The effects of ethephon on stomatal resistance, water potential, osmotic potential, turgor potential, and ethylene production were determined on leaves of a drought-resistant (KS 65) and a drought-sensitive (IA 25) genotype of sorghum [Sorghum bicolor (L.) Moench] grown under wellwatered or drought-stressed conditions. With both sufficient and limited water supply, ethephon had no effect on the adaxial, abaxial, or total stomatal resistance of either genotype. For both water treatments, the adaxial stomatal resistance of the drought-sensitive genotype was higher than that of the drought-resistant genotype. Ethephon increased the amount of ethylene produced by the plants under both levels of water. For plants with sufficient water, water potentials of both genotypes were lowered by ethephon. Ethephon had no effect on the water potentials under drought or on the osmotic potentials under either water regime. With drought, the turgor potential of the drought-sensitive genotype, but not that of the drought-resistant, was increased by ethephon.

Leaves of drought-resistant and drought-sensitive genotypes of maize (Zea mays L.) were sprayed with ethephon to determine its effect on water and osmotic potentials. With both sufficient and limited water supplies, ethephon decreased the water potential, but not the osmotic potential, of the drought-sensitive maize. It had no effect on the water and osmotic potentials of the drought-resistant maize under either water regime. The results showed that the response of a genotype of maize to ethephon depends upon its susceptibility to drought.

Genotypes of sorghum [Sorghum bicolor (L.) Moench] vary in drought resistance. Yet it is not known if their hydraulic resistances vary. The objective of this study was to determine if the hydraulic resistance of a drought-resistant sorghum was the same as that of a drought-sensitive sorghum. Leaf water and osmotic potentials were measured daily, during a 14-d period, in leaves of a drought-resistant (‘KS9’) and a drought-sensitive (‘IA25’) sorghum, which had the roots in pots with a commercial potting soil that was either well watered or allowed to dry. Soil water potential, adaxial stomatal resistance, and transpiration rate were determined daily. Hydraulic resistance of the plants was calculated from the slope of the line relating soil water potential minus leaf water potential versus transpiration rate. When the soil was not watered, the drought-sensitive sorghum had a water potential that averaged −0.50 MPa lower and an osmotic potential that averaged −0.57 MPa lower, but a similar adaxial stomatal resistance (1.19 s mm−1), compared with the drought-resistant sorghum. Seven days after the beginning of the experiment, the water potential of the soil with the drought-sensitive sorghum was −0.25 MPa lower than that of the soil with the drought-resistant sorghum. With the water-limited conditions, the drought-sensitive sorghum depleted the soil-water reserve more quickly and died 2 d before the drought-resistant sorghum. Under well watered conditions, the two sorghums had similar water potentials (−1.64 MPa), osmotic potentials (−2.83 MPa), and adaxial stomatal resistances (0.78 s mm−1). The calculated hydraulic resistance of the two sorghums did not differ and averaged 3.4 × 107 MPa s m−1. The results suggested that the variation in susceptibility to drought between the two genotypes was due to differences in rate of soil-water extraction. Water and osmotic potentials were measured in leaves of a drought-sensitive ('Ponca') and a drought-resistant ('KanKing') cultivar of winter wheat ( Triticum aestivum L . em. Thell.) to determine if the potentials of the drought-sensitive cultivar could be made similar to those of the drought-resistant cultivar through application of abscisic acid (ABA). Stomatal resistance was also measured. Plants were sprayed with ABA and grown in soil, which was watered or allowed to dry. In well-watered plants, ABA closed the stomata of both cultivars. Stomatal resistance of plants grown without added water and with ABA was less than that of plants grown without added water and without ABA. Under ample water supply, ABA decreased water and osmotic potentials of the drought-sensitive cultivar (Ponca), but had no effect on these potentials in the drought-resistant cultivar (KanKing). Under water-deprived conditions, ABA increased water and osmotic potentials of Ponca, but did not change these potentials in KanKing. The overall effect of ABA was to decrease the differences in the water and osmotic potentials between the two cultivars.

The tepary bean ( Phaseolus acutifolius Gray var. latifolius ), a drought resistant species, was compared under water stress conditions with the more drought susceptible P. vulgaris L. cvs Pinto and White Half Runner (WHR). In order to better understand the basis for the superior drought resistance of tepary, this study was designed to determine the relationships among leaf water potential, osmotic potential, turgor potential, and relative water content (RWC).
Plants were prestressed by withholding irrigation water. These stress pretreatments changed the relation between leaf water potential and relative water content of both species so that prestressed plants had lower water potentials than controls at the same leaf RWC. Tepary had lower water potentials at given RWC levels than Pinto or WHR; this can account for part of the superior resistance of tepary. In all genotypes, prestressed plants maintained osmotic potentials approximately 0.2 MPa lower than controls. Tepary reached osmotic potentials that were significantly lower (0.15 to 0.25 MPa) than Pinto or WHR. Both control and prestressed tepary plants had 0.05 to 0.25 MPa more turgor than Pinto or WHR at RWC values between 65 and 80%. Both prestressed and control tepary plants had greater elasticity (a lower elastic modulus) than Pinto or WHR. This greater turgor of tepary at low RWC values could be caused by several factors including greater tissue elasticity, active accumulation of solutes, or greater solute concentration.
Tepary had significantly lower osmotic potentials than the P. vulgaris cultivars, but there was little difference in osmotic potential between Pinto and WHR. Knowledge of differences in osmotic and turgor potentials among and within species could be useful in breeding for drought resistance in Phaseolus.

The water permeability of the leaves of three deciduous plants (Acer campestre, Fagus sylvatica, Quercus petraea) and two evergreen plants (Hedera helix, Ilex aquifolium) was analysed in order to assess its role as a mechanism of drought resistance. Cuticular permeances were determined by measurement of the water loss through adaxial, astomatous leaf surfaces. Minimum conductances after complete stomatal closure were obtained by leaf drying curves. The comparison of the water permeabilities determined with these two experimental systems revealed good agreement in the case of Acer, Fagus, Quercus, and Ilex. For Hedera the minimum conductance was 3-fold higher than the cuticular permeance indicating a significant contribution of residual stomatal transpiration. The leaf water potential was measured as a function of water content and analysed by pressure-volume curves. The influence of water potential as a component of the driving force for transpirational water loss was assessed in order to identify modifications of the cuticular barrier by the leaf water content. The ecophysiological meaning of the water relations parameters describing transpiration under drought conditions (cuticular transpiration, minimum transpiration, residual stomatal transpiration, effect of leaf water content on transpiration) and the water relations parameters derived from pressure-volume curves (osmotic potential at full saturation, turgor loss point, bulk modulus of elasticity) are discussed with regard to adaptations for drought resistance.

Water potential, osmotic potential and turgor measurements obtained by using a cell pressure probe together with a nanoliter osmometer were compared with measurements obtained with an isopiestic psychrometer. Both types of measurements were conducted in the mature region of Tradescantia virginiana L. leaves under non-transpiring conditions in the dark, and gave similar values of all potentials. This finding indicates that the pressure probe and the osmometer provide accurate measurements of turgor, osmotic potentials and water potentials. Because the pressure probe does not require long equilibration times and can measure turgor of single cells in intact plants, the pressure probe together with the osmometer was used to determine in-situ cell water potentials, osmotic potentials and turgor of epidermal and mesophyll cells of transpiring leaves as functions of stomatal aperture and xylem water potential. When the xylem water potential was-0.1 MPa, the stomatal aperture was at its maximum, but turgor of both epidermal and mesophyll cells was relatively low. As the xylem water potential decreased, the stomatal aperture became gradually smaller, whereas turgor of both epidermal and mesophyll cells first increased and afterward decreased. Water potentials of the mesophyll cells were always lower than those of the epidermal cells. These findings indicate that evaporation of water is mainly occurring from mesophyll cells and that peristomatal transpiration could be less important than it has been proposed previously, although peristomatal transpiration may be directly related to regulation of turgor in the guard cells.

Apple trees are very drought tolerant, having the capability to grow and carry on photosynthesis even at low water potentials. Much of the tolerance is due to the ability of apple leaves to maintain turgor potentials at levels conducive to growth and stomatal opening. Diurnally, leaf turgor is maintained through decreases in osmotic potentials (due to active solute accumulation), osmotic adjustment, or to concentration of solutes via tissue water loss. These two processes combined may decrease osmotic potentials by as much as 1.65 MPn during the day. Seasonally, osmotic potentials remain fairly constant, but leaf elasticity increases, allowing growth to continue and stomata to remain open us water and turgor potentials become progressively lower. Release of stored water from plant tissues to the transpiration stream is another means of preventing water potentials from reaching critical values for stomatal closure. A combination of a number of these physiological adaptations may account for much of the drought tolerance in apple trees.

In order to ascertain the stomatal and photosynthetic responses of mustard to ethylene under varying N availability, photosynthetic characteristics of mustard grown with optimal (80 mg N kg(-1) soil) or low (40 mg N kg(-1) soil) N were studied after the application of an ethylene-releasing compound, ethephon (2-chloroethyl phosphonic acid) at 40 days after sowing (DAS). The availability of N influenced ethylene evolution and affected stomatal conductance and photosynthesis. The effect of ethylene was smaller under deficient N where plants contained higher glucose (Glc) sensitivity, despite high ethylene evolution even in the absence of ethephon, potentially because the plants were less sensitive to ethylene per se. Ethephon application at each level of N increased ethylene and decreased Glc sensitivity, which increased photosynthesis via its effect on the photosynthetic machinery and effects on stomatal conductance. Plants grown with sufficient-N and treated with 200 μl l(-1) ethephon exhibited optimal ethylene, the greatest stomatal conductance and photosynthesis, and growth. These plants made maximum use of available N and exhibited the highest nitrogen-use efficiency (NUE).

The relationships between the vulnerability of stem xylem to cavitation, stomatal conductance, stomatal density, and leaf and stem water potential were examined in six hybrid poplar (P38P38, Walker, Okanese, Northwest, Assiniboine and Berlin) and balsam poplar (Populus balsamifera) clones. Stem xylem cavitation resistance was examined with the Cavitron technique in well-watered plants grown in the greenhouse. To investigate stomatal responses to drought, plants were subjected to drought stress by withholding watering for 5 (mild drought) and 7 (severe drought) days and to stress recovery by rewatering severely stressed plants for 30 min and 2 days. The clones varied in stomatal sensitivity to drought and vulnerability to stem xylem cavitation. P38P38 reduced stomatal conductance in response to mild stress while the balsam poplar clone maintained high leaf stomatal conductance under more severe drought stress conditions. Differences between the severely stressed clones were also observed in leaf water potentials with no or relatively small decreases in Assiniboine, P38P38, Okanese and Walker. Vulnerability to drought-induced stem xylem embolism revealed that balsam poplar and Northwest clones reached loss of conductivity at lower stem water potentials compared with the remaining clones. There was a strong link between stem xylem resistance to cavitation and stomatal responsiveness to drought stress in balsam poplar and P38P38. However, the differences in stomatal responsiveness to mild drought suggest that other drought-resistant strategies may also play a key role in some clones of poplars exposed to drought stress.

The influence of water shortage and ethylene (ethephon) application on ear fertility and tillering of barley plants were compared. In both cases, highest sensitivity was observed during jointing and pre-anthesis (Feekes 7–9). The ear initial with the surrounding tissue was identified as the site of ethylene action. Treating this region of barley plants with AVG before wilting partly prevented drought effects. These results, in connection with rising ethylene values in ear-bearing stem segments of wilting barley plants (more obvious in increasing ACC and MACC levels), especially in the drought-sensitive stages, favors a role for ethylene in the development of cereal plants under drought.

The objective of this research was to study the effect of drought on nutrient content and leaf water status in alfalfa (Medicago sativa L. cv Aragón) plants inoculated with a mycorrhizal fungus and/or Rhizobium compared with noninoculated ones. The four treatments were: a) plants inoculated with Glomus fasciculatum and Rhizobium meliloti 102 F51 strain, (MR); b) plants inoculated with R. meliloti only (R); c) plants with G. fasciculatum only (M); and d) noninoculated plants (N). Nonmycorrhizal plants were supplemented with phosphorus and nonnodulated ones with nitrogen to achieve similar size and nutrient content in all treatments. Plants were drought stressed using two cycles of moisture stress and recovery. The components of total leaf water potential (osmotic and pressure potentials at full turgor), percentage of apoplastic water volume and the bulk modulus of elasticity of leaf tissue were determined. Macronutrient (N, P, K, Ca, S and Mg) and micronutrient (Co, Mo, Zn, Mn, Cu, Na, Fe and B) content per plant were also measured. Leaves of N and R plants had decreased osmotic potentials and increased pressure potentials at full turgor, with no changes either in the bulk modulus of elasticity or the percentage of apoplastic water upon drought conditions. By contrast, M and MR leaves did not vary in osmotic and turgor potentials under drought stress but had increased apoplastic water volume and cell elasticity (lowering bulk modulus). Drought stress decreased nutrient content of leaves and roots of noninoculated plants. R plants showed a decrease in nutrient content of leaves but maintained some micronutrients in roots. Leaves of M plants were similar in content of nutrients to N plants. However, roots of M and MR plants had significantly lower nutrient content. Results indicate an enhancement of nutrient content in mycorrhizal alfalfa plants during drought that affected leaf water relations during drought stress.

Based upon two different research studies in the mediterranean regions of France and Turkey, drought resistance strategies were investigated in a broad group of species. The diurnal and seasonal patterns of the water relations of different lifeforms from the thermo-mediterranean to submediterranean lifezones were compared. Three sites near Montpellier, in Southern France, and five sites near Antalya, Turkey were used for this comparison. Xylem pressure potential and relative stomatal aperture were the key water relations parameters collected in France while these parameters as well as osmotic potential and leaf conductance were studied in Turkey. From the 26 different study species investigated in France, 7 distinct types of stomatal control were observed, with the deciduous lifeforms showing the least control, the sclerophyllous and coniferous evergreens the greatest control and the malacophyllous shrublets intermediate levels of control. Predawn water potential values provided a means of classifying species according to their temporal and spatial utilization of site water reserves. The comparison of turgor potentials (difference between water and osmotic potentials) gave an insight into leaf adaptations to site moisture. Species with high predawn water potentials generally maintain positive turgor even at midday during the summer, whereas species with low predawn values were frequently at zero turgor even at predawn. Phlomis grandiflora was the most extreme species with mid-summer predawns and midday water potentials of –6 MPa and osmotic potentials never more negative than –2.4 MPa.

The sequence of physiological events during drought strongly impacts plants' overall performance. Here, we synthesized the global data of stomatal and hydraulic traits in leaves and stems of 202 woody species to evaluate variations in the water potentials for key physiological events and their sequence along the climatic gradient. We found that the seasonal minimum water potential, turgor loss point, stomatal closure point, and leaf and stem xylem vulnerability to embolism were intercorrelated and decreased with aridity, indicating that water stress drives trait co-selection. In xeric regions, the seasonal minimum water potential occurred at lower water potential than turgor loss point, and the subsequent stomatal closure delayed embolism formation. In mesic regions, however, the seasonal minimum water potential did not pose a threat to the physiological functions, and stomatal closure occurred even at slightly more negative water potential than embolism. Our study demonstrates that the sequence of water potentials for physiological dysfunctions of woody plants varies with aridity, that is, xeric species adopt a more conservative sequence to prevent severe tissue damage through tighter stomatal regulation (isohydric strategy) and higher embolism resistance, while mesic species adopt a riskier sequence via looser stomatal regulation (anisohydric strategy) to maximize carbon uptake at the cost of hydraulic safety. Integrating both aridity-dependent sequence of water potentials for physiological dysfunctions and gap between these key traits into the hydraulic framework of process-based vegetation models would improve the prediction of woody plants' responses to drought under global climate change.

Seasonal Changes in the Cytokinin Content of Ginkgo biloba Leaves
Young growth-chamber-grown cotton plants were subjected to a series of eight periods of soil water stress, which served as a preconditioning treatment. After preconditioning, water was withheld and changes in the stomatal resistance and leaf water potential were determined and compared with similar well watered control plants. The stomatal response of stress preconditioned plants adjusted such that the diffusion resistance of the lower surface of the leaf did not reach a value greater than 20 s cm?1 until the leaf water potential dropped 14 bars below that required to reach the same resistance on previously unstressed plants. The resistance—leaf water potential relation for the adaxial surface was unaltered by the preconditioning treatment. Adjustment of the osmotic potential of the guard cells on the abaxial surface provides at least a partial explanation of this change in response. The lack of adjustment of stomatal response on the adaxial surface of the leaves was correlated with a lack of adjustment in osmotic potential of guard cells on that surface.

Critical Water Potential for Stomatal Closure in Sitka Spruce
Steady state rates of net photosynthesis and stomatal conductance at high water potentials were measured under controlled conditions in a leaf chamber on Sitka spruce [Picea sitchensis (Bong.) Carr.] shoots detached from the forest canopy or on seedlings. The water supply to the seedlings was terminated by excision and the shoot water potential (or critical water potential) and osmotic potential at the onset of stomatal closure measured. The turgor potential was calculated. The initial osmotic potential before insertion of the shoot into the chamber was also measured. Shoot water potential and osmotic potential at stomatal closure, and initial osmotic potential were significantly higher (less negative) in foliage from the lowest level in the canopy compared with foliage in the upper canopy, and higher in shoots of seedlings transferred to low light than in those at high light. Critical water potential also varied with season, being higher in July than in October and November. In all except one instance, turgor potential at the onset of stomatal closure was negative, possibly because of dilution of the cell sap by the extracellular water during the estimate of osmotic potential. Over all the experiments variation in critical water potential was correlated with variation in critical osmotic potential and, to a lesser extent, the initial osmotic potential. However, turgor potential at the critical potential varied from +0.6 to -4.6 bar. This suggests that difference in turgor between the guard cells and subsidiary cells, which controls stomatal aperture, is only loosely coupled with the bulk leaf turgor and hence that bulk leaf turgor is not a good index of the turbor relations of the guard cells.

The impact of osmotic stress on growth, physiology, and metabolism of winter oilseed rape(Brassica napus L.)was investigated by detailed analysis of biomass traits, hormone metabolites and osmolytes in two genetically unrelated drought-tolerant genotypes and two unrelated drought-sensitive genotypes. Seedlings were grown in vitro under controlled conditions and osmotic stress was simulated by applying a gradual treatment with polyethylene glycol(PEG6000), followed by hypo-osmotic treatment of variants used for metabolite determination. The results provide a basis for the identification of reliable selection criteria for drought resistance in oilseed rape. The in vitro cultivation system established during this study enabled effective discrimination of early osmotic stress responses between drought-resistant and -susceptible oilseed rape genotypes that also show large differences in relative seed yield under drought conditions in the field. Clear physiological and metabolic differences were observed between the drought-resistant and drought-sensitive genotypes, suggesting that osmotic adjustment is a key component of drought response in oilseed rape. Unexpected ly, however, the drought-resistant genotypes did not show typical hormonal adjustment and osmolyte accumulation suggesting that they possess alternative physiological mech anisms enabling avoidance of stress symptoms.
