Summary Patterns of colonization by queens and incipient nest survival of the leaf-cutting antsAcromyrmex niger andAcromyrmex balzani were studied by means of belt transects and individually marked incipient nests. No relation was found between colony density and the number of colonization attempts. Both species are not claustral, and high rates of queen mortality were attributed to conspecific executions and predation. Of other discernable mortality factors, failure of fungal garden establishment was the most important. Only 34 of 296A. balzani and 13 of 154A. niger marked colonies were alive at the end of one year. These figures are higher than those reported for species ofAtta. These results are contrasted with those of claustral-foundingAtta species. Small colonies are occasionally raided by larger colonies which robbed brood.  相似文献

Streptomyces griseus produces an antibiotic substance against Colletotrichum lindemuthianum because, in dual culture on potato dextrose agar (PDA), a large inhibitory zone was formed between the colonies of S. griseus and C.lindemuthianum. When S. griseus was placed adjacent to a colony of C. lindemuthianum in PDA, S.griseus grew poorly in PDA but it thrived and sporulated profusely when it came in contact with a colony of C. lindemuthianum indicating that S.griseus might require some essential nutrients from C.lindemuthianum. Scanning electron microscopy showed that hyphae of S.griseus in contact with C.lindemuthianum produced appressorium-like swellings or simply grew on the hyphal surface of C.lindemuthianum. Internal parasitism was evidenced by the presence of hyphae and conidia of the mycoparasite inside the host hyphae. Internal parasitism resulted in formation of several blebs. Cell walls of the parasitized hyphae degenerated having a sponge-like texture. The blebs broke off and left rounded holes on the hyphae.  相似文献

Fungal mycelia are exposed to heterogenic substrates. The substrate in the central part of the colony has been (partly) degraded, whereas it is still unexplored at the periphery of the mycelium. We here assessed whether substrate heterogeneity is a main determinant of spatial gene expression in colonies of Aspergillus niger. This question was addressed by analyzing whole-genome gene expression in five concentric zones of 7-day-old maltose- and xylose-grown colonies. Expression profiles at the periphery and the center were clearly different. More than 25% of the active genes showed twofold differences in expression between the inner and outermost zones of the colony. Moreover, 9% of the genes were expressed in only one of the five concentric zones, showing that a considerable part of the genome is active in a restricted part of the colony only. Statistical analysis of expression profiles of colonies that had either been or not been transferred to fresh xylose-containing medium showed that differential expression in a colony is due to the heterogeneity of the medium (e.g., genes involved in secretion, genes encoding proteases, and genes involved in xylose metabolism) as well as to medium-independent mechanisms (e.g., genes involved in nitrate metabolism and genes involved in cell wall synthesis and modification). Thus, we conclude that the mycelia of 7-day-old colonies of A. niger are highly differentiated. This conclusion is also indicated by the fact that distinct zones of the colony grow and secrete proteins, even after transfer to fresh medium.  相似文献

The ecology of Chroothece was studied in the highly calcareous Río Chícamo, south-east Spain, in order to explain its success there, but rarity elsewhere. The river, which originates mainly from an underground aquifer, has water with high conductivity, sulphate and nitrate but low phosphate concentrations, the latter mainly organic. Chroothece occurs in mats and in lobed colonies reaching 4 cm in the broadest dimension. The colony surface consists of one layer of cells, each of which is attached to a stalk, which dichotomizes when the cell divides; stalks often extend to the colony base. The central region of many mat cells and almost all colony cells has a yellow to orange-brown colour, associated with the numerous lipid droplets densely covering the surface of the pyrenoid and arms of the star-shaped chloroplast. Field material and laboratory isolates indicate that stalk formation occurs under moderate P limitation and both stalks and cell sheath show high phosphatase activities. This also occurred in a culture collection strain maintained for 30 years in a very P-rich medium, but then transferred to a moderately P-limiting medium (c. 0.9 mg l?1). We suggest that colony formation is initiated by aggregation of motile cells following P pulses in the water. Comparisons are made with Rivularia, a competitor in this nitrate-rich river, in spite of being a N2-fixer. One difference is that Chroothece cells lie at the periphery of the colonies and are therefore exposed to maximum sunlight, whereas Rivularia trichomes grow inside colonies with photoprotection by scytonemin. However, the ability to withstand heavy grazing pressure may be an especially important factor favouring Rivularia here.  相似文献

Antifungal activity of two imidazoles (miconazole and ketoconazole) and one polyene (amphotericin B) was evaluated using an automatic growth analysis system. Spores ofAspergillus niger were inoculated on the polylysine-coated glass bottom of a culture vessel. A colony formed in liquid medium was exposed to an antifungal agent and subsequently washed. Based on the dynamic growth rate of a test hypha selected from the colony in response to the antifungal agent, the minimum inhibitory concentration (MIC) was evaluated. The influence of time of reading (1, 2 and 3 h after washing) on the MIC determined was investigated. MICs for test hyphae subjected to antifungal pre-treatment were compared with those for hyphae without pre-treatment. Hyphae pre-treated with an antifungal agent for 1 h were found to become adapted and tolerant to that antifungal agent. Hyphae exposed and adapted to an imidazole obtained tolerance to amphotericin B as well as to the other imidazole.  相似文献

The algal genus Scenedesmus is famous for its highly phenotypic plasticity in response to various environmental factors. In laboratory, axenic cultures of Scenedesmus often fail to form colonies and remain in unicellular morph. To examine whether unicellular Scenedesmus can form colonies after exposure to natural lakes, dialysis bags and plastic bottles which contained the precultured Scenedesmus obliquus were exposed in two lakes with different nutrient levels for four weeks. Results showed S. obliquus grew well in dialysis bags but not in plastic bottles; S. obliquus can form colonies when incubated in dialysis bags which allow exchange with the aquatic environment of the substances, regardless of the nutrient levels of the two lakes. However, no colonies were observed in S. obliquus incubated in plastic bottles exposed in both lakes. This suggested that active growth and zooplankton infochemicals contributed together to the colony formation of S. obliquus exposed in situ environment.  相似文献

Aspergillus niger hyphae were found to grow with unliquefied potato starch under aerobic conditions, but did not grow under anaerobic conditions. The raw culture ofA. niger catalyzed saccharification of potato starch to glucose, producing approximately 12 g glucose/L/day/ The extracellular enzyme activity was decreased in proportion to incubation time, and approximately 64% of initial activity was maintained after 3 days. At 50°C,A. niger hyphae growth stopped, while the extracellular enzyme activity peaked. On the basis of theA. niger growth property and enzyme activity, we designed a serial bioreactor system composed of four different reactors. Fungal hyphae were cultivated in reactor I at 30°C, uniquefied starch was saccharified to glycose by a fungal hyphae culture in reactors II and III at 50°C, and glucose was fermented to ethanol bySaccharomyces cerevisiae in reactor IV. The total glucose produced by fungal hyphae in reactor I and saccharification in reactor II was about 42 g/L/day. Ethanol production in reactor IV was approximately 22 g/L/day, which corresponds to about 79% of the theoretical maximum produced from 55 g starch/L/day.  相似文献

THE MORPHOGENESIS AND POSSIBLE EVOLUTIONARY ORIGINS OF FUNGAL SCLEROTIA   总被引:2,自引:0,他引:2  
1. Fungal sclerotia are able to survive adverse conditions for long periods and they are formed by many important plant pathogens. An understanding of the factors involved in their initiation and development may lead to a method of repressing their formation in nature, thereby reducing the chances of survival of fungi that depend on them as persistent resting stages in their life-cycles. Also, data on sclerotial morphogenesis may be applicable to other multihyphal fungal structures. 2. There are three types of sclerotial development. The most primitive and least common is the loose type, which is illustrated by Rhizoctonia solani. The sclerotium forms by irregular branching of the mycelium followed by intercalary septation and hyphal swelling. When mature, it consists of loosely interwoven hyphae that are rich in food reserves and darkly pigmented. The main types of development are terminal and lateral. The former develops from the coalescence of initials that are produced by a well-defined pattern of branching at the tip of a hypha or tips of closely associated hyphae, e.g. Botrytis cinerea. Lateral sclerotia are formed by the interweaving of side branches of one or several main hyphae. When only one main hypha is involved the sclerotium is of the lateral, simple type, e.g. Sclerotinia gladioli. If several main hyphae give rise to a sclerotium, the term strand type has been used. Sclerotium rolfsii is the classical example. 3. There is a considerable literature on the effects of environmental conditions on the initiation, development and maturation of sclerotia but few attempts have been made to interpret the data. Phenolics and/or polyphenol oxidases have been found to be connected with morphogenesis of the protoperithecium of Neurospora crassa, the perithecium of Podospora anserina and of Hypomyces sp. and the basidiocarp of Schixophyllum commune. A close correlation has been shown between melanin synthesis and microsclerotial development by Verticillium but there appears to be no literature on the role of phenolics and polyphenol oxidases in the morphogenesis of sclerotia. Possibly these substances may inhibit growth of the apices of main hyphae by changing the permeability of the membrane, by inducing a thickening of the cell wall at the tip or by reducing the plasticity of the wall. Such a check in growth could trigger-off the formation of initials close to the margin of the colony or elsewhere in the culture. Sulphydryl groups and disulphide bonds are of great significance in morphogenesis of organisms and are probably involved in sclerotial initiation. The formation of a large number of hyphal branches is a prerequisite for sclerotial initiation and mycelial branching is possible only if there is plasticity of hyphal walls. The ability of the wall to be moulded is possibly related to changes in the sulphur linkages of the protein of the protein-carbohydrate complexes of the cell wall and could be influenced by sulphur availability or the activity of specific enzymes. 4. After a sclerotial primordium has been initiated, further increase in size will depend on the continued, active translocation of nutrients to the site of development. Movement of nutrients to sclerotia is through a few translocatory hyphae. Presumably, nutrients will continue to move into the young sclerotium as long as a concentration or pressure gradient is maintained. Energy and substances for the formation of new branches are supplied in this way and as the requirements for hyphal branches are reduced, excess nutrients become available for conversion to inactive or insoluble reserves and for exudation. The exudates are often complex, consisting of proteins, including enzymes, lipids and carbohydrates. Many sclerotia have a mucilaginous matrix in which the medullary hyphae are embedded. Sclerotium-forming, fungal species that are not regarded as having such a matrix appear to secrete a layer of mucilage over the surface of sclerotial hyphae. This mucilage could have a morphogenetic function and serve as an adhesive which loosely binds hyphae together. More permanent unions are by hyphal fusions or anastomoses. 5. The sclerotium matures within a few days of attaining its maximum size. The rind effectively seals off the medullary hyphae from the surroundings and the translocatory hyphae cease to function. Thus the sclerotium is isolated both physiologically and nutritionally. The endogenous reserves enable the structure to exist in the absence of exogenous nutrients and then, when conditions become suitable, to germinate. 6. The sclerotium appears to provide an example of convergent evolution whereby analogous structures, which have become adapted to resist adverse conditions, have evolved. Data are available mainly for Typhula spp. and ScZerotinia spp. Sclerotia may be degenerate sexual reproductive structures, hyphal aggregates that have developed from closely interwoven conidiophores and undifferentiated conidia or they may be modified vegetative structures.  相似文献

1. The performance of ant colonies depends on different factors such as nest site, colony structure or the presence of pathogens and social parasites. Myrmica ants host various types of social parasites, including the larvae of Maculinea butterflies and Microdonmyrmicae (Schönrogge) hoverfly. How these social parasites affect host colony performance is still unexplored. 2. It was examined how the presence of Maculinea teleius Bergsträsser, Maculinea alcon (Denis & Schiffermüller), and M. myrmicae larvae, representing different feeding and growth strategies inside host colonies, is associated with worker survival, the number of foragers, and colony productivity parameters such as growth and reproduction. 3. It was found that the presence of social parasites is negatively associated with total colony production and the production of ant larvae and gynes. Male production was lower only in nests infested by M. teleius, whereas the number of worker pupae was significantly higher in all types of infested colonies than in uninfested colonies. Laboratory observations indicated that nests infested by Maculinea larvae are characterised by a higher number of foragers compared to uninfested nests but we did not find differences in worker survival among nest types. 4. The observed pattern of social parasite influence on colony productivity can be explained by the feeding strategies of parasitic larvae. The most negative effect was found for M. teleius, which feeds on the largest host brood and eliminates a high number of sexual forms. The strong, adverse influence of all studied parasite species on gyne production may result in low queen production in Myrmica populations exposed to these social parasites.  相似文献

Monoclonal antibodies are an important tool in the study of botryllid ascidians’ immunology and developmental biology. Here we describe the development of a panel of 38 monoclonal antibodies that are specific to Botryllus schlosseri (Ascidiacea; subfamily Botryllinae) cell surface antigens. Many of these hybridomas recognize (by enzyme-linked immunosorbent assay and immunohistochemistry) epitopes of Botrylloides subpopulations (SP) II and III from the Mediterranean coast of Israel and show, on blood cell smear assays, reactions with subsets of Botryllus circulating blood cells. Fluorescence-activated cell sorting analyses using antibodies positive for botryllid tissues revealed up to 3.6% positive cells. ELISA screenings were performed with 64 new monoclonal antibodies on 5 different individual botryllid ascidian colonies (B. schlosseri, Botrylloides). The positive antibodies in this panel identified a large number of different antigenic determinants, some of which distinguish Botryllus versus Botrylloides colonies, and other, different colonies within these two species, or different cell types within tissues, embryos, and buds of individual colonies. Only 21 monoclonal antibodies tested positive with all colonies. Cross-reactivity with at least one Botrylloides colony was recorded in 49 hybridomas that identified Botryllus cells. This wide panel of monoclonal antibodies is the first such detailed set of monoclonals available for studies on botryllid ascidians.  相似文献

In this study, we used two common ant species (Lasius niger and Lasius neoniger) to assay how they translate variation in the diet (both in composition and frequency) into growth. We measured colony development for over 8 months and measured several phenotypic traits of the worker caste, and examined whether forager preference corresponded with diet quality. Optimal colony growth was a balance between survival and growth, and each of these was maximized with different nutrient regimes. Interestingly, forager preference was not totally aligned with the diet that maximized colony growth. Our results highlight that: (a) organism and superorganism size are controlled by the same nutrients, and this may reflect a common molecular basis for size across life's organizational levels, (b) there are nutrient trade‐offs that are associated with life‐history trade‐offs, likely leading to selection for a balanced diet, and (c) the connection between the preference of foragers for different nutrients and how nutrient combinations affect colony success and demographics are complex and only beginning to be understood.  相似文献

Abstract Colonially nesting birds have been implicated in the destruction of their nest site vegetation, the most likely mechanism being through an overload or imbalance of soil nutrients. The numbers of egrets in coastal New South Wales colonies have increased rapidly in the second half of this century since cattle egrets (Ardeola ibis) were introduced. This species now nests colonially with three native species: great (Egretta alba), intermediate (Egretta intermedia) and little egrets (Egretta garzetta). Thus there is potential for more rapid degradation of the colony site. We measured the levels of phosphorus in surface sediments, and phosphorus and nitrogen in water at eight colony site wetlands, and compared each to four other wetlands in the same region where there were no egret colonies. We conclude that levels of both nutrients were elevated in colony wetlands. The implications of this result for death of colony site vegetation and the conservation of the three native egret species are briefly discussed.  相似文献

Ants are highly abundant generalist predators and important ecosystem engineers which can strongly affect the composition of animal communities. We manipulated the density of the ant species Lasius niger with baits in a small‐scale field experiment to study the role of intraguild predation, top‐down control and bottom‐up effects of ants in a dry grassland surrounded by agricultural fields. Two different kinds of baits (honey and tuna) were presented near to the nests and at a distance of 2 m from six L. niger colonies in a dry grassland habitat, where L. niger was a highly abundant, omnipresent species. The experiments were performed for 1 month in spring. Additionally, the natural abundance of L. niger varying with the distance to their nests was used to study the effects on spiders and potential prey groups. The activity of L. niger was significantly higher at tuna baits compared with that at honey baits and empty control dishes. We found no effects of higher activity of L. niger on the arthropod community. However, there is evidence for a facilitation effect of ants on Collembola near to their colonies, probably due to habitat modification, which also influenced the density of Linyphiidae. Both groups had up to four times higher denisities next to L. niger colonies than at a distance of 2 m. Furthermore, δ13C values demonstrated that linyphiid spiders and L. niger predominantly feed on Collembola. We conclude that there is no evidence of top‐down effects of L. niger in a grassland in spring, but we found a facilitation of linyphiid spiders and their prey by the ants, which acted as ecosystem engineers.  相似文献

Foraging behavior is a critical adaptation by insects to obtain appropriate nutrients from the environment for development and fitness. Bumble bees (Bombus spp.) form annual colonies which must rapidly increase their worker populations to support rearing reproductive individuals before the end of the season. Therefore, colony growth and reproduction should be dependent on the quality and quantity of pollen resources in the surrounding landscape. Our previous research found that B. impatiens foraging preferences to different plant species were shaped by pollen protein:lipid nutritional ratios (P:L), with foragers preferring pollen species with a ~5:1 P:L ratio. In this study, we placed B. impatiens colonies in three different habitats (forest, forest edge, and valley) to determine whether pollen nutritional quality collected by the colonies differed between areas that may differ in resource abundance and diversity. We found that habitat did not influence the collected pollen nutritional quality, with colonies in all three habitats collecting pollen averaging a 4:1 P:L ratio. Furthermore, there was no difference in the nutritional quality of the pollen collected by colonies that successfully reared reproductives and those that did not. We found however, that “nutritional intake,” calculated as the colony‐level intake rate of nutrient quantities (protein, lipid, and sugar), was strongly related to colony growth and reproductive output. Therefore, we conclude that B. impatiens colony performance is a function of the abundance of nutritionally appropriate floral resources in the surrounding landscape. Because we did not comprehensively evaluate the nutrition provided by the plant communities in each habitat, it remains to be determined how B. impatiens polylectic foraging strategies helps them select among the available pollen nutritional landscape in a variety of plant communities to obtain a balance of key macronutrients.  相似文献

Morphological variations of Gloeotaenium loitlesbergarianum Hansgirg were studied both in cultures and in nature. In cultures, the alga exhibits considerable variation in the number of cells per colony, ranging from unicells to colonies with more than four cells. The characteristic band was also absent in cultures. In nature, colonies resembling the culture material of Gloeotaenium also occur. The morphology of the alga varies depending on the nature and composition of the nutrients available. The study shows that Gloeotaenium may exhibit polymorphism in nature as well.  相似文献

Nostoc parmelioides colonies housing dipteran larvae (Cricotopus) had higher rates of weight specific photosynthesis than colonies without the larvae. A change in colony shape, which allowed the alga to be exposed to higher light intensities, occurred, in the presence of the larvae. This change in morphology together with potential nutrient additions by the larvae and other effects may have caused the increase in photosynthetic rates. Nostoc colonies were typically found in open areas of small streams in western Oregon mountains where the ability to respond to high light would be advantageous in supporting the metabolically expensive process of nitrogen fixation.  相似文献

The dimorphic yeasts Candida boidinii and Yarrowia lipolytica were applied as model organisms to study mycelial growth. A mathematical model of hybrid cellular automaton type was developed to analyze the impact of different biological assumptions on the predicted development of filamentous yeast colonies. The one-dimensional model described discrete cells and continuous distribution of nutrients. The simulation algorithm accounted for proliferation of cells, diffusion of nutrient, as well as biomass decay and recycling inside the mycelium. Simulations reproduced the spatio-temporal development of C. boidinii colonies when a diffusion-limited growth algorithm based on the growth of pseudohyphal cells was applied. Development of Y. lipolytica colonies could only be reproduced when proliferation was restricted to the colony boundary, and cell decay and biomass recycling were incorporated into the model. The results suggested that cytoplasm, which served as the secondary nutrient resource, had to be translocated inside the hyphal network.  相似文献
