本文研究了蒙脱土/还原氧化石墨烯负载纳米铜(MMT-rGO-CuNPs)复合抗菌材料的制备及其抗菌性能。通过X射线衍射图谱的检测,确定MMT-rGO-CuNPs复合物已经成功。为了研究复合物的抗菌活性,通过观察大肠杆菌和金黄色葡萄球菌的表面形貌变化和细菌体外离子浓度变化,证明MMT-rGO-CuNPs复合物对金黄色葡萄球菌的抗菌性能要强于对大肠杆菌。  相似文献

考察了不同分子量壳聚糖对大肠杆菌的抑菌性能,利用壳聚糖的席夫碱反应对其氨基加以保护,探讨了壳聚糖对大肠杆菌的抗菌机理。研究结果表明:壳聚糖分子量越小,对大肠杆菌的抗菌作用越明显;壳聚糖对大肠杆菌的抑菌作用与其氨基的质子化有关。  相似文献

以茶多酚磷脂复合物为研究对象,以茶多酚磷脂复合物对大肠杆菌的抑菌圈直径作为抑菌效果指标,对茶多酚磷脂复合物的抑菌条件进行了研究。考察茶多酚磷脂复合物浓度、壳聚糖浓度、柠檬酸浓度对大肠杆菌抑菌圈值的影响。在单因素试验的基础上,采用3因素3水平的响应面分析法对茶多酚磷脂复合物对大肠杆菌的抑制条件进行了优化,依据回归分析确定最优提取条件。结果表明,最佳抑菌条件为:茶多酚磷脂复合物的浓度为3.22 g/L,壳聚糖浓度为31.06 g/L,柠檬酸浓度为2.45 g/L。在此抑菌条件下,茶多酚磷脂复合物对大肠杆菌的抑制圈值达到最大,为18.1 mm。  相似文献

本文研究了纳米银胶/壳聚糖抗菌剂的制备及其形貌的表征分析,以大肠杆菌为代表菌株,研究了复合抗菌剂在洗涤产品中的抗菌效率及抗菌的稳定性,结果说明复合抗菌剂在洗涤产品中添加1.0%时,其抗菌效率达99%,经180 d长期分析,其抗菌活性仍保持95%左右。此外,复合抗菌剂对不同菌株的抗菌性能也均较强。  相似文献

内源抗坏血酸对水稻种子萌发及幼苗生长的影响   总被引:1,自引:0,他引:1  
为了解内源抗环血酸在水稻(Oryza sativa)种子中的作用,以野生型品种‘中花11’(ZH-11)、抗坏血酸合成关键酶GLDH基因的上调(超表达)株系GO-2及下调(干涉)株系GI-2为材料,研究了抗环血酸含量对其种子萌发和幼苗生长的影响。结果表明,GO-2株系的种子萌发率比野生型ZH-11高6%,比干涉型GI-2高60%。外源添加1 mmol L–1抗坏血酸后,干涉型GI-2萌发率提高了22%,GO-2及ZH-11则没有明显增加。GO-2株系在幼苗的根长、株高、分蘖数和鲜重等指标上均高于ZH-11和GI-2。实时荧光定量测定结果表明,GO-2株系叶片的GLDH基因表达量显著上调,而GI-2株系则显著下调。这说明抗坏血酸有助于维持水稻种子活力和促进水稻种子发芽和幼苗生长。  相似文献

分析原核生物中RNA与蛋白质的相互作用,给了我们很多的信息和启示。到目前为止,大肠杆菌核糖体是研究最多的RNP复合物,而大肠杆菌中的谷氨酰胺基tRNA合成酶和相关的tRNA是目前唯一制成RNA一蛋白质共结晶,并揭示其结构的RNA一蛋白质复合物。最近在真核生物系统中的研究不仅发现了由RNA-蛋白质结合介导的基因表达调控的机理,而且鉴定出结合RNA的蛋白家族中存在有共同的短段(Common motifs)。本文拟对目前研究的饶有兴趣的5SRNA-结合蛋白复合物以及RNP-致序列进行阐述和讨论。  相似文献

本文报道了用大肠杆菌注射处理中蜂Apis cerana Fabricius后,用含菌培养基平板测活法,就中蜂抗菌物质的产生规律及抗菌活性进行了初步研究。结果表明,不同诱导源均可诱导中蜂产生抗菌物质,但诱导的抗菌物质的抗菌活性则有一定的差异。用大肠杆菌重复诱导则使中蜂的成活率和抗菌物质的抗菌活性有很大提高。注射大肠杆菌后冲蜂抗菌物持产生高峰在48小时左右。诱导后产生的抗菌物质具有广谱性,对大肠杆菌Ascherichia coli、枯草芽孢杆菌 Bacillus  subtilis、苏云金杆菌 Bacillus thuring-iensis、巨大芽孢杆菌 Bacillus megatherium、黑腐菌 Xanthomonas campestris 等均有抑菌作用,而对真菌白僵菌Beauveria bassiana未发现抑菌作用。中蜂麻醉方法以冷冻法最为简便、易行。  相似文献

肠出血性大肠杆菌O157∶H7是一种重要的致病菌,加深其致病机理的基础研究将为相关疫苗研究及疾病控制等提供新的思路和依据.串联亲和纯化(TAP)技术是最近发展的分离纯化天然状态蛋白质复合物进而研究蛋白质相互作用的新方法.用我们自己构建的原核表达串联亲和标签载体,在大肠杆菌O157∶H7中表达了标签融合蛋白GroEL-TAP,建立了非变性条件下制备蛋白复合物的方法,并且对串联亲和纯化过程中的相关实验条件进行了探索和优化,最终得到了高纯度的GroEL-TAP与天然GroEL形成的嵌合型多聚体复合物.这表明我们建立的串联亲和纯化技术能高度特异地纯化靶蛋白参与形成的复合物,为后续寻找O157∶H7中毒力蛋白参与形成的复合物奠定了实验基础.  相似文献

Seven monoclonal antibodies (MAbs) have been prepared to phytochrome from green oat (Avena sativa L. cv. Garry) leaves. One of these MAbs (GO-1) cross-reacts with apoprotein of the phytochrome that is most abundant in etiolated oat shoots as assessed by immunoblot assay of fusion proteins expressed in Escherichia coli. The epitope for this MAb is located between amino acids 618 and 686 in the primary sequence of type 3 phytochrome (Hershey et al. 1985, Nucleic Acids Res. 13, 8543–8559), which is one of the predominant phytochromes in etiolated oats. Three other MAbs (GO-4, GO-5, GO-6) immunoprecipitate phytochrome isolated from green oat leaves, as evaluated by photoreversibility assay. GO-1, GO-4, GO-5 and GO-6 are therefore directed to phytochrome. While evidence obtained with the other three MAbs (GO-2, GO-7, GO-8) strongly indicates that they are also directed to phytochrome, this evidence is not as rigorous. Recognition of antigen by any of these seven MAbs is not significantly reduced by periodate oxidation, indicating that their epitopes probably do not include carbohydrate. All but GO-1 bind either very poorly or not at all the phytochrome that is abundant in etiolated oat shoots. These data reinforce earlier observations made with antibodies directed to phytochrome from etiolated oats, indicating (1) that the phytochromes that predominate in etiolated and green oats differ immunochemically and (2) that phytochrome preparations from green oat leaves contain very little of the phytochrome that is abundant in etiolated shoots. The antigenic relationships of Bifidobacterium bifidum 1 peptidoglycans with different strains of this species (LVA-3, 791, GO-4), bifidobacteria of other species (B. adolescentis GO-13, B. breve 79-38, B. lactentis 79-41, B. longum GO-3) and bacteria of remote taxonomic groups (Streptococcus faecalis 6-3. Staphylococcus aureus COM 885, S. epidermidis COM 2124. Lactobacillus plantarum 1, Escherichia coli M-17) were studied on the basis of a highly sensitive test system permitting the registration of normal human antibodies to peptidoglycans. The level of cross reactions with staphylococci and streptococci correspond to intraspecific and antigenic affinity to L. plantarum and E. coli was considerably less pronounced. Copying a number of epitopes of bifidobacteria, S. aureus peptidoglycan seems to possess additional antigenic determinants which participate in the formation of immunological responsiveness in man.  相似文献

New approaches to antibiotic design are desperately needed. The design of simple oligomers that capture the shape and biological function of natural antimicrobial peptides could prove to be versatile and highly successful. We discuss the use of aromatic backbones to design facially amphiphilic (FA) -sheet like structures which are potently antimicrobial. These oligomers capture the physiochemical properties of peptides like the Magainins and Defensins, which fold into specific conformations that are amphiphilic resulting in antimicrobial activity. However, natural peptides are expensive to prepare and difficult to produce on large scale. The design of polymers and oligomers that mimic the complex structures and remarkable biological properties of proteins is an important endeavor and provides attractive alternatives to the difficult synthesis of natural peptides. We therefore have designed a series of FA oligomers that are easy to prepare from inexpensive monomers. They adopt structures very reminiscent of amphiphilic -sheets and have significant activity with minimal inhibitory concentrations at 6 h in the low microgram per ml range (M to nM). They are active against a broad spectrum of bacteria including gram-positive and gram-negative as well as antibiotic resistant strains.  相似文献

以天然松香酸衍生物——脱氢松香胺为原料,合成了6种脱氢松香基季铵盐。对合成化合物结构进行了元素分析、紫外、红外光谱及核磁共振谱鉴定。药理实验结果表明,合成产品对金黄色葡萄球菌及大肠杆菌具有一定抗菌活性,其最低抑菌浓度分别为7. 18~31. 25μ/ml和250~500μg/ml。同时对产品结构对抗菌活性影响也进行了比较研究。  相似文献

Demand for beef produced from pasture-based diets is rising as it is perceived to be healthier, animal friendly and good for the environment. Animals reared on a solely grass forage diet, however, have a lower growth rate than cereal-fed animals and consequently are slaughtered at an older age. This study focused on the former by conducting life cycle assessments of beef production systems offering only fresh or conserved grass, and comparing them to a conventional pasture-based beef production system offering concentrate feeding during housing. The four suckler weanling-to-beef production systems simulated were: (i) Steers produced to slaughter entirely on a grass forage diet at 20 months (GO-20); (ii) Steers produced to slaughter entirely on a grass forage diet at 24 months (GO-24); (iii) Steers produced to slaughter on a grass forage diet with concentrate supplementation during housing (GC-24), and (iv) Steers produced to slaughter entirely on a grass forage diet at 28 months (GO-28). Two breed types were evaluated: early-maturing and late-maturing (LM). The environmental impacts assessed were global warming potential (GWP), non-renewable energy (NRE), acidification potential (AP), eutrophication potential (marine (MEP) and freshwater) were expressed per animal, per kg live weight gain (LWG), kg carcass weight gain, and kg meat weight gain (MWG). The GO-20 production system had the lowest environmental impact across all categories and functional units for both breeds. Extending age at slaughter increased environmental impact across all categories per animal. The LWG response of EM steers to concentrate feed supplementation in GC-24 was greater than the increase in total environmental impact resulting in GC-24 having a lower environmental impact across categories per kg product than GO-24. Concentrate feed supplementation had a similar effect on LM steers with the exception of NRE and AP. The increase in daily LWG in the third grazing season in comparison to the second grazing and housing resulted in GO-28 having lower GWP, NRE, AP, and MEP per kg product than GO-24. Early-maturing steers had lower environmental impact than LM when expressed per kg LWG. However the opposite occurred when impacts were expressed per kg MWG, despite LM steers producing the least LWG. The LM steers compensated for poor LWG performance by having superior carcass traits, which caused the breed to have the lowest environmental impact per kg MWG. The results reaffirms the importance of functional unit and suggests reducing the environmental impact of LWG does not always translate into improvements in the environmental performance of meat.  相似文献

Hydroxides of magnesium and zinc, aluminum oxide, zinc phosphate, and co-precipitated Ca3(PO4)2 and Mg(OH)2 were efficient in binding extracellular glucose oxidase (GO) of P. adametzii LF F-2044.1 in a culture liquid filtrate (CLF). Basic Al2O3 was the most appropriate adsorbent for GO isolation from the CLF of the fungus. A GO isolation method was developed, which allowed for obtaining an enzyme with a high degree of purification. Spectral properties of the enzyme, its catalytic activity, and stability were characterized. The GO of P. adametzii LF F-2044.1 exhibited high pH stability, retaining activity within the range 4.5-9.0. The rate that GO-catalyzed D-glucose oxidation increased as the temperature increased (up to approximately 60 degrees C). The catalytic activity and thermal stability of GO depended on its concentration in the medium. Under optimum conditions, the fractions GO-1 and GO-2 were characterized by KM values of 1.56 x 10(-2) and 2.19 x 10(-2) M, respectively; the corresponding values of kcat equaled 235.1 and 318.2 s(-1).  相似文献

为提升杀虫剂防效, 减少杀虫剂用量, 提高农产品安全, 本研究采用虫体浸渍法研究了阳离子助剂1227和C8-10及有机硅助剂Breakthru S240对高效氯氰菊酯、 毒死蜱和溴虫腈在甜菜夜蛾Spodoptera exigua幼虫和棉铃虫Helicoverpa armigera幼虫上的增效作用, 并测定了3种助剂对清水物理性状的影响。结果表明: (1)供试助剂在两试虫上对3种杀虫剂均有显著增效作用, 且增效比与助剂浓度存在显著相关关系。相同浓度下, 除Breakthru S240对毒死蜱在棉铃虫上增效作用高于1227外, 3种助剂在两试虫上对3种杀虫剂增效作用均为C8-10>1227>Breakthru S240。3种助剂在甜菜夜蛾上对3种杀虫剂的最佳增效浓度均为900 mg/L。3种助剂在棉铃虫上对3种杀虫剂的最佳增效浓度分别为900, 300 和900 mg/L。(2)3种助剂对清水物理性状均有显著影响。相同浓度下, 供试助剂对清水表面张力的降低能力、 对扩展直径和最大稳定持留量的增强能力均为 Breakthru S240>1227>C8-10。综上所述, 阳离子助剂1227和C8-10对3种杀虫剂在两试虫上的增效作用显著高于有机硅助剂Breakthru S240, 但后者对于提高杀虫剂在叶表沉积量的能力高于前者, 研究结果为3种助剂在杀虫剂领域的应用提供了一定的依据。  相似文献

Biological activities of a series of palladium(II) complexes (M1–M9) bearing NN, NS, and NO chelating ligands are reported. The palladium complexes were tested for their cytotoxic properties against human cervical cancer (HeLa) cells and antibacterial activity against Gm+ve and Gm–ve bacteria. Among the palladium complexes studied (M1-M9), the complex M5, M8, and M9 were found to be more effective in inhibiting the proliferation of HeLa cells. Hence, these complexes were further investigated for their potential role in cellular damage and apoptosis. DCFDA staining, Rhodamine 123 staining and DNA cleavage assay revealed that complex M5, M8 and M9 induced apoptotic cell death in HeLa cells through ROS generation, DNA damage and mitochondrial depolarization. Computational and titration studies also indicated strong electrostatic interaction with DNA groove. Most of the complexes exhibited good antibacterial activity against both Gm+ve and Gm−ve bacteria. The antibacterial activity of the compounds could not be correlated with their anticancer activity indicating a differential mechanism at their effective concentrations. The detailed study on the antibacterial mechanism of the most potent complex M7 revealed that it exerted its antibacterial activity by inhibiting the function of FtsZ and perturbing the localization of the Z-ring at the mid cell.  相似文献
