Common bean (Phaseolus vulgaris L.) is grown over a wide rangeof environments, including sites with low or high soil temperaturesat sowing time. To describe the temperature responseof seedgermination, 20 bean genotypes were evaluated using a rolledpaper towel system with 11 constant temperatures ranging from12 to 34 °C. Germination response was characterized by fittingcumulative counts using a maximum-likelihood analysis. Rateof germination increased from abase temperature (Tb) typicallynear 8 °C to an optimal development temperature (To) of29 to 34 °C. Tb did not differ among common bean genotypes.Mesoamerican germplasm showed slightlyhigher To than Andeangermplasm, but there was large variation in To within each ofthe two gene pools. The single accession of tepary bean (P.acutifolius) evaluated appeared to be the mosttolerant to highgermination temperatures. Key words: Common bean, seed germination rate, temperature  相似文献

HSP70 expression in the CNS in response to exercise and heat stress in rats   总被引:7,自引:0,他引:7  
We havepreviously documented the regional distribution of 70-kDa heat shockprotein (HSP70) in brains of rats made hyperthermic by brief exposureto high-powered microwaves (HPM; 2.06 GHz). We now compare HSP70expression induced by HPM exposure to that induced by exertionaland/or environmental heat stress. Rats were chronicallyimplanted with a temperature probe guide in the hypothalamic region ofthe brain (Tbr). After recovery,the following treatment groups were examined: HPM; sham exposed;treadmill exercise at room temperature (24°C; Ex-1); treadmillexercise in a warm environment (34°C; Ex-2); and sedentary groups(Sed-1 and Sed-2), in which ambient temperature was adjusted so thatthe Tbr mimicked the Tbr in the corresponding exercisegroups. Significant HSP70 expression occurred only in the hyperthermic(Ex-2, Sed-2, and HPM) groups. The pattern of HSP70 expression wassimilar among Ex-2 and Sed-2 rats but differed from that in HPM rats.We conclude that 1) the pattern ofHSP70 expression differs between HPM and nonmicrowave heating, and2) exercise alone was not sufficientto induce central HSP70 expression.

Eighteen poplar (Populus) clones, varying in growth capacitywere grown in plastic containers under outdoor conditions. Duringthe first year of growth their net CO2 exchange rate (NCER)was studied by infra-red gas analysis as a function of photosyntheticphoton flux density (PPFD) under controlled environment conditions.Maximal NCER (under saturating PPFD) and the PPFD compensationpoint were significantly correlated with the first year's shootlength and hence with the above ground biomass production ofthe various clones. Specific S-glycoproteins were isolated from three Brassica campestriscultivars homozygous with respect to the S-alleles S8, S9 andS12. Amino acid sequences of various peptide fragments of theS-proteins were determined using a gas-phase protein sequencer,and a nearly complete amino acid sequence of the S8-glycoproteinwas determined on the basis of the revised cDNA sequence ofthe B. oleracea S-specific glycoprotein. The lysyl endopeptidasefragments of S9 and S12-glycoproteins were aligned in comparisonwith the sequence of the S8-glycoprotein. Although extensivesequence homology was evident among the three S-glycoproteins,the sequences of the middle part were relatively different fromeach other. The pedicel region of Zea mays kernels contains a unique formof maize glutamine synthetase (GS), GSpl. RNA blot analysisusing GS gene-specific probes revealed that the expression ofthe GS.2 gene was specific to the pedicel and that it increasedin the kernels during development. This pattern of the maizeGS.2 gene expression is consistent with the tissue specificityof the GSpl protein and suggests that it encodes the GSpl isoformof maize GS. The recessive gene na results in peas with extremely short internodes(phenotype nana). In intact plants, na prevents expression ofthe Le/le gene pair which determine the tall/dwarf difference.na blocks a step early in the gibberellin biosynthetic pathwaywhile le prevents conversion of gibberellin A20 to GA1. Background and Aims: The grass Alloteropsis semialata is the only plant species withboth C3 and C4 subspecies. It therefore offers excellent potentialas a model system for investigating the genetics, physiologyand ecological significance of the C4 photosynthetic pathway.Here, a molecular phylogeny of the genus Alloteropsis is constructedto: (a) confirm the close relationship between the C3 and C4subspecies of A. semialata; and (b) infer evolutionary relationshipsbetween species within the Alloteropsis genus. Methods: The chloroplast gene ndhF was sequenced from 12 individuals,representing both subspecies of A. semialata and all four ofthe other species in the genus. ndhF sequences were added tothose previously sequenced from the Panicoideae, and used toconstruct a phylogenetic tree. Key Results: The phylogeny confirms that the two subspecies of A. semialataare among the most recently diverging lineages of C3 and C4taxa currently recognized within the Panicoideae. Furthermore,the position of the C3 subspecies of A. semialata within theAlloteropsis genus is consistent with the hypothesis that itsphysiology represents a reversion from C4 photosynthesis. Thedata point to a similar evolutionary event in the Panicum stenodesP.caricoidesP. mertensii clade. The Alloteropsis genusis monophyletic and occurs in a clade with remarkable diversityof photosynthetic biochemistry and leaf anatomy. Conclusions: These results confirm the utility of A. semialata as a modelsystem for investigating C3 and C4 physiology, and provide moleculardata that are consistent with reversions from C4 to C3 photosynthesisin two separate clades. It is suggested that further phylogeneticand functional investigations of the Alloteropsis genus andclosely related taxa are likely to shed new light on the mechanismsand intermediate stages underlying photosynthetic pathway evolution.  相似文献

The present study illustrates elements ofthe signal cascades involved in the activation of taurine effluxpathways in myotubes derived from skeletal muscle cells. Exposingprimary skeletal muscle cells, loaded with 14C-taurine, to1) hypotonic media, 2) the phospholipaseA2 (PLA2) activator melittin, 3)anoxia, or 4) lysophosphatidyl choline (LPC) causes anincrease in 14C-taurine release and a concomitantproduction of reactive oxygen species (ROS). The antioxidants butulatedhydroxy toluene and vitamin E inhibit the taurine efflux after cellswelling, anoxia, and addition of LPC. The muscle cells possess twoseparate taurine efflux pathways, i.e., a swelling- andmelittin-induced pathway that requires 5-lipoxygenase activity foractivation and a LPC-induced pathway. The two pathways aredistinguished by their opposing sensitivity toward the anion channelblocker DIDS and cholesterol. These data provide evidence forPLA2 products and ROS as key mediators of the signalcascade leading to taurine efflux in muscle.

Endogenous levels of gibberellins in shoots and ears of twodwarf rice (Oryza sativa L.) cultivars, Tan-ginbozu (dx mutant)and Waito-C (dy mutant), were analyzed and compared with thoseof normal rice cultivar, Nihonbare. The endogenous levels of13-hydroxylated gibberellins in Tan-ginbozu were much lowerthan those in Nihonbare. In Waito-C, the levels of GA19 andGA20 in the shoots were higher but that of GA1 was lower thanthe levels of these gibberellins in Nihonbare. These resultssupport the hypothesis that the dy gene controls the 3ß-hydroxylationof GA20 to GA1 while the dx gene controls a much earlier stepin the gibberellin biosynthesis. Our results indicate that GA1is the active gibberellin that regulates the vegetative growthof rice. The endogenous levels of GA4 in the ears of the twodwarf cultivars of rice were higher than the level of GA4 inthe ears of the normal cultivar, Nihonbare suggesting that thebiosynthesis of gibberellin is not blocked in the anthers ofthe dwarf rice. A single striata mutant (green and yellowish-white striped leaves)was observed in first generation (A1) barley plants grown fromaged seeds. In the following generation (A2) normal, striataand virido-albina (yellowish-white leaves and pale yellowish-greentips) phenotypes were observed. The segregation of the progenywas examined in A2 and A3 and it was found that the large majorityof normal and virido-albina phenotypes bred true, whereas mostof the A2 striata phenotypes segregated normal, striata andvirido-albina phenotypes in variable ratios. The expression of both mutant phenotypes is temperature-dependentand normally only observed when the initial temperature forseedling growth is about 5 °C. In the vast majority of individualsthe symptoms disappear on transfer to 7 °C or more. Evenat 5 °C the expression of the striata phenotype is variable,ranging from seedlings which are little different from the normalphenotype to those which are difficult to distinguish from thevirido-albina phenotype. The genetical status of this mutant is discussed in relationto these observations and previous reports of the anomolousbehaviour of the striata character. It is suggested that thesymptoms could be the result of a single gene mutation showingincomplete dominance, and that the anomalous segregation ratiosresult mainly from difficulties in categorizing the heterozygousgenotype at the extremes of its phenotypic expression. However,there is insufficient evidence to exclude the possibility thatthe symptoms derive from a cytoplasmically inherited extrachromosomalmutation. The levels of endogenous GA, and GA4 in the anthers of a standardcultivar and the corresponding male-sterile, single gene mutantof rice, Oryza saliva L. (Japonica), were measured by radioimmunoassayusing GA4-specific antiserum that showed highly specific immunoreactivitywith GA, and GA4. The recessive of gene, producing tendrils in place of leaves,and the recessive st gene, reducing stipule size, produce phenotypesof pea that are termed leafless (afafstst) and semi-leafless(afafStSt). Photosynthesis and growth of these two types werecompared with the conventional phenotype (AfAfStSt) during thefirst 9 days of post-emergent growth. The conventional seedlingshowed faster net photosynthesis per unit dry weight than theleafless phenotype, whilst the semi-leafless seedlings wereintermediate. Differences in dark respiration were small butleafless seedlings had significantly higher rates soon afteremergence. Where the three phenotypes used were isogenic, except for ofand st, the rates of shoot growth were in the same ranking orderas net CO2 uptake. With three other genotypes, representingthe three phenotypes, more similar shoot growth was found betweenthe conventional and semi-leafless phenotype, possibly becauseof compensating differences in embryonic axis size. The ratesof growth of roots and the rates of dry weight loss from thecotyledons showed no consistent differences between phenotypes. The results are discussed in relation to the potential for thesemi-leafless phenotype as an alternative to the conventionalphenotype for the dried pea crop. Distribution of iron-containing oxidases in aging nodal rootsof rice and wheat was studied. Activities of cytochrome c oxidase(1.9.3.1 [EC] , cytochrome c : O2 oxidoreductase), catalase (1.11.1.6 [EC] ,H2O2: H2O2 oxidoreductase) and peroxidase (1.11.1.7 [EC] , donor:H2O2 oxidoreductase) in wheat roots were comparatively higherthan were those in rice roots at corresponding stages. Cytochromec oxidase in roots remained active throughout the lives of therice and wheat crops. In rice roots, catalase seemed to playa distinct role around the panicle formation stage. Decay ofcatalase activity took place earlier than did that of peroxidaseand cytochrome c oxidase activities. In wheat roots similarenzyme activity changes were not observed. A critical stage in flower bud growth in the spike of Gladioluswhich is initiated by gibberellic acid (GA3) and sustained bysucrose has been identified. This corresponds to the stage atwhich separation of the outer bract occurs. In buds at differentdevelopmental stages isolated and held in water, it is the samebud stage that first shows increased growth. Buds not inducedby light were shown to respond more significantly to GA3 andsucrose than those induced by light. NADP-malic enzyme (NADP-ME) and phosphoenolpyruvate carboxykinase(PCK) are specifically expressed in bundle sheath cells (BSCs)in NADP-ME-type and PCK-type C4 plants, respectively. Unlikethe high activities of these enzymes in the green leaves ofC4 plants, their low activities have been detected in the leavesof C3 plants. In order to elucidate the differences in the geneexpression system between C3 and C4 plants, we have producedchimeric constructs with the ß-glucuronidase (GUS)reporter gene under the control of the maize NADP-Me (ZmMe)or Zoysia japonica Pck (ZjPck) promoter and introduced theseconstructs into rice. In leaves of transgenic rice, the ZmMepromoter directed GUS expression not only in mesophyll cells(MCs) but also in BSCs and vascular cells, whereas the ZjPckpromoter directed GUS expression only in BSCs and vascular cells.Neither the ZjPck nor ZmMe promoters induced GUS expressiondue to light. In rice leaves, the endogenous NADP-Me (OsMe1)was expressed in MCs, BSCs and vascular cells, whereas the ricePck (OsPck1) was expressed only in BSCs and vascular cells.Taken together, the results obtained from transgenic rice demonstratethat the expression pattern of ZmMe or ZjPck in transgenic ricewas reflected by that of its counterpart gene in rice. Some plant species show constant rates of respiration and photosynthesismeasured at their respective growth temperatures (temperaturehomeostasis), whereas others do not. However, it is unclearwhat species show such temperature homeostasis and what factorsaffect the temperature homeostasis. To analyze the inherentability of plants to acclimate respiration and photosynthesisto different growth temperatures, we examined 11 herbace-ouscrops with different cold tolerance. Leaf respiration (Rarea)and photosynthetic rate (Parea) under high light at 360 µll–1 CO2 concentrations were measured in plants grown at15 and 30°C. Cold-tolerant species showed a greater extentof temperature homeostasis of both Rarea and Parea than cold-sensitivespecies. The underlying mechanisms which caused differencesin the extent of temperature homeostasis were examined. Theextent of temperature homeostasis of Parea was not determinedby differences in leaf mass and nitrogen content per leaf area,but by differences in photosynthetic nitrogen use efficiency(PNUE). Moreover, differences in PNUE were due to differencesin the maximum catalytic rate of Rubisco, Rubisco contents andamounts of nitrogen invested in Rubisco. These findings indicatedthat the temperature homeostasis of photosynthesis was regulatedby various parameters. On the other hand, the extent of temperaturehomeostasis of Rarea was unrelated to the maximum activity ofthe respiratory enzyme (NAD-malic enzyme). The Rarea/Parea ratiowas maintained irrespective of the growth temperatures in allthe species, suggesting that the extent of temperature homeostasisof Rarea interacted with the photosynthetic rate and/or thehomeostasis of photosynthesis.  相似文献

Studies have been mode of the growth curvatures induced in theroots of Lepidium sativum, the coleoptiles of Avena saliva andthe sporangiophores of Phycomyces blakesleeanus by exposureto an intense magnetic gradient during continuous rotation ona slow klinostat (0.5 rpm). Attempts have been made to determinethe relationships of the curvature responses to the parametersof the magnetic field (H, dH/dx, and H. dH/dx). Analysis ofthe data was by a progressive regression analysis up to thethird order coupled with an analysis of variance to test theimprovement of fit at each stage. Response is characterizedby an initial lag (reaction time, T) followed by a constantrate of curvature down the magnetic gradient for the first 20or 30? of curvature. The reciprocal of T was the most satisfactorymeasure of response (i. e. a measure of the rate of inductionof the response while the organ was in a constant magnetic environment)and in Avena and Phycomyces was best fitted by linear regressionson logedH/dx and on logeH. dH/dx. For Lepidium, with much morevariable data, the best fit was with a linear regression onlogeH. The significance of these results is discussed.  相似文献
