ABSTRACT

Introduction: Mass spectrometry imaging (MSI) techniques are nowadays widely used to obtain spatially resolved metabolite information from biological tissues. Since (phospho)lipids occur in all animal tissues and are very sensitively detectable, they are often in the focus of such studies. This particularly applies for phosphatidylcholines (PC) which are very sensitively detectable as positive ions due to the permanent positive charge of their choline headgroup.

Areas covered: After a short introduction of lipid species occurring in biological systems and approaches normally used to obtain spatially resolved mass spectra (with the focus on matrix-assisted laser desorption/ionization coupled to time-of-flight (MALDI-TOF) MSI) a survey will be given which diseases have so far been characterized by changes of the PC composition.

Expert commentary: Since PC species are very sensitively detectable by MS, sensitivity is not a major issue. However, spatial resolution is still limited and cellular dimensions can be hardly resolved by MALDI-TOF MSI, which is a critical point of the available approaches. BackgroundBrain Computer Interface (BCI) systems have been widely used to develop sustainable assistive technology for people suffering from neurological impairments. A major limitation of current BCI systems is that they are based on Subject-dependent (SD) concept. The SD based BCI system is time consuming and inconvenient for physical or mental disables people and also not suitable for limited computer resources. In order to overcome these problems, recently subject-independent (SI) based BCI concept has been introduced to identify mental states of motor disabled people but the expected outcome of the SI based BCI has not been achieved yet. Hence this paper intends to present an efficient scheme for SI based BCI system. The goal of this research is to develop a method for classifying mental states which can be used by any user. For attaining this target, this study employs a supervised spatial filtering method with four types of feature extraction methods including Katz Fractal Dimension, Sub band Energy, Log Variance and Root Mean Square (RMS) and finally the obtained features are used as input to Linear Discriminant Analysis (LDA) classification model for identifying mental states for SI BCI system.ResultsThe performance of the proposed design is evaluated in several ways such as considering different time window length; different frequency bands; different number of channels. The mean classification accuracy using Katz feature is 84.35% which is the maximum output compare to other features that outperforms the existing methods.ConclusionsOur proposed design will help to make a new technology for development of real-time SI based BCI systems that can be more supportive for the motor disabled patients.  相似文献

Background  

Aromatase, the cytochrome P-450 enzyme (CYP19) responsible for estrogen biosynthesis, is an important target for the treatment of estrogen-dependent breast cancer. In fact, the use of synthetic aromatase inhibitors (AI), which induce suppression of estrogen synthesis, has shown to be an effective alternative to the classical tamoxifen for the treatment of postmenopausal patients with ER-positive breast cancer. New AIs obtained, in our laboratory, by modification of the A and D-rings of the natural substrate of aromatase, compounds 3a and 4a, showed previously to efficiently suppress aromatase activity in placental microsomes. In the present study we have investigated the effects of these compounds on cell proliferation, cell cycle progression and induction of cell death using the estrogen-dependent human breast cancer cell line stably transfected with the aromatase gene, MCF-7 aro cells.  相似文献

Abstract

This study aimed to evaluate whether natural or synthetic steroid hormones could directly modulate the activity of the different superoxide dismutase (SOD) isoforms found in human blood fractions without changing enzyme expression. Enzyme samples of human erythrocytes, the human platelet-rich plasma fraction (PRP) or isolated CuZnSOD, which was purified from human erythrocytes were pre-incubated with natural steroids (17β-estradiol 17-acetate and progesterone) and their synthetic derivatives (β-estradiol 3-benzoate and medroxyprogesterone 17-acetate). Then, CuZn and MnSOD activities were measured using the xanthine/xanthine oxidase/nitroblue tetrazolium method. Hormones had no effect on MnSOD activity from the PRP, but we show for the first time that natural and synthetic steroid hormones have a direct, bell-shaped effect on the activity of CuZnSOD from both male and female human erythrocytes. Low (physiological) hormone concentrations caused a dose-dependent increase in enzyme activity, which disappeared at higher hormone concentrations. In addition, the combination of synthetic and natural estrogens and progestins had a synergistic stimulatory effect on the activity of CuZnSOD from human erythrocytes. The molecular interaction between CuZnSOD and steroid hormones was preliminarily studied. Natural hormones did not change the electrophoretic mobility of SOD under denaturing conditions, but they did increase the absorption spectra of SOD in the 230–290 nm range. These data suggest that hormone-mediated modulation of CuZnSOD is related to subtle changes in protein conformation, possibly related to Trp and Phe residues. We propose that this effect may account for the physiological regulation of enzyme activity during conditions where steroid hormones undergo alterations as the ovulatory cycle.  相似文献

植物组织空间排列对植食性哺乳动物功能反应的作用,是觅食生态学的热点问题之一。以新鲜紫花苜蓿叶片为食物,改变苜蓿叶片大小调控根田鼠口量,改变叶片间距调控叶片密度,设置叶片空间异质性斑块。在空间异质性斑块上测定根田鼠的觅食行为及其参数,检验植食性哺乳动物4种功能反应模型的预测性。除复合模型参数Rmax、h和Vmax及口量模型参数Rmax的最大似然估计值与测定值近似外,其它模型参数的估计值与测定值均存在较大差异。根田鼠摄入率测定值与4种模型预测值的线性回归均显著(P0.01),但与复合模型预测值的线性拟合效果最佳。表明,复合模型能很好地解释根田鼠觅食与行走的竞争对摄入率调节的动态。根田鼠复合模型存在调节其功能反应机制转变的距离临界值(d)。叶片间距大于该值时,叶片密度调节摄入率;叶片间距小于该值时,口量调节摄入率。结果充分地验证了提出的特定假设:在植物密集条件下,植物大小能调节植食性小型哺乳动物根田鼠的摄入率;在植物稀疏条件下,植物密度调节其摄入率。  相似文献

Abstract

In the present study, we report a reverse-phase high-performance liquid chromatography (RP-HPLC) method for separation of the regio-isomers of Glyceryl MonoRicinoleate (GMR) identified using position specificity of lipases. The approaches explored to identify these regio-isomers include LC-mass spectrometry, UV spectroscopy, and selective hydrolysis with lipases. A distinct UV absorption spectrum and λmax values for each isomer were noted, and mass spectral analysis further revealed their molecular weight. Lastly, the purified regio-isomers were subjected to hydrolysis with two distinctive regio-specific lipases to identified as sn-2 and sn-1(3) GMR. The current methodology of using analytic tool and enzyme specificity provides a useful platform for identifying regio-isomers for structured lipid synthesis.  相似文献

Clonal organisms persist at a range of population sex ratios, from equal numbers of males and females to single-sex systems. When intersexual competition is strong enough to drive one sex locally extinct, the maintenance of the sexes is facilitated by the semi-independent dynamics of populations within a metapopulation. These semi-independent dynamics are influenced by dispersal and recolonization rates, which are affected by the spatial arrangement of populations. To establish the quantitative relationship between spatially complex metapopulations and the maintenance of the sexes, we used a mathematical model of the liverwort Marchantia inflexa. This clonal organism is found in discrete patches on rocks and along the banks of streams, which form single-sex and two-sex metapopulations. In this system, asexual propagules mainly disperse short distances. Long-distance between-patch dispersal and recolonization mainly occurs via sexual propagules, which require both sexes to be present. Dispersal of these two types of propagules could interact with the spatial arrangement of populations to affect the maintenance of the sexes. With our mathematical model, we found that at intermediate distances between populations, metapopulations maintained both sexes, and the spatial arrangement of populations changed the threshold at which one sex was lost. On the other hand, when populations were close to one another, one sex was lost and the single-sex metapopulation persisted through dispersal of asexual propagules. When populations were far apart, one sex was lost, and the metapopulation either went extinct due to lack of recolonization by asexual propagules or persisted because clumped populations facilitated recolonization. These idealized spatial arrangements help clarify the effects of the spatial arrangement on the maintenance of the sexes and the persistence of metapopulations of clonal organisms, which can help explain geographic parthenogenesis and the distribution of asexual populations, the persistence of asexual species, and inform the conservation of clonal organisms.  相似文献

Abstract

Receptor tyrosine kinases (PTKs) play key roles in the pathogenesis of numerous human diseases, including cancer, and therefore PTK inhibitors are currently under intense investigation as potential drug candidates. PTK inhibitor screening data are, however, poorly comparable because of the different assay technologies used. Here we report a comparison of ELISA-based assays for screening epidermal growth factor receptor (EGFR) tyrosine kinase (TK) inhibitory compound libraries to study interassay variations. All assays were based on the same protocol, except for the source of EGFR-TK enzymes. In the first protocol, the enzyme was isolated from A431 cells without affinity purification. In the second protocol, commercial EGFR-TK (Sigma) isolated from A431 cells by affinity-purification was employed. In the third protocol, an enzyme preparation obtained from a recombinant (Baculovirus transfected Sf9 cells) expression system was used. All assays employed the synthetic peptide substrate poly-(Glu, Tyr)1:4 and an ELISA-based system to detect phosphorylated tyrosine residues by a monoclonal antibody. We observed significant differences in both the activity of the enzymes and in the EGFR-TK inhibitory effect of our reference compound PD153035. The differences were significant in case of A431 cell lysate compared to affinity purified EGFR-TKs derived from either A431cells or Baculovirus transfected Sf9 cells, whereas the latter two showed comparable results. Our data suggest that differences in terms of interassay variation are not related to the source of the enzyme but to its purity; changes in the mode of detection can markedly influence the reproducibility of results. In conclusion, normalization of the EGFR activity used for inhibitor screening and standardization of detection methods enable safe comparison of data.  相似文献

分析比较了山西芦芽山不同海拔处分布的亚高山草甸(样地A,海拔2756.3 m;样地B,海拔2542.3 m)和云杉林(样地C,海拔2656.8 m;样地D,海拔2387.2 m)土壤有机碳和全氮的小尺度空间异质性特征。结果表明:相同植被类型下海拔较高的样地有机碳含量较高(A:49.84 g/kg,B:38.33 g/kg,C:47.06 g/kg,D:40.67 g/kg),而较低海拔的样地土壤有机碳含量的异质性较高;除样地A以外的其他3个样地均表现为高度空间依赖性。亚高山草甸土壤全氮含量的异质性远远高于云杉纯林,四个样地中均表现出强的空间自相关性。亚高山草甸样地土壤有机碳和全氮含量均在较大尺度上空间自相关,云杉纯林样地则表现为较小尺度的空间自相关变异。  相似文献

The spatial distribution of soil invertebrates is aggregated with high-density patches alternating with low-density zones. A high degree of spatio-temporal organization generally exists with identified patches of specific species assemblages, in which species coexist according to assembly rules related to competitive mechanisms for spatial and trophic resources occur. However, these issues have seldom been addressed. The spatio-temporal structure of a native earthworm community in a natural savanna and a grass–legume pasture in the Colombian “Llanos” was studied during a 2-year-period. A spatially explicit sampling design (regular grid) was used to discern the distribution pattern of species assemblages in both systems. Earthworms were collected from small soil pits at three different sampling dates. Data collected from 1 m2 soil monoliths were also used in the present study. Data were analyzed with the partial triadic analysis (PTA) and correlograms, while niche overlap was computed with the Pianka index. The PTA and correlogram analysis revealed that earthworm communities displayed a similar stable spatial structure in both systems during the 2-year study period. An alternation of population patches where different species' assemblages dominated was common to all sampling dates. The medium-sized Andiodrilus sp. and Glossodrilus sp. exhibited a clear spatial opposition in natural savanna and the grass–legume pasture for the duration of the study. The Pianka index showed a high degree of niche overlapping in several dimensions (vertical distribution, seasonality of population density) between both species. The inclusion of space-time data analysis tools as the PTA and the use of classical ecological indices (Pianka) in soil ecology studies may improve our knowledge of earthworm assemblages' dynamics.  相似文献

In this study we evaluated the effect of quercetin on D-galactose-induced aged mice using the Morris water maze (MWM) test. Based on the free radical theory of aging, experiments were performed to study the possible biochemical mechanisms of glutathione (GSH) level and hydroxyl radical (OH) in the hippocampus and cerebral cortex and the brain tissue enzyme activity of the mice. The results indicated that quercetin can enhance the exploratory behavior, spatial learning and memory of the mice. The effects relate with enhancing the brain functions and inhibiting oxidative stress by quercetin, and relate with increasing the GSH level and decreasing the OH content. These findings suggest that quercetin can work as a possible natural anti-aging pharmaceutical product.  相似文献

Theory predicts that hosts and pathogens will evolve higher resistance and aggressiveness in systems where populations are spatially connected than in situations in which populations are isolated and dispersal is more local. In a large cross‐inoculation experiment we surveyed patterns of host resistance and pathogen infectivity in anther‐smut diseased Viscaria alpina populations from three contrasting areas where populations range from continuous, through patchy but spatially connected to highly isolated demes. In agreement with theory, isolated populations of V. alpina were more susceptible on average than either patchily distributed or continuous populations. While increased dispersal in connected systems increases disease spread, it may also increase host gene flow and the potential for greater host resistance to evolve. In the Viscaria–Microbotryum system, pathogen infectivity mirrored patterns of host resistance with strains from the isolated populations being the least infective and strains from the more resistant continuous populations being the most infective on average, suggesting that high resistance selects for high infectivity. To our knowledge this study is the first to characterize the impacts of varying spatial connectivity on patterns of host resistance and pathogen infectivity in a natural system.  相似文献

BackgroundLongstanding, successful use of combinations of phytopharmaceuticals in traditional Chinese medicine (TCM) has caught the attention of several pharmacologists to natural medicines. However, the development and popularisation of TCM is mainly limited because of the unavailability of reports clarifying the mechanisms of action and pharmacologically active ingredients in such formulations. Previous studies on natural medicines have mostly focused on their dominant components using forward pharmacology which often neglects trace components. It is necessary to assess the pharmacological and therapeutic superiority of many such trace components in comparison with single constituents.PurposeIn this study, we aimed to propose a new pharmacological research strategy for TCM. In particular, we presented the possibility that the effective mechanism of action of trace components of TCM is based on synthetic lethality. We sincerely hope to explore this theory further.MethodWe obtained retrieve published research information related to synthetic lethality, phytochemicals and Chinese medicine from PubMed and Google scholar. Based on the inclusion criteria, 71 studies were selected and discussed in this review.ResultsAs an interaction among genes, synthetic lethality can amplify co-regulatory biological effects exponentially. Synthetic strategies have been successfully applied for research and development of anti-tumour agents, including poly ADP-ribose polymerase inhibitors and clinical combination of chemotherapeutic agents for efficacy enhancement and toxicity reduction. TCM drugs contain several secondary metabolites to combat environmental stresses, providing a multi-component basis for corresponding synergistic targets. Therefore, we aimed to study whether this method could be used to identify active components present in trace amounts in TCM drugs. Based on a reverse concept of target–component–effect and identified synergistic targets, we explored the mechanisms of action of weakly active components present in trace amounts in TCM drugs to assess combinations of potential synergistic components.ConclusionThis pattern of synthetic lethality not only elucidated the mechanisms of action of TCM drugs from a new perspective but also inspired future studies on discovering naturally occurring active components.  相似文献

Purpose

Stakeholders from across supply chains have been prompted to explore ways to reduce the environmental burdens of corn production. To effectively manage these environmental impacts, spatially explicit information accounting for the differences in growing conditions and production practices across the production landscape is essential, allowing for high impact intensity corn to be identified and prioritized for improvement. To support these sustainability efforts, this study examines the spatially explicit life cycle greenhouse gas emissions of US county corn production, providing the most comprehensive assessment to date.

Methods

A streamlined spatial life cycle assessment is conducted, focusing on the three key hotspots of corn production for spatial differentiation at the county scale across the contiguous USA, accounting for almost 60% of total average cradle-to-farm gate impacts. Variations in nitrogen fertilization types and rates, N2O emission rates, and irrigation emission rates are specifically revealed. Spatially distinguished hotspot inputs and related emissions are combined with static national average emission estimates from all other inputs used in corn production to gain a full picture and understand the relative contributions to total cradle-to gate impacts.

Results and discussion

Results show significant variation across corn producing counties, states, and regions. High impact priority locations are highlighted and key contributors of impact for each location are illuminated, providing critical information on the spatially explicit levers to reduce impacts. Results increase the generalizability of emission estimates using expected yields to characterize emission intensity, enabling more practical integration into company supply chain sustainability assessments to align with the time horizons in which decisions are made.

Conclusions

Streamlined life cycle assessment methods are an effective way to characterize spatial heterogeneity around key contributors of impact, helping deliver the necessary information for companies, stakeholders, and policy makers to target their influence to reduce these emissions through various engagement efforts.

The enzyme dipeptidyl-peptidase IV (DPP-IV) is recognized to be a promising target for the management of type 2 diabetes. Over the last decade, numerous synthetic molecules and more recently, peptides from dietary proteins, have been reported to be able to inhibit DPP-IV activity. Most studies that have investigated the in vitro effect of these inhibitors have used porcine or human DPP-IV. Although structurally alike, it is unclear whether these two species display similar inhibition patterns. Therefore, the objective of this study was to compare the effects of protein-derived peptides on the activity of porcine and recombinant human DPP-IV. The two species showed different inhibition susceptibility to 43 of the 62 peptide sequences investigated. While 37 protein-derived peptides were more effective at inhibiting the porcine DPP-IV, only six caused a stronger inhibition of the activity of the human enzyme. Although the peptides WR, IPIQY and WCKDDQNPHS were found to be among the most potent inhibitors of both species, the inhibitory effect was greater on the porcine enzyme than on human DPP-IV (αKi or Ki = 11.5, 13.4, 13.3 μM and 31.4, 28.2, 75.0 μM for porcine and human DPP-IV, respectively). Investigation into the mode of action of the most effective inhibitory peptides revealed that both species were inhibited in a similar manner by short fragments (≤5 amino acid residues), but that some of the longer peptides acted differently on the enzymes. This study shows that porcine DPP-IV is generally inhibited with greater potency by protein-derived peptides than is the human enzyme.  相似文献

ABSTRACT

Horseradish peroxidase (HRP) isoenzyme C1a is one of the most widely used enzymes for various analytical methods in bioscience research and medical fields. In these fields, real-time monitoring of HRP activity is highly desirable because the utility of HRP as a reporter enzyme would be expanded. In this study, we developed a simple assay system enabling real-time monitoring of HRP activity by using biolayer interferometry (BLI). The HRP activity was quantitatively detected on a BLI sensor chip by tracing a binding response of tyramide, a substrate of HRP, onto an immobilized protein. This system could be applied to analyses related to oxidase activity, as well as to the functional analysis of recombinant HRP.  相似文献

The South-European nemoral region. - The objective of this paper is to present a checklist and a biogeographical arrangement of the main vegetation types on the whole submediterranean zone (and supramediterranean belt in the mountains) of the European southern nemoral region. This revision has been made possible by the recent publication of a number of synthetic vegetation maps. Background

The detailed characterization of arabinoxylan-active enzymes, such as double-substituted xylan arabinofuranosidase activity, is still a challenging topic. Ad hoc chromogenic substrates are useful tools and can reveal subtle differences in enzymatic behavior. In this study, enzyme selectivity on natural substrates has been compared with enzyme selectivity towards aryl-glycosides. This has proven to be a suitable approach to understand how artificial substrates can be used to characterize arabinoxylan-active α-l-arabinofuranosidases (Abfs).

Methods

Real-time NMR using a range of artificial chromogenic, synthetic pseudo-natural and natural substrates was employed to determine the hydrolytic abilities and specificity of different Abfs.

Results

The way in which synthetic di-arabinofuranosylated substrates are hydrolyzed by Abfs mirrors the behavior of enzymes on natural arabinoxylo-oligosaccharide (AXOS). Family GH43 Abfs that are strictly specific for mono-substituted d-xylosyl moieties (AXH-m) do not hydrolyze synthetic di-arabinofuranosylated substrates, while those specific for di-substituted moieties (AXH-d) remove a single l-arabinofuranosyl (l-Araf) group. GH51 Abfs, which are supposedly AXH-m enzymes, can release l-Araf from disubstituted d-xylosyl moieties, when these are non-reducing terminal groups.

Conclusions and general significance

The present study reveals that although the activity of Abfs on artificial substrates can be quite different from that displayed on natural substrates, enzyme specificity is well conserved. This implies that carefully chosen artificial substrates bearing di-arabinofuranosyl d-xylosyl moieties are convenient tools to probe selectivity in new Abfs. Moreover, this study has further clarified the relative promiscuity of GH51 Abfs, which can apparently hydrolyze terminal disubstitutions in AXOS, albeit less efficiently than mono-substituted motifs.  相似文献
