Growth of two actinorhizal species was studied in relation tothe form of N supply in water culture. Non-nodulated bog myrtle(Myrica gale) and grey alder (Alnus incana) were grown withNH4+, NH4NO3 or NO3 (4 mol m–3 N). A nodulatedseries of bog myrtle was also cultivated in N-free medium. Relative growth rate (RGR), utilization rate of N, and shoot/rootratio were highest for the two species with the N completelysupplied as NH4+. In both species, nitrate was largely reducedin the roots and the presence of NO3 in combined-N supplyalways affected the RGR and N utilization rate negatively. BothN2 fixation and complete NO3 nutrition represented conditionsof relative N-deficiency resulting in relatively low tissue-Nconcentrations and a greater allocation of dry mass to the roots.The physiological N status of nodulated M. gale plants was comparativelygood, as indicated by a normal nodule weight ratio and a relativelyhigh N2-fixing rate per unit nodule mass. However, whole-plantN2-fixing capacity remained relatively low in comparison withacquisition rates of N in combined-N plants. The anion charge from the nitrate reduction was largely directlyexcreted as an OH efflux. H + /N ratios generally agreedwith the theory. In comparison with NH4+ nutrition, carboxylateconcentrations were higher in N2-fixing M. gale plants and theH + /N ratio in nodulated plants was less than unity below thevalue for ammonium plants as previously found for other actinorhizalspecies. Therefore, NH4+ should be an energetically more attractiveN source for actinorhizal plants than N2. The results agree with commonly accepted views on energeticsof N uptake and assimilation in higher plants and support theconcept of a basically similar physiological behaviour betweennon-legumes and legumes. Photoautotrophic cell suspension cultures of Chenopodium rubrumrequire high concentrations of nitrate and ammonium. Duringthe growth phase total NH4+ and the greater portion of NH3were consumed. During the stationary phase nitrate uptake continuedbut at a substantially smaller rate than during the growth phase.During growth the bulk of the absorbed N was incorporated intoprotein, the amount of which was then maintained constant untilsenescence. NH3 was accumulated upon transition betweenthe growth and the stationary phase. NH3, like the freeamino acids, was deposited in the vacuole but, unlike thesecompounds, could not be remobilized upon transfer of the cellsinto N-free medium. Readdition of NH4+ to the medium, however,resulted in a mobilization of the vacuolar NH3-pool.Reutilization of both vacuolar N-storage pools must have beenaccomplished by recycling from the vacuole to the cytoplasmbecause N-metabolizing enzymes could not be detected in isolatedvacuoles. Transfer of the cells of the stationary phase intomedium containing NH3 and NH4+ resulted in an inductionof nitrate uptake by the cells, but only after a lag phase of4–5 days. The growth of four heathland species, two grasses (D. flexuosa,M. caerulea) and two dwarf shrubs (C. vulgaris, E. tetralix),was tested in solution culture at pH 4.0 with 2 mol m–3N, varying the N03/NH4+ ratio up to 40% nitrate. In addition,measurements of NRA, plant chemical composition, and biomassallocation were carried out on a complete N03/NH4+ replacementseries up to 100% nitrate. With the exception of M. caerulea, the partial replacement ofNH4+ by NO3 tended to enhance the plant's growth ratewhen compared to NH4+ only. In contrast to the other species,D. flexuosa showed a very flexible response in biomass allocation:a gradual increase in the root weight ratio (RWR) with NO3increasing from 0 to 100%. In the presence of NH4+, grassesreduced nitrate in the shoot only; roots did not become involvedin the reduction of nitrate until zero ambient NH4+. The dwarfshrubs, being species that assimilate N exclusively in theirroots, displayed an enhanced root NRA in the presence of nitrate;in contrast to the steady increase with increasing NO3in Calluna roots, enzyme activity in Erica roots followed arather irregular pattern. Free nitrate accumulated in the tissuesof grasses only, and particularly in D. flexuosa. The relative uptake ratio for NO3 [(proportion of nitratein N uptake)/(proportion of nitrate in N supply)] was lowestin M. caerulea and highest in D. flexuosa. Whereas M. caeruleaand the dwarf shrubs always absorbed ammonium highly preferentially(relative uptake ratio for NO3 <0.20), D. flexuosashowed a strong preference for NO3 at low external nitrate(the relative uptake ratio for N03 reaching a value of2.0 at 10% NO3). The ecological significance of thisprominent high preference for NO3 at low NO3/NH4+ratio by D. flexuosa and its consequences for soil acidificationare briefly discussed. Effects of chilling (5 °C) period, light and applied nitrogen(N) on germination (%), rate of germination (d to 50% of totalgermination; T50%) and seed imbibition were examined inClematisvitalba L. In the absence of chilling, light and N, germinationwas minimal (3%). When applied alone, both chilling and N increasedgermination. Chilling for 12 weeks increased germination to64%, and 2.5 mM NO-3or NH+4increased germination to 10–12%.Light did not increase germination when applied alone, but didwhen applied in combination with chilling and/or N. Half theseed germinated when light was combined with 2.5 mM NO-3or NH+4.The influence of chilling, light and/or N on germination wasgreater when combined, than when either factor was applied alone.Both oxidized (NO-3) and reduced (NH+4) forms of N increasedgermination, but non-N-containing compounds did not, suggestingthe response was due to N and not ionic or osmotic effects. Without additional N, T50%decreased from 16–20 d at zerochilling, to around 5 d at 8 and 12 weeks chilling. AlthoughT50%was not influenced by an increase in NO-3or NH+4from 0.5to 5.0 mM , it did increase with additional applied N thereafter.However, the magnitude of the N effect was small compared tothat of chilling. Like germination, seed imbibition increasedwith a longer chilling period, but in contrast imbibition decreasedslightly with increased applied NO-3or NH+4. It is argued thatincreased imbibition is not directly related to an increasein total germination, but that it may be related to the rateof germination. Possible mechanisms involved in the reductionin dormancy ofC. vitalba seed are discussed. Ammonium influx into roots and N translocation to the shootswere measured in 3-week-old hydroponically grown rice seedlings(Oryza sativa L., cv. IR72) under conditions of N deprivationand NH4+ resupply, using 13NH4+as a tracer. Root NH4+ influxwas repressed in plants continuously supplied with NH4+ (at0.1 mM), but a high proportion of absorbed N (20 to 30%) wastranslocated to the shoot in the form of N assimilates duringthe 13-min loading and desorption periods. Interruption of exogenousNH4+ supply for periods of 1 to 3 d caused NH4+ influx to bede-repressed. This same treatment caused N translocation tothe shoot to decline rapidly, until, by 24 h, less than 5% ofthe absorbed 13N was translocated to the shoot, illustratinga clear priority of root over shoot N demand under conditionsof N deprivation. Upon resupplying 1 mM NH4+, root NH4+ influxresponded in a distinct four-phase pattern, exhibiting periodsin which NH4+ influx was first enhanced and subsequently reduced.Notably, a 25 to 40% increase in root influx, peaking at {smalltilde}2 h following re-exposure was correlated with a 4- to5-fold enhancement in shoot translocation and a repression ofroot GS activity. The transient increase of NH4+ influx wasalso observed in seedlings continuously supplied with NO3and subsequently transferred to NH4+. Extended exposure to NH4+caused root NH4+ influx to decrease progressively, while shoottranslocation was restored to {small tilde}30% of incoming NH4+.The nature of the feedback control of NH4+ influx as well asthe question of its inducibility are discussed. Potassium-Ammonium Uptake Interactions in Tobacco Seedlings   总被引:6,自引:0,他引:6  
Short-term (< 12 h) uptake experiments were conducted with6–7-week-old tobacco (Nicotiana tabacum L. cv. Ky 14)seedlings to determine absorption interactions between K+ andNH4+. At equal solution concentrations (0.5 mol m–3) netK+ uptake was inhibited 30–35% by NH4+ and NH4+ uptakewas decreased 9–24%. Removal of NH4+ resulted in completerecovery in K+ uptake rate, but NH4+ uptake rate did not recoverwhen K+ was removed. In both cases, inhibition of the uptakerate of one cation saturated as the concentration of the othercation was increased up to 0.5 mol m–3. The relative effectof K+-NH4+ interactions was not altered when Cl- was replacedwith SO42–, but the magnitudes of the uptake rates wereless in the absence of Cl-. The Vmax for NH4+ uptake was reducedfrom 128 to 105 µmol g–1 dry wt. h–1 in thepresence of 0.5 mol m–3 K+ and the Km for NH4+ doubledfrom 12 to 27 mmol m–3 in the presence of K+. The resultsof these K+-NH4+ experiments are interpreted as mixed-noncompetitiveinteractions. However, an enhanced efflux of K+ coupled to NH4+influx via an antiporter cannot be ruled out as contributingto the decrease in net K+ uptake. Relationships between nitrate (NO-3) supply, uptake and assimilation,water uptake and the rate of mobilization of seed reserves wereexamined for the five main temperate cereals prior to emergencefrom the substrate. For all species, 21 d after sowing (DAS),residual seed dry weight (d.wt) decreased while shoot plus rootd.wt increased (15–30%) with increased applied NO-3concentrationfrom 0 to 5–20 mM . Nitrogen (N) uptake and assimilationwere as great with addition of 5 mM ammonium (NH+4) or 5 mMNO-3but NH+4did not affect the rate of mobilization of seedreserves. Chloride (Cl-) was similar to NO-3in its effect onmobilization of seed reserves of barley (Hordeum vulgare L.).Increased rate of mobilization of seed reserves with additionalNO-3or Cl-was associated with increases in shoot, root and residualseed anion content, total seedling water and residual seed watercontent (% water) 21 DAS. Addition of NH+4did not affect totalseedling water or residual seed water content. For barley suppliedwith different concentrations of NO-3or mannitol, the rate ofmobilization of seed reserves was positively correlated (r >0.95)with total seedling water and residual seed water content. Therate of mobilization of seed reserves of barley was greaterfor high N content seed than for low N content seed. Seed watercontent was greater for high N seed than for low N seed, 2 DAS.Additional NO-3did not affect total seedling water or residualseed water content until 10–14 DAS. The effects of seedN and NO-3on mobilization of seed reserves were detected 10and 14 DAS, respectively. It is proposed that the increasedrate of mobilization of seed reserves of temperate cereals withadditional NO-3is due to increased water uptake by the seedlingwhile the seed N effect is due to increased water uptake bythe seed directly. Uptake capabilities for ammonium (NH4+) and urea by diatoms(Thalassiosira pseudonana and Skeletonema costatum) growingon oxidized forms of nitrogen were studied in short-term uptakeexperiments. Even when nutrient-saturated, an enhanced uptakecapability not coupled with the growth rate was present forNH4+ and urea. No such enhanced uptake ability was seen forNO2 or NO3 under either nutrient-saturated ornutrient-depleted conditions. The presence of NH4+ decreasedthe enhanced ability to take up urea, but the urea uptake ratein 5 min incubations remained greater than the growth rate evenwhen NH4+ was present.  相似文献

Allen, S. and Smith, J A. C. 1986. Ammonium nutrition in Ricinuscommunis: its effect on plantgrowth and the chemical compositionof the whole plant, xylem and phloem saps.—J. exp. Bot.37: 1599–1610. The growth and chemical composition of Ricinus communis cultivatedhydroponically on 12 mol m – 3 NO3-N were comparedwith plants raised on a range of NH4+-N concentrations. At NH4+-Nconcentrations between 0·5 and 4·0 mol m–3,fresh- and dry-weight yields of 62-d-old plants were not significantlydifferent from those of the NO3-N controls. Growth wasreduced at 0·2 mol m–3 NH4+-N and was associatedwith increased root. shoot and C: organic N ratios, suggestingthat the plants were N-limited. At 8·0 mol m–3NH4+-N, growth was greatly restricted and the plants exhibitedsymptoms of severe ‘NH4+ toxicity’. Plants growingon NH4+-N showed marked acidification of the rooting medium,this effect being greatest on media supporting the highest growthrates. Shoot carboxylate content per unit dry weight was lower at mostNH4+-N concentrations than in the NO3-N controls, althoughit increased at the lowest NH4+-N levels. Root carboxylate contentwas comparable on the two N sources, but also increased substantiallyat the lowest NH4+-N levels. N source had little effect on inorganic-cationcontent at the whole-plant level, while NO3 and carboxylatewere replaced by Cl as the dominant anion in the NH4+-N plants.This was reflected in the ionic composition of the xylem andleaf-cell saps, the latter containing about 100 mol m–3Cl in plants on 8·0 mol m–3 NH4+. Xylem-saporganic-N concentration increased more than threefold with NH4+-N(with glutamine being the dominant compound irrespective ofN source) while in leaf-cell sap it increased more than 12-foldon NH4+-N media (with arginine becoming the dominant species).In the phloem, N source had little or no effect on inorganic-cation,sucrose or organic-N concentrations or sap pH, but sap fromNH4+-N plants contained high levels of Cl and serine. Collectively, the results suggested that the toxic effects ofhigh NH4+ concentrations were not the result of medium acidification,reduced inorganic-cation or carboxylate levels, or restrictedcarbohydrate availability, as is commonly supposed. Rather,NH4+ toxicity in R. communis is probably the result of changesin protein N turnover and impairment of the photorespiratoryN cycle. This work aimed to study the impacts of acquisition and assimilationof various nitrogen sources, i.e. NO3, NH4+ or NH4NO3,in combination with gaseous NH3 on plant growth and acid-basebalance in higher plants. Plants of C3 Triticum aestivum L.and C4 Zea mays L. grown with shoots in ambient air in hydroponicculture solutions with 2 mol m–3 of nitrogen source asNO3, NH4+ or NH4NO3 for 21 d and 18 d, respectively,had their shoots exposed either to 320 µg m–3 NH3or to ambient air for 7 d. Variations in plant growth (leaves,stubble and roots), and OH and H+ extrusions as wellas the relative increases in nitrogen, carbon and carboxylatewere determined. These data were computed as H+/N, H+/C, (C-A)/N,and (C-A)/C to analyse influences of different nitrogen sourceson acid-base balance in C3 Triticum aestivum and C4 Zea maysplants. Root growth in dry weight gain was significantly reduced bytreatment with 320 µg m–3 NH3 in Triticum aestivumand Zea mays growing with different N-forms, whereas leaf growthwas not significantly affected by NH3. In comparison with C3Triticum aestivum, non-fumigated C4 Zea mays had low ratiosof OH/N in NO3–3-grown plants and of H+/N in NH4+- and NH 4NO3-grown plants. Utilization of NH3 from the atmospherereduced both the OHN ratios in NO3 -grown plantsand the H+/N ratio in NH4+ - and NH4NO3 -grown plants of bothspecies. Furthermore, Zea mays had higher ratios of (C-A)/Nin NH4+ - and NH4NO3-grown plants than Triticum aestivum. Thismeans that C4 Zea mays had synthesized more organic anion perunit increase in organic N than C3 Triticum aestivum plants.Within both species, different nitrogen sources altered theratios of (C-A)/N in the order: NH4NO3>NH4+>NO3.Fumigation with NH3 increased organic acid synthesis in NO3- and NH4+ - grown plants of Triticum aestivum, whereas it decreasedorganic acid synthesis in Zea mays plants under the same conditions.Furthermore, these differences in acid-base regulation betweenC3 Triticum aestivum and C4 Zea mays plants growing with differentnitrogen sources are discussed. The processes of NO3 uptake and transport and the effectsof NH4+ or L-glutamate on these processes were investigatedwith excised non-mycorrhizal beech (Fagus sylvatica L.) roots.NO3 net uptake followed uniphasic Michaelis-Menten kineticsin a concentration range of 10µM to 1 mM with an apparentKm of 9.2 µM and a Vmax of 366 nmol g–1 FW h–1.NH4+, when present in excess to NO3, or 10 mM L-glutamateinhibited the net uptake of NO3 Apparently, part of NO3taken up was loaded into the xylem. Relative xylem loading ofNO3 ranged from 3.21.6 to 6.45.1% of NO3 netuptake. It was not affected by treatment with NH4+ or L-glutamate.16N/13N double labelling experiments showed that NO3efflux from roots increased with increasing influx of NO3and, therefore, declined if influx was reduced by NH4+ or L-glutamateexposure. From these results it is concluded that NO3net uptake by non-mycorrhizal beech roots is reduced by NH4+or L-glutamate at the level of influx and not at the level ofefflux. Shelp, B. J. 1987. Plant characteristics and nutrient compositionand mobility of broccoli (Brassica oleracea var. italica) suppliedwith NH+4, NO3 or NH4NO3.—J. exp. Bot. 38: 1603–1618. The effects of varying NH+4, NO3 or NH4NO3 concentration onthe final plant characteristics, element composition, and accumulationof NO3-N, NH+4-N and organic-N were evaluated in broccoli (Brassicaoleracea var. italicacv. Futura and Premium Crop) plants culturedin vermiculite under greenhouse conditions supplemented withlight. NH+4-grown plants were stunted and exhibited signs ofmarginal necrosis on the old leaves, accompanied by an accumulationof NH4. The tissue levels of N, P, Mn, Cu, Zn and B were generallyincreased by NH+4 versus NO3 nutrition whereas the reverse wastrue for Ca; Mg and K were only slightly affected, if at all.These results are attributed to: changes in element availabilityresulting from reduced rhizosphere pH due to NH+4uptake ratherthan NO 3uptake; competition of Ca uptake by NH+4; and dilutionof N by increased vegetative growth with NO3-nutrition. Theelement concentrations of N, P or K were similar in all tissueswhereas Ca, B and Mn were markedly less in the florets and youngleaves compared to mature leaves; this supports literature indicatingthat the former elements are phloem-mobile whereas the latterare not. Assuming that the nutrient supply for mature leavesis delivered principally via the xylem stream, the data suggestthat nutrients for developing leaves and florets are suppliedpredominantly in the phloem. If so, under our experimental conditions.Zn and Cu were also readily mobile in the phloem whereas Mgmovement was restricted. NH4+ versus NO4+ J nutrition alteredthe distribution of these elements. The two broccoli cultivarstested under the greenhouse environment differed in NH+4 toleranceand in the distribution of K and Cu suggesting there was a geneticbasis for cultivar variation in mineral acquisition and redistribution. Light-enhanced active pyruvate uptake into mesophyll chloroplastsof C4 plants was reported to be mimicked by either of the twotypes of cation jump: H+-jump in maize and phylogenically relatedspecies (H+-type) and Na+-jump in all the other C4 species tested(Na+-type) [Aoki, N., Ohnishi, J. and Kanai, R. (1992) PlantCell Physiol. 33: 805]. In this study, medium and stromal pH was monitored in the suspensionof C4 mesophyll chloroplasts. Medium alkalization lasting for5 to 10 seconds after pyruvate addition was detected by a pHelectrode and observed only in the light and only in mesophyllchloroplasts from H+-type species, Zea mays L. and Coix lacryma-jobiL., but not in those from Na+-type species Panicum miliaceumL., Setaria italica (L.) Beauv. and Panicum maximum Jacq. Theinitial rate of H+ consumption showed good correlation with[14C]pyruvate uptake measured by silicone oil filtering centrifugation,both being inhibited by N-ethylmaleimide and 7-chloro-4-nitrobenzo-2-oxa-l,3-diazole to the same degree. The ratio of the rate of H+ uptaketo that of pyruvate uptake was always about 1. Pyruvate-inducedacidification of the stroma was observed in maize mesophyllchloroplasts. Inorganic carbon-dependent O2 evolution, nitrate utilizationand nitrogen recycling.—J. exp Bot. 36: 1373–1386 Scenedesmus obtusiusculus Chod. was grown on an inorganic mediumflushed with either air or air supplemented with 3% CO2. Inair-grown cells, O2 evolution dependent on low, but not high,HCO3 concentrations was strongly inhibited by the carbonicanhydrase inhibitor acetazolamide. Cells grown with 3% CO2 exhibitedlow rates of O2 evolution at low external inorganic C; however,after 30 min in air O2 evolution rates at low inorganic C approachedthose of air-grown cells. These results are compatible withthe view that Scenedesmus develops a ‘CO2 concentratingmechanism’ in air, with carbonic anhydrase as an importantconstituent When 3% CO2-grown cells were subjected to air-level of CO2,just a transient decline in NO3 utilization was observed,but in the presence of acetazolamide the rate of the processdecreased drastically in response to the decrease in the CO2level. In CO2-free air NO3 was taken up at high ratesbut in a deregulated manner, leading to release of NH4+. A portionof the NO3 taken up in the absence of CO2 was apparentlyassimilated Cellular nitrate reductase (NR) activity initially decreasedbut subsequently recovered after a transition from 3% CO2 toair. In the presence of acetazolamide, a persistent decreasein NR activity was observed. Cellular glutamine synthetase (GS)activity increased after transition from 3% CO2 to air, theactivity increase being unaffected by acetazolamide. NH4+ releaseto the medium in the presence of L-methionine-D, L-sulphoximine(MSO) transiently increased in 3% CO2-grown cells in responseto a transfer to air. MSO-induced NH4+ release was in fact higherin air-grown cells than in 3% CO2-grown cells. Glycollate wasinitially released after transition from 3% CO2 to air, butthere was no difference in glycollate release between MSO-treatedand untreated cells. The effects of NO-3 and NH+4 nutrition on hydroponically grownwheat (Triticum aestivum L.) and maize (Zea mays L.) were assessedfrom measurements of growth, gas exchange and xylem sap nitrogencontents. Biomass accumulation and shoot moisture contents ofwheat and maize were lower with NH+4 than with NO-3 nutrition.The shoot:root ratios of wheat plants were increased with NH+4compared to NO-3 nutrition, while those of maize were unaffectedby the nitrogen source. Differences between NO-3 and NH+4-fedplant biomasses were apparent soon after introduction of thenitrogen into the root medium of both wheat and maize, and thesedifferences were compounded during growth. Photosynthetic rates of 4 mM N-fed wheat were unaffected bythe form of nitrogen supplied whereas those of 12 mM NH+4-fedwheat plants were reduced to 85% of those 12 mM NO-3-fed wheatplants. In maize supplied with 4 and 12 mM NH+4 the photosyntheticrates were 87 and 82% respectively of those of NO-3-fed plants.Reduced photosynthetic rates of NH+4 compared to NO-3-fed wheatand maize plants may thus partially explain reduced biomassaccumulation in plants supplied with NH+4 compared to NO-3 nutrition.Differences in the partitioning of biomass between the shootsand roots of NO-3-and NH+4-fed plants may also, however, arisefrom xylem translocation of carbon from the root to the shootin the form of amino compounds. The organic nitrogen contentof xylem sap was found to be considerably higher in NH+4- thanin NO-3-fed plants. This may result in depletion of root carbohydrateresources through translocation of amino compounds to the shootin NH+4-fed wheat plants. The concentration of carbon associatedwith organic nitrogen in the xylem sap of maize was considerablyhigher than that in wheat. The relationship between thef-ratio [NO3 uptake/(NO3+ NH4+) uptake] and ambient nitrate concentration was evaluatedfor eight data sets from coastal waters. The f-ratio increasedasymptotically with increase in nitrate concentration in mostdata sets. However, the rate at which f-ratio increased at lownitrate concentration (slope = m) and the maximum attained f-ratio(fmax) varied among regions; the initial slope varied most withvalues ranging in excess of an order of magnitude. The datawere analyzed in relation to environmental factors and methodologicalconsiderations known to influence the f-ratio. Ambient ammoniumconcentration was important in accounting for regional differencesin the f versus NO3 relationship. A further analysisof the data, relating f-ratio to the ratio of NO3/(NO3+ NH4+) concentrations yielded a much more regionally consistentand approximately linear relationship; slopes varied by lessthan a factor of two in the extreme cases. Inclusion of knownalternative (aside from NH4+) sources of reduced-N (e.g. urea)and correction for methodological/computational errors (isotopedilution) systematically reduce f-ratio estimates. Other factors,e.g. reduced-N uptake by microheterotrophs, may systematicallyincrease the f-ratio.  相似文献

Ricinus communis L. var. Gibsonii was grown in Long Ashton nutrientmedium with either 12mol m–3 NO3 or 8.0 mol m–3NH+4 as N source. Two plants from each N treatment were harvestedtwice a week and analysed for C, N, P, S, NO3, SO2–4ClK+Na+, Ca2+ Mg2+ and ash alkalinity. Statistical analysis of thedata showed that the effect of age and N source was differentfor the chemical variables analysed. Thus [Na+] was unaffectedby age or N source, and for both N sources [Mg2+] started atthe same level and decreased at the same rate as the plantsmatured. With NH+4 as N source, [SO2–4] was higher thanwith NO3, but did not alter with age. The concentrations,in mmol g–1 dry wt, of C, organic N, K+ and Ca2+ weredifferent for the two N sources, but the levels of these variablesaltered with age in the same way for both N sources; i.e. therewas no age x N interaction. In the case of P, NO3, Cl and COO, however,age-related variations were different for the two N sources.It is concluded, inter alia, that [Na+] is determined by external[Na+] alone, and that K+, Ca2+ and Cl are the inorganicions actively involved in charge balance during ion uptake bythe roots. Respiratory oxygen consumption by roots was 1·4- and1·6-fold larger in NH+4-fed than in NO-3-fed wheat (Triticumaestivum L.) and maize (Zea mays L.) plants respectively. Higherroot oxygen consumption in NH+4-fed plants than in NO-3-fedplants was associated with higher total nitrogen contents inNH+4-fed plants. Root oxygen consumption was, however, not correlatedwith growth rates or shoot:root ratios. Carbon dioxide releasewas 1·4- and 1·2-fold larger in NO+3-fed thanin NH+4-fed wheat and maize plants respectively. Differencesin oxygen and carbon dioxide gas exchange rates resulted inthe gas exchange quotients of NH-4-fed plants (wheat, 0·5;maize, 0·6) being greatly reduced compared with thoseof NO-3-fed plants (wheat, 1·0; maize, 1·1). Measuredrates of HCO-3 assimilation by PEPc in roots were considerablylarger in 4 mM NH+4-fed than in 4 NO-3 plants (wheat, 2·6-fold;maize, 8·3-fold). These differences were, however, insufficientto account for the observed differences in root carbon dioxideflux and it is probable that HCO-3 uptake is also importantin determining carbon dioxide fluxes. As rice can use both nitrate (NO3-) and ammonium (NH4+), we have tested the hypothesis that the shift in the pattern of cultivars grown in Jiangsu Province reflects the ability of the plants to exploit NO3- as a nitrogen (N) source. Four rice cultivars were grown in solution culture for comparison of their growth on NO3- and NH4+ nitrogen sources. All four types of rice, Xian You 63 (XY63), Yang Dao 6 (YD), Nong Keng 57 (NK) and Si You 917 (SY917), grew well and produced similar amounts of shoot biomass with 1 mmol/L NH4+ as the only N source. However, the roots of NK were significantly smaller in comparison with the other cultivars. When supplied with 1 mmol/L NO3-, YD produced the greatest biomass; while NK achieved the lowest growth among the four cultivars. Electrophysiological measurements on root rhizodermal cells showed that the NO3--elicited changes in membrane potential (ΔEm) of these four rice cultivars were significantly different when exposed to low external NO3- (<1 mmol/L); while they were very similar at high external NO3- (10 mmol/L). The root cell membrane potentials of YD and XY63 were more responsive to low external NO3- than those of NK and SY917. The ΔEm values for YD and XY63 rhizodermal cells were almost the same at both 0.1 mmol/L and 1 mmol/L NO3-; while for the NK and SY917 the values became larger as the external NO3- increased. For YD cultivar, ΔEm was measured over a range of NO3- concentrations and a Michaelis-Menten fit to the data gave a Km value of 0.17 mmol/L. Net NO3- uptake depletion kinetics were also compared and for some cultivars (YD and XY63) a single-phase uptake system with first order kinetics best fitted the data; while other cultivars (ND and SY917) showed a better fit to two uptake systems. These uptake systems had two affinity ranges: one had a similar Km in all the cultivars (0.2 mmol/L); the other much higher affinity system (0.03 mmol/L) was only present in NK and SY917. The expression pattern of twelve different NO3- transporter genes was tested using specific primers, but only OsNRT1.1 and OsNRT2.1 expression could be detected showing significant differences between the four rice cultivars. The results from both the physiological and molecular experiments do provide some support for the hypothesis that the more popular rice cultivars grown in Jiangsu Province may be better at using NO3- as an N source.  相似文献

Plants of Lolium perenne L. cv. S23 were grown in sand culturesupplied with either ammonium (NH4+) or nitrate (NO3)in an otherwise complete nutrient solution at 12°C or 20°C.Three weeks after germination, plants were clipped weekly tosimulate grazing. After 10 weeks growth all nitrogen (N) wassupplied enriched with 15N to quantify the effects of form ofN supply and temperature on the relative ability of currentroot uptake and remobilization to supply N for laminae regrowth. The form of N supply had no effect on the dry matter partitioning,while at 20°C more dry weight was allocated to laminae regrowthand less to the remaining plant material. The current root uptakeof N, which subsequently appeared in the laminae regrowth, wassimilar for plants supplied with NH4+ or NO3, and bothwere equally reduced at the lower temperature of growth. Remobilizationof N to laminae regrowth was greater for plants receiving NH4+than NO3; remobilization with either form of N supplywas reduced at the lower temperature of growth. Remobilizationwas reduced to a lesser extent at 12°C than current rootuptake. It was concluded that remobilization became relativelymore important in supplying N for regrowth of laminae at lowertemperatures.
