Aim

Our objectives were to compare effects of root charge properties on Al adsorption by the roots of rice that differed in Al-tolerance, and to examine effects of different nitrogen forms on charge properties of rice roots and Al adsorption.

Methods

Streaming potential and chemical methods were used to measure root zeta potential and investigate Al chemical forms adsorbed on the roots of rice obtained from solution culture experiments.

Results

Rice roots of the Al-sensitive variety Yangdao-6 carried greater negative charge than the Al-tolerant variety Wuyunjing-7, which meant the roots of Yangdao-6 adsorbed more exchangeable and complexed Al. When both rice varieties were grown in NH4 +-containing nutrient solutions, there were less functional groups and lower negative surface charge on their roots, which reduced Al adsorption compared to the rice grown in NO3 ? containing nutrient solutions. The decline in nutrient solution pH due to NH4 + uptake by rice roots was responsible for the reduced numbers of functional groups and the lower negative surface charge on the roots compared to the rice grown in NO3 ? containing solutions.

Conclusions

Integrated root surface charge, as expressed by zeta potential, played an important role in Al adsorption by the roots of rice with different Al-tolerance.

The phytotoxicity of aluminium (Al) ions can be alleviated by ammonium (NH4+) in rice and this effect has been attributed to the decreased Al accumulation in the roots. Here, the effects of different nitrogen forms on cell wall properties were compared in two rice cultivars differing in Al tolerance. An in vitro Al‐binding assay revealed that neither NH4+ nor NO3? altered the Al‐binding capacity of cell walls, which were extracted from plants not previously exposed to N sources. However, cell walls extracted from NH4+‐supplied roots displayed lower Al‐binding capacity than those from NO3?‐supplied roots when grown in non‐buffered solutions. Fourier‐transform infrared microspectroscopy analysis revealed that, compared with NO3?‐supplied roots, NH4+‐supplied roots possessed fewer Al‐binding groups (‐OH and COO‐) and lower contents of pectin and hemicellulose. However, when grown in pH‐buffered solutions, these differences in the cell wall properties were not observed. Further analysis showed that the Al‐binding capacity and properties of cell walls were also altered by pHs alone. Taken together, our results indicate that the NH4+‐reduced Al accumulation was attributed to the altered cell wall properties triggered by pH decrease due to NH4+ uptake rather than direct competition for the cell wall binding sites between Al3+ and NH4+.  相似文献

Aims

Rice is known as an ammonium (NH4 +)-tolerant species. Nevertheless, rice can suffer NH4 + toxicity, and excessive use of nitrogen (N) fertilizer has raised NH4 + in many paddy soils to levels that reduce vegetative biomass and yield. Examining whether thresholds of NH4 + toxicity in rice are related to nitrogen-use efficiency (NUE) is the aim of this study.

Methods

A high-NUE (Wuyunjing 23, W23) and a low-NUE (Guidan 4, GD) rice cultivar were cultivated hydroponically, and growth, root morphology, total N and NH4 + concentration, root oxygen consumption, and transmembrane NH4 + fluxes in the root meristem and elongation zones were determined.

Results

We show that W23 possesses greater capacity to resist NH4 + toxicity, while GD is more susceptible. We furthermore show that tissue NH4 + accumulation and futile NH4 + cycling across the root-cell plasma membrane, previously linked to inhibited plant development under elevated NH4 +, are more pronounced in GD. NH4 + efflux in the root elongation zone, measured by SIET, was nearly sevenfold greater in GD than in W23, and this was coupled to strongly stimulated root respiration. In both cultivars, root growth was affected more severely by high NH4 + than shoot growth. High NH4 + mainly inhibited the development of total root length and root area, while the formation of lateral roots was unaffected.

Conclusions

It is concluded that the larger degree of seedling growth inhibition in low- vs. high-NUE rice genotypes is associated with significantly enhanced NH4 + cycling and tissue accumulation in the elongation zone of the root.  相似文献

Poplar plants are cultivated as woody crops, which are often fertilized by addition of ammonium (NH4 +) and/or nitrate (NO3 ?) to improve yields. However, little is known about net NH4 +/NO3 ? fluxes and their relation with H+ fluxes in poplar roots. In this study, net NH4 +/NO3 ? fluxes in association with H+ fluxes were measured non-invasively using scanning ion-selective electrode technique in fine roots of Populus popularis. Spatial variability of NH4 + and NO3 ? fluxes was found along root tips of P. popularis. The maximal net uptake of NH4 + and NO3 ? occurred, respectively, at 10 and 15 mm from poplar root tips. Net NH4 + uptake was induced by ca. 48 % with provision of NO3 ? together, but net NO3 ? uptake was inhibited by ca. 39 % with the presence of NH4 + in poplar roots. Furthermore, inactivation of plasma membrane (PM) H+-ATPases by orthovanadate markedly inhibited net NH4 +/NO3 ? uptake and even led to net NH4 + release with NO3 ? co-provision. Linear correlations were observed between net NH4 +/NO3 ? and H+ fluxes in poplar roots except that no correlation was found between net NH4 + and H+ fluxes in roots exposed to NH4Cl and 0 mM vanadate. These results indicate that root tips play a key role in NH4 +/NO3 ? uptake and that net NH4 +/NO3 ? fluxes and the interaction of net fluxes of both ions are tightly associated with H+ fluxes in poplar roots.  相似文献

In the present study, we investigated whether growth and main nutrient ion concentrations of cabbage (Brassica campestris L.) could be increased when plants were subjected to different NH4^+/NO3- ratios. Cabbage seedlings were grown in a greenhouse in nutrient solutions with five NH4^+/NO3- ratios (1:0; 0.75:0.25; 0.5:0.5; 0.25:0.75; and 0:1). The results showed that cabbage growth was reduced by 87% when the proportion of NH4^+-N in the nutrient solution was more than 75% compared with a ratio NH4^+/NO3- of 0.5:0.5 35 d after transplanting, suggesting a possible toxicity due to the accumulation of a large amount of free ammonia in the leaves. When the NH4+/NO3- ratio was 0.5:0.5, fresh seedling weight, root length, and H2PO4- (P), K^+, Ca^2+, and Mg^2+ concentrations were all higher than those in plants grown under other NH4^+/NO3- ratios. The nitrate concentration in the leaves was the lowest in plants grown at 0.5: 0.5 NH4^+/NO3-. The present results indicate that an appropriate NH4^+/NO3- ratio improves the absorption of other nutrients and maintains a suitable proportion of N assimilation and storage that should benefit plant growth and the quality of cabbage as a vegetable.  相似文献

We examined the effect of soil NH4+ and NO3? content upon the root systems of field-grown tomatoes, and the influence of constant, low concentrations of NH4+ or NO3? upon root growth in solution culture. In two field experiments, few roots were present in soil zones with low extractable NH4+ or NO3?; they increased to a maximum in zones having 2μg-N NO3? g?1 soil and 6 μg-N NO3= g?1 soil, but decreased in zones having higher NH4+ or NO3? levels. Root branching was relatively insensitive to available mineral nitrogen. Plants maintained in solution culture at constant levels of NH4+ or NO3?, had similar shoot biomass, but all root parameters – biomass, length, branching and area – were greater under NH4 nutrition than under NO3?. These results suggest that the size of root system depends on a functional equilibrium between roots and shoots (Brouwer 1967) and on the balance between soil NH4+ and NO3?.  相似文献

To address the questions of whether allocation of carbohydrates to roots is influenced by ionic form of nitrogen absorbed and whether allocation of carbohydrates to roots in turn influences proportionality between NH4+ and NO3? uptake from mixed sources, NH4+ and NO3? were supplied separately to halves of a split-root hydroponic system and were supplied in combination to a whole-root system. Dry matter accumulation in the split-root system was 18% less in the NH4+-fed axis than in the NO3?-fed axis. This, however, does not indicate that partitioning of carbohydrate between the two axes was different. Most of the reduction in dry matter accumulation in the NH4+-fed axis can be accounted for by the retransport of CH2O equivalents from the root back to the shoot with amino acids produced by NH4+ assimilation. Uptake of NH4+ or NO3? by the respective halves of the split-root system was proportional to the estimated allocation of carbohydrate to that half. When NH4+ and NO3? were supplied to separate halves of the split-root system, the cumulative NH4+ to NO3? uptake ratio was 0.81. When supplied in combination to the whole-root system, the cumulative NH4+ to NO3? uptake ratio was 1.67. Thus, while the shoot may affect total nitrogen uptake through the export of carbohydrates to roots, the shoot (common for halves of the split-root system) apparently does not exert a direct effect on proportionality of NH4+ and NO3? uptake by roots. For whole roots supplied with both NH4+ and NO3?, the restriction in uptake of NO3? may involve a stimulation of NO3? efflux rather than an inhibition of NO3? influx. While only the net uptake of NH4+ and NO3? was measured by ion chromatography, monitoring at approximately hourly intervals during the first 3 days of treatment revealed irregularly occurring intervals of both depletion (net influx) and enrichment (net efflux) in solutions. In the case of NH4+, numbers of net efflux events were similar (21 to 24 out of 65 sequential sampling intervals) whether NH4+ was supplied with NO3? to whole-root systems or separately to an axis of the split-root system. In the case of NO3?, however, the number of net efflux events increased from 8 when NO3? was supplied to a separate axis of the split-root system to between 19 and 24 when NO3? was supplied with NH4+ to whole-root systems.  相似文献

Background and aims

Plant responses to S supply are highly dependent on N nutrition. We investigated the effect of S status on metabolic, nutritional, and production variables in Brachiaria brizantha treated with different N forms. Additionally, 15N and 34S root influx were determined in plants under short- and long-term S deprivation.

Methods

Plants were submitted to soil fertilization treatments consisted of combinations of N forms [without N, ammonium (NH4 +), nitrate (NO3 ?) or NH4 ++NO3 ?] at S rates (0, 15, 30, or 45 mg dm?3). N and S influx capacity was determined in hydroponically-grown plants.

Results

Shoot production due to S supply increased 53, 145 and 196 % with NH4 +, NH4 ++NO3 ? and NO3 ? treatments, respectively. No or low S impaired protein synthesis and led to high accumulation of N-NO3 ? and asparagine in NO3 ?-fed plants, both alone and with NH4 +. Proline accumulation was observed in NH4 +-fed plants. Short- and long-term S deprivation did not promote considerable changes in 15N influx. 34S absorption decreased depending on the N form provided: NH4 ++NO3 ? > only NH4 + > only NO3 ? > low N.

Conclusions

Including both NH4 + and NO3 ? forms in fertilizer increases N and S intake potential and thereby enhances plant growth, nutritional value and production.  相似文献

In order to investigate the effects of homogeneous and localized supply of different nitrogen forms (nitrate, NO3 ? vs ammonium, NH4 +) on the growth of tomato seedlings, root morphology and six cytokinin (CTK) fractions in xylem sap were analyzed. Whole roots were supplied with different ratios of NO3 ? to NH4 + (100:0, as 100-0NA; 75:25, as 75-25NA; 50:50, as 50-50NA) under homogeneous supply. In split-root experiments, three treatments were compared: a sole NO3 ? supply (N|N), a spatially separated supply of NO3 ? and NH4 + (N|A), and a spatially separated supply of NO3 ? and a mixture of NO3 ? and NH4 + nutrition at a ratio of 75:25 (N|AN). All concentrations of total N were set at 5 mM. The results showed that (1) homogeneous 75% NO3 ? plus 25% NH4 + supply to the whole root zone led to maximum shoot and root dry matter (DM), root surface area (RS) and root volume (RV). The spatially separated supply of NO3 ? and NH4 + (N|A) resulted in a contrasting effect on root morphology: in comparison to N|N, root DM in the NO3 ?-containing pot was increased by 50% whereas it was depressed by 50% in the NH4 +-containing pot. The 75% NO3 ? plus 25% NH4 + supply in the split-root experiment led to no significant effects either on shoot DM and root DM, or on RS and RV when compared to N|N. (2) The presence of NH4 + in the external medium led to a significantly reduced total xylem-CTK concentration, and a close negative correlation was found between xylem NH4 + and total CTK concentration irrespective of culture mode. A relatively high level of zeatin riboside (ZR) was maintained both in 75-25NA and N|A treatments. It was concluded that, in addition to the percentage of NH4 + to NO3 ? in the nutrient solution, whether NH4 + was supplied to the whole root system or to only part of the root system was also an important factor affecting plant growth. The fact that the 75-25NA and N|A treatments resulted in optimal growth of tomato seedlings might be attributed to the higher ZR concentration in xylem.  相似文献

Tomato growth was examined in solution culture under constant pH and low levels of NH4+ or NO3?. There were five nitrogen treatments: 20 mmoles m?3 NH4+, 50 mmoles m?3 NO3?, 100 mmoles m?3 NH4+ 200 mmoles m?3 NO3?, and 20 mmoles m?3 NH4++ 50 mmoles m?3 NO3?. The lower concentrations (20 mmoles m?3 NH4+ and 50 mmoles m?3 NO3?) were near the apparent Km for net NH4+ and NO3? uptake; the higher concentrations (100 mmoles m?3 NH4+ and 200 mmoles m?3 NO3?) were near levels at which the net uptake of NH4+ or NO3? saturate. Although organic nitrogen contents for the higher NO3? and the NH4++ NO3? treatments were 22.2–30.3% greater than those for the lower NO3? treatment, relative growth rates were initially only 10–15% faster. After 24 d, relative growth rates were similar among those treatments. These results indicate that growth may be only slightly nitrogen limited when NH4+ or NO3? concentrations are held constant over the root surface at near the apparent Km concentration. Relative growth rates for the two NH4+ treatments were much higher than have been previously reported for tomatoes growing with NH4+ as the sole nitrogen source. Initial growth rates under NH4+ nutrition did not differ significantly (P≥ 0.05) from those under NO3? or under combined NH4++ NO3?. Growth rates slowed after 10–15 d for the NH4+ treatments, whereas they remained more constant for the NO3? and mixed NH4++ NO3? treatments over the entire observation period of 24–33 d. The decline in growth rate under NH4+ nutrition may have resulted from a reduction in Ca2+, K+, and/or Mg2+ absorption.  相似文献

The negative effects of water stress on rice can be alleviated by NH4+ nutrition. However, the effects of mixed nitrogen (N) nutrition (NO3? + NH4+) on resistance to water stress are still not well known. To investigate the response of rice growth to water stress and its relationship with photosynthetic characteristics, a hydroponic experiment supplying different N forms was conducted. Compared with NO3? nutrition, mixed‐N and NH4+ nutrition greatly alleviated the reduction of leaf area, chlorophyll content, and photosynthesis under water stress, whilst subsequently maintaining higher biomass. In contrast, water stress inhibited the root‐shoot ratios in NH4+‐ and mixed‐N‐supplied plants, indicating reduced root growth and higher photosynthate availability to shoots. The following key observations were made: (1) a similar stomatal limitation and low proportion of activated Rubisco were observed among the three different N nutrition regimes; (2) increased mesophyll conductance in NH4+‐ and mixed‐N‐supplied plants simultaneously stimulated leaf photosynthesis and improved the water use efficiency and (3), the maximum carboxylation rate and actual photochemical efficiency of photosystem II in NH4+‐ and mixed‐N‐supplied plants were significantly higher than that in NO3?‐supplied plants, thus resulting in higher photochemical efficiency under water stress. In conclusion, mixed‐N and NH4+ nutrition may be used to develop strategies for improved water stress resistance and stimulated biomass production under conditions of osmotic stress and possibly drought.  相似文献

To investigate whether mixed-N (NO3 ??+?NH4 +) nutrition can enhance rice growth under water-deficit condition, a hydroponic experiment in which rice plants were supplied with different N forms (NO3 ?, NH4 + and mixed-N) was conducted, and the intrinsic mechanisms involved in photosynthesis, root-shoot carbon partitioning, and hormone signalling were investigated. Water stress was found to decrease rice biomass, leaf area, chlorophyll and Rubisco contents. However, mixed-N nutrition substantially alleviated these inhibitions compared with NO3 ? nutrition alone. Mixed-N nutrition also maintained a higher electron transport rate, actual photochemical efficiency of PSII, and non-photochemical quenching, causing higher photosynthesis and photochemical efficiency. Water stress up-regulated leaf sucrose-phosphate synthase (SPS), but down-regulated acid invertase (InvA). However, leaf InvA and root sucrose synthase in the cleavage direction (SSc) in NO3 ? nutrition was higher than that in mixed-N nutrition. Water stress decreased indole acetic acid (IAA) content in leaves and cytokinins content in roots, but their contents in mixed-N nutrition were higher than those in NO3 ? nutrition. In mixed-N nutrition, the up-regulation of SPS and IAA in leaves and the reduction of sucrose metabolism (SSc and InvA) in roots jointly resulted in the accumulation of sucrose in leaves and the inhibition of its transportation to roots, finally reducing the root:shoot ratio (R/S). The reduced R/S provides more photosynthates for shoots and increases the utilisation efficiency, thereby strengthening the water-deficit tolerance of plants. We concluded that the strengthened water-deficit tolerance in mixed-N-supplied rice was closely associated with higher accumulation of dry matter mainly via improvement of photosynthesis and photochemical efficiency, hormone balance, and coupling with root-shoot carbon partitioning.  相似文献

As a major antioxidant in plants, ascorbic acid (AsA) plays a very important role in the response to aluminum (Al) stress. However, the effect of AsA on the mitigation of Al toxicity and the mechanism of nitrate nitrogen (NO3 ?–N) uptake by plants under Al stress are unclear. In this study, a hydroponic experiment was conducted using peak 1 A rice (sterile line, Indica) with weaker resistance to Al and peak 1 superior 5 rice (F1 hybrid, Indica) with stronger resistance to Al to study the effects of exogenous AsA on the physiological and biochemical responses to NO3 ?–N uptake by rice roots exposed to 50 μmol L?1 Al. Al stress induced increases in the concentrations of H2O2 and malondialdehyde (MDA) and in the activities of antioxidant enzymes [such as superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and ascorbate peroxidase (APX)]. Plasma membrane (PM) H+-ATPase and H+-pump activities, endogenous AsA content and NO3 ?–N uptake in rice roots decreased under Al stress. After treatment with 2 mmol L?1 exogenous AsA combined with Al, concentrations of H2O2 and MDA in roots notably decreased, and endogenous AsA content and activities of SOD, POD, CAT, and APX in rice roots increased significantly; furthermore, the interaction of PM H+-ATPase and the 14-3-3 protein was also enhanced significantly compared with that in control plants without AsA treatment, which clearly increased NO3 ?–N uptake. Based on all of these data, the application of AsA significantly reduced the accumulation of H2O2 and MDA and increased the activities of PM H+-ATPase and the H+-pump by increasing the endogenous AsA content, the antioxidant enzyme activities, and the interaction of PM H+-ATPase and the 14-3-3 protein in the roots of the two rice cultivars under Al stress, thereby improving the uptake of NO3 ?–N in rice.  相似文献

InMucuna pruriens var.utilis, grown with nitrate-N in a hydroponic split-root system, an Al avoidance reaction of root growth was observed, which was ascribed to local P stress in the Al containing compartment. The Al avoidance reaction was similar to the avoidance ofMucuna roots of acid subsoil in the field where roots grew preferentially in the topsoil. In the present paper the effect of different N forms (NO3 and NH4 +) on the reactions ofMucuna to Al were studied, since in acid soils N is present as a mixture of NO3 and NH4 +. No interaction between the N form and Al toxicity was found. A hydroponic split-root experiment with NH4NO3 nutrition, which is comparable to the situation in the field, showed that under these conditions Al avoidance did not occur. Excessive use of nitrogen (N) fertilizer has increased ammonium (NH4+) accumulation in many paddy soils to levels that reduce rice vegetative biomass and yield. Based on studies of NH4+ toxicity in rice (Oryza sativa, Nanjing 44) seedlings cultured in agar medium, we found that NH4+ concentrations above 0.75 mM inhibited the growth of rice and caused NH4+ accumulation in both shoots and roots. Use of excessive NH4+ also induced rhizosphere acidification and inhibited the absorption of K, Ca, Mg, Fe and Zn in rice seedlings. Under excessive NH4+ conditions, exogenous γ‐aminobutyric acid (GABA) treatment limited NH4+ accumulation in rice seedlings, reduced NH4+ toxicity symptoms and promoted plant growth. GABA addition also reduced rhizosphere acidification and alleviated the inhibition of Ca, Mg, Fe and Zn absorption caused by excessive NH4+. Furthermore, we found that the activity of glutamine synthetase/NADH‐glutamate synthase (GS; EC 6.3.1.2/NADH‐GOGAT; EC1.4.1.14) in root increased gradually as the NH4+ concentration increased. However, when the concentration of NH4+ is more than 3 mM, GABA treatment inhibited NH4+‐induced increases in GS/NADH‐GOGAT activity. The inhibition of ammonium assimilation may restore the elongation of seminal rice roots repressed by high NH4+. These results suggest that mitigation of ammonium accumulation and assimilation is essential for GABA‐dependent alleviation of ammonium toxicity in rice seedlings.  相似文献

A study was conducted to examine the hypothesis that the effects of external Al on NO3? uptake and assimilation depend upon the concentration of Al present. Young soybean seedlings [Glycine max (L.) Merrill, cv. Essex], growing under moderate acidity stress at pH 4-2, were exposed to a range of {A13+} in solution for 3d, and to labelled 99 atom %15NO3? during the final hour of Al exposure. Uptake of 15NO3?g?1 root dry weight was increased by about 28% in the presence of Al at {A13+} below 10 mmolm?3, and NO3? uptake was decreased by about 12% when the {A13+} increased to 44mmoln?3. The stimulation phase closely paralleled stimulation of root elongation. At higher {A13+}, the inhibition of root elongation was much more severe than that of NO3? uptake. There was no indication of a separate effect of Al on root 15NO3? reduction in situ, as the accumulation of reduced 15N in the root remained a similar percentage of 15NO3? uptake at all {A13+}. At higher {A13+}, the atom %15N enrichment of the insoluble reduced-N (protein) fraction of root tips increased. This suggests that the Al inhibition of root elongation did not result from disruption of the N supply to the root apex.  相似文献

In vivo 31P nuclear magnetic resonance (NMR) was used to characterize the effect of the N form (NO3 vs. NH4) and the external pH (4, 6, and 8), on the intracellular pH of root tips (0–5 mm) and root segments (5–30 mm). Ammonium-grown root tips were the most sensitive to changes in the external pH. In vivo 15N NMR was used to characterize the pathway of primary ammonium assimilation in the ammonium-grown roots and to compare the activity of the apical and more-basal root parts. The kinetics of 15NH4 + incorporation showed that primary assimilation in both root tips and root segments followed the glutamine synthetase (GS) pathway. In agreement with the reported gradient of GS along the seminal root of maize, incorporation of label into glutamine amide was more rapid in tips than in segments. It is suggested that this higher GS activity increases the endogenous proton production and thus contributes to the greater dependence of the cytoplasmic pH on the external pH in the ammonium-treated root tips.  相似文献

The effects of nitrogen form and phosphorus source on the growth, nutrient uptake and rhizosphere soil property of tea (Camellia sinensis L.) were investigated in a pot experiment. The experiment was performed with a compartmental cropping device, which enables the collection of rhizosphere soil at defined distances from the root of tea plant. Nitrogen was supplied as nitrate or ammonium in combination with soluble phosphorus as Ca(H2PO4)2 or insoluble P as rock phosphate. The leaf dry matter production of tea was significantly greater in the treatments with NH4 + than NO3 -, whereas dry matter production of root and stem was not significantly affected. Addition of phosphorus as either source did not influence the dry matter production. The concentrations of K in root, Mg and Ca in both the shoot and root supplied with NO3 - were significantly higher than in NH4 + and influence of P sources was minor. On the contrary, Al and Mn concentrations were significantly larger in NH4 --fed plants which could be attributed to remarkably increased availability of Al and Mn caused by acidification of the rhizosphere soil (the first 1-mm soil section from the root surface) with NH4–N nutrition. The concentration of N in shoot was also significantly higher in NH4- than in NO3-fed plants, indicating higher use efficiency of NH4–N. Whatever the phosphate source, rhizosphere pH declined in ammonium compared to in nitrate treatment. The pH decrease was much larger when no P or soluble P were applied and reached 0.85–1.30 units which extended to 3–5 mm away from the root surface. Exchangeable acidity, content of exchangeable Al and Mn were also considerably higher in the rhizosphere soils of NH4 + fed tea plants. Significant amounts of P dissolved from rock phosphate accumulated in rhizosphere of NH4 +, not NO3 -, suggesting that the dissolution of rock phosphate was induced by the proton excreted by tea root fed with ammonium. With soluble P addition, shoot and root P concentrations were greater in NH4 + than in NO3 - treatment and it appeared that this difference could not be sufficiently explained by the available P content in soil which was only slightly higher in NH4 + treatment. With rock phosphate addition, the shoot and root P concentrations were hardly affected by nitrogen form, although the available P content was much higher and accumulated in the rhizosphere soil supplied with ammonium. Lolium perenne L. cv. 23 (perennial ryegrass) plants were grown in flowing solution culture and acclimatized over 49 d to low root temperature (5°C) prior to treatment at root temperatures of 3, 5, 7 and 9°C for 41 d with common air temperature of 20/15°C day/night and solution pH 5·0. The effects of root temperature on growth, uptake and assimilation of N were compared with N supplied as either NH4 or NO3 at 10 mmol m?3. At any given temperature, the relative growth rate (RGR) of roots exceeded that of shoots, thus the root fraction (Rf) increased with time. These effects were found in plants grown with the two N sources. Plants grown at 3 and 5°C had very high dry matter contents as reflected by the fresh weight: freeze-dried weight ratio. This ratio increased sharply, especially in roots at 7 and 9°C. Expressed on a fresh weight basis, there was no major effect of root temperature on the [N] of plants receiving NHJ but at any given temperature, the [N] in plants grown with NHJ was significantly greater than in those grown with NO3. The specific absorption rate (SAR) of NH+4 was greater at all temperatures than SAR-NO3. In plants grown with NH+, 3–5% of the total N was recovered as NH+4, whereas in those grown with NO?3 the unassimilated NO?3 rose sharply between 7 and 9°C to become 14 and 28% of the total N in shoots and roots, respectively. The greater assimilation of NH+4 lead to concentrations of insoluble reduced N (= protein) which were 125 and 20% greater, in roots and shoots, respectively, than in NO?3-grown plants. Plants grown with NH+4 had very much greater glutamine and asparagine concentrations in both roots and shoots, although other amino acids were more similar in Concentration to those in NO?3 grown plants. It is concluded that slow growth at low root temperature is not caused by restriction of the absorption or assimilation of either NH+4 or NO?3. The additional residual N (protein) in NH+4 grown plants may serve as a labile store of N which could support growth when external N supply becomes deficient.  相似文献

The effect of ectomycorrhizal association of Pinus pinaster with Hebeloma cylindrosporum was investigated in relation to the nitrogen source supplied as mineral (NH4+ or NO3?) or organic N (L ‐glutamate) and at 5 mol m?3. Plants were grown for 14 and 16 weeks with mineral and organic N, respectively, and samples were collected during the last 6 weeks of culture. Total fungal biomass was estimated using glucosamine amount and its viability was assessed using the glucosamine to ergosterol ratio. Non‐mycorrhizal plants grew better with NH4+ than with NO3? and grew very slowly when supplied with L ‐glutamate. The presence of the fungus decreased the growth of the host plant with mineral N whereas it increased it with L ‐glutamate. Whatever the N source, most of the living fungal biomass was associated with the roots, whereas the main part of the total biomass was assayed outside the root. The form of mineral N did not significantly affect N accumulation rates over the 42 d in control plants. In mycorrhizal plants grown on either N source, the fungal tissues developing outside of the root were always the main N sink. The ectomycorrhizal association did not change 15NH4+ uptake rate by roots, suggesting that the growth decrease of the host‐plant was related to the carbon cost for fungal growth and N assimilation rather than to a direct effect on NH4+ acquisition. In contrast, in NO3?‐grown plants, in addition to draining carbon for NO3? reduction the fungus competed with the root for NO3? uptake. With NH4+ or NO3? feeding, although mycorrhizal association improved N accumulation in shoots, we concluded that it was unlikely that the fungus had supplied the plant with N. In L ‐glutamate‐grown plants, the presence of the fungus increased the proportion of glutamine in the xylem sap and improved both N nutrition and the growth rate of the host plant.  相似文献
