Rapidly dividing photoautotrophic cell suspensions from Chenopodium rubrum L. assimilated about 85 μmol CO2 (mg chlorophyll)−1 h−1. During the late stationary phase of culture growth, CO2 fixation rate was reduced to about 60 μmol CO2 (mg chlorophyll)−1 h−1. Actively dividing cells characteristically incorporated a smaller proportion of 14C into starch than cells from non-dividing stationary phases. In rapidly dividing cells, [14C]-turnover from free sugars, sugar-phosphates, organic and amino acids was substantially higher compared to non-dividing cells from stationary growth phase. Higher proportions of photosynthetically fixed carbon were channelled into proteins, lipids and structural components in actively dividing cells than in non-dividing cells. In the latter. 14C was preferentially channeled into starch, and a striking increase in starch accumulation was observed. The transfer of non-dividing, stationary growth-phase cells into fresh culture medium resulted in an increase in the maximum extractable activities of some enzymes involved in the glycolytic and dark respiratory pathways and in the citric acid cycle. In contrast, the maximum extractable activities of the chloroplastic enzymes, ribulose-1,5-bisphosphate carboxylase (EC 4.1.1.38) and NADP+-glyceraldehyde-3-phosphate dehydrogenase (EC 1.2.1.13) were highest after the cells had reached the stationary growth phase.  相似文献

Specific superantigens activate different T-cell fractions with distinct TCR Vβ elements in association with MHC class II molecules and also induce SDCC against MHC class II+ target cells. In the present study, to determine whether the responsiveness of each CD8+ T-cell fraction expressing a different TCR Vβ element is primarily determined by the TCR Vβ, we compared the levels of proliferation and SDCC in Vβ3+ and Vβ11+ T cells upon stimulation with SEA. Upon stimulation with SEAwt, the levels of proliferation were higher in Vβ3+ T cells than in Vβ11+ T cells. The levels of SDCC were also higher for the combination of Vβ3+ T cells and SEAwt than for the combination of Vβ11+ T cells and SEAwt during both the induction phase and the effector phase. In addition, upon stimulation with SEAm, the levels of proliferation were higher in Vβ11+ T cells than in Vβ3+ T cells. And then, the levels of SDCC were also higher for the combination of Vβ11+ T cells and SEAm than for the combination of Vβ3+ T cells and SEAm during both the induction phase and the effector phase. These results suggest that the SAG-responsiveness of each CD8+ T-cell fraction expressing a different TCR Vβ element is primarily determined by the interaction between the TCR Vβ element and the SAG.  相似文献

Aims:  The ability to transform Vibrio spp. is limited by the extracellular nuclease that their cells secrete. The reported transformation efficiency of this organism is 102–105 transformants per microgram DNA. We tried different buffers and conditions, aiming to elevate its transformation efficiency.
Methods and Results:  MgCl2 and sucrose are often included in the washing and/or electroporation buffers to stabilize the cell membrane. However, Mg2+ is required for production and activity of the extracellular nuclease. A simple electroporation buffer lacking Mg2+ was found to increase transformation efficiency dramatically, to levels 50-fold more than the buffers containing Mg2+. To maintain the stability of the cell membranes, Mg2+ was replaced with high concentrations of sucrose, from 272 to 408 mmol l−1. With the new buffers, the transformation efficiency of Vibrio parahaemolyticus was increased to 2·2 × 106 transformants per microgram DNA.
Conclusions:  Mg2+ in the buffer adversely affected transformation of V. parahaemolyticus by electroporation. The cell membranes of vibrio can be stabilized by high concentration of sucrose when Mg2+ is absent.
Significance and Impact of the Study:  A greater transformation efficiency can facilitate the genetic analysis of an organism and its pathogenicity. Buffers lacking Mg2+ can be used for other nuclease-producing organisms.  相似文献

Abstract: Cell dissociates from embryonic chick dorsal root ganglia, incubated for 6 h with 22Na+, accumulated four to six times more radioactivity in the absence than in the presence of Nerve Growth Factor (NGF). The accumulation of radioactivity paralleled the external Na+ concentration, indicating that the cells may have been reaching equilibrium with the medium. Delayed presentation of NGF to 22Na+-loaded cells caused a rapid loss of radioactivity, even with extracellular 22Na+ still present, demonstrating that NGF caused an overall efflux of Na+ rather than an accelerated equilibration. The Na+ exclusion from 22Na+-loaded cells was dependent upon NGF concentration. Use of nutrient-rich medium, serum, and certain hormones and other proteins did not prevent the Na+ accumulation in the absence of NGF or its reversion by delayed NGF administration. Incubation of the ganglionic cells with ouabain or dinitrophenol during the 22Na+ loading period (no NGF) increased the rate, but not the magnitude, of loading. The same incubation carried out in a Na+-free medium and followed by 22Na+ presentation resulted in fast radioactive loading that was identical to that occurring in drug-free, NGF-deprived cells and was not prevented by presentation of NGF together with the 22Na+. These data are consistent with a model in which NGF acts through a Na+ pump rather than by restricting Na+ influxes.  相似文献

A new epidemic, NTED, has recently occurred in Japan. The cause of NTED is a bacterial superantigen, TSST-1. The aim of the present study was to analyze the change in Vβ2+ T cells reactive to TSST-1 in NTED in order to establish T-cell-targeted diagnostic criteria for NTED. Blood samples from 75 patients with clinically diagnosed NTED were collected from 13 neonatal intensive care units throughout Japan. We investigated the percentages of Vβ2+, Vβ3+ and Vβ12+ T cells and their CD45RO expressions in the samples using flow cytometry. In 18 of the 75 patients, we conducted multiple examinations of the T cells and monitored serial changes. The Vβ2+ T-cell population rapidly changed over three phases of the disease. Whereas the percentage of Vβ2+ T cells was widely distributed over the entire control range, CD45RO expression on Vβ2+ T cells in CD4+ in all 75 patients was consistently higher than the control range. Patients cannot necessarily be diagnosed as having NTED based on expansion of Vβ2+ T cells alone in the early acute phase. Instead, CD45RO expression on specific Vβ2+ cells is a potential diagnostic marker for a rapid diagnosis of NTED. We present three diagnostic categories of NTED. Fifty patients (66.7%) were included in the category 'definitive NTED'. It is important to demonstrate an increase of Vβ2+ T cells in the following phase in cases of 'probable NTED' or 'possible NTED'.  相似文献

Abstract: Na+ flux was studied in cultured neuroblastoma cells grown in medium containing increased glucose or L - fucose concentrations. Chronic exposure of neuroblastoma cells to 30 m M glucose or 30 m M L-fucose caused a decrease in ouabain-sensitive and veratridine-stimulated 22Na+ uptake compared with cells cultured in unsupplemented medium. The Na+ current, determined by using whole-cell configuration of the patch clamp, was also decreased in these cells. Tetrodotoxin (3 μ M ), which blocked whole cell Na+ currents, also blocked veratridine-stimulated 22Na+ accumulation. Culturing cells in medium containing 30 m M fructose as an osmotic control had no effect on Na+ flux. Specific [3H] saxitoxin binding was not affected by 30 m M glucose or 30 m M L-fucose compared with cells grown in unsupplemented medium, suggesting that the number of Na+ channels was not decreased. These studies suggest that exposing cultured neuronal cells to conditions that occur in the diabetic milieu alters Na+ transport and Na+-channel activity.  相似文献

Domination of Carnobacterium divergens LV13 by a bacteriocin-producing (bac+) organism Carnobacterium piscicola LV17 was dependent on the level of inoculum of the producer strain and its bacteriocin production. When C. piscicola LV17 was grown in APT broth from an initial inoculum of α-104 cfu ml-1, bacteriocin was not produced (bac-) although maximum population was reached. The culture remained bac- during subsequent inoculation at 102-107 cfu ml-1 unless it was first grown on solid medium or if heat-treated supernatant fluids from a bac+ culture of C. piscicola LV17, LV17A or LV17B were added to the culture prior to the stationary phase of growth. Use of purified carnobacteriocins from C. piscicola LV17A and LV17B confirmed their role in regulation of the bac+ phenotype. The need for induction might account in part for differences in bacteriocin production by cultures in liquid and on solid growth media.  相似文献

The mechanism of cobalt uptake was investigated using cells of the giant alga Chara corallina in which it is possible to resolve separately uptake by the cell wall and actual influx across the cell membrane. The absorption of 60Co by Chara cells appeared to saturate within 2 h, but this was mainly due to rapid uptake into the cell wall which accounted for 87–92% of the total activity. Even after prolonged desorption most of the cell‐associated 60Co was found on the cell wall. The intracellular distribution of absorbed 60Co was investigated by fractionating the cell into cytoplasm and vacuole. It was shown that 60Co influx to the vacuole occurs simultaneously with influx to the cytoplasm. The transported species appears to be Co2+ rather than the less charged Co(OH)+ or Co(OH)2. 60Co influx is pH dependent (optimum pH 7–9), and is sensitive to some other divalent metals. Influx from solutions containing 1 µ M 60Co was inhibited by 5 µ M Cd2+, Cu2+, and Zn2+, but Mn2+ and Ni2+ had no significant effect. The sensitivity of Co uptake to N ‐ethyl maleimide (NEM) and cysteine suggests that the transport system involves direct binding of CO2+ to ‐SH groups.  相似文献

Abstract— Desheathed rat dorsal root ganglia were incubated in a medium containing amino-oxyacetic acid and [3H]GABA. Under these conditions, [3H]GABA is taken up exclusively by the satellite glial cells in the ganglia. Efflux of [3H]GABA from the tissue was measured after passing the ganglia through a series of wash solutions. The spontaneous efflux of radioactivity, mostly [3H]GABA, was more rapid in the absence of amino-oxyacetic acid in the incubation and wash media.
Raising the potassium concentration in the wash media caused an increase in the efflux of [3H]GABA. This increase was sigmoidally related to the potassium concentration in the wash media, reaching a maximum at 64 m m -K+. The releasing effect of K+ was inhibited by removing calcium from the media. Reducing the calcium and raising the magnesium concentration in the wash solutions inhibited the increased efflux of [3H]GABA due to 64 m m -K+ by 48 per cent, while 5 mM-La3+ and diphenylhydantoin (0·005 and 0·5 m m ) had no effect on this increase.
Only a small increase in the efflux of [14C]glutamate was produced by 64 m m -K+ and it had no effect upon the effluxes of [3H]glycine, [3H]alanine or [3H]leucine. The efflux of lactate dehydrogenase was similarly unaffected by 64 mM-K+. The results suggest that glial cells in spinal ganglia can respond to depolarizing concentrations of potassium by releasing GABA in a calcium-dependent process.  相似文献

A rather specific inhibitor of calmodulin-regulated functions, compound 48/80. inhibited 36Cl, 86Rb+ and 22Na+ absorption by Hordeum vulgare, Phaseolus mungo and Zea mays roots. The compound did not inhibit plasmalemma or tonoplast Mg:2-ATPase activity; neither did it inhibit or uncouple respiration, or affect ATP content and seemed also not to cause depolarization of the membrane potential of roots cells. Compound 48/80 inhibited the ΔpH-mediated Cl-H+ symport in ATP-depleted roots.  相似文献

The photosynthetic characteristics of the terrestrial cyanobacterium, Nostoc flagelliforme , after complete recovery by rewetting, was investigated to see whether it could use bicarbonate as the external inorganic carbon source when submerged. The photosynthesis–pH relationship and high pH compensation point suggested that the terrestrial alga could use bicarbonate to photosynthesize when submerged. The photosynthetic oxygen evolution rates were significantly inhibited in Na + -free and Na + + Li + media but were not affected by the absence of Cl , implying that the bicarbonate uptake was associated with Na + / HCO3 symport rather than Cl /HCO3 exchange system.  相似文献

Abstract: We have previously reported that insulin/insulin-like growth factor (IGF)-I induced the α1 isoform of Na+,K+-ATPase in cultured astrocytes. In this study the effects of insulin/IGF-I on Na+,K+-ATPase activity and cell proliferation were examined in astrocytes cultured under the various conditions, to test the possible involvement of the enzyme activity in the mitogenic action of IGF-I on astrocytes. Insulin increased Na+,K+-ATPase activity and stimulated cell proliferation in subconfluent astrocytes (cultured for 7–14 days in vitro). In contrast, these effects were not observed in confluent cells (cultured for 28 days). Furthermore, insulin stimulated neither the enzyme activity nor [3H]thymidine incorporation in astrocytes preincubated in fetal calf serum-free medium for 2 days (quiescent cells) and treated with dibutyryl cyclic AMP (differentiated cells). The increases in Na+,K+-ATPase activity and expression of the α1 mRNA preceded the mitogenic effect. 125I-IGF-I binding experiment showed that all the cells used here had similar binding characteristics. The insulin-induced increase in enzyme activity was not affected by 1-(5-isoquinolinesulfonyl)-2-methylpiperazine (H-7), and it was observed even in Ca2+-free medium. The stimulation by IGF-I of [3H]thymidine incorporation was attenuated by ouabain and a low external K+ level. These findings suggest that stimulation of Na+,K+-ATPase activity is involved in the mitogenic action of IGF-I on cultured astrocytes.  相似文献
