The genetic and hormonal control of body colouration is investigated using two recessive genetic mutant strains, the reddish–brown (RB) mutant and an albino mutant, as well as a normal (pigmented) strain of the desert locust Schistocerca gregaria. The colour patterns of the RB nymphs are similar to those of a normal strain, although the intensity of the melanization is weaker in the former. Reciprocal crosses between the RB and albino mutants produce only normal phenotypes in the F1 generation. In the F2 generation, the normal, RB and albino phenotypes appear in a ratio of 9 : 3 : 4, indicating that two Mendelian units might determine the appearance of dark body colour and the intensity of melanization, respectively. In other words, at least two steps of regulation might be involved in the expression of body colour. Injections of [His7]‐corazonin, a neuropeptide inducing dark colour in this locust, fail to induce dark colour in albino nymphs but show a dose‐dependent darkening in RB nymphs in the range, 10 pmol to 1 nmol. Some RB nymphs become indistinguishable from normal individuals after injection of the peptide. Implantation of corpora cardiaca (CC) taken from RB mutants into other RB individuals induces darkening in the latter and CC from RB, albino and normal strains have similar dark colour‐inducing activity when implanted into albino Locusta migratoria. These results suggest the possibility that the RB mutant gene regulates the intensity of melanization, possibly through controlling the pathway of pigment biosynthesis associated with [His7]‐corazonin.  相似文献

The albino Okinawa strain of Locusta migratoria is deficient in the neurohormone [His7]-corazonin. This peptide induces darkening of the cuticle, one of the typical features of gregarious locusts. As part of a broader study on the possible role of [His7]-corazonin in phase transition, we explored whether corazonin-deficiency might be associated with differences in behavior and morphometrics between albino and normal phenotypes of L. migratoria. Using a modification of the logistic-regression assay of behavioral phase state previously derived for Schistocerca gregaria, we found that there were strain dependent behavioral differences between crowd-reared nymphs of the albino Okinawa and the normally colored African strain, with no evidence of the albino strain being obligatorily solitarious. However, upon isolation, a shift towards more solitarious behavior occurs in both strains, even more profoundly in the Okinawa albinos. A shift could also be recorded in morphometrics. The conclusion is that the albino strain, although showing some solitarious features even when crowd-reared, is not, as has been suggested, obligatory solitarious and, as a consequence, the complete absence of corazonin is not sufficient to bring about the solitarious state.  相似文献

Abstract.  An albino strain that had originated from Okinawa, Japan, and a normally coloured strain that had originated from West Africa, were used to study the darkening response to injection of graded doses of dark-colour-inducing neurohormone of locusts (DCIN) ([His7]-corazonin) of gregarious and solitarious adults of the migratory locust, Locusta migratoria (L.). By repeated crossings, congenic albinos and normal phenotypes were obtained, both with a 99.6% West African genome, and their darkening response was compared with the original Okinawa and West African strains. Within each of these four strains, no difference was found in DCIN-induced darkening between gregarious and solitarious adults despite previous publications in the literature claiming an absence of 'fire-darkening' in gregarious adults of this species. Okinawa albino adults showed a markedly higher darkening response than the other three strains, including albinos with a 99.6% West African genome. This finding demonstrates that the differential darkening response of the Okinawa albinos is caused not by albinism, but by the geographical origin (Okinawa) of the strain. This is the first report of geographical-strain-dependent differences in the response of an insect to a neurohormone. The darkening response of adults reached a maximum on day 10 after injection; subsequently, the dark colour faded slowly. Adults injected 1 day after their moult showed a greater darkening response than those injected after 14 or 28 days.  相似文献

Paranosema (Antonospora) locustae (Canning) is a microsporidium with an extensive host range including >100 reported insect hosts from the order Orthoptera. The susceptibility of two species of locusts (Orthoptera: Acrididae) – the migratory locust, Locusta migratoria subsp. migratorioides (Fairmaire & Reiche), and the desert locust, Schistocerca gregaria Forsskål – to P. locustae was compared under laboratory conditions at a decreased temperature of 27 °C to enhance susceptibility to infection. Locusta migratoria was found highly vulnerable as infection percentages exceeded 70% at 104, 105, and 106 spores per insect, and mortality increased with increasing dosage. Conversely, P. locustae spores were not found in S. gregaria throughout the experiment. Only at 107 spores per insect, as many as 20% of S. gregaria were infected. This suggests that the desert locust is resistant to P. locustae infection, as opposed to the migratory locust. The laboratory models of these parasite–host systems may be useful for elucidating mechanisms of insect immunity to microsporidia.  相似文献

The dark-colour-inducing effect of several peptides in comparison to that of the dark-colour-inducing neurohormone (DCIN, [His(7)]-corazonin) of locusts was investigated by a bioassay based on nymphs of a DCIN-deficient albino mutant of Locusta migratoria. The study was aimed at elucidating the active part of the DCIN and to explore the contribution of its amino acids to the activity. Graded doses of all peptides were injected in oil. [Arg(7)]-corazonin and DCIN were equally effective. Certain arthropod neuropeptides having the -SXGW- partial sequence (a part of the DCIN and of [Arg(7)]-corazonin; X=His and X=Arg, respectively) yielded the following findings: Scg-AKH-II (adipokinetic hormone II of Schistocerca gregaria X=Thr), Grb-AKH ( adipokinetic hormone of Gryllus bimaculatus X=Thr) and RPCH (red pigment concentrating hormone of crustaceans X=Pro) evoked a moderate darkening response, but Lom-AKH-II (adipokinetic hormone II of L. migratoria X=Ala) was ineffective. Step by step shortening of the sequence of the DCIN at the N-terminal, from pGlu-3-11DCIN to pGlu-9-11DCIN, resulted in a decreasing activity, but even pGlu-9-11DCIN induced a weak response with high doses. Shortening of the DCIN from the C-terminal revealed a moderate activity of 1-7DCIN-NH(2) and a weak activity of 1-5DCIN-NH(2). An octadecapeptide which induces dark colour in moth larvae, having the pentamer FTPRL-NH(2) at its C-terminal, evoked no darkening in the albino locusts. We conclude that although the -SXGW- partial sequence has some role in induction of darkening, for obtaining maximal effect the whole sequence of the DCIN (or of [Arg(7)]-corazonin) is necessary.  相似文献

The two plague locusts, Schistocerca gregaria and Locusta migratoria, exhibit density-dependent phase polyphenism. Nymphs occurring at low population densities (solitarious forms) are uniformly colored and match their body color to the background color of their habitat, whereas those occurring at high population densities (gregarious) develop black patterns. An injection of the neuropeptide, corazonin (Crz) has been shown to induce black patterns in locusts and affect the classical morphometric ratio, F/C (F, hind femur length; C, maximum head width). We herein identified and cloned the CRZ genes from S. gregaria (SgCRZ) and L. migratoria. A comparative analysis of prepro-Crz sequences among insects showed that the functional peptide was well conserved; its conservation was limited to the peptide region. Silencing of the identified SgCRZ gene in gregarious S. gregaria nymphs markedly lightened their body color and shifted the adult F/C ratio toward the value typical of solitarious forms. In addition, knockdown of the gene in solitarious nymphs strongly inhibited darkening even after a transfer to crowded conditions; however, these individuals developed black patterns after being injected with the Crz as a rescue treatment. SgCRZ was constitutively expressed in the brains of S. gregaria during nymphal development in both phases. This gene was highly expressed not only in the brain in both phases, but also in the corpora allata in the gregarious phase. This conspicuous phase-dependent difference in SgCRZ gene expression may indicate a functional role in the control of phase polyphenism in this locust.  相似文献

Behavioural responses of nymphs and adults in the gregarious phase of the desert locust,Schistocerca gregaria (Forskal) (Orthoptera: Acrididae) were investigated in a single-chamber bioassay system to a choice of two columns of air, one permeated with airborne volatiles emanating from either sex of nymphs or adults and the other untreated. There was no sexual differentiation in the production of or response to nymphal volatiles. Young adults of either sex did not produce a stimulus with significant activity. Of the older adults, only the males produced the aggregation stimulus to which both sexes were equally responsive. Charcoal-trapped volatiles from the two sexes of nymphs and adults evoked similar aggregation responses. Antennae of the older adults showed significantly higher EAG responses than those of fifth instar nymphs to all four volatile collections, of which volatiles from older adult males were the most stimulatory and evoked the highest EAG amplitudes.  相似文献

Abstract The effect of the dark-colour-inducing neurohormone (DCIN = [His7]-corazonin) of locusts was investigated in field-collected young and old adults of the grasshopper, Oedipoda miniata (Pallas) (Orthoptera: Acrididae). This species shows homochromy, but neither green-brown, nor phase-dependent colour polymorphism. By injecting graded doses of synthetic DCIN in 2 µL of olive oil, young adults were tested within a week of their last moult, and old adults 3.5 months later, a few weeks before natural termination of their reproductive summer diapause. Colour changes were followed for 28 days after injection. Darkening of the young adults was considerable, but their response to DCIN is more moderate than that of conspecific nymphs, by exhibiting a higher threshold, slower response and weaker maximal response. Old adults also show a clear effect, but their response is even slower and less marked than the response of the young adults. It is concluded that the response to DCIN decreases from nymphs to young adults and it is further decreasing with ageing of the adults.  相似文献

In the 11-residue long dark-color-inducing neurohormone (DCIN = [His7]-corazonin), of locusts, from residue 2 to residue 11, one amino acid at each time was substituted by d-phenylalanine (d-Phe). The dark-color-inducing effect of these peptides was investigated in comparison with unaltered DCIN by a bioassay based on nymphs of a DCIN-deficient albino mutant of the migratory locust, Locusta migratoria. Substitution of any single amino acid by d-Phe always reduced the activity, but did not abolish it completely. Maximum inactivation was obtained after substitution of Gln4, Ser6, or Trp9. The latter two residues are within the partial sequence -Ser-Xxx-Gly-Trp- (Xxx = His in the DCIN) that seems to be important for the dark-color-inducing activity, as found also in another study (Insect Biochem. Mol. Biol. 32, 2002, 909). Gln4, however, is outside of this partial sequence. Minimal, although still considerable, inactivation occurred after substitution of Gly8, Phe3, or Asn11, despite the fact that Gly8 is within the -Ser-Xxx-Gly-Trp- partial sequence. In conclusion, no single active core was found, indicating that the whole sequence of the DCIN is necessary to induce maximum darkening effect. No difference was found in the activity of the peptides in which Gly8 was substituted by d-Phe or by l-Phe. Therefore the -Ser-Xxx-Gly-Trp- partial sequence does not seem to be stabilized by a type II beta-turn. Nevertheless, existence of another kind of turn that includes this partial sequence is feasible. A single unsuccessful attempt was made to discover an antagonist to the DCIN.  相似文献

Previously, we reported a chain of effects induced by phenylacetonitrile, the gregarious‐phase adult cohesion pheromone of the desert locust, Schistocerca gregaria (Forskål) (Orthoptera: Acrididae), on conspecific hoppers. Specifically, the nymphs became hyperactive and displayed high levels of intraspecific predation, as surviving individuals gradually shifted to the solitary phase. The findings suggested that the pheromone could induce a significant level of mortality of the nymphs and predispose them to greater sensitivity to lower doses of insecticides. In this study, we compared the effects of the pheromone and various doses of three pesticides (fipronil, malathion, and carbosulfan) to pheromone‐exposed and unexposed crowd‐reared hoppers in semi‐field enclosures. The pheromone on its own displayed a high level of cumulative mortality of the nymphs (89%). Although combinations with fractional pesticide doses gave control efficiencies that were, in most cases, higher than with individual pesticides, pheromonal and pesticidal effects were only partially complementary, probably because of the feeding‐deterrent effects of the pesticides and the resulting reduction in cannibalism. However, our results demonstrate the possibility of significantly reducing the levels of pesticides used in hopper control and, thus, their negative environmental effects.  相似文献

A comparison was made of the relationship between deterrence and toxicity of eight secondary compounds to two acridids. The grass specialist Locusta migratoria was compared with the polyphagous Schistocerca gregaria. L. migratoria was deterred by all compounds presented at natural concentrations to which S. gregaria showed a variety of behavioral responses. Overall, L. migratoria was significantly more susceptible to toxic effects of the chemicals when injected into the hemolymph than was S. gregaria and was more sensitive behaviourally to them. There was a significant correlation in both species between deterrency and toxicity of injected compounds, but little evidence of a relationship between deterrency and oral toxicity.
Résumé La comparaison a porté sur les relations entre répulsion et toxicité de huit substances secondaires chez deux acridiens: Locusta migratoria, spécialiste de graminées, et Schistocerca gregaria, polyphage. L. migratoria est repoussé par toutes les substances proposées, aux concentrations naturelles, pour lesquelles S. gregaria a présenté des comportements trés divers. Mais surtout, L. migratoria a été significativement plus sensible que S. gregaria aux effets toxiques de ces substances quand elles sont injectées dans son haemolymphe; S. gregaria a réagi à ces substances plus par des modifications de son comportement. Il y a une corrélation significative pour les deux espèces, entre la répulsion et la toxicité des substances injectées.

The effect of the dark-colour-inducing neurohormone (DCIN=[His(7)]-corazonin) of locusts was investigated in nymphs of the grasshopper, Oedipoda miniata, which exhibit strong homochromy, but neither green-brown, nor phase colour polymorphism. Graded doses of synthetic DCIN were injected in 1 μl of olive oil into 0-24-h-old penultimate nymphs. DCIN induced dose-dependent darkening in the recipients, seen 4 days after injection (still in the penultimate nymphal instar), as well as in the subsequent last-instar nymphs. The dose-range obtained between discernible and maximum (almost black) darkening extended over three orders of magnitude, from 1 pmol to 1 nmol of DCIN. In spite of the darkening observed already in the penultimate nymphal instar, the exuviae in the moult from this to the last nymphal instar did not show marked dark patches. However, the exuviae of the next moult (from last-instar nymph to adult) showed such dark patches, which increased with the increase of the dose, revealing the presence of exocuticular melanin. We conclude that DCIN, or a very close peptide, is the endogenous hormone which plays the major or sole role in the control of homochromy of O. miniata and possibly also of other acridids which exhibit homochromy.  相似文献

The dark-color-inducing neurohormone (DCIN) of locusts and corazonin of a cockroach, both 11 residue-long peptides, induce dark coloration in albino nymphs of Locusta migratoria when injected after a nymphal molt. These peptides differ at position 7 (His in DCIN and Arg in corazonin) and elicit an almost identical darkening response. The three-dimensional structures of these peptides, dissolved in dimethylsulfoxide (DMSO), were determined by NMR. Structural elements determined at atomic resolution may provide insight into the biological activity of these two neurohormones. The calculated structures of DCIN and corazonin indicate clear, prevalent conformations with similar secondary features. The generated low-energy solution structures of each show structural elements within residues Phe3 to Trp9 with a turn situated at the core of the peptide from which the sidechains of residue 7 of each peptide protrude. A calculated negative electrostatic potential surface almost completely covers both neurohormones and only the 7th residue sidechains of each peptide emerge in their entirety. Within these residues there is a partial sequence seen in several neurohormones that control various physiological functions in Arthropods: -Ser-X-Gly-Trp- (X=His in DCIN and Arg in corazonin). This partial sequence may play a role in the physiological activity of some Arthropod neurohormones.  相似文献

Aiolopus thalassinus thalassinus (Fabricius) (Orthoptera: Acrididae) is a non-migratory grasshopper of widespread geographical distribution, also endemic in the Tel-Arad region of the Northern Negev of Israel, where it is liable to sporadically damage agricultural crops. Periodic sampling in uncultivated ‘batha’ and agricultural fields, conducted during 1990/1991, indicate thatA. thalassinus populations exhibit seasonal fluctuations in density. Local spatial and temporal distribution, within this region, are dependent on food availability. Field observations and laboratory studies suggest that the local population ofA. thalassinus exhibits genetic heterochromy unaffected by density. The duration of nymphal developmental is 34 days for nymphs reared in isolation, whereas crowded nymphs complete their development within only 21 days. No striking density related changes in gross morphometric features are evident, but, in adults from isolated culture, an increased abundance ofsensilla coeloconica, presumably involved in olfactory chemoreception, may be functionally related to enhanced (23.6-fold higher) activity of nymphs reared under crowded conditions. This higher level of activity is correlated to higher levels of energy reserves in the haemolymph — lipids and carbohydrates — and to increased respiration. Finally, the individual food consumption of nymphs from crowded culture is almost 5-fold higher than that of isolated nymphs and may increase the potential for crop damage. These results are similar to those obtained with the migratory locustLocusta migratoria and support the hypothesis that non-migratory grasshoppers exhibit some density-dependent physiological characteristics of locusts, but do not exhibit overt chromatic or morphometric phase characteristics.  相似文献

Details are given of techniques for preparing surface spreads of locust spermatocytes for light and electron microscopy. The pachytene synaptonemal complex (SC) karyotypes of Locusta migratoria and Schistocerca gregaria are analysed and compared. Up to six different SCs can be identified in Locusta migratoria based on lengths, centromere positions, and possession of nucleolar organiser regions, but only two SCs are identifiable in Schistocerca gregaria. The total SC length is significantly greater in Schistocerca gregaria than in Locusta migratoria, and this difference is almost exactly proportional to the difference in the genomic DNA contents of the two species.  相似文献

Here, we describe a single micro‐CT scan with a spatial resolution of 10 μm of a 10‐day‐old adult male Schistocerca gregaria (Forskål) (Orthoptera: Acrididae) and we compare our tracheal volume (VT) determination with published work on the subject. We also illustrate the feasibility of performing non‐invasive ‘virtual dissection’ on insects after performing micro‐CT. These post‐processing steps can be performed using free downloadable 3‐D software. Finally, the values of producing stereo‐lithography (STL) files that can be viewed or used to print out 3‐D models as teaching aids are discussed.  相似文献

Three new species of larval Charletonia Oudemans, 1910 from Rhodes, Greece are described: C. dalegori from an undetermined orthopteran, C. glifadaensis from Oedipoda sp. (Orthoptera: Acrididae: Oedipodinae) and C. kaliksti from Aiolopus sp. (Orthoptera: Acrididae: Oedipodinae). C. krendowskyi is recorded for the first time from Greece. The glucosinolates of a Saharan crucifer Schouwia purpurea (Forskål) (Brassicaceae) were determined by liquid chromatography. Two of these glucosinolates and sinigrin were tested for their deterrent effect on Schistocerca gregaria (Forskål) (Orthoptera: Acrididae). Glucobrassicin, three indolyls and epigoitrin were synthesized for this purpose. Epiprogoitrin was extracted from Crambe seeds. Choice tests on artificial substrate compared feeding responses to glucosinolates and to related breakdown products released when the plant is eaten. Breakdown products were more efficient in deterring the generalist locust than were glucosinolates. Two patterns of dose responses were recorded: glucosidic compounds deterred or stimulated feeding, depending on the concentration tested; aglycones did not stimulate feeding at any concentration. Allyl isothiocyanate, a volatile compound, was a 100-fold higher deterrent than its substrate (sinigrin).  相似文献

Black‐eyed Locusta migratoria appeared in albino locusts as a result of crossing between a short‐winged strain originating from Tsushima Island, Japan, and an albino strain originating from Okinawa Island. The black eye trait was recessive to the white eye trait because the crosses between black‐ and white‐eyed albino locusts produced only individuals with white eyes in the F1 generation. In the F2 generation, black‐ and white‐eyed individuals appeared in a ratio of 1:3, indicating that the black eye trait was controlled by a simple Mendelian unit. The black eye trait showed no genetic association with other traits including wing morph, adult body dimensions and classical morphometric ratios such as hind femur length / head width and forewing length / hind femur length.  相似文献
