The relationship between chlorophyll a (Chl a) and primary productivity(PP) in the uppermost water layer and the water column-based(0–15 m) integral values of those variables were examinedusing measurements taken in Lake Kinneret (Israel) from 1990to 2003. In 81% of all Chl a profiles examined, the distributionwas fairly uniform within the entire 0–15 m water column,and 12.3% of instances showed a prominent subsurface maximum,when the lake phytoplankton was dominated by the dinoflagellatePeridinium gatunense. Chl a can be reliably estimated by remotesensing techniques in the productive and turbid water of LakeKinneret, since Chl a concentration at surface layers can beextrapolated to the entire water column. Light vertical attenuationcoefficient average for wavelengths from 400 to 700 nm, Kd,ranged from 0.203 to 1.954 m–1 and showed high degreeof temporal variation. The maximal rate of photosynthetic efficiency,PBopt [average 3.16 (±1.50)], ranged from 0.25 to 8.85mg C m–3 h–1 mg Chl a–1. Using measured dataof Chl a, PBopt, and light as an input, a simple depth-integratedPP model allowed plausible simulation of PP. However, a lackof correlation between photosynthetic activity and temperature(or other variable with remotely sensed potential) renders theuse of models that require input of photosynthetic efficiencyto calculate integrated PP of little value in the case of productiveand turbid Lake Kinneret.  相似文献

The effect of ultraviolet A (UVA) on growth and photosyntheticrate was studied in diatoms (Melosira spp.) of the phytoplanktonof a eutrophic lake and a cultured green alga Chloretla ellipsoidea.The cells were incubated under photosynthetically active radiation(PAR) (–UVA) or PAR + UVA conditions (+UVA). Growth ofC.ellipsoidea was retarded under +UVA, as shown by an increasein the lag period, but the rate of exponential growth was almostthe same in + and –UVA conditions. The photosyntheticrate was depressed markedly by UVA in Chlorella cells grownunder –UVA. In contrast, cells grown in +UVA showed onlyslight inhibition by UVA and after exposure to UVA for 6 daysthere was no inhibition. During the growth experiment, the cellularchlorophyll a content was higher in +UVA than +UVA grown cells.A similar effect was observed in diatoms from the eutrophicLake Suwa. In vivo fluorescence with (Fa) and without 3-(3,4-dichloropheny)-l,l-dimethylurea (DCMU) (Fb) and the photosynthetic rate were measured forC.ellipsoidea and the diatoms for 5 h under + and –UVAconditions. Soon after C.ellipsoidea had been subjected to +UVA,Fb and Fa / Fb decreased quickly and reached minima after 40min and 1 h, respectively. The suppressed in vivo fluorescenceresumed and full recovery was achieved after 4 h. This suggeststhat reactivation of the photosystem is acquired under prolongedexposure to UVA. A similar shift of Fa + Fb, but no change inFb, was found in diatoms by exposure to UVA. Changes in photosyntheticoxygen evolution by C.ellipsoidea under +UVA were similar tochanges in Fa + Fb. Degradation of chlorophyll a extracted inmethanol was enhanced by UVA. The rate of degradation by UVAwas independent of temperature from 15 to 34°C, suggestinga photochemical reaction. The results indicate that C.ellipsoideaand Melosira spp. acclimatize to prolonged UVA exposure by reactivationof the photosystem and enhanced cellular chlorophyll a synthesis.The ecological importance of these results to phytoplanktonproductivity in natural aquatic environments is discussed.  相似文献

The absorption changes of chlorophyll (Chl) a-like pigments(C670) were studied by ns-ms laser spectroscopy at 77 K in theuntreated and urea-treated homodimeric reaction center (RC)complex of the green sulfur bacterium Chlorobium tepidum. Theuntreated RC complex contained 9 molecules of C670 in additionto 41 molecules of Bchl a and 0.9 molecules of menaquinone-7per one primary electron donor Bchl a dimer (P840). Upon photo-oxidationof P840, C670 showed an absorption change of a red-shift withan isosbestic wavelength at 668 nm. The absorption change ofP840 decayed with time constants (t1/e) of 55 and 37 ms at 283and 77 K, respectively, and was assigned to represent the chargerecombination between P840+ and FeS. In the urea-treatedRC complex, a bleach peaking at 670 nm with a shoulder peakat 662 nm, which is ascribable to the reduced primary electronacceptor A0, was detected after the laser excitationin addition to the shift at 668 nm indicating the formationof the P840+A0 state. The P840+A0 state decayedwith a t1/e of 43 ns at 77 K and produced a triplet state p840Tdue to the suppression of the forward electron transfer. Theseresults indicate the two different types of C670 species inthe RC complex; the one peaking at 670 nm functions as A0, whilethe other peaking at 668 nm shows the electrochromic shift,which presumably functions as the accessory pigment locatedin the close vicinity of P840. The effects of 3-(4'-chlorophenyl)-1, 1-dimethylurea (CMU)onthe fluorescence of photosynthetic pigments in vivo wereinvestigatedin blue-green, red and brown algae and in isolatedspinach chloroplasts.CMU caused an increase in steady statelevel of fluorescenceof chlorophyll a, but did not influencethe fluorescence ofphycobilins. The spectrum of the fluorescenceincrement hada peak at 685 m/µ and a shoulder at 730–740mµ.These two bands probably arise from chlorophyll a(Cf684) belongingto pigment system II.
  2. On excitation of chlorophylla in a red alga, Porphyra yezoensis,a fluorescence band witha peak at 720 mµ was observedbesides a shoulder at 685mµ. The 720 m band is inferredto arise from chlorophylla (probably, Cf-1) in pigment systemI.
  3. On addition of CMUto the algal cells, the induction of fluorescencewas modifiedto take a simple time course. The induction wasobserved onlywith respect to the fluorescence of chlorophylla, but not inthe fluorescence of phycobilins. The spectrumof the "transient"fluorescence showed two emission bands ofchlorophyll a at 685mµ and 740 mµ, and was quitesimilar in form tothe spectrum of the CMU-caused increase insteady state fluorescence.

A photoactive reaction center (RC) complex was isolated fromthe green sulfur bacterium Chlorobium tepidum by solubilizationof membranes with Triton X-100, followed by sucrosedensity gradientcentrifugation, DEAE Bio-Gel A chromatography, and hydroxyapatitechromatography. The purified RC complex contained about 50–70bacteriochlorophyll molecules (BChl) per P840, as assayed byphotooxidafion. It showed a near-infrared BChl a absorptionpeak at 814 nm and shoulders at about 800 and 835 nm at roomtemperature. SDS-PAGE analysis revealed 6 polypeptides withapparent molecular masses of 100, 65, 41, 32, 23, and 18 kDa.The RC complex binds functional P840 and Cyt c551, which werephotooxidized by continuous illumination at room temperature.Upon flash excitation, the bound Cyt c551 was oxidized, andrereduced in the dark with a half-time of 16 and 400 ms in thepresence and absence of 0.1 mM 2,6-dichlorophenol indophenol,respectively, at room temperature. At 551 nm, the amount ofthe Cyt c photooxidized by continuous illumination was 60% ofthe amount determined by chemical oxidation-reduction. The functionalCyt c551/P840 ratio was calculated to be 1.2–1.7. EPRspectroscopy at cryogenic temperatures revealed that the RCcomplex binds three photoreducible Fe-S centers designated tobe CFA, CFB and CFX (C for Chlorobium). The light-harvesting system of photosynthesis was studied infour strains of Porphyra yezoensis differing in their phycoerythrin(PE) content; the red strain, richer in PE than the wild strain,and the green and the yellow strains, poorer in PE. Specialattention was given to possible alteration of pigment systemin response to PE content, especially in the green and the yellowstrains. The relative quantum yields of Chi a fluorescence at–196°C and O2 evolution were compared. Four strains commonly showed a low yield in pigment system II(PS II) fluorescence on Chl a excitation. The yield was as lowas in those algae in which PS II has only a small portion ofChl a as the light harvester. Measurement of O2 evolution gavethe same results. Results indicate that the functional compositionof Chl a system remains unaltered in four strains with differentPE content. PS II in the green and the yellow strains reflectsa reduction in the size of light-harvesting components, suggestingthat pigmentation in these strains is fixed genetically as asun-type. 4 On leave from University of Washington, Seatle, Washington,U.S.A. Guard cell protoplasts were isolated enzymatically from theepidermis of Vicia faba L. and their photosynthetic activitieswere investigated. Time courses of light-induced changes inthe chlorophyll a fluorescence intensity of these protoplastsshowed essentially the same induction kinetics as found formesophyll protoplasts of Vicia. The transient change in thefluorescence intensity was affected by DCMU, an inhibitor ofphotosystem II; by phenylmercuric acetate, an inhibitor of ferredoxinand ferredoxin NADP reductase; and by methyl viologen, an acceptorof photosystem I. Low temperature (77 K) emission spectra ofthe protoplasts had peaks at 684 and 735 nm and a shoulder near695 nm. A high O2 uptake (175 µmol mg–1 Chl hr–1)was observed in guard cell protoplasts kept in darkness, whichwas inhibited by 2 mM KCN or NaN3 by about 60%. On illumination,this O2 uptake was partially or completely suppressed, but itssuppression was removed by DCMU, which indicates that oxygenwas evolved (150 µmol mg–1 Chl hr–1) photosynthetically.We concluded that both photosystems I and II function in guardcell chloroplasts and that these protoplasts have high respiratoryactivity. Conditions for measuring fluorescence induction curves (time-scalems) of in vivo chlorophyll a were studied using cultures ofDunaliella tertiolecta Butcher (Chlorophyceae) and of Thalassiosirapseudonana Hustedt (3H) (Bacillariophyceae), and samples ofnatural phytoplankton populations from the Grand Banks. Thearea above the fluorescence induction curve (ADCMU) and themaximum fluorescence intensity (Fmax) measured in the presenceof 3-(3,4-dichlorophenyl)-1, 1-dimethylurea (DCMU) were computedby microcomputer. Cells must be ‘conditioned’ or‘adapted’ prior to obtaining a fluorescence inductioncurve; dark-adaptation resulted in a lower ADCMU and Fmax thandid adaptation in far-red (720 nm) light, and was the conditioningmethod chosen. ADCMU and Fmax increased linearly with increasingirradiance up to 32.8 W m–2 the highest actinic irradianceavailable. Information on the light history of D. tertiolectawas obtained by following the time-course of change in ADCMUand in Fmax for cells exposed for 10 min to far-red or to bluelight. The rise-time of the fluorescence induction curve andvalues of Fmax were greater for samples of D. tertiolecta concentratedonto glass-fiber filters than for liquid samples, however, valuesof ADCMU for filtered and liquid samples were not significantlydifferent. Samples of Grand Banks phytoplankton collected ontoglass-fiber filters and frozen for 28 d exhibited a significantdecrease in Fmax and in ADCMU relative to the same freshly-filteredsamples. Fluorescence spectra of the pigment system at –196°Cin membrane fragments of Anabaena variabilis and A. cylindricawere investigated. The fluorescence spectra of membrane fragments having four emissionbands at 645–655, 685, 695 and 725 nm were basically similarto those reported for intact cells of blue-green algae, thoughthe emission from phycocyanin (645–655 nm) was far strongerwith membrane fragments than with intact algal cells. Incubation of membrane fragments of A. variabilis in a dilutebuffer (10–2M, pH 7.5) caused an increase in the 645 nmfluorescence and slight decreases in the 685 and 695 nm fluorescences,but had no influence on the 725 nm fluorescence. The decreasein the 685 and 695 nm fluorescences of A. cylindrica was moremarked and had the same kinetics as the inactivation of photosystemII reaction measured by DPIP-photoreduction. When membrane fragments of A. cylindrica were incubated in thebuffer solution at room temperature or in the presence of MgCl2(10–3M) at 0°C; phycobilin aggregates, which emittedthe 655 and 685 nm fluorescence, were solubilized. Chlorophyll-fluorescence parameters of three mosses, Rhytidiadelphusloreus (Hedw.) Warnst.,Anomodon viticulosus (Hedw.) Hook &Tayl. and Grimmia pulvinata (Hedw.) Sm. were measured duringdrying, and following remoistening after a period of desiccation.Most changed little with drying until the full-turgor pointwas approached, when rapid changes accompanied loss of cellwater. In R. loreus all parameters decreased together down toa water content of about 30% d.wt (RWC approx. 0.15). In situ light measurements were used to obtain information oninherent and apparent optical properties. The average verticalattenuation coefficient Kd(ave) varied from 1.1 to 4.6 In unitsm–1 During three periods the variation in Kd(ave) correlatedwith changes in chlorophyll a concentration and specific attenuationcoefficients Ks, of 0.013, 0.014 and 0.022 m2 mg Chl a–1were calculated. Chlorophyll-specific diffuse absorption coefficients(A,) for these periods were 0.012. 0.013 and 0.017 m2 mg Chla–1 and only varied significantly from estimates of Ksin the period when scattering was intense. Absorption coefficientsa(zmid) and scattering coefficients b(zmid) calculated for themid-point of the euphotic zone ranged between 0.45 and 2.9 mand 3.5–52.0 m respectively. Chlorophyll-specific absorptioncoefficients Ka, of 0.005, 0.006 and 0.007 m2 mg Chl a–1and scattering coefficients Kb of 0.05. 0.09 and 0.191 m2 mgChl a–1 were measured during the three periods. The highKb value occurred when gas-vacuolate cyanobactena were dominant.Algal photosynthesis and light absorption were related throughthe maximum quantum yield m which varied between 0.019 and 0.11mol C Einstein–1 while average quantum yields a, variedbetween 0.006 and 0.024 with a mean of 0.013 mol C Einstein–1A comparison of changes in the mean irradiance of the mixedzone and chlorophyll concentration indicated that growth waslight limited below 0.04–0.05 Einsteins absorbed mg Chla–1 day–1.  相似文献

The calanoid copepod, Eudiaplomus graciloides, was reared fromegg to adult on uni-algal diets (0.1. 0.5 and 2.5 mg dry wt1–1) using the green alga, Chlamydomonas reinhardtii,as food, or on a mixed diet consisting of Lake Esrom water filteredthrough a plankton net with pore size 45 µm and supplementedwith C. reinhardtii (2.5 mg dry wt 1–1). On the mixeddiet at 21.0°C growth in body dry wt (W, µg dry wt)was exponential, and the growth constants were 0.21 day–1in the early to mid juvenile stage (N1 - C4) and 0.11 day–1in the late juvenile to early adult stage (C4-A). At 14.5°Cthe corresponding growth rate constants were 0.10 and 0.08 day–1.Similar growth rates were found at uni-algal concentrationsof 0.5 and 2.5 mg dry wt I–1, and it was argued that thethreshold concentration for growth in Eudiaptomus was closeto 0.1 mg dry wt I–1. The clearance (C, ml h–1)of copepodites was measured on the uni-algal diets. The constantsof the regression (C = aWb) were: a = 0.125, b = 0.858 (2000C. reinhardtii ml–1), a = 0.068, b = 0.849 (10 000), a= 0.028, b = 0.875 (50 000). Ingestion rates were calculatedfrom the clearances and the average algal concentrations. Atthe three food levels the average daily rations were 30, 67and 125% of body dry wt. The respiration rate (R, nl O2 h–1)was measured in individuals reared on the mixed diet. The constantsof the regression (R = aWb) were: a = 4.82, b = 1.07 (nauplii,14.5°C), a = 4.17, b = 0.904 (copepodites and adults, 14.5°C),a = 6.87, b = 0.757 (copepodites and adults, 21.0°C). Nosignificant difference in the respiration rate of copepoditesreared on uni-algal diets and the mixed diet could be demonstrated.Energy budgets were calculated. The assimilation efficiencyand the gross growth efficiency of copepodites decreased markedlywith increasing food concentration, the net growth efficiencyvaried from an average of 0.44 at the lowest algal concentrationto 0.60 on the mixed diet. In a coastal area of southern Chile (41° S), the major ammoniumassimilating enzyme glutamine synthetase (GS) was detected ina green dinoflagellate bloom during April 2003. High chlorophylla concentrations (1000 µg L–1) attributable to Gymnodiniumcf. chlorophorum in surface waters were associated with highand very low nitrate reductase activities. Coincident with thebloom, dissolved inorganic nitrogen concentrations were nearthe detection limit (NO3 + NH4+ <0.5 µM). SinceGS correlates with the use of ammonium as an external nitrogensource, we suggest that GS activity seems to be a good indicatorof ammonium utilization in a period dominated by a single dinoflagellatespecies.  相似文献

When grown in pots and well-watered, the relative growth ratesof the above ground parts of two species of Moricandia (M. arvensis,an intermediate C3–C4 species, and M. moricandioides,a C3 species) were inferior to those of two cultivated Brassicaspecies (B. campestris and B. napus). The Moricandia specieshad thicker leaves (greater d.wt per unit leaf area) with morechlorophyll than the Brassica species and had slightly greaterrates of photosynthesis per unit leaf area at an irradiance(400–700 nm) of 2000 µmol quanta m–2 s –1.Leaves of M. arvensis, known to have a CO2 compensation pointbetween that of C3 and C4 species, had a lower ratio of theintercellular to atmospheric partial pressure of CO2 (C1/Ca)and a greater instantaneous water use efficiency (WUE) thanthose of M. moricandioides and the Brassica species. The effects of ultraviolet-B radiation (UV-B, 250–315nm) were determined on Euglena gracilis with respect to speed,phototactic orientation, specific growth rate, and in the presenceof humic substances. Humic substances had a protective effectwhen studying the speed and specific growth rate. However, thedegree of phototactic orientation decreased in UV-B radiationboth with and without humic substances. The inhibition of O2evolution and speed was most pronounced when using cutoff filtersWG280 and WG295. The photosynthetic inhibitor DCMU (10–6M) did not have any effect on the speed, but the 02 evolutiondecreased to zero. The effect of different wavelengths in theUV-B region on the speed of E.gracilis showed the maximum sensitivityat 280 and 290 nm.  相似文献

Nutrient-sufficient cultures of a Trondheimsfjord (Norway) cloneof the marine centric diatom Skeleionema costatum (Grev.) Clevewere grown at 75 µmol m–2 s–1 and 15C at24 and 12 h daylength to study diurnal variations and the effectof daylength on pigment and chemical composition, photosyntheticparameters, dark respiration rates and scaled fluorescence excitationspectra (F), the latter used as estimates for the absorptionof energy available to Photosystem II. Specific growth rateswere 1.06 and 0.56 day in 24 and 12 h daylength, respectively,while dark respiration rates were generally 85% of the net growthrate. The Chla-normalized photosynthetic coefficients PBm andaB were {small tilde}20–25% higher in continuous lightthan at 12 h daylength, while the Chla:C ratio was {small tilde}15%lower (0.051 versus 0.061 w:w). Thus, the carbon-normalizedcoefficients Pcm and ac were <11% lower at 24 h than at 12h daylength. The maximum quantum yield max, the Chla:C ratioand F differed negligibly, as did the light saturation indexlk, the N:C ratio and the ratios Chlc:Chla and Fucoxanthin:Chla. PBm and lk did not exhibit diurnal variations at 24 hdaylength, and varied within 23% of the daily mean at 12 h daylength.Predictions of the daily gross photosynthetic rate based ondata for a given time of the day should thus not be >10%in error relative to an integrated value based on several datasets collected through 24 h. max was 0.084–0.117 mol O2(mol photons) for gross oxygen evolution. However, ifused in mathematical models for predicting the gross and netgrowth rates (i.e. the gross and net carbon turnover rates),‘practical’ values of 0.076 and 0.040 g-at C (molphotons), respectively, should be employed. In situ growth and development of Neocalanus flemingeri/plumchrusstage C1–C4 copepodites were estimated by both the artificial-cohortand the single-stage incubation methods in March, April andMay of 2001–2005 at 5–6°C. Results from thesetwo methods were comparable and consistent. In the field, C1–C4stage durations ranged from 7 to >100 days, dependent ontemperature and chlorophyll a (Chl a) concentration. Averagestage durations were 12.4–14.1 days, yielding an averageof 56 days to reach C5, but under optimal conditions stage durationswere closer to 10 days, shortening the time to reach C5 (fromC1) to 46 days. Generally, growth rates decreased with increasingstage, ranging from 0.28 day–1 to close to zero but weretypically between 0.20 and 0.05 day–1, averaging 0.110± 0.006 day–1 (mean ± SE) for single-stageand 0.107 ± 0.005 day–1 (mean ± SE) forartificial-cohort methods. Growth was well described by equationsof Michaelis–Menten form, with maximum growth rates (Gmax)of 0.17–0.18 day–1 and half saturation Chl a concentrations(Kchl) of 0.45–0.46 mg m–3 for combined C1–3,while Gmax dropped to 0.08–0.09 day–1 but Kchl remainedat 0.38–0.93 mg m–3 for C4. In this study, in situgrowth of N. flemingeri/plumchrus was frequently food limitedto some degree, particularly during March. A comparison withglobal models of copepod growth rates suggests that these modelsstill require considerable refinement. We suggest that the artificial-cohortmethod is the most practical approach to generating the multispeciesdata required to address these deficiencies.  相似文献

The fluorescence characteristics of a freshwater assemblageof picocyanobacteria weredetermined in a mesotrophic lake usingmicrospectrofluorometry. In Jack's Lake. Ontario. 72–98%of the assemblage was comprised of cells with a single excitationpeak for chlorophyll (Chl) a (emission at 680 nm) at 565 ±3)nm. theexcitation spectra for Chl a resembling the spectralcomposition of downwelling irradiance. The assemblage was, therefore,dominated by a single phycobiliproteinpigment type, similarto type 2 phycoerythrin (PE) marine Synechococcus strains. Theshape of excitation spectra did not change significantly withdepth down to 0.6% of incident irradiance or between samplingdates, although the relative intensity of the PE excitationpeak was generally greater for populations below the thermoclinecompared to surface populations during summer stratification.Two separate populations of PE-containing picocyanobacteria,distinguished based on their morphology and plane of division,could be further separated based on their emission spectra (usingblue excitation): a Synechocystis type cell (PE-Sys) consistentlyhad a more pronounced peak at 665 nm from allophycocyanin comparedto a Synechococcus (PE-Syn) type cell. In addition, the ratioof the PE to Chl a peak emissions was higher in PE-Sys and increasedsignificantly with depth below the thermocline. While nitrogenwas limiting in the lake in summer, experimental additions ofnitrogen did not significantly affect this ratio in surfacewater populations, but increased the ratio in PE-Syn populationsat the base of the photic zone. Rhizosolenia mats conduct extensive vertical migrations in theoligotrophic central North Pacific (cNP) gyre that permit thesediatoms to acquire nitrate at depth and return to the surfacefor photosynthesis. The ultimate fate of this N within the ecosystemis unknown, but may include remineralization by grazing, lossto depth by sinking biomass, or N excretion by Rhizosoleniamats. Direct release of N by mats into the mixed layer wouldrepresent an upward biological pump that circumvents the diffusionbarriers and nutrient sinks at the base of the oceanic euphoticzone. We examined Rhizosolenia mat N release along a transect(28–31° N) in the summer of 2002 (Hawaii to California)and 2003 (Hawaii to west of Midway Island) using sensitive fluorometricand chemiluminescence methods. Nitrate, NO2 and NH4+release was determined. Nitrate and NH4+ release by the matsoccurred in both 2002 (22.84 ± 6.04 and 3.69 ±1.74 nmol N µg–1 Chl a h–1, respectively)and 2003 (23.74 ± 3.54 and 3.60 ± 0.74 nmol Nµg–1 Chl a h–1, respectively). Nitrite releaseonly occurred in the 2003 summer period but occurred in bothyears when Fe chelators were added. Fv/Fm values decreased westwardin 2003 suggesting a gradient of increasing physiological stresstowards the west. The various physiological measures are consistentwith concurrent Fe stress; however, other possibilities exist.Nitrate excretion was the dominant form of N release in bothyears and provided a substantial addition to the ambient nitratepool in the mixed layer. Rhizosolenia mat nitrate release suppliesat least 4–7% of the nitrate pool on daily basis, andpossibly as much as 27%. Rhizosolenia mats are part of a largephytoplankton community that appears to migrate, and rates couldbe significantly higher. Literature reports suggest little orno nitrification in the upper euphotic zone, and thus biologicaltransport and release of nitrate may be a major source to thisregion. This N release is uncoupled from upward CO2 transportand, like N2 fixation, provides a component of the N pool availablefor net carbon removal.  相似文献

Fluorescence transients were investigated with the diatom Phaeodactylumtricornutum. Supplementary experiments were done with Chaetocerossp. Under weak excitation ({small tilde}103 erg/cm2sec), fluorescencetransients were induced simply by die oxidation-reduction reactionof Q, the primary reductant of photosystem II. The action spectraindicated that the electron transfer components between thetwo photosystems were in the most reduced state when fucoxanthinwas excited. The transients were observed with the 681 run emissionand with the 707 nm emission at room temperature. At –196°C,induction due to the reduction of Q. appeared both at the 681and 707 nm emissions. Similar results were also obtained withChaetoceros sp. Under strong excitation (104–105 erg/cm2-sec), the fluorescencetransients due to the interconversion between States 1 and 2of die pigment system (cf. ref. 27, 29) were observed. The transientswere induced by die alternate excitation of chlorophyll a andfucoxanthin or chlorophyll c. Conversion from State 2 to State1 was inhibited by DNP and CCCP, indicating that die processwas energy-dependent. Fluorescence spectra at –196°Cwere not altered by die state-conversion of die pigment system. These results suggest diat all die fluorescence bands whichappeared at room temperature and at –196°C were dueto die chlorophyll a of pigment system II in Phaeodactylum andChaetoceros.
