In the United States alone, the National Heart, Lung, and Blood Institute (NHLBI) has invested several hundred million dollars in pursuit of myocardial infarct-sparing therapies. However, due largely to methodological limitations, this investment has not produced any notable clinical application or cardioprotective therapy. Among the major methodological limitations is the reliance on animal models that do not mimic the clinical situation. In this context, the limited use of conscious animal models is of major concern. In fact, whenever possible, studies of cardiovascular physiology and pathophysiology should be conducted in conscious, complex models to avoid the complications associated with the use of anesthesia and surgical trauma. The mouse has significant advantages over other experimental models for the investigation of infarct-sparing therapies. The mouse is inexpensive, has a high throughput, and presents the ability of one to create genetically modified models. However, successful infarct-sparing therapies in anesthetized mice or isolated mouse hearts may not be successful in more complex models, including conscious mice. Accordingly, a conscious mouse model of myocardial ischemia and reperfusion has the potential to be of major importance for advancing the concepts and methods that drive the development of infarct-sparing therapies. Therefore, we describe, for the first time, the use of an intact, conscious, and unrestrained mouse model of myocardial ischemia-reperfusion and infarction. The conscious mouse model permits occlusion and reperfusion of the left anterior descending coronary artery in an intact, complex model free of the confounding influences of anesthetics and surgical trauma. This methodology may be adopted for advancing the concepts and ideas that drive cardiovascular research.  相似文献

Anthropology has conventionally taken as some of its most cherished foundational categories the precise opposites of the key concepts that animate this inquiry: rather than “bare life,” anthropology has tended always to emphasize the fullness and complexity of social and political life; instead of labor in the abstract, which we recognize in its commodified form as “labor-power,” anthropology has produced exquisite inventories of concrete laboring activities and the “cultural” content of productive work; against the impermanence and mutability of lives characterized by their mobility, the ethnographic enterprise has been deeply attached to the sedentarist presuppositions of lasting settlement, dwelling, and “community”; and contrary to the task of apprehending space on a global scale, ethnographic study has been overwhelmingly localized and place-bound. Rethinking these elementary premises of the ethnographic endeavor and situating these critical concepts at the center of our epistemological frameworks are crucial theoretical and practical tasks for any meaningful social inquiry today. In this regard, the Marxian theoretical arsenal is simply indispensable. But, in the derisive words of so many disciplinary forebears and overseers, is this properly “anthropological”? The prospective convergence of genuinely critical sociopolitical inquiry with the techniques and insights of anthropology must remain for us the locus of an urgent problem—an open question on an open horizon.  相似文献

1-800 COCAINE has provided assistance to over two million callers to date. It has supplied epidemiologic data regarding cocaine use, with increasing proportions of female users since 1983, decrease in average age and income of callers since 1983, and numerous social and medical consequences of use. In addition, it has provided data regarding timing of the progression of cocaine abuse and confirmation that cocaine abuse is an addictive illness for those calling to seek help. It has corroborated other studies in documenting the psychosocial and medical consequences of addiction and has been a source of insight into trends in cocaine addiction. 800-COCAINE is, by its existence and name recognition, a primary prevention project.  相似文献

Recently, research of stem cells has garnered great attention and has shown promise by changing the view of traditional therapeutics, with broad impact on gene therapy, carcinogenesis, organ development, tissue injury, regeneration and almost all aspects of the life cycle and all living systems. A century's scientific progress has significantly improved controls for infectious diseases and many other disorders. However, many remaining problems (i.e. cancer, AIDS, diabetes, Parkinson's disease and Marburg infection) appear to be even harder than those that have already been solved. In particular, respiratory stem cell research has been less active and has moved more slowly than that of many other organs. This is probably due to the complexity of the lung and airway system, particularly owing to the many types of cells (>40), unique structures and functions, and technical difficulty in analyzing this system at the genetic, biochemical, molecular and cellular level. Compared with other epithelial cells (i.e., gastrointestinal epithelium), respiratory epithelia have a very low turnover rate and minimal regenerative activity. This review will discuss the current state of pulmonary stem cells, their origin, development, differentiation, and regenerative application, with a particular focus on potential impact on cancer development and lung injury repair.  相似文献

In modern industrial nations, the traditional positive correlation between female body fat and social class has turned strongly inverse, thinness in women is admired and plumpness is a handicap. This recent reversal of what had seemed to be stable aspects of human nature is analyzed as a potentially adaptive response to two ecological novelties: chronic food surplus and the breakdown of barriers between men's and women's work, which, together, may have made thinness helpful to women competing for status and resources in both mating and job markets. Whether status and resources still promote long-term Darwinian fitness is an open question. Progress in understanding the unique properties of the human mind depends on widespread recognition that the mind has been designed by natural selection to seek and sometimes find adaptive solutions to the novel problems we ourselves create. Adaptive flexibility and cultural change are two sides of the same coin.  相似文献

Ferritin is one of the major proteins of iron metabolism. It is almost ubiquitous and tightly regulated by the metal. Biochemical and structural properties of the ferritins are largely conserved from bacteria to man, although the role in the regulation of iron trafficking varies in the different organisms. Recent studies have clarified some of the major aspects of the reaction between iron and ferritin, which results in the formation of the iron core and production of hydrogen peroxide. The characterization of cellular models in which ferritin expression is modulated has shown that the ferroxidase catalytic site on the H-chain has a central role in regulating iron availability. In turn, this has secondary effects on a number of cellular activities, which include proliferation and resistance to oxidative damage. Moreover, the response to apoptotic stimuli is affected by H-ferritin expression. Altered ferritin L-chain expression has been found in at least two types of genetic disorders, although its role in the determination of the pathology has not been fully clarified. The recent discovery of a new ferritin specific for the mitochondria, which is functionally similar to the H-ferritin, opens new perspectives in the study of the relationships between iron, oxidative damage and free radicals.  相似文献

Although Australia has comparatively few individuals seeking asylum, it has had a mandatory detention policy in place since 1992. This policy has been maintained by successive governments despite the overwhelmingly negative impact mandatory detention has on mental health. For mental health professionals working in this environment, a number of moral, ethical, and human rights issues are raised. These issues are discussed here, with a focus on dual loyalty conflicts and drawing on personal experience, the bioethics and human rights literature, and recent parliamentary inquiries. The culture concept has been central to anthropology since the formational period of the discipline. Yet for much of the discipline's history it was used without explicit definition. Recent attempts to define it have yielded a range of varied formulations in the subdisciplines of archaeology and sociocultural anthropology. Does this mean that the center of anthropology—shared belief in a unified culture concept—has been destroyed? Quite the opposite, the author concludes—the debate has yielded benefits.  相似文献

Cytoskeletal disruption is one of the distinguishing characteristics of the vulnerable neurons in Alzheimer disease (AD). It has been suggested that these cytoskeletal changes occur secondarily to covalent modifications of the protein components. Despite the abundance and probable importance of these changes, there has been very little data regarding the identity of the modified proteins or the precise chemistry of the modifications. Here we review a specific type of modification, namely carbonylation of proteins, which has been shown to be a common result of cellular oxidative stress. Hopefully, the following discussion will help elucidate the relationship between oxidative stress, protein modification and the pathogenesis of AD.  相似文献

The freedom of a doctor to treat an individual patient in the way he believes best has been markedly limited by the concept of evidence-based medicine. Clearly all would wish to practice according to the best available evidence, but it has become accepted that "evidence-based" means that which is derived from randomized, and preferably double-blind, clinical trials. The history of clinical trial development, which can be traced to the use of oranges and lemons for the treatment of scurvy in 1747, has reflected a progressive need to establish whether smaller and smaller effects of treatment are real. It has led to difficult concepts such as "equivalence" and aberrations such as "meta-analysis." An examination of evidence-based practice shows that it has usually been filtered through the opinions of experts and journal editors, and "opinion-based medicine" would be a more appropriate term. In the real world of individual patients with multiple diseases who are receiving a number of different drugs, the practice of evidence-based (or even opinion-based) medicine is extremely difficult. For each patient a judgment has to be made by the clinician of the likely balance of risks and benefits of any therapy. Good practice still requires clinical freedom for doctors.  相似文献

During the last 10–15 years significant progress has been achieved in all directions of studies of the glutathione system. A series of new enzymes involved into metabolism of glutathione has been discovered. Many of these enzymes are polyfunctional and their new activities have been recognized. The enzymes interact with hormones and signal transduction systems. Significant progress has been achieved in the studies of intracellular, intercellular and inter-organ transport. The protease which initiates the massive protein degradation early in bacterial spore germination has been purified from Bacillus megaterium spores. The enzyme has a molecular weight of 160,000 and contains four apparently identical subunits, but only the tetramer is enzymatically active. A radioimmunoassay has been developed for this enzyme and has been used to show that the protease is absent from growing cells, but appears early in sporulation within the developing forespore. In contrast, the protease antigen disappears rapidly during spore germination, in parallel with the loss in enzyme activity.  相似文献

The cell has been represented as a charged liquid drop. Contrary to the DLVO-theory, the effect of the surface potential upon the value of the interfacial tension of the cell membrane has also been taken into consideration. The cell membrane has visco-elastic properties and its constituents may move against each other. Cell movement is caused by the appearance of a small number of the electrically charged constituents of the cell membrane on the leading edge of the cell. This produces a local decrease in the surface tension and the cell membrane expansion. At the moment of contact between two cells proton transfers occur between the strongly negatively charged microvilli of one cell and the body of the other, analogous to a condenser breakdown. This, through the effect on the surface tension, causes contact inhibition of movement. The distribution of the proton dissociable groups modifies the interaction between the cells (differentiation) and between the cell and the substratum (adhesion). Adsorption of the charged compounds at the surface of the cell membrane, decreasing the surface potential and increasing the surface tension, causes the phenomena of chemotaxis, phagocytosis and pinocytosis. Cell division, considered in the terms of the surface energy, requires an adequate supply of considerable quantities of energy inversely proportional to the surface potential value. In case of a reduction of the distance between the cells, their surface potential and the energetic barrier of the cell division processes increases, and causes contact inhibition of cell division. Due to their high charge, division of neoplastic cells is inhibited much later than division of normal cells, or is completely ininhibited due to geometric conditions. Fusion of the cell membrane in the intra-cellular and intercellular processes is a reverse process in relation to the cell division.  相似文献

An unrecognized pentahydroxy bile alcohol has been isolated from the bile and feces of patients with cerebrotendinous xanthomatosis (CTX). Its structure, 5 β-cholestane-3 α, 7 α, 12 α, 25, 26-pentol has been deduced by spectroscopic methods and confirmed by comparison with a synthetic analog.  相似文献
