为探究先后提取枸杞多糖及枸杞色素时对各自得率的影响。本研究通过分别考察这两种成分在提取过程中的提取溶剂、提取温度、料液比、提取次数及提取时间等因素对枸杞色素和枸杞多糖的得率影响,确定枸杞色素和枸杞多糖在提取次序不同时,两者的最佳提取工艺以及对DPPH·和·OH自由基清除率。结果表明,首先提取枸杞多糖后,枸杞色素的最佳提取工艺为采用正己烷,80℃时,料液比1∶10,提取2次,每次1 h,枸杞色素得率为2.48%,此时枸杞多糖得率为7.45%;而首先提取枸杞色素后,采用了超声辅助提取的方式提取枸杞多糖,发现超声效率为25%,料液比1∶10,提取20 min,枸杞多糖得率为5.23%,此时枸杞色素得率为3.93%。因此,首先提取枸杞多糖,使其平均得率为7.45%,而后提取枸杞色素,其平均得率为2.48%;总体上,枸杞色素1和枸杞多糖1对DPPH·自由基清除率都较高,枸杞多糖1对·OH自由基清除率较高,其抗氧化活性都接近Vc。  相似文献

Polysaccharides were extracted from Asparagus officinalis. A novel ultrasonic circulating extraction (UCE) technology was applied for the polysaccharide extraction. Three-factor-three-level Box-Behnken design was employed to optimize ultrasonic power, extraction time and the liquid-solid ratio to obtain a high polysaccharide yield. The optimal extraction conditions were as follows: ultrasonic power was 600 W, extraction time was 46 min, the liquid-solid ratio was 35 mL/g. Under these conditions, the experimental yield of polysaccharides was 3.134%, which was agreed closely to the predicted value. The average molecular weight of A. officinalis polysaccharide was about 6.18 × 104 Da. The polysaccharides were composed of glucose, fucose, arabinose, galactose and rhamnose in a ratio of 2.18:1.86:1.50:0.98:1.53. Compared with hot water extraction (HWE), UCE showed time-saving, higher yield and no influence on the structure of asparagus polysaccharides. The results indicated that ultrasonic circulating extraction technology could be an effective and advisable technique for the large scale production of plant polysaccharides.  相似文献

Taking into account the rising trend of the incidence of cancers of various organs, effective therapies are urgently needed to control human malignancies. However, almost all chemotherapy drugs currently on the market cause serious side effects. Fortunately, several studies have shown that some non‐toxic biological macromolecules, including algal polysaccharides, possess anti‐cancer activities or can increase the efficacy of conventional chemotherapy drugs. Polysaccharides are characteristic secondary metabolites of many algae. The efficacy of polysaccharides on the normal and cancer cells is not well investigated, but our investigations proved a cell specific effect of a newly isolated extracellular polysaccharide from the red microalga Porphyridium sordidum. The investigated substance was composed of xylose:glucose and galactose:manose:rhamnose in a molar ratio of 1:0.52:0.44:0.31. Reversible electroporation has been exploited to increase the transport through the plasma membrane into the tested breast cancer tumor cells MCF‐7 and MDA‐MB231. Application of 75 µg/mL polysaccharide in combination with 200 V/cm electroporation induced 40% decrease in viability of MDA‐MB231 cells and changes in cell morphology while control cells (MCF10A) remained with normal morphology and kept vitality.  相似文献

Polysaccharides production from Poria cocos was carried out using aqueous NaOH with the assistance of ultrasonic. Experimental design was used to investigate the effect of three parameters (extraction time, extraction concentration of NaOH, and ratio of aqueous NaOH to raw material) on polysaccharides yields. The ranges of the factors investigated were 1–3 min for extraction time (X1), 0.5–1.0 mol/L for extraction concentration of NaOH (X2), and 30–50 for ratio of aqueous NaOH to raw material (X3). The statistical analysis of the experiment indicated that extraction concentration of NaOH had significant effect on P. cocos polysaccharides yields. The central composite design showed that polynomial regression models were in good agreement with the experimental results with the coefficients of determination of 0.9935 for P. cocos polysaccharides yield. The optimal condition for P. cocos polysaccharides yield within the experimental range of the variables studied was at 2.44 min, 0.789 mol/L, and 53.0. At this condition, the predicted yield of polysaccharides extracted was 82.3%.  相似文献

为利用红托竹荪菌托,采用酶解法提取菌托多糖,优化多糖提取工艺,并测定多糖分子量、单糖组成、抗氧化及降血糖活性。结果表明,最佳酶解法提取工艺为纤维素酶2.5%、果胶酶0.4%、木瓜蛋白酶1.5%,50 ℃酶解1 h,料液比1:60、提取温度80 ℃、时间3 h,该条件下多糖提取率达15.37%,比热水浸提法提高39.60%。酶解法多糖分子量为3 344 Da (Mn)、522 208 Da (Mw)、2 929 Da (Mp),主要由葡萄糖、甘露糖、葡萄糖醛酸、半乳糖和岩藻糖等组成,葡萄糖占最高,达48.82%。菌托多糖为2.0 mg/mL时,DPPH·清除率为93.83%,Fe3+还原能力为0.140 7,α-葡萄糖苷酶活性抑制率为54.62%、α-淀粉酶活性抑制率为56.45%,与热水浸提法相比差异极显著或显著。酶解法提取红托竹荪菌托多糖,提取率较高,具有较高的抗氧化、降血糖活性,具有推广应用价值。  相似文献

An aqueous extract of polysaccharides from longan pulp was chromatographed on a DEAE anion-exchange column to yield four fractions (LPI-IV). Immunomodulatory activities of these polysaccharides were also evaluated in vitro. The purified products, neutral polysaccharide LPI, polysaccharide-protein complex LPII and acidic polysaccharides LPIII and LPIV, exhibited conspicuous differences in their monosaccharide composition, molecular mass and glycosidic linkages. Except for LPI, the other three significantly stimulated lymphocyte proliferation in the dose range of 100-400 μg/mL compared with the normal control (P < 0.05), and might electively stimulate B cells, but not T cells. Furthermore, their stimulations on normal/lipopolysaccharide-induced proliferation and depressions on concanavalin A-induced proliferation could be ordered as LPIII > LPIV > LPII > LPI. All the fractions had the optimal dose of 100 μg/mL on enhancing macrophage phagocytosis. Among them, LPII had the considerable yield and activity for exploiting as a potential immunoadjuvant.  相似文献

This study emphasis the production of yellow pigment from endolichenic Bacillus sp. isolated from the lichen Dirinaria aegialita (Afzel. ex Ach.) B.J. Moore. Yellow pigment-producing twenty different strains were investigated. The hyperactive pigment-producing bacterial strain was identified as Bacillus gibsonii based on 99 % sequence similarity. Maximum bacterial pigment production appeared in Luria Bertani medium. Methanol extraction of the pigment and its partial purification using TLC was carried out. Furthermore, isolated pigments were characterized using UV-visible spectroscopy, FTIR spectroscopy, and GC-MS results related to the possibility of the carotenoid occurrence. The pigment also exhibited efficient antifungal activity against selected fungal pathogens of economic importance. Likewise, the pigment extract evaluated for the total antioxidant potential using Phosphomolybdenum and Ferric reducing antioxidant power assay and the results represented in Ascorbic Acid Equivalent (AAE)- 21.45 ± 1.212 mg/mL. The SC50 of the pigment extract found to be 75.125 ± 0.18 µg/ml determined by the ABTS assay.  相似文献

The purpose of this study is to optimize the extraction process and study antioxidant activity of Polysaccharide extracted from Centipeda minima. The Box-Behnken design-response surface methodology was adopted to optimize the extraction process of polysaccharides from Centipeda minima. We purified the crude polysaccharides from Centipeda minima, as well as determined the purity, monosaccharide composition, and molecular weight of the purified fraction. Fourier transform infrared spectrometer (FT-IR) and scanning electron microscopy (SEM) were used to analyze the structural features of the polysaccharides. Further, we investigated the antioxidant activities of different fractions of polysaccharides. Consequently, the results showed that the optimum extraction conditions for polysaccharides were: a liquid-solid ratio of 26 mL/g, extraction temperature of 85.5 °C, and extraction time of 2.4 h. Moreover, the yield of polysaccharides measured under these conditions was close to the predicted value. After purification, we obtained four components of Centipeda minima polysaccharides (CMP). The purity, monosaccharide composition, molecular weight, and structural characteristics of CMP were different, but with similar infrared absorption spectra. CMP exhibited a typical infrared absorption characteristic of a polysaccharide. Besides, CMP displayed good antioxidant activity, with potential to scavenge DPPH radical, hydroxyl radical, and superoxide radical. Therefore, this study provides a reference for future research on the structure and biological activity of CMP, and lays a theoretical foundation for food processing and medicinal development of CMP.  相似文献

Medical fungi polysaccharides belong to a very important species of biological macromolecules, which are the basic substances that effectively maintain and ensure the normal operation of biological life activities. However, research on extraction and biological activity of Inonotus cuticularis polysaccharides has never been reported. In this study, the optimum yield of Inonotus cuticularis polysaccharides was determined by the orthogonal experimental design. The highest yield of 3.10±0.06 % was obtained with extraction temperature of 80 °C, extraction time of 150 min, and water to raw material ratio of 30 mL/g and repeated twice. After deproteinization for 5 times, the protein removal rate reached 70.10±1.75 %, and the content of polysaccharides and protein were 46.64 and 0.42 %. Infrared spectrometer indicated that Inonotus cuticularis polysaccharides are typical β‐pyranose with characteristic peaks of polysaccharides. Subsequently, the activities of scavenging free radicals for the deproteinated polysaccharides were studied. When the concentration of Inonotus cuticularis polysaccharides was 0.3 mg/mL, the scavenging activities of the sample on DPPH., .OH, ABTS.+ and O2.? reached 83.67±0.27, 65.21±4.82, 43.45±1.36 and 80.28±2.30 %, respectively, and the reducing power reached 0.46±0.01. The IC50 values scavenging DPPH., .OH, ABTS.+ and O2.? were 0.139±0.13, 0.162±0.14, 0.317±0.30 and 0.121±0.10 mg/mL, respectively. Results showed that Inonotus cuticularis polysaccharides present potential stronger antioxidant activities, especially .OH scavenging activity and reducing power. Experimental results could provide research basis of Inonotus cuticularis polysaccharides for further exploitation and utilization.  相似文献

奶油栓孔菌Trametes lactinea是一种生物活性丰富的大型真菌。本研究在单因素试验的基础上,通过响应面法优化其菌丝体多糖的提取工艺,利用DEAE-Cellulose-52阴离子交换柱和Sephadex G-200层析柱对粗多糖进行分离纯化,获得TLMPS-0、TLMPS-1和TLMPS-3均一多糖组分。采用化学组成分析、UV-vis、FTIR、刚果红实验对3种多糖组分进行结构分析,并检测了多糖清除自由基的能力和还原力。结果表明,奶油栓孔菌菌丝体多糖最优提取工艺为:提取温度99℃、料液比1:30 (g/mL)、提取时间5h,提取次数2次。在此工艺条件下,多糖提取率为4.01%。TLMPS-0、TLMPS-1和TLMPS-3的糖醛酸含量分别为12.91%±0.44%、8.24%±0.22%、7.50%±0.66%,硫酸基含量分别为22.24%±1.88%、14.55%±0.56%、18.68%±0.69%,并且证明TLMPS-0是一种α-吡喃型多糖或β-吡喃型多糖,而TLMPS-1是一种β-吡喃型多糖,均不具备三螺旋空间构象,此外,3种多糖组分均具有一定的清除DPPH自由基、ABTS自由基、羟基自由基的能力和铁离子还原能力,其中TLMPS-0抗氧化活性最强。研究结果为奶油栓孔菌多糖的功能研究与挖掘提供了研究基础与理论依据。  相似文献

为优化雪松松针多糖超声波酶法的提取工艺,并研究多糖结构及其抗氧化性。通过响应面法分析确定最佳提取参数为:3. 0 g松针粉末,液料比20∶1(m L∶g),提取温度80℃,超声功率560 W,超声时间47 min,纤维素酶用量12 FPU/g原料,提取两次,多糖得率高达10. 39%。采用高效液相色谱、红外光谱和核磁共振光谱等对松针多糖进行了结构表征,松针多糖以β-糖苷键为主要连接方式,并由葡萄糖、果糖、阿拉伯糖和半乳糖等单糖组成。体外抗氧化性研究结果表明:松针粗多糖对羟基自由基(·OH)和1,1-二苯基-2-三硝基苯肼自由基(DPPH·)的清除能力远高于纯化多糖,呈现出良好的量效关系,粗多糖对·OH和DPPH·的半抑制浓度IC50分别为0. 47 g/L和0. 076 g/L。  相似文献

通过响应面设计得到桦褐孔菌子实体Inonotus obliquus多糖的优化提取条件,利用酶和细胞体外实验,探究桦褐孔菌子实体多糖的降血糖活性,确定活性多糖组分,并初步分析其降糖机理及单糖组成。优化条件下多糖得率为(2.79±0.03)%。粗多糖经乙醇分级沉淀分别获得IOP30、IOP60、IOP80组分。其中IOP30显示出最强的α-葡萄糖苷酶抑制活性(IC50=38.20μg/mL),且在酶动力学上属于非竞争性抑制。HepG2细胞经0.01、0.10、1.00mg/mL的IOP30处理,葡萄糖消耗与对照组相比差异极其显著(P<0.01);同时添加10-7mmol/L的胰岛素和0.10、1.00mg/mL的IOP30后葡萄糖消耗增加更显著(P<0.01)。IOP30主要由鼠李糖(7.93%)、阿拉伯糖(3.12%)、木糖(11.15%)、甘露糖(19.08%)、葡萄糖(39.17%)和半乳糖(19.15%)等单糖组成。桦褐孔菌子实体IOP30多糖组分具有显著的降血糖活性,其降糖可通过抑制α-葡萄糖苷酶活性减少餐后血糖的形成,促进细胞周围组织对葡萄糖的代谢,加强肝细胞对胰岛素的敏感性。  相似文献

In the current investigation, the active principles of the methanol extracts of Rhododendron arboreum leaves (MEL) and flowers (MEF) were investigated with the help of ultra-high performance liquid chromatography (UHPLC), amino acid analyzer and gas chromatography mass spectrometry (GC-MS). UHPLC revealed different polyphenols present in the extracts. GC-MS identified 20 phytochemicals in leaves and 17 in the flowers, whereas, amino acid analyzer confirmed 11 amino acids in leaves and 10 in the flowers. The extracts were subjected to the investigation of biological activity through analysis of antioxidant activity in different in vitro assays, antimutagenic activity in Ames assay and cancer cell growth inhibition activity by MTT (3-4,5 dimethylthiazol-2,5 diphenyltetrazolium bromide) assay. MEL showed higher antioxidant activity in lipid peroxidation inhibition assay (95.32 ± 0.37%) than MEF (77.09 ± 4.17%) with IC50103.6 µg/ml for MEL and 271.17 µg/ml for MEF. In nitric oxide scavenging assay, an activity of 94.46 ± 0.32% (IC50 150.13) was observed in MEF followed by 83.71 ± 0.74% (IC50 179.52) in MEL. The antimutagenic activity of both the extracts was evaluated against sodium azide, 4-nitro-O-phenylenediamine and 2-aminofluorene mutagens in TA-98 and TA-100 strains of Salmonella typhimurium. The analysis was carried out using pre- and co-incubation modes. However, both extracts were observed to possess considerable antimutagenic activity against different known mutagens, flowers came out to be more effective than the leaves in terms of % inhibition. The extracts also exhibited significant cancer cell growth inhibition activity, when tested against 3 cancer cell lines namely, Human cervical cancer cell line (HeLa), Breast cancer cell line (MCF7) and Lung cancer cell line (A549). In case of HeLa and A549, MEL showed higher activity of 64.62 ± 2.65 and 75.08 ± 1.68% as compared to 53.11 ± 2.84 and 45.92 ± 2.43% in MEL, respectively. The EC50 values for MEL in HeLa and A549 were noted to be 232.76 and 155.38 µg/ml, respectively, whereas, MEF had EC50 of 395.50 µg/ml in HeLa and 660.26 µg/ml in A549. Further, MEF showed higher cytotoxicity in MCF7 cell line (84.93 ± 1.17%) followed by the MEL (73.57 ± 1.27%) with EC50 value of 95.16 µg/ml for MEF followed by 172.19 µg/ml for MEL. The study aimed to assess the proficiency of secondary metabolites (SMs) synthesized by actinobacteria isolated from the rhizospheric soil of Rauwolfia serpentina for its antimicrobial and anti-biofilm activity. After morphological and biochemical identification of actinobacteria, primary and secondary screening was done for specific metabolite production. The secondary metabolites were then tested for their antioxidant, antibacterial, and antibiofilm potential. Out of 29 bacterial colonies isolated, only one emerged as a novel isolate, Microbacterium LA2(R). Partial 16S rRNA gene sequence of the isolate LA2(R) was deposited in NCBI GenBank with accession number MN560041. The highest antioxidant capacity of the methanolic extract the novel isolate was found to be 474.183 µL AAE/mL and 319.037 µL AAE/mL by DPPH assay and ABTS assay respectively; three folds higher than the control. These results were further supported by the high total phenolic (194.95 gallic acid equivalents/mL) and flavonoid contents (332.79 µL quercetin equivalents/mL) of the methanolic extract. GC–MS analysis revealed the abundance of antibacterial compounds; where, n-Hexadecanoic acid was found to be the major compound present with a peak of 14 min retention time (RT) and 95% similarity index. MIC value of the metabolite was noted to be around 132.28 ± 84.48 μg/mL. The IC50 value was found to be 74.37, 71.33, 66.28 and 84.48 μg/mL against Escherichia coli, Staphylococcus aureus, Klebsiella pneumonia, and Salmonella abony, respectively. Treatment with IC50 of the extract decreased the biofilm formation up to 70%–80% against pathogenic strains viz. Escherichia coli, Staphylococcus aureus, Klebsiella pneumoniae and Salmonella abony. These significant activities of Microbacterium sp. LA2(R) suggests that it could be utilized for antibiotic production for human welfare and in various important industrial applications.  相似文献

Owing to extremely high salinity and harsh environmental conditions, T. articulata is one of the most abundant wild plants growing in the deserts of Saudi Arabia. Such plants may contain novel compounds to display promising biological activities. Here, in this study, we evaluate the biological activities of methanolic extracts of fresh leaves, dry leaves, stem, and roots of T. articulata. The antioxidant activity was determined by 2, 2-diphenyl-1-picrylhydrazyl (DPPH) and total phenolic and flavonoid content were determined using standard colorimetric methods. Whereas antimicrobial and ant-proliferative activities were determined by standard well-diffusion and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) methods, respectively. Our results demonstrate that all methanolic extracts of T. articulata showed antioxidant activity, however, the methanolic extract of dry leaves exhibits promising antioxidant effect with IC₅₀ value 49.08 ± 1.98, which was strongly supported by total phenolic (409.92 ± 6.03 mg GAE/g DW) and flavonoid (177.71 mg QE/g DW) content. Although, antimicrobial activity was also exhibited by all the methanolic extracts, however, methanolic extract of dry leaves exhibits promising antimicrobial activity in Gram-positive bacteria Staphylococcus epidemidis. Furthermore, MTT assay revealed that all methanolic extracts exhibit antiproliferative activity in MCF-7 (breast cancer) and RKO (colorectal cancer) cells with IC₅₀ values ranges from 219 ± 5.112 µg/ml to 253 ± 5.231 µg/ml and 220 ± 4.330 µg/ml to 325 ± 6.213 µg/ml, respectively. However, the most promising antiproliferative effect was displayed by methanolic extract of dry leaves with IC₅₀ values 219 ± 5.112 µg/ml and 220 ± 4.330 µg/ml, respectively. In summary, these findings provide evidence that T. articulata has promising biological activities and can be used for many pharmaceutical activities in the future.  相似文献

研究石耳属地衣Umbilicaria muehlenbergii多糖的提取工艺及其体外抗氧化、抗肿瘤能力。在单因素试验基础上,通过Box-Behnken design(BBD)和响应面分析法优化得出热水浸提法提取U. muehlenbergii多糖的最佳工艺条件。通过测定地衣多糖对1,1-二苯基-2-苦肼基自由基(1,1-diphenyl-2-picrylhydrazyl,DPPH)、羟自由基、超氧阴离子和亚硝酸盐的清除率,对其体外抗氧化能力、抗肿瘤能力进行了评价。结果表明,U. muehlenbergii多糖提取最佳工艺参数为温度90℃,时间每次4.5h,液固比40:1,提取次数5次。在此工艺条件下,U. muehlenbergii多糖实际提取率为10.63%。同时,该多糖具有较好的抗氧化、抗肿瘤能力,是一种潜在的开发资源。  相似文献

Marine natural products have displayed numerous advantageous effects on biological activities, including antioxidants and cytotoxicity. The total lipids, carotenoids, chlorophyll a and b content, total phenolic content (TPC), total flavonoid content (TFC), and antioxidant activity of methanolic crude extract of the green seaweed Halimeda opuntia were all measured in this study. The TPC of the extracts was determined according to the Folin-Ciocalteu method, yielding a result of 55.04 ± 0.98 mg GAE/g of extract. As determined by the aluminium chloride colorimetric method, the TFC of the extract was 40.02 ± 0.02 mg QE/g of extract. Antioxidant activity was determined by using a 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay with different concentrations that ranged between 200 and 1000 µg/mL, noted H. opuntia as the highest in DPPH reduction (63.61 %) at 1000 µg/mL concentration. Total antioxidant capacity (TAC) of the extract was 57.36 ± 0.004 mg AAE/g extract at concentration of 1.0 mg/mL. The cytotoxic activity of this seaweed was pre-screened against a panel of cell lines including estrogen receptor-positive human breast adenocarcinoma (MCF-7), estrogen negative human breast adenocarcinoma (MDA-MB-231), human colorectal adenocarcinoma (HT-29), human hepatocellular carcinoma (HepG2), and mouse embryonic fibroblast (3T3) using the MTT assay. The content of total lipids in H. opuntia was 1.60 ± 0.002 %. Total carotenoids were 115.57 ± 0.98 µg/g, while chlorophyll a and b were 148.73 ± 2.60 µg/g and 290.83 ± 9.46 µg/g, respectively. In terms of cytotoxicity activity, methanolic extract of H. opuntia was found to be highly cytotoxic to MCF-7 cells, with an IC50 of 25.14 ± 1.02 g/mL, and slightly less so to 3T3 cells (IC50 65.23 ± 0.25 µg/mL). This study's findings suggest that natural pigments (carotenoids and chlorophyll), phytochemicals like phenolic and flavonoid compounds found in this species may play an important role and could be used as a natural cancer treatment.  相似文献

To probe the effects of renewal regime on the production of polysaccharides, Porphyridium cruentum was cultured semi-continuously in flat plate photobioreactor. Uniform design was used to optimize renewal conditions. Quadratic mathematic models related to productivity, total recovery yield of biomass and polysaccharides were set up to clarify the influence of individual factors and their interactions. According to the mathematic models, the optimal semi-continuous condition for total yield of polysaccharide was NaNO3 3.5 g/L, renewal rate 27%, renewal period 2.91 days. The optimal condition for polysaccharide output rate was NaNO3 0.5 g/L, renewal rate 5%, renewal period 7 days. With the optimal renewal regime, the maximal total recovery yields of polysaccharide achieved at 29.4 g, which was 1.57 times higher than that of batch cultivation. The maximum output rate of polysaccharide was 68.64 mg/L per day, which was 2.02 times higher than previous reported data.  相似文献

Fucose-containing sulfated polysaccharides can be extracted from the brown seaweed, Sargassum sp. It has been reported that fucose-rich sulfated polysaccharides from brown seaweeds exert different beneficial biological activities including anti-inflammatory, anticoagulant, and anti-viral effects. Classical extraction of fucose-containing sulfated polysaccharides from brown seaweed species typically involves extended, multiple-step, hot acid, or CaCl2 treatments, each step lasting several hours. In this work, we systematically examined the influence of acid concentration (HCl), time, and temperature on the yield of fucose-containing sulfated polysaccharides (FCSPs) in statistically designed two-step and single-step multifactorial extraction experiments. All extraction factors had significant effects on the fucose-containing sulfated polysaccharides yield, with the temperature and time exerting positive effects, and the acid concentration having a negative effect. The model defined an optimized single-step FCSPs extraction procedure for Sargassum sp. (a brown seaweed). A maximal fucose-containing sulfated polysaccharides yield of ~7% of the Sargassum sp. dry matter was achieved by the optimal extraction procedure of: 0.03?M HCl, 90°C, 4?h. HPAEC-PAD analysis confirmed that fucose, galactose, and glucuronic acid were the major constituents of the polysaccharides obtained by the optimized method. Lower polysaccharide yield, but relatively higher fucose content was obtained with shorter extraction time. The data also revealed that classical multi-step extraction with acid ≥0.2?M HCl at elevated temperature and extended time had a detrimental effect on the FCSPs yield as this treatment apparently disrupted the structural integrity of the polymer and evidently caused degradation of the carbohydrate chains built up of fucose residues.  相似文献

Microwave-assisted extraction was employed for the isolation of polysaccharides from Posidonia oceanica (PPO). The extracting parameters were optimized adopting response surface methodology. The highest polysaccharide yield (2.55 ± 0.09%), which is in concordance with the predicted value (2.76%), was obtained under the following conditions: extraction time 60 s, liquid–solid ratio of 50:1 (mL/g) and power of 800 W. This polysaccharide, with molecular weight of 524 KDa, characterized by gas chromatography–mass spectrometry showed that PPO was mainly composed of galactose, glucose, and arabinose with molar percentages 25.38, 24.37, and 21.64%, respectively. The pharmacological evaluation of PPO using animal models at the dose of 100 mg/kg indicated a significant anti-inflammatory activity with a percentage of inhibition of edema of 54.65% and a significant antinociceptive activity with 78.91% inhibition of writhing for peripheral analgesic activity and an increase in the hot plate reaction time for central analgesic activity.  相似文献
