Soybean seedlings treated with ethylene exhibited small increases in ribonucleic acid content in the elongating section of the hypocotyl. Chromatin isolated from the elongating section of ethylene-treated seedlings showed a 35 to 60% increase in the capacity for RNA synthesis. The ethylene-induced response was saturated at 1 microliter/liter of ethylene and was fully expressed after 3 hours. Auxin caused marked accumulation of RNA and DNA in the elongating and basal tissue of the hypocotyl. Chromatin isolated from these auxin-treated tissues showed an 8- to 10- fold increase in RNA synthetic capacity as measured in vitro. Ethylene added with auxin reduced the auxin enhancement of nucleic acid synthesis in the elongating and basal tissues. Both ethylene and auxin treatment of the seedlings inhibited nucleic acid accumulation and chromatin activity in the apical tissue. Ethylene did not appear to mediate the auxin effects on nucleic acid synthesis in soybean hypocotyl with the possible exception of inhibition in the apical tissue.  相似文献

Summary Regeneration of adventitious shoots was obtained in over 80% of explants, consisting of wounded cotyledonary nodes of Acacia mangium, by culturing germinated seedlings on DKW medium with combinations of N6-benzyladenine and either thidiazuron or N-(2-chloro-4-pyridyl)-N-phenylurea. Electron microscopy showed the presence of adventitious buds arising from wound tissue of the cotyledons and cotyledonary nodes. Shoot regeneration was also obtained at lower frequency in isolated cotyledon explants cultured with 6% sucrose alone (10%), or with 3% sucrose and 30.0 mg l−1 (0.1 μM) 2–4-dichlorophenoxyacetic acid (2,4-D; 16%). With 2,4-D,>60% of explants produced organized structures but these did not develop into shoots or somatic embryos. Shoot formation was not induced in either hypocotyl or root explants.  相似文献

DNA-synthesis in the hypocotyls of Sinapis alba L. was studied with H3-thymidine labelling. Cells from hypocotyl segments were stained by the Feulgen-method and squash preparations were made. The following labelling patterns were observed: 1. Labelling of the chromocentres only. 2. Nuclear area evenly labelled. 3. No radioactivity in the chromocentres. This pattern was rarely seen. — The frequency of the first two types in different tissue segments is not equal. In segments with more differentiated cells there was an increase in the percentage of nuclei with radioactivity only in the chromocentres. This could be due to a prolongation of the phase of synthesis in the chromocentres in this tissue. — The total number of labelled nuclei decreases basipetally as well as with the age of the hypocotyl. In hypocotyls of seedlings older than 52 hrs radioactivity appeared only sporadically in the nuclei. The decrease in the number of labelled nuclei is faster than the decline of the corresponding measurable total DNA synthesis in the hypocotyl. This can either be due to extra nuclear DNA synthesis or depend on an increase in DNA synthesis in the later replicating heterochromatic region of the nucleus.  相似文献

Summary Germination of tamarind seeds in medium containing thidiazuron (TDZ) resulted in induction of nodular protrusions in and around the cotyledonary node meristem. The structures developed radially in well-defined circles and subsequently spread towards the cotyledonary bridge and also in the proximal part of the hypocotyl. The structures developed into shoots on transfer to medium devoid of growth regulators. Histological studies revealed that the protrusions initiated from the nodal meristem and extended to the non-meristematic region between the two meristems and also in the proximal part of the hypocotyl in seedlings germinated in 9.08 μM TDZ. Newly formed cell layers and less-differentiated meristematic protrusions were also seen. With the increase in the distance from the meristem, the buds were less differentiated; in the proximal part of the hypocotyl only the multiple layers of meristematic cells were noted. With extension of the period of incubation, the TDZ-induced meristematic activity extended laterally in circles towards the neighboring region. The radial spread of the meristematic activity from the center of the nodal meristem was also evident at 18.16 μM TDZ. From the pattern of the morphogenic development and the histological studies it may be hypothesized that in tamarind, TDZ influences the existing meristems specifically. Subsequently de novo organogenesis is triggered in the neighboring cells.  相似文献

Seed and developing seedlings of Populus deltoides Bartr. ex Marsh., through 2 wk old, were embedded in Paraplast or Spurr's resin and serially sectioned at 7 or 2 μm, respectively, to examine vascular development in the root-hypocotyl transition region. The procambial template is well established in the dormant embryo. Protophloem forms a continuous system connecting the root with the cotyledons but protoxylem is restricted primarily to the cotyledons. With the onset of germination there is rapid elongation of both the root and hypocotyl. The basipetal progression of protoxylem from an initiating center at the cotyledonary node is correlated with the establishment of a second initiating center in the collet at the base of the hypocotyl. The relation between procambium, initiating layer, and metacambium, and their derivatives is similar to that described for the cottonwood shoot. The primary vascular system of cottonwood should be considered a single unit, both conceptually and functionally. The vascular pattern of the root may be, in part, determined by a basipetal stimulus from the cotyledonary node.  相似文献

Soybean seedlings (Glycine max L.) were germinated and dark-grown in water-saturated vermiculite (water potential = −0.01 megapascal) for 48 hours, then transferred either to water-saturated vermiculite or to low water potential vermiculite (water potential = −0.30 megapascal). A decrease in growth rate was detectable within 0.8 hour post-transfer to low water potential vermiculite. A fourfold increase in the abscisic acid content of the elongating region was observed within 0.5 hour. At 24 hours post-transfer, hypocotyl elongation was severely arrested and abscisic acid reached its highest measured level: 3.7 nanograms per milligram dry weight (74-fold increase). A comparison of the polyA+ RNA populations isolated at 24 hours post-transfer from the elongating region of water-saturated and low water potential vermiculite-grown seedlings was made by two-dimensional (isoelectric focusing-sodium dodecyl sulfate) polyacrylamide gel analysis of in vitro translation products. It revealed both increases and decreases in the relative amounts of a number of translation products. Rewatering seedlings grown in low water potential vermiculite at 24 hours post-transfer led to a total recovery in growth rate within 0.5 hour, while abscisic acid in the elongating hypocotyl region required 1 to 2 hours to return to uninduced levels. Application of 1.0 millimolar (±) abscisic acid to well-watered seedlings resulted in a 48% reduction in hypocotyl growth rate during the first 2 hours after treatment. Plants treated with abscisic acid for 24 hours had a lower polysome content than control plants. However, hypocotyl growth inhibition in abscisic acid-treated seedlings preceded the decline in polysome content.  相似文献

Regeneration of subterranean clover (Trifolium subterraneum L.) was achieved by both shoot organogenesis and somatic embryogenesis. Shoots derived via organogenesis were initiated from the hypocotyls of mature imbibed seed. The hypocotyl, including the emerging radicle, was sliced longitudinally into two halves and cultured on shoot induction medium. After 30 days, adventitious shoots were formed from the hypocotyl region while the radicle showed no development. Shoots were then subcultured onto shoot multiplication medium and finally onto a root initiation medium. Histological studies revealed that shoots arose de novo and did not originate from pre-existing meristems. In the second regeneration protocol, shoot apical meristems from young seedlings were induced to form callus. Following four to six weeks culture in the dark, somatic embryos appeared spontaneously on the calli. A majority of embryos had a well-defined root pole, two cotyledonary lobes, and were capable of germination, albeit at a low frequency. Regenerated plants obtained from both protocols appeared phenotypically normal.  相似文献

Young seedlings of Lavatera cretica L. exhibit positive phototropism. The hypocotyl perceives unilateral illumination with blue light and curves towards the light source by unequal growth. In addition, the cotyledonary laminas perceive the vectorial component of unilateral illumination with blue light and reorient normal to the beam by creating a turgor differential in their pulvini. Excision of one cotyledon resulted in negative organotropic curvature of the hypocotyl, away from the remaining cotyledon. Illumination of the cotyledonary lamina did not participate in the phototropic curvature of the hypocotyl, so long as the lamina was free to reorient to face the beam. When the lamina was continuously exposed to vectorial photoexcitation, elongation of the hypocotyl on the side carrying the cotyledon could be enhanced, or inhibited, depending on the direction of the beam striking its lamina.  相似文献

Hormonal and plant factors regulating the development of the inhibited cotyledonary buds of Lycopersicon esculentum Mill. cv. `Fireball' seedlings were studied. Excision of the immature plumular leaves of 5- to 20- millimeter length significantly stimulated bud development after 2 to 4 days, but excision of leaves exceeding 20-millimeter length was without effect. Apical application of 20 microliters of 5 millimolar abscisic acid significantly promoted development of the cotyledonary buds after 6 days. A subapical ring of 0.1 millimolar concentration of 2,3,5-triiodobenzoic acid (TIBA) in lanolin significantly promoted cotyledonary bud development after 11 days. Twenty microliters of 0.1 millimolar 6-benzylaminopurine (BAP) applied directly to the cotyledonary bud loci significantly promoted bud development, but 1 micromolar gibberellin A4/7 was ineffective. Application of 0.1 millimolar BAP in lanolin to the petiole or hypocotyl was ineffective. However, application of 0.1 millimolar TIBA as a ring around the petioles of the cotyledons or 1-centimeter on the hypocotyl below the cotyledons significantly promoted cotyledonary bud development.  相似文献

Summary Plant regeneration, which is the major limiting factor for transformation of Cajanus cajan, has been obtained via multiple shoot formation from the cotyledonary node region of seedlings germinated on MS medium containing 2 mgl–1 6-benzylaminopurine. A mass of multiple shoot-initials formed at the axillary bud region of the cotyledonary node of the seedlings within two weeks. The cotyledonary node along with the mass of shoot-initials excised from the seedling, continued to form new shoot-initials on MS medium containing 6-benzylaminopurine (2 mgl–1) and supplemented topically with indole-3-acetic acid. The formation of new shoot-initials was also observed from the cotyledonary nodal explant, after cutting off its surface layers to completely remove the pre-existing shoot-initials and culturing it on 6-benzylaminopurine (2 mgl–1) containing medium. The shoots elongated rapidly on basal MS medium and rooted efficiently in MS medium supplemented with indole-3-butyric acid (0.5 mgl–1). Obtention of epinasty in the cotyledons of Sinapis alba under continuous far-red illumination. Relation to the development of hypocotyl and cotyledons. Treatment by far red light induces epinasty of the cotyledonary leaf. The effects of light treatments of different intensities were recorded for hypocotyl length, percentage of epinasty and the cotyledon leaf area. The percentage of epinasty increases as a function of treatment duration and light intensity. As from the second day of treatment a correlation is obtained between the percentage of epinasty and hypocotyl length when the seedlings are given far-red light treatment of different durations.  相似文献

Summary The appearance of the first vertical wall in the terminal hemispherical cell of the proembryo ofPotamogeton indicus is indicative of the origin of the cotyledonary and epicotylary loci. The cotyledonary locus grows at a faster rate while the epicotylary region conspicuously lags behind. Topographically the cotyledonary and epicotylary loci are adjacent to each other, both being derived from the terminal tier of the proembryo. The subjacent tiers, including them tier, do not take part in the organization of the shoot apex but only contribute to the formation of the hypocotyl and root regions.  相似文献

Seedlings of Brassica napus L. 2–11 days after germination were used. However, the most investigation was concentrated on the 6-day old seedlings. The primary root has a diarch protostele, the two groups of primary phloem alternate with the primary xylem. At higher level, the metaxylem is gradually differentiated in a lateral direction. Being coincident with this changes of the metaxylem, the groups of phloem cell are extended. The stele of the lower hypocotyl is root-like and has no pith. In the middle hypocotyl, there is a further lateral differentiation of the metaxylem. At the higher level, four metaxylem arms appear and the groups of phloem are extended circumferentially to form two crescent shaped sectors. In the upper hypocotyl below 0.2 cm of the cotyledonary node, a central pith has been formed which separates the differentiating primary xylem into two distinct units. At a slightly higher level, each primary phloem divides into two small groups, at this time, each xylem unit and the two adjacent groups of phloem constitute a cotyledonary trace. The foliar traces of the first two foliage leaves appear in the inter-cotyledonary plane between the vascular elements of the cotyledonary traces. At this level, the vascular tissue of the hypocotyl forms a siphonostele made up of two cotyledonary traces and the two foliage leaves, where the root-stem transition has nearly been completed, while the endarch condition is not attained in the hypocotyl. At incresing distances from the cotyledonary node upwards, in the cotyledonary petiole, the protoxylem occupies a more and more adaxial position and the metaxylem a more and more abaxial direction and, thus, the endarch condition is attained. The primary system of the root, hypocotyl, and cotyledons forms a complete circular system, the plumular vascular elements are directly connected by secondary elements formed by the cambium in the region of the hypocotyl. As for the results mentioned above, the authers have not detected that the primary xylem has a rotation of 180˚, as described by Van Tieghem.  相似文献

Summary The synthesis of DNA, RNA, protein and dry matter was followed in the elongating cells of hypocotyl of Lupinus (Lupinus angustifolius L.) germinating in total darkness or in continuous light. Light strongly inhibits the synthesis of DNA, RNA and protein. It was shown by histofluorometric DNA determinations that a reduced synthesis of DNA in continuous light is accompanied by a lower level of endomitosis in the cortex cells of the hypocotyl. In the dark cortex nuclei become 8 C while in the light they become only 4 C. The more pronounced differences between dark and light germinated hypocotyls of lupinus in comparison with pea epicotyls is explained to a great extent by mitotic growth.  相似文献

A quick procedure for the isolation of nuclei with good yield from soybean hypocotyl (Glycine max var. Wayne) was developed. The isolated nuclei appeared to retain their structural integrity. They were typically ellipsoidal with minima and maxima diameter of about 6 and 8 to 10 micrometers. While the nuclei were similar in size, the nucleoli were significantly larger in nuclei from auxin-treated tissue. The DNA content per nucleus was 4 ± 1 picograms for both untreated and auxin-treated tissues. The DNA: RNA: protein ratio of isolated nuclei in untreated and auxin-treated tissues was 1: 3.1: 11 and 1: 5.4: 21.7, respectively. The purified nuclei were active in RNA synthesis; the level of RNA polymerase II activity expressed in the nuclei from untreated tissue was 50 to 60% higher than RNA polymerase. I. The nuclei from auxin-treated tissues contained about 2.5 times as much RNA polymerase I activity as nuclei from untreated tissue. The purified nuclei from both untreated and auxin-treated tissues were also active in the incorporation of 3H-TTP into DNA.  相似文献

The changes in the endogenous growth as well as in the cell wall composition were studied along the hypocotyl of Pinus pinaster Aiton. Cell elongation decreased as the distance from the cotyledonary node increased. Pectic polysaccharides underwent an important depolymerization accompanied by a decrease in their uronic acid content from the apical to basal region of the hypocotyl. Additionally, the molecular mass of pectic polysaccharides strongly decreased from the apical to the basal regions. Watersoluble hemicellulosic polysaccharides extracted with 4% KOH decreased notably from the cotyledonary node towards the base, while water-soluble polysaccharides extracted with 24% KOH showed few differences along the hypocotyl. The molecular mass of xyloglucan present in both hemicellulosic fractions was lower in the upper hypocotyl region as compared with the basal region. These findings are in agreement with an active xyloglucan depolymerization in the upper region as would be expected in a region exhibiting very active growth.  相似文献

Procedures for callus induction, plantlet regeneration, andAgrobacterium-mediated transformation ofBrassica juncea were optimized by studying several factors, including explant types, and various plant growth regulators and adjuvants, such as silver nitrate, sucrose and agar. The highest shoot regeneration frequency was obtained from hypocotyl and cotyledonary petiole explants on MS medium containing 3 mg/L benzylaminopurine (BA) and 2 mg/L α-naphthaleneacetic acid (NAA). Transformation was affected by a number of factors, including explant type, selection agents, preculture duration, pre-selection conditions, and coculture temperature. Transformation efficiencies for hypocotyl and cotyledonary petiole explants were at 65% and 69%, respectively.  相似文献
