Benzyladenine (BA) stimulated 5-aminolevulinic acid (ALA) accumulation in the presence of levulinic acid during illumination with 43 μmol m−2 s−1 light in excised etiolated cotyledons of cucumber ( Cucumis sativus L. cv. Aonagajibai). A short dark-pretreatment (6 h) with BA eliminated the lag phase of ALA accumulation. The rate of ALA accumulation during the steady-state phase in cotyledons pretreated with BA for a long period (14 h) was considerably accelerated compared to that in cotyledons pretreated with BA for 6 h. The rate of ALA accumulation during the lag phase was saturated at a very low light fluence (<1.4 μmol m−2 s−1) in both BA-pretreated and water-control cotyledons. The steady-state rate of ALA accumulation increased with increasing light fluence up to 43 μmol m−2 s−1 (parallel to that of Chl formation) in water-control cotyledons. In contrast, in cotyledons pretreated with BA for either 6 or 14 h, the steady-state rate reached a plateau at a very low light fluence. Based on the above results together with our finding that there are two components of Chl formation (M. Dei, 1984. Physiol. Plant. 62: 521–526) possible intermediate steps of Chl biosynthesis pathway affected by BA and light intensity are discussed.  相似文献

Treatment of 3-day-old excised etiolated cotyledons of cucumber ( Cucumis sativus L. cv. Aonagajibai) with benzyladenine (BA) in the dark stimulates chlorophyll (Chl) formation during the lag phase (designated as 'lag elimination') and accelerates the steady-state rate of Chl formation under subsequent continuous illumination with white light. The separation of this two-fold effect is possible using two different methods of BA treatment in darkness: a brief BA treatment followed by various periods of water treatment in darkness, or various periods of continuous dark BA treatment. In either treatment, BA rapidly eliminates the lag phase (the fast-appearing effect) and after a longer time period accelerates the steady-state rate (the late-appearing effect). With a brief BA treatment, both effects decay rapidly. In contrast, with continuous BA treatment, none of the effects decay after reaching their maxima, particularly in cotyledons excised 2 days after sowing and aged for a long period before the onset of BA treatment. These facts indicate that BA acts as a trigger in stimulating Chl formation. The relationship between the actions of BA and light is discussed.  相似文献

In darkness, sporalation of Penicillium claviforme Bainier CBS strain 126–23 was uniform. A single 10 J m−2 blue light pulse (in the range 360 to 520 nm) was sufficient to elicit an endogenous zonation rhythm (coremia formation); the higher the fluence, the longer the rhythm expression. The period was 30 to 36 h as long as sufficient fluence rate (3 μW m−2 for broad band blue light and 18.1 nmol m−2 s−1 for wavelengths 433, 457 or 465 nm) was continuously maintained; this rhythm became desynchronized in about a week. The period increased rapidly and reached 72 to 120 h as soon as the fluence rate was too low or dark was established. The 445,479 and 495 nm radiations evoked the rhythm in pulse experiments, whereas the rhythm was immediately desynchronized in continuous light. The participation of two photosensitive reactions in the rhythm regulation of P. claviforme is postulated.  相似文献

Previous studies utilizing cotyledon explants of radiata pine ( Pinus radiata D. Don) revealed that cytokinin was required for shoot formation. This was confirmed and extended in the present study, which also showed that light was required. As little as three days of exposure to 16 h photoperiod light at a photon fluence rate of ca 80 μmol m−2 s−1 was sufficient to give some meristematic tissue formation. Longer exposure to light increased this formation. While cytokinin must be present during the first three days in culture for shoot formation, the exposure of the cotyledons to light could be delayed at least until day 10, but after 21 days in darkness, transfer to light did not permit shoot formation. Anatomical examination of the cotyledons confirmed the morphogenetic interactions of light and cytokinin in shoot formation. The data suggest that cytokinin is directly involved in the induction of shoot initiation and both light and cytokinin are required for the development of meristematic tissue and subsequent shoot formation.  相似文献

Maize ( Zea mays L. hybrid ZP-704) and black pine ( Pinus nigra Arn.) were grown for five days at low fluence rate (0.4–4.0, μmol m–2 s−1) in blue or red light. Compared to red light of the same fluence rate, blue light effects in maize were repressive for the accumulation of Chita, b , carotenoids and light-harvesting complex-2 (LHC-2) proteins. The maximal reduction of proteins bound to the light-harvesting complex of photosystem 2 and pigments was attained at different fluence rate levels. In black pine, blue light compared to the red of the same fluence rate level either activated or reduced accumulation of pigments and LHC proteins, the effect being dependent on its fluence rate level. At fluence less than 3.0 μmol m−2 s−1 blue light was more efficient for the synthesis of Chi a, b and carotenoids, hut for LHC-2 complexes, fluence rates between 0.4 and 1.5 [μmol m−2 s−1 were more effective. In pine the effects of the two lights on the accumulation of pigments and LHC proteins were demonstrated separately and were dependent on fluence rate level. This suggests irradianoe-controlled activation/deactivation of the photoreceptor at the level of the cell.  相似文献

Light effects on in vitro adventitious root formation in axillary shoots of a 95-year-old black cherry ( Prunus serotina Ehrh.) were examined using microcuttings derived from cultured vegetative buds. Three studies were performed: 1) complete darkness and 4 levels of continuous white light irradiance were tested at 70, 278, 555 and 833 μmol m−2 s−1; 2) white, red, yellow and blue light were tested to assess the importance of spectral quality; and 3) the effect of blue light at intensities of 7,15, 22 and 30 μmol m−2 s−1 was also studied, Measurements included rooting percentage, total number of roots per shoot, and shoot and root dry weight. There was a strong negative effect of white light intensity upon root formation. Blue light between 15 and 22 μmol m−2: s−1 significantly retarded root formation and completely inhibited it at 36 μmol m−2 s−1. Shoots treated with yellow light exhibited the highest rooting percentage, mean number of roots per shoot, and root dry weight.  相似文献

The dependence of the Chl content and the rate of CO2 gas exchange (RGE) on both blue and red quanta fluence rates have been studied in primary leaves of barley ( Hordeum vulgare L. cv. Viner). Empirical equations connecting the two photosynthetic indices with fluence rates of blue or red light were developed. These equations consist of 3 (Chl content) or 2 (RGE) terms, each reflecting the involvement of a specific reaction in the long-term light control of the development of the photosynthetic apparatus. On the basis of the equations the effects of mixed blue plus red light on both the Chl content and RGE were calculated. An additive mode of the co-action of blue and red light in the range of high PFDs (10–170 μmol m−2 s−1) becomes evident from the comparison of the experimental results and calculated data. The results indicate the involvement of phytochrome, cryptochrome and chlorophyll in the long-term regulation of the Chl content and RGE.  相似文献

The circadian rhythm in growth of the red macroalga Porphyra umbilicalis (Linnaeus) J. Agardh was investigated under different spectral light conditions in laboratory-grown thalli. A free-running rhythm was observed in constant green or red light at irradiances of 2.5 to 20 μmol photons·m−2·s−1, whereas arhythmicity occurred in constant blue light at 6–20 μmol photons·m−2·s−1. The circadian oscillator controlling growth rhythmicity in Porphyra uses most of the visible sunlight spectrum and possibly multiple photoreceptors with a high sensitivity for blue light and a lower sensitivity for red light. This was inferred from three experimental results: (1) The free-running period, τ, of the growth rhythm decreased with increasing irradiance, from approximately 25 h at 2.5 μmol photons·m−2·s−1 to 22 h at 20 μmol photons·m−2·s−1 in red or green light, (2) Dark pulses of 3 h duration, interrupting otherwise continuous green or red light, caused advances during the subjective day and delays during the subjective night; the circadian oscillator in Porphyra can discriminate darkness from green or red light, and (3) Low-irradiance blue light pulses (2.5 μmol photons·m−2·s−1) shifted the growth rhythm in red light of higher irradiance (e.g. 10 μmol photons·m−2·s−1), and a strong, high amplitude, type 0 phase response curve was obtained that is usually observed with light pulses shifting a circadian rhythm in otherwise continuous darkness.  相似文献

Gyrodinium dorsum Kofoid responds photophobically to flashes of blue light. The photophobic response consists of a cessation of movement (stop-response). Without background light and after a flash fluence above 10 J m−2, 75–85% of the cells show a stop-response, while only 50% of the cells show this response at 5 J m−2. With a flash fluence of 5 J m−2, background light of different wavelengths either increases (614 nm. 5.5–18.2 μmol m−2 s−1) or decreases (700 nm, 18.4–36.0 μmol m−2 s−1) the stop-response. Two hypotheses for the mechanism of the modulation by background light of the photophobic response are discussed: an effect of light on the balance of the photosynthetic system (PS I/PS II) or an effect on a phytochrome-like pigment (Pr/Pfr). This study supports the idea that a phytochrome-like pigment works in combination with a blue light-absorbing pigment. It was also found that cells of Gyrodinium dorsum cultured in red light (39.8 μmol m−2) had a higher absorption in the red region of the absorption spectra than those cultured in white light (92.7 μmol m−2).  相似文献

The effects of UV-C (254 nm), UV-A (365 nm) and broad-band UV (280–380 nm) on guard cells of Vicia faba L. cv. Long Pod were investigated in the presence of white light (450 μmol m−2 s−1). UV-C (7 μmol m−2 s−1) was found to cause leakage of 86Rb+ from guard cells, while UV-A (0.3 μmol m−2 s−1) stimulated increased uptake in these cells. A relatively small stimulatory effect was observed by broad-band UV (3 μmol m−2 s−1) during the first 30 min of irradiation with an apparent equilibration of influx and efflux thereafter. Leakage of 86Rb+ from guard cells continued despite the removal of UV-C and an increase in the amount of white light from 450 to 1500 μmol m−2 s−1, suggesting that membranes were irreversibly damaged. Irradiation of guard cells with UV-C for 30, 45 and 90 min indicated that these cells began to be affected already by 30 min UV-C irradiation.  相似文献

In crude extracts of the unicellular green alga Chlorella kessleri Fott et Novákóva grown in red light the activity of the glycolytic enzyme phosphofructokinase (PFK, EC 2.7.1.11) is about 40% higher compared to white light conditions giving the same dry matter production. Application of cycloheximide and density labelling with D2O indicate that this increase depends on the de novo synthesis of the enzyme: Twelve h of illumination at a fluence rate of 7 × 1018 quanta m−2 s−1 (11.6 μmol m−2 s−1) suffice to saturate the effect. In autotrophically grown algae maximal increase in enzyme saturate the effect. In autotrophically grown algae maximal increase in enzyme activity is reached in light of 680 nm, while in 3-(3,4-dichlorophenyl)-1,1-dimethyl urea (DCMU)-poisoned, glucose-fed cells, light of wavelengths around 727 nm is most effective. Involvement of a phytochrome-like photoreceptor is discussed.  相似文献

Rooting ability was studied for cuttings derived from pea plants ( Pisum sativum , L. cv. Alaska) grown in controlled environment rooms. When the cuttings were rooted at 70 μmol m−2 s, 1 (photosynthetic photon flux density) or more, a stock plant irradiance at 100 μmol m−2 s−1 decreased rooting ability in cuttings compared to 5 μmol m−2, s−1, However, cuttings rooted at 160 μmol m−2 s−1 formed more roots compared to 5 (μmol m−2 s−1. Although a high irradiance increased the number of roots formed, it could not overcome a decreased potential for root formation in stock plants grown at high irradiance. Light compensation point and dark respiration of cuttings decreased by 70% during the rooting period, and the final levels were strongly influenced by the irradiance to the cuttings. Respiratory O2 uptake decreased in the apex and the base of the cutting from day 2 onwards, whereas a constant level was found in the leaves. Only the content of extractable fructose, glucose, sucrose and starch varied during the early part of the rooting period. We conclude that the observed changes in the cuttings are initiated by excision of the root system, and are not involved in the initiation of adventitious roots.  相似文献

The significance of photosynthetic photon flux (PPF) and sugar feeding for the production of plants in vitro is only poorly understood. Nicotiana tabacum L. plantlets were grown photoautotrophically and photomixotrophically (3% sucrose) at two different PPFs (60 µmol m−2 s−1 and 200 µmol m−2 s−1) to investigate the effect of these culture parameters on photosynthetic performance and growth. Photomixotrophically‐grown plantlets showed an increase in carbohydrate content, mainly in glucose and fructose. Plant growth, dry matter accumulation and total leaf area were higher under photomixotrophic than photoautotrophic conditions. Not only biomass formation but also photosynthesis was positively affected by exogenous sucrose; the chlorophyll (Chl) content and the light‐saturated rate of photosynthetic oxygen evolution were higher in photomixotrophic plantlets. Photoinhibition occurred in plantlets that were grown photoautotrophically at the higher PPF. It became apparent as a loss in Chl content and photochemical efficiency. Photoinhibited plantlets showed a decrease in the D2/LHCII and CP47/LHCII ratios, suggesting a preferential loss of proteins from the photosystem II (PSII) core. The increased content of xanthophyll cycle pigments in photoinhibited plantlets indicated that also protective mechanisms were activated. Photomixotrophic growth of the plantlets prevented the occurrence of photoinhibitory symptoms. Therefore, we conclude that culture on sugar medium increases not only the photosynthetic potential but also the high light resistance of plantlets grown in vitro.  相似文献

We conducted a series of experiments to assess the effects of oxidative stress on chlorophyll biosynthesis in the vascular plant Cucumis sativus (cucumber). Specifically, cucumber cotyledons were treated with 100 μ M methyl viologen (MV) and subsequently exposed to dark (0 μE m−2 s−1), low light (40–45 μE m−2 s−1), or high light (1500–1600 μE m−2 s−1). Following treatment, extracts of these samples were subjected to high-performance liquid chromatography (HPLC) to quantitate the accumulation of chlorophyll biosynthetic pathway intermediates. The results of these analyses revealed significant accumulation of Mg-protoporphyrin IX monomethyl ester (Mg-proto IX ME) in green (14-h illuminated) as well as in etiolated cotyledons with MV treatment. These data suggest that MV-induced oxidative stress may have inhibited Mg-proto IX ME cyclase activity. Upon exposure to high light, in the presence or absence of MV, both green and etiolated cotyledons predominantly accumulated protoporphyrin IX (Proto IX). These elevated levels of Proto IX might be attributable to attenuated activity of any or all of the following enzymes: Mg-chelatase, Fe-chelatase and protoporphyrinogen IX oxidase. We also observed that MV-induced oxidative stress impacts on chlorophyll biosynthesis to a greater extent than on photosystem II. These results demonstrate that oxidative stress impedes key steps in chlorophyll biosynthesis by either directly or indirectly inhibiting the activity of these enzymes.  相似文献

A high-altitude ecotype of tomato ( Lycopersicon hirsutum f. typicum Humb. and Bonpl.) has previously been shown to resist further loss of photosynthetic function after three to four days of chilling stress. This study examined the influence of PPFD prior to, and during chilling on the development of protective zeaxanthin and energy-dependent quenching mechanisms in this ecotype. Five-week-old tomato plants were acclimated to either low PPFD (60 μmol m−2 s−1) or high PPFD (550 μmol m−2 S−1) at 25/20°C (day/night) for three days, and then exposed to a temperature of 5/5°C and a PPFD of either 60 or 550 μmol m−2 s−1 for three days. The plants acclimated to low PPFD had lower Chl a/b ratio, and lower level of total Chl per leaf area, total xanthophyll cycle pool and β-carotene. The capacity of their photosynthetic system to resist photoinhibition and to recover photosynthetic function was also lower compared to that of the plants acclimated at high PPFD but exposed to the same chilling stress. In the plants chilled at low PPFD, energy-dependent quenching preceded the formation of zeaxanthin on the first day of chilling and there was an overall reduction in the conversion of violaxanthin to zeaxanthin as compared to the plants chilled at high PPFD. During the last day of chilling-induced photoinhibition, energy-dependent quenching in any of the treatments did not increase, but zeaxanthin levels increased continuously throughout the three days of chilling. Our results suggest that light-acclimation before chilling affects the capacity of the plants to resist chilling-induced photoinhibition. In addition, photoinhibitory quenching appears to be a major component for quenching excessive energy at the latter stage of long-term chilling.  相似文献

Abstract: Very large numbers (3466 ml−1) of ciliated protozoa were found living beneath the oxic-anoxic boundary in a stratified freshwater pond. Most ciliates (96%) contained symbiotic algae ( Chlorella spp.). Peak abundance was in anoxic water with almost 1 mol free CO2 m−3 and a midday irradiance of 6 μmol photon m−2 s−1. Photosynthetic rate measurements of metalimnetic water indicated a light compensation point of 1.7 μmol photon m−2 s−1 which represents 0.6% of sub-surface light. We calculate that photosynthetic evolution of O2 by symbionts is sufficient to meet the demand of the host ciliates for 13 to 14 hours each day. Each 'photosynthetic ciliate' may therefore become an aerobic island surrounded by anoxic water.  相似文献

Corn ( Zea mays L. cv. OP Golden Bantum) was grown under low irradiance blue, red or blue plus red light. Red was more effective than blue light for synthesis of Chl a, b and light-harvesting proteins (LHC-2) associated with photosystem 2(PS2). Blue light was slightly more effective for synthesis of light-harvesting proteins (LHC-1) associated with photosystem 1 (PS1), but below a fluence rate of 1 μmol m−2 s−1 the response to blue vs that to red depended on irradiance level. Blue light containing a small amount of red light was as effective as red light for Chl a and b synthesis, but no more effective than blue light for LHC-2 synthesis. Adding small amounts of blue light to red repressed the effect of red light on LHC-2 synthesis and produced irradiance response curves similar to those produced by blue alone for LHC-2 synthesis. This repression by blue light depended on the ratio of red to blue and the level of the blue light.  相似文献

  An experiment was conducted on intact algal assemblages of stream periphyton to test their response to fluctuating and constant light regimes having the same mean intensity. The light regimes (in μmol·m−2·s−1) were constant light at 100, light fluctuating between 50 and 150 with a period of 5 min, and light fluctuating between 10 and 460 with periods of either 4:1 or 8:2 min. Compared to the rates measured under 100 in μmol·m−2·s−1 constant light conditions, fluctuations ranging between 50 and 150 in μmol·m−2·s−1 with a 5-min period produced a 23% greater rate of photosynthesis. Conversely, fluctuations between 10 and 460 in μmol·m−2·s−1 led to a 59%–74% decrease in photosynthetic activity. Detailed examination of periphytic algal responses to fluctuating light revealed that higher light intensities produced steeper photosynthesis/time slopes, but it was the combined interaction with lower light intensity that ultimately determined overall photosynthetic rate for a given light regime. This study offers compelling evidence that variable light regimes have important consequences for algal photosynthesis in natural streams.  相似文献

A study of the kinetics of chlorophyll (Chl) synthesis in cotyledons of etiolated cucumber seedlings ( Cucumis sativus L . cv. Delilah) treated with 5×10-5 M -ben-zyladenine (BA) showed that cytokinin, like a red light pulse, could inhibit as well as promote pigment accumulation depending on the length of the dark period following induction. Spraying intact, dark-grown seedlings with BA, 24 h prior to white light exposure, eliminated the lag phase in Chl synthesis, while treatment with hormone 72 h before greening not only delayed the onset of synthesis, but it also reduced the amount of Chl accumulated after 24 h continuous white light. Impairment of Chl formation was correlated with inhibited regeneration of protochlorophyll and delayed appearance of the light harvesting Chl alb polypeptide. Application of σ-aminolevulinic acid (15 m M ) 2 h before white light exposure shortened the lag phase in Chl synthesis in control as well as in inhibited cotyledons, but the adverse effect of the red light and BA treatments on long-term Chl accumulation (24 h) was not reversed. Application of glutamate did not stimulate Chl production. Simultaneous treatment with hormone and red light 72 h before greening enhanced their separate inhibitory effects on Chl synthesis, but when given together 24 h prior to white light, their promotive effects on pigment accumulation were not additive.  相似文献

The effects of light on in vitro proliferation and subsequent in vivo rooting and acclimatisation of Vaccinium corymbosum were investigated. The shoots were exposed in vitro to different irradiances (total radiation ranging from 55 to 240 μmol m−2 s−1) for 7 to 60 days. In vitro growth and proliferation and the possible consequences on in vivo rooting were observed.
As compared to the control treatment (55 μmol m−2 s−1), higher irradiances improved proliferation and rooting ratios only with short applications (7 days). Short but high (210 μmol m−2 s−1) exposures applied at the end of the proliferation phase increased in vivo growth and rooting of the shoots. The shoots treated with strong light for longer times (14 and 28 days) showed both inhibition of growth and red colour of leaves and sprouts, and were less vigorous when transferred in vivo.  相似文献
