中国五台山多目涡虫(涡虫纲,三肠目)染色体及核型分析   总被引:3,自引:0,他引:3  
利用空气干燥法对采自山西茶铺和镇海寺的五台山多目涡虫Polycelis wutaishanica Liu,1996的核型进行分析,结果表明:五台山多目涡虫体细胞有42条染色体,为2倍体,核型公式2n=2x=42 =28m 14sm,其中28条为中部着丝粒染色体, 14条为亚中部着丝粒染色体,第1对中部着丝粒染色体明显比其它染色体长。五台山多目涡虫的核型属于2C型。  相似文献

利用骨髓细胞蒸汽固定法制备染色体标本,研究了分布于河南新乡的花背蟾蜍(Bufo raddei)的核型、C-带和Ag-NORs。结果表明:河南产花背蟾蜍体细胞染色体数为22条,核型公式为2n=22(20m 2sm),全部为中部或亚中部着丝点染色体,NF=44。Ag-NORs具有多态现象。C-带核型显示22条染色体的着丝点均正染,No.1染色体近着丝粒处有不恒定插入型C-带,No.4染色体具有恒定近端部插入型C-带,随体部位被正染的仅占所观察细胞的15%。  相似文献

Karyotypes of females and males of the brackish-water polychaete Hediste japonica sensu stricto, collected from the Ariake Sea, Japan, were examined by using regenerating tails. We used the Giemsa staining method and a computer-assisted image-analyzing system for the identification of each chromosome pair. The somatic chromosome number was 2n=28. The presence of an XX-XY (male heterogametic) sex chromosome system was determined from well-spread metaphase plates of somatic cells. The type of sex chromosomes related with phenotype exactly. The metacentric Y chromosome was much larger than the submetacentric X chromosome. All autosomes were metacentric. The karyotype of this species was compared with those of the other two closely related species (H. diadroma and H. atoka). The karyotypes of all the three species were similar to one another.  相似文献

Linkage group identities and homologies were determined for metaphase chromosomes of Sorghum bicolor (2n = 20) by FISH of landed BACs. Relative lengths of chromosomes in FISH-karyotyped metaphase spreads of the elite inbred BTx623 were used to estimate the molecular size of each chromosome and to establish a size-based nomenclature for sorghum chromosomes (SBI-01-SBI-10) and linkage groups (LG-01 to LG-10). Lengths of arms were determined to orient linkage groups relative to a standard karyotypic layout (short arms at top). The size-based nomenclature for BTx623 represents a reasonable choice as the standard for a unified chromosome nomenclature for use by the sorghum research community.  相似文献

Cytogenetic analysis of Astylus antis using mitotic and meiotic cells was performed to characterize the haploid and diploid numbers, sex determination system, chromosome morphology, constitutive heterochromatin distribution pattern and chromosomes carrying nucleolus organizer regions (NORs). Analysis of spermatogonial metaphase cells revealed the diploid number 2n = 18, with mostly metacentric chromosomes. Metaphase I cells exhibited 2n = 8II+Xyp and a parachute configuration of the sex chromosomes. Spermatogonial metaphase cells submitted to C-banding showed the presence of small dots of constitutive heterochromatin in the centromeric regions of nearly all the autosomes and on the short arm of the X chromosome (Xp), as well as an additional band on one of the arms of pair 1. Mitotic cells submitted to double staining with base-specific fluorochromes (DAPI-CMA(3) ) revealed no regions rich in A+T or G+C sequences. Analysis of spermatogonial mitotic cells after sequential Giemsa/AgNO (3) staining did not reveal any specific mark on the chromosomes. Meiotic metaphase I cells stained with silver nitrate revealed a strong impregnation associated to the sex chromosomes, and in situ hybridization with an 18S rDNA probe showed ribosomal cistrons in an autosomal bivalent.  相似文献

采用常规空气干燥法制片,对寄生于黄鳝(Monopterus albus)体腔内的胃瘤线虫(Eustrongylidesignotus)染色体核型进行分析。结果表明:胃瘤线虫体细胞有12条染色体,为二倍体,核型公式为2n=12=10 m+2 sm。由5对常染色体和1对性染色体组成,性别决定模式为XX-XY,其中X、Y和1~4号染色体都为中着丝粒染色体,5号为亚中着丝粒染色体。每对染色体都有特定的G-带带型。  相似文献

A karyometric analysis of the chromosome set of the marine turbellariansMonocelis fusca, M. lineata andParotoplana macrostyla has been carried out. The karyotype of the twoMonocelis species investigated (2n=6) is formed by three pairs of small and similarly sized chromosomes: InM. fusca, chromosome 1 is metacentric, chromosome 2 acrocentric and chromosome 3 is subtelocentric.M. lineata also presents one pair of metacentric chromosomes (chromosome 2), while chromosomes 1 and 3 are submetacentric.P. macrostyla (2n=12) reveals two pairs of large metacentric and four pairs of small chromosomes, three of which are metacentric, whereas the last is subtelocentric.  相似文献

Peripheral blood lymphocyte metaphase chromosomes of three Bovoidean species have been studied using Quinacrine fluorescence and Giemsa banding techniques to give Q-, G-, and C-banding patterns. Q- and G-banding characteristics, coupled with chromosome length, enabled all of the chromosomes in each of the chromosome complements to be clearly distinguished, although some difficulties were encountered with the very smallest chromosomes. A comparison of G-banding patterns between the species revealed a remarkable degree of homology of banding patterns. Each of the 23 different acrocentric autosomes of the domestic sheep (2n=54) was represented by an identical chromosome in the goat (2n=60) and the arms of the 3 pairs of sheep metacentric autosomes were identical matches with the remaining 6 goat acrocentrics. A similar interspecies homology was evident for all but two of the autosomes in the ox (2n=60). This homology between sheep metacentric and goat acrocentric elements confirms a previously suggested Robertsonian variation. The close homology in G-banding patterns between these related species indicates that the banding patterns are evolutionarily conservative and may be a useful guide in assessing interspecific relationships. —The centromeric heterochromatin in the autosomes of the three species was found to show little or no Q-or G-staining, in contrast to the sex chromosomes. This lack of centromeric staining with the G-technique (ASG) contrasts markedly with results obtained with other mammalian species. However, with the C-banding technique these regions show a normal intense Giemsa stain and the C-bands in the sex chromosomes are inconspicuous. The amount of centromeric heterochromatin in the sheep metacentric chromosomes is considerable less than in the acrocentric autosomes or in a newly derived metacentric element discovered in a goat. It is suggested that the pale G-staining of the centromeric heterochromatin in these species might be related to the presence of G-Crich satellite DNA.  相似文献

本文首次报道虫草蝠蛾(鳞翅目,蝙蝠蛾科,蝠蛾属)的有丝分裂染色体核型。应用醋酸分离和热干燥技术,研究了云南的两种虫草蝠蛾Hepialus zhayuensis Chu et Wang和Hepialus sp.的有丝分裂染色体,它们的染色体数目为2n=64。在有丝分裂的早中期染色体上清晰地呈现出散漫着丝粒。然而,分裂中期和较晚的中期阶段,每条染色体都具显著的初级着丝粒(即主缢痕)。它们的雄性中期核型中都有一对典型的异形性染色体,X染色体着色稍淡,且都具中或亚中着丝粒;Y染色体比X染色体长,染色很深。 在雄性的分裂间期细胞中,观察到异固缩性染色质体,此异固缩体是Y染色体。  相似文献

In the progeny of a trisomic type for chromosome 6, Purple, a 16-chromosome type was obtained, which had a pair of new metacentric chromosome 6 in excess. The new metacentric chromosome 6 was shorter than any of the 14 chromosomes of normal barley complement and showed a heteropycnotic nature at late prophase in somatic mitosis. At metaphase I in the plants with 14+one metacentric chromosome 6 (2n=15) the chromosome configuration was exclusively 7II+1I indicating that the extra metacentric chromosome 6 could not associate with the normal chromosome 6. At diakinesis and metaphase I in the new 16-chromosome plants most of the sporocytes showed 8IIor 7II+2I. Neither tetravalents nor trivalents were observed at meiosis. The chromosome behaviour at anaphase I and later stages of meiosis was regular in general, resulted in a fairly high pollen fertility of about 61 per cent. Seed fertility however, was very low. The transmission rate of the new metacentric chromosome 6 through the pollen was extremely low in 16-chromosome plants. Karyotypic and cytogenetic characteristics of catfish Harttia carvalhoi (Paraíba do Sul River basin, S?o Paulo State, Brazil) were investigated using differential staining techniques (C-banding, Ag-staining) and fluorescent in situ hybridization (FISH) with 18S and 5S rDNA probes. The diploid chromosome number of females was 2n = 52 and their karyotype was composed of nine pairs of metacentric, nine pairs of submetacentric, four pairs of subtelocentric and four pairs of acrocentric chromosomes. The diploid chromosome number of males was invariably 2n = 53 and their karyotype consisted of one large unpaired metacentric, eight pairs of metacentric, nine pairs of submetacentric, four pairs of subtelocentric, four pairs of acrocentric plus two middle-sized acrocentric chromosomes. The differences between female and male karyotypes indicated the presence of a sex chromosome system of XX/XY1Y2 type, where the X is the largest metacentric and Y1 and Y2 are the two additional middle-sized acrocentric chromosomes of the male karyotype. The major rDNA sites as revealed by FISH with an 18S rDNA probe were located in the pericentromeric region of the largest pair of acrocentric chromosomes. FISH with a 5S rDNA probe revealed two sites: an interstitial site located in the largest pair of acrocentric chromosomes, and a pericentromeric site in a smaller metacentric pair of chromosomes. Translocations or centric fusions in the ancestral 2n = 54 karyotype is hypothesized for the origin of such multiple sex chromosome systems where females are fixed translocation homozygotes whereas males are fixed translocation heterozygotes. The available cytogenetic data for representatives of the genus Harttia examined so far indicate large kayotype diversity.  相似文献

A karyological study was carried out in order to compared the chromosome numbers, chromosome morphologies and karyotypes of the oriental liver fluke, Clonorchis sinensis (Trematoda: Opisthorchiidae), collected from Korea and China. Chromosome preparations were made by means of air-drying method. The chromosome number was 2n = 56 in both Korean and Chinese flukes, and chromosomes were divided into two groups based on this size; consisting of 8 pairs of large and 20 pairs of small chromosomes. However, the karyotypes showed some differences between Korean and Chinese flukes. The karyotype of liver flukes from Korea consisted of three metacentric pairs, one meta-/submetacentric pair, 16 submetacentric pairs and eight subtelocentric pairs of chromosomes. On the other hand, liver flukes from China consisted of two metacentric pairs, two meta-/submetacentric pairs, 16 submetacentric pairs and eight subtelocentric pairs of chromosomes.  相似文献

Mitotic analyses of the brook trout (Salvelinus fontinalis) x arctic char (S. alpinus) hybrids (sparctic trout) revealed a mode of 2n = 82 with 18 metacentric and 64 acrocentric chromosomes. The brook trout had 2n = 84 with 16 metacentric chromosomes and the arctic char had 2n = 80 with 20 metacentric chromosomes; both species are derivatives of a single tetraploid event. Variable multivalent-like configurations that may be centromeric associations of bivalents were observed in C-banded pachytene figures of female sparctic trout. Metaphase I analyses of sparctic trout males indicated that two fusions of nonhomologous acrocentric chromosomes representing two duplicated chromosome sets must have occurred in the arctic char after its evolutionary divergence from the brook trout. A mode of seven tetravalent rods per cell suggests that preferential multivalent pairing occurs in the sparctic hybrid; metaphase I analyses of S. alpinus males revealed a mode of only five tetravalent rods per cell. The presence of multivalents implies that the arctic char, like the brook trout, is still undergoing diploidization. Cytochemical detection of the nucleolar organizer region (NOR) revealed intra- and interspecific as well as intraindividual variability in the numbers and types of chromosomes (metacentric or acrocentric) on which NORs appeared in arctic char and sparctic trout. Brook trout only had NORs on acrocentric chromosomes. This may indicate that different chromosomal fusions occurred in the evolution of brook trout from arctic char.  相似文献

We made a cytogenetic study of the fish Rhamdia quelen collected from the Bodoquena Plateau, an isolated national park region in Mato Grosso do Sul State, Brazil. The diploid number was 2n = 58, with 36 metacentric + 16 submetacentric + 6 subtelocentric chromosomes. We found one to three B chromosomes, which were metacentric and submetacentric and of medium size, showing both intra- and interindividual variation. The nucleolus organizer region (NOR) was located in the terminal region of the short arm of submetacentric pair 20. Staining with CMA3 fluorochrome revealed the NOR location, while there was no evidence of fluorescent staining with DAPI. C banding revealed heterochromatin mainly in the terminal regions of the chromosome arms, including the NOR pair. In addition, metacentric pair 2 showed three heterochromatic blocks in the terminal portions and in the pericentromeric region. The B chromosomes appeared euchromatic. The CB + CMA3 staining combination demonstrated only one chromosome pair with fluorescence, probably the NOR-bearing one, while CB + DAPI gave various fluorescent signals, including metacentric pair 2, indicating that these heterochromatic regions are AT-rich in this population of R. quelen. The R. quelen population in this isolated region of Brazil is chromosomally distinct from that of other populations that have been studied.  相似文献

Mitotic metaphase plates of a new species of the genus Macrostomum were studied using conventional Giemsa staining, Orcein staining and semi-thin sections stained after Richardson. Because of the ease in culturing it, this species has the potential to become the new model organism for developmental and evolutionary studies among the lower metazoa. The chromosome number was found to be 2 n  = 8. Reaching a relative length of 45.9% of the haploid genome size, the chromosomes of one pair were significantly larger than all other chromosomes. Of the smaller pairs, chromosomes of one pair were slightly larger (relative length of 21.3%) than the chromosomes of the remaining two pairs (each with a relative length of 16.4%). All chromosomes were metacentric (2 m  + 2 m  + 2 m  + 2 m ). For the first time, a diploid chromosome set of four pairs was described for the genus, as compared with previous studies showing predominantly 2 n  = 6 for 20 different Macrostomum species.  相似文献
