We studied the algal community of the Great Salt Plains (GSP), an expansive (65 km2) salt flat situated in north‐central Oklahoma, USA that has been designated as the Salt Plains Microbial Observatory (SPMO) by the National Science Foundation. The GSP offered a unique opportunity to study a terrestrial, hypersaline algal community that experiences wide‐ranging environmental conditions. We were able to show that ammonium‐N, rather than salinity, was the most important predictor of total algal biomass. However, salinity was found to be a significant controlling variable in diatom distribution at the GSP, where diatom abundance was negatively correlated with porewater salinity concentrations. Overall, chlorophytes (likely dominated by Dunaliella spp.) were the most abundant algal group at the consistently hypersaline (>300 ppt) south site. Diatom and cyanobacterial biomass were on average highest at the central site, which experienced greater fluctuations in salinity. While taxonomic diversity was limited to three algal groups (chlorophytes, diatoms, and cyanobacteria), the salinity preferences and halotolerance ranges of isolated strains were quite variable. Although porewater salinities at the GSP are commonly near saturation (>300 ppt), the large majority of isolates had halotolerance ranges below 150 ppt. This suggests that algae at the GSP rarely achieve maximum growth rates, and could only do so when intermittent rain events reduce salinity to optimal levels. Because the vast majority of our strains were isolated from salt‐saturated soil samples, maintaining viability (rather than growth efficiency) appears to be the most successful adaptation to the extreme conditions at the GSP.  相似文献

The Great Salt Plains (GSP), an unvegetated, barren salt flat that is part of the Salt Plains National Wildlife Refuge near Cherokee, Oklahoma, is the site of the Salt Plains Microbial Observatory. At the GSP the briny remains of an ancient sea rise to the surface, evaporate under dry conditions, and leave crusts of white salt. Adaptation to this environment requires development of coping mechanisms providing tolerance to desiccating conditions due to the high salinity, extreme temperatures, alkaline pH, unrelenting exposure to solar UV radiation, and prevailing winds. Several lines of evidence suggest that the same DNA repair mechanisms that are usually associated with UV light or chemically induced DNA damage are also important in protecting microbes from desiccation. Because little is known about the DNA repair capacity of microorganisms from hypersaline terrestrial environments, we explored the DNA repair capacity of microbial isolates from the GSP. We used survival following exposure to UV light as a convenient tool to assess DNA repair capacity. Two species of Halomonas (H. salina and H. venusta) that have been isolated repeatedly from the GSP were chosen for analysis. The survival profiles were compared to those of Escherichia coli, Pseudomonas aeruginosa, and Halomonas spp. from aquatic saline environments. Survival of GSP organisms exceeded that of the freshwater organism P. aeruginosa, although they survived no better than E. coli. The GSP isolates were much more resistance to killing by UV than were the aquatic species of Halomonas reported in the literature [Martin et al. (2000) Can J Microbiol 46:180–187]. Unlike E. coli, the GSP isolates did not appear to have an inducible, error-prone repair mechanism. However, they demonstrated high levels of spontaneous mutation.  相似文献

Dunaliella, a commercially important chlorophyte, is globally distributed in saline habitats. Morphological species have not been definitively reconciled with phylogenetic analyses. Considerable genetic diversity continues to be discovered in new isolates, especially from soil and benthic habitats. Twenty‐nine new isolates from Great Salt Lake, Utah, many from benthic or supralittoral habitats, were phylogenetically analyzed using ITS1+5.8S+ITS2 in comparison to a broad sampling of available sequences. A few new isolates align in one branch of a bifurcated monophyletic Dunaliella salina clade and several cluster within monophyletic D. viridis. Several others align with relatively few unnamed strains from other locations, comprising a diverse clade that may represent two or more new species. The overall Dunaliella clade is relatively robust, but the nearest outgroups are ambiguously placed with extremely long branches. About half of the isolates, all from benthic or supralittoral habitats, have been persistently sarcinoid in liquid media since isolation. This trait is spread across the Dunaliella phylogeny. The morphology of two sarcinoid strains was documented with light microscopy, revealing an extensive glycocalyx. Clumping behavior of unicellular and sarcinoid strains was unaffected by presence or absence of Mg2+ or Ca2+, addition of lectin‐inhibiting monosaccharides, or water‐soluble factors from morphologically opposite strains. Results from this investigation have significantly expanded our current understanding of Dunaliella diversity, but it seems likely that much remains to be discovered with additional sampling.  相似文献

Unlike most other green algae, trebouxiophyceans are predominantly aerophytic and contain many symbiotic representatives. In recent years, a number of new terrestrial trebouxiophycean taxa were described from soils, tree bark, and lichens. The present phylogenetic study reveals three new lineages of free‐living trebouxiophyceans found in North American desert soil crusts and proposes new generic names to accommodate them: Desertella, Eremochloris, and Xerochlorella. This survey of desert isolates also led to discovery of representatives of seven existing genera of trebouxiophyceans. Two of these genera have never been reported to contain desert representatives and one was known previously only from aquatic habitats. Furthermore, we expand the known geographic range of the recently described genus Chloropyrula, heretofore only known from the Ural Mountains. We demonstrate that the diversity of trebouxiophyceans is still underestimated and poorly understood, and that most major trebouxiophycean lineages contain desert‐dwelling taxa.  相似文献

The Great Salt Plains (GSP) in north-central Oklahoma, USA is an expansive salt flat (∼65 km2) that is part of the federally protected Salt Plains National Wildlife Refuge. The GSP serves as an ideal environment to study the microbial diversity of a terrestrial, hypersaline system that experiences wide fluctuations in freshwater influx and diel temperature. Our study assessed cyanobacterial diversity at the GSP by focusing on the taxonomic and physiological diversity of GSP isolates, and the 16S rRNA phylogenetic diversity of isolates and environmental clones from three sites (north, central, and south). Taxonomic diversity of isolates was limited to a few genera (mostly Phormidium and Geitlerinema), but physiological diversity based on halotolerance ranges was strikingly more diverse, even between strains of the same phylotype. The phylogenetic tree revealed diversity that spanned a number of cyanobacterial lineages, although diversity at each site was dominated by only a few phylotypes. Unlike other hypersaline systems, a number of environmental clones from the GSP were members of the heterocystous lineage. Although a number of cyanobacterial isolates were close matches with prevalent environmental clones, it is not certain if these clones reflect the same halotolerance ranges of their matching isolates. This caveat is based on the notable disparities we found between strains of the same phylotype and their inherent halotolerance. Our findings support the hypothesis that variable or poikilotrophic environments promote diversification, and in particular, select for variation in ecotype more than phylotype.  相似文献

Some species of the diving beetle tribe Hygrotini (subfamily Hydroporinae) are among the few insects able to tolerate saline concentrations more than twice that of seawater. However, the phylogenetic relationships of the species of Hygrotini, and the origin and evolution of tolerance to salinity in this lineage, are unknown. In this work, we aim to reconstruct how many times salinity tolerance did evolve in Hygrotini, whether this evolution was gradual or if tolerance to hypersalinity could evolve directly from strictly freshwater (FW) species, and to estimate the probabilities of transition between habitats. We build a phylogeny with ca. 45% of the 137 species of Hygrotini, including all major lineages and almost all of the known halophile or tolerant species. We used sequence data of four mitochondrial (COI‐5′, COI‐3′, 16S + tRNA and NADH1) and three nuclear (28S, 18S and H3) gene fragments, plus ecological data to reconstruct the history of the salinity tolerance using Bayesian inference. Our results demonstrate multiple origins of the tolerance to salinity, although most saline and hypersaline species were concentrated in two lineages. The evolution of salinity was gradual, with no direct transitions from FW to hypersaline habitats, but with some reversals from tolerant to FW species. The oldest transition to saline tolerance, at the base of the clade with the highest number of saline species, was dated in the late Eocene‐early Oligocene, a period with decreasing temperature and precipitation. This temporal coincidence suggests a link between increased aridity and the development of tolerance to saline waters, in agreement with recent research in other groups of aquatic Coleoptera.  相似文献

ABSTRACT. A new ciliate, Trimyema koreanum n. sp., isolated from hypersaline water (salinity of 293‰) from a solar saltern in Korea, was investigated using live observation, protargol impregnation, and gene sequencing. Trimyema koreanum is about 30 × 13 μm in vivo, has usually 23 longitudinal ciliary rows forming two distinct ciliary girdles visible both in vivo and in protargol impregnation. A third indistinct ciliary girdle as well as a girdle of mucocysts is distinguishable only in impregnated cells. We suggest T. koreanum as a new species, differing from the most similar species, T. marinum, by the presence of two distinct ciliary girdles (T. marinum usually has six ciliary girdles clearly visible in living cells and three anterior spirals that encircle the cell completely). Although the number of known 18S rRNA sequences in the genus Trimyema was limited, the Trimyema group including T. koreanum forms a strong clade. The phylogenetic position confirms that the isolate belongs to the genus Trimyema and is different from previously sequenced species. Trimyema koreanum is able to consume both prokaryotes and small eukaryotes (specifically, the alga Dunaliella sp.).  相似文献

Two new isolates of halotolerant chlorophyte algae from the Salt Plains National Wildlife Refuge in Oklahoma, USA, tentatively identified as Dunaliella sp. Teodoresco and Nannochloris sp. Naumann, were characterized with respect to interaction between growth salinity and short‐term heat tolerance. Cells were cultured at 23–25° C over a wide range of salinity. In both species, salinity alone had little effect on maximum photochemical yield (measured by pulse modulated fluorescence) and integrity of the light harvesting system (77 K fluorescence emission spectra). In contrast, Nannochloris exhibited decreasing growth rate (μ), light‐saturated photosynthetic capacity (Pcellmax), respiration (Rd), light‐harvesting efficiency (αcell), and chl content with increasing salinity. Cultures were heated for 2 h near their upper temperature limits (41.5° C for Dunaliella and 45° C for Nannochloris grown at 50 psu). Dunaliella was progressively more heat‐tolerant with increasing salinity. Photochemical yield of cells at 100 and 50 psu was inhibited by about 15% and 40%, respectively, and largely recovered within 30 min after return to 23° C. Thermal inhibition of photochemical yield in Nannochloris was about 45% at both 50 and 100 psu, but recovery was slower at 100 psu. At 20 psu, both species were almost 90% inhibited by high temperature and required more than a day to recover. In both species, 2 h of heating increased the PSI:PSII fluorescence emission ratio (714:690 nm) at all salinities. This ratio largely recovered within 24 h in Dunaliella at 50 and 100 psu and partially recovered in Nannochloris at 100 psu, but cells of both species heated at 20 psu were chlorotic the next day.  相似文献

Recent culture‐based studies demonstrate the distinctiveness of the microbial eukaryote biota of very hypersaline environments. In contrast, microscopy‐based faunistic studies suggest that the biota of habitats of more moderate hypersalinity (60–150‰) overlaps substantially with that of marine environments, but this has barely been studied with modern techniques. To investigate the diversity and salinity tolerance range of these organisms, eight cultures of heterotrophic stramenopiles were established from (or from nearby) moderately hypersaline locations. These isolates represent five independent groups; Groups A, B and C are bicosoecids; Groups D and E belong to Placididea. One isolate (Group A) is a strain of the widespread marine species Cafeteria roenbergensis, and cannot grow above 100‰ salinity. The other isolates – Groups B–E – can all grow at 150–175‰ salinities and are probably moderate halophiles. Groups B–E all represent previously unsequenced species or even genera, although Group B is the sister group of the borderline extreme halophile Halocafeteria. The high level of novelty en countered suggests that moderately hypersaline environments may harbour a heterotrophic stramenopile biota distinct from that of marine environments. Interestingly, our new isolates are all most closely related to marine or halophilic forms, and our phylogenies show large clades defined by saline/non‐saline habitats within bicosoecids, placidomonads and related lineages. In particular, most freshwater/soil bicosoecids form one well‐supported clade. The sole major exception is Bicosoeca, which is intermixed with marine environmental sequences originally referred to as ‘MAST‐13’, which are from brackish water, not typical seawater. It seems that the freshwater/marine barrier has been crossed very few times in the evolutionary history of these heterotrophic stramenopile flagellates.  相似文献

Recently, heterotrophic nanoflagellates (HNF) have been reported to actively ingest prokaryotes in high salinity waters. We report the isolation and culture of an HNF from a Korean saltern pond of 300‰ salinity. The organism is biflagellated with an acronematic anterior flagellum and never glides on surfaces. The mitochondria have tubular cristae. Neither transitional helix nor spiral fiber were observed in the transition zones of the flagella. The cell has a cytostome supported by an arc of eight microtubules, suggesting that our isolate is a bicosoecid. Our isolate had neither mastigonemes, lorica, body scales, nor cytopharynx and thus could not be placed in any of the presently described bicosoecid genera. Phylogenetic analysis of 18S rRNA gene sequences from stramenopiles confirmed the bicosoecid affinities of our isolate, but did not place it within any established genus or family. Its closest relatives include Caecitellus and Cafeteria. The optimal range of growth temperature was 30–35°C. The isolated HNF grew optimally at 150‰ salinity and tolerated up to 363‰ salinity, but it failed to grow below 75‰ salinity, indicating that it could be a borderline extreme halophile. On the basis of its morphological features and position in 18S rRNA trees we propose a novel genus for our isolate; Halocafeteria, n. gen. The species name Halocafeteria seosinensis sp. nov. is proposed.  相似文献

Aims: The aims of the present investigation were to characterize and identify serovars of Riemerella anatipestifer and Riemerella‐like isolates genetically and to test the specificity of PCR tests reported for the identification of R. anatipestifer. Methods and Results: A total of 50 isolates from poultry tentatively classified with Riemerella anatipestifer were characterized genetically by partial sequencing of rpoB and by nearly full sequencing of the 16S rRNA gene for selected isolates. The results obtained were compared with the data from 13 reference strains by phylogenetic analysis. A total of 41 isolates were identified as R. anatipestifer, three as Wautersiella falsenii like, a single isolate as Pelistega europaea, while five isolates were classified as new, unnamed taxa. None of the reported PCR tests for identification of R. anatipestifer were found specific. Conclusions: Characterization of R. anatipestifer and related bacteria by traditional methods is often inconclusive because of inconsistent reactions and phenotypic diversity. For the same reason, gene sequencing and phylogenetic analysis are essential to allow proper classification and identification as demonstrated in the present study. Significance and impact of the Study: The present investigations demonstrated that isolates of R. anatipestifer are often misidentified, and that new serovars should not be accepted unless they have been properly characterized by relevant genetic methods such as gene sequencing. In addition, we showed that the published PCR tests are not specific for this species. Finally, two new taxa were outlined, the final taxonomic positions of which remain to be identified.  相似文献

The Dagong Brine Well, the earliest man-made brine well in record, located in Zigong, Sichuan, China, had the construction and facilities mainly made of bamboo, wood and stone that are eroded easily by halophiles colonizing in the brine of the well. To better preserve this historic placemark, we initiated an investigation on the microbial community and the diversity of the halophilic bacteria in the brine of Dagong well for the first time. A total of 112 aerobic halophile strains were isolated and characterized according to their phenotype, cellular fatty acids composition and 16S rRNA. Furthermore, the intra specific phylogeny of closely related strains was also screened by PCR fragment length polymorphism of 16S–23S ribosomal RNA intergenic spacer regions (ISR). The result showed that the halophilic isolates found in current study were closely related to the following genera: Planococcus, Halomonas, Halobacillus, Oceanobacillus and Virgibacillus, a lineage of the domain Bacteria. Halomonas and Halobacillus were the dominant genera as represents 27% and 40% of the total isolates, respectively. The distribution of other genera was as follows: Oceanobacillus (18%), Virgibacillus (10%) and Planococcus (5%). The ISR analysis disclosed variation of banding pattern in some isolates related to Oceanobacillus and Halobacillus which was observed with phenotypic and physiological characterizations as well. It was clear that these halophiles have adapted to the special man-made hypersaline environment by the basic physiological evolution during phylogenesis and thus resulted in phenotypic and genotypic diversity in the Dagong Brine Well.  相似文献

Euryhaline halophiles grow over a wide range of salinity, from <3% NaCl (seawater equivalent) to >15% NaCl and even saturation level (about 30% NaCl). Several species of euryhaline halophiles occur worldwide, especially in marine environments and also in aquatic and terrestrial habitats of the Antarctic ice-free areas. A biogeographic view of Antarctic halophiles is that their migration among lakes on land is more difficult than in marine setting. Ponds and lakes on land may thus serve as “islands” which facilitate the selection and separation of unique species. We isolated euryhaline halophiles from the saline lake, Suribati Ike, near Syowa Station and placed them into seven groups, each demonstrating a clear depth-related distribution. Six of the seven groups probably represent new species of the genera Halomonas and Marinobacter. This result suggests that Antarctic saline lakes exhibit high selectivity of unique euryhaline halophiles and possibly of other microbial groups.  相似文献

To study how archaeal community responds to environmental changes, we investigated archaeal community structures in waters of three Tibetan saline lakes in northwestern China (Gahai, Xiaochaidan, and Charhan Lakes) with 16S rRNA gene phylogenetic analysis. Temperature, pH, and water chemistry (major anions and cations) of the lakes were measured. Three archaeal clone libraries were constructed with a total of 297 sequences. Incorporating our previous data obtained from other lakes on the Tibetan Plateau, we performed statistical analyses to identify dominant environmental parameters that could account for the observed variations in archaeal community structure. We concluded that salinity and water chemistry (Na and bicarbonate concentration in particular) played an important role in shaping archaeal community. In particular, the relative abundance of archaeal 16S rRNA genes affiliated with the Halobacteriales of the Euryarchaeota increased with salinity, whereas that of crenarchaeotal 16S rRNA gene sequences showed the opposite trend. Crenarchaeotal 16S rRNA gene sequences were retrieved from lake waters with salinity up to 28.3%. These results have important implications for our understanding of response of archaeal community to environmental changes in high-altitude lake ecosystems.  相似文献

Cyanobacteria and eukaryotic algae are important primary producers in a variety of environments, yet their distribution and response to environmental change in saline lakes are poorly understood. In this study, the community structure of cyanobacteria and eukaryotic algae in the water and surface sediments of six lakes and one river on the Qinghai–Tibetan Plateau were investigated with the 23S rRNA gene pyrosequencing approach. Our results showed that salinity was the major factor controlling the algal community composition in these aquatic water bodies and the community structures of water and surface sediment samples grouped according to salinity. In subsaline–mesosaline lakes (salinity: 0.5–50 g L?1), Cyanobacteria (Cyanobium, Synechococcus) were highly abundant, while in hypersaline lakes (salinity: >50 g L?1) eukaryotic algae including Chlorophyta (Chlorella, Dunaliella), Bacillariophyta (Fistulifera), Streptophyta (Chara), and Dinophyceae (Kryptoperidinium foliaceum) were the major members of the community. The relative abundance ratio of cyanobacteria to eukaryotic algae was significantly correlated with salinity. The algae detected in Qinghai–Tibetan lakes exhibited a broader salinity range than previously known, which may be a result of a gradual adaptation to the slow evolution of these lakes. In addition, the algal community structure was similar between water and surface sediment of the same lake, suggesting that sediment algal community was derived from water column.  相似文献

Oxyrrhynchium speciosum (Brid.) Warnst. is a relatively uncommon moss that can be found in wetland habitats across Europe and New Zealand. In this paper, we address the morphological, molecular and ecological relationships between the aquatic form of O. speciosum discovered in a deep, Chara-dominated lake (Budzis?awskie, NW Poland) and its terrestrial relatives. Morphological assessments revealed a considerable difference between terretrial and deep-water samples of O.speciosum. The aquatic form of O. speciosum was, on average, 12 times larger, with more branched stems (4–30 times) and longer leaves (1–2 times).

Investigation of ITS1-5·8S-ITS2-26S region sequences, however, revealed no distinction between the terrestrial and aquatic forms. An analysis of the ecological data suggests that the presence of O. speciosum in deep lake habitats is not incidental (as with many land species developing in aquatic environments). On the contrary, aquatic forms have a distinct ecological niche. Our results confirm that both forms of O. speciosum are closely related to O. hians (Hedw.) Loeske (a terrestrial species). The data indicate that O. speciosum is nested within a well-supported O. hians clade and separation of these species may be connected with polyploidization that occurred during their evolution.  相似文献

Interactions between bacterial microbiota and mosquitoes play an important role in mosquitoes’ capacity to transmit pathogens. However, microbiota assemblages within mosquitoes and the impact of microbiota in environments on mosquito development and survival remain unclear. This study examined microbiota assemblages and the effects of aquatic environment microbiota on the larval development of the Aedes albopictus mosquito, an important dengue virus vector. Life table studies have found that reducing bacterial load in natural aquatic habitats through water filtering and treatment with antibiotics significantly reduced the larva‐to‐adult emergence rate. This finding was consistent in two types of larval habitats examined—discarded tires and flowerpots, suggesting that bacteria play a crucial role in larval development. Pyrosequencing of the bacterial 16S rRNA gene was used to determine the diversity of bacterial communities in larval habitats and the resulting numbers of mosquitoes under both laboratory and field conditions. The microbiota profiling identified common shared bacteria among samples from different years; further studies are needed to determine whether these bacteria represent a core microbiota. The highest microbiota diversity was found in aquatic habitats, followed by mosquito larvae, and the lowest in adult mosquitoes. Mosquito larvae ingested their bacterial microbiota and nutrients from aquatic habitats of high microbiota diversity. Taken together, the results support the observation that Ae. albopictus larvae are able to utilize diverse bacteria from aquatic habitats and that live bacteria from aquatic habitats play an important role in larval mosquito development and survival. These findings provide new insights into bacteria's role in mosquito larval ecology.  相似文献

外来植物往往可以入侵多种生境并受到多种昆虫的采食,而不同生境条件将可能会影响这些入侵植物对昆虫采食的防御策略。以入侵我国的克隆植物——空心莲子草为研究对象,分别选择生长在水生生境、水陆两栖生境和陆生生境中的无性个体(分株),通过50%去叶处理模拟昆虫采食,分析不同生境下空心莲子草对模拟昆虫采食处理的生长及化学防御响应的差异。模拟昆虫采食处理显著抑制了陆生生境、水陆两栖生境以及水生生境下空心莲子草的根、茎、叶和总生物量,但对3种生境下空心莲子草的生物量分配(根冠比、根生物量分配、茎生物量分配和叶生物量分配)均无显著影响。陆生生境下空心莲子草根、茎和总生物量显著高于水陆两栖生境和水生生境,根冠比显著低于水陆两栖生境和水生生境。模拟昆虫采食处理显著降低了空心莲子草的木质素含量,而对单宁和总酚含量影响不显著。生境对木质素含量无显著影响,但陆生生境下空心莲子草单宁含量显著高于水陆两栖生境和水生生境,且总酚含量显著高于水陆两栖生境,表明陆生生境中空心莲子草具有更强的防御能力。空心莲子草木质素含量与总生物量无显著相关性,但在模拟采食情况下,其总酚含量与总生物量呈显著负相关,而无论模拟昆虫采食处理存在与否,空心莲子草单宁含量与总生物量均呈显著正相关。因此,空心莲子草存在昆虫介导的生长和化学防御之间的权衡,在昆虫采食的情况下可通过减少生长来增加对化学防御物质的投入,但生境对空心莲子草这种生长-防御权衡的影响十分有限。  相似文献

水花生(Alternanthera philoxeroides)因其表型可塑性、高生长速率和快速无性繁殖能适应水、陆生境。该文利用光学显微镜和荧光显微镜对水、陆生境的水花生不定根、茎解剖结构、组织化学特征及质外体通透性进行了研究。结果表明:(1)水生境下,其不定根皮层中具较大裂生型通气组织,无次生生长,内皮层具凯氏带且栓质化,皮层和皮下层明显木质化。(2)在陆生环境下,其不定根有次生生长,胞间具通气组织,内皮层具凯氏带且栓质化,皮层和皮下层略木质化;此外,不定根还具额外形成层,产生次生维管束、薄壁组织和不定芽;多年生不定根中具直接分裂的薄壁组织,周皮具凯氏带,且栓质化和木质化。(3)水、陆生境下,其匍匐茎具髓和中空髓腔,发生次生生长,具裂-溶生型通气组织、单层内皮层、厚角组织和木质化且栓质化的角质层,陆生匍匐茎周皮栓质化且木质化。(4)水花生质外体屏障结构组成复杂,黄连素无法穿透质外体屏障结构。水花生的上述解剖学特征,是水花生适应水、陆生境的有力证据。  相似文献
