Nerve growth factor-stimulated mitogen-activated protein kinase (pp42/44MAP) kinase was characterized by sequential column chromatography on DEAE-Sephacel, phenyl-Sepharose CL4B, and S-200. The kinase displayed an apparent molecular mass of 42 kDa and reacted with an antiphosphotyrosine antibody. Peptide mapping of myelin basic protein revealed the presence of one phosphopeptide that was phosphorylated on Thr-97. pp42/44MAP kinase activity was dependent on Mg2+ and inhibited by K252a both in vitro and in vivo. Nerve growth factor-stimulated kinase activation was diminished by down-regulation of protein kinase C with 200 nM 12-phorbol 13-myristate acetate or with staurosporine (1 nM), a protein kinase C inhibitor. Genistein, a protein tyrosine kinase inhibitor, blocked nerve growth factor-mediated neurite extension as well as diminished activation of pp42/44MAP kinase. Our data demonstrate that activation of this kinase system by nerve growth factor displays a requirement for both protein kinase C as well as protein tyrosine kinase. In addition, other agents that are capable of promoting neurite outgrowth in PC12 cells, such as fibroblast growth factor or dibutyryl cyclic AMP, do so independently of activating this kinase system.  相似文献

Goldfish pituitary contains two types of growth hormones. One with five cysteine residues (type-I) similar to other Cyprinid GHs, and the other with four Cys residues (type-II) similar to those of other fish and tertapod species. Recombinant goldfish type II GH (gfGH-II) was produced in Escherichia coli using the pRSETB expression vector. The gfGH-II was produced fused to a leader sequence, which sequestered into inclusion bodies after expression. The inclusion bodies were solubilized using sodium hydroxide and the fusion protein purified by chelating affinity chromatography. Subsequently, gfGH-II was cleaved and analyzed by Western blotting, using a specific antiserum. For comparison we also produced recombinant common carp GH (cGH) which has 95% similarity to gfGH-II, and tested their growth promoting activity in goldfish. Both forms of GH significantly increased the growth rate of goldfish (P<0.05), although cGH was found to have a somewhat higher potency than gfGH-II.  相似文献

生长激素/胰岛素样生长因子-1(GH/IGF-1)轴的合成、分泌、调节及生物学活性与阿尔茨海默病(AD)有密切关系。生长激素(GH)的合成和分泌受生长激素释放激素(GHRH)正向调节。GH/IGF-1轴活性下降导致一系列生理功能变化。GH/IGF-1缺乏可引起衰老及神经退行性变(AD)而导致认知功能的下降,相应激素的补给可以抑制或逆转这种认知障碍。越来越多的证据表明:GH/IGF-1参与AD型痴呆病理过程,对AD有很好的治疗应用前景。本文就生长激素/胰岛素样生长因子1在AD发病中的机理和药理学研究做一综述。  相似文献

The anaerobic oxidation of methane (AOM) is an important methane sink in marine ecosystems mediated by still uncultured Archaea . We established an experimental system to grow AOM communities in different sediment samples. Approaches to show growth of the slow-growing anaerobic methanotrophs have been either via nucleic acids (quantitative PCR) or required long-term incubations. Previous long-term experiments with 13C-labelled methane led to an unspecific distribution of the 13C-label. Although quantitative PCR is a sensitive technique to detect small changes in community composition, it does not determine growth yield. Therefore, we tested an alternative method to detect a biomass increase of AOM microorganisms with 15N-labelled ammonium as N-source. After only 3 weeks, significant 15N-labelling became apparent in amino acids as major structural units of microbial proteins. This was especially evident in methane-containing incubations, showing the methane-dependent uptake of the 15N-labelled ammonium by microorganisms. Cell counts demonstrated a two- and fourfold increase at ambient or elevated methane concentrations. With denaturing gradient gel electrophoresis, over 6 months incubation no changes in community composition of sulphate-reducing bacteria and archaea were detected. These data indicate doubling times for AOM microorganisms between 2 and 3.4 months. In conclusion, the 15N-labelling approach proved to be a sensitive and fast way to show growth of extremely slow-growing microorganisms.  相似文献

The fibroblast growth factors (FGFs) are involved in hematopoiesis and tumorigenesis. However, little is known about the contribution of the FGFs identified within the past 10 years to leukemogenesis. To elucidate whether these FGFs (FGF-8, -9, -10, -11, -12, -13, -14, -16, -17, -18, -19, -20, and -21) are expressed in leukemic cells, we performed RT-PCR analyses using 28 cell lines. The members of a fetal-oncogenic subfamily, FGF-8/-17/-18, were often expressed (53.5%, 25.0%, and 32.1%) with the co-expression of their receptors. Realtime quantitative-PCR analysis showed that FGF-8/-17 were aberrantly expressed in patients with acute leukemia. Moreover, cell proliferation assays revealed the proliferation activity of FGF-17 on leukemic cells expressing its receptors. These results demonstrated that certain recently identified FGFs play an important role in the growth of leukemic cells, possibly with an autocrine mode of action, and that these FGFs will become novel biomarkers for hematopoietic tumors.  相似文献

Abstract

In the current study, the PGP (production of siderophore and IAA, fixation of nitrogen and solubilization of phosphate) potential of forty-five bacterial isolates comprising sixteen genera (Marinibacillus, Zhihengliuella, Bacillus, Nesterenkonia, Halobacillus, Promicromonospora, Terribacillus, Jeotgalibacillus, Staphylococcus, Oceanobacillus, Virgibacillus, Halomonas, Gracilibacillus, Planococcus, Thalassobacillus and Microbacterium) was investigated. Among these forty-five bacterial isolates, most of the isolates (40 isolates) possess plant growth promoting potential. Four bacterial isolates [Marinibacillus sp. (EM1), Zhihengliuella salsuginis (EM30), Planococcus sp. (EK9), and Promicromonospora sp. (EK12)] possessing PGP traits were applied on the barley grown under salt (NaCl) stress in a hydroponic system. Comparing to the un-inoculated group, all the four bacterial isolates significantly improved plant growth that inhibited with NaCl (200?mM) stress. The growth rate of the plants inoculated with Zhihengliuella salsuginis (EM30), Promicromonospora sp. (EK12), Planococcus sp. (EK9), and Marinibacillus sp. (EM1) was 81.86, 80.51, 70.62 and 69.22, respectively. As far as we know, this is the first report showing the PGP potential of Marinibacillus sp. (EM1), Bacillus horikoshii (EM9), Bacillus patagoniensis (EM12), Bacillus agaradhaerens (EK13), Oceanobacillus profundus (EM26) Thalassobacillus devorans (EM34) and Promicromonospora sp. Kuris A. M. 1980. Effect of exposure to Echinostoma liei miracidia on growth and survival of young Biomphalaria glabrata snails. International Journal for Parasitology10: 303–308. Exposure to miracidia of Echinostoma liei resulted in increased mortality and reduced growth of 1–2 mm albino Biomphalaria glabrata snails whether or not the snails became infected. Growth rates for infected and exposed but uninfected snails were significantly more variable than growth rates of unexposed snails. Retarded growth and increased mortality were detected as rapidly as seven to nine days after exposure. Neither growth nor survivorship of 4–6 mm snails was altered upon exposure to or infection by E. liei.  相似文献

The functioning of the biosynthetic pathways of the amino acids alanine, glycine, aspartic acid, glutamic acid and tyrosine, and of nucleosides in the photosynthetic bacterium Chlorobium thiosulfatophilum during heterotrophic growth on 13CO2 and unlabelled acetate was investigated using 13C-NMR as the method for determination of the labelling patterns of the separated substances. On the basis of the analysis of the multiplet structure of the spectra of the tightly-coupled systems, the conclusion was drawn that the Calvin cycle does not function in the experimental conditions used. The labelling pattern of the glutamic acid indicated that about 30% of the amino acid molecules were synthesized through the reactions of the reductive carboxylic acid cycle, the remaining 70% being derived from oxaloacetate and exogenous acetate through the reactions of the Krebs cycle. Labelling patterns of the nucleosides were in agreement with their known biosynthetic pathways.  相似文献

Auxin-mediated elongation growth of maize ( Zea mays L.) coleoptile segments can be nullified by lowering the turgor pressure by 0.45 MPa. Under these conditions irreversible segment length (lin) measured after freezing/thawing increases steadily over a period of 8 h although the in vivo length (ltot) remains constant. This phenomenon, designated as 'cryptic growth', is an indication of a wall-stiffening process which appears to be an intrinsic component of irreversible cell wall extension. Using a range of metabolic inhibitors it is demonstrated that cryptic growth is caused by a temperature-sensitive biochemical process in the cell wall which depends on the presence of O2 and active peroxidase, but not on ATP and protein synthesis. Inhibition of cryptic growth by anaerobic conditions can be alleviated by extermal H2O2. Moreover, cryptic growth can be partially inhibited by the antioxidant ascorbate. It is concluded that cryptic growth represents a wall-stiffening reaction mediated by peroxidase-catalyzed, H2O2-dependent cross-linking of phenolic residues of wall polymers. The experimental demonstration of a wall-stiffening reaction in a rapidly growing organ supports the concept that irreversible cell elongation (growth) is caused by an interplay of two chemorheological reactions, a turgor-dependent wall-loosening reaction and a separate wall-stiffening reaction which fixes the viscoelastically extended wall structure through oxidative cross-linking and thus conferring irreversibility to wall extension.  相似文献

Tomato growth was examined in solution culture under constant pH and low levels of NH4+ or NO3?. There were five nitrogen treatments: 20 mmoles m?3 NH4+, 50 mmoles m?3 NO3?, 100 mmoles m?3 NH4+ 200 mmoles m?3 NO3?, and 20 mmoles m?3 NH4++ 50 mmoles m?3 NO3?. The lower concentrations (20 mmoles m?3 NH4+ and 50 mmoles m?3 NO3?) were near the apparent Km for net NH4+ and NO3? uptake; the higher concentrations (100 mmoles m?3 NH4+ and 200 mmoles m?3 NO3?) were near levels at which the net uptake of NH4+ or NO3? saturate. Although organic nitrogen contents for the higher NO3? and the NH4++ NO3? treatments were 22.2–30.3% greater than those for the lower NO3? treatment, relative growth rates were initially only 10–15% faster. After 24 d, relative growth rates were similar among those treatments. These results indicate that growth may be only slightly nitrogen limited when NH4+ or NO3? concentrations are held constant over the root surface at near the apparent Km concentration. Relative growth rates for the two NH4+ treatments were much higher than have been previously reported for tomatoes growing with NH4+ as the sole nitrogen source. Initial growth rates under NH4+ nutrition did not differ significantly (P≥ 0.05) from those under NO3? or under combined NH4++ NO3?. Growth rates slowed after 10–15 d for the NH4+ treatments, whereas they remained more constant for the NO3? and mixed NH4++ NO3? treatments over the entire observation period of 24–33 d. The decline in growth rate under NH4+ nutrition may have resulted from a reduction in Ca2+, K+, and/or Mg2+ absorption.  相似文献

Abstract: To study how growth factors affect stimulus-secretion coupling pathways, we examined the effects of nerve growth factor (NGF), epidermal growth factor (EGF), and insulin on ATP-induced [Ca2+]i rise and dopamine secretion in PC12 cells. After a 4-day incubation of cells, all three factors increased ATP-induced dopamine secretion significantly. We then examined which step of ATP-induced secretion was affected by the growth factors. Cellular levels of dopamine-β-hydroxylase and catecholamines were increased by NGF treatment but were not affected by EGF or insulin. The ATP-induced [Ca2+]i rise was also enhanced after growth factor treatment. The EC50 of ATP for inducing [Ca2+]i rise and dopamine secretion was increased by NGF treatment but not by treatment with EGF or insulin. Accordingly, the dependence on [Ca2+]i of dopamine secretion was increased significantly only in NGF-treated cells. Our results suggest that for EGF- and insulin-treated PC12 cells, the increase in secretion is mainly due to increased potency of ATP in inducing [Ca2+]i rise. NGF treatment not only increased the potency of ATP but also decreased the Ca2+ sensitivity of the secretory pathway, which as a result becomes more tightly regulated by changes in [Ca2+]i.  相似文献

【目的】丝氨酸/苏氨酸蛋白激酶K(Serine/Threonine protein kinases K)是分枝杆菌类似真核样的蛋白激酶,预测在分枝杆菌的生长和新陈代谢等生理过程中起着重要的作用,解析PknK的生物功能及作用机制,将为结核病的防治提供一定的理论基础。【方法】通过基因敲除等遗传方法获得结核分枝杆菌疫苗株BCG的pknK敲除菌株△pknK、回补菌株pMV361-pknK/△pknK和过表达菌株pMV261-pknK/BCG;对获得的菌株进行生长曲线测定和抗药性分析;通过pulldown-MS方法及生物信息学方法鉴定了PknK相互作用蛋白。【结果】监测各种分枝杆菌△pknK、pMV361-pknK/△pknK和pMV261-pknK/BCG生长,确定PknK负调控BCG生长;抗药性分析显示PknK降低BCG的耐药性;pulldown-MS方法显示PknK与丝氨酸/苏氨酸蛋白激酶PknA和双组分系统中的反应调节因子MtrA、TrcR、MoxR等蛋白相互作用。【结论】研究发现PknK调控分枝杆菌的生长和耐药性,我们的研究为深入研究PknK在结核分枝杆菌中的功能奠定了基础。  相似文献

Effects of the current (38 Pa) and an elevated (74 Pa) CO2 partial pressure on root and shoot areas, biomass accumulation and daily net CO2 exchange were determined for Opuntia ficus-indica (L.) Miller, a highly productive Crassulacean acid metabolism species cultivated worldwide. Plants were grown in environmentally controlled rooms for 18 weeks in pots of three soil volumes (2 600, 6 500 and 26 000 cm3), the smallest of which was intended to restrict root growth. For plants in the medium-sized soil volume, basal cladodes tended to be thicker and areas of main and lateral roots tended to be greater as the CO2 level was doubled. Daughter cladodes tended to be initiated sooner at the current compared with the elevated CO2 level but total areas were similar by 10 weeks. At 10 weeks, daily net CO2 uptake for the three soil volumes averaged 24% higher for plants growing under elevated compared with current CO2 levels, but at 18 weeks only 3% enhancement in uptake occurred. Dry weight gain was enhanced 24% by elevated CO2 during the first 10 weeks but only 8% over 18 weeks. Increasing the soil volume 10-fold led to a greater stimulation of daily net CO2 uptake and biomass production than did doubling the CO2 level. At 18 weeks, root biomass doubled and shoot biomass nearly doubled as the soil volume was increased 10-fold; the effects of soil volume tended to be greater for elevated CO2. The amount of cladode nitrogen per unit dry weight decreased as the CO2 level was raised and increased as soil volume increased, the latter suggesting that the effects of soil volume could be due to nitrogen limitations.  相似文献

Abstract. Despite recent interest in the effects of high CO2 on plant growth and physiology, very little is known about the mechanisms by which plants sense changes in the concentration of this gas. Because atmospheric CO2 concentration is relatively constant and because the conductance of the cuticle to CO2 is low, sensory mechanisms are likely to exist only for intercellular CO2 concentration. Therefore, responses of plants to changes in atmospheric CO2 will depend on the effect of these changes on intercellular CO2 concentration. Although a variety of plant responses to atmospheric CO2 concentration have been reported, most of these can be attributed to the effects of intercellular CO2 on photosynthesis or stomatal conductance. Short-term and long-term effects of CO2 on photosynthesis and stomatal conductance are discussed as sensory mechanisms for responses of plants to atmospheric CO2. Available data suggest that plants do not fully realize the potential increases in productivity associated with increased atmospheric CO2. This may be because of genetic and environmental limitations to productivity or because plant responses to CO2 have evolved to cope with variations in intercellular CO2 caused by factors other than changes in atmospheric CO2.  相似文献

A simple in vitro method is described which allows the study of effects of mineral nutrient supply on growth rate of the tuber. Single node cuttings carrying axillary tubers are cultured under semi-sterile conditions in cooled (+ 1°C) nutrient solutions. Tuber growth is monitored volumetrically and mineral nutrient uptake calculated from the nutrient solution taken up by the cuttings.  相似文献

The moulting cycle and growth of the larval integument of Drosophila melanogaster has been studied by light and electron microscopy. Growth during the first, second and third larval instars is accompanied by 3.0-, 3.4- and 3.7-fold increases in surface area, respectively. Growth in surface area occurs continuously during the larval stages, with no detectable relationship to the moulting cycle. Measurements of the thickness of the cuticular layers show that the endocuticle grows in thickness by apposition and in surface area by stretching. The pre-apolytic epicuticle remains at fairly constant thickness during the increase in surface area, indicating that it grows by intussusception of new components. Post-apolytic epicuticle becomes thinner and increases in surface area by stretching. The epicuticle and pre-ecdysial endocuticle are traversed by filaments, but these do not penetrate the endocuticle secreted after ecdysis. We suggest that the filaments transport breakdown products from the old cuticle inward to the epidermis for reutilization. The growth and deposition of cuticle in two larval growth mutants, lethal (2) giant larvae and Chubby Tubby, involves mechanisms similar to those found in wild-type larvae, but in Chubby Tubby the endocuticle contains inclusions which are ultrastructurally similar to dense epicuticle.  相似文献

A viable wheat hybrid intermediate of the same height as the parents was obtained by crossing the female parent of tall variety NP4 with the male parent of the dwarf variety HD2160. Seeds of the hybrid and its parents were germinated and their growth pattern as well as the activities of peroxidase, indolyl-3-acetic acid oxidase and amylase in extracts made from them were studied at the early seedling stages i.e. up to 96 h.
A positive correlation existed between the length of the axis at the early seedling stage and at mature plant height as far as the parental varieties are concerned but no such correlation was observed with the hybrid. Growth of the hybrid seedlings was less than of its parents. Light appeared to stimulate the longitudinal growth of the axis to different extents in the parents and hybrid. Higher activities of peroxidase, indolyl-3-acetic acid oxidase and amylase were observed in the hybrid as compared to both of its parents. Lethal wheat hybrid also exhibits increased activities of amylase, indolyl-3-acetic acid oxidase and peroxidase. Therefore, it appears that seedling growth and enzyme activities at the seedling stage are not always correlated with hybrid vigour.  相似文献

Sexually mature strobilated adults of Mesocestoides corti were grown consistently from undifferentiated tetrathyridia in vitro using a conventional diphasic culture system. Development (growth, strobilisation and maturation) was compared in vitro and in vivo. Although growth and strobilisation were comparable in vitro and in vivo, during the first 18 days, total length and numbers of proglottids decreased in vivo but continued to increase in vitro after day 18. Both male and female reproductive systems appeared to develop normally in vitro and self copulation was frequently observed in cultured worms.
