Potato tubers artificially inoculated with Fusarium solani var. coeruleum or F. sulphureum 3 months after harvest were uniformly wounded and held at 5, 10 or 15°C for up to 21 days before immersion in fungicide suspensions. Holding tubers for 14 days at 15°C (curing conditions) or at 5°C did not alter the incidence of dry rot subsequently developing on tubers stored at 10°C, and holding tubers for up to 21 days at 15°C slightly increased disease caused by both pathogens. Thiabendazole, imazalil and prochloraz applied to tubers immediately after wounding almost completely prevented dry rot. Treatment after 3 days was less effective and the amount of disease increased with further delay; fungicides were more effective on tubers held at 5°C than at 10 or 15°C before treatment and storage, and efficacy of the fungicide was decreased by increasing the amount of inoculum on tubers. Wounds became less susceptible to infection by F. solani var. coeruleum and F. sulphureum when tubers were held at 15°C before inoculation, and the incidence of rots was decreased by 70–80% by delaying inoculation for 7 days. Treating tubers with dichlorophen immediately after wounding slightly increased the disease. The effects of fungicide treatment, curing conditions and wound healing are discussed.  相似文献

Over 6 yr the effectiveness of imazalil, prochloraz and fenpiclonil, applied either alone or in a mixture, in controlling gangrene, dry rot, skin spot and silver scurf on potato tubers in store was compared with that of 2-aminobutane and thiabendazole. An assessment was also made of their efficiency in controlling the development of these diseases on the daughter tubers of seed tubers treated at harvest or before planting. Overall, 2-aminobutane was more effective in controlling gangrene (Phoma foveata) in store than the spray-applied fungicides. Deposits of imazalil, thiabendazole and fenpiclonil were greater when sprays were applied with an electrostatic sprayer than with a hydraulic sprayer. The opposite was found with the mixture of prochloraz Mn and tolclofos-methyl. More effective gangrene control was associated with the highest deposits. Fenpiclonil and the mixture of thiabendazole and imazalil were more effective in controlling dry rot (Fusarium solani var. coeruleum) than imazalil alone. The development of dry rot was, however, increased by 2-aminobutane treatment on eight out of 14 stocks. 2-aminobutane gave the greatest reduction (83%) in the severity of skin spot during storage whereas thiabendazole alone, and the mixture of thiabendazole and imazalil, gave mean reductions of 70% and 65% respectively. This mixture and fenpiclonil gave the greatest reduction in the severity of silver scurf although, in general, reductions in silver scurf with fungicide treatment were less than with skin spot. The type of sprayer used to apply a fungicide did not affect the effectiveness of the fungicides in controlling either skin spot or silver scurf on tubers in store, or on the daughter tubers. The incidence of gangrene and dry rot on daughter tubers was not reduced consistently by fungicide treatment of seed tubers of the six stocks tested. However, the severity of skin spot and silver scurf was reduced by fungicide treatments of all eight stocks but the reduction in disease was greater for skin spot than for silver scurf. All fungicides gave reductions in the severity of skin spot, and fenpiclonil and the mixture of thiabendazole and imazalil were the most effective for silver scurf. The effectiveness of the fungicides in controlling these diseases was similar for seed treated at harvest and that treated before planting.  相似文献

To determine the relative importance of seed tubers and stems as sources of inoculum for potato gangrene in progeny tubers, different levels of inoculum of Phoma exigua var. foveata were established in field experiments by planting rotting or contaminated seed tubers and by inoculating stems shortly before haulm desiccation. The pathogen was only occasionally detected by isolation from inside green stems in June and July on plants growing from contaminated but unrotted seed. The incidence of pycnidia on desiccated stems increased with increasing concentration of inoculum contaminating seed tubers and with increasing time interval between desiccation and harvest. Stem infection was probably derived from inoculum on seed tubers spreading via the soil to the stem bases. Stem inoculation of plants growing from uninoculated or uncontaminated seed greatly increased both the gangrene potential of progeny tubers (defined as % wounds with gangrene after uniformly damaging tubers and storing them at 5°C for 12 wk), and tuber contamination (defined as % wounds with gangrene after spreading tuber-borne soil onto test tuber slices and storing them at 5°C for 8 wk). However, when stems of plants growing from rotting or contaminated seed were cut at ground level and removed before desiccation, gangrene potential of progeny tubers was only slightly less than that of tubers from untreated plots. There was no evidence that soil inoculum or aerial spread played a significant role in disease development. Gangrene potential and contamination of progeny tubers were related to Contamination levels on seed tubers. Some transmission also occurred from rotted seed tubers to progeny. Inoculum levels around progeny tubers increased rapidly after desiccation even in plots where stems had previously been cut at soil level and removed to eliminate pycnidial development above ground as a source of inoculum. Both gangrene potential and contamination of progeny tubers were greater on cv. Ulster Sceptre than on cv. Pentland Crown. The results showed that the inoculum on seed tubers, whether from rots or surface contamination, contributed more to the contamination of progeny tubers at harvest than did the inoculum from pycnidia on stems following desiccation of the haulm.  相似文献

In experiments extending over 3 yr, King Edward tubers harvested on various dates from early September until early November were uniformly wounded and kept at 15 oC and c. 95% r.h. for periods of up to 21 days for wounds to cure. When tubers were subsequently inoculated with Phoma exigua var.foveata and incubated at 5 oC for 12 wk, curing for 3 and 7 days decreased gangrene incidence to respectively c. 13 and 4% of the non-cured controls. When tubers were inoculated before curing and immediately after wounding, curing for 7 days decreased gangrene incidence to only c. 68% of the non-cured controls. Curing was also progressively less effective the later the date of harvesting, suggesting that there would be advantages in harvesting potato crops early in the autumn.  相似文献

The susceptibility to gangrene infection of wounds of various shapes and depths on potato tubers was studied by inflicting wounds using differently-shaped brass teeth and rods of different diameters. Inoculating wounds with spore suspensions or damaging tubers which had been previously contaminated with Phoma exigua var. foveata or which had been recently lifted from plots of field experiments showed that wounds in which tissue was crushed were most susceptible to infection. Over a wide range of inoculum concentrations and in experiments using several different cultivars the incidence of infection of any wound type was compared to that of the standard severe cut and crush wound. Using a probit transformation a linear relationship was established, the slope of the line indicating the relative susceptibility of the wound. In 1977 and 1978, crops of cv. Pentland Dell were surveyed for damage incidence, inoculum and inoculum potential on arrival at a commercial bulk store. Nets of tubers buried among the tuber bulk were recovered after storage and gangrene incidence compared with damage and inoculum assessments. Inoculum potential and incidence of severe damage both influenced disease development but damage incidence was of greater importance, showing that priority should be given to decreasing damage and to curing to promote rapid wound healing in endeavours to control the disease.  相似文献

When potato seed tubers (Solanum tuberosum cv. Pentland Javelin) were stored in darkness or diffuse daylight at 9°C and transferred at intervals to conditions suitable for sprouting, their capacity for sprout growth was unaffected by the presence or absence of light during previous storage. When similar tubers were stored at 10°C, 18°C or 25°C, sprout growth commenced earliest at 25°C, but the date was unaffected by fluorescent light. It was concluded that light did not affect the length of the dormant period, but only the rate of sprout elongation after that period had ceased. When tubers with growing sprouts at 10°C or 18°C were transferred from darkness into fluorescent light, sprout growth virtually ceased. Transfer from light into darkness resulted in immediate sprout growth, at a rate comparable with tubers stored continuously in the dark. Tubers of three Peruvian cultivars, stored in farm-scale diffuse-daylight stores, grew progressively shorter sprouts with increasing daily exposure to light from 30 min to 12 h. Storage of cv. Wilja under 21 Wm-2 (total) of white fluorescent light for 10 h per day maintained the sprouts at the same length as ten times this light intensity for 1 h per day. In a subsequent experiment with cv. Bintje the 10 h, low-intensity light regime gave slightly shorter sprouts. It appeared that the total light energy falling on the tubers was the dominant factor controlling sprout growth.  相似文献

In an experiment designed to measure the effect of foliar sprays of fungicides on gangrene in stored tubers thiabendazole but not captafol reduced yield by 30%. Although both fungicides reduced disease incidence in stems and tubers the extent of the reduction was so small that foliar sprays were considered unimportant as a method of control. Sprays of thiabendazole and captafol applied to the haulm 1 wk or 1 wk and 2 wk after desiccation did not reduce the incidence of gangrene in storage.  相似文献

An assessment was made over 3 years of the efficacy of thiabendazole, 2–aminobutane and a formulation of a mixture of thiabendazole and 2–aminobutane in controlling gangrene, skin spot and dry rot when applied either at harvest or after grading at different dates during the storage season.
After harvest applications, the incidence of gangrene was least on tubers treated with the mixture and generally greatest with thiabendazole. When applied after grading the efficacy of the fungicides was not affected by date of grading. Although 2–aminobutane applications at harvest or after grading were equally effective in controlling gangrene, thiabendazole and the mixture were less effective after grading where gangrene was prevalent in a stock. Thiabendazole and the mixture gave good control of dry rot when applied at harvest but were less effective when applied after grading, particularly late in the storage season. Dry rot was sometimes more prevalent on tubers fumigated with 2–aminobutane, especially those treated after grading. Skin spot was more effectively controlled by the mixture and 2–aminobutane than by thiabendazole but, with applications after grading, the efficacy of all fungicides declined the later the date of grading.
Plants grown from tubers treated with the mixture or its 2–aminobutane glycollate constituent exhibited characteristics associated with genetic variation.  相似文献

The influence of temperature on total glycoalkaloid (TGA) synthesis in tubers exposed to light (250 jumol m“2 s”2 PAR, Photosynthetically Active Radiation) or dark environments for 96 h was examined in three potato cultivars. Following 96 h light or dark the tubers were stored without light at 5°C or 24°C and TGA concentrations monitored over the subsequent 30 and 90 days. Exposure to light and cultivar were found to be major factors influencing TGA concentrations; temperature had no significant effect. TGA content in illuminated tubers of cvs ‘Pentland Hawk’ and ‘Kerrs Pink’ were significantly higher (P < 0.01) compared with tubers placed in the dark. TGA concentrations in cv. ‘Desiree’ increased significantly only following exposure to light at low temperatures (P < 0.05). Removal of tubers from storage at 5°C and immediate illumination at 24°C altered the ratio of glycoalkaloids in cvs ‘Pentland Hawk’ and ‘Kerrs Pink’. Regardless of cultivar and storage temperature TGA concentrations were higher at the end of the storage period compared with initial TGA concentrations. During storage TGA concentrations fluctuated widely and gradual accumulation of glycoalkaloids with time was rarely demonstrated except in cv. ‘Desiree’. Tubers stored at 24°C accumulated higher TGA concentrations than those stored at 5°C in cv. ‘Kerrs Pink’ but not in cvs ‘Pentland Hawk’ and ‘Desiree’. Tubers of cv. ‘Kerrs Pink’ exposed to light prior to storage accumulated glycoalkaloids more rapidly than unexposed tubers during storage at 24°C and occasionally at 5°C. Light enhanced glycoalkaloids are not degraded over time.  相似文献

In 3 years, seed tubers of cv. King Edward were planted in field experiments and diseases assessed on the daughter tubers after storage for 24–26 wk. In 1991 and 1992, the seed tubers were treated with a mixture of thiabendazole and imazalil at two rates. Tubers were harvested on several dates from late August to early October and dried under different conditions before storage. In 1990, silver scurf after storage was decreased equally by dry curing for 2 wk and windrowing for 2 h whereas skin spot was decreased most by dry curing, and these effects were greater the earlier tubers were harvested. In 1991, dry curing for 2 wk decreased skin spot and silver scurf more than dry curing for 1 wk, and skin spot was slightly decreased by windrowing for 2 h; windrowing improved the control of both diseases when tubers were subsequently dry cured. In 1991 and 1992 fungicide treatment of seed tubers greatly decreased skin spot after storage and was also effective against silver scurf, but increased black dot when tubers were harvested in September or October. Skin spot was controlled equally well by the two rates of fungicide but silver scurf was less well controlled when the dose was decreased by 75%. It is suggested that amounts of fungicide applied to seed tubers could be decreased provided that the efficiency of application is improved and that crops are harvested early and dried before storage.  相似文献

The incidence of wounds infected by Phoma exigua var. foveata was increased if freshly damaged tubers (recipients) were shaken in a bag with diseased tubers (donors) to simulate the tuber-to-tuber contact that occurs during potato handling. An increase in the number of gangrene rots on damage points also occurred if the recipient tubers were wounded after contact with the diseased tubers, rather than before, and when the donor tubers were heavily infested with P. exigua var. foveata but were free of gangrene lesions. Increasing the proportion of donor to recipient tubers increased the percentage of infected wounds on recipients. Increased incidences of infection in recipient tubers also occurred after they had been passed over an elevator digger when it was lifting stocks of tubers heavily infested with P. exigua var. foveata. When spores of an E +ve isolate of P. exigua var. foveata were sprayed onto the webs of manned potato harvesters, tubers harvested immediately after developed gangrene rots from many of which the E +ve isolate was cultured. An E +ve isolate was also used to demonstrate the transfer of P. exigua var. foveata inoculum from tubers onto soil on riddles of a potato grader and from these soil-coated surfaces onto other tubers during grading.  相似文献

The effect of vacuum packaging on the shelf-life and handling of Pesta granules and seed treatment made with chlamydospores of Fusarium oxysporum strains Foxy2, PSM197 or their mixture was studied at 4°C and 22±3°C over 1 year. In addition, the effects of co-incorporated amendments [urea in Pesta or co-delivered fungicides (Ridomil Gold®, Apron XL®) on coated sorghum seeds], and coating material (Arabic gum ‘AG’, SUET Binder ‘SB’) on the viability of Striga-mycoherbicides were evaluated. Storage under vacuum packaging did not enhance shelf-life of the formulated Striga-mycoherbicidal products after 12 months of storage regardless of the treatment used. The co-incorporated urea into Pesta granules significantly reduced the viability of mycoherbicides, but less so at 4°C (58% strain-stability after 12 months). No significant differences between the coating materials in maintaining the viability of mycoherbicides were observed. The shelf-life of isolates on coated seeds significantly decreased when adding Ridomil Gold®. However, at 4°C, the fungicide Apron XL® allowed better survival of Foxy2 and PSM197 by maintaining their averaged half-lives (t 0.5) by an additional 6 months compared to Ridomil Gold®. In general, Striga-mycoherbicidal product combinations exhibited a significantly higher shelf-life when stored at 4°C than at 22±3°C. The absence of a positive effect of vacuum packaging on shelf-life of Striga-mycoherbicidal products reflects the tolerance of the formulated fungal propagules (chlamydospores) to withstand an oxygen enriched environment and allows their handling and distribution through ordinary packaging systems in Africa. The high compatibility between Striga-mycoherbicides and the co-delivered fungicide Apron XL®, and the fungal storage stability allows simultaneous control of Striga and fungal cereal diseases within an integrated pest management (IPM).  相似文献

The efficiency of thiabendazole in controlling gangrene on potato tubers harvested on different dates was assessed over two years in Scotland (East Craigs) and England (Rothamsted). At East Craigs the incidence of gangrene on wounded and cold-stored tubers was greater than on those tubers kept at ambient store temperatures and graded in January, but the assessments were correlated. The incidence of gangrene increased with delay in harvest after haulm destruction although the pattern differed with year and site. Dipping tubers in thiabendazole or sodium hypochlorite almost eliminated gangrene on all harvest dates at Rothamsted but at East Craigs the incidence of gangrene on dipped tubers was much greater and tended to increase the longer the interval between haulm destruction and harvest. Furthermore, dipping tubers in thiabendazole was much more effective than spraying even though the fungicide residues were twice as large with the latter treatment. All 43 isolates of Phoma foveata collected in Scotland were sensitive to thiabendazole in agar plate tests. The results are discussed in relation to disease development at each of the sites.  相似文献

The germination of seeds of three species of forage grasses, Lolium perenne, Festuca pratensis and Dactylis glomerata, was studied after storage for 3–5 years under five different storage conditions: in aluminium foil packets at —25°C, 0°C and laboratory temperature (c. 18°C), and in manilla paper packets at 0°C and laboratory temperature. With Lolium perenne and Festuca pratensis high germination values at 3 and 7 days were obtained from seed stored at — 25 °C and 0°C in foil packets (5% moisture), but at laboratory temperatures, seed from foil packets gave lower germination values than those from manilla paper packets. At all three temperatures Dactylis glomerata germination after 7 and 14 days was higher in seed stored in foil than in manilla packages. With all three species stored in manilla packets, germination was higher after laboratory than cold storage.  相似文献

The aim of the present investigation was to determine the impact of relative humidity (RH) and temperature on conidial germination, nuclear position and effect of important fungicides on growth and conidial germination of Cercospora canescens. Germination of conidia was observed at RH range 92–100% at 5–35°C. Significant interaction between temperature and RH indicated that higher humidity and high temperature promoted quick germination both in the presence and absence of free moisture. Although in absence of free moisture at 92–95% RH higher temperatures 25–35°C promoted quick evaporation of moisture and no conidial germination. Number of germtube was increased significantly at the optimum temperature 25–30°C and higher humidity (98–100%). But higher temperature 25–35°C with lower RH did not support the conidial germination. This finding is very important for disease forecasting using meteorological data. The spray of Carbendazim as contact fungicide may not be useful since it is not effective against the conidia of C. canescens. Triadimefon did not inhibit the conidia germination but completely inhibited mycelium development at 50 μg/ml. Propriconazole inhibited both conidia germination and mycelial development. Therefore, Propiconazole may be taken as protective as well as curative spray. In non-systemic fungicide, Copper oxychloride gave anticipated result by inhibiting both conidial germination and mycelium development. Therefore, copper oxychloride can be used as protectant fungicides for Cercospora leaf spot caused by C. canescens.  相似文献

The incidence of Phoma exigua var. foveata and Polyscytalum pustulans in dry soil and dust from potato stores was assessed at 10 farms in Scotland producing virus-tested stocks of seed potatoes derived from stem cuttings (VTSC). Samples were collected on three occasions during 1976: during the storage period (February-March), soon after the potatoes were planted (May) and just before the new crop was lifted and stored (August-October). Both pathogens were detected at all three sampling times but P. exigua var. foveata was more frequently detected at the last one than P. pustulans. Soil and dust from various sites within the stores, such as floors, ledges, graders and boxes, were contaminated by these pathogens. Propagules of both pathogens remained airborne for at least 12–17 min after the floors were swept. P. exigua var. foveata remained viable for at least 16 months in dry soil from tuber surfaces stored at 4–6°C. When VTSC tubers were dusted before planting with a dry store soil contaminated by P. exigua var. foveata the gangrene potential of the progeny tubers was much greater than that from untreated tubers or from tubers treated with an uncontaminated soil. The role of inoculum surviving in dry soil is discussed in relation to reinfection of VTSC stocks.  相似文献

Blastospores of three strains of Metarhizium anisopliae were stored in 18 liquids at 4°C, 20°C and 35°C for 18 weeks, 12 weeks or 9 days respectively. Viability was quantified by determination of their germination. In bioassays the virulence of stored blastospores was studied using adults and third instars of Locusta migratoria migratorioides (R. & F.) and compared to those of freshly produced blastospores and conidia. Generally, there was great variability in the viability of blastospores, depending on the fungal strain and the liquids used. Blastospores survived best at 4°C in 10% hydroxyethyl starch; for example, germination of M. anisopliae strain 97 still amounted to more than 80% after storage for 18 weeks. Other suitable liquids were deionized water, 25% Ringer's solution and 1% sodium alginate. The viability of blastospores stored at 20°C was considerably shorter than at 4°C. During storage for 12 weeks at 20°C the best protective liquids for M. anisopliae strain 97 were 25% Ringer's solution (43% germination), deionized water (23%) and 10% hydroxyethyl starch (23%). At 35°C, 45% of M. anisopliae strain 97 blastospores still germinated after storage for 7 days in 25% glycerol. The bioassays revealed that the virulence of blastospores after storage was comparable to that of fresh ones and even better than that of fresh conidia. In general, the LT50 was about 4–6 days at an alternating day/night temperature of 28/20°C.  相似文献

Copper fungicides and mycoinsecticides based on entomopathogenic fungi Beauveria spp. are the most common pesticides used in organic crop production systems. The in vitro effects of the copper fungicides copper oxide, copper hydroxide, copper oxychloride, copper sulphate, dicopper chloride trihydroxide and tribasic copper sulphate were investigated for mycelial growth, sporulation and conidial germination of the ATCC 74040 commercial strain of Beauveria bassiana. Mycelial growth was evaluated on potato dextrose agar plates with 100%, 75%, 50%, 25%, 12.5%, 6.25% and 0% of the recommended application rates of each fungicide at 15 and 25°C. Sporulation and conidial germination were determined at the recommended field doses of each fungicide at 25°C. All copper fungicides had fungistatic or fungicidal effects on B. bassiana that varied according to the dose. Only in two cases, copper oxide at 15°C and copper hydroxide at 25°C, at the lowest concentration of 6.5%, was mycelial growth not statistically significantly inhibited. Inhibition of mycelial growth depended both on the fungicide and its concentration, and partly on temperature. Both sporulation and conidial germination of B. bassiana were significantly inhibited by all fungicides. All fungicides inhibited the sporulation in a similar way (99.8%–100%). With the exception of copper oxychloride (inhibition, 13.6%), the other fungicides showed high detrimental effects on conidial germination (inhibition, 91.7%–100%). The fungus was strongly affected by some fungicides even at the lowest doses. The biological index used for the B. bassiana with copper fungicides ranged from 0.6 (copper sulphate) to 18.1 (copper oxychloride). Therefore, the tested fungicides were classified into the upper half of the highly toxic (T) category and are considered incompatible with the entomopathogenic fungus B. bassiana strain ATCC 74040 under in vivo experimental conditions. These results need to be further verified in vitro under both greenhouse and open-field conditions.  相似文献

The effects of various fungicides on mycelial growth and spore germination of Ascochyta rabiei were determined by incorporating them into potato dextrose agar and measuring colony diameter and observing colony growth and spore germination at 20 ± 2°C. Eight fungicides prevented spore germination of the pathogen at concentrations of 0.125–2 μg/ml, three hindered mycelial growth at 2–4 μg/ml and seven failed to inhibit mycelial growth even at 128 μg/ml. The reference fungicide for the pathogen, chlorothalonil, stopped conidial germination at low rates but did not prevent mycelial growth at 128 μg/ml. Thirteen fungicides were tested against seed infections of the pathogen, and benomyl + thiram, carbendazim and carbendazim + chlorothalonil seed treatments gave more than 85% inhibition on both vacuum‐infiltrated and naturally infected seeds. Coating the seeds with polymers did not increase the effectiveness of fungicides. Three fungicides; (azoxystrobin, chlorothalonil and mancozeb), gave the highest protection in the field but protection decreased with increased inoculum pressure. Addition of humic acid to fungicide suspensions did not affect their performance.  相似文献

Potato tuber worm (PTW), Phthorimaea operculella (Zeller), is a world-wide pest of potato. In rustic stores, PTW larvae can infest 100% of stored tubers. Treatment of tubers in rustic stores with the PTW granulovirus (PoGV) has been demonstrated to protect stored tubers. This is the first study to show the effects of PoGV for protection of tubers stored in refrigerated warehouse conditions. Tubers were treated by dipping in aqueous suspensions of PoGV or water. An estimated 0.0819 larval equivalents of virus or 1.88×109 viral occlusion bodies were deposited on each kilogram of tubers. They were held at 16°C for 11 days before lowering the temperature by 0.5°C per day until 10°C was reached. The tubers were stored at this temperature for 53 days. Mean numbers of infested tubers at the end of the assay was affected by both pre-infestation rate and virus treatment. Mean numbers of infested tubers in the control treatment was 3 tubers per chamber higher than in the virus treatment providing strong evidence that PoGV controlled larvae and minimized spread into un-infested tubers. Of the larvae that were retrieved in virus-treated infested tubers, the mean mortality was 87% compared to 37% in controls.  相似文献
