The response of several leaf gas exchange parameters were monitored with decreasing leaf water potential in Phaseolus vulgaris L. leaflets. These included photosynthesis, transpiration, CO2 compensation point, ribulose 1,5-diphosphate carboxylase activity, boundary layer plus stomatal, and mesophyll resistance to diffusion of CO2. Mesophyll resistance was calculated under two assumptions: (a) the CO2 concentration at the chloroplast was zero, and (b) it was equal to the CO2 compensation point.  相似文献

Rates of net photosynthesis and translocation, CO2 diffusive resistances, levels of carbohydrates, total protein, chlorophyll, and inorganic phosphate, and ribulose 1,5-diphosphate carboxylase activity were measured in soybean (Glycine max L. Merrill) leaves to ascertain the effect of altered assimilate demand. To increase assimilate demand, the pods, stems, and all but one leaf (the “source leaf”) of potted plants were completely shaded for 6 or 8 days and the responses of the illuminated source leaf were monitored. Rate of net photosynthesis in the source leaf of the shaded plants was found to increase curvilinearly to a maximum on the 8th day. The source leaf of the control plants (no sink shading) maintained a constant photosynthetic rate during this period. Vapor-phase resistance to CO2 diffusion did not vary with treatment, but mesophyll (liquid phase) resistance was significantly lower in the source leaf of the shaded plants.  相似文献

Wheat was grown in the field with different levels of nitrogenousfertilizer, and the rate of photosynthesis and the activityof ribulose 1,5-diphosphate carboxylase in the flag leaves determined.Additional nitrogen increased the dry-weight and leaf area ofthe plants, but did not increase grain yield; the rate of photosynthesisof the flag leaves was unchanged but the activity of ribulose1,5-diphosphate carboxylase increased. Clonal cuttings of Picea sitchensis (Bong.) Carr. were grown in a controlled environment and, after completion of shoot extension and maturation, subjected to a drying cycle. Photosynthesis and stomatal conductance were measured in situ using 14CO2 and a porometer, respectively. Shoot water potential was measured with the pressure chamber. Photosystem and carboxylase activities of chloroplast preparations were measured in vitro. A considerable fall in photosynthetic rate occurred at low water potential. This was associated with stomatal closure and a decrease in CO2 transfer or fixation processes in the mesophyll. Little change in activity of photosystem I, photosystem II, and ribulose 1,5-diphosphate carboxylase was detected during the drying cycle. Any decline in activity of the photosynthetic partial processes in vivo under severe water stress (Ψ < – 30 bar) was probably masked in vitro as a result of rehydration prior to assay.  相似文献

Rate of net CO2 assimilation by soil-grown soybean plants were studied over a range of relative leaf water contents at each of four levels of irradiance. There was a large interaction between light level and leaf water deficit on the rate of CO2 assimilation. The effect of leaf water deficit on assimilation became larger as irradiance increased. Both stomatal resistance to CO2 transport and mesophyll resistance to CO2 assimilation increased as leaf-water deficit increased. The increase in both resistance with changing leaf-water content was largest at high irradiance and became smaller as irradiance decreased. Relief of soil-moisture stress by watering induced large oscillations of CO2 assimilation, stomatal resistance, and mesophyll resistance. The oscillation of the mesophyll resistance occurred in the absence of changes in relative water content and appeared to be related to oscillations in leaf temperature. The observed increase in mesophyll resistance with decreasing leaf-water content under nonoscillative conditions may be caused by changes in leaf temperature rather than leaf water content.  相似文献

Maize plants (Zea mays L.) were subjected to soil flooding for 72, 96, and 120 h. A noticeable decrease in the rate of net photosynthesis (PN) and the activity of ribulose-1,5-bisphosphate carboxylase (RuBPC, EC 4.1.1.39) were observed. The values of intercellular CO2 concentrations (ci) increased in all flooded plants without significant changes in stomatal conductance (gs). The activity of phosphoenolpyruvate carboxylase (PEPC, EC 4.1.1.31) increased twofold 120 h after soil flooding. Flooding of maize plants led to a decrease in chlorophyll and protein levels and to slight increase of proline content. Flooded plants exhibited a large accumulation of leaf acidity. An increase in the values of some important parameters associated with oxidative stress, namely peroxides production, lipid peroxidation, and electrolyte leakage, confirmed the suggestion that root oxygen deficiency caused photooxidative damage in maize leaves.  相似文献

Sugar beet leaves (Beta vulgaris L.) infected with powdery mildew (Erysiphe polygoni D.C.) show declining rates of net photosynthesis as the disease develops; relative to healthy controls, reductions of 35, 70, and 75% were observed at 9, 16, and 22 days after inoculation, respectively. A leaf gas exchange procedure in which an air stream flowed through the leaf showed that mesophyll conductance declined in parallel with photosynthesis in mildew-infected leaves. Viscous flow conductance of diseased leaves also declined over the same period suggesting that stomatal aperture was reduced. From the magnitude and time course of disease effects on stomatal aperture and mesophyll conductance, it appears that the effects at the mesophyll level were primarily responsible for mediating the decline in net photosynthesis. Changes in mesophyll conductance were closely correlated with reduced activity of ribulose-1,5-bisphosphate carboxylase on a leaf area basis. This decrease could be attributed to a reduction in the concentration of the enzyme, a reduction which was greater than the reduction in total soluble protein. The quantum efficiency of light use was also decreased by the disease. Mildew-infected leaves had quantum yields that were reduced, relative to healthy leaves, by 17 and 22% at 14 and 18 days after inoculation, respectively.  相似文献

Mesophyll resistance to CO2 uptake was calculated from gas exchange data on intact leaves of 12 species of woody plants. Plants studied were native to habitats ranging from streamsides to deserts. Gas exchange measurements were made at light saturation and constant temperature to eliminate possible effects of light and temperature on estimates of mesophyll resistance. Cuticular transpiration was measured and used in calculation of stomatal resistances from whole leaf transpiration rates. In all species examined, an increase in mesophyll resistance was observed as leaves dried. The increase in mesophyll resistance in all cases occurred at the same water potential as the initial decline in net photosynthesis, and was accompanied by an increase in stomatal resistance.  相似文献

Photosynthesis in C3 plants is CO2 limited and therefore any increase in Rubisco carboxylation substrate may increase net CO2 fixation, unless plants experience acclimation or other limitations. These aspects are largely unexplored in grapevine. Photosynthesis analysis was used to assess the stomatal, mesophyll, photochemical and biochemical contributions to the decreasing photosynthesis observed in Tempranillo grapevines (Vitis vinifera) from veraison to ripeness, modulated by CO2, temperature and water availability. Photosynthesis and photosystem II photochemistry decreased from veraison to ripeness. The elevated CO2 and temperature increased photosynthesis, but transiently, in both well irrigated (WI) and water‐stressed plants. Photosynthetic rates were maxima 1 week after the start of elevated CO2 and temperature treatments, but differences with treatments of ambient conditions disappeared with time. There were not marked changes in leaf water status, leaf chlorophyll or leaf protein that could limit photosynthesis at ripeness. Leaf total soluble sugars remained at ripeness as high as 2 weeks after the start of treatments. On the other hand, and as expected, CO2 diffusional limitations impaired photosynthesis in grapevine plants grown under water scarcity, stomatal and mesophyll conductances to CO2 decreased and in turn low chloroplastic CO2 concentrations limited photosynthetic CO2 fixation. In summary, photochemistry and photosynthesis from veraison to ripeness in Tempranillo grapevine were dominated by a developmental‐related decreasing trend that was only transiently influenced by elevated CO2 concentrations.  相似文献

This work aimed to evaluate the effects of lower water levels on leaf intercellular spaces and to assess their relations with the gas exchange, anatomy, and growth of Sorghum bicolor. Experiments were conducted in a greenhouse, in which plants were subjected to three water conditions (ten replicates, n = 30): well-irrigated, decreased irrigation, and limited irrigation. Lower water levels had no significant effect on the growth of S. bicolor but increased the biomass of the roots. Moreover, the number of leaves, leaf area, and leaf size as well as the chlorophyll content were not affected by lower water levels, and no significant changes were detected for whole plant photosynthesis, transpiration, or stomatal conductance. The water content of the plants and the water potential remained unchanged. However, compared with other treatments, the decreased irrigation decreased water loss and increased the water retention. Lower water levels increased the intercellular CO2 percentage, mesophyll area, and proportion of stomatal cavities and promoted minor changes in leaf tissue and stomatal traits. The increased stomatal cavities provided higher CO2 uptake and prevented excessive water loss. Thus, modifications to the intercellular spaces promoted conditions to avoid excessive water loss while concurrently improving CO2 uptake, which are important traits for drought-tolerant plants.

The relationship between loss of ribulose-1,5-bisphosphate carboxylase (RuBPCase) and the decline in photosynthesis during the senescence of barley primary leaves was assessed. Loss of RuBPCase accounted for about 85% of the decrease in soluble protein. RuBPCase was highly correlated with in vitro RuBPCase activity (r = 0.95) and gross photosynthesis (r = 0.96). However, the rate of photosynthesis per milligram RuBPCase increased during the early stages of leaf senescence. The concentration of nonreducing sugars was negatively correlated (1% level) with photosynthesis. Free α-amino N, in contrast to nonreducing sugars, declined markedly during senescence. A decrease in chlorophyll and an increase in in vitro protease activity was observed, but these changes did not appear to be closely related to the decline in photosynthesis and RuBPCase. Mesophyll resistance increased at the same rate that photosynthesis and RuBPCase declined. Stomatal resistance increased more rapidly than mesophyll resistance and accounted for about 24% of the total increase in resistance to CO2 diffusion. The concentration of CO2 in the intercellular air spaces decreased during the last stage of senescence. Although loss of RuBPCase probably is the primary event responsible for the decline in photosynthesis during leaf senescence, other factors such as in vivo regulation and stomatal aperture must also be considered.  相似文献

The seasonal change in leaf water potential and its components, stomatal resistance, specific leaf weight, photosynthesis rate, the activities of ribulose-1,5-bisphosphate carboxylase and nitrate reductase, and soluble proteins were measured in flag leaves (ninth from base in position), seventh and fifth leaves of wheat Triticum aestivum L. cv Kalyansona. Flag leaves had a lower water and solute potential and lower or equal turgor pressure than seventh and fifth leaves. These differences were found to be independent of environment. The rate of photosynthesis and nitrate reductase activity were always lower in fifth and seventh leaves than in flag leaf. The photosynthetic efficiency in flag leaves appeared to be associated with lower stomatal resistance and higher specific leaf weight. The relations between leaf water potential and relative water content showed a change with leaf position. This change possibly allows flag leaf to maintain its functional efficiency despite its lower water potential.  相似文献

CO2 and water vapor exchange studies of intact plants of black needle rush (Juncus roemerianus Scheele) were conducted in an undisturbed marsh community on Sapelo Island, Georgia. The seasonal patterns of the light and temperature responses of net photosynthesis, transpiration, leaf diffusive conductance, water-use efficiency and respiration were determined five times over the year. Internal resistances to CO2 uptake were also evaluated. Net photosynthesis was highest in early spring, but declined only slightly through the year. A distinct and moderate temperature optimum of net photosynthesis was observed with decreasing rates above 30 C. Leaf conductances to water vapor were similar at all seasons and were high at cooler temperatures and decreased with increasing temperature. Transpiration was relatively high and constant during all seasons. The water-use efficiency of photosynthesis was high below 25 C, but decreased sharply above that temperature. Dark respiration was relatively low. Seasonal changes reflected changes in leaf density. Decreasing stomatal conductances and increasing respiration rates reduced net photosynthesis at higher temperatures. The stomatal resistance increased and internal resistances to CO2 uptake decreased over the year, but the total resistance remained constant. The internal resistance to CO2 uptake was consistently higher than the stomatal resistance. These seasonal response patterns show that J. roemerianus is well adapted to the seasonal changes in ambient temperature and irradiance and other microenvironmental factors in the high marsh. These physiological characteristics permit this C3 species to maintain a high productivity in a seasonally hot and stressful environment.  相似文献

Supra-optimal levels of zinc in primary leaves of Phaseolus vulgaris increased the CO2 compensation point and inhibited net photosynthesis. Leaf morphology was modified: mesophyll intercellular area, stomatal slit length and interstomatal distance were reduced, but stomatal density increased. Internal and stomatal conductances to CO2 diffusion decreased. These changes are discussed in relation to the observed effects on leaf gas exchange and to the previously reported inhibition of different photosynthetic and photorespiratory enzymes.  相似文献

The effects of light, temperature, and salinity on growth, net CO2 exchange and leaf anatomy of Distichlis spicata were investigated in controlled environment chambers. When plants were grown at low light, growth rates were significantly reduced by high substrate salinity or low temperature. However, when plants were grown at high light, growth rates were not significantly affected by temperature or salinity. The capacity for high light to overcome depressed growth at high salinity cannot be explained completely by rates of net photosynthesis, since high salinity caused decreases in net photosynthesis at all environmental conditions. This salinity-induced decrease in net photosynthesis was caused largely by stomatal closure, although plants grown at low temperature and low light showed significant increases in internal leaf resistance to CO2 exchange. Increased salinity resulted in generally thicker leaves with lower stomatal density but no significant differences in the ratio of mesophyll cell surface area to leaf area. Salinity and light during growth did not significantly affect rates of dark respiration. The mechanisms by which Distichlis spicata tolerates salt appear to be closely coulpled to the utilization of light energy. Salt-induced leaf succulence is of questionable importance to gas exchange at high salinity in this C4 species.  相似文献

Nutrients such as phosphorus may exert a major control over plant response to rising atmospheric carbon dioxide concentration (CO2), which is projected to double by the end of the 21st century. Elevated CO2 may overcome the diffusional limitations to photosynthesis posed by stomata and mesophyll and alter the photo-biochemical limitations resulting from phosphorus deficiency. To evaluate these ideas, cotton (Gossypium hirsutum) was grown in controlled environment growth chambers with three levels of phosphate (Pi) supply (0.2, 0.05 and 0.01 mM) and two levels of CO2 concentration (ambient 400 and elevated 800 μmol mol−1) under optimum temperature and irrigation. Phosphate deficiency drastically inhibited photosynthetic characteristics and decreased cotton growth for both CO2 treatments. Under Pi stress, an apparent limitation to the photosynthetic potential was evident by CO2 diffusion through stomata and mesophyll, impairment of photosystem functioning and inhibition of biochemical process including the carboxylation efficiency of ribulose-1,5-bisphosphate carboxylase/oxyganase and the rate of ribulose-1,5-bisphosphate regeneration. The diffusional limitation posed by mesophyll was up to 58% greater than the limitation due to stomatal conductance (gs) under Pi stress. As expected, elevated CO2 reduced these diffusional limitations to photosynthesis across Pi levels; however, it failed to reduce the photo-biochemical limitations to photosynthesis in phosphorus deficient plants. Acclimation/down regulation of photosynthetic capacity was evident under elevated CO2 across Pi treatments. Despite a decrease in phosphorus, nitrogen and chlorophyll concentrations in leaf tissue and reduced stomatal conductance at elevated CO2, the rate of photosynthesis per unit leaf area when measured at the growth CO2 concentration tended to be higher for all except the lowest Pi treatment. Nevertheless, plant biomass increased at elevated CO2 across Pi nutrition with taller plants, increased leaf number and larger leaf area.  相似文献

The effect of salinity on C(4) photosynthesis was examined in leaves of maize, a NADP-malic enzyme (NADP-ME) type C(4) species. Potted plants with the fourth leaf blade fully developed were treated with 3% NaCl solution for 5d. Under salt treatment, the activities of pyruvate orthophosphate dikinase (PPDK), phosphoenolpyruvate carboxylase (PEPCase), NADP-dependent malate dehydrogenase (NADP-MDH) and NAD-dependent malate dehydrogenase (NAD-MDH), which are derived mainly from mesophyll cells, increased, whereas those of NADP-ME and ribulose-1,5-bisphosphate carboxylase, which are derived mainly from bundle sheath cells (BSCs), decreased. Immunocytochemical studies by electron microscopy revealed that PPDK protein increased, while the content of ribulose-1,5-bisphosphate carboxylase/oxygenase protein decreased under salinity. In salt-treated plants, the photosynthetic metabolites malate, pyruvate and starch decreased by 40, 89 and 81%, respectively. Gas-exchange analysis revealed that the net photosynthetic rate, the transpiration rate, stomatal conductance (g(s)) and the intercellular CO(2) concentration decreased strongly in salt-treated plants. The carbon isotope ratio (δ(13)C) in these plants was significantly lower than that in control. These findings suggest that the decrease in photosynthetic metabolites under salinity was induced by a reduction in gas-exchange. Moreover, in addition to the decrease in g(s), the decrease in enzyme activities in BSCs was responsible for the decline of C(4) photosynthesis. The increase of PPDK, PEPCase, NADP-MDH, and NAD-MDH activities and the decrease of NADP-ME activity are interpreted as adaptation responses to salinity.  相似文献

Leaf gas exchange parameters and the content of ribulose‐1,5‐bisphosphate carboxylase/oxygenase (Rubisco) in the leaves of two 2‐year‐old aspen (Populus tremuloides Michx.) clones (no. 216, ozone tolerant and no. 259, ozone sensitive) were determined to estimate the relative stomatal and mesophyll limitations to photosynthesis and to determine how these limitations were altered by exposure to elevated CO2 and/or O3. The plants were exposed either to ambient air (control), elevated CO2 (560 p.p.m.) elevated O3 (55 p.p.b.) or a mixture of elevated CO2 and O3 in a free air CO2 enrichment (FACE) facility located near Rhinelander, Wisconsin, USA. Light‐saturated photosynthesis and stomatal conductance were measured in all leaves of the current terminal and of two lateral branches (one from the upper and one from the lower canopy) to detect possible age‐related variation in relative stomatal limitation (leaf age is described as a function of leaf plastochron index). Photosynthesis was increased by elevated CO2 and decreased by O3 at both control and elevated CO2. The relative stomatal limitation to photosynthesis (ls) was in both clones about 10% under control and elevated O3. Exposure to elevated CO2 + O3 in both clones and to elevated CO2 in clone 259, decreased ls even further – to about 5%. The corresponding changes in Rubisco content and the stability of Ci/Ca ratio suggest that the changes in photosynthesis in response to elevated CO2 and O3 were primarily triggered by altered mesophyll processes in the two aspen clones of contrasting O3 tolerance. The changes in stomatal conductance seem to be a secondary response, maintaining stable Ci under the given treatment, that indicates close coupling between stomatal and mesophyll processes.  相似文献

Photosynthetic gas exchange, vegetative growth, water relations and fluorescence parameters as well as leaf anatomical characteristics were investigated on young plants of two Olea europaea L. cultivars (Chemlali and Zalmati), submitted to contrasting water availability regimes. Two-year-old olive trees, grown in pots in greenhouse, were not watered for 2 months. Relative growth rate (RGR), leaf water potential (ΨLW) and the leaf relative water content (LWC) of the two cultivars decreased with increasing water stress. Zalmati showed higher values of RGR and LWC and lower decreased values of ΨLW than Chemlali, in response to water deficit, particularly during severe drought stress. Water stress also caused a marked decline on photosynthetic capacity and chlorophyll fluorescence. The net photosynthetic rate, stomatal conductance, transpiration rate, the maximal photochemical efficiency of PSII (F v/F m) and the intrinsic efficiency of open PSII reaction centres (F′ v/F′ m) decreased as drought stress developed. In addition, drought conditions, reduced leaf chlorophyll and carotenoids contents especially at severe water stress. However, Zalmati plants were the less affected when compared with Chemlali. In both cultivars, stomatal control was the major factor affecting photosynthesis under moderate drought stress. At severe drought-stress levels, the non-stomatal component of photosynthesis is inhibited and inactivation of the photosystem II occurs. Leaf anatomical parameters show that drought stress resulted in an increase of the upper epidermis and palisade mesophyll thickness as well as an increase of the stomata and trichomes density. These changes were more characteristic in cv. ‘Zalmati’. Zalmati leaves also revealed lower specific leaf area and had higher density of foliar tissue. From the behaviour of Zalmati plants, with a smaller reduction in relative growth rate, net assimilation rate and chlorophyll fluorescence parameters, and with a thicker palisade parenchyma, and a higher stomatal and trichome density, we consider this cultivar more drought-tolerant than cv. Chemlali and therefore, very promising for cultivation in arid areas.  相似文献
