Calcifying and a noncalcifying strains of Emiliania huxleyi were cultured in nutrient replete turbidostats under a photon flux density (PFD) gradient from 50 to 600 μmol E·m?2·s?1. For both strains, growth was PFD‐saturated at 300 μmol E·m?2·s?1. The strains, although with clearly different physiological properties due to the presence or absence of calcification, showed the same trends and magnitude of change in their pigment compliment as a function of PFD. Light‐controlled pigment composition and the trends of change in pigment composition were identical in both strains. Fucoxanthin (Fuco) was the major carotenoid in the calcifying strain, while in the noncalcifying strain this role was assumed by 19′ hexanoyloxyfucoxanthin (19 Hex). The photoprotective pigments and 19 Hex, normalized to chl a, increased with increasing light, while chl a content per cell and chl c's and Fuco, normalized to chl a, decreased with increasing PFD. The sum of all carotenoids normalized to chl a was remarkably similar in all PFDs used. Collectively, our results suggest that 19 Hex was synthesized from Fuco with light as a modulating factor and that the total amount of carotenoids is strain‐specific and synthesized/catabolized in tandem with chl a to a genetically predefined level independent of PFD.  相似文献

In 56 samples of freshwater fish, most were low in fat, ≤ 5% of dry weight (D.W.), and the sum of all fatty acids (ΣFA) was about 2% of D.W. Trout, whitefish, and grayling had the highest content of the long-chained FA. of ω3 type, EPA and DHA (1·7–2·6% of D.W.). Two large, low-fat pikes with ΣFA of about12–3% of D.W. and a medium-fat whitefish had the highest ω3/ω6 ratios, 8–9, whereas the fattiest fishes, eels from two lakes and the Baltic (ΣFA =17–26% of D.W.) had lower ω3/ω6 ratios, 1·1–1·8 (ω3 and ω6 FA are two important series of FA). The results indicate that ΣA controls the content of saturated FA (SAFA) and monounsaturated FA (MUFA), whereas the polyunsaturated FA (PUFA) was independent of ΣFA after a break point of about 10%ΣFA of D.W. The P/S ratio (PUFA/SAFA) and the PUFA/ΣFA ratio decreased with increased ΣFA, whereas the ω3/ω6 ratio showed no clear correlation to ΣFA. The difference in fatty acid patterns lay between low-fat and high-fat fishes, rather than between marine and freshwater fishes. The variation, both within and between species of the separate FA is small in fish with similar ΣFA content. Also, low-fat and medium-fat fishes tend to be more dietarily favourable than high-fat fishes, when considering the latest criteria for high nutritional value to humans. The photoprotective response in the dinoflagellate Glenodinium foliaceum F. Stein exposed to ultraviolet‐A (UVA) radiation (320–400 nm; 1.7 W · m2) and the effect of nitrate and phosphate availability on that response have been studied. Parameters measured over a 14 d growth period in control (PAR) and experimental (PAR + UVA) cultures included cellular mycosporine‐like amino acids (MAAs), chls, carotenoids, and culture growth rates. Although there were no significant effects of UVA on growth rate, there was significant induction of MAA compounds (28 ± 2 pg · cell?1) and a reduction in chl a (9.6 ± 0.1 pg · cell?1) and fucoxanthin (4.4 ± 0.1 pg · cell?1) compared to the control cultures (3 ± 1 pg · cell?1, 13.3 ± 3.2 pg · cell?1, and 7.4 ± 0.3 pg · cell?1, respectively). In a second investigation, MAA concentrations in UVA‐exposed cultures were lower when nitrate was limited (P < 0.05) but were higher when phosphate was limiting. Nitrate limitation led to significant decreases (P < 0.05) in cellular concentration of chls (chl c1, chl c2, and chl a), but other pigments were not affected. Phosphate availability had no effect on final pigment concentrations. Results suggest that nutrient availability significantly affects cellular accumulation of photoprotective compounds in G. foliaceum exposed to UVA.  相似文献

Cultures and field samples of the toxic dinoflagellate Gymnodinium catenatum Graham from Tasmania, Australia, were analyzed for pigment, fatty acid, and sterol composition. Gymnodinium catenatum contained the characteristic pigments of photosynthetic dinoflagellates, including chlorophyll a, chlorophyll c2, and the carotenoids peridinin, dinoxanthin, diadinoxanthin, diatoxanthin, and β,β-carotene. In midlogarithmic and early stationary phase cultures, the chlorophyll a content ranged 50–72 pg · cell?1, total lipids 956–2084 pg · cell?1, total fatty acids 426–804 pg · cell?1, and total sterols 8–20 pg · cell?1. The major fatty acids (in order of decreasing abundance) were 16:0, 22:6(n-3), and 20:5(n-3) (collectively 65–70% of the total fatty acids), followed by 16:1(n-7), 18:2(n-6), and 14:0. This distribution is characteristic of most dinoflagellates, except for the low abundance (<3%) of the fatty acid 18:5(n-3), considered by some authors to be a marker for dinoflagellates. The three major sterols were 4α-methyl-5α-cholest-7-en-3β-ol, 4α,23,24-trimethyl-5α-cholest-22E-en-3β-ol (the dinoflagellate sterol, dinosterol), and 4α,23,24-trimethyl-5α-cholest-7-en-3β-ol. These three sterols comprised about 75% of the total sterols in both logarithmic and early stationary phase cultures, and they were also found in high proportions (22–25%) in natural dinoflagellate bloom samples. 4-Desmethyl sterols, which are common in most microalgae, were only present in trace amounts in G. catenatum. The chemotaxonomic affinities of G. catenatum and the potential for using specific signature lipids for monitoring toxic dinoflagellate blooms are discussed.  相似文献

Growth rate, pigment composition, and noninvasive chl a fluorescence parameters were assessed for a noncalcifying strain of the prymnesiophyte Emiliania huxleyi Lohman grown at 50, 100, 200, and 800 μmol photons·m?2·s?1. Emiliania huxleyi grown at high photon flux density (PFD) was characterized by increased specific growth rates, 0.82 d?1 for high PFD grown cells compared with 0.38 d?1 for low PFD grown cells, and higher in vivo chl a specific attenuation coefficients that were most likely due to a decreased pigment package, consistent with the observed decrease in cellular photosynthetic pigment content. High PFD growth conditions also induced a 2.5‐fold increase in the pool of the xanthophyll cycle pigments diadinoxanthin and diatoxanthin responsible for dissipation of excess energy. Dark‐adapted maximal photochemical efficiency (Fv/Fm) remained constant at around 0.58 for cells grown over the range of PFDs, and therefore the observed decline, from 0.57 to 0.33, in the PSII maximum efficiency in the light‐adapted state, (Fv′/Fm′), with increasing growth PFD was due to increased dissipation of excess energy, most likely via the xanthophyll cycle and not due to photoinhibition. The PSII operating efficiency (Fq′/Fm′) decreased from 0.48 to 0.21 with increasing growth PFD due to both saturation of photochemistry and an increase in nonphotochemical quenching. The changes in the physiological parameters with growth PFD enable E. huxleyi to maximize rates of photosynthesis under subsaturating conditions and protect the photosynthetic apparatus from excess energy while supporting higher saturating rates of photosynthesis under saturating PFDs.  相似文献

Fatty acid (FA), total lipid, protein, amino acid, carbon, nitrogen, and phosphorus content was analyzed in 24 samples of freshwater microalgae. The samples originated from batch, continuous, or mass cultures in various growth phases and from net samples from lakewater. FA were analyzed quantitatively by using an internal standard in a GLC system and expressed as mg·g?1 dry weight (DW). The FA of one group of blue-greens (e.g. Oscillatoria and Microcystis) were similar to those of the greens with higher amounts of 18C acids of the ω3 type compared to the ω6 type, whereas the other group (e.g. Anabaena and Spirulina) contained mostly ω6 acids. The flagellates, a taxonomically diverse group, were characterized by high amounts of long-chained (20–22 C) polyunsaturated FA (PUFA), particularly of the ω3 type. The ω3/ω6 ratio appears to be highest in algae in the exponential growth phase. The increased lipid content in stressed algae was mostly due to increased saturated fatty acids and ω6 acids, whereas the valuable ω3 acids were unchanged or even decreased. Amino acid composition (% of total amino acids) did not vary much betaken species, but when analyzed quantitatively (mg-g?1 DW), varied considerably between species and within species in different growth phases. The nitrogen and phosphorus contents were variable in all three algal groups. The relationship between PUFA and phosphorus content differed among the algal groups. The data suggest that PUFA in the phospholipids consist mostly ω3 acids.  相似文献

Lipid content and lipid class composition were determined in stream periphyton and the filamentous green algae Cladophora sp. and Spirogyra sp, Sterols and phospholipids were compared to chlorophyll a (chl a) as predictors of biomass for stream periphyton and algae. Chlorophyll a, phospholipids, and sterols were each highly correlated with ash-free dry mass (AFDM) (r2 > 0.98). Stream periphyton exposed naturally to high light (HL) and low light (LL) had chl a concentrations (μg chl a-mg?1AFDM) of 7.9± 0.7 and 12.4 ± 2.9, respectively, while the sterol concentrations of these HL and LL stream periphyton (1.6 ± 0.4) were not significantly different (P > 0.05). Periphyton exposed to an irradiance of 300 μmol photons·m?2s?1 in the laboratory for 60 h had 5.6 ± 0.55 μg chl a·mg?1 AFDM, but the same periphyton exposed to 2% incident light for the same amount of time had 11.0 ± 0.56 μg chl mg?1 AFDM. Sterol concentrations in these periphyton communities remained unchanged (1.5 ± 0.3 μg·mg?1AFDM), Similar results (i.e. changes in chl a but stability of sterol concentrations in response to irradiance changes) were also found for Cladophora and Spirogyra in laboratory experiments. Sterols can be quantified rapidly from a few milligrams of algae and appear to be a useful predictor of eukaryote biomass, whereas cellular levels of chl a vary substantially with light conditions. Phospholipids (or phospholipid fatty acids) are considered to be a reliable measure of viable microbial biomass. Nevertheless, phospholipid content varied substantially and unpredictably among algae and periphyton under different light regimes. Irradiance also had a significant effect on storage lipids: HL Cladophora and HL periphyton had 2 × and 5 × greater concentrations of triacylglycerols, respectively, compared to their LL forms. HL and LL algae also differed in the concentration of several major fatty acids. These light-induced changes in algal lipids and fatty acids have important implications for grazers.  相似文献

Clones of Skeletonema costatum (Grev.) Cl. isolated from Narragansett Bay, R.I., during different seasons were grouped according to their electrophoretic banding patterns. The growth rates, pg chlorophyll · cell?1, carbon uptake · cell?1· h?1, and carbon uptake · pg chl?1· h?1 were measured at 20°C, in a 14:10 h L:D cycle at 180 μE · m?2· s?1. Statistically significant sources of variation were found among groups of clones in growth rate, pg chl · cell?1, and carbon uptake · pg chl?1· h?1. It was concluded that there is a significant relationship between the physiological characteristics of clones isolated from populations in different seasons and patterns of genetic variation inferred from the electrophoretic studies. However, genetic diversity detected by banding patterns tends to underestimate the total genetic diversity in natural populations. The groups of clones most common in summer bloom populations had significantly higher growth rates, lower values of pg chl · cell?1, and higher rates of carbon uptake · pg chl?1· h?1 at 20°C than did the group of clones most common in winter bloom populations. However, differences among groups in these parameters at 20°C alone cannot account for the seasonal cycling of genetically variable populations of Skeletonema in Narragansett Bay. The range of growth rates among clones of this species is 0.1–5.0 divisions · d?1 under a single set of temperature and light conditions. Chlorophyll concentrations range from 0.2–1.7 pg chl · cell?1 and carbon uptake · pg chl?1· h?1 varies by a factor of 7 among clones. The range of physiological variation in this species means that it is difficult to use laboratory studies of single clones to analyze the responses of natural populations of Skeletonema.  相似文献

1. The value of algal fatty acids (FA) as diet biomarkers for benthic harpacticoid copepods was investigated. A high proportion of 18:1ω9 and 18:2ω6 FA was observed in the lipid reserve fraction of copepods fed with cyanobacteria. In contrast, a high proportion of 16:1ω7 and ω3 FA (including eicosapentaenoic) was present in the lipid reserve fraction of copepods grown on diatoms. 2. Copepods that were grown on cyanobacteria showed reduced survival and took 26% more time to develop from the first copepodid stage to adult than copepods that were grown on diatoms. Copepods feeding on the cyanobacteria showed reduced FA content when compared with animals fed with diatoms. This reduction in FA content was more pronounced in the apolar lipid fraction (mainly reserve lipids) than in the polar (mainly structural) lipid fraction. 3. The FA profiles of algae were used to calculate a function discriminating between diatoms and cyanobacteria. This function was applied to the FA profiles in the reserve lipid fraction of copepods and correctly classified copepod diet. 16:1ω7, 18:2ω6 and 20:5ω3 were the most important FA in the discriminant function. The suitability of this chemometric method to infer copepod diet was further tested by using algal class FA data from literature to derive the discriminant functions. The correct classification of the diet when the functions were applied to FA composition of the copepod reserve lipids suggests that this method may be employed in trophic web studies. 18:3ω3, 18:1ω9 and 16:1ω7 were the most important FA in the functions discriminating diatoms, cyanobacteria and green algae. The identification and quantification of the whole suit of 16:1ω7, 18:1ω9, 18:2ω6, 18:3ω3 and 20:5ω3 in trophic web studies is therefore of paramount importance to infer diet origin of aquatic herbivores. 4. The FA profile of copepod polar lipids did not reflect that of the diet. The presence of long chain polyunsaturated FAs in the polar lipid fraction of copepods feeding on the cyanobacterium suggests that C18 FAs from the diet may be elongated and desaturated by the copepod. The ability to elongate and desaturated FAs may reduce the importance of some FAs as diet biomarkers while it may turn the copepods into valuable trophic intermediaries in transferring organic matter from microorganisms to higher trophic levels.  相似文献

Phaeodactylum tricornutum Bohlin was maintained in exponential growth over a range of photon flux densities (PFD) from 7 to 230 μmol·m?2s?1. The chlorophyll a-specific light absorption coefficient, maximum quantum yield of photosynthesis, and C:N atom ratio were all independent of the PFD to which cells were acclimated. Carbon- and cell-specific, light-satuated, gross photosynthesis rates and dark respiration rates were largely independent of acclimation PFD. Decreases in the chlorophyll a-specific, gross photosynthesis rate and the carbon: chlorophyll ratio and increases of cell- or carbon-specific absorption coefficients were associated with an increase in cell chlorophyll a in cultures acclimated to low PFDs. The compensation PFD for growth was calculated to be 0.5 μmol·m?2s?1. The maintenance metabolic rate (2 × 10?7s?1), calculated on the basis of the compensation PFD, is an order of magnitude lower than the measured dark respiration rate(2.7 × 10?6mol O2·mol C?1s?1). Maintenance of high carbon-specific, light-saturated photosynthesis rates in cells acclimated to low PFDs may allow effective use of short exposures to high PFDs in a temporally variable light environment.  相似文献

1. We studied variation in the composition of fatty acids in the seston of a small freshwater reservoir with changes in phytoplankton composition during four growth seasons. We focused on the dynamics of the ω3 fatty acids because of their potential importance for zooplankton nutrition. 2. Total diatoms were related to the 20:5ω3 fatty acid (eicosapentaenoic, EPA) content in seston. Among two dominant diatom genera, Cyclotella was not associated with EPA content. In contrast, there was a significant correlation between Stephanodiscus and the percentage contribution and content of EPA throughout the study. Hence, freshwater diatoms can differ strongly in content of the essential EPA. 3. We considered abundant cyanobacteria as a potential source of 18:3ω3 fatty acid (linolenic, ALA) to aquatic food webs. Among four dominant cyanobacteria species, two (Anabaena flos‐aquae and Planktothrix agardhii) showed significant correlation with the ALA content of the seston, while the other two (Aphanizomenon flos‐aquae and Microcystis aeruginosa) did not. 4. Dinophyta had a relatively high level of 22:6ω3 (docosahexaenoic, DHA) for freshwater species and can be also a source of EPA to aquatic food webs. 5. Our results show that various species of diatoms as well as cyanobacteria can be of contrasting nutritional value for zooplankton because of their different content of the essential PUFAs. Diatoms, which are low in EPA, could not be considered as a valuable food, while some field populations of cyanobacteria might be valuable sources of essential ALA.  相似文献

Lipid profiles of three strains (Mexico, Australia, Japan) of Chattonella marina (Subrahmanyan) Hara et Chihara were studied under defined growth (phosphate, light, and growth phase) and harvest (intact and ruptured cells) conditions. Triacylglycerol levels were always <2%, sterols <7%, free fatty acids varied between 2 and 33%, and polar lipids were the most abundant lipid class (>51% of total lipids). The major fatty acids in C. marina were palmitic (16:0), eicosapentaenoic (EPA, 20:5ω3), octadecatetraenoic (18:4ω3), myristic (14:0), and palmitoleic (16:1ω7c) acids. Higher levels of EPA were found in ruptured cells (21.4–29.4%) compared to intact cells (8.5–25.3%). In general, Japanese N‐118 C. marina was the highest producer of EPA (14.3–29.4%), and Mexican CMCV‐1 the lowest producer (7.9–27.1%). Algal cultures, free fatty acids from C. marina, and the two aldehydes 2E,4E‐decadienal and 2E,4E‐heptadienal (suspected fatty acid‐derived products) were tested against the rainbow trout fish gill cell line RTgill‐W1. The configuration of fatty acids plays an important role in ichthyotoxicity. Free fatty acid fractions, obtained by base saponification of total lipids from C. marina showed a potent toxicity toward gill cells (median lethal concentration, LC50 (at 1 h) of 0.44 μg · mL?1 in light conditions, with a complete loss of viability at >3.2 μg · mL?1). Live cultures of Mexican C. marina were less toxic than Japanese and Australian strains. This difference could be related to differing EPA content, superoxide anion production, and cell fragility. The aldehydes 2E,4E‐decadienal and 2E,4E‐heptadienal also showed high impact on gill cell viability, with LC50 (at 1 h) of 0.34 and 0.36 μg · mL?1, respectively. Superoxide anion production was highest in Australian strain CMPL01, followed by Japanese N‐118 and Mexican CMCV‐1 strains. Ruptured cells showed higher production of superoxide anion compared to intact cells (e.g., 19 vs. 9.5 pmol · cell?1 · hr?1 for CMPL01, respectively). Our results indicate that C. marina is more ichthyotoxic after cell disruption and when switching from dark to light conditions, possibly associated with a higher production of superoxide anion and EPA, which may be quickly oxidized to produce more toxic derivates, such as aldehydes.  相似文献

This study was conducted to create a nutritional database on brown seaweeds and to popularize their consumption and utilization in Iran. The fatty acid contents, amino acids profiles, and certain mineral elements composition of some brown seaweeds, Padina pavonica (L.) Thivy, Dictyota dichotoma (Huds.) J. V. Lamour., and Colpomenia sinuosa (Mert. ex Roth) Derbés et Solier were determined. Total lipid content ranged from 1.46 ± 0.38 to 2.94 ± 0.94 g · 100 g?1dry weight (dwt), and the most abundant fatty acids were C16:0, C18:1, C20:4 ω6, and C20:5 ω3. The unsaturated fatty acids predominated in all species and had balanced sources of ω3 and ω6 acids. Highest total polyunsaturated fatty acid (PUFA) levels occurred in C. sinuosa. The protein content of D. dichotoma was 17.73 ± 0.29 g · 100 g?1dwt, significantly higher than the other seaweeds examined. Among amino acids essential to human nutrition, methionine (Met; in D. dichotoma and P. pavonica) and lysine (Lys; in C. sinuosa) were present in high concentrations. The crude fiber content varied by 9.5 ± 11.6 g · 100 g?1dwt in all species. Chemical analysis indicated that ash content was between 27.02 ± 0.6 and 39.28 ± 0.7 g · 100 g?1dwt, and that these seaweeds contained higher amounts of both macrominerals (7,308–9,160 mg · 100 g?1dwt; Na, K, Ca) and trace elements (263–1,594 mg · 100 g?1dwt; Fe, Ni, Mn, Cu, Co) than have been reported for edible land plants. C. sinuosa had the highest amount of Ca, Fe, and a considerable content of Na was measured in P. pavonica.  相似文献

1. While the balance of light and nutrients is known to influence the food quality of herbivores by altering algal phosphorus and nitrogen content, the combined effects of light and nutrients on fatty acid synthesis in freshwater periphyton are relatively unknown. In this study, we manipulated light and phosphorus concentration in large, flow‐through experimental streams to examine their effects on both elemental stoichiometry and fatty acid content in periphyton. 2. Two levels of phosphorus (4 and 80 μg L?1) and three of light (17, 40, 110 μmol photons m?2 s?1) were applied in a factorial design in two separate experiments. Diatoms dominated periphyton communities in both experiments, comprising >95% of algal biovolume. Periphyton growth in the streams was simultaneously affected by both resources, even at low rates of supply. 3. Periphyton C/P and C/N ratios increased with light augmentation and decreased with phosphorus enrichment, and consistent with the light : nutrient hypothesis (LNH). Light effects were strongest in streams with low phosphorus concentrations. 4. Periphyton fatty acids reflected the dominance of diatoms : palmitic (16 : 0), palmitoleic (16 : 1ω7) and eicosapentanoic (20 : 5ω3) were the principal saturated (SAFA), monounsaturated (MUFA) and polyunsaturated fatty acids (PUFA), respectively. Linoleic (18 : 2ω6) and linolenic (18 : 3ω3) acids, characteristic of chlorophytes and cyanophytes, were rare, comprising <2% of total fatty acids. 5. Periphyton fatty acid profiles were highly sensitive to light and phosphorus. The proportion of fatty acids comprised by SAFA and MUFA increased with light augmentation and decreased with phosphorus enrichment, whereas PUFA decreased with light and increased with phosphorus. Light effects on fatty acid composition were strongest in phosphorus‐poor streams. PUFA declined with increasing light/phosphorus ratios in the streams, whereas ‘energy’ fatty acids (16 : 0 and 16 : 1) increased. The ratio of SAFA/PUFA was strongly and positively correlated with C/P and C/N ratios. SAFA and MUFA, normalised to dry mass, increased two‐ to threefold with increasing light, while PUFA normalised to dry mass was not significantly affected by light. 6. Similarities in the responses of fatty acids and elemental stoichiometry to light and phosphorus treatments suggested that they were influenced by a common mechanism. Both components of food quality appeared to be sensitive to light‐regulated rates of carbon fixation which, when coupled with insufficient supplies of phosphorus, caused diatom cells to store surplus carbon in SAFA, MUFA and other carbon‐rich compounds that diluted both essential fatty acids and mineral nutrients.  相似文献

The effects of the triazine herbicide, simazine, on photosynthetic oxygen evolution and growth rate in photoacclimated populations of Anabaena circinalis Rabenhorst were investigated. Chemostat populations were acclimated to photon flux densities (PFDs) of 50, 130, and 230 μmol·m?2·s?1 of photosynthetic active radiation (PAR), Decreases in chlorophyll a (Chl a). c-phycocyanin (CPC), and total carotenoid (TCar) contents and CPC: Chl a and CPC: TCar ratios of populations coincided with increasing PFD, Polynomial regression models that characterize inhibition of photosynthesis for populations acclimated to 50 and 130 μmol photons·m?2·s?1 PAR were distinct from the model for populations acclimated to 230 μmol photons·m?2·s?1 PAR. Simazine concentrations that, depressed oxygen evolution 50% compared to controls decreased with increasing PFD. Increases and decreases in both biomass and growth rate coincided with increasing PFD and simazine concentration, respectively. Simazine concentrations that depressed growth rate 50% compared to controls increased with decreasing PFD. The marine diatom Thalassiosira pseudonana (Hustedt, clone 3H) Hasle and Heimdal was cultured under three different light regimes: 100 μmol photon · m?2· s?1 on 12:12 h light : dark (L:D) cycles; 50 μmol photon · m?2· s?2 on 24:0 h L:D; and 100 μmol photon · m?2· s?1 on 24:0 h L:D. It was harvested during logarithmic and stationary phases for analysis of biochemical composition. Across the different light regimes, protein (as % of organic weight) was highest in cells during logarithmic phase, whereas carbohydrate and lipid were highest during stationary phase. Carbohydrate concentrations were most affected by the different light regimes; cells grown under 12:12 h L:D contained 37–44% of the carbohydrate of cells grown under 24:0 h L:D. Cells in logarithmic phase had high proportions of polar lipids (79 to 89% of total lipid) and low triacylglycerol (≤10% of total lipid). Cells in stationary phase contained less polar lipid (48 to 57% of total lipid) and more triacylglycerol (22 to 45% of total lipid). The fatty acid composition of logarithmic phase cells grown under 24:0 h L:D were similar, but the 100 μmol photon · m?2· s?1 (12:12 h L:D) cells at the same stage contained a higher proportion of polyunsaturated fatty acids (PUFAs) and a lower proportion of saturated and monounsaturated fatty acids due to different levels of 16:0, 16:1(n-7), 16:4(n-1), 18:4(n-3), and 20:5(n-3). With the onset of stationary phase, cells grown at 100 μmol photon · m?2· s?1 (both 12:12 and 24:0 h L:D) increased in proportions of saturated and monounsaturated fatty adds and decreased in PUFAs. Concentrations (% organic or dry weight) of 14:0, 16:0, 16:1(n-7), 20:5(n-3), and 22:6(n-3) increased in cells of all cultures during stationary phase. The amino acid compositions of cells were similar irrespective of harvest stage and light regime. For mariculture, the recommended light regime for culturing T. pseudonana will depend on the nutritional requirements of the animal to which the alga is fed. For rapidly growing bivalve mollusc larvae, stationary-phase cultures grown under a 24:0 h L:D regime may provide more energy by virtue of their higher percentage of carbohydrate and high proportions and concentrations of energy-rich saturated fatty acids.  相似文献

Eight species of marine phytoplankton showed significant variation in the relative amount of some fatty acids (FAs) in response to variation in temperature. Large changes in relative amounts of certain FAs occurred as a result of a 15° C change in growth temperature. For example, 14:0 increased from ?4% of total FAs at 10° C to > 20% at 25° C for Chaetoceros simplex and Isochrysis aff. galbana but decreased for Phaeodactylum tricornutum. The percentage of the polyunsaturated fatty acid (PUFA) 16:ω1 was consistently greater at 10° C than at 25° C, and the converse was usually true for 16: 4ω3. Calculated over all eight species, there was a modest but significant inverse relationship between the percentage of PUFAs and temperature. Only for Thalassiosira pseudonana was the percentage of either of the PUFAs and nutritionally essential fatty acids (EFAs) also an inverse function of temperature. For T. pseudonana, the percentage of the EFA 22:6ω3 decreased linearly with increasing temperature over the range from 10 to 25° C. For three species, the ratio of unsaturated/saturated FAs was correlated with growth rate when growth rate was controlled by variation in irradiance and temperature. Only for Thalassiosira pseudonana was the ratio of unsaturated/saturated FAs also an inverse function of temperature alone.  相似文献

Intra‐thallus variation in fatty acid and pigment contents and profiles was investigated in five species of Laminariales (Alaria esculenta, Laminaria digitata, Laminaria hyperborea, Saccharina latissima, and Saccorhiza polyschides), and three Fucales (Ascophyllum nodosum, Fucus serratus, and Himanthalia elongata). Significant variation occurred across all species and compounds examined. Total fatty acids were generally higher in the fronds, with highest levels and largest variability observed in A. nodosum (1.5% of dry weight (DW) in the base, 6.3% of DW in frond tips). Percentages of the omega‐3 fatty acids 18:4 n‐3 and 20:5 n‐3 were generally higher in more distal parts, while 20:4 n‐6 exhibited a contrasting pattern, with higher levels in basal structures and holdfasts. Trends for pigments were similar to those for fatty acids in Laminariales. In the Fucales, highest levels were detected in the mid‐fronds, with lower concentrations in meristematic areas. Highest levels and greatest variability in pigments (e.g., chl a) was observed in F. serratus (1.07 mg · g?1 DW in the base, 3.04 mg · g?1 DW in the mid frond). Intra‐thallus variability was attributed to physiological functions of the respective thallus sections, e.g., photosynthetic activity, meristematic tissue, and to variations in physical attributes of the structures investigated. Regarding potential commercial nutritional applications, fronds appeared to represent most suitable source materials, due to higher levels of pigments, polyunsaturated fatty acids, and more preferable omega‐3/omega‐6 ratios.  相似文献

The effects of irradiance on the biochemical composition of the prymnesiophyte microalga, Isochrysis sp. (Parke; clone T-ISO), a popular species for mariculture, were examined. Cultures were grown under a 12:12 h light: dark (L:D) regime at five irradiances ranging from 50 to 1000 μE·m 2·s?1 and harvested at late-logarithmic phase for analysis of biochemical composition. Gross composition varied aver the range of irradiances. The highest levels of protein were present in cells from cultures grown at 100 and 250 μE·m 3·s1, and minimum levels of carbohydrate and lipid occurred at 50 μE·m?2·s?1. Because the cell dry weight was reduced at lower irradiances, different trends were evident when results were expressed as percentage of dry weights. Protein percentages were highest at Wand 100 μE·m?2·s?1 and carbohydrate at 100 μE·m?2·s?1. The composition of amino acids did not differ over the range of irradiances. Glutamate and aspartate were always present in high proportions (9.0–13.5%); histidine. methionine, tryptophan, cystine, and hydroxy-proline were minor constituents (0.0–2.6%). Glucose was the predominant sugar in all cultures, ranging from 23.0% (50 μE·m?2·s?1) to 45.0% (100 μE·m?2·s?1) of total polysaccharide. No correlation was found between the proportion of any of the sugars and irradiance. The proportions of the lipid class components and fatty acids showed little change with irradiance. The main fatty acids were 14:0, 16:0, 16:1(n-7), 18:1(n-9), 18:3(n-3). 18:4(n-3), 18:5(n-3), and 22:6(n-3). Proportions of 22: 6(n-3) increased, whereas l8:3(n-3). 18:3(n-6). and 18:4(n-3) decreased, with increasing irradiance. Pigment concentrations were highest in cultures grown at 50 μE·m?2·s?1, except for fucoxanthin and diadinoxanthin (100 μE·m?2·s?1). The concentrations of accessory pigments correlated with chlorophyll a, which decreased in concentration with increasing irradiance. On the basts of biochemical composition, an irradiance of 100 μE·m?1·s?1 (12:12 h L:D cycle)for the culture of Isochrysis sp. (clone T-ISO) may provide optimal nutritional value for maricultured animals, although feeding trials are now necessary to substantiate this.  相似文献

The composition of fatty acids and contents of eicosapentaenoic acid (EPA) and polyunsaturated fatty acids (PUFAs) of the economically important marine diatom, Phaeodactylum tricornutum (Bohlin), were investigated to see whether reducing the culture temperature enhances the production of EPA and PUFAs. The contents of EPA and PUFAs of P. tricornutum were found to be higher at lower temperature when cultured at 10, 15, 20, or 25°C. When the cells grown at 25°C were shifted to 20, 15, or 10°C, the contents per dry mass of PUFAs and EPA increased to the maximal values in 48, 24, and 12 h, respectively. The highest yields of PUFAs and EPA per unit dry mass (per unit volume of culture) were 4.9% and 2.6% (12.4 and 6.6 mg·L?1), respectively, when temperature was shifted from 25 to 10°C for 12 h, both being raised by 120% compared with the control. The representative fatty acids in the total fatty acids, when temperature was lowered from 25 to 10°C, decreased proportionally by about 30% in C16:0 and 20% in C16:1(n?7) but increased about 85% in EPA. It was concluded that lowering culture temperature of P. tricornutum could significantly raise the yields of EPA and PUFAs.  相似文献
