Farm workers are often exposed to high concentrations of airborne organic dust and fungal conidia, especially when working with plant materials. The purpose of this investigation was to study the possibility of exposure to the mycotoxin ochratoxin A (OTA) through inhalation of organic dust and conidia. Dust and aerosol samples were collected from three local cowsheds. Aerosol samples for determination of total conidia and dust concentrations were collected by stationary sampling on polycarbonate filters. Total dust was analysed by gravimetry, and conidia were counted using scanning electron microscopy. A method was developed for extraction and determination of OTA in small samples of settled dust. OTA was extracted with a mixture of methanol, chloroform, HCI, and water, purified on immunoaffinity column, and analysed by ion-pair HPLC with fluorescence detection. Recovery of OTA from spiked dust samples (0.9–1.0 μg/kg) was 74% (quantitation limit 0.150 μg/kg). OTA was found in 6 out of 14 settled dust samples (0.2–70 μg/kg). The total concentration of airborne conidia ranged from < 1.1 × 104 to 3.9 × 155 per m3, and the airborne dust concentration ranged from 0.08 to 0.21 mg/m3. Conidia collected from cultures of Penicillium verrucosum and Aspergillus ochraceus contained 0.4–0.7 and 0.02–0.06 pg OTA per conidium, respectively. Testing of conidial extracts from these fungi in a Bacillus subtilis bioassay indicated the presence of toxic compounds in addition to OTA. The results show that airborne dust and fungal conidia can be sources of OTA. Peak exposures to airborne OTA may be significant, e.g., in agricultural environments. In 12 selected flats in Sosnowiec, Upper Silesia, the concentrations of particulate aerosol, bioaerosol and bacterial endotoxin were examined. Concentrations of particulate aerosol, bacteria, fungi and endotoxin were in the order of 101–102 μg/m3, 101–103 cfu/m3, 100–102 cfu/m3 and 10−2–10−1 ng/m3, respectively. The most numerous group of microorganisms in indoor air during the winter season were Gram positive mesophilic bacteria. They were more common in flats polluted with tobacco smoke. The concentrations of airborne endotoxins were higher in flats polluted with tobacco smoke in all size ranges. The highest level of endotoxins was found in the fraction of fine particles below 5μm.  相似文献

Cyanobacteria were a major constituent of phototrophic communities in the lakes, ponds and streams of Bylot Island, in the Canadian high Arctic. The waters spanned a range of temperatures (1.8–16.8°C in late July), pH regimes (6.2–9.2) and conductivities (1.5–1700 μS cm−1) but nutrient concentrations were consistently low (< 1 μg dissolved reactive P l−1 at all sites; < 10 μg NO3-N l−1 at most sites). Picoplanktonic species (Synechococcus spp.) were often the numerical dominants in the plankton, and periphytic filamentous species (Oscillatoriaceae) commonly formed thick (5–50 mm) benthic mats. Bloom-forming species of cyanobacteria were either absent or poorly represented even in Chla-rich ponds. The total community biomass ranged from 0.1 to 29.8 μg Chla l−1 in the plankton and from 1.1 to 34.8 μg Chla cm−2 in the benthos. The in vivo absorbance characteristics of isolates from these environments indicated a genetically diverse range of species in each group of Arctic cyanobacteria. Growth versus irradiance relationships were determined for each of the isolates and similarly revealed large genetic differences (maximum growth rates from 0.17 to 0.61 day−1), even between morphologically identical taxa. A comparison of nutrients, pigment concentrations and species composition underscores the strong similarities between freshwater ecosystems in the north and south polar zones. A marine Bacillus circulans DMS-2 was able to grow and produce biosurfactant on glucose mineral salts medium (GMSM) with a reduction in the surface tension up to 27 mN m−1. The microorganism produced 1.64 ± 0.1 g l−1 of crude biosurfactant. The lipopeptide nature of the produced biosurfactant was confirmed by primulin and ninhydrin assays using High Performance Thin Layer Chromatography (HPTLC). Preparative thin layer chromatography (TLC) was performed to purify the lipopeptides from the crude biosurfactant. The critical micelle concentrations (CMC) of the crude and purified products were found to be 90 and 40 mg l−1 respectively. Fourier transform infrared spectrophotometer (FTIR) and matrix assisted laser desorption/ionization time of flight (MALDI-ToF) mass spectral analysis revealed the identity of the produced lipopeptides as surfactin (m/z 1,023 Da) and fengycin (m/z 1,495 Da) isoforms. The purified marine lipopeptides displayed a significant antiproliferative activity against the human colon cancer cell lines HCT-15 (IC50 80 μg ml−1) and HT-29 (IC50 120 μg ml−1).  相似文献

Light irradiation had remarkable effects on callus growth of Oldenlandia affinis with an optimum intensity of 35 μmol m−2 s−1. Biosynthesis of kalata B1, the main cyclic peptide in O. affinis, was induced and triggered with rising irradiation intensities. The highest concentration of kalata B1, 0.49 mg g−1 DW characterised by the maximum productivity of 3.88 μg per litre and day was analysed at 120 μmol m−2 s−1, although callus growth was repressed. The light saturation point was established to be 35 μmol m−2 s−1, where kalata B1 productivity was in a similar order (3.41 μg per day) due to the higher growth index. O. affinis suspension cultures were shown to accumulate comparable specific kalata B1 concentrations in a delayed growth associated production pattern. These were dependent on irradiation intensity (0.16 mg g−1 at 2 μmol m−2 s−1; 0.28 mg g−1 at 35 μmol m−2 s−1). The batch cultivation process resulted in a maximum productivity of 27.30 μg per litre and day with culture doubling times of 1.16 d−1. Submers operation represented a 8-fold product enhancement compared to callus cultivation.  相似文献

Indoor and outdoor airborne fungal propagule concentrations in Mexico City   总被引:7,自引:0,他引:7  
Thirty homes of asthmatic adults located in Mexico City were examined to determine the predominant culturable fungi and the changes in their airborne concentrations. Fungi were cultured and identified microscopically from air samples collected in naturally ventilated homes, during both wet (July–August) and cool dry (November–December) seasons, and from settled dust from the same homes. Airborne dust from indoor yielded 99–4950 cfu m−3, and settled dust 102–106 cfu g−1 on DG18 agar. The indoor geometric mean concentration of airborne fungi during the cool dry season was 460 cfu m−3 while in the wet season it was 141 cfu m−3. Similarly, numbers of airborne fungal propagules out of doors decreased 60% between the dry and wet season. In general, the total fungal concentrations in indoor air were less than 103 cfu m−3 and a large proportion of them was collected in Stage-2 of the Andersen sampler. Moreover, the ratio between indoor and outdoor concentrations was <3:1. Five of the 30 sampled homes yielded >500 cfu m−3 of one genus, with up to 1493Cladosporium cfu m−3 or 2549Penicillium cfu m−3. Also, these two genera were predominant in both airborne and settled dust, and their concentrations were greater indoors than out, indicating a possible indoor source of fungal propagules. The predominant species wereCladosporium herbarum, Penicillium aurantiogriseum andP. chrysogenum. These results suggest that exposure to large concentrations of fungi occurs indoors and is associated with both seasons of the year and with particular home characteristics.  相似文献

Studies on the chemical and biological properties of annual pack ice at a coastal station in Terra Nova Bay (74°41.72′S, 164°11.63′E) were carried out during austral spring at 3-day intervals from 5 November to 1 December 1997. Temporal changes of nutrient concentrations, algal biomasses, taxonomic composition, photosynthetic pigment spectra and P–E relationships were studied. Quantity, composition and degradation rates of organic matter in the intact sea ice were also investigated. In addition, microcosm experiments were carried out to evaluate photosynthetic and photo-acclimation processes of the sympagic flora in relation to different light regimes. High concentrations of ammonia were measured in four ice-cores (weighted mean values of the cores ranged from 4.3 ± 1.9 μM to 7.2 ± 3.4 μM), whereas nitrate and phosphate displayed high concentrations (up to 35.9 μM and 7.6 μM, respectively) only in the bottom layer (135–145 cm depth). Particulate carbohydrate and protein concentrations in the intact sea ice ranged from 0.5 to 2.3 mg l−1 and 0.2 to 2.0 mg l−1, respectively, displaying a notable accumulation of organic matter in the bottom colored layer, where bacterial enzymatic activities also reached the highest values. Aminopeptidase activity was extremely high (up to 19.7 μM l−1 h−1 ± 0.05 in the bottom layer), suggesting a rapid turnover rate of nitrogen–enriched organic compounds (e.g. proteins). By contrast, bacterial secondary production was low, suggesting that only a very small fraction of mobilized organic matter was converted into bacterial biomass (<0.01‰). The sympagic autotrophic biomass (in terms of chlorophaeopigments) of the bottom layer was high, increasing during the sampling period from 680 to 2480 μg l−1. Analyses of pigments performed by HPLC, as well as microscope observations, indicated that diatoms dominated bottom communities. The most important species were Amphiprora sp. and Nitschia cfr. stellata. Bottom sympagic communities showed an average P B max of 0.12 mgC mg Chl−1 and low photoadaptation index (E k=18 μE m−2 s−1, E m=65 μE m−2 s−1). Results of the microcosm experiment also indicated that communities were photo-oxidized when irradiance exceeded 100 μE m−2 s−1. An aerobiological study was performed to evaluate the potential exposure of animals and workers to dust constituents generated during routine animal house work. Different rooms of air conditioned (A, control) and passively ventilated (B, non-air conditioned) animal facilities were sampled, in order to evaluate total airborne culturable fungi and bacteria, fungal spore concentrations and particle levels. Airborne room particles were analyzed gravimetrically and for endotoxin content. All parameters, except for culturable fungi, were higher in facility B and statistically significant, with respect to those from the control facility A. Median values for airborne particle concentration, endotoxin and fungal spores in facility B were: 115 µg m–3, 25 EU m–3, and 2173 spores m–3, respectively. Median values for facility A were: 66 µg m–3, 9 EU m–3, and 248 fungal spores m–3. Broncheoalveolar lavage from rats kept in the rat room of B, presented median concentrations of total cells and lactate dehydrogenase, higher than those found in the control facility (4.4 × 105 vs. 1.1 × 105 and 2.7 UmL-1 vs. 0.39 UmL–1, respectively). Values of total and biological particles of both facilities, as well as the time spent in different rooms, showed that worker exposure was higher during cage washing. It was especially high in the passively ventilated facility (airborne particles 686 µg m–3 3.5 h–1 vs. 976 µg m–3 3.5 h–1, endotoxin 70 EU m–3 3.5 h–1 vs. 108 EU m–3 3.5 h–1). The type of basidiospores and ascospores found, as well as the significant correlation between particle levels and endotoxin contents suggests that wood chip bedding disturbance during cage washing is an important source for airborne biological particles. The changes in broncheoalveolar lavage components found in rats from these facilities and previously reported changes in pro-inflammatory cellular responses found in workers, indicate that these relatively low levels of exposure are enough to induce a biological response. Studies considering the composition of mixed organic dusts, would be needed to reevaluate current occupational standards.  相似文献

Tehran is densely populated possessing unhealthy and polluted air. There are 22 municipality districts in Tehran, and considerable numbers of industries are located within each district. Due to adverse environmental and health effects of trace elements in urban air, this study has been carried out to measure and compare concentration of trace elements including Ag, As, Br, Ce, Co, Cr, Cs, Cu, Mn, Hf, La, Sb, Sc, Sm, Th, and Zn in industrial (district 21) and residential (district 22) areas. For this purpose, total suspended particles were collected in two different time intervals in the year 2008. As a result, maximum and minimum concentrations in residential area associated with Mn (2.9 μg/m3) and Hf (2.92 × 10−4 μg/m3), respectively. Also, in industrial sites, these concentrations are higher for Mn (4.79 × 10−1 μg/m3) and Hf (6.65 × 10−4 μg/m3). The concentrations of Mn (2.07 μg/m3) and Hf (7.46 × 10−4 μg/m3) in traffic zone are close to residential values. General, concentrations of Ce, Co, Cs, La, Sc, Sm, Th, Br, and Hf in industrial area were higher than in residential area. The source of these elements in Tehran's air is mainly anthropogenic. According to the achieved results, the concentration levels of As, Cr, and Zn were close together in both residential and industrial zones. Therefore, these elements could have been originated from natural sources. Also, results show that levels of Ag, Mn, Sb, and Cu in residential area were higher than in industrial area. Finally, obtained data indicated that the trends of average concentration of trace elements in industrial area (from low to high) follow as Hf<Cs<Sm<Th<La<Ce<Sc<Co<Ag<As<Sb<Cr<Br<Zn<Cu<Mn and, in residential area, are in order of Hf<Cs<La<Th<Ce<Sc<Sm<Co<As<Ag<Br<Sb<Cr<Zn<Cu<Mn.  相似文献

Response to irradiance of leaf net photosynthetic rates (P N) of four carrot cultivars: Cascade, Caro Choice (CC), Oranza, and Red Core Chantenay (RCC) were examined in a controlled environment. Gas exchange measurements were conducted at photosynthetic active radiation (PAR) from 100 to 1 000 μmol m−2 s−1 at 20 °C and 350 μmol (CO2) mol−1(air). The values of P N were fitted to a rectangular hyperbolic nonlinear regression model. P N for all cultivars increased similarly with increasing PAR but Cascade and Oranza generally had higher P N than CC. None of the cultivars reached saturation at 1 000 μmol m−2 s−1. The predicted P N at saturation (P Nmax) for Cascade, CC, Oranza, and RCC were 19.78, 16.40, 19.79, and 18.11 μmol (CO2) m−2 s−1, respectively. The compensation irradiance (I c) occurred at 54 μmol m−2 s−1 for Cascade, 36 μmol m−2 s−1 for CC, 45 μmol m−2 s−1 for Oranza, and 25 μmol m−2 s−1 for RCC. The quantum yield among the cultivars ranged between 0.057–0.033 mol(CO2) mol−1(PAR) and did not differ. Dark respiration varied from 2.66 μmol m−2 s−1 for Cascade to 0.85 μmol m−2 s−1 for RCC. As P N increased with PAR, intercellular CO2 decreased in a non-linear manner. Increasing PAR increased stomatal conductance and transpiration rate to a peak between 600 and 800 μmol m−2 s−1 followed by a steep decline resulting in sharp increases in water use efficiency. Thailand border market is where the local Thais, Cambodians, Laotians, and Burmeses exchange their goods and culture at the border checkpoints. It is considered to be the source of aerial disease transmission especially for foreigners because it is always very crowded with people from all walks of life. Unhealthy air quality makes this area high risk of spread of airborne diseases. This study assessed airborne concentrations of bacteria and fungi in a border market to improve exposure estimates and develop efficient control strategies to reduce health risk. The density and distribution of airborne bacteria and fungi were investigated in the Chong Chom border market in Surin Province, Thailand. Eighteen air sampling sites were taken from outdoors and various work environments including indoor footpaths, wooden handicraft shops, electronic shops, the secondhand clothing shops, and fruit market areas. Exposed Petri plate method and liquid impinger sampler were used for sampling at the breathing zone, 1.5 m above the floor level, during weekend and holiday. Meteorological factors such as relative humidity, temperature, and light intensity were collected by portable data logger. The relative humidity was 67–73%, and temperature 29–33°C, and light varied between 18 and 270 Lux m−2. Gram-positive and Gram-negative bacteria were found at a mean value of 104 CFU m−3, and airborne fungi of 103 CFU m−3 were recorded. The highest concentration of culturable airborne microorganisms was found along the indoor footpath (9.62 × 104 CFU m−3 and 750.00 CFU/plate/h for impingement and sedimentation methods, respectively), the fruit market area (7.86 × 104 CFU m−3 and 592.42 CFU/plate/h for impingement and sedimentation methods, respectively), and the secondhand clothing shop (4.59 × 103 CFU m−3 and 335.42 CFU/plate/h for impingement and sedimentation methods, respectively) for Gram-positive bacteria, Gram-negative bacteria, and fungi, respectively. The lowest concentration of Gram-positive bacteria, Gram-negative bacteria, and fungi was found only at the outdoor area at 1.53 × 104 CFU m−3, 0.93 × 104 CFU m−3 and 0.80 × 103 CFU m−3 by means of impingement method and 136.67 CFU/plate/h, 69.25 CFU/plate/h, and 62.00 CFU/plate/h by means of sedimentation methods for Gram-positive bacteria, Gram-negative bacteria, and fungi, respectively. The most frequently present airborne bacteria were identified as Bacillus, Corynebacteria, Diplococcus, Micrococcus, Acinetobacter, Alcaligenes, Enterobacter, and spore former rods. Acremonium, Aspergillus, Cladosporium, Penicillium, and Sporotrichum were the most frequently found aerosol fungi genera. The distribution of airborne microorganisms correlated with relative humidity and light factors based on principal component analysis. In conclusion, the border market is a potential source of aerial disease transmission and a various hazards of bioaerosols for workers, consumers, sellers, and tourists. The bioaerosol concentration exceeded the standard of occupational exposure limit. Many major indicators of allergenic and toxigenic airborne bacteria and fungi, Acinetobacter, Enterobacter, Pseudomonas, Cladosporium, Alternaria, Aspergillus, and Penicillium, were found in the various market environments.  相似文献

Airborne bacteria are important biological components of bioaerosol and play an important role in ecosystem. Bacteria at a high concentration in the atmosphere can result in biological air pollution and all kinds of diseases. In this study, a systematical survey on the culturable airborne bacteria was carried out for 1 year at three sites in Beijing urban area. Results showed that concentrations of culturable bacteria ranged from 71 colony forming units (CFU)/m3 to 22,100 CFU/m3, and the mean was 2,217 CFU/m3. Bacterial concentrations at the human activity-enriched site (RCEES) and the highly trafficked site (XZM) were virtually the same point. They were significantly higher than those at the greener site (BBG). Significant variation in bacterial concentrations in different seasons was observed at RCEES and XZM with higher concentrations in summer and autumn. In a single day, significantly lower concentrations were detected at 13:00 hours through all sampling sites. In this study, 165 species in 47 genera of culturable bacteria were identified. Micrococcus was one of the most dominant bacterial groups and contributed to approximately 20∼30% of the total bacterial concentration, followed by Staphylococcus, Bacillus, Corynebacterium, and Pseudomonas. The bacterial species with a high concentration percentage included Micrococcus luteus and Micrococcus roseus.  相似文献

The Florida Everglades is a mosaic of short and long-hydroperiod marshes that differ in the depth, duration, and timing of inundation. Algae are important primary producers in widespread Everglades’ periphyton mats, but relationships of algal production and community structure to hydrologic variability are poorly understood. We quantified differences in algal biomass and community structure between periphyton mats in 5 short and 6 long-hydroperiod marshes in Everglades National Park (ENP) in October 2000. We related differences to water depth and total phosphorus (TP) concentration in the water, periphyton and soils. Long and short-hydroperiod marshes differed in water depth (73 cm vs. 13 cm), periphyton TP concentrations (172μg g−1 vs. 107 μg g−1, respectively) and soil TP (284 μg g−1 vs. 145 μg g−1). Periphyton was abundant in both marshes, with short-hydroperiod sites having greater biomass than long-hydroperiod sites (2936 vs. 575 grams ash-free dry mass m−2). A total of 156 algal taxa were identified and separated into diatom (68 species from 21 genera) and “soft algae” (88 non-diatom species from 47 genera) categories for further analyses. Although diatom total abundance was greater in long-hydroperiod mats, diatom species richness was significantly greater in short- hydroperiod periphyton mats (62 vs. 47 diatom taxa). Soft algal species richness was greater in long-hydroperiod sites (81 vs. 67 soft algae taxa). Relative abundances of individual taxa were significantly different among the two site types, with soft algal distributions being driven by water depth, and diatom distributions by water depth and TP concentration in the water and periphyton. Periphyton communities differ between short and long-hydroperiod marshes, but because they share many taxa, alterations in hydroperiod could rapidly promote the alternate community. Sampling was conducted from June 2007 to May 2008 in an enclosed rabbit house to investigate composition and variability of airborne fungi. Samples were collected using an Andersen-6 sampler, with Sabouraud culture medium as sampling medium. The results showed that monthly mean concentration was 2.79–5.46 × 103 colony forming unit/m3 air (CFU/m3 air), with the maximum level in October, and the minimum level in January. Within a day, the maximum level occurred at 09:00, followed by 17:00 and then 13:00. A total of 6,523 fungal colonies, belonging to 17 genera and 36 species, were obtained. The predominant genera included Cladosporium, Penicillium, Aspergillus and Altemaria, comprising 71.45% of the colony count. The obtained fungi of the year were mainly centralized in the stage D of the sampler (2.0–3.0 μm), accounting for 37.8% of the colonies. The minimum value occurred at stage F (<0.65 μm), accounting for 1.10% of the colonies.  相似文献

The purpose of the study was to determine the concentration of trace elements present in scalp hair sample of schizophrenic patients and to find out the relationship between trace elements level and nutritional status or socioeconomic factors. The study was conducted among 30 schizophrenic male patients and 30 healthy male volunteers. Patients were recruited from Bangabandhu Sheikh Mujib Medical University by random sampling. Hair trace element concentrations were determined by flame atomic absorption spectroscopy and analyzed by independent t test, Pearson’s correlation analysis, regression analysis, and analysis of variance (ANOVA). Mn, Zn, Ca, Cu, and Cd concentrations of schizophrenic patients were 3.8 ± 2.31 μg/gm, 171.6 ± 59.04 μg/gm, 396.23 ± 157.83 μg/gm, 15.40 ± 5.68 μg/gm, and 1.14 ± 0.89 μg/gm of hair sample, while those of control subjects were 4.4 ± 2.32 μg/gm, 199.16 ± 27.85 μg/gm, 620.9 ± 181.55 μg/gm, 12.23 ± 4.56 μg/gm, and 0.47 ± 0.32 μg/gm of hair sample, respectively. The hair concentration of Zn and Ca decreased significantly (p = 0.024; p = 0.000, respectively) and the concentration of Cu and Cd increased significantly (p = 0.021; p = 0.000, respectively) in schizophrenic patients while the concentration of Mn (p = 0.321) remain unchanged. Socioeconomic data reveals that most of the patients were poor, middle-aged and divorced. Mean body mass indices (BMIs) of the control group (22.26 ± 1.91 kg/m2) and the patient group (20.42 ± 3.16 kg/m2) were within the normal range (18.5−25.0 kg/m2). Pearson’s correlation analysis suggested that only Ca concentration of patients had a significant positive correlation with the BMI (r = 0.597; p = 0.000) which was further justified from the regression analysis (R 2 = 44%; t = 3.59; p = 0.002) and one-way ANOVA test (F = 3.62; p = 0.015). A significant decrease in the hair concentration of Zn and Ca as well as a significant increase in the hair concentration of Cu and Cd in schizophrenic patients than that of its control group was observed which may provide prognostic tool for the diagnosis and treatment of this disease. However, further work with larger population is suggested to examine the exact correlation between trace element level and the degree of disorder.  相似文献

Physical, biogeochemical and photosynthetic parameters were measured in sea ice brine and ice core bottom samples in the north-western Weddell Sea during early spring 2006. Sea ice brines collected from sackholes were characterised by cold temperatures (range −7.4 to −3.8°C), high salinities (range 61.4–118.0), and partly elevated dissolved oxygen concentrations (range 159–413 μmol kg−1) when compared to surface seawater. Nitrate (range 0.5–76.3 μmol kg−1), dissolved inorganic phosphate (range 0.2–7.0 μmol kg−1) and silicic acid (range 74–285 μmol kg−1) concentrations in sea ice brines were depleted when compared to surface seawater. In contrast, NH4 + (range 0.3–23.0 μmol kg−1) and dissolved organic carbon (range 140–707 μmol kg−1) were enriched in the sea ice brines. Ice core bottom samples exhibited moderate temperatures and brine salinities, but high algal biomass (4.9–435.5 μg Chl a l−1 brine) and silicic acid depletion. Pulse amplitude modulated fluorometry was used for the determination of the photosynthetic parameters F v/F m, α, rETRmax and E k. The maximum quantum yield of photosystem II, F v/F m, ranged from 0.101 to 0.500 (average 0.284 ± 0.132) and 0.235 to 0.595 (average 0.368 ± 0.127) in the sea ice internal and bottom communities, respectively. The fluorometric measurements indicated medium ice algal photosynthetic activity both in the internal and bottom communities of the sea ice. An observed lack of correlation between biogeochemical and photosynthetic parameters was most likely due to temporally and spatially decoupled physical and biological processes in the sea ice brine channel system, and was also influenced by the temporal and spatial resolution of applied sampling techniques.  相似文献

The number of airborne microorganisms in the area of large-scale composting facilities with different composting techniques (A: open facility using the intensive decomposition process [4000 t/year]. B: closed facility with compost containers [7000 t/year], C: closed facility with table-pile compositing and automatic turning equipment [22 000 t/year]) was investigated using impactor sampling systems (Andersen samplers). All counts carried out inside the closed facilities, especially during the turning process, showed values of >5.0 × 105 CFU/m3 for viable bacteria and moulds with a proportion ofAspergillus fumigatus of up to 64%. Depending on the type of facility, different median values were determined inside the plant area. Counts were highest in the immediate area around the biofilter outside of Facility C (1.7 × 104 CFU/m3 for bacteria and 9.5 × 103 CFU/m3 for moulds). In view of the high load of ambient airborne microorganisms inside the composting facilities, adequate occupational health measures are urgently required. Counts determining the hazard to neighbourhood residents at distances of between 150 and 2000 m showed, depending on the facility, annual median values of 170–330 CFU/m3 for bacteria, 75–340 CFU/m3 for moulds, and 15–52 CFU/m3 forA. fumigatus. Higher individual counts — up to 3 × 103 CFU/m3 for moulds and up to 350 CFU/m3 forA. fumigatus — were found as a result of specific climatic influences, (e.g. winds) and activities as well poor operation. Given the high proportion ofA. fumigatus in the exhaust air, this mould can serve as an indicator for the evaluation of the health risk. However, the maximum values found in the present study, may also be caused by other events in rural areas, (e.g. agricultural activities). With regard to neighbourhood residents, odour complaints are more important than pollution by microorganisms.  相似文献

Chlorophyll a and nutrient concentrations along with temperature and salinity values were measured at 22 CTD stations along a 735-km transect running to the northwest of the island of South Georgia, Southern Ocean. Measurements were repeated during five summer surveys (January and February 1994, January 1996, December 1996, January 1998) and one spring survey (October 1997). The transect sampled Sub-Antarctic Zone water in the north, Polar Frontal Zone water and Antarctic Zone water in the south. Chlorophyll a concentrations were lowest to the north of the transect and frequently high (up to 17 mg m−3) in the deep open ocean of the Antarctic Zone. Sub-surface peaks were measured in all zones and chlorophyll a was detectable to a depth of 150 m. There was a clear latitudinal temperature gradient in the near-surface waters (0–50 m), the warmest water occurring in the north (∼12 °C), and the coolest in the Antarctic Zone (∼2 °C). There was also a well-defined latitudinal gradient in summer near-surface silicate concentrations (∼2, 4, and 10 mmol m−3 in the Sub-Antarctic Zone, the Polar Frontal Zone and the Antarctic Zone, respectively), increasing to >20 mmol m−3 near South Georgia. Distinct differences in silicate concentrations were also evident in all three zones to a depth of 500 m. Near-surface nitrate and phosphate concentrations were relatively low to the north of the transect (∼14 and 1 mmol m−3, respectively) and higher in the Polar Frontal Zone and Antarctic Zone (∼18 and 1.4 mmol m−3, respectively). Ammonium and nitrite were restricted to the upper 200 m of the water column, and exhibited sub-surface concentration peaks, the lowest being in the Sub-Antarctic Zone (0.68 and 0.25 mmol m−3, respectively) and the highest in the Antarctic Zone (1.72 and 0.29 mmol m−3, respectively). Surface (∼6 m) spring nutrient measurements provided an indication of pre-bloom conditions; ammonium and nitrite concentrations were low (∼0.27 and 0.28 mmol m−3, respectively), while silicate, nitrate and phosphate concentrations were high and similar to previously measured winter values (e.g. ∼26, 23, 2 mmol m−3, respectively in the Antarctic Zone). Although the values measured were very variable, and there was some evidence of a seasonal growth progression, the chlorophyll a and nutrient distribution patterns were dominated by intercruise (interannual) factors. Approximate nutrient depletions (spring minus summer) appeared similar in the Polar Frontal Zone and Antarctic Zone for nitrate and phosphate, while silicate showed a marked latitudinal increase from north to south throughout the transect. Highest chlorophyll a concentrations coincided with the highest apparent silicate depletions over the deep ocean of the Antarctic Zone. In this area, relatively warm, easterly flowing Antarctic Circumpolar Current water meets cooler, westerly flowing water that is influenced by the Weddell-Scotia Confluence and is rich in nutrients, especially silicate. Increased bioaerosol loadings in downwind plumes from concentrated animal feeding operations (CAFOs) may increase the risk for allergy and infection in humans. In this study, we monitored airborne concentrations of culturable bacteria and fungi at upwind (background) and downwind sites at a 10,000 milking cow dairy over the course of a year. The average bacterial concentrations at the upwind site were 8.4 × 103 colony forming units (CFU) m−3 and increased to 9.9 × 105 CFU m−3 at the downwind edge of the cattle lots, decreasing to 6.3 × 104 CFU m−3 200 m farther downwind. At the same sites, the average fungal concentrations were 515, 945, and 1,010 CFU m−3, respectively. Significant correlations between the ambient weather conditions and airborne fungal and bacterial concentrations were identified. Sequence analysis of PCR-amplified DNA from bacterial clones and fungal isolates revealed genus and species level differences between upwind and downwind sites. Although we could not cultivate gram-negative bacteria, bacterial clones at downwind sites identified as being gram-negative matched with the following genera: Acinetobacter, Bradyrhizobium, Escherichia, Idiomarina, Methylobacterium, Ralstonia, and Novosphingobium. Fungal isolates from downwind matched with the following genera: Acremonium, Alternaria, Ascomycte, Aspergillus, Basidiomycete, Cladosporium, Davidiella, Doratomyces, Emericella, Lewia, Onygenales, Penicillium, Rhizopus, and Ulocladium. None of the bacterial and fungal sequence matches were affiliated with genera and species known to be pathogenic to humans. Overall, the data suggest that exposure to bioaerosols in the downwind environment decreases with increasing distance from the open-lot dairy.  相似文献

Effects of short-term ozone (O3) fumigation on radish (Raphanus sativus L.) plants were examined in growth chambers under controlled environment conditions. Plants were exposed to 0 μg/m3 (reference), 80 μg/m3, 160 μg/m3 and 240 μg/m3 O3 concentrations for 7 h per day for five days. Day/night temperature was 21°C/14°C and photoperiod 16 h. Chlorophyll content was evaluated spectrophotometrically. Chromatographic analysis of saccharides was also undertaken. The results showed that elevated O3 inhibited the growth of radish rhizocarps, net assimilation rate and biomass accumulation. O3 induced leaf desiccation, necrosis and premature senescence, but a typical reaction of plants to O3 stress was the rapid regeneration of new leaves. O3 inhibited accumulation of carotenoids more than chlorophylls. The higher photosynthetic pigment content in newly generated radish leaves may be regarded as an adaptation of the photosynthetic system to O3. Leaf saccharide metabolism and incorporation depended on O3 concentration. Rapid regeneration of new leaves and increased content of photosynthetic pigments is the typical reaction of radish plants to O3 stress.  相似文献
