Localizing genes that contribute to drought avoidance in a quantitative way should enable the exploitation of these genes in breeding through marker-assisted selection, and may lead to the discovery of gene identity and function. Between 110 and 176 F6 recombinant inbred lines from a mapping population derived from a cross of upland rice varieties Bala and Azucena have been evaluated for indicators of drought avoidance in sites in the Philippines and West Africa over two dry seasons. A molecular map with 102 RFLP, 34 AFLP and six microsatellite markers has been used to map (by composite interval mapping) quantitative trait loci (QTLs) for the visual scores of leaf rolling and leaf drying and leaf relative water content. QTLs were mapped for each site and across sites. A total of 17 regions were identified which contained QTLs with a LOD score greater than 3.2. For leaf rolling, Bala was the parent contributing the majority of positive alleles whilst for the other traits, Bala and Azucena contributed more evenly. Six of the 17 regions influenced more than one trait, explaining the phenotypic correlations between traits that were observed. Three QTLs appeared to be specific to the Philippines experiments. One QTL had opposing effects in the Philippines and West Africa. QTLs for relative water content were detected on chromosome 8, congruent with an osmotic adjustment QTL identified in another population. Only three of the QTLs identified here have not been reliably identified in the two other populations that have been screened for drought avoidance. By using several populations assessed for drought avoidance in different sites, the distribution and utility of QTLs for drought avoidance in rice is being elucidated.  相似文献

利用籼稻窄叶青8号(ZYQ8)和粳稻京系17(JX17)衍生的加倍单倍体(DH)群体127个株系,2002年在杭州采用田间断水法栽培,在水分胁迫下,对叶片的卷叶、相对含水量和电导率3个性状进行了评价和QTL分析。结果表明,3个性状在DH群体中均存在双向超亲分离,接近正态分布,受数量性状基因的控制;检测到影响这些性状的6个QTL,其中卷叶3个(qLR—1,qLR—5和qLR—11)、相对含水量2个(qRWC—1和qRWC—6和电导率1个(qERC—6)。旱胁迫时,目测卷叶方便易行,适于对大批品种或资源筛选,对抗旱栽培稻品种的筛选和利用具有一定的指导意义。  相似文献

To investigate the genetic basis of drought tolerance in soybean ( Glycine max L. Merr.) a recombinant inbred population with 184 F2:7:11 lines developed from a cross between Kefeng1 (drought tolerant) and Nannong1138-2 (drought sensitive) were tested under water-stressed and well-watered conditions in field and greenhouse trials. Traits measured included leaf wilting coefficient, excised leaf water loss and relative water content as indicators of plant water status and seed yield. A total of 40 quantitative trait loci (QTLs) were identified: 17 for leaf water status traits under drought stress and 23 for seed yield under well-watered and drought-stressed conditions in both field and greenhouse trials. Two seed yield QTLs were detected under both well-watered and drought-stressed conditions in the field on molecular linkage group H and D1b, while two seed yield QTLs on molecular linkage group C2 were found under greenhouse conditions. Several QTLs for traits associated with plant water status were identified in both field and greenhouse trials, including two leaf wilting coefficient QTLs on molecular linkage group A2 and one excised leaf water loss QTL on molecular linkage group H. Phenotypic correlations of traits suggested several QTLs had pleiotropic or location-linked associations. These results will help to elucidate the genetic basis of drought tolerance in soybean, and could be incorporated into a marker-assisted selection breeding program to develop high-yielding soybean cultivars with improved tolerance to drought stress.  相似文献

A recombinant inbred population developed from a cross between high-yielding lowland rice (Oryza sativa L.) subspecies indica cv. IR64 and upland tropical rice subspecies japonica cv. Cabacu was used to identify quantitative trait loci (QTLs) for grain yield (GY) and component traits under reproductive-stage drought stress. One hundred fifty-four lines were grown in field trials in Indonesia under aerobic conditions by giving surface irrigation to field capacity every 4 days. Water stress was imposed for a period of 15 days during pre-flowering by withholding irrigation at 65 days after seeding. Leaf rolling was scored at the end of the stress period and eight agronomic traits were evaluated after recovery. The population was also evaluated for root pulling force, and a total of 201 single nucleotide polymorphism markers were used to construct the molecular genetic linkage map and QTL mapping. A QTL for GY under drought stress was identified in a region close to the sd1 locus on chromosome 1. QTL meta-analysis across diverse populations showed that this QTL was conserved across genetic backgrounds and co-localized with QTLs for leaf rolling and osmotic adjustment (OA). A QTL for percent seed set and grains per panicle under drought stress was identified on chromosome 8 in the same region as a QTL for OA previously identified in three different populations.  相似文献

Drought stress is a major limitation to rice (Oryza sativa L.) yields and its stability, especially in rainfed conditions. Developing rice cultivars with inherent capacity to withstand drought stress would improve rainfed rice production. Mapping quantitative trait loci (QTLs) linked to drought resistance traits will help to develop rice cultivars suitable for water-limited environments through molecular marker-assisted selection (MAS) strategy. However, QTL mapping is usually carried out by genotyping large number of progenies, which is labour-intensive, time-consuming and cost-ineffective. Bulk segregant analysis (BSA) serves as an affordable strategy for mapping large effect QTLs by genotyping only the extreme phenotypes instead of the entire mapping population. We have previously mapped a QTL linked to leaf rolling and leaf drying in recombinant inbred (RI) lines derived from two locally adapted indica rice ecotypes viz., IR20/Nootripathu using BSA. Fine mapping the QTL will facilitate its application in MAS. BSA was done by bulking DNA of 10 drought-resistant and 12 drought-sensitive RI lines. Out of 343 rice microsatellites markers genotyped, RM8085 co-segregated among the RI lines constituting the respective bulks. RM8085 was mapped in the middle of the QTL region on chromosome 1 previously identified in these RI lines thus reducing the QTL interval from 7.9 to 3.8 cM. Further, the study showed that the region, RM212–RM302–RM8085–RM3825 on chromosome 1, harbours large effect QTLs for drought-resistance traits across several genetic backgrounds in rice. Thus, the QTL may be useful for drought resistance improvement in rice through MAS and map-based cloning.  相似文献

Soil drying causes leaf rolling in rice, but the relationship between leaf rolling and drought tolerance has historically confounded selection of drought‐tolerant genotypes. In this study on tropical japonica and aus diversity panels (170–220 genotypes), the degree of leaf rolling under drought was more affected by leaf morphology than by stomatal conductance, leaf water status, or maintenance of shoot biomass and grain yield. A range of canopy temperature and leaf rolling (measured as change in normalized difference vegetation index [ΔNDVI]) combinations were observed among aus genotypes, indicating that some genotypes continued transpiration while rolled. Association mapping indicated colocation of genomic regions for leaf rolling score and ΔNDVI under drought with previously reported leaf rolling genes and gene networks related to leaf anatomy. The relatively subtle variation across these large diversity panels may explain the lack of agreement of this study with earlier reports that used small numbers of genotypes that were highly divergent in hydraulic traits driving leaf rolling differences. This study highlights the large range of physiological responses to drought among rice genotypes and emphasizes that drought response processes should be understood in detail before incorporating them into a varietal selection programme.  相似文献

A population of recombinant inbred rice lines from a cross between the upland japonica cultivar Azucena and the upland indica cultivar Bala was evaluated in a series of upland field experiments. Water stress was imposed during the reproductive stage by managed irrigation during the dry season, while control treatments were maintained in aerobic, well-irrigated conditions. Water deficit resulted in a yield reduction of 17 to 50%. The genetic correlation between stress and control yields was quite high when stress was mild, and the heritability of yield was similar in stress and control treatments across both years of this study. Genetic correlations between secondary traits such as leaf rolling and drying and yield under stress varied from high (leaf drying) to insignificant (leaf rolling). Lines with superior yield tended to have fewer panicles and larger grain size than the high-yielding parent, Bala, even though the panicle number was positively correlated with yield and the thousand-grain weight was not associated with yield for the population as a whole. Analysis of quantitative trait loci (QTLs) for yield and yield components allowed the identification of 31 regions associated with growth or yield components. Superior alleles came from either parent. Several of the regions identified had also been reported for root mass at depth or maximum root length in this population in other studies made under controlled environments, and for leaf drying (LD) in field studies. However, the direction of the effect of QTLs was not consistent, which indicates that there was not necessarily a causal relationship between these secondary traits and performance. Rice is one of the most important food crop drastically affected by drought in lowland rice ecosystem. Dissecting out the traits of importance and genomic regions influencing the response of drought tolerance and yield traits on grain yield will aid the breeders to know the genetic mechanism of drought tolerance of rice leads to the development of drought tolerant varieties. Grain yield and its components on drought situation of recombinant inbred population (IR 58821/IR 52561) were investigated under lowland managed stress situation in 2003 and 2004 by given importance to the relative water content. Water deficit resulted in significant effect on phenology and grain yield. Best lines were selected for further varietal development programme. Variability studies showed the traits viz., days to 70% relative water content, leaf rolling, leaf drying, harvest index, biomass yield and grain yield offer high scope for improvement for drought tolerance by way of simple selection technique. Correlation and path analysis indicated that, to harness high yielding combined with drought tolerance breeders should give selection pressure on relative water content, panicle length, grains per panicle, harvest index, biomass yield, root/shoot ratio and root length in positive direction, and low scores of leaf rolling, leaf drying and drought recovery rate. Analysis of quantitative trait loci for drought tolerance, yield and its components allowed the identification of 38 regions associated with both drought tolerant and yield traits. Out of these, 18 were closely linked with DNA markers could be used for marker assisted selection in breeding for drought tolerance in rice. Pleiotropism and G × E effects interaction were noticed in some of the traits. Parent IR 58821 contributed favorable alleles for the entire drought related and most of the yield component traits. Identification of traits of importance and their nature of relationship by morphological and molecular level under lowland condition will be useful to improve drought tolerance of rice.  相似文献

Abscisic acid (ABA) concentration is a quantitatively inherited trait which plays a pivotal role in the response of plants to drought stress. A recent study identified 17 quantitative trait loci (QTLs) controlling bulk-leaf ABA concentration (L-ABA) in a maize (Zea mays L.) population of 80 F4 random families tested for two years under droughted field conditions. Sixteen of the QTL regions influencing L-ABA also harboured QTLs for one or more of the following traits: stomatal conductance, a drought sensitivity index, leaf temperature, leaf relative water content, anthesis-silking interval, and grain yield. The analysis of the effects of each QTL region on the investigated traits indicated that L-ABA mainly represented an indicator of the level of drought stress experienced by the plant at the time of sampling because an increase in L-ABA was most commonly associated with a decrease in both stomatal conductance and grain yield as well as an increase in leaf temperature. Opposite results were observed at one QTL region on chromosome 7 near the RFLP locus asg8. Drought is a major abiotic stress limiting rice production and yield stability in rainfed ecosystems. Identifying quantitative trait loci (QTL) for rice yield and yield components under water limited environments will help to develop drought resilient cultivars using marker assisted breeding (MAB) strategy. A total of 232 recombinant inbred lines of IR62266/Norungan were used to map QTLs for plant phenology and production traits under rainfed condition in target population of environments. A total of 79 QTLs for plant phenology and production traits with phenotypic variation ranging from 4.4 to 72.8% were detected under non-stress and drought stress conditions across two locations. Consistent QTLs for phenology and production traits were detected across experiments and water regimes. The QTL region, RM204-RM197-RM217 on chromosome 6 was linked to days to 50% flowering and grain yield per plant under both rainfed and irrigated conditions. The same genomic region, RM585-RM204-RM197 was also linked to harvest index under rainfed condition with positive alleles from Norungan, a local landrace. QTLs for plant production and drought resistance traits co-located near RM585-RM204-RM197-RM217 region on chromosome 6 in several rice genotypes. Thus with further fine mapping, this region may be useful as a candidate QTL for MAB, map-based cloning of genes and functional genomics studies for rainfed rice improvement.  相似文献

Drought stress is the major constraint to rice (Oryza sativa L.) production and yield stability in rainfed ecosystems. Identifying genomic regions contributing to drought resistance will help to develop rice cultivars suitable for rainfed regions through marker-assisted breeding. Quantitative trait loci (QTLs) linked to leaf epicuticular wax, physio-morphological and plant production traits under water stress and irrigated conditions were mapped in a doubled haploid (DH) line population from the cross CT9993-5-10-1-M/IR62266-42-6-2. The DH lines were subjected to water stress during anthesis. The DH lines showed significant variation for epicuticular wax (EW), physio-morphological and plant production traits under stress and irrigated conditions. A total of 19 QTLs were identified for the various traits under drought stress and irrigated conditions in the field, which individually explained 9.6%–65.6% of the phenotypic variation. A region EM15_10-ME8_4-R1394A-G2132 on chromosome 8 was identified for leaf EW and rate of water loss i.e., time taken to reach 70% RWC from excised leaves in rice lines subjected to drought stress. A large effect QTL (65.6%) was detected on chromosome 2 for harvest index under stress. QTLs identified for EW, rate of water loss from excised leaves and harvest index under stress in this study co-located with QTLs linked to shoot and root-related drought resistance traits in these rice lines and might be useful for rainfed rice improvement.  相似文献

选用协青早B//东乡野生稻/协青早B回交重组系及其亲本共79份水稻材料,研究了水稻发芽期、苗期、孕穗期、成熟期的种子萌发抗旱指数等31个指标与东乡野生稻回交重组系抗旱性的关系.结果表明:15% PEG-6000溶液处理的相对发芽势、20% PEG-6000溶液处理的种子萌发抗旱指数和相对发芽势及水分胁迫后最长根长、苗高、根鲜质量、根干质量、根系相对含水量、萎蔫率、叶片可溶性糖含量、叶片脯氨酸含量、叶片丙二醛含量、叶片相对含水量、卷叶级别、株高、单株分蘖数、单株有效穗数、穗实粒数、着粒密度、结实率、千粒重等21个指标的相对值与抗旱系数或抗旱指数的相关性显著.通过逐步回归分析筛选出9个抗旱性鉴定指标:20% PEG-6000溶液处理的相对发芽势及水分胁迫后根干质量、根系相对含水量、叶片可溶性糖含量、叶片脯氨酸含量、叶片丙二醛含量、单株有效穗、结实率和千粒重的相对值.根据这些指标和偏相关系数,建立了不同时期的抗旱性评价(D值)方程和评价体系,可对东乡野生稻回交重组系抗旱性进行较好的评价.  相似文献

为探究叶片水势(LWP)相关基因在水稻(Oryza sativa)抗旱中的作用及其遗传机制, 以热研2号(Nekken2)和华占(HZ)为亲本以及构建的120个重组自交系(RILs)群体为实验材料, 对水稻分蘖期叶片水势进行检测, 并利用前期基于高通量测序构建的分子遗传连锁图谱进行数量性状基因座(QTL)分析。结果表明, 共检测到5个与水稻分蘖期叶片水势相关的QTLs, 分别位于第2、3、4、11和12号染色体上, LOD值均达2.5以上, 其中位于4号染色体物理距离24 066 261- 30 847 136 bp内QTL的LOD值高达5.15。对这些QTL区间内与水势相关的候选基因进行定量分析, 发现LOC_Os02g56630LOC_Os02g57720LOC_Os02g57580LOC_Os04g43730LOC_Os04g46490LOC_Os04g44570LOC_Os04g44060这7个基因在双亲间表达量差异显著。位于4号染色体QTL区间内LOC_Os04g46490基因的表达在两亲本间存在显著差异。对基因LOC_Os04g46490进行测序分析, 发现该基因在两亲本间共存在6处差异, 从而导致氨基酸序列的改变。通过QTL挖掘及相关基因表达分析, 发现这些基因与水稻叶片水势调控相关, 可能间接影响水稻的抗旱性。检测到的QTL位点对水势相关基因精细定位和克隆具有重要参考价值, 有助于进一步理解水稻叶片水势的遗传基础, 并为培育耐旱水稻新品种提供有利的基因资源。  相似文献

亚铁毒是潜育性水稻土中限制水稻产量的主要因子。利用龙杂8503/IR64的F2和等价的F3群体,在营养液中培养来定位耐亚铁毒的QTLs。通过构建101SSR标记的遗传连锁图谱来确定耐亚铁毒QTLs的位置和特性。借助叶片棕色斑点指数、株高和最大根长3个性状,利用营养液在水稻苗期来评价F2单株、F3群体和亲本龙杂8503、IR64,共检测到叶片棕色斑点指数、株高和最大根长的QTLs20个,分布在水稻的10条染色体上,表明这些性状受多基因控制。控制叶片棕色斑点指数的QTLs分别定位在第1染色体的RM315-RM212、第2染色体的RM6-RM240和第4染色体的RM252-RM451之间。与前人的研究结果比较发现:1)位于第4染色体RM252-RM451之间的控制叶片棕色斑点指数的QTL与水稻功能图谱上控制叶绿素含量减少的QTL的位置一致。另一个位于第1染色体的RM315-RM212之间的控制叶片棕色斑点指数的QTL与水稻功能图谱上位于C178-R2635之间控制叶绿素含量的QTL连锁。2)位于第2染色体RM6-RM240之间的第3个控制叶片棕色斑点指数的QTL与位于RZ58-CD0686的控制钾吸收的QTL连锁。  相似文献

Drought tolerance is one of the most important but complex traits of crops. We looked for quantitative trait loci (QTLs) that affect drought tolerance in maize. Two maize inbreds and their advanced lines were evaluated for drought-related traits. A genetic linkage map developed using RFLP markers was used to identify QTLs associated with drought-related traits. Twenty-two QTLs were detected, with a minimum of one and a maximum of nine for drought-related traits. A single-QTL was detected for sugar concentration accounting for about 52.2% of the phenotypic variation on chromosome 6. A single-QTL was also identified for each of the traits root density, root dry weight, total biomass, relative water content, and leaf abscisic acid content, on chromosomes 1 and 7, contributing to 24, 0.2, 0.4, 7, and 19% of the phenotypic variance, respectively. Three QTLs were identified for grain yield on chromosomes 1, 5, and 9, explaining 75% of the observed phenotypic variability, whereas four QTLs were detected for osmotic potential on chromosomes 1, 3, and 9, together accounting for 50% of the phenotypic variance. Nine QTLs were detected for leaf surface area on chromosomes 3 and 9, with various degrees of phenotypic variance, ranging from 25.8 to 42.2%. Four major clusters of QTLs were identified on chromosomes 1, 3, 7, and 9. A QTL for yield on chromosome 1 was found co-locating with the QTLs for root traits, total biomass, and osmotic potential in a region of about 15 cM. A cluster of QTLs for leaf surface area were coincident with a QTL for osmotic potential on chromosome 3. The QTLs for leaf area also clustered on chromosome 9, whereas QTLs for leaf abscisic acid content and relative water content coincided on chromosome 7, 10 cM apart. Co-location of QTLs for different traits indicates potential pleiotropism or tight linkage, which may be useful for indirect selection in maize improvement for drought tolerance.  相似文献

Genetic relations among the contents of Rubisco, soluble protein and total leaf nitrogen (N) in leaves of rice (Oryza sativa L.) were studied by quantitative trait loci (QTL) analysis with a population of backcross inbred lines (BILs) of japonica Nipponbarexindica Kasalath. The ratio of Rubisco to total leaf N in leaves is the main target in improving photosynthetic N-use efficiency in plants. QTLs controlling Rubisco content were not detected near QTLs for total leaf N content. These results indicate that contents of Rubisco and total leaf N are controlled by different genetics. QTLs that controlled the ratio of Rubisco to total leaf N (CORNs) were detected. These results suggest that some mechanism(s) may be involved in determining this ratio, while the contents of Rubisco and total leaf N are controlled in other ways. In elite BILs, the ratios of Rubisco to total leaf N were higher than those of both parents. These results suggest a good possibility of improving N-use efficiency by CORNs in cultivated rice. A QTL controlling Rubisco content was mapped near a QTL for soluble protein content on chromosome 8 at 5 d after heading and on chromosome 9 at 25 d. In each chromosome region, the peaks of both QTLs overlapped accurately, giving a high possibility of pleiotropic effects by the same genes. Different QTLs controlling soluble protein or Rubisco were detected from those detected at 5 d or 25 d after heading. This suggests that these traits are genetically controlled depending on the growth stages of leaves.  相似文献

植物激素ABA参与不同的生理过程,尤其是在种子发育和非生物逆境的适应都需要ABA的调控.以水稻珍汕97和旱稻IRAT109为亲本的重组自交系群体为材料,分别调查种子发芽和苗期对ABA的敏感程度.种子发芽阶段以ABA处理下的相对发芽势(Relative germination vigor,RGV)和相对发芽率(Relative germination rate,RGR)为指标,苗期以ABA喷施处理下的卷叶程度(Leaf rolling scores by ABA spaying,LRS)和含ABA水培条件下的卷叶程度(Leaf rolling scores by ABA culturing,LRC)为指标.性状相关分析表明发芽阶段的相对发芽势与苗期卷叶程度呈显著正相关.用复合区间作图法和混合线性模型对ABA敏感性QTL定位和上位性效应分析.两种软件检测到的主效QTL位点大致相同.共检测到5个单位点QTL和6对上位性QTL与发芽阶段的ABA敏感性有关;8个单位点QTL和5对上位性QTL与水稻苗期对ABA的敏感性有关;在苗期,两种ABA处理条件下共检测到两个共同的QTL;仅一个共同的QTL同时控制发芽阶段和苗期对ABA的敏感性.这些研究结果说明,水稻对ABA的敏感性同时受单位点的多基因和上位性基因控制;而且控制种子萌发阶段发芽势和苗期对ABA敏感性的遗传基础有很大的不同.  相似文献

Drought stress has long been a major constraint in maintaining yield stability of soybean (Glycine max (L.) Merr.) in rainfed ecosystems. The identification of consistent quantitative trait loci (QTL) involving seed yield per plant (YP) and drought susceptibility index (DSI) in a population across different environments would therefore be important in molecular marker-assisted breeding of soybean cultivars suitable for rainfed regions. The YP of a recombinant line population of 184 F2:7:11 lines from a cross of Kefengl and Nannong1138-2 was studied under water-stressed (WS) and well-watered (WW) conditions in field (F) and greenhouse (G) trials, and DSI for yield was calculated in two trials. Nineteen QTLs associated with YP-WS and YP-WW, and 10 QTLs associated with DSI, were identi- fied. Comparison of these QTL locations with previous findings showed that the majority of these regions control one or more traits re- lated to yield and other agronomic traits. One QTL on molecular linkage group (MLG) K for YP-F, and two QTLs on MLG C2 for YP-G, remained constant across different water regimes. The regions on MLG C2 for YP-WW-F and MLG H for YP-WS-F had a pleiotropic effect on DSI-F, and MLG A1 for YP-WS-G had a pleiotropic effect on DSI-G. The identification of consistent QTLs for YP and DSI across different environments will significantly improve the efficiency of selecting for drought tolerance in soybean.  相似文献

Plant photosynthetic traits such as net photosynthetic rate (Pn), stomata conductance (gs), transpiration rate (Tr), and intercellular CO2 concentration (Ci), are known to relate to drought tolerance in plants, but the genetic basis of these traits remains largely uncharacterized because of the difficulty in phenotyping physiological traits in a large mapping population. In this study, a set of 55 overlapping introgression lines (ILs) in the Teqing (indica) background were used to genetically dissect several morph-physiological traits and their relationship with grain yield under water stress and non-stress conditions. These traits included specific leaf weight (SLW), chlorophyll content (CC), leaf stomata frequency (SF), Pn, gs, Tr, and Ci. A total of 40 QTLs affecting the measured traits were identified and mapped to 21 genomic regions in the rice genome. Clustered QTLs affecting Pn, gs, Tr, and Ci in the same genomic regions suggest common genetic bases for the physiological traits. Low or no phenotypic correlations between leaf morphological traits and photosynthetic traits and between morph-physiological traits and grain yield (GY) appeared to be due to inconsistence in QTL effect for clustered QTLs, unlinked QTLs affecting different traits, and to possible epistasis that could not be adequately addressed in this study. Our results indicate that improving drought tolerant (DT) of rice by selecting any single secondary traits is not expected to be effective and the identified QTLs for GY and related morph-physiological traits should be carefully confirmed before to be used for improving DT in rice by MAS.  相似文献
