从污水处理厂分离到一株紫色非硫细菌3p ,该菌株具避光性,能形成孢囊,具片层状光合内膜,在波长798 nm 处的吸收峰很低,需硫胺素和维生素B12 作生长因子。该菌株可以琥珀酸为碳源,最佳生长温度为34 ℃~41 ℃,体内未发现R 体结构,醌类的主要成分是Q9 。对菌株3p 的16SrRNA 基因的分子系统学分析结果表明,菌株3p 与世纪红篓菌( Rhodocista centenaria) 关系最近,相似性为95 % ,二者可归为同一属;与Rc.Centenaria 的杂交结果表明,二者的DNA 相关性为56 % ,故可把3p 定为一个新种:北京红篓菌( Rhodocistapekinense sp .Nov) .  相似文献

通过对猪生长激素(pGH)基因的cDNA进行测序,得到pGHcDNA的全序列,并与Seeburg等报道的序列进行了比较和讨论。然后利用具人工合成启动子和多角体蛋白XIV启动子的转移载体质粒pSXIVVI^+X3/4构建出含pGH基因的重组质粒pX3/4-pGH。将pX3/4-pGH与致死缺失型线性化AcMNPV-OCC^-DNA共转染Sf9细胞,构建出既能形成多角体又能表达pGH基因的苜蓿丫纹夜蛾  相似文献

由含有BHBV-1(BovineHerpesVirus-1)前早期基因的基因组片段亚克隆BICPO(BHV-1InfectedCellProteinO)的DNA序列至表达载体pSVK3,构建质粒pSV2.9。将该质粒与pBLTR-Luc共转染小牛肺细胞,检测转染细胞裂解物的荧光素酶活性,BICPO的表达产物可以显著地激活BIVLTR启动子控制下的荧光素酶基因的表达。根据pSV2.9与含有BIVLTR不同区段缺失的质粒pD-319-Luc、pD-115-Luc、pD-52-Luc共转染小牛肺细胞的实验结果,推测BIVLTR-319位上游区的DNA序列影响BICPO基因产物对BIVLTR表达的激活作用。  相似文献

以携带质粒pAM120(Ap^r,Tc^r/Tn916)的大肠杆菌(E.coli CG120)为供体菌株,与受体菌巴西固氮螺菌采用膜接合法进行接合转移,在选择平板上得到具较高频率的接合子(10^-5/每个供体菌,选择四环素抗性)。从846株四环素抗性接合子中进一步用奈氏法筛选得到氨分泌突变株3株。在无氮培养基上,其氨分泌量可达7.5 ̄14.0mmol/L。用乙炔还原法分析氨分泌突变株在不同浓度氮源  相似文献

通过有机试剂抽提,CF-11纤维素柱层析,从感染水稻齿叶矮缩病毒菲律宾分离株(RiceRaggedStuntVirus,Philippineisolate,简称RRSV-P)的水稻植株中获取该病毒的全基因组,即获得从Segment1到Segment10(S1-S10)的10条双链RNA(dsRNA),然后设计合适的引物,用RT-PCR方法得到S9的cDNA并将其克隆到pUC119质粒上扩增,以双链测序法测定该cDNA的全序列。同时又将此cDNA克隆到大肠杆菌表达质粒pGEX-3X上,在大肠杆菌菌株DE3中用IPTG诱导表达,经超声波破菌、离心、Glutathione-sepharose4B亲和层析,得到纯化的分子量为64kD的融合蛋白。  相似文献

发现于甘肃灵台任家沟上第三系红粘土中的哺乳动物化石包括5目7科9种,Nyctereutes sinensis,Chardinomys sp,Gomphotheeriidae, Rhinocerotidae,Hipparion houfenense Paracamelus sp., Gazella blacki,  Gazella sp.及Antilospira licenti.化石野外的产出层位距第四系黄土底界约22m,古地磁测定为3.4~3.5Ma,即晚上新世早期(MN16下部)。  相似文献

从人新鲜的胎盘组织中提取总RNA,以RT-PCR法获取了人组织金属蛋白酶抑制-3成熟蛋白的cDNA。序列分析表明,TIMP-3成熟蛋白含有188个氨基酸残基,其中12个半胱氨酸残基在TIMP家族中高度保守。将人TIMP-3cDNA插入含7启动子的质粒pET-24构建表达质粒pET-TIMP3,转化大肠杆菌BL21,筛选表达菌株BLTIMP3。  相似文献

A new purple bacterium (strain T4), capable of heterotrophic aerobic and phototrophic anaerobic growth, was isolated from waste water of a noodle factory near Hanoi, Vietnam. A comparison of 16S rDNA sequences revealed its association with the genus Rhodocista. The isolate, tentatively named "Rhodocista hanoiensis", forms cysts after growth on butyrate-containing plates at 42 degrees C. The vegetative cells form short (under aerobic conditions) or long curve-shaped rods. In contrast to other species of this genus T4 does not need any supplines (growth factors, not synthesized by the organisms). Comparative studies of T4 with Rhodocista centenaria (Rhodospirillum centenum) and Rhodocista pekingensis revealed a remarkable physiological versatility regarding nutrient spectra and survival properties of this genus.  相似文献

A gene for photoactive yellow protein (PYP) was previously cloned from Rhodobacter capsulatus (Rc), and we have now found it to be associated with genes for gas vesicle formation in the recently completed genome sequence. However, the PYP had not been characterized as a protein. We have now produced the recombinant RcPYP in Escherichia coli as a glutathione-S-transferase (GST) fusion protein, along with the biosynthetic enzymes, resulting in the formation of holo-RcPYP following cleavage of the GST tag. The absorption spectrum (with characteristic peaks at 435 and 375 nm) and the photocycle kinetics, initiated by a laser flash at 445 nm, are generally similar to those of Rhodobacter sphaeroides (RsPYP) but are significantly different from those of the prototypic PYP from Halorhodospira halophila (HhPYP), which has a single peak at 446 nm and has slower recovery. RcPYP also is photoactive when excited with near-ultraviolet laser light, but the end point is then above the preflash baseline. This suggests that some of the PYP chromophore is present in the cis-protonated conformation in the resting state. The excess 435 nm form in RcPYP, built up from repetitive 365 nm laser flashes, returns to the preflash baseline with an estimated half-life of 2 h, which is markedly slower than that for the same reaction in RsPYP. Met100 has been reported to facilitate cis-trans isomerization in HhPYP, yet both Rc and RsPYPs have Lys and Gly substitutions at positions 99 and 100 (using HhPYP numbering throughout) and have 100-fold faster recovery kinetics than does HhPYP. However, the G100M and K99Q mutations of RcPYP have virtually no effect on kinetics. Apparently, the RcPYP M100 is in a different conformation, as was recently found for the PYP domain of Rhodocista centenaria Ppr. The cumulative results show that the two Rhodobacter PYPs are clearly distinct from the other species of PYP that have been characterized. These properties also suggest a different functional role, that we postulate to be in regulation of gas vesicle genes, which are known to be light-regulated in other species.  相似文献

The genera Azospirillum, Skermanella and Rhodocista form a phylogenetic subgroup within the alfa subclass of Proteobacteria. Based on comparative 16S rRNA sequence analysis a nested set of new oligonucleotide probes was designed. It comprises probes for the whole genus cluster Azospirillum-Skermanella-Rhodocista, for the Azospirilli subcluster I including A. lipoferum, A. doebereinerae, A. largimobile, A. brasilense and A. halopraeferens, for the Azospirilli subcluster II including A. amazonense, A. irakense and the genus Skermanella, for the genus Rhodocista as well as for all Azospirilli species or species cluster. The new probes allow a fast and reliable in situ identification of bacteria belonging to the Azospirillum-Skermanella-Rhodocista-cluster at different phylogenetic levels. The specificity of the new probes was tested with 56 strains of the Azospirillum-Rhodocista-Skermanella-cluster and selected reference cells from other genera by hybridising with the complete probe set. In addition, applications of the fluorescently labelled probes for in situ identification of isolates and for the in situ localisation of A. brasilense on maize roots were demonstrated using confocal laser scanning microscopy.  相似文献

从舟山册子岛船舶压载水泥样中分离到一株细菌S3-22,其与已知细菌的16S rDNA序列相似性低于97%,G+C mol%为54.9 mol%,主要脂肪酸iso-C17:1ω9c(24.99%),细胞醌型为甲基萘醌MK-5。革兰氏染色阴性,最适生长条件为30~37℃、pH7、3%NaCl。嗜盐,氧化酶、接触酶、淀粉酶、酯酶呈阳性,可还原硝酸盐。依据其16S rDNA序列相似性、系统发育学分析及细胞与分子水平的鉴定表明,该菌是Kordiimonas属的一个新种,菌株S3-22的16S rDNA序列登陆号为FJ847942。  相似文献

从形态、生理生化、16S rDNA3个方面确定了番茄青枯菌拮抗菌株3-1-16的分类地位。光学显微镜下观察到菌体为杆状细胞,革兰氏染色均匀,并可见菌体染成蓝紫色。透射电镜进一步观察到细胞内有许多颗粒状物质,无伴胞晶体。Biolog鉴定,3-1-16与巨大芽孢杆菌(Bacillus megaterium)具有最高相似率为98%。16S rRNA分析,3-1-16与巨大芽孢杆菌MO31同源性最高为99.4%。聚类分析显示3-1-16与3株巨大芽孢杆菌聚成一支,支持度为100%。生理生化特征及培养特征测定结果表明,菌株3-1-16鉴定为巨大芽孢杆菌(Bacillus megaterium)。盆栽试验表明该菌株对番茄青枯病防病效果达到81.3%。  相似文献

A facultatively psychrophilic bacterium, previously described as Pseudomonas sp. strain E-3, has been reassigned by phenotypic characterization, chemotaxonomic analysis, DNA-DNA hybridization, and 16S rRNA gene phylogenetic analysis. The organism was a gram-negative, aerobic. straight rod with polar flagella. It was catalase positive and oxidase positive, able to grow at -1 degree C but not at 40 degree C, and produced acid from D-glucose under aerobic conditions. The major isoprenoid quinone was ubiquinone-9, and the DNA G + C content was 57.2 mol%. Phylogenetic analysis based on 16S rRNA gene sequencing indicated that the bacterium is a member of the genus Pseudomonas and was closest to Pseudomonas fragi. Determination of the DNA-DNA relatedness between strain E-3 and P. fragi revealed too low a level of homology (47.9%-51.3%) to identify them as the same species. On the basis of phenotypic characteristics, phylogenetic analysis, and DNA-DNA relatedness data, it is concluded that strain E-3 represents an individual species. Accordingly, the name Pseudomonas psychrophila is proposed. The type strain is E-3T (= JCM 10889).  相似文献

A total of 58 isolates of β-glucosidase-producing microorganisms were isolated from soil around the wild ginseng roots under forest using Esculin-R2A agar. Among these isolates, strain GS33 showed a strong ability to convert ginsenosides Rb1, Rb2, Rc, and Rd into F2, Rg3, C-K, and convert ginsenoside Rg1 into Rh1, and F1. Fermented ginseng products can inhibit ES-2 cells growth and the IC50 value was 0.73 mg ml?1. Phylogenetic analysis based on 16S rRNA gene sequences indicated that the strain GS33 belongs to the genus Penicillium and is most closely related to Penicillium simplicissimum (99 %).  相似文献

目的对分离自啮齿类实验动物的17株螺杆菌进行鉴定,以期获得生物学、遗传学特性典型的菌株作为模式菌株。方法通过生物学特性、16S rRNA序列测定、系统发育树及超微结构分析,对螺杆菌进行种的鉴定。结果确定所分离的17株菌株分为两大类,其中一类为胆汁螺杆菌(H.bilis);另外一类属于螺杆菌属成员。结论所分离到的菌株分别是胆汁螺杆菌和未鉴定到种的螺杆菌,其中胆汁螺杆菌与ATCC51630模式菌株16SrRNA序列比较,相似性达99.6%,可作为检测用的模式菌株。  相似文献

对已经鉴定的嗜酸热硫球菌 (Sulfosphaerellusthermoacidophilumgen.nov .sp .nov)S5菌株的进一步研究发现 ,它既能在好氧条件也能在厌氧条件下代谢元素硫进行化能自养生长 ,结合其 1 6SrRNA基因的分子系统学分析 ,S5菌株应归于Acidianus属。另外 ,S5菌株与Acidianus属中三个已知种基因组DNA的同源性分别仅有 44%、2 2 %和 2 3% ,DNA中G +Cmol%为 38,与已知种 31 0和 32 7有较大差异 ;而且 ,在代谢特性上S5菌株为专性化能无机营养型 ,与Acidianusbrierleyi有明显不同。因此S5菌株应是Acidianus属中一个新种 ,建议定名为 :腾冲嗜酸两面菌 (Acidianustengchongensessp .nov .)。  相似文献

16S ribosomal RNA gene sequences from seven strains of Aquaspirillum peregrinum, Aqu. itersonii, Aqu. polymorphum, and Oceanospirillum pusillum were compared with homologous sequences from other members of helical-shaped bacteria. The bootstrapped neighbor-joining tree, inferred from 887 aligned sites, placed the spirillum taxa assigned to Aquaspirillum, Oceanospirillum, Azospirillum, Magnetospirillum, Rhodospirillum, and Rhodocista of the Proteobacteria in seven clusters of alpha Proteobacteria separately from other shapes of bacteria. Aqu. peregrinum and Aqu. itersonii grouped together in 88% bootstrap support. They were more related to Rhodospirillum rubrum and Rsp. photometricum than Aqu. polymorphum. Aqu. polymorphum was close to Magnetospirillum gryphiswaldense, Mag. magnetotacticum, Rsp. fulvum, and Rsp. molischianum, and more close to Mag. gryphiswaldense. Oce. pusillum was not related to other spirillum taxa and was placed in a separate branch. Rhodocista was very closely related to Azospirillum. Photosynthesis and magnetotaxis, as phenotypic characters, were not important in the classification of helical bacteria.  相似文献
