Photosynthesis, primary productivity, N content, and N2 fixation were determined as a function of applied NH4+ in peas (Pisum sativum L. cv. Alaska) which were inoculated or not inoculated with Rhizobium leguminosarum. Cabon dioxide exchange rate (CER) increased 10-fold, total N content 7-fold, and total dry weight 3-fold in 26-day-old uninoculated plants as applied NH4+ was increased from 0 to 16 millimolar. In inoculated plants of the same age CER and dry weight were maximal at 2 millimolar NH4+, and total N content increased between 0 and 2 millimolar NH4+ but did not change significantly with higher NH4+ applications. Per cent N content of uninoculated plants was significantly lower than that of inoculated plants except at the highest NH4+ concentration (16 millimolar). Symbiotic N2 fixation by inoculated plants was maximal in peas grown with 2 millimolar NH4+; and apparent relative efficiency of N2 fixation, calculated from C2H2 reduction and H2 evolution, was maximal in the 2 to 4 millimolar NH4+ concentration range. The capacity to fix N2 through the Rhizobium-legume symbiosis significantly enhanced the rate and efficiency of photosynthesis and plant N content when NH4+ concentration in the nutrient solution was below 8 millimolar. Above 8 millimolar NH4+ concentration uninoculated plants had greater CER, N content, and dry weight.  相似文献

Carbon and nitrogen limitations on symbiotically grown soybean seedlings (Glycine max [L.] Merr.) were assessed by providing 0.0, 1.0, or 8.0 millimolar NH4NO3 and 320 or 1,000 microliters CO2/liter for 22 days after planting. Maximum development of the Rhizobium-soybean symbiosis, as determined by acetylene reduction, was measured in the presence of 1.0 millimolar NH4NO3 under both levels of CO2. Raising NH4NO3 from 0.0 to 8.0 millimolar under 320 microliters CO2/liter increased plant dry weight by 251% and Kjeldahl N content by 287% at 22 days after planting. Increasing NH4NO3 from 1.0 to 8.0 millimolar under 320 microliters CO2/liter increased total dry weight and Kjeldahl N by 100 and 168%, respectively, on day 22. Raising CO2 from 320 to 1,000 microliters CO2/liter during the same period had no significant effect on Kjeldahl N content of plants grown with 0.0 or 1.0 millimolar NH4NO3. The maximum CO2 treatment effects were observed in plants supplied with 8.0 millimolar NH4NO3, where dry weight and Kjeldahl N content were increased 64% and 20%, respectively. An increase in shoot CO2-exchange rate associated with the CO2-enrichment treatment was reflected in a significant increase in leaf dry weight and starch content for plants grown with 1,000 microliters CO2/liter under all combined N treatments. These data show directly that seedling growth in symbiotically grown soybeans was limited primarily by N availability. The failure of the CO2-enrichment treatment to increase total plant N significantly in Rhizobium-dependent plants indicates that root nodule development and functioning in such plants was not limited by photosynthate production.  相似文献

Either NO3 (16 millimolar) or NH4+ (1 millimolar) completely inhibited infection and nodulation of white clover seedlings (Trifoliin repens) inoculated with Rhizobium trifolii. The binding of R. trifolii to root hairs and the immunologically detectable levels of the plant lectin, trifoliin, on the root hair surface had parallel declining slopes as the concentration of either NO3 or NH4+ was increased in the rooting medium. This supports the role of trifoliin in binding R. trifolii to clover root hairs. Agglutination of R. trifolii by trifoliin from seeds was not inhibited by these levels of NO3 or NH4+. The results suggest that these fixed N ions may play important roles in regulating an early recognition process in the Rhizobium-clover symbiosis, namely the accumulation of high numbers of infective R. trifolii cells on clover root hairs.  相似文献

Similar NH4+ and NO3?.uptake kinetic patterns were observed in Neoagardhiella baileyi (Harvey ex Kiitzing) Wyinne & Taylor and Gracilaria foliifera (Forssk?l) Borgesen. NO3? was taken up in a rate-sturating fashion described by the Michaelis-Menten equation. NH4+ uptake was multicomponent: a saturable component was accompanied by a diffusive or a high K component showing no evidence of saturation (at ≤50 μM [NH4+]). Nitrogen starved plantsi(C/N atom ratios > ca. 10) showed higher transient rates of NH4+ uptake at a given concentration than plants not N-Iimited. Only plants with high N content exhibited diel changes inNH4+ uptake rates, and showed transient rates of NH4+ accumulation which did not greatly exceed the capacity to incorporate N in steady-state growth. NH4+ was preferred over NO3?even in plants preconditioned on NO3?as the sole N. source, NO3? uptake was suppressed at 5μM [NH4+], but simultaneous uptake occurred at unsurpressed rates at lower concentrations. Potential for N accumulation was greater via NH4+uptake than via NO3?uptake. Changing capacity for NH4+ uptake with N content appears to be a mechanism whereby excessive accumulation of N was avoided by N-.satiated plants but a large accumulation was possible for N-depleted plants.  相似文献

The negative effects of water stress on rice can be alleviated by NH4+ nutrition. However, the effects of mixed nitrogen (N) nutrition (NO3? + NH4+) on resistance to water stress are still not well known. To investigate the response of rice growth to water stress and its relationship with photosynthetic characteristics, a hydroponic experiment supplying different N forms was conducted. Compared with NO3? nutrition, mixed‐N and NH4+ nutrition greatly alleviated the reduction of leaf area, chlorophyll content, and photosynthesis under water stress, whilst subsequently maintaining higher biomass. In contrast, water stress inhibited the root‐shoot ratios in NH4+‐ and mixed‐N‐supplied plants, indicating reduced root growth and higher photosynthate availability to shoots. The following key observations were made: (1) a similar stomatal limitation and low proportion of activated Rubisco were observed among the three different N nutrition regimes; (2) increased mesophyll conductance in NH4+‐ and mixed‐N‐supplied plants simultaneously stimulated leaf photosynthesis and improved the water use efficiency and (3), the maximum carboxylation rate and actual photochemical efficiency of photosystem II in NH4+‐ and mixed‐N‐supplied plants were significantly higher than that in NO3?‐supplied plants, thus resulting in higher photochemical efficiency under water stress. In conclusion, mixed‐N and NH4+ nutrition may be used to develop strategies for improved water stress resistance and stimulated biomass production under conditions of osmotic stress and possibly drought.  相似文献

Soybeans (Glycine max L. Merr. cv Tracy and Ransom) were grown under N2-dependent or NO3-supplied conditions, and the partitioning of photosynthate and dry matter was characterized. Although no treatment effects on photosynthetic rates were observed, NO3-supplied plants in both cultivars had lower starch accumulation rates than N2-dependent plants. Leaf extracts of NO3-supplied plants had higher activities of sucrose phosphate synthase (SPS) and cytoplasmic fructose-1,6-bisphosphatase (FBPase) than N2-dependent plants. The variation in starch accumulation was correlated negatively with the activity of SPS, but not the activity of FBPase, UDP-glucose pyrophosphorylase, or ADP-glucose pyrophosphorylase. These results suggested that starch accumulation is biochemically controlled, in part, by the activity of SPS. Leaf starch content at the beginning of the photoperiod was lower in NO3-supplied plants than N2-dependent plants in both cultivars which suggested that net starch utilization as well as accumulation was affected by N source.

Total dry matter accumulation and dry matter distribution was affected by N source in both cultivars, but the cultivars differed in how dry matter was partitioned between the shoot and root as well as within the shoot. The activity of SPS was correlated positively with total dry matter accumulation which suggested that SPS activity is related to plant growth rate. The results suggested that photosynthate partitioning is an important but not an exclusive factor which determines whole plant dry matter distribution.

Additions of methionine sulfoximine (MSX), an inhibitor of glutamine synthetase (GS), result in an increase in NH3 in seedling leaves of C3 (wheat [Triticum aestivum cv. Kolibri] and barley [Hordeum vulgare var Perth]) and C4 (corn [Zea mays W6A × W182E] and sorghum [Sorghum Vulgare var MK300]) plants. NH3 accumulation is higher in C3 (about 17.8 micromoles per gram fresh weight per hour) than in C4 (about 4.7 micromoles) leaves. Under ideal conditions, when photosynthesis is not yet inhibited by the accumulation of NH3, the rate of NH3 accumulation is about 16% of the apparent rate of photosynthesis. A maximum accumulation of NH3 was elicited by 2.5 millimolar MSX and was essentially independent of the addition of NO3 during either the growth or experimental period. When O2 levels in the air were reduced to 2%, MSX resulted in some accumulation of NH3 (6.0 micromoles per gram fresh weight per hour). At these levels of NH3, there was no significant inhibition of rates of CO2 fixation. There was also a minor, but significant, accumulation of NH3 in corn roots treated with MSX. Inhibitors of photorespiration (isonicotinic hydrazide, 70 millimolar; 2-pyridylhydroxymethanesulfonic acid, 20 millimolar) or transaminase reactions (aminooxyacetate, 1 millimolar) inhibited the accumulation of NH3 in both C3 and C4 leaves. These results support the hypothesis that GS is important in the assimilation of NH3 in leaves and that the glycine-serine conversion is a major source of that NH3.  相似文献

Optimization of nitrogen (N) use by grasses is a central issue of the current work. The effects of different N concentrations (0, 0.25, 0.5, 1.0, 2.5, and 5.0 mM NH4NO3) on growth of Brachypodium distachyon were assessed on controlled hydroponic culture. Maximal growth (132% of control) was obtained at 0.5 mM NH4NO3, critical N level, and was maintained at higher N concentrations. The highest N level (5.0 mM) has a similar effect on growth as 0.5 mM NH4NO3. It has no significant effects on water status, and total and reduced N contents in shoots while, increased those in roots, compared to plants receiving 0.5 mM NH4NO3. The high N availability, however, increased nitrate contents in shoots and roots by 3- and 20-folds, respectively, compared to those of plant receiving 0.5 mM NH4NO3. In addition, high N availability reduced the nitrogen use efficiency (NUE) by 18% compared to that of plant receiving only 0.5 NH4NO3. In view of B. distachyon productivity and environmental concerns, it is concluded that the critical level of N application should be 0.5 mM NH4NO3 and the excess fertilization led to a high nitrate accumulation.  相似文献

Nodulated white clover plants (Trifolium repensL.) of a Norwegian ecotype from Pasvik (70°N) were grown in flowing solution culture. Root temperature was 17°C until 51d after sowing, when it was lowered decrementally over 5d to 7°C in four of the eight plant culture units. After a further 24h, mineral N was supplied automatically at 20μMNH4NO3in three culture units at each root temperature (7 and 17°C) over 17d. The remaining two units provided control plants solely dependent on N2fixation at 7 and 17°C.The supply of NH4NO3greatly reduced the nodule biomass per plant at 17°C over 17d compared with control plants, but had little effect at 7°C. The nodule decline at 17°C accompanied an acute and progressive decrease in specific rate of N2fixation, from 9mmolN d-1g-1nodule d.wt on day 0 to zero by day 10. Whilst initial rates of N2fixation were lower at 7°C, the mineral N-induced decrease in fixation rates was also less severe than at 17°C and specific fixation rates recovered after reaching a minimum on day 11. N2fixation accounted for 36% of the total uptake of N by +min.N plants during the treatment period at 7°C as opposed to only 13% at 17°C. The total N2fixed at 7°C was 86% of that fixed at 17°C, although the specific growth rate (d.wt) at 7°C was only 55% of that at 17°C. Addition of NH4NO3at 7°C had little effect on the gross amount of N2fixed subsequently. In contrast, total N2fixation by +min.N plants at 17°C was only 24% of that fixed by the corresponding controls. The possible mechanisms by which mineral N affects N2fixation are discussed.  相似文献

Growth, chemical composition, and nitrate reductase activity (NRA) of hydroponically cultured Rumex crispus, R. palustris, R. acetosa, and R. maritimus were studied in relation to form (NH4 +, NO3 -, or both) and level of N supply (4 mM N, and zero-N following a period of 4mM N). A distinct preference for either NH4 + or NO3 - could not be established. All species were characterized by a very efficient uptake and utilization of N, irrespective of N source, as evident from high concentrations of organic N in the tissues and concurrent excessive accumulations of free NO3 - and free NH4 +. Especially the accumulation of free NH4 + was unusually large. Generally, relative growth rate (RGR) was highest with a combination of NH4 + and NO3 -. Compared to mixed N supply, RGR of NO3 -- and NH4 +-grown plants declined on average 3% and 9%, respectively. Lowest RGR with NH4 + supply probably resulted from direct or indirect toxicity effects associated with high NH4 + and/or low Ca2+ contents of tissues. NRA in NO3 - and NH4NO3 plants was very similar with maxima in the leaves of ca 40 μmol NO2 - g-1 DW h-1. ‘Basal’ NRA levels in shoot tissues of NH4 + plants appeared relatively high with maxima in the leaves of ca 20 μmol NO2 - g-1 DW h-1. Carboxylate to organic N ratios, (C-A)/Norg, on a whole plant basis varied from 0.2 in NH4 + plants to 0.9 in NO3 - plants. After withdrawal of N, all accumulated NO3 - and NH4 + was assimilated into organic N and the organic N redistributed on a large scale. NRA rapidly declined to similar low levels, irrespective of previous N source. Shoot/root ratios of -N plants were 50–80% lower than those from +N plants. In comparison with +N, RGR of -N plants did not decline to a large extent, decreasing by only 15% in -NH4 + plants due to very high initial organic-N contents. N-deprived plants all exhibited an excess cation over anion uptake (net proton efflux), and whole-plant (C-A)/Norg ratios increased to values around unity. Possible difficulties in interpreting the (C-A)/Norg ratio and NRA of plants in their natural habitats are briefly discussed.  相似文献

Toxic Effect of Cadmium on Rice as Affected by Nitrogen Fertilizer Form   总被引:1,自引:0,他引:1  
A nutrient solution experiment was conducted to determine the influence of N forms on growth, oxidative stress, and Cd and N uptake in rice plants. The treatments were consisted of two Cd levels (0 and 1 μmol) and three N forms (NH4)2SO4, NH4NO3 and Ca(NO3)2. The results indicated that without Cd addition in the culture solution, the N forms had no significant effect on all measured parameters, including plant growth, photosynthetic traits, malondialdehyde (MDA) concentration, superoxide dismutase (SOD) activity, and Cd and N concentration, while Cd addition in the medium resulted in significant differences in measured parameters among the three forms of N fertilizers. The least inhibition of growth was noted in (NH4)2SO4-fed plants, and the largest in Ca(NO3)2-fed plants, when plants were exposed to Cd stress. The highest photosynthetic rate and chlorophyll content was also recorded in (NH4)2SO4-fed plants. Addition of Cd caused a remarkable increase in SOD activity and MDA content in plants, and the extent of increase varied with N form, with (NH4)2SO4-fed plants being smallest. In comparison with the control plants, the N concentration in roots and shoots was not significantly affected in (NH4)2SO4-fed plants, but significant decrease in root N concentration was found for the NH4NO3 and Ca(NO3)2-fed plants under Cd stress. Moreover, the significant differences were also noted among the three N forms in both root and shoot Cd concentrations, with (NH4)2SO4-fed plants being the lowest. The results indicated that the toxic effect of Cd on rice varied with the form of N fertilizer.  相似文献

Partitioning and utilization of assimilated C and N were compared in nonnodulated, NO3-fed and nodulated, N2-fed plants of white lupin (Lupinus albus L.). The NO3 regime used (5 millimolar NO3) promoted closely similar rates of growth and N assimilation as in the symbiotic plants. Over 90% of the N absorbed by the NO3-fed plants was judged to be reduced in roots. Empirically based models of C and N flow demonstrated that patterns of incorporation of C and N into dry matter and exchange of C and N among plant parts were essentially similar in the two forms of nutrition. NO3-fed and N2-fed plants transported similar types and proportions of organic solutes in xylem and phloem. Withdrawal of NO3 supply from NO3-fed plants led to substantial changes in assimilate partitioning, particularly in increased translocation of N from shoot to root. Nodulated plants showed a lower (57%) conversion of C or net photosynthate to dry matter than did NO3-fed plants (69%), and their stems were only half as effective as those of NO3-fed plants in xylem to phloem transfer of N supplied from the root. Below-ground parts of symbiotic plants consumed a larger share (58%) of the plants' net photosynthate than did NO3-fed roots (50%), thus reflecting a higher CO2 loss per unit of N assimilated (10.2 milligrams C/milligram N) by the nodulated root than by the root of the NO3-fed plant (8.1 milligrams C/milligram N). Theoretical considerations indicated that the greater CO2 output of the nodulated root involved a slightly greater expenditure for N2 than for NO3 assimilation, a small extra cost due to growth and maintenance of nodule tissue, and a considerably greater nonassimilatory component of respiration in root tissue of the symbiotic plant than in the root of the NO3-fed plant.  相似文献

An hydroponic experiment with a simulated water stress induced by PEG (6000) was conducted in a greenhouse to study the effects of nitrate (NO3 ), ammonium (NH4 +) and the mixture of NO3 and NH4 +, on water stress tolerance of rice seedlings. Rice (Shanyou 63) was grown under non- or simulated water stress condition (10% (w/v) PEG, MW6000) with the 3 different N forms during 4 weeks. Under non-stressed condition no difference was observed among the N treatments. Under simulated water stress, seedlings grown on N-NO3 were stunted. Addition of PEG did not affect rice seedling growth in the treatment of only NH4 + supply but slightly inhibited the rice seedling growth in the treatment of mixed supply of NO3 and NH4 +. Simulated water stress, when only N-NH4 + was present, did not affect leaf area and photosynthesis rate, however, both parameters decreased significantly in the NO3 containing solutions. Under water stress, Rubisco content in newly expanded leaves significantly increased in the sole NH4 + supplied plants as compared to that in plants of the other two N treatments. Under water stress, the ratio of carboxylation efficiency to Rubisco content was, respectively, decreased by 13 and 23% in NH4 + and NO3 treatments, respectively. The influence of nutrient nitrate level (0-20 millimolar) on the effects of NO2 (0-0.5 parts per million) on nodulation and in vivo acetylene reduction activity of the roots and on growth and nitrate and Kjeldahl N concentration in shoots was studied in bean (Phaseolus vulgaris L. cv Kinghorn Wax) plants. Exposing 8-day old seedlings for 6 hours each day, for 15 days, to 0.02 to 0.5 parts per million NO2 decreased total nodule weight at 0 and 1 millimolar nitrate, and nitrogenase (acetylene reduction) activity at all concentrations of nitrate. The pollutant had little effect on root fresh or dry weights. Shoot growth was inhibited by NO2. The NO2 exposure increased nitrate concentration in roots only at 20 millimolar nutrient nitrate. Exposure to NO2 markedly increased Kjeldahl N concentration in roots but generally decreased that in shoots. The experiments demonstrated that nutrient N level and NO2 concentration act jointly in affecting nodulation and N fixing capability, plant growth and composition, and root/shoot relationships of bean plants.  相似文献

Abstract. Wild radish plants deprived of, and continuously supplied with solution NO?3 for 7 d following 3 weeks growth at high NO?3 supply were compared in terms of changes in dry weight, leaf area, photosynthesis and the partitioning of carbon and nitrogen (NH2-N and NO?3-N) among individual organs. Initial levels of NO?3-N accounted for 25% of total plant N. Following termination of NO?3 supply, whole plant dry weight growth was not significantly reduced for 3 d, during which time plant NH2-N concentration declined by about 25% relative to NO?3-supplied plants, and endogenous NO?3-N content was reduced to nearly zero. Older leaves lost NO?3 and NH2-N, and roots and young leaves gained NH2-N in response to N stress. Relative growth rate declined due both to decreased net assimilation rate and a decrease in leaf area ratio. A rapid increase in specific leaf weight was indicative of a greater sensitivity to N stress of leaf expansion compared to carbon gain. In response to N stress, photosynthesis per unit leaf area was more severely inhibited in older leaves, whereas weight-based rates were equally inhibited among all leaf ages. Net photosynthesis was strongly correlated with leaf NH2-N concentration, and the relationship was not significantly different for leaves of NO3?-supplied compared to NO?3-deprived plants. Simulations of the time course of NO?3 depletion for plants of various NH2-N and NO?3 compositions and relative growth rates indicated that environmental conditions may influence the importance of NO?3 accumulation as a buffer against fluctuations in the N supply to demand ratio.  相似文献

This work aimed to study the regulation of K+/Na+ homeostasis and the physiological responses of salt-treated sorghum plants [Sorghum bicolor (L.) Moench] grown with different inorganic nitrogen (N) sources. Four days after sowing (DAS), the plants were transferred to complete nutrient solutions containing 0.75 mM K+ and 5 mM N, supplied as either NO3 ? or NH4 +. Twelve DAS, the plants were subjected to salt stress with 75 mM NaCl, which was applied in two doses of 37.5 mM. The plants were harvested on the third and seventh days after the exposure to NaCl. Under the salt stress conditions, the reduction of K+ concentrations in the shoot and roots was higher in the culture with NO3 ? than with NH4 +. However, the more conspicuous effect of N was on the Na+ accumulation, which was severely limited in the presence of NH4 +. This ionic regulation had a positive influence on the K+/Na+ ratio and the selective absorption and transport of K+ in the plants grown with NH4 +. Under control and salt stress conditions, higher accumulation of free amino acids and soluble proteins was promoted in NH4 + grown roots than NO3 ? grown roots at both harvesting time, whereas higher accumulation of soluble sugars was observed only at 7 days of salt stress exposure. Unlike the NH4 + grown plants, the gas exchanges of the NO3 ? grown plants were reduced after 7 days of salt stress. These results suggest that external NH4 + may limit Na+ accumulation in sorghum, which could contribute to improving its physiological and metabolic responses to salt stress.  相似文献

The response of non-nodulated cowpea (Vigna unguiculata (L.) Walp. cv Caloona) to a wide range of NO3 levels in the rooting medium was studied 40 days after sowing by in vitro assays of plant organs for NO3 reductase (EC 1.6.6.1) and analyses of root bleeding (xylem) sap for nitrogenous solutes. Plants fed 1, 5, 10, 20, and 40 millimolar NO3 showed, respectively, 64, 92, 94, and 91% of their total reductase activity in shoots and 34, 30, 66, 62, and 58% of the total N of their xylem sap as NO3. These data, and the absence in the plants of significant pools of stored NO3, indicated that shoots were major organs of NO3 assimilation, especially at levels of NO3 (10 to 40 millimolar) that maintained plant growth at near maximum rates. Partitioning and utilization of C and N were studied in nodulated, minus NO3 plants and non-nodulated plants fed 10 or 20 millimolar NO3, the levels of NO3 which gave rates of growth and N assimilation closest to those of the symbiotic plants. The conversion of the C of net photosynthate to dry matter was similar in nodulated plants (67%) and NO3-grown plants (64%), but greater proportions of photosynthate were translocated to below ground parts of nodulated plants (37%) than of NO3-fed plants (23 to 26%). Greater photosynthate consumption by nodulated roots was associated with proportionately greater root growth and respiration and 2-fold greater export of C in xylem than in the NO3-fed plants. Theoretical considerations suggest that the elevated CO2 output of nodulated roots was due not only to CO2 loss associated with nodule function, but also to a much greater nonassimilatory component of respiration in the supporting root of the nodulated plant compared to roots of the NO3-fed plants. The effect of exogenous NH4+ on NO3 uptake and in vivo NO3 reductase activity (NRA) in roots of Phaseolus vulgaris L. cv Witte Krombek was studied before, during, and after the apparent induction of root NRA and NO3 uptake. Pretreatment with NH4Cl (0.15-50 millimolar) affected neither the time pattern nor the steady state rate of NO3 uptake.

When NH4+ was given at the start of NO3 nutrition, the time pattern of NO3 uptake was the same as in plants receiving no NH4+. After 6 hours, however, the NO3 uptake rate (NUR) and root NRA were inhibited by NH4+ to a maximum of 45% and 60%, respectively.

The response of the NUR of NO3-induced plants depended on the NH4Cl concentration. Below 1 millimolar NH4+, the NUR declined immediately and some restoration occurred in the second hour. In the third hour, the NUR became constant. In contrast, NH4+ at 2 millimolar and above caused a rapid and transient stimulation of NO3 uptake, followed again by a decrease in the first, a recovery in the second, and a steady state in the third hour. Maximal inhibition of steady state NUR was 50%. With NO3-induced plants, root NRA responded less and more slowly to NH4+ than did NUR.

Methionine sulfoximine and azaserine, inhibitors of glutamine synthetase and glutamate synthase, respectively, relieved the NH4+ inhibition of the NUR of NO3-induced plants. We conclude that repression of the NUR by NH4+ depends on NH4+ assimilation. The repression by NH4+ was least at the lowest and highest NH4+ levels tested (0.04 and 25 millimolar).

Infection by the fungal endophyte Acremonium coenophialum affected the accumulation of inorganic and organic N in leaf blades and leaf sheaths of KY 31 tall fescue (Festuca arundinacea Schreb.) grown under greenhouse conditions. Total soluble amino acid concentrations were increased in either the blade or sheath of the leaf from infected plants. A number of amino acids were significantly increased in the sheath, but only asparagine increased in the blade. Infection resulted in higher sheath NH4+ concentrations, whereas NO3 concentrations decreased in both leaf parts. The effects on amino acid, NO3, and NH4+ concentrations were dependent upon the level of N fertilization and were usually apparent only at the high rate (10 millimolar) of application. Administration of 14CO2 to the leaf blades increased the accumulation of 14C in their amino acid fraction but not in the sheaths of infected plants. This may indicate that infection increased amino acid synthesis in the blade but that translocation to the sheath, which is the site of fungal colonization, was not affected. Glutamine synthetase activity was greater in leaf blades of infected plants at high and low N rates of fertilization, but nitrate reductase activity was not affected in either part of the leaf. Increased activities of glutamine synthetase together with the other observed changes in N accumulation and metabolism in endophyte-infected tall fescue suggest that NH4+ reassimilation could also be affected in the leaf blade.  相似文献
