the mutant lines produced less dry weight than the control, with root dry weights being more affected than shoot dry weights. The nodulation trait has been stable through the M5 generation in all three mutants.  相似文献
