The random amplified polymorphic DNA (RAPD) molecular marker technique was used to determine the sex of Calamus simplicifolius C. F. Wei In the present study, DNA samples were extracted individually from 10 male and 10 female plants. After a total of 1 040 decamer primers had been tested, an approximate 500-bp male-specific DNA fragment was generated with the S 1443 primer. It is feasible to identify sex at the early stages of plant life, which is beneficial for improving breeding programs of this dioecious species. In addition, we have obtained a proper RAPD protocol that is useful for other species of rattan.  相似文献

Responses of the photosynthetic activity of Phaeodactylum tricornutum (Bacillariophyta) to organic carbon glycerol were investigated. The growth rate, photosynthetic pigments, 77 K fluorescence spectra, and chloroplast ultrastructure of P. tricornutum were examined under photoautotrophic, mixotrophic, and photoheterotrophic conditions. The results showed that the specific growth rate was the fastest under mixotrophic conditions. The cell photosynthetic pigment content and values of Chl a/Chl c were reduced under mixotrophic and photoheterotrophic conditions. The value of carotenoid/Chl a was enhanced under mixotrophic conditions, but was decreased under photoheterotrophic conditions. In comparison with photoautotrophic conditions, the fluorescence emission peaks and fluorescence excitation peaks were not shifted. The relative fluorescence of photosystem (PS) Ⅰ and PS Ⅱ and the values of F685/F710 and F685/F738 were decreased. Chloroplast thylakoid pairs were less packed under mixotrophic and photoheterotrophic conditions. There was a strong correlation between degree of chloroplast thylakoid packing and the excitation energy kept in PS Ⅱ. These results suggested that the PS Ⅱ activity was reduced by glycerol under mixotrophic conditions, thereby leading to repression of the photosynthetic activity.  相似文献

The status of the names, Physalis minima L. and P. maxima Mill. (Solanaceae), and their alleged presence on the Indian subcontinent are discussed. The issues of nativity and identity of Linnaean Physalis minima are long-debated while the use of the name P. maxima Mill. and its report from India are recent. The available evidence indicates that the name "P. minima L." is misapplied to two different elements, viz., P. angulata L. and P. lagascae Roem. & Schult. The name Physalis minima L. may be rejected as nomen confusum, for which the paper provides the primary information. As on today, it is submerged under the synonymy of P. angulata L. The correct name for the widely known P. minima is P. lagascae. The name "P. maxima Mill." applied to the escape and naturalized weed in the Indian subcontinent and elsewhere is to be substituted by P. pruinosa L., a name misapplied to P. grisea (Waterf.) M. The REL2 gene plays an important role in innate immunity against both Gram (+) and Gram (-) bacteria and malaria parasites in Anopheles gambiae, the main vector of malaria in Africa. Through alternative splicing, REL2 produces two protein products, REL2F (with a Rel-homology domain as well as an inhibitory ankyrin repeat region) and REL2S (without the ankyrin repeats). In the immune-competent cell line SualB from An. gambiae, REL2 has been shown to be a key regulator for cecropin A (or CEC1). The high level expression of CEC1 in SualB was postulated to be the result of constitutive activation of REL2F. Here we showed that REL2F is indeed processed, albeit at a low level, in the SualB cell line. The primary cleavage requires residue 678 (an aspartic acid). Proteolytic cleavage of REL2F can be enhanced by challenge with bacteria Escherichia coli and Bacillus subtilis, but not with fungus Beauveria bassiana. The inducible cleavage can be substantially reduced by RNA interference against PGRP-LC and CASPL1. Over-expression of REL2S or a constitutively active form of REL2F (REL2F380C or REL2F678) in An. gambiae cell line can further increase expression of CEC1 and other antimicrobial peptide genes. Over-expression of these constitutive active proteins in an immune naive cell line, MSQ43, from Anopheles stephensi, results in even more dramatic increased expression of antimicrobial peptides.  相似文献

The alterations of cardiac and lymphocyte β-adrenoceptors were observed in the rats with chronic heart failure produced by constriction of both abdominal aorta and renal artery. The results showed that β1-adrenocep-tor density and mRNA levels were increased, whereas these levels remained unchanged for β2 The concentration-contractile response curve for isoproterenol was shifted to the right in cardiac atrium, whereas the concentration-cAMP accumulation response curve for isoproterenol in myocardium was not changed. The number of β-adrenoceptors in blood lymphocyte was markedly reduced. Thus in the heart-failure rats the density of cardiac β-adrenoceptor was increased accompanying reduced β-adrenoceptor-mediated positive inotropic response, suggesting a post adenylate cyclase dys-function or impaired contractile components. In contrast, the alteration of β-adrenoceptor in lymphocyte is consistent with the reduced β-adrenoceptor-mediated inotropic response in heart.  相似文献

Coptotermes curvignathus Holmgren is capable of feeding on living trees. This ability is attributed to their effective digestive system that is furnished by the termite's own cellulolytic enzymes and cooperative enzymes produced by their gut microbes. In this study, the identity of an array of diverse microbes residing in the gut of C. curvignathus was revealed by sequencing the near-full-length 16S rRNA genes. A total of 154 bacterial phylotypes were found. The Bacteroidetes was the most abundant phylum and accounted for about 65% of the gut microbial profile. This is followed by Firmicutes, Actinobacteria, Spirochetes, Proteobacteria, TM7, Deferribacteres, Planctomycetes, Ferrucomicrobia, and Termite Group 1. Based on the phylogenetic study, this symbiosis can be a result of long coevolution of gut enterotypes with the phylogenic distribution, strong selection pressure in the gut, and other speculative pressures that determine bacterial biome to follow. The phylogenetic distribution of cloned rRNA genes in the bacterial domain that was considerably different from other termite reflects the strong selection pressures in the gut where a proportional composition of gut microbiome of C. curvignathus has established. The selection pressures could be linked to the unique diet preference of C. curvignathus that profoundly feeds on living trees. The delicate gut microbiome composition may provide available nutrients to the host as well as potential protection against opportunistic pathogen.  相似文献

326 species of spiders belonging to 26 families are recorded from the Bolshekhekhtsyrski State Nature Reserve,of them 70 are new records for the reserve and six are new to the fauna of Russia:Asperthorax borealis Ono et Saito,2001; Cyclosa kumadai Tanikawa,1992; Cyclosa okumae Tanikawa,1992(earlier it was identified as C. argenteoalba Bosenberg et Strand,1906); Haplodrassus taepaikensis Paik,1992; Hypselistes fossilobus Fei et Zhu,1993; and Pachygnatha gaoi Zhu et al.,2003. The name Pronous minutus (S. Saito,1939) is synonymized with Pronoides brunneus Schenkel,1936. The male of H. taepaikensis is illustrated for the first time. Composition of the fauna is briefly discussed; 41% of the recorded species have their ranges confined to the SE Palaearctics. By its species diversity,the reserve's fauna is the second largest local fauna eastward of the Urals. An expected spider diversity of this reserve is likely to be over 400 species.  相似文献

Transformation and Functional Expression of the rFCA-RRM2 Gene in Rice   总被引:2,自引:0,他引:2  
The primary aim of the present study was to investigate the overexpression of the rice (Oryza sativa L. subsp, japonica var. Zhonghua 11) flowering control gene (rFCA-RRM2) in monocotyledonous model rice. Constitutive expression of rFCA-RRM2 from the Actl-5 rice promoter caused late flowering in transgenic rice and increased grain weight that was more than 50% higher than that of control plants, which is the first demonstration of rFCA-RRM2 being able to increase rice production. Late flowering was accompanied by strong phenotype and some morphological modifications. These observations suggest that rFCA-RRM2 is a useful tool for phenotype improvement and yield enhancement in cereal crops.  相似文献

Plant quality can directly and indirectly affect the third trophic level. The predation by all the instars of green lacewing, Chrysoperla carnea (S.) (Neuroptera: Chrysopidae) on the cereal aphids, Rhopalosiphum padi (L.), and Sitobion avenae (F.) at varying nitrogen fertilizer levels was calculated under laboratory conditions. Wheat plants were grown on four nitrogen fertilizer levels and aphids were fed on these plants and subsequently offered as food to the C. carnea. Aphid densities of 10, 30, and 90 were offered to first, second, and third instar larvae of green lacewing. Increased nitrogen application improved nitrogen contents of the plants and also the body weight of cereal aphids feeding on them. Aphid consumption by green lacewings was reduced with the increase in nitrogen content in the host plants of aphids. Predation of both aphid species by first, second, and third instars larvae of C. carnea was highest on aphids reared on plants with the lowest rate of fertilization, suggesting a compensatory consumption to overcome reduced biomass (lower aphid size). Total biomass devoured by C. carnea on all nitrogen fertilizer treatments was not statistically different. Additionally, the heavier host prey influenced by the plant nutrition had an effect on the life history characteristics of green lacewings. The larval duration, pupal weight, pupal duration, fecundity, and male and female longevity were significantly affected by the level of nitrogen fertilization to the aphid's host plants, except for pupal duration when fed on S. avenae. This study showed that quantity of prey supplied to the larvae affects the prey consumption and thereafter the life history characteristics of green lacewings.  相似文献

Aims As an endangered wild species with extremely small populations, Ferrocalamus strictus is narrowly distributed in South Yunnan with a small number of individuals. The survey of population structure and community characteristics of the wild population of F. strictus can facilitate understanding its endangered system and mechanisms and provide preliminary research basis for its protection. Methods We investigated the community and population structure of F. strictus, including species composition, population density, population survivorship curve and death factors by plot surveying and sampling. Important findings The community in which population of F. strictus is located in Mojiang has some characteristics of mountain rainforests in terms of appearance and species composition. The population density of F. strictus was 2.04 ind.·m–2. The survivorship curve of F. strictus was between Deevey-I and Deevey-II. The net proliferation rate (R0) of F. strictus population was 1.10, which indicates an expanding population of F. strictus. The death of F. strictus is caused by human logging, natural death, shoot degradation and insects feeding. Among them, artificial cutting accounts for the largest proportion. Ferrocalamus strictus is a species of forest bamboos distributed in the South Asian subtropics, which is a medium-sized bamboo species. Its internode length change suddenly from the base 3–4 nodes. The longest ones exceed 1 m, which ranks at the top of all bamboo species and is closely related to its adaptation to the tropical mountain rainforest environment. © Chinese Journal of Plant Ecology.  相似文献

A low-pathogenicity isolate of Listeria monocytogenes from cow's milk, as screened in mouse and chicken embryonated egg models, was examined for virulence-related phenotypic traits. Corresponding virulence genes (iap, prfA, picA, hly, mpl, actA, plcB, InlA and lnlB) were compared with L. monocytogenes reference strains 10403S and EGD to elucidate the possible molecular mechanisms of low virulence. Although L. monocytogenes H4 exhibited similar patterns to strain 10403S in terms of hemolytic activity, in vitro growth and invasiveness and even had higher adhesiveness, faster intracellular growth and higher phospholipase activity in vitro, it was substantially less virulent than the strain 10403S in mouse and chicken embryo models (50% lethal dose: 10^8.14 VS. 10^5.49 and 10^6.73 VS. 10^1.9, respectively). The genes prfA, picA and mpl were homologous among L. monocytogenes strains H4, 10403S and EGD (〉98%). Genes iap, hly, plcB, lnlA and lnlB of L. monocytogenes 10403S had higher homology to those of strain EGD (〉98%) than isolate H4. The homology of the gene hly between strain 10403S and isolate H4 was 96.9% at the nucleotide level, but 98.7% at the amino acid level. The actA gene of isolate H4 had deletions of 105 nucleotides corresponding to 35 amino acid deletions falling within the proline-rich region. Taken together, this study presents some clues as to reduced virulence to mice and chicken embryos of the isolate H4 probably as a result of deletion mutations of actA.  相似文献

Partial DNA sequences of three genes, that is, mitochondrial large ribosomal subunit (16S), nuclear large ribosomal subunit (28S D2) and mitochondrial NADH1 dehydrogenase (NADH1) gene, were sequenced from different microgas trine species(Braconidae: Microgastrinae) collected fresh from paddy fields. The DNA sequences were used to determine the extent of sequence variation among species in order to evaluate the specific status of each species. Cladistic analysis was also used to infer a phylogenetic relationship among these species. The results showed that sequence divergence among species of the same genus Cotesia was much lower than those among different genera, such as Cotesia, Exoryza and Apanteles; the sequence similarity of 16S rDNA and NADH 1 genes between Cotesia sp. and C. chilonis was higher than that between C. sp. and C. ruficrus.Phylogenetic analyses suggested that four species of Cotesia were always grouped in the same clade regardless of using different analysis methods; Cotesia sp. and C. chilonis are more closely related to each other than to C. ruficrus, different from previous morphological results. Additionally, sequence analyses indicated that NADH1 gene has more parsimony informative characters than 28S rDNA D2 and 16S rDNA at the species-level analysis,indicating that NADH1 gene might be a useful marker for species-level analysis.  相似文献

The content of polyphenolic compounds including chlorogenic acid, vitexin-4"-O-glucoside, vitexin rhamnoside, rutin, vitexin, hyperoside in leaf of Crataegus pinnatifida Bunge var. major N. E. Br. at different times was determined by HPLC. The results show that the content of polyphenolic compounds in different times is various. In April, the total content of polyphenolic compounds and the contents of chlorogenic acid, vitexin-4"-O-glucoside, vitexin rhamnoside, rutin reach the peak. And the content of every compound and the total content of these compounds decrease gradually with the plant growing, and reach the lowest point in July to August. Therefore, in order to extract polyphenolic compounds, the optimal harvest time for C. pinnatifida var. major leaf is April.  相似文献

Mouse TGF-β1 gene was microinjected into male pronuclei of F2 hybrid fertilized eggs obtained by mating CSJLF1 and C57BL/6J inbred strains to generate transgenic mice with over-expressed TGF-β1 gene.The rate of founder production is 31% and Southern blot analysis of founder mice tail DNAs gave an integration efficiency of 33%.TGF-β1 gene could be stably integrated to the chromosomes of transgenic mice and transmitted to their progeny at a rate of 33% in the second generation.Dot blot analysis of tail RNA of some transgenic mice indicated a moderate expression of the transgene.The most interestin finding of the present work is the striking eviation from the normal male:female sex ratio in transgenic mice,with an average ratio of 6.7:1.The possible nature of the predominance of male sex in transgenic mice overexpressing TGF-β1 is discussed.  相似文献

The response of desiccation-sensitive plant tissues to dehydration is significantly affected by dehydration conditions, particularly the rate of drying. Consequently it is important to be able to quantify drying rate. The aim of the study was to assess two models that have been proposed to describe drying kinetics, and thus to provide a quantification of non-linear drying rates, of embryonic axes excised from recalcitrant seeds. These models are an exponential drying time course, and a modified inverse relationship, respectively. For the six species investigated here the inverse function was generally found to fit drying data better than the exponential function under both rapid and slow drying conditions, and so is recommended. The rate of drying, under the conditions used here, was determined by axis size and possibly the nature of the axis outer coverings, rather than the water activity difference between the tissue and surrounding air.  相似文献

The species of the genus Chrysotus Meigen from Shanxi, China are reviewed. Two species are described as new to science: C. shanxiensis sp. nov. and C. thornpenis sp. nov. The effects of ethylene on tension wood formation were studied in 3-year-old Fraxinus mandshurica Rupr. var. japonica Maxim. seedlings in two separate experiments. In experiment 1, ethylene evolution of buds and stems was measured using gas chromatography after 0, 2, 4, 7, 14, and 21 d of treatment; in experiment 2, both aminoethoxyvinylglycine (AVG) and AgNO3 were applied to the horizontally-placed stems, and the cell numbers on sites of applications were measured after 40 d. Ethylene evolution from buds was found to be much greater in tilted seedlings than in upright ones. The cell numbers of wood fibers in shoots and 1-year-old stems were reduced in treatments with 12.5×10^-7μmol/L AVG, 12.5×10^-8μmol/L AVG, and 11.8×10^-8μmol/Lmol/L AgNO3; whereas the horizontal and vertical diameters were reduced by treatment of 12.5×10^-7μmol/L AVG. Ethylene evolutions of shoots and 1-year-old stems were inhibited greatly in comparison with the control by applying 12.5×10^-7μmol/L AVG. The formation of a gelatinous layer of wood fibers was affected by neither AVG nor AgNO3 application. These results suggest that ethylene regulates the quantity of wood production, but does not affect G-layer formation in F. mandshurica Rupr. var.japonica Maxim. seedlings.  相似文献
