In the present study, antiproliferative, apoptotic and especially neurotoxic effects of Caulerpa racemosa var. cylindracea dry and wet extracts on mouse neuroblastoma cell line, NA2B were investigated by neurotoxicity screening test (NST). C. racemosa var. cylindracea wet and dry extracts were obtained by methanol (MT) extraction. The effect of the extracts on viability and proliferation was measured by MTT. NA2B cells were induced to differentiate using 1 μM dcAMP and the amount of inhibition of growing neurites in different dilutions (50, 35, 25, 15, 10 and 5 μl/ml) by extracts was measured. The number of apoptotic cells was computed by TUNEL method using cells in culture. It was found that majority of the cells died with dry extract above the level of 15 μl/ml due to the MT effect. Below this level, on the other hand, presence of cell death and antiproliferative effect was noted due to the toxic effects of C. racemosa var. cylindracea which was independent of MT. In all doses of wet extracts, similar but less prominent dose-dependent effects were observed. Below the level of 15 μl/ml, mild toxic effect presented itself with neurite inhibition. In addition to the toxic, apoptotic and antiproliferative effects of C. racemosa var. cylindracea, its neurotoxic effects possessing property at low concentrations which manifesting itself by neurite inhibition was also showed. This species offers a potential for developing new drugs due to its antiproliferative, toxic and apoptotic effects. Nevertheless, its neurotoxic effect is a factor to be considered as multifunctional agents especially in neuronal metabolism.  相似文献

Alien plant species, such as Caulerpa racemosa var. cylindracea, that invade Mediterranean marine vegetated habitats can affect habitat structure. In turn, changes in habitat structure may affect the associated invertebrate assemblages, either through changes in habitat selection or as a result of altered predation efficiency. In order to test for effects of changes in habitat structure resulting from colonization by C. racemosa on prey availability for predators, the importance of amphipods as a trophic resource in natural vegetated habitat was first assessed, and later experiments were undertaken to assess the effects of the alien alga on predation by Thalassoma pavo of two dominant amphipods: Elasmopus brasiliensis (Gammaridea) and Caprella dilatata (Caprellidea). Laboratory experiments were conducted in separate aquaria with five vegetation habitat types: Halopteris scoparia, Jania rubens, C. racemosa without detritus, C. racemosa with detritus, Cymodocea nodosa, together with controls. The vegetation was first defaunated, and then 30 amphipods were introduced to each aquarium and exposed to a single Thalassoma pavo individual for 1 h, after which the fish’s gut contents were examined. Consumption (per fish per hour) of caprellids (11.7 ± 1.4) was higher overall than that of gammarids (8.7 ± 1.5) and likely reflects different microhabitat use by amphipods, which affects susceptibility to predators. Consumption of amphipods also varied by habitat type. The highest predation rate was found in the C. nodosa habitat (12.7 ± 2.19) and the lowest in the C. racemosa habitats with detritus (4.1 ± 1.78) and without detritus (5.2 ± 0.55), which did not differ. The pattern of predation across habitats, however, was similar for both caprellid and gammarid amphipods, indicating a more general effect of habitat on amphipod predation. Our findings showed that invasive species such as C. racemosa can decrease feeding by predators such as T. pavo. Changes in predator–prey interactions could have consequences for food web support in the Mediterranean.  相似文献

Caulerpa racemosa var. cylindracea has been invading various types of substrates in wide areas throughout the Mediterranean Sea. However, the effects of the distribution of this alga on zoobenthos are scanty. The aim of this study was to evaluate the effects of C. racemosa invasion on the feeding habits of some zoobenthic taxa inhabiting the upper infralittoral rocky shores. This was done by identifying the isotopic N and C ratios of several potential food sources and testing differences in isotope composition among the taxa collected from areas invaded and not-invaded areas by C. racemosa. Results suggest that C. racemosa detritus was a significant food source for the polychaete Syllis prolifera, the gammarid Corophium sextonae and the gastropods Cerithium rupestre and Pisinna glabrata. They would also suggest that stable carbon and nitrogen isotope ratios deserve further attention as a possible valuable approach to understand the overall effect of C. racemosa spread on the trophic interactions among the species.  相似文献

Growth of the invasive algae Caulerpa racemosa var. cylindracea in shallow habitats may influence the faunal assemblage composition. We studied its effects on caprellid assemblages associated with shallow-water habitats of hard and soft bottoms from the SE Iberian Peninsula (native rocky-bottom algae, C. racemosa from hard and soft bottoms, and Caulerpa prolifera, Cymodocea nodosa and Posidonia oceanica from soft bottoms). Samples were taken in two different sampling periods (September 2004 and March 2005). A total of seven caprellid species were identified, with important differences in their distribution in different habitats. Total abundance of caprellids was very high in March on native algae on hard bottoms, and on C. racemosa on both soft and hard bottoms. On both hard and soft bottoms, abundances of Caprella hirsuta recorded from C. racemosa were low. On the other hand, a higher abundance of other species, namely C. acanthifera, C. santosrosai, Phtisica marina and Pseudoprotella phasma, was recorded from C. racemosa. The results indicate that C. racemosa may have a positive influence on some caprellid species, while seasonal changes are also evident. It is concluded that introduced C. racemosa may serve as a new habitat, promoting and maintaining caprellid populations in shallow Mediterranean habitats.  相似文献

Bemisia tabaci Genn. (Hemiptera: Aleyrodidae) and Frankliniella occidentalis (Thysanoptera: Thripidae) are major pests in greenhouse grown cucumber crops. Recently, Amblyseius swirskii Athias-Henriot (Acari: Phytoseiidae) was shown an effective biological control agent of both pests. Hence, perhaps both pests can be controlled simultaneously by this predator. However, with simultaneous infestation of both pests, synergistic effects, or interference could affect biological control and perhaps require changes in release rates of the predator. Thus, the aim of the present study was to evaluate different release rates of A. swirskii to control both pests under a worst case scenario of rapid immigration into a cucumber greenhouse. Two experiments were conducted, one simulating the influx of whiteflies alone (whitefly experiment) and the other immigration of whiteflies and thrips together (whitefly plus thrips experiment). Three treatments were compared in the whitefly experiment: (1) B. tabaci alone, (2) B. tabaci + 25 A. swirskii m−2 and (3) B. tabaci + 75 A. swirskii m−2. The high release rate was more effective than the low rate in controlling B. tabaci alone. The high rate was subsequently tested against B. tabaci and F. occidentalis for the whitefly and thrips experiment in which five treatments were compared: (1) B. tabaci alone, (2) F. occidentalis alone, (3) B. tabaci + 75 A. swirskii m−2, (4) F. occidentalis + 75 A. swirskii m−2 and (5) B. tabaci + F. occidentalis + 75 A. swirskii m−2. This rate of A. swirskii controlled whiteflies and thrips either alone or together. Therefore, 75 A. swirskii m−2 should be an adequate rate for controlling both pests either alone or simultaneously in cucumber greenhouses.  相似文献

A genetic transformation system has been developed for callus cells of Crataegus aronia using Agrobacterium tumefaciens. Callus culture was established from internodal stem segments incubated on Murashige and Skoog (MS) medium supplemented with 5 mg l−1 Indole-3-butyric acid (IBA) and 0.5 mg l−1 6-benzyladenine (BA). In order to optimize the callus culture system with respect to callus growth and coloration, different types and concentrations of plant growth regulators were tested. Results indicated that the best average fresh weight of red colored callus was obtained on MS medium supplemented with 2 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-D) and 1.5 mg l−1 kinetin (Kin) (callus maintenance medium). Callus cells were co-cultivated with Agrobacterium harboring the binary plasmid pCAMBIA1302 carrying the mgfp5 and hygromycin phosphotransferase (hptII) genes conferring green fluorescent protein (GFP) activity and hygromycin resistance, respectively. Putative transgenic calli were obtained 4 weeks after incubation of the co-cultivated explants onto maintenance medium supplemented with 50 mg l−1 hygromycin. Molecular analysis confirmed the integration of the transgenes in transformed callus. To our knowledge, this is the first time to report an Agrobacterium-mediated transformation system in Crataegus aronia.  相似文献

Caulerpa spp. are clonal green marine algae which often act as invasive species when growing outside their native biogeographical borders. Over the two past decades, Caulerpa taxifolia has spread along the Mediterranean coast, presently occurring at 70 sites and covering nearly 3,000 ha of subtidal area. New genetic markers (microsatellites) have been developed to assess clonal structure and genetic diversity of recently established populations of the invasive species C. taxifolia and Caulerpa racemosa in comparison with populations of the native Caulerpa prolifera in the Mediterranean. Our results show that nine polymorphic markers have been developed for C. prolifera, seven for C. taxifolia, and three for C. racemosa. Genetic diversity in Caulerpa was assessed in two geographical scales: one at a population scale where 40 thalli units were collected from C. prolifera in Cala d’Or, Mallorca, Spain, and another at a species scale, where 30 sample units were analyzed for C. prolifera, 24 for C. taxifolia, and 24 for C. racemosa from different sites in the Mediterranean, Atlantic, and Pacific Ocean. Number of alleles, expected heterozygosity, and marker amplification success are provided in each case.  相似文献

Invasive epiphyte Lophocladia lallemandii macroalga induces changes in the erect bryozoan Reteporella grimaldii at shallow Posidonia oceanica meadows at a Mediterranean pristine location. Bryozoan densities at noninvaded seagrass plots (88.32 ± 3.11 colonies m−2) are higher than those at invaded plots (13.39 ± 1.09 colonies m−2) with a fourfold decrease in number of colonies. Activation of enzymatic pathways (catalase, superoxide dismutase, glutathione peroxidase) and increase in lipid peroxidation malondialdehyde (MDA) [0.80 ± 0.06 nmol/mg prot at Posidonia oceanica plots to 1.08 ± 0.04 nmol/mg prot at L. lallemandii (P < 0.05)] is observed on sessile bryozoans as response to anoxia caused by L. lallemandii. δ13C of bryozoan isotopic composition differed among treatments, covering a broad range (−19.30‰ invaded to −2.84‰ at noninvaded plots), suggesting modification of food sources. Induced shifts of a filter-feeding erect bryozoan by dense algal turfs at invaded seagrasses are demonstrated, highlighting the need to further address interaction across natural communities and alien species invaded systems before further cascade effects are driven.  相似文献

The behavior of Streptomyces peucetius var. caesius N47 was studied in a glucose limited chemostat with a complex cultivation medium. The steady-state study yielded the characteristic constants μ max over 0.10 h−1, Y XS 0.536 g g−1, and mS 0.54 mg g−1 h−1. The product of secondary metabolism, ɛ-rhodomycinone, was produced with characteristics Y PX 12.99 mg g−1 and m P 1.20 mg g−1 h−1. Significant correlations were found for phosphate and glucose consumption with biomass and ɛ-rhodomycinone production. Metabolic flux analysis was conducted to estimate intracellular fluxes at different dilution rates. TCA, PPP, and shikimate pathway fluxes exhibited bigger values with production than with growth. Environmental perturbation experiments with temperature, airflow, and pH changes on a steady-state chemostat implied that an elevation of pH could be the most effective way to shift the cells from growing to producing, as the pH change induced the biggest transient increase to the calculated ɛ-rhodomycinone flux.  相似文献

Ethylene synthesis is accelerated in response to various environmental stresses like salinity. Ten rhizobacterial strains isolated from wheat rhizosphere taken from different salt affected areas were screened for growth promotion of wheat under axenic conditions at 1, 5, 10 and 15 dS m−1. Three strains, i.e., Pseudomonas putida (N21), Pseudomonas aeruginosa (N39) and Serratia proteamaculans (M35) showing promising performance under axenic conditions were selected for a pot trial at 1.63 (original), 5, 10 and 15 dS m−1. Results showed that inoculation was effective even in the presence of higher salinity levels. P. putida was the most efficient strain compared to the other strains and significantly increased the plant height, root length, grain yield, 100-grain weight and straw yield up to 52, 60, 76, 19 and 67%, respectively, over uninoculated control at 15 dS m−1. Similarly, chlorophyll content and K+/Na+ of leaves also increased by P. putida over control. It is highly likely that under salinity stress, 1-aminocyclopropane-1-carboxylic acid-deaminase activity of these microbial strains might have caused reduction in the synthesis of stress (salt)-induced inhibitory levels of ethylene. The results suggested that these strains could be employed for salinity tolerance in wheat; however, P. putida may have better prospects in stress alleviation/reduction.  相似文献

Seagrasses are well known ecosystem engineers that can significantly influence local hydrodynamics and the abundance and biodiversity of macrobenthic organisms. This study focuses on the potential role of the seagrass canopy structure in altering the abundance of filter-feeding organisms by modifying the hydrodynamic driven food supply. We quantified the effect of two ecosystem engineers with contrasting canopy properties (i.e. Zostera noltii and Cymodocea nodosa) on the food intake rate of a suspension-feeding bivalve Cerastoderma edule living in these seagrass meadows. Field experiments were carried out in two seagrass beds (Z. noltii and C. nodosa) and bare sediment, located on sandflat characterised by a relatively high hydrodynamic energy from waves and currents. Results demonstrated that the filter-feeding rate was almost twofold increased when C. edule was inhabiting Z. noltii meadows (1.10 ± 0.24 μg Chl g Fresh Weight−1) when compared to cockles living on the bare sediment (0.65 ± 0.14 μg Chl g FW−1). Intermediate values were found within C. nodosa canopy (0.97 ± 0.24 μg Chl g FW−1), but filter feeding rate showed no significant differences with values for Z. noltii meadows. There were no apparent correlations between canopy properties and filter-feeding rates. Our results imply that food refreshment within the seagrass canopies was enough to avoid food depletion. We therefore expect that the ameliorated environmental conditions within vegetated areas (i.e. lower hydrodynamic conditions, higher sediment stability, lower predation pressure…) in combination with sufficient food supply to prevent depletion within both canopies are the main factors underlying our observations.  相似文献

The re-emergence of Gymnodinum catenatum blooms after a 10 year hiatus of absence initiated the present investigation. This study aims to evaluate the exposure of small pelagic fishes to paralytic shellfish toxins (PST) during blooms of G. catenatum. Sardines (Sardina pilchardus) were selected as a representative fish species. In order to assess toxin availability to fish, both intracellular PSTs (toxin retained within the algal cells) and extracellular PSTs (toxin found in seawater outside algal cells) were quantified, as well as toxin levels within three fish tissue matrices (viscera, muscle and brain). During the study period, the highest cell densities of G. catenatum reached 2.5 × 104 cells l−1 and intracellular PST levels ranged from 3.4 to 398 ng STXeq l−1 as detected via an enzyme linked immunosorbent assay (ELISA). Measurable extracellular PSTs were also detected in seawater (0.2–1.1 μg STXeq l−1) for the first time in Atlantic waters. The PST profile in G. catenatum was determined via high performance liquid chromatography with fluorescence detection (HPLC-FLD) and consisted mostly of sulfocarbamoyl (C1+2, B1) and decarbamoyl (dcSTX, dcGTX2+3, dcNEO) toxins. The observed profile was similar to that reported previously in G. catenatum blooms in this region before the 10-year hiatus. Sardines, planktivorous fish that ingest a large number of phytoplankton cells, were found to contain PSTs in the viscera, reaching a maximum of 531 μg STXeq kg−1. PSTs were not detected in corresponding muscle or brain tissues. The PST profile characterized in sardine samples consisted of the same sulfocarbamoyl and decarbamoyl toxins found in the algal prey with minor differences in relative abundance of each toxin. Overall, the data suggest that significant biotransformation of PSTs does not occur in sardines. Therefore, planktivorous fish may be a good tracer for the occurrence of offshore G. catenatum blooms and the associated PSTs produced by these algae.  相似文献

The regeneration potential and antioxidative enzyme activities of economically important Brassica rapa var. turnip were evaluated. Calli were induced from leaf explants of seed-derived plantlets on Murashige and Skoog (MS) medium incorporated with different concentrations of various plant growth regulators (PGRs). The highest leaf explant response (83%) was recorded for 2.0 mg l−1 benzyladenine (BA) and 1.0 mg l−1 α-naphthaleneacetic acid (NAA). Subsequent subculturing of callus after 3 weeks of culture, on medium with similar compositions of PGRs, induced shoot organogenesis. The highest shoot induction response (83%) was recorded for 5.0 mg l−1 BA after 5 weeks of transfer. However, 7.8 shoots/explant were recorded for 2.0 mg l−1 BA. The transferring of shoots to elongation medium resulted in 5.1-cm-long shoots on 10 mg l−1 of gibberellic acid (GA3). Rooted plantlets were obtained on MS medium containing different concentrations of indole butyric acid (IBA). The determination of activities of antioxidative enzymes (superoxide dismutase [SOD], ascorbate peroxidase [APX], catalase [CAT], glutathione peroxidase [GPX], and peroxidase [POD]) revealed involvement of these enzymes in callus formation and differentiation. All of the activities were interlinked with each other and played significant roles in the scavenging of toxic free radicals. This study will help in the advancement of a regeneration protocol for B. rapa var. turnip and the understanding of the functions of antioxidative enzymes in plant differentiation.  相似文献

Two oligosaccharides, a heptasaccharide (HS) and an octasaccharide (OS), isolated from Paris polyphylla var. yunnanensis, stimulated the growth and saponin accumulation of Panax ginseng hairy roots at 5–30 mg l−1. HS and OS at 30 mg l−1, fed separately to hairy root cultures at 10 days post-inoculation, increased the root biomass dry weight by more than 70% to ∼20 g l−1 from 13 g l−1 and the total saponin content of roots by more than 1-fold to ∼3.5% from 1.6% (w/w). The results suggest that the two oligosaccharides may have plant growth-regulatory activity in plant tissue cultures.  相似文献

Two experiments were performed to determine how application of the cytokinin benzyladenine (BA) influenced flowering in Doritaenopsis and Phalaenopsis orchid clones. In the first experiment, two vegetative orchid clones growing in 15-cm pots were transferred from a 28°C greenhouse that inhibited flowering to a 23°C greenhouse for flower induction (day 0). A foliar spray (0.2 L m−2) containing BA at 100, 200, or 400 mg L−1 or 25, 50, or 100 mg L−1 each of BA and gibberellins A4 + A7 (BA+GA) was applied on days 0, 7, and 14. Plants treated with BA alone at 200 or 400 mg L−1 had a visible inflorescence 3–9 days earlier and had a mean of 0.7–3.5 more inflorescences and 3–8 more flowers per plant than nontreated plants. The application of BA+GA had no effect on inflorescence number and total flower number at the rates tested. In the second experiment, three orchid clones received a single foliar spray of BA at 200 mg L−1 at six time points relative to time of transfer from 29°C to 23°C (−1, 0, +1, +2, +4, or +6 weeks). A separate group of plants received a BA application at week 0 but was maintained at 29°C. Inflorescence number was greatest in all three orchid clones when plants were treated with BA 1 week after the temperature transfer. Plants that were sprayed with BA and maintained at 29°C did not initiate inflorescences. The promotion of flowering by the application of BA suggests that cytokinins at least partially regulate inflorescence initiation of Doritaenopsis and Phalaenopsis, but its promotion is conditional and BA application cannot completely substitute for an inductive low temperature.  相似文献

Two 60-day experiments were conducted to study the influence of photon flux density (PFD) and temperature on the attachment and development of Gloiopeltis tenax and Gloiopeltis furcata tetraspores. In the first experiment, tetraspores of the two Gloiopeltis species were incubated at five temperature ranges (8°C, 12°C, 16°C, 20°C, 24°C) under a constant PFD of 80 μmol photons m−2 s−1 with a photoperiod of 12:12. In a second experiment, tetraspores were incubated under five PFD gradients (30, 55, 80, 105, 130 μmol photons m−2 s−1) at a constant temperature of 16°C with a photoperiod of 12:12. Maximum density of attached tetraspores was observed at 16°C for both species. Maximum per cent of spore germinating into disc was recorded at 12–16°C for G. tenax and 8–12°C for G. furcata. Maximum per cent of discs producing erect axes for G. tenax and G. furcata were recorded at 24°C and 20°C, respectively. Light had no significant effect on tetraspore attachment and developing into disc, but it affected the growth, sprouting and survival of its discs. Under 30–55 μmol photons m−2 s−1, the discs of the two species of Gloiopeltis did not form thallus until the end of the experiment. Optimum PFD range for G. tenax discs was 80–105 μmol photons m−2 s−1, whilst it was 80–130 μmol photons m−2 s−1 for G. furcata. Results presented in this study are expected to assist the progress of artificial seeding of Gloiopeltis.  相似文献

Recent studies have identified common variants in or near GC, CYP2R1 and NADSYN1/DHCR7 to be associated with 25-hydroxyvitamin D [25(OH)D] levels in European populations. We aimed to examine whether these variants also influence 25(OH)D levels in Chinese. Seven common variants were successfully genotyped and tested for associations with plasma 25(OH)D levels in a population-based cohort of 3,210 Chinese Hans from Beijing and Shanghai. Six common variants at GC (rs4588, rs7041, rs2282679 and rs1155563) and NADSYN1/DHCR7 (rs3829251 and rs1790349) loci were all significantly associated with lower plasma 25(OH)D levels (−0.036 ≤ β ≤ −0.076 per risk-allele, P ≤ 5.7 × 10−5), while CYP2R1-rs2060793 showed a trend toward association with 25(OH)D levels in the Shanghai subpopulation (P = 0.08), but not in the Beijing subpopulation (P = 0.82). Haplotype-based association analyses of the four GC variants showed that only the haplotype that contained all risk-alleles (TACC) was significantly associated with lower plasma 25(OH)D levels (β = −0.085, P = 2.3 × 10−9), while the haplotype containing the risk-alleles of rs4588 and rs2282679 (TATC) was marginally associated with lower 25(OH)D levels (β = −0.054, P = 0.0562) when compared with GCTA haplotype carrying the four protective alleles. Most notably, conditional analyses showed that only GC-rs4588 and GC-rs2282679 (r 2 = 0.97) remained significantly associated with 25(OH)D concentrations (P ≤ 1.9 × 10−5) after adjusting for the other two SNPs in GC. In conclusion, GC and NADSYN1/DHCR7 loci individually and collectively contribute to variation in plasma vitamin D levels in Chinese Hans.  相似文献

Sparse Ulmus pumila woodlands play an important role in contributing to ecosystem function in semi-arid grassland of northern China. To understand the key attributes of soil carbon cycling in U. pumila woodland, we studied dynamics of soil respiration in the canopy field (i.e., the projected crown cover area) and the open field at locations differing in distance (i.e., at 1–1.5, 3–4, 10, and >15 m) to tree stems from July through September of 2005, and measured soil biotic factors (e.g., fine root mass, soil microbial biomass, and activity) and abiotic factors [e.g., soil water content (SWC) and organic carbon] in mid-August. Soil respiration was further separated into root component and microbial component at the end of the field measurement in September. Results showed that soil respiration had a significant exponent relationship with soil temperature at 10-cm depth. The temperature sensitivity index of soil respiration, Q 10, was lower than the global average of 2.0, and declined significantly (P < 0.05) with distance. The rate of soil respiration was generally greater in the canopy field than in the open field; monthly mean of soil respiration was 305.5–730.8 mg CO2 m−2 h−1 in the canopy field and 299.6–443.1 mg CO2 m−2 h−1 in the open field from July through September; basal soil respiration at 10°C declined with distance, and varied from ~250 mg CO2 m−2 h−1 near tree stems to <200 mg CO2 m−2 h−1 in the open field. Variations in soil respiration with distance were consistent with patterns of SWC, fine root mass, microbial biomass and activities. Regression analysis indicated that soil respiration was tightly coupled with microbial respiration and only weakly related to root respiration. Overall, variations in SWC, soil nutrients, microbial biomass, and microbial activity are largely responsible for the spatial heterogeneity of soil respiration in this semi-arid U. pumila woodland.  相似文献
