The Effect of Light on the Structure and Organization of Lemna Peroxisomes   总被引:1,自引:0,他引:1  
The effect of light on a number of Lemna minor enzyme activitieswas investigated. The levels of activity of glycolate oxidase,catalase and RuBPCase increased with increasing irradiance,paralleling the increase in Lemna growth rate. In contrast withresults obtained for other species, no glycolate oxidase activitycould be detected in etiolated Lemna fronds or when these weretreated with light or glycolate, in vivo or in vitro, for upto 24 h. The number of peroxisome profiles per cell section was determinedin Lemna grown under different light conditions. When frondswere grown under dim light, the number of peroxisome profilesper cell section appeared to increase with increasing irradiance,although no further increase in the peroxisome number was apparentwhen the fronds were grown under higher irradiances. The levelof glycolate oxidase activity per peroxisome was shown to increasewith increasing irradiance, whereas that of catalase remainedrelatively constant, indicating that differential addition ofenzymes to pre-existing peroxisomes is possible. Peroxisomes from Lemna grown under high irradiance were subjectedto serial sectioning and examined under the electron microscope.Some peroxisomes were found to have a three dimensional structuresuggesting either fission and/or fusion or branching of theseorganelles, supporting the hypothesis of a peroxisomal reticulum.The dynamic relationship between the various shapes is discussed. Abstract Climatic data and collection records for the cactophilic Drosophila aldrichi and Drosophila buzzatii for 97 localities were used to examine the effects of geographical location, season, host plant species and climatic factors on their range and relative abundance. Temporal variation in relative abundance was assessed from monthly collections over 4 years at one locality. Effects of weather variables over the 28 days before each collection were examined. A generalized linear model of the spatial data showed significant geographical variation in relative abundance, and significant climatic effects, with the proportion of D. aldrichi higher in the warm season, and increasing as temperature variation decreased and moisture indices increased. The temporal data gave generally concordant results, as D. aldrichi proportion was higher in summer and autumn, and increased as maximum and minimum temperatures increased, and as variation in maximum temperature decreased. In a laboratory competition experiment, D. aldrichi eliminated D. buzzatii at 31°C, but was itself eliminated at 18°C and 25°C. The range of D. buzzatii is constrained only by availability of its host plant, Opuntia species, although its relative abundance is reduced in the northern part of its distribution. The range of D. aldrichi, from central Queensland to northern NSW, Australia, is entirely within that of D. buzzatii, and its relative abundance decreases from north to south. Both climate and weather, particularly temperature variability, have direct effects on the relative abundances of the two species, and both likely act indirectly by influencing the outcome of interspecific competition.  相似文献

We describe here an integration of hydraulic and chemical signals which control stomatal conductance of plants in drying soil, and suggest that such a system is more likely than control based on chemical signals or water relations alone. The determination of xylem [ABA] and the stomatal response to xylem [ABA] are likely to involve the water flux through the plant. (1) If, as seems likely, the production of a chemical message depends on the root water status (Ψr), it will not depend solely on the soil water potential (Ψs) but also on the flux of water through the soil-plant-atmosphere continuum, to which are linked the difference between Ψr and Ψs. (2) The water flux will also dilute the concentration of the message in the xylem sap. (3) The stomatal sensitivity to the message is increased as leaf water potential falls. Stomatal conductance, which controls the water flux, therefore would be controlled by a water-flux-dependent message, with a water-flux-dependent sensitivity. In such a system, we have to consider a common regulation for stomatal conductance, leaf and root water potentials, water flux and concentration of ABA in the xylem. In order to test this possibility, we have combined equations which describe the generation and effects of chemical signals and classical equations of water flux. When the simulation was run for a variety of conditions, the solution suggested that such common regulation can operate. Simulations suggest that, as well as providing control of stomatal conductance, integration of chemical and hydraulic signalling may also provide a control of leaf water potential and of xylem [ABA], features which are apparent from our experimental data. We conclude that the root message would provide the plant with a means to sense the conditions of water extraction (soil water status and resisance to water flux) on a daily timescale, while the short-term plant response to this message would depend on the evaporative demand.  相似文献

Whole-plant ABA flux and the regulation of water loss in Cedrella odorata   总被引:2,自引:0,他引:2  
Three-month-old Cedrella odorata seedlings were exposed to a soil-drying treatment. During this period, xylem sap was periodically collected from the plant by applying pneumatic pressure to the roots. This also allowed whole-plant water status to be measured by recording the balancing pressure applied. The concentration of ABA in xylem sap (C) was related to the whole-plant transpiration rate (V) which was measured with a sap flow gauge. The analysis of these paired measurements centred on how the reciprocal of C (R) varied with respect to V. This revealed that (1) the observed increases in C could not be explained by the reductions in V alone, (2) initially, decreases in V were associated with proportional increases in the whole-plant ABA flux (M), and (3) this relationship broke down at low values of V since zero flow was associated with a finite value for C estimated to be 41 pmol ABA mmol?1 H2O. A simple static model is developed from the observations that is able to explain the data well, and the results are discussed in terms of the effects of ABA on stomatal conductance (gsw).  相似文献

SYNOPSIS. One intact and 2 splenectomized calves were infected with Babesia divergens and the persistence of the parasites in the blood was followed by periodic subinoculations into susceptible splenectomized calves. After periods varying from 3–7 years the parasites failed to be demonstrable by this method. When the immunity of the animals was challenged by the intravenous injection of blood containing Babesia divergens , they were all resistant to reinfection. These observations add support to the suggestion that sterile immunity may play a part in the resistance of cattle to reinfection with B. divergens.  相似文献

[14C]Sucrose, [14C]glucose and [14C]fructose were introducedinto potato tubers held at 10 °C and the redistributionof label chased over a 65 d period in storage. Respiratory losseswere identical in all treatments, as was the partitioning of14C between soluble and insoluble forms. Sucrose was the predominantlabelled sugar in the tubers after 20 h, regardless of the original[14C]sugar introduced, and was loaded and distributed throughoutthe tubers by the internal phloem system. After 20 h the proportionsof labelled sugars bore no relationship to those of the unlabelledendogenous sugars. However, with time the percentage of 14Cin sucrose fell while that in glucose increased and by 65 dthe proportions of the labelled sugars more closely resembledthe endogenous pools. Fructose represented a consistently lowproportion of both the labelled and unlabelled sugars. By 21d a considerable proportion of the soluble 14C had been convertedto starch (approx. 25% of the total tuber 14C), this value remainingrelatively constant for the remainder of the storage period.Sprouts which formed on the tubers contained up to 6% of thetotal tuber 14C but less than 0.2% of the tuber dry matter.It is suggested that the bulk of the translocated [14C]sucroseentered the symplast and exchanged slowly with the bulk of thesugars in the storage cell vacuoles. In Verticillium dahliae isolates (‘wild type’) obtained prior to the advent of benzimidazole fungicides, growth of germ tubes and extension of mycelium were typically inhibited by benomyl at 0–3 ppm and 0.5-1.0 ppm respectively. Culture of such isolates in the presence of increasing concentrations of benomyl resulted in the selection of variants able to grow at 12.0 ppm. Conidia, and mycelium grown for 24 b, were each more sensitive to inhibition by benomyl than mycelium grown for 48 h. Mycelium of both wild-type and selected variants was still viable after treatment with 20 ppm benomyl in a nutrient medium for 112 days. Long-term treatments at 20 ppm caused the enlargement of mycelial cells and conidia, thickening of the cell walls and formation of large spherical inclusions apparently comprising or containing lipids.  相似文献

SYNOPSIS. Examination of littoral fish Blennius pholis and Cottus bubalis caught at Aberystwyth and Porth Cwyfan, Wales. U.K., revealed 2 species of coccidia.
Eimeria dingleyi sp. n. Oocysts spherical (16.1–19.2) to subspherical (13.9–14.2 × 18.8–20.0) μm, with thin walls; sporulation outside the host to produce ellipsoid sporocysts; endogenous phases in epithelial cells throughout intestine; 26 of 58 B. pholis infected.
Eimeria variabilis (Thélohan) Reichenow. Oocysts spherical (11.9–14.6) to subspherical (9.2–10.9 × 13.9–14.3) μm; sporulation in lining of pyloric ceca and rectum; previously unrecorded schizonts and gametocytes present; 21 of 25 C. bubalis infected.
Electron microscopy revealed that the oocyst wall of E. variabilis consists of a thin membrane whereas the sporocyst wall is thick and 3-layered. Typical oocyst wall-forming bodies were absent from the macrogamete. Cytochemical tests on the endogenous stages of E. dingleyi and E. variabilis indicated that in general they resembled other coccidia in their chemical constitution.  相似文献
