The preferences of some woodland understorey species for ammoniumand nitrate were investigated by measuring the potential nitrification(conversion of ammonium to nitrate) in the rhizosphere comparedwith the bulk soil. Less acid-tolerant species, which usuallyprefer nitrate or a mixture of ammonium and nitrate in hydroponicculture, should have a higher potential nitrification in therhizosphere compared to the bulk soil due to a low uptake ofammonium (since ammonium is relatively immobile). Acid-tolerantspecies should have a high uptake of ammonium and thereby loweror equal potential nitrification in the rhizosphere comparedto the bulk soil. The hypothesis was tested in a field investigationof five understorey herb species, Deschampsia flexuosa, Convallariamajalis, Poa nemoralis, Geum urbanum andAegopodium podagrariaperformed in oak forests in southern Sweden. Overall, the twoless acid-tolerant species, Geum urbanum and Aegopodium podagraria,had high potential nitrification in the rhizosphere comparedto the bulk soil (indicating a relatively low uptake of ammonium),whilst the acid tolerant species, Deschampsia flexuosa andConvallariamajalis , had approximately equal potential nitrification inthe rhizosphere compared to the bulk soil (indicating a relativelyhigh uptake of ammonium). In the case of Poa nemoralis, a specieswhich grows in both acid and less acid soils, we found the potentialnitrification in the rhizosphere and in the bulk soil to besimilar at low inorganic nitrogen concentrations, but the difference(rhizosphere > bulk) increased when nitrification in thebulk soil was enhanced (i.e. when the nitrogen availabilityincreased). The potential nitrification in the bulk soil variedbetween 0 and 16 nmol g-1h-1and was positively correlated withpH. The chromosomal DNAs of nine strains of seven Bacteroides speciesincluding B. fragilis, the type species of the genus Bacteroides,were digested with rare-cutting restriction enzymes I-Ceu I,Not I, and Asc I and analysed by pulsed-field gel electrophoresis.The genome sizes of B. fragilis, B. distasonis, B. eggerthii,B. ovatus, B. thetaiotaomicron, B. uniformis, and B. vulgatuswere determined to be 5.3, 4.8, 4.4, 6.9, 4.8, 4.6, and 5.1Mbp, respectively. B. distasonis and B. vulgatus, and also B.uniformis and B. eggerthii, showed simillar I-Ceu I restrictionprofiles. I-Ceu I cut B. uniformis and B. eggerthii genomesinto four, B. ovatus into five, B. fragilis and B. thetaiotaomicroninto six, and B. distasonis and B. vulgatus into seven fragments.On the basis of genome size, restriction profile, and I-CeuI fragment number, a phylogenetic tree of the Bacteroides specieswas proposed. This was in overall agreement with the previousphylogenetic tree obtained by 16S rRNAdata, with the exceptionsof B. distasonis and B. ovatus.  相似文献

Fructifications of basidiomycetes have been collected at LakenheathWarren, and thirty-nine species have been obtained from fivegrassland soils. The different soils have been found to havedifferent species growing upon them and most species occur onthe alkaline-slightly acid grasslands. Highly acid grasslandat Lakenheath has a poor fungus flora. Studies have been made of the regions in the soil in which themycelia of certain of these basidiomycetes occur, and attemptshave been made to isolate these fungi from soil. Some specieshave been found to inhabit the litter zone rather than the minerallayers of the soil. A few form extensive rhizomorph systemswhich may penetrate deeply into the soil. A few species of ringfungi, such as Marasmius oreades, Psalliota arvensis, and Tricholomanudum, have been found to possess a well-defined mycelial zonein the soil, from which the fungus could be isolated. Profile studies have shown that the mycelial zones of thesespecies contain a restricted population of microfungi, bothfewer species and fewer colonies, as compared with the normalsoil around. Ascomycetes, such as Arachniotus, Chaetomium, Gymnoascus,and Penicillium, have been isolated more frequently from mycelialzones than from normal soil.  相似文献

Three species of forage grasses (Festuca arundinacea, Eragrostiscurvula, Sporobolus stapfianus) commonly grown in the Mediterraneanregion were subjected to a soil drying treatment. Leaf growthrate in F. arundinacea was highly sensitive to soil drying andlow growth rates were associated with high laminar turgors.The production of ABA was stimulated by soil drying and therewas a clear relation between increasing ABA accumulation andreduction in leaf growth. Leaf growth of E. cutvula, a C4 warmseason grass, was relatively insensitive to soil drying whichwas not accompanied by a substantial increase in leaf ABA content.S. stapfianus, a resurrection plant, was highly sensitive todecreasing soil water availability. In these two latter species,leaf growth was substantially restricted before ABA accumulationoccurred. It is suggested that reductions in laminar turgorof E. curvula and S. stapfianus may be limiting leaf growthas soil dries. The results indicated a different mechanism ofsensing and responding to reduction in soil water availabilityfor the three species studied. The relative importance of thechemical and hydraulic control of leaf growth is discussed. Variation in genome size of 24 populations belonging to 11 NWPatagonian species of Berberis was analysed as a function ofthe environment and geographical location. The variation showedthree levels of discontinuity, two of which corresponded todiploid species (2 n = 28) while the third corresponded to polyploidspecies (2 n = 56). Diploids with DNA content ranging from 1.463pg to 1.857 pg includedBerberis cabrerae , B. chillanensis,B. montana, B. serrato-dentata and B. bidentata. Diploids withDNA content ranging from 2.875 pg to 3.806 pg included B. linearifolia,B. darwinii, B. parodii and B. empetrifolia. The genome sizeof the polyploid species B. buxifolia and B. heterophylla rangedfrom 5.809 pg to 6.844 pg. Principal component analysis (PCA)was applied to represent the variability of environmental conditions.The eigenvectors of the principal component axes showed thatPC1 discriminates the populations according to rainfall, typesof vegetation and geomorphology; altitude and latitude, on theother hand, contribute to PC2 and PC3, respectively. From theseresults it is concluded: (1) that diploids with lower DNA contentgrow in high-elevation sites having greater rainfall but lowerwater availability; (2) diploids with higher DNA content areassociated with half-elevation forests where the vegetativeperiod is longer, the water availability is greater and thetemperatures are higher; and (3) the distribution pattern ofpolyploids is considerably wider than that of diploids, whichare geographically and ecologically restricted to forest areas.These results suggest that the C-value plays an important rolein the ability of the species to adapt to different growingconditions. Allozyme comparisons were made for adult Bulinus forskalii,B. camerunensis, and B. senegalensis from populations in Cameroonusing starch gel electrophoresis. Fifteen loci were examinedbut no differences were found between B. forskalii and B. camerunensis,bringing into question the validity of B. camerunensis as aseparate species. Bulinus senegalensis differed from B. forskaliiand B. camerunensis in allozymes for acid phosphatase, alpha-glycero-phosphatedehydxogenase, hydroxybutyrate dehy-drogenase, and phosphoglucomutase.No differences were found, as reported elsewhere, between B.forskalii and B. senegalensis for aspartate amino-transferase,isocitrate debydrogenase, phosphogluco-isomerase, and xanthineoxidise. No polymorphism was seen in B. camerunensis or variousB. senegalensis populations examined. Three alleles for isocitratedehydrogenase were observed for B. forskalii. Biomphalaria tenagophila tenagophila, B. tenagophila guaibensisand B. occidentalis are indistinguishable on the basis of shellmorphology and the majority of organs of the genital system.Only B. t. tenagophila is susceptible to infection with Schistosomamansoni. The identification of this species is important forepidemiological studies of schistosomiasis. Snails from differentsites in Brazil, Argentine and Uruguay were studied using thePolymerase Chain Reaction (PCR) and Restriction Fragment LengthPolymorphism (RFLP) of the Ribosomal RNA Internal TranscribedSpacers (ITS) using seven enzymes. Profiles resulting from digestionwith AluI showed some invariant species-specific products allowingcorrect identification of B. t. tenagophila, B. occidentalisand B. t. guaibensis. Profiles obtained with other enzymes didnot permit species identification as extensive intraspecificpolymorphism or invariant RFLP profiles were observed. Restriction profiles obtainedwith all enzymes were used to calculate the percentage of bandsharing between all individual snails and these data were usedfor a cluster analysis. A closer relationship between B. occidentalisand B. t. guaibensis than B. t. tenagophila and the subspeciesB. t. guaibensis was observed. Based on previous morphologicaldata and these molecular data, we propose grouping B. t. tenagophila,B. occidentalis and B. t. guaibensis into a B. tenagophila complex. Veronica spicata and Phleum phleoides are calcicole plants,mainly occurring on neutral or alkaline soil. An experimentof 16 weeks duration was performed in a glasshouse with theobjective of elucidating the influence of soil moisture levelon soil solution chemistry, and biomass concentrations and uptakeof mineral nutrients by the plants. Seven levels of moisture,corresponding to 35–85% of the water holding capacity(WHC) of the soil, were tested. Soil solution HCO3, P and Mnconcentrations, and pH, increased, whereas Ca, Mg and Zn concentrationsdecreased, with increasing soil moisture. Concentrations ofK were highest at 50–70% WHC. Concentrations and amountsof P, Zn and Mn in the two species were usually related to soilsolution concentrations; these are elements with low solubilityand availability in calcareous soils. Concentrations of nutrientsin biomass were more influenced by soil moisture in V. spicatathan inP. phleoides . This indicates that P. phleoides is morecapable of controlling its uptake of mineral nutrients, whereasV. spicata is sensitive to variations in soil moisture. It isconcluded that variation in soil moisture regime may greatlyinfluence concentrations of mineral nutrients in calcareoussoil solutions and their uptake by plants. Species able to utilizethese solubility fluctuations may have an advantage in competitionfor nutrients. Variation in soil moisture content might evenbe a prerequisite for adequate acquisition of mineral nutrientsand growth of plants on limestone soils, thereby influencingthe field distribution of native plants among habitats. Bacillus licheniformis was able to utilize gluconate as thesole carbon source as efficiently as Bacillus subtilis did.Southern analysis indicated that B. licheniformis likely possessesonly one gnt determinant. The nucleotide sequence (6278 bp)of the B. licheniformis DNA containing the gnt operon was determined,revealing the five complete open reading frames (ORF; genes).The putative product of the first gene, oug, did not show anysignificant homology to known proteins, but those of the secondto fifth genes exhibited striking homology to the gntRKPZ genesof B. subtilis, respectively, indicating that they are the correspondinggnt genes of B. licheniformis. Not only is the organizationof the gnt genes of these two Bacilli highly conserved, butso are the cis regulatory elements of their gnt operon. Sequenceanalysis of the upstream regions of these two gnt operons impliedthat a chromosome rearrangement in B. subtilis might have occurredimmediately upstream of the gnt operon during evolution, causingit to diverge from a common ancestor into B. licheniformis andB. subtilis.  相似文献

The relative abilities of freshwater calanoid copepods to grazealgae and filamentous cyanobacteria were examined in three speciesof Boeckella (B.triarticulata, B.hamata, B.dilatata) fed Cydotella,Cryptomonas and two species of Nostoc and Anabaena. Althoughrates of ingestion were highest on Cryptomonas, all the Boeckellaspecies consumed cyanobacteria, some species at similar ratesto Cyclolella. Faecal production on the foods was highest onCydotella and, in B.triarticulata, higher on A.flos-aquae andNostoc sp. 2 than on Cryptomonas. At 1 mg l–1 of thesecyanobacteria, female B.triarticulata voided 70% by volume ofthe material they ingested as compact faecal pellets. By theirgrazing they also decreased dramatically the trichome lengthsof A.flos-aquae, A.oscillarioides and Nostoc sp. 2, therebypossibly altering the potential susceptibility of these filamentouscyanobacteria to grazing by other zooplankton.  相似文献

Studies in recent years on chromosome number in freshwater snailsbelonging to the Bulinus truncatus/tropicus complex demonstratedthe occurrence in Kenya of one diploid species, B. tropicus,and 2 tetraploid species, B. truncatus and B. permembranaceus.Further observations are now described on the morphologies ofthe tetraploid species, and their distributions. Observations were made on B. truncatus from 26 localities (11newly reported) and B. permembranaceus from 26 localities (7newly reported), in respect of chromosome number, egg proteins,enzymes (5 systems), shell (10 variables measured), male genitalsystem (normal or aphallic) and radula (1st lateral tooth, sizeand mesocone shape). B. permembranaceus differs most clearly from B. truncatus inegg proteins, enzyme-loci and in lacking aphallic individuals.Mesocone shape is generally less angular in B. permembranaceus.The shell of B. permembranaceus grows larger, has the columellamore commonly concave and its spire is proportionally higherand more acute. To distinguish these tetraploid species fromthe partly sympatric B. tropicus, observations on chromosomenumber and biochemical characters are necessary. The tetraploids are allopatric, B. truncatus being found rarelyas high as 1900 m altitude, whereas B. permembranaceus occupiesthe altitude range 1940–2760 m. B. truncatus may be expandingits distribution in Kenya in man-made waterbodies, while B.permembranaceus possibly is restricted by adaptation to coolconditions and by interaction with B. tropicus. Water movement to and from a root depends on the soil hydraulicconductivity coefficient (Lsoil), the distance across any root-soilair gap, and the hydraulic conductivity coefficient of the root(LP). After analytical equations for the effective conductanceof each part of the pathway are developed, the influences ofsoil drying on the soil water potential and Lsoil are describedduring a 30 d period for a loamy sand in the field. The influencesof soil drying on LP for three desert succulents, Agave deserti,Ferocactus acanthodes, and Opuntia ficus-indica, are also describedfor a 30 d period. To quantify the effects of soil drying onthe development of a root-soil air gap, diameters of 6-week-oldroots of the three species were determined at constant watervapour potentials of –1.0 MPa and –10 MPa as wellas with the water vapour potential decreasing at the same rateas soil drying during a 30 d period. The shrinkage observedfor roots initially 2·0 mm in diameter averaged 19% duringthe 30d period. The predominant limiting factor for water movementwas LP of the root for the first 7 d of soil drying, the root-soilair gap for the next 13 d, and Lsoil thereafter. Compared withthe ease of water uptake from a wet soil, the decrease in conductancesduring soil drying, especially the decrease in Lsoil causedthe overall conductance to decrease by 3 x 103-fold during the30 d period for the three species considered, so relativelylittle water was lost to the dry soil. Such rectifier-like behaviourof water movement in the soil-root system resulted primarilyfrom changes in Lsoil and, presumably, is a general phenomenonamong plants, preventing water loss during drought but facilitatingwater uptake after rainfall. In vitro Regeneration from Excised Leaf Discs of Three Brassica Species   总被引:5,自引:0,他引:5  
Excised leaf discs of three Brassica species, B. oleracea, B.napus, and B. campestris were induced to produce adventitiousbuds and subsequently entire plants by culture on media withspecific combinations of 6-benzylaminopurine (BAP) and -naphthylaceticacid (NAA). Each species required a particular hormone concentrationfor optimum growth and differentiation: B. oleracea, BAP 10mg–1 and NAA 1 mg 1–1; B. napus, BAP 10 mg 1–1and NAA 10 mg 1–1; B. campestris, BAP 1 mg 1–1 andNAA 10 mg 1–1. In a more detailed study on one of these species, namely B.oleracea, the relative influence of other media components suchas amino acids and other organic additives was examined. Itwas also found that the source and size of the explant greatlyaffected the growth response, as did the size of the culturevessel. The regenerated plants dislayed a range of ploidy as well asphenotypic abnormalities. Findings are discussed in relation to results from other tissueculture systems.  相似文献

In acidic soils, monomeric aluminium (Al3+) can reach levelsthat are toxic to plants, thus preventing many species fromgrowing there. Organic acids chelate Al and render it non-toxic.It has been shown that exudation of organic acids by Al-tolerantcrops increases their tolerance to Al. We have extended thisobservation to wild plants by comparing the ability of ten herbsto exude organic acids in response to elevated Al levels. Wehypothesized that exudation of organic acids was related tothe ability of plants to grow on Al-rich soils. Two grasseswere grown in rhizotrons in soils with 41 and 63 µM reactiveAl. Organic acids were sampled from root tips connected to anintact plant-root system.Deschampsia flexuosa (L.) Trin. exudedmore malic acid when grown in the soil with the highest Al content.Five forbs and five grasses were also exposed to three Al levels(0, 25 and 75 µM) in a hydroponic system.Rumex acetosellaL. and Viscaria vulgaris Bernh. increased exudation of oxalicacid and Galium saxatile auct. non L. and Veronica officinalisL. increased exudation of citric acid in response to elevatedAl. The distribution of the forbs in the field as describedby soil pH was negatively related to the amount of organic acidsexuded in response to Al. In contrast, none of the grasses exudedhigher amounts of organic acids with increasing Al concentrationin the hydroponic experiment. Water uptake by Agave deserti and Ferocatus acanthodes was predictedusing a two-dimensional simulation model in which the soil arounda plant was divided into a series of layers and concentric cylindricalshells. Root lengths in 0.05 m thick soil layers were determinedfor both species in the field, where mean root depths were only0.11 m for A. deserti and 0.10 m for F. acanthodes. For a yearwith average precipitation (159 mm), 42 per cent of the annualprecipitation could be taken up by A. deserti and 25 per centby F. acanthodes. Predicted water uptake by both species wasgreater from the upper soil layers (above 0.15 m) for averageand dry years, but was greater from the deeper layers for awet year. The actual root distribution for both species ledto more water uptake than when all of the roots were in a singlelayer. The large number of days per year when the soil temperaturesexceeded 57 °C (the temperature for 50 per cent inhibitionof uptake of a vital stain by root cells) may exclude rootsfrom the 0.00–0.05 m soil layer, even though water uptakewhen all roots were located there was predicted to be maximal.Therefore, the observed root distribution of A. deserti andF. acanthodes may be limited near the soil surface by high temperaturesand at maximum depths by water availability for all but wetyears. Single populations of three hexaploid species of wheat, Triticumaestivum, Triticum spelta and Triticum macha, and two populationsof the tetraploid wheat, Triticum dicoccum (Pontus and Bordeaux),were grown in a greenhouse experiment at a range of soil floodingregimes: free draining, two levels of transient flooding andcontinuous flooding. Increasing severity of flooding treatment resulted in increasedsoil reduction and an increase in the concentration of reducediron and manganese in the experimental soil, and also resultedin a reduction in vegetative growth, number of inflorescences,grain number and grain weight. There were, however, large differencesbetween the wheat populations in the degree of reduction inyield caused by flooding. The population of T. macha was muchmore flooding-tolerant than the other hexaploid species andthe ‘Pontus’ population of the emmer wheat, T. dicoccum,was more tolerant than the ‘Bordeaux’ populationof this species and than T. spelta and T. aestivum.
