Abstract

The content and composition of lipids and fatty acids of taproots and lateral roots of Vicia faba were investigated with plants grown under saline (80 mM NaCl) and non-saline conditions. Lipids of both types of faba bean roots are constituted of ~80% phospholipids, of ~15% glycolipids and of some 5% of wax-esters. Phosphatidylethanolamine (PE) and phosphatidylcholine are the main constituents of both root types. Di-phosphatyidylglycerol and phosphatidylinositol are present in medium concentrations, whereas phosphatidylglycerol and phosphatidylserine are present only in trace amounts. The content of sulpholipids was much lower in salt-treated roots as compared with that of the controls. The content of cardiolipins and PE was higher in lateral roots than in the taproots. Roots of salt-treated plants had some 18% lower ether-soluble lipid content in the lateral roots, and approximately 28% lower than the taproots. Less than 25% of the fatty acids of the extracted phospholipids were saturated, with palmitic acid as the main constituent (13 – 18%). Linoleic acid comprised 65 – 70% of the unsaturated acids. Differences in the composition of some fatty acids were found between taproots and lateral roots of salt-treated plants. The observed differences in composition between root types suggests that the reported physiological differences between such roots could be based, at least in part, on structural or compositional differences in their lipids.  相似文献

Arabidopsis plants responding to phosphorus (P) deficiency increase lateral root formation and reduce primary root elongation. In addition the number and length of root hairs increases in response to P deficiency. Here we studied the patterns of radical oxygen species (ROS) in the roots of Arabidopsis seedlings cultured on media supplemented with high or low P concentration. We found that P availability affected ROS distribution in the apical part of roots. If plants were grown on high P medium, ROS were located in the root elongation zone and quiescent centre. At low P ROS were absent in the elongation zone, however, their synthesis was detected in the primary root meristem. The proximal part of roots was characterized by ROS production in the lateral root primordia and in elongation zones of young lateral roots irrespective of P concentration in the medium. On the other hand, plants grown at high or low P differed in the pattern of ROS distribution in older lateral roots. At high P, the elongation zone was the primary site of ROS production. At low P, ROS were not detected in the elongation zone. However, they were present in the proximal part of the lateral root meristem. These results suggest that P deficiency affects ROS distribution in distal parts of Arabidopsis roots. Under P-sufficiency ROS maximum was observed in the elongation zone, under low P, ROS were not synthesized in this segment of the root, however, they were detected in the apical root meristem.  相似文献

Abstract

This paper investigates the modification of root architecture of Spartium junceum L. seedlings grown in slope condition. It is reported that 50% of the total number of lateral roots are concentrated in few centimetres of the taproot near the collar. The anatomical analysis of transverse sections along the taproot axis reveals that this taproot zone is characterised by two types of lateral roots: one with a trace extending to the centre of the vascular cylinder by following the path of a medullar ray; one with a trace which ends in the vascular cambium. The first type may be lateral roots originated from the taproot primary structure; the second type seems to be lateral roots developing later when a secondary structure has completely substituted the primary structure. The emission of this second type of lateral roots seems to be strongly controlled by environmental conditions with considerable consequences upon the overall root architecture. In the example reported in this paper, young plants growing under mechanical stress due to a slope develop asymmetric root architecture with lateral roots elongating in two prevalent directions: up-slope or down-slope. This asymmetric architecture is produced in the zone of the taproot where a secondary structure is present and represents the plant response to the need of increasing its anchorage strength.  相似文献

Abstract

Our study was carried out in bench rhizotrons using the Camarosa variety of strawberry (Fragaria × ananassa Duch.), by exciding the apex of fast-growing primary roots at two distances (1 or 8 cm) from the apex. It was demonstrated that new lateral meristems were rapidly induced by excision of the root apex at either distance; after 24 h, histone H4 in situ hybridisation detected groups of cell-organising root primordia just a few millimetres below the cut. After a further 24 h, new lateral roots were about to protrude from the original root. Results show that lateral roots can be formed anywhere along the primary roots of strawberry plants from a few stem cells distributed along the pericycle close to the protoxylem arches.  相似文献

Root exudate composition and quantity vary in relation to plant nutritional status, but the impact of the differences on rhizosphere microbial communities is not known. To examine this question, we performed an experiment with barley (Hordeum vulgare) plants under iron-limiting and iron-sufficient growth conditions. Plants were grown in an iron-limiting soil in root box microcosms. One-half of the plants were treated with foliar iron every day to inhibit phytosiderophore production and to alter root exudate composition. After 30 days, the bacterial communities associated with different root zones, including the primary root tips, nonelongating secondary root tips, sites of lateral root emergence, and older roots distal from the tip, were characterized by using 16S ribosomal DNA (rDNA) fingerprints generated by PCR-denaturing gradient gel electrophoresis (DGGE). Our results showed that the microbial communities associated with the different root locations produced many common 16S rDNA bands but that the communities could be distinguished by using correspondence analysis. Approximately 40% of the variation between communities could be attributed to plant iron nutritional status. A sequence analysis of clones generated from a single 16S rDNA band obtained at all of the root locations revealed that there were taxonomically different species in the same band, suggesting that the resolving power of DGGE for characterization of community structure at the species level is limited. Our results suggest that the bacterial communities in the rhizosphere are substantially different in different root zones and that a rhizosphere community may be altered by changes in root exudate composition caused by changes in plant iron nutritional status.  相似文献

Abstract

Pseudomonas fluorescens strains which are proven biocontrol agents in black pepper against foot rot (caused by Phytophthora capsici ) were also found to enhance root proliferation and fibre root production. Experiments conducted in the greenhouse with five efficient strains of P. fluorescens (IISR-6, IISR-8, IISR-11, IISR-13 and IISR-51) showed that the bacterial strains could significantly increase the root biomass of the plants (30 – 135%). Parameters for total root length, root area and root tips were estimated by scanning the entire root system and analysis through GS Root® software (PP systems, Winterstreet, USA). All the strains increased the root length in the treated plants (12 – 127%), the highest being with IISR-6, which was on a par with IISR-11 and IISR-51. A similar trend was observed with the total root area after bacterization (43 – 200%). The P. fluorescens treated plants had a higher number of feeder roots as evidenced by the increased number of root tips (82 – 137%). The enhanced growth parameters upon root bacterization could be corroborated with the production of the plant growth hormones IAA & GA by the bacterial strains and their P-solubilization potential.  相似文献

Strains ofKlebsiella pneumoniae, Klebsiella terrigena, Enterobacter agglomerans andAzospirillum lipoferum were compared as diazotrophic inoculants in association withPoa pratensis andTriticum aestivum. Each strain colonized both plants in numbers ranging from 104 to 107 bacteria per root, and electron microscopy and immunofluorescence staining of inoculated roots revealed bacteria mainly on root hairs. Indirect immunofluorescence with specific antifimbriae antibodies showed that the enteric bacteria expressed their fimbria in both associations. All associations were positive in an acetylene reduction test but only in half of them was atmospheric nitrogen transferred to the plant. In the inoculated plants, variable effects in the dry matter and N yields in both hosts were observed and no correlation was found between dry matter, nitrogen content or the amount of fixed nitrogen. In infected plants, the number of root hairs and lateral roots increased and the length of the zone of elongation decreased. The changes in root morphology were more evident in associations with enteric bacteria than with Azospirillum. The results give further evidence on the importance of bacterial adhesion in associative N2 fixation and suggest that bacteria-induced physiological changes in plant roots may be more important than the amount of nitrogen transferred to the plant.  相似文献

Root growth in Lupinus angustifolius is greatly decreased when the nutrient solution has a pH above 6.0. This study examined the water relations in this species (cv. Yandee) in response to high pH in solution culture in a glasshouse.The dry weight of roots, the length of taproots and lateral roots and the number of lateral roots were significantly reduced at day 12 after transfer to solution with a pH of 7.5 compared to pH 5.2. This resulted in a marked reduction of total root surface area. However, shoot growth and total leaf area were not affected. In comparison with pH 5.2, plants grown at pH 7.5 in the nutrient solution had a 14–38% more-negative leaf water potential, and their stomatal resistance had increased by 67%.The observations indicate that the impairment of the water relations by high pH is mainly caused by decreased root growth.  相似文献

Protozoan communities around roots with different types of ectomycorrhizae were distinct. These protozoan communities differed both qualitatively and quantitatively with the host (Pinus ponderosa, Pseudotsuga menziesii, Picea sitchensis, Tsuga heterophylla and Abies grandis) and the ectomycorrhizal fungal species. Based on the species identified and the numbers of individuals of each species, six communities of protozoa were found associated with specific ectomycorrhizae. Previous researchers have shown that mycorrhizal colonization of roots alters the amounts and types of exudates produced by roots, which in turn alters the bacterial community present. Most likely, mycorrhizal colonization of roots influences the protozoan community around roots by controlling the bacterial community. However, the protozoan community may in turn influence the successional dynamics of ectomycorrhizal fungi on different host root systems by a variety of mechanisms. These mechanisms could include: (1) preying upon individuals and perhaps removing particular species of bacteria from the mycorrhizosphere; and (2) controlling nitrogen mineralization in the rhizosphere. Further work needs to be performed to determine the interaction between these quadrate (plant-bacteria-fungi-protozoa) associations.  相似文献

Is it possible to manipulate root anchorage in young trees?   总被引:1,自引:1,他引:0  
The optimal root system architecture for increased tree anchorage has not yet been determined and in particular, the role of the tap root remains elusive. In Maritime pine (Pinus pinaster Ait.), tap roots may play an important role in anchoring young trees, but in adult trees, their growth is often impeded by the presence of a hard pan layer in the soil and the tap root becomes a minor component of tree anchorage. To understand better the role of the tap root in young trees, we grew cuttings (no tap root present) and seedlings where the tap root had (?) or had not (+) been pruned, in the field for 7 years. The force (F) necessary to deflect the stem sideways was then measured and divided by stem cross-sectional area (CSA), giving a parameter analogous to stress during bending. Root systems were extracted and root architecture and wood mechanical properties (density and longitudinal modulus of elasticity, E L ) determined. In seedlings (?) tap roots, new roots had regenerated where the tap root had been pruned, whereas in cuttings, one or two lateral roots had grown downwards and acted as tap roots. Cuttings had significantly less lateral roots than the other treatments, but those near the soil surface were 14% and 23% thicker than plants (+) and (?) tap roots, respectively. Cuttings were smaller than seedlings, but were not relatively less resistant to stem deflection, probably because the thicker lateral roots compensated for their lower number. Apart from stem volume which was greater in trees (+) tap roots, no significant differences with regard to size or any root system variable were found in plants (?) or (+) tap roots. In all treatments, lateral roots were structurally reinforced through extra growth along the direction of the prevailing wind, which also improved tap root anchorage. Predictors of log F/CSA differed depending on treatment: in trees (?) tap roots, a combination of the predictors stem taper and %volume allocated to deep roots was highly regressed with log F/CSA (R 2 = 0.83), unlike plants (+) tap roots where the combined predictors of lateral root number and root depth were best regressed with log F/CSA (R 2 = 0.80). In cuttings, no clear relationships between log F/CSA and any parameter could be found. Wood density and E L did not differ between roots, but did diminish with increasing distance from the stem in lateral roots. E L was significantly lower in lateral roots from cuttings. Results showed that nursery techniques influence plant development but that the architectural pattern of Maritime pine root systems is stable, developing a sinker root system even when grown from cuttings. Anchorage is affected but the consequences for the long-term are still not known. Numerical modelling may be the only viable method to investigate the function that each root plays in adult tree anchorage.  相似文献

Summary. Viral invasion of the root system of Nicotiana benthamiana was studied noninvasively with a tobacco mosaic virus (TMV) vector expressing the green-fluorescent protein (GFP). Lateral root primordia, which developed from the pericycle of primary roots, became heavily infected as they emerged from the root cortex. However, following emergence, a progressive wave of viral inhibition occurred that originated in the lateral-root meristem and progressed towards its base. Excision of source and sink tissues suggested that the inhibition of virus replication was brought about by the basipetal movement of a root meristem signal. When infected plants were inoculated with tobacco rattle virus (TRV) expressing the red-fluorescent protein, DsRed, TRV entered the lateral roots and suppressed the host response, leading to a reestablishment of TMV infection in lateral roots. By infecting GFP-expressing transgenic plants with TMV carrying the complementary GFP sequence it was possible to silence the host GFP, leading to the complete loss of fluorescence in lateral roots. The data suggest that viral inhibition in lateral roots occurs by a gene-silencing-like mechanism that is dependent on the activation of a lateral-root meristem. Trifolium tomentosum and T. glomeratum are small (< 0·5 mg) seeded pasture legumes which are considered to be waterlogging tolerant and intolerant, respectively. The root porosity of the two species was compared for plants raised for 10 d in aerated nutrient solution and then transferred to either aerated (0·25 mol O2 m–3) or ‘hypoxic’ (0·031–0·069 mol O2 m–3) solutions for a further 7 and 21 d. After 21 d, T. tomentosum developed a root porosity of 11·2% in ‘hypoxic’ solution, which was significantly higher than the 6·1% developed by T. glomeratum. When grown in aerated solution, T. tomentosum also had a larger constitutive porosity (6·7%) than T. glomeratum (3·9%). Cylindrical root-sleeving O2 electrodes were used to measure the rates of radial O2 loss (ROL) from roots of the two species when in an O2-free medium. In general, roots previously grown in ‘hypoxic’ solution had higher rates of ROL than roots grown in aerated solution. Moreover, the rates of ROL along the main root of T. tomentosum were ≈ 5-fold faster than from equivalent locations along roots of T. glomeratum. Manipulations of the shoot O2 concentration resulted in rapid changes in ROL near the root tip of T. tomentosum plants raised in aerated or ‘hypoxic’ solutions, whereas for T. glomeratum ROL only increased for roots of plants raised in ‘hypoxic’ solution. Thus, the cortical air spaces in roots of both species raised in ‘hypoxic’ solution formed a continuous, low resistance pathway for O2 diffusion from the shoots to the roots. ROL from the lateral roots was also evaluated and it was 3-fold faster from T. tomentosum than from T. glomeratum. Moreover, ROL from lateral roots of T. tomentosum was 10–20-fold higher than from a position on the primary root axis the same distance from the root/shoot junction. Relatively, high rates of ROL were also recorded for young (40 mm in length) lateral roots of T. glomeratum which were previously grown in ‘hypoxic’ solution, but the ROL was low for the older lateral roots of this species. The substantial amounts of ROL from the lateral roots may limit O2 supply to the lower parts of the primary root axis, so that the laterals probably become the main functional root system in waterlogged soils.  相似文献

The effect of root temperature on growth and yield of rockwool-grown tomato plants infected with Phytophthora cryptogea was investigated. Measurements of shoot and root growth were taken at high (25oC) and low (15oC) root temperatures during the generative phase of growth. The growth of roots of healthy and P. cryptogea-infected tomato plants in rockwool blocks was higher in plants grown with roots at 25oC than at 15oC after 60 days and a similar effect was found in slabs after 98 days. Under sub-optimal conditions for growth the disease became severe when root temperatures were low. Growth of roots was greatest when roots were maintained at a high temperature in combination with an ambient air temperature of c. 15oC and the response was greater in cv. Counter than cvs Calypso and Marathon. Water-soluble carbohydrates of roots were higher in those produced in blocks than slabs and were reduced by infection compared to healthy plants with roots at 15oC and 25oC. Reduced transpiration rates were found 17 days after inoculation in symptomless plants grown at a root temperature of 25oC. Infection, regardless of the temperature of the roots or cultivar, led to reduced stem growth. The plants grown at 25oC were taller than those with a root temperature of 15oC. After 9 wk of harvest, the cumulative fruit yields in infected cvs Counter and Calypso grown at 25oC were comparable to that in healthy plants grown at either temperature and cumulative fruit numbers followed a similar pattern. High root temperatures led to delayed fruit ripening between weeks 3–10 and a larger number of unripe fruit. The weight of unripe fruit from infected plants grown at 25oC at the terminal harvest was higher than from healthy plants with roots maintained at 15oC.  相似文献

The effect of direct inoculation of seeds with the plant growth promoting rhizobacteria (PGPR) Azospirillum lipoferum CRT1 was assessed on maize (Zea mays) grown for 35 days after sowing (d.a.s.) in controlled conditions (greenhouse) in a luvisol soil from south-eastern France. WhinRhizo® software was used to describe the following changes in the root system morphology for each plant: distribution and average root diameter, root surface and the number of tips. The stress at breakage and stiffness of the roots in tension were also determined. Evaluation of biochemical components of roots was achieved by direct Attenuated Total Reflectance (or reflection) (ATR)-Fourier transform infrared (FTIR) on root section. Inoculated roots exhibited significantly larger numbers of tips and extending surface to rhizosphere when compared to controls. Measured mechanical parameters of inoculated roots showed a slight increase in rupture stress up to the largest diameter (1.2 mm) when compared to controls. Stiffness (Young’s modulus) values were nearly constant for inoculated plants with higher values than for non-inoculated plants at day 26 and day 35. Using Principal Components Analysis of ATR-FTIR profiles, the polysaccharide enrichment of inoculated roots compared to controls was found at day 35. Noticeable absorbance at wavenumber specific to aromatic ether (lignin) was observed in control plants. All these data had a pattern of immature root properties, when maize was inoculated with Azospirillum lipoferum CRT1. Observed modifications of root development are possibly conducive to unseen beneficial effects, like water retention, resistance to mechanical stress, or root litter quality. Studies on more mature plants are required to assess if the differences between inoculated and control plants would persist or become accentuated with time until harvest.  相似文献

The effects of salt stress on endophytic prokaryotic communities in plants are largely unknown, and the distribution patterns of bacterial and archaeal endophytes in different tissues of a plant species are rarely compared. We investigated the endophytic bacterial and archaeal communities in roots, stems and leaves of the common reed, Phragmites australis, collected from three tidal zones along a salinity gradient, using terminal restriction fragment (T-RF) length polymorphism analysis of the 16S rRNA genes. The results showed that the bacterial diversity in the roots was significantly higher than that in the leaves, whereas similar archaeal diversity was revealed for either plant tissues or tidal zones. Network analysis revealed that T-RFs were grouped largely by tissue, and the major groups were generally linked by a few common T-RFs. Unique T-RFs in roots were mainly present in plants growing in the supratidal zone, but unique T-RFs in stems and leaves were mainly present in those from the middle and high tidal zones. Non-metric multidimensional scaling ordination and analysis of similarity revealed that bacterial communities were significantly different among tissues (P < 0.05), but similar among tidal zones (P = 0.49). However, the archaeal communities differed among tidal zones (P < 0.05), but were similar among tissues (P = 0.89). This study indicates that: (1) the endophytic archaeal communities are influenced more significantly than the endophytic bacterial communities by soil salinity, and (2) the differential distribution patterns of bacterial and archaeal endophytes in plant tissues along a salinity gradient imply that these two groups play different roles in coastal hydrophytes.  相似文献

【背景】植物内生菌长期与宿主共生,对宿主生长发育产生影响。葛根作为重要的药食两用作物,葛根内生菌的研究具有重要实践意义。【目的】对广西葛根根部内生细菌进行分离、鉴定及促植物生长特性分析,旨在了解该药食同源植物内生细菌种群结构及其促生特性,为分析内生菌群体在药食同源植物产量和品质形成的作用及其内生细菌资源的开发利用提供参考。【方法】采用6种不同的培养基从广西葛根的根瘤、根系和根愈伤组织分离内生细菌,16S rRNA基因测序和系统发育分析内生细菌的分布特征和遗传多样性,采用生理生化方法测定分离菌株的固氮活性、溶磷特性、产生嗜铁素、分泌吲哚乙酸(indole-3-aceticacid,IAA)等促生特性。【结果】从葛根根瘤、根系和根部愈伤组织中共分离得到223个菌株,16S rRNA基因测序鉴定这些菌株隶属于2门4纲10科19属,其中芽孢杆菌属、假单胞菌属、土壤杆菌属、肠杆菌属为葛根优势菌群;内生细菌数量和群落组成存在明显的组织特异性,其数量表现为根瘤>根系>根愈伤组织,但其种群多样性表现为根愈伤组织>根系>根瘤。不同培养基分离出的细菌种群丰富度有差异。从供试菌株中筛...  相似文献

The effects of Cd on the growth and Cd uptaking in corn (Zea mays L.) were explored under different Cd stress. The results showed that no reduction in shoot and root dry matter yields were noted when the plants were grown at Cd supply levels ≤100 μmol l?1 nutrient solution. The Cd concentration in the shoots and roots of corn increased sharply with increasing external Cd supply levels, peaked at 50 μmol l?1, and then decreased slowly with further increasing Cd levels due to high Cd toxic effects on root growth. The concentrations of chlorophyll a, chlorophyll b and chlorophyll a + b in the leaf of corn decreased slowly with increasing external Cd supply levels. Proline concentrations of corn increased when the plants were grown under the external Cd influence. The lower concentration of Cd treatments did not influenced the growth of corn significantly, and increased the uptake of Cd, the higher levels of Cd supply caused significantly physiological resposes and decreased the Cd uptaking.  相似文献

Abstract

It has yet to be established whether or not root architecture results from a metameric organization similar to that recognizable in the stem. To address this question, we have reviewed the data on the major cytological, histological and anatomical events underlying root development and on the intrinsic factors controlling these events. The evidence emerging from this review indicates that root architecture has a metameric organization that can be ‘deranged’ when environmental factors interfere with the intrinsic factors rhythmically controlling lateral root development. Metameric patterning occurs in the primary body of a root, but not in the secondary body. This difference can be attributed to the fact that primary and secondary body roots arise from completely different tissues. The root system of a woody plant is very complex, and its architecture is largely governed by roots with a secondary body organization that lack metameric patterning. The architectural contribution of the portion of roots with a primary body organization, where metameric patterning could be present, is negligible. This explains why it is difficult to recognize metameric patterning in the root architecture of these plants.  相似文献

Summary This study examined the effects of aggregate size on root impedance and developed an equation to describe the root pressure necessary to avoid deflection around an aggregate. This critical root pressure was predicted to increase with increasing aggregate size, decreasing root diameter, and decreasing deflection angle. In growth chamber experiments, maize (Zea mays L.) seedlings were grown in A horizon material of Groseclose silt loam (Clayey, mixed, mesic, Typic Hapludult). The soil had been moist sieved into different aggregate sizes (0–1, 1–2, 2–3, and 3–6 mm diameter). The larger aggregates did constitute a slight root impedance as roots were deflected around them. Diameters of roots grown in 3–6 mm aggregates increased significantly, whereas root lengths were not always signficantly decreased. The smaller aggregates did not impede root growth and were readily displaced by roots. Large aggregates were more of an impedance to lateral roots than to main axes.  相似文献
