卵孢小奥德蘑是一种珍贵的食药用真菌。本研究选用果渣、酒糟与菌糠为部分替代料,采用正交试验优化了野生卵孢小奥德蘑的母种、原种及栽培料配方,并测定了栽培子实体的含水量、蛋白质、总糖、维生素C、多酚含量及醇提物对DPPH、ABTs+及羟基自由基的清除能力。结果表明,最佳母种培养基配方为麦麸35 g/L、葡萄糖20 g/L、磷酸氢二钾3.5 g/L、硫酸镁2 g/L和琼脂20 g/L,确定了葡萄糖与磷酸氢二钾是影响菌丝生长的重要因素;最优原种培养基配方为果渣45%、豆秸20%、麦麸15%、石灰3%、石膏1%和刺芹侧耳菌糠16%,在培养基中添加果渣能够显著促进卵孢小奥德蘑原种菌丝的生长;最佳栽培料配方为酒糟35%、棉籽壳30%、麦麸20%、石灰1%、石膏1%和玉米芯13%,酒糟对子实体产量的影响达到了显著水平;卵孢小奥德蘑的营养成分显示其富含蛋白质、糖类、维生素C及多酚,由9种配方栽培出的子实体中最高含量可分别达到42.78 g/100 g、23.54 g/100 g、4.02 mg/100 g及4.19 mg/g,且栽培料的不同组分及用量对其营养物质的含量具有显著差异。此外,卵孢小奥德蘑的醇提物具有较强的抗氧化能力且随着用量的增加抗氧化活性增强,当用量为150、40和250 μL时配方Z1对DPPH和ABTs+及配方Z2对羟基自由基的清除能力可分别达到88.64%、99.81%和93.48%,本研究结果为卵孢小奥德蘑的栽培、生理活性、药理研究及进一步的开发利用提供理论依据。  相似文献

为明确昆诺阿藜链格孢叶斑病病原,在山西省昆诺阿藜种植区采集典型症状的标本分离病原菌,选择代表性菌株LGB-b和LGB-h对其形态学、分子生物学、致病性及生物学特性进行了研究。综合形态学和多基因系统发育(Alt a 1endoPG 和OPA10-2)分析,确定昆诺阿藜链格孢叶斑病病原为Alternaria alternata。致病性测定发现接种6 d后病斑呈灰绿至黄绿色,表面具有灰棕至灰褐色霉层,周围具黄绿色晕圈,与田间症状基本一致。菌株LGB-b和LGB-h均可侵染昆诺阿藜、藜和台湾藜。菌株LGB-b菌丝生长的最适培养基为V8 蔬菜汁琼脂培养基(V8)、温度为25-30 ℃、水活度≥0.98、pH为6-7;分生孢子萌发的最适水活度≥0.98、pH为6-7。菌株LGB-h菌丝生长的最适培养基为马铃薯胡萝卜琼脂培养基(PCA)、温度为20-25 ℃、水活度≥0.98、pH为6-7;分生孢子萌发的最适水活度≥0.98、pH为7-8。  相似文献

高雄山虫草Cordyceps tenuipes是一种重要的珍稀野生虫草,无性型为细脚棒束孢Isaria tenuipes。对采集的野生无性型高雄山虫草生物学特性进行了研究,采用小麦和大米为栽培基质,通过添加不同营养成分进行人工驯化和栽培条件优化,对后续提高其孢梗束产量和商业化栽培具有重要意义。试验结果表明,该虫草菌丝在固体和液体培养条件下生长所需的最佳营养组成一致,通过单因素和多因素正交试验得出菌丝在固体和液体条件下生长的最佳培养基配方为蔗糖30g/L、酵母粉15g/L、磷酸二氢钾2g/L、七水硫酸镁2g/L,最佳培养条件为温度25℃,pH 7。以小麦为培养基栽培的孢梗束生物学效率介于(45.750±6.062)%-(67.820±6.018)%,总体优于大米培养基(23.410±5.242)%-(36.880±8.812)%。以添加蚕蛹粉的小麦培养基栽培的孢梗束生物转化率最高,达67.820%,高于已有的报道。通过生物学特性分析和人工驯化条件的优化,为高雄山虫草的规模化人工栽培提供参考。  相似文献

通过对尖顶羊肚菌液体培养基质与条件的研究,明确其菌丝生长的最适pH值、最适温度、适宜光照条件、适宜葡萄糖和蛋白胨浓度、适宜培养基,以便应用于尖顶羊肚菌液体菌种的生产和工业发酵。结果表明:菌丝的最适生长温度为2 5℃;最适生长pH值为6 ;葡萄糖和蛋白胨最适浓度分别为2 0 0g/L和10g/L ;菌丝在黑暗环境下生长良好,光照对菌丝生长具有抑制作用;用胡萝卜酵母膏培养基振荡培养形成的菌丝球多,菌丝生长量大;菌丝球在不同培养基中生长,可引起培养液pH值的上升或者下降;菌丝球可利用培养基内的氨基酸,使氨基酸降解,在胡萝卜酵母膏培养基中振荡培养8d的菌液总氨基酸含量较原液减少了36 71% ,亮氨酸、异亮氨酸和甲硫氨酸含量的下降幅度最大  相似文献

桔梗匍柄霉叶斑病是近年来发现的新病害,病原菌为桔梗匍柄霉Stemphylium platycodontis。为了更好地了解病害的发生规律,本研究对该病原菌的生物学特性及药剂敏感性进行了分析。结果表明:25℃为该病原菌的最适生长温度和产孢温度;菌丝生长最适pH值为4,而最适合产孢的pH值为8;24h黑暗处理,对病原菌的菌丝生长和产孢都最有利;PCA培养基对菌丝生长最有利,而桔梗汁液培养基对病原菌产孢最有利;可溶性淀粉对菌丝生长最有利,而蔗糖最利于病原菌产孢;乙酰铵对病原菌生长及产孢都最有利;26.8μg/mL氟硅唑对桔梗匍柄霉的抑制效果最好,毒力持效性最强。  相似文献

对采自祁连山地区的一株野生食用菌进行分离,通过形态学及其ITS序列的分子系统发育分析鉴定为絮缘蘑菇Agaricus subfloccosus。以分离纯化获得的菌株作为实验材料,对该种的生物学特性和驯化栽培进行研究。检测了不同碳源、氮源、温度、pH、无机盐、维生素在固体培养条件下对絮缘蘑菇菌丝生长的影响,对以上6个因素进行单因素试验,从中挑选出3个较优因素的最优水平进行正交试验。结果表明:实验范围内,絮缘蘑菇菌丝生长最佳碳源为蔗糖,最佳氮源为硝酸钾,最佳无机盐为硫酸镁,最佳维生素为VB12,最佳温度为16℃,最佳pH为6。影响较大的3个因素为碳源、无机盐和pH,絮缘蘑菇最适的营养因子最佳组合配方为:蔗糖25g/L,硝酸钾2g/L,硫酸镁2g/L,pH为5.0。驯化栽培过程中,栽培配方为:木屑79%、高粱籽5%、玉米面5%、石膏1%、麸皮10%。温度16℃使絮缘蘑菇菌丝在90d左右满袋,覆土可培育出子实体。  相似文献

Growth conditions, including incubation times, temperature, agitation rate and initial pH of medium, that affect xylanase production by Aspergillus carneus M34 were studied sequentially use the classical “change-one-factor-at-a-time” method. Our results showed that there was a similar trend between cellular xylanase activity and extracellular xylanase activity. The optimal conditions for xylanase production, different from their cell growth, were on the third day, 30 °C, 100 rpm and pH 4, respectively, in this test. Response surface methodology (RSM) was further introduced to optimize the cultivation conditions and to evaluate the significance of these factors. The optimal cultivation conditions predicted from canonical analysis of this model were achieved by incubation at 35.08 °C with an agitation rate of 111.9 rpm and an initial pH of 5.16. In addition, temperature was the most critical factor for xylanase production by A. carneus M34. Xylanase activity of 22.2 U/mL was verified using the predicted optimal conditions and confirmed the fitness and applicability of the model. The optimal temperature and pH of the crude xylanase activity was observed at 60 °C and acidic pH, respectively. Sustained xylanase activity in the crude extract was also detected over a broad range of pH from 3 to 10. Considering its higher specificity toward agricultural wastes, especially corn cob and coba husk, this strain can be used to develop low-cost media for the mass-production of xylanase.  相似文献

The hatchability of eggs and the fecundity and survival of adult Bulinus (Physopsis) africanus was investigated in different salinities. Experimental results revealed egg masses and hatchlings to be considerably more sensitive to salinity than the adult snails. Egg-laying was recorded in salinities 4·5 ‰ and further increases in salinity resulted in a progressive reduction in the hatching success up to a lethal concentration of 5·25 ‰ Survival of these hatchings was adversely affected by salinities as low as 1·0 ‰ and a salinity of 4·5 ‰ was lethal within 6 days. In contrast, adult survival was unaffected in salinities < 3·5 ‰ while further increases in salinity resulted in significant reductions in survival up to a lethal salinity of 8·7 ‰, which caused 100% mortality within 24 h. The survival of B. africanus infected with Schistosoma haematobium and Schistosoma mattheei was lower in the different salinities and control than that of their uninfected counterparts.  相似文献

对我国皱环球盖菇Stropharia rugosoannulata 20个主栽菌株进行了ISSR标记遗传多样性分析,使用的28个ISSR引物中22个具有多态性,UPGMA聚类分析显示遗传相似性水平在0.68-0.86之间,在0.72时可将菌株分为6个类群,类群间遗传差异较大。在PDA培养基上,除菌株Sr-03和Sr-05最适生长pH值为8.0外,其他菌株均为pH 5.0-6.0;在5-30℃的温度范围内,除菌株Sr-01、Sr-08、Sr-11在30℃时长速受到抑制外,其他菌株长速随温度升高而加快,但在35℃时仅有5个菌株在培养20d时具有活性;皱环球盖菇菌丝在以木屑为主的原种培养料中长速较慢,为0.73-1.08mm/d。在菌株的农艺性状比较中,菌株Sr-12的菇型比例最好、产量最高,生物学效率达98.09%,菌株Sr-08和Sr-12菇体硬度较大,菌盖颜色与其他菌株差异显著,菌株Sr-19和Sr-20的菌褶颜色较其他菌株差异较大。本研究筛选出了7个具有遗传差异的优异种质,产量高、抗逆性强,并在菇型、菌盖颜色、菌褶颜色等农艺性状上具有较大的差异,可为皱环球盖菇育种及遗传研究提供菌株选择。  相似文献

Models of the cultivation process of Actinobacillus sp. cells in two media, rich (NB) and minimal (M9) that includes phenol as a sole carbon source, have not been described in the available literature. For these reasons, several single-substrate inhibition models (Monod, Andrew, and Tesseir) were investigated in order to determine the mathematical expression of Actinobacillus sp. growth rate. The experimental data for both nutrient broth and M9 media were fitted to the above models mentioning that Andrews' model best fits these data adequately for both media with regression coefficient of 0.973 and 0.962, respectively. The maximum predicted growth rate by this model is 0.37 h- 1 for both media obtained when the initial concentration of phenol is 100 mg/L. The half-saturation concentration constant, KP, is 1.00 mg/L, which represents the phenol concentration when μ is equal to half μmax. On the other hand, the inhibition constant, Kp is 13,000.00 mg/L for broth medium and 12,000 mg/L for M9 medium, which is a measure of sensitivity to inhibition by inhibitory substances. When cells are grown in nutrient broth and minimal media, the rate of cell production with time can be expressed by the Reccati and Voltera models. Voltera model better fits in the case of M9 minimal medium plus phenol as sole carbon source. The pH of 7, the incubation temperature of 35°C to 37°C, and the agitation rate of 150 rpm are the optimal conditions for achieving the higher percentage of phenol degradation by Actinobacillus sp. Succinic acid and glycine as carbon and nitrogen source, respectively, were the most efficient of the cosubstrates (out of 10 substrates tested) for removal of phenol on an mg/L basis.  相似文献

A new technique for the selective isolation of actinomycetes from natural mixed microbial populations is described. A nutrient agar medium was overlaid with a 0.22- to 0.45-microns-pore cellulose ester membrane filter, and the surface of the filter was inoculated. During incubation, the branched mycelia of the actinomycetes penetrated the filter pores to the underlying agar medium, whereas growth of nonactinomycete bacteria was restricted to the filter surface. The membrane filter was removed, and the agar medium was reincubated to allow the development of the isolated actinomycete colonies. This procedure selects actinomycetes on the basis of their characteristic mycelial mode of growth, offers a general method for their selective isolation, and does not rely on the use of special nutrient media or of antibacterial antibiotics.  相似文献

【背景】大球盖菇和斑玉蕈是食药兼用且具有开发潜力的珍稀食用菌,培养基对菌丝生长及子实体发育具有重要作用,优化培养基显得尤为重要。【目的】筛选出最适合培养大球盖菇、斑玉蕈的新型培养基。【方法】使用添加海藻糖的新型培养基,对不同培养基培养的大球盖菇及斑玉蕈菌株的菌丝生长状况、生长速度和生物量及纤维素酶、漆酶活性进行测定与分析。【结果】相较于PDA培养基,添加海藻糖的培养基能够提高菌丝生长速度、增加生物量,海藻糖添加的比例对纤维素酶和漆酶的影响较大,对大球盖菇及斑玉蕈菌丝的生长产生显著的促进作用,但不会改变其蛋白质的组成。【结论】大球盖菇最适合选用PTA-5培养基,斑玉蕈的最佳培养基是PDTA培养基。  相似文献

In order to evaluate the potential use of Phoma exigua isolate PFC 2705 (PFC2705) as a biological control agent for salal (Gaultheria shallon), effect of cultural and environmental parameters on growth, conidia production, and pathogenicity of P. exigua were characterized in studies conducted under laboratory and greenhouse conditions. Within a range of 5-30°C, the optimum growth and germination temperature range was 20-25°C. The effect of pH on mycelial growth and conidial germination was not significant from pH 5 to 10. Fluorescent light significantly enhanced sporulation of the fungus on most agar media tested, yet was not necessary for growth. The type of culture media significantly affected mycelium growth, sporulation, and conidia germination. Age of mycelia used as inoculum affected the disease severity on salal. PFC2705 suppressed the growth of mature salal plant by inciting lesions on leaves, branch tips, and axillary buds and caused 56% death of the total biomass above ground. Characteristics such as easy inoculum production, wide range of growth environments, and high infectivity on salal increased the potential of P. exigua as a biocontrol agent for management of salal.  相似文献

The shore crab, Carcinus maenas L. (Portunidae), is a coastal and estuarine species, which can live and reproduce under brackish water conditions; freshly hatched larvae have been observed in the field at salinities below 15‰. In the present laboratory study, the tolerance of hypo-osmotic stress was experimentally investigated in early larvae of a marine (North Sea) population of C. maenas reared at four different salinities (15, 20, 25, 32‰). Two and 4 days after hatching, the Zoea I larvae were moult-staged microscopically, and their rates of respiration and growth (changes in dry weight, W, carbon, C, nitrogen, N, and hydrogen, H) were measured. Survival and development were monitored until the megalopa was reached: 15‰ did not allow for development beyond the first zoeal stage, while metamorphosis to the megalopa was reached at salinities ≥20‰. At 20‰, development was significantly delayed and mortality enhanced as compared with 25 and 32‰. Rates of growth and respiration decreased during exposure to reduced salinities ≤25‰. Hence, the suppression of growth could not be explained as a consequence of enhanced metabolic losses per larva. Instead, a partial C budget indicates that the Zoea I larvae suffered from decreased capabilities of assimilating ingested and subsequently converting assimilated matter to tissue growth. Net growth efficiency (K2, C-based) was at 25 and 32‰ initially high (>60% during the postmoult and intermoult stages of the Zoea I moult cycle), but decreased during the later stages (down to ≤30% in premoult). An inverse pattern of C partitioning was observed at ≤20‰, with initially low K2 values (≤21% during the first 2 days of the moult cycle), and a later increase (up to ≥46% in premoult). Thus, larval growth was initially suppressed under conditions of reduced salinity, but this was later (during premoult) partially compensated for by an increase in C assimilation and K2. Our observations indicate that Zoea I shore crab larvae react during the late stages of their moulting cycle less sensitively against reduced salinities than during postmoult and intermoult. This suggests that the transition between moult cycle stages C and D0 may be a critical point for effects of hypo-osmotic stress, similarly as already known in relation to effects of nutritional stress. Negative effects were found also when freshly hatched Zoea I shore crab larvae were exposed only transitorily (for 24–72 h) to 20‰, with significantly lower rates of survival, development, growth, respiration, and K2. 为获得冬虫夏草菌固体发酵产分生孢子的最优工艺,以野生分离的冬虫夏草菌为材料,对其固体发酵产分生孢子的培养基及培养条件进行了研究。试验结果表明:泥炭土为最佳基础培养基,该培养基中冬虫夏草菌气生菌丝生长一般,但产分生孢子最多,可达4.2×103个/g;泥炭土培养基中添加0.1‰ IAA(吲哚乙酸)、0.1‰ IBA(吲哚丁酸)和0.1‰ NAA(萘乙酸)能促进冬虫夏草菌气生菌丝的生长和分生孢子的产生,其分生孢子达8.1×103个/g;该基础培养基中,冬虫夏草菌于18℃培养30d后,在10℃、相对湿度45%、蓝光照射进行诱导,分生孢子可达1.0×104个/g。本研究建立了一种大量获取冬虫夏草菌分生孢子的方法,为冬虫夏草繁育奠定了基础。  相似文献

Humoral and cellular responses to homologous parasite extracts were studied in C57BL mice infected with Nematospiroides dubius or Nippostrongylus brasiliensis, to determine whether these parasites induced specific immunosuppression which might facilitate their survival. IgG and IgM titres to adult, excretory-secretory, larval and egg antigens from N. dubius and adult antigen from N. brasiliensis increased progressively for several weeks, irrespective of parasite rejection. Delayed-type hypersensitivity responses to the same antigens peaked after about 2 weeks and then remained constant in N. dubius -infected mice, but declined after the rejection of N. brasiliensis. The specific lymphoproliferative responses of spleen cells to these extracts reached peak values 1–3 weeks after infection and were anti-Thy 1.2-sensitive. They then declined sharply, but remained above the levels seen in uninfected mice. These results should be considered in the interpretation of any investigations into the stable host-parasite relationship which exists during a primary N. dubius infection.  相似文献
