The freshwater rotifer, Brachionus calyciflorus is one of the live food organisms used for the mass production of larval fish. In this study possibility of obtaining high density cultures of the freshwater rotifer B. calyciflorus were investigated. The two culture systems used differed in their air and dissolved oxygen supplies using three temperatures in each case: 24, 28 and 32 °C. Rotifers were batch-cultured using 5 l-vessels and fed with the freshwater Chlorella. The growth rate of rotifers significantly increased with an increase in temperature. The maximum density of the rotifers with air-supply at 24 °C, 6500 ind. ml–1, was significantly lower than those cultured at 28 and 32 °C, i.e. 8600 and 8100 ind. ml–1, respectively. Dissolved oxygen levels decreased with time and ranged from 0.8 to 1.4 mg l–1 when the density of freshwater rotifer was the highest at each temperature. The highest density (19200 ind. ml–1) of freshwater rotifer was obtained in cultures with a supply of oxygen at 28 °C. Densities of 13500 and 17200 ind. ml–1 were found at 24 and 32 °C, respectively. Levels of NH3-N increased with time and a dramatic increase of NH3-N was observed at high temperatures. Levels of NH3-N at 24, 28 and 32 °C were 13.2, 18.5 and 24.5 mg l–1, respectively. These levels coincided with the highest rotifer density at each of the three temperatures. When rotifers were cultured with an oxygen-supply and pH was adjusted to 7, the maximum density of rotifer reached 33500 ind. ml–1 at 32 °C . These results suggested that high density culture of freshwater rotifer, B. calyciflorus could be achieved under optimal conditions with DO value of exceeding 5 mg l–1 and NH3-N values of lower than 12.0 mg l–1.  相似文献

The oxygen consumption rate (OCR) is a cumulative index of metabolic losses during aerobic metabolism. The generalized relationship of oxygen consumption rate (R, n1 O2 ind–1 h–1) and dry body mass (M, µg) for rotifers is described by the equation: R = 9.15M0.716. The level of rotifer metabolism is slightly lower than that of multicellular poikilothermic animals. Differences of OCR values in ontogenesis are substantial. Embryos and senile individuals are characterized by minimal OCR values. The OCR of oviparous females in the beginning of reproduction exceeds 2–3 times OCR values of juveniles. Differences in oxygen consumption intensity (OCI) are not so essential. OCR depends on food concentration. An increase of food concentration from 1.4 to 7.0 µg dry mass m1–1 resulted in Brachionus calyciflorus in an OCR escalation of 2.5 times at 30°C, and 0.5 times at 25°C. Maximal OCR values occur at food concentration close to the saturation concentration for population growth rate. An exponential equation is adequate to describe R-t dependence for animals, long-term adapted to different constant temperatures (2 < Q10 < 3). Acclimation effects observed during sharp temperature changes are determined by peculiarities of compensation reactions in species and separate populations. The formation of a zone of relative temperature independence of OCR (Q10 1) at fluctuating temperature is observed. It is necessary to study enzymatic activities parallel to OCR and OCI measurements.  相似文献

Ammonia-nitrogen excretion rates were measured in natural summer and cultured populations of Daphnia pulex from Silver Lake, Clay County, Minnesota, USA during 1973. The mean rate of ammonia-nitrogen excretion for the summer populations was 0.20 µg N animal–1 day–1 or 5.11 µg N mg–1 dry body weight day–1 (N = 80) measured at 15°, 20°, and 25°C. These rates appear to be temperature and weight dependent, but they are probably affected by factors other than temperature and dry body weight. Ammonia-nitrogen excretion rates of Daphnia pulex cultured on Chlamydomonas reinhardi yielded the following relationship with temperature: Log10E = (0.061) T 1.773, where E is µg N animal–1 day–1 and T is temperature °C. The ammonia-nitrogen excretion on a mg–1 dry body weight day–1 basis was related to temperature according to the following similar expression Log10E = (0.043) T + 0.153, where E is µg N mg–1 dry body weight day–1, and T is temperature °C. The length-weight relationship of Daphnia pulex for the summer populations (N = 1583) was log10W = (0.526) Log10L + 1.357, where W is weight in µg and L is length in mm.  相似文献

Population growth and life table demography of the predatory rotifer A. girodi using spineless Brachionus calyciflorus and spined Brachionus havanaensis as prey at densities of 1, 2, 4 and 8 ind. ml–1 at 25°C were studied. Regardless of the prey species, the population of A. girodi increased with increasing availability of Brachionus in the medium. At any given prey density, A. girodi fed B. calyciflorus showed consistently better growth than when fed B. havanaensis. The maximum population densities of A. girodi varied from 0.28 to 1.8 ind. ml–1 depending on the prey species and the density. The rate of population increase observed in population growth studies varied from 0.17 to 0.43 day–1 when fed B. calyciflorus and 0.09 to 0.27 day–1 when fed B. havanaensis. Male population of A. girodi was closely related to female density. The lowest average lifespan was observed for A. girodi when fed B. havanaensis at 1 ind. ml–1, while the converse was the case when fed B. calyciflorus at comparable prey concentration. Net reproductive rates varied from 16 to 26 offspring female–1 lifespan–1 depending on the prey species and concentration. Generation time of A. girodi decreased with increasing food concentrations for both the prey species. The rates of population increase obtained from life table demography were lower for A. girodi when fed B. havanaensis than when fed B. calyciflorus.  相似文献

To determine the contribution of food ingestion (trophic pathway) to PCB contamination of zooplankton in the river Meuse (Belgium), we used 14C-labelled algae (Dictyosphaerium ehrenbergianum) to measure ingestion and assimilation rates in the rotifer species Brachionus calyciflorus. When the concentration of algae in the culture medium varied from 20 103 to 200 103 algal cells ml–1 (0.12 to 1.18 mg Cl–1), the Brachionus calyciflorus ingestion rate varied from 0.25 ± 0.12 to 1.52 ± 0.43 ng C ind–1 h–1 at 15 °C and from 0.74 ± 0.17 to 5.93 ± 0.61 ng C ind–1 h–1 at 20 °C. The assimilation efficiency (ratio of the assimilation rate to the ingestion rate) measured in a culture medium containing 200 103 algal cells ml–1 was 55.7 ± 5.8%. Since the PCB concentration measured in the phytoplankton of the river Meuse is about 3 µg PCBs g–1 D.W., the estimated PCB contamination of zooplankton ascribable to the trophic pathway ranges from 0.22 ± 0.17 to 1.31 ± 0.77 µg PCBs g–1 D.W. at 15 °C and from 0.64 ± 0.34 to 5.10 ± 2.10 µg PCBs g–1 D.W. at 20°C. The lower figure based on measurements effected at 20 °C is comparable to the actual level measured in zooplankton samples collected in the river Meuse (0.69 ± 0.20 µg PCBs g–1 D.W.). The applicability of the formula used in our estimate was checked in a 48-hour in vitro experiment in which the rotifers were fed contaminated algae. The PCB accumulation measured in the rotifers was found to coincide with the calculated PCB contamination. Additional experiments were carried out to determine the contribution of the direct pathway to PCB contamination of zooplankton living in the river Meuse (0.02 µg PCBs l–1 of water; average dissolved organic matter: 3 mg C 1–1). The PCB concentration in zooplankton resulting from direct uptake of PCBs from the water was estimated at 0.19 ± 0.05 µg PCBs g–1 D.W. These results show that in zooplankton living in polluted ecosystems, PCBs are likely to accumulate via the trophic pathway to concentrations up to 30 times higher than by direct contamination. Furthermore, our estimates of PCB contamination via the trophic pathway coincide quite well with actual concentrations measured in situ.  相似文献

We studied the outcome of competition between a large (Brachionus calyciflorus) and a small (Anuraeopsis fissa) rotifer species at five algal (Scenedesmus acutus) concentrations (0.5 × 106 to 40.5 × 106 cells ml–1) and with varying initial densities in mixed populations (100 to 0% of B. calcyciflorus or A. fissa), the combined initial biomass being 0.2 µg ml–1 in all test jars. Experiments were conducted at 28 ± 1 °C.Regardless of food concentration, B. calcyciflorus showed a greater increase in biomass than A. fissa, peak densities (mean ± standard error) at the lowest food concentration in the controls being 1.34 ± 0.31 µg dry weight ml–1 and 0.82 ± 0.08 dry weight ml–1, respectively. At the lower food concentrations, A. fissa displaced B. calyciflorus and vice versa at the higher food concentrations. At the intermediate food concentrations of 4.5 × 106 cells ml–1, B. calyciflorus outcompeted A. fissa only if its initial population density was three times higher. The rates of population growth in controls varied from 0.792 ± 0.06 d–1 to 1.492 ± 0.13 d–1 for B. calyciflorus and 0.445 ± 0.04 to 0.885 ± 0.01 for A. fissa depending on food level. When both species were introduced together, low food levels favoured higher abundance of A. fissa than B. calyciflorus, suggesting, in nature, it is likely that small Anuraeopsis colonize oligotrophic water bodies more successfully than larger Brachionus. The results also suggest that the outcome of competition depends not only on the size of the competing species and food availability but also on their colonizing density.  相似文献

Respiration data for different stages of Brachionus calyciflorus, fed with three concentrations of Kirchneriella lunaris at 20°C, are presented. Increasing oxygen consumption from 4.1 to 4.6 .10–3 µl/h × ind. with food decreasing from 5.106 to 106 and 4.105 cells/ml has been fourid for adult females with one egg, but other age groups showed divergent results. Based on the respiration data for age groups o to 12 and 12 to 24 h old and some other results and calculations-e.g. dry weight and caloric content of eggs and females, ingestion rates/h for the different concentrations of food-energy budgets for juvenile, growing B. calyciflorus are presented.  相似文献

S. cerevisiae was grown in a blackstrap molasses containing medium in batch and fed-batch cultures. The following parameters were varied: pH (from 4.0 to 6.5), dissolved oxygen (DO) (from 0 to 5.0 mg O2L–1) and sucrose feeding rate. When glucose concentration (S) was higher than 0.5 g L–1 a reduction in the specific invertase activity of intact cells (v) and an oscillatory behavior of v values during fermentation were observed. Both the invertase reduction and the oscillatory behavior of v values could be related to the glucose inhibitory effect on invertase biosynthesis. The best culture conditions for attainingS. cerevisiae cells suitable for invertase production were: temperature=30°C; pH=5.0; DO=3.3 mg O2L–1; (S)=0.5 g L–1 and sucrose added into the fermenter according to the equations: (V–Vo)=t2/16 or (V–Vo)=(Vf–Vo)·(e0.6t–1)/10.This work was supported by FAPESP  相似文献

We evaluated the effect of algal food density (1.5 × 106, 3.0 × 106 and 4.5 × 106 cells ml−1 of Chlorella) and temperature (22° and 28 °C) on competition among the rotifers Brachionus calyciflorus, Brachionus havanaensis, Brachionus patulus and Brachionus rubens, based on population growth experiments for 24 days. The growth experiments were conducted seperately for each individual rotifer species (i.e., controls), and in mixtures of all four species in equal initial proportions (i.e., under competition). The population growth of B. calyciflorus, B. havanaensis, B. patulus and B. rubens grown separately at two temperatures and at three algal food densities showed typical patterns of lag, exponential and retardation phases in the controls. This pattern differed considerably under competition. In general, we observed that in all of the test species, the highest growth rates were observed at higher food levels and in the absence of congenerics. At 22 °C, under the lowest food level, the differences in the population abundances of B. havanaensis, B. patulus and B. rubens grown alone and in the presence of competition were large. However, these differences reduced as food density was increased from 0.5 × 106 to 4.5 × 106 cells ml−1. At 28 °C and at the lowest food level, all of the other rotifer species eliminated B. havanaensis in mixed cultures. Each brachionid species had a higher rate when grown alone than when cultured with other species. The highest r (mean ± standard error: 0.54 ± 0.01 day−1) was recorded for B. havanaensis at 28 °C under 4.5 × 106 cells ml−1 of algal food density. At 28 °C at low algal food density, the presence of competitors resulted in negative population growth rates for three of the four rotifer species tested.  相似文献

Competitive laboratory experiments between Brachionus calyciflorus and B. patulus were conducted at low (1×106 cells ml–1) and high (3×106 cells ml–1) densities of Chlorella vulgaris and four initial inoculation densities (numerically, 100% B. calyciflorus; 75% B. calyciflorus + 25% B. patulus; 50% each of the two species; 25% B. calyciflorus + 75% B. patulus and 100% B. patulus). Population densities were enumerated and the medium was changed daily for 20 days. B. patulus was a superior competitor in low food density regardless of inoculation density. At high food density, B. calyciflorus showed higher population growth in the first week but thereafter was outcompeted by B. patulus regardless of initial density. When grown alone, B. calyciflorus reached peak abundances (mean ± standard error) of 31±3 and 81±7 individuals ml–1 at low and high food densities, respectively. The corresponding values for B. patulus were 130±2 and 306±13. The adverse effects of B. patulus on the peak abundances of B. calyciflorus were higher at low food concentration. Data on egg ratios (eggs female–1) revealed an inverse relation with population abundance of both tested rotifer species. Our results indicated that the rate of population increase of a species was not a good indicator of its competitive ability. Instead, the ability to reproduce under continuously diminishing food resources (until a threshold level) was responsible for the competitive edge of B. patulus over B. calyciflorus. This was further influenced by the relative inoculation densities of the tested rotifer species and the offered food densities.  相似文献

The recombination reactions of Photosystem II have been investigated in vivo in rice leaves by using the thermoluminescence (TL) emission technique. Excitation of dark-adapted leaf segments at 0 °C with different number of single turn-over flashes induced the appearance of complex TL glow curves. The mathematical analysis of these curves showed the existence of four TL components: B1-band (temperature maximum, tmax, at 24 °C, originating from S3QB recombination), B2-band (tmax at 35 °C, from S2QB), AG-band (tmax at 46 °C) and C-band (tmax at 55 °C, from TyrD+QA). Their contributions to the total TL signal were different depending on the number of flashes given. AG-band seems to reflect a special electron transfer from some unknown stroma donor to PS II. Q-band (tmax at 19 °C), originating from S2QA recombination, was recorded after flashing samples incubated in the presence of DCMU. The recombination halftimes (t1/2) at 20 °C of S2QA, S3QB, S2QB and TyrD+QA were, respectively, 0.8 s, 48 s, 74 s and about 1 h. A sharp AG-band (tmax at 50 °C and t1/2 of 210 s) could be also observed after illumination of leaves with far-red light and after a dark incubation period of whole plants. Incubation of leaf segments with 0.5 M NaCl abolished the inductions of AG-band by darkness and far-red illumination, significantly decreased Q-band intensity, whereas induced a strong increase in C-band intensity. The possible inhibition of S2/S3 formation and quinone oxidation by saline stress are discussed.  相似文献

The calanoid copepod Calanus sinicus was reared in the laboratory under excess food conditions, and its development and growth rates were measured at various temperatures. Egg development time (DH, days) was dependent on temperature (T °C), and was expressed as DH = 55.3 (T + 0.7)–1.44. Post-embryonic development followed the equiproportional rule. The stage duration was short in NI and NII, but compensatingly longer in NIII. Between NIV and CII, it was nearly isochronal, and beyond CII, it tended to increase gradually. The time from egg to adult was expressed as DCVI = 1258 (T + 0.7)–1.44. The specific growth rate was also temperature-dependent and highest from CI to CIII, intermediate from NII to CI and from CIII to CV, and lowest from CV to CVI. The growth rates of C. sinicus are higher than those of co-occurring small copepods such as Paracalanus, Acartia and Microsetella.  相似文献

Rotifer cultures of Brachionus plicatilis (SINTEF-strain, length 250 m) rich in 3 fatty acids were starved for > 5 days at variable temperature (0–18 °C). The net specific loss rate of rotifer numbers were 0.04 day–1 (range 0–0.08 day–1) at 5–18 °C, but reached values up to 0.25 day–1 at 0–3 °C. The loss rate was independent on culture density (range 40–1000 ind ml–1), but was to some extent dependent on the initial physiological state of the rotifers (i.e., egg ratio).The loss rate of lipids was 0.02–0.05 day–1 below 10 °C, where the potential growth rate of the rotifer is low (0–0.09 day–1). The loss rate of lipids increased rapidly for higher temperatures where the rotifer can maintain positive growth, and reached 0.19 day–1 at 18 °C. The Q10 for the lipid loss rate versus temperature was higher than the Q10 for respiration found in other strains. This may suggest that other processes than respiration were involved in lipid catabolism. The content of 3 fatty acids became reduced somewhat faster than the lipids (i.e. in particular 22:6 3), but the fatty acid per cent distribution remained remarkably unaffected by the temperature during starvation.The results showed that rotifer cultures could be starved for up to 4 days at 5–8 °C without essential quantitative losses of lipids, 3 fatty acids, and rotifers. The rotifers exhausted their endogenous lipids through reproduction (anabolism) and respiration (including enhanced locomotion) at higher temperatures. Summary The effect of temperature on the response properties of primary auditory fibres in caiman was studied. The head temperature was varied over the range of 10–35 ° C while the body was kept at a standard temperature of 27 °C (Ts). The temperature effects observed on auditory afferents were fully reversible. Below 11 °C the neural firing ceased.The mean spontaneous firing rate increased nearly linearly with temperature. The slopes in different fibres ranged from 0.2–3.5 imp s–1 °C–1. A bimodal distribution of mean spontaneous firing rate was found (<20 imp s–1 and >20 imp s–1 at Ts) at all temperatures.The frequency-intensity response area of the primary fibres shifted uniformly with temperature. The characteristic frequency (CF) increased nearly linearly with temperature. The slopes in different fibres ranged from 3–90 Hz °C–1. Expressed in octaves the CF-change varied in each fibre from about O.14oct °C–1 at 15 °C to about 0.06 oct °C–1 at 30 °C, irrespective of the fibre's CF at Ts. Thresholds were lowest near Ts. Below Ts the thresholds decreased on average by 2dB°C–1, above Ts the thresholds rose rapidly with temperature. The sharpness of tuning (Q10db) showed no major change in the temperature range tested.Comparison of these findings with those from other lower vertebrates and from mammals shows that only mammalian auditory afferents do not shift their CF with temperature, suggesting that a fundamental difference in mammalian and submammalian tuning mechanisms exists. Summary The effect of doubling the saturated fatty acid content on the electrophysiology ofNeurospora crassa membranes was studied. Intracellular membrane input resistance (R m ) and potential (E m ) were measured for wild-type (w/t) andcel (Tween 40) organisms as a function of temperature. Over the 0 to 40°C temperature range studied, meanE m values of bothw/t andcel (Tw 40) organisms increased from –160 to –210 mV. This difference is greater than that expected from Nernst potential considerations, indicating an active component ofE m . This active component is insensitive to a doubling of the saturated fatty acid content.R m exhibits a temperature dependence and hysteresis. Averaged data indicate an increase inR m with decreased temperature. The slope of the temperature dependence varies among individual hyphae. Above 17.5°Ccel (Tw 40) hyphae averaged greater than 70% higher values ofR m thanw/t. Below 17.5°Cw/t R m data divided into low and high temperature dependence groups, whilecel data exhibited a low temperature dependence. The results are discussed in relation to gel-liquid crystal phase transitions, membrane fluidity, and the contribution of fatty acid structure to membrane electrical properties.  相似文献

The CO2 production of individual larvae of Apis mellifera carnica, which were incubated within their cells at a natural air humidity of 60–80%, was determined by an open-flow gas analyzer in relation to larval age and ambient temperature. In larvae incubated at 34 °C the amount of CO2 produced appeared to fall only moderately from 3.89±1.57 µl mg–1 h–1 in 0.5-day-old larvae to 2.98±0.57 µl mg–1 h–1 in 3.5-day-old larvae. The decline was steeper up to an age of 5.5 days (0.95±1.15 µl mg–1 h–1). Our measurements show that the respiration and energy turnover of larvae younger than about 80 h is considerably lower (up to 35%) than expected from extrapolations of data determined in older larvae. The temperature dependency of CO2 production was determined in 3.5-day-old larvae, which were incubated at temperatures varying from 18 to 38 °C in steps of 4 °C. The larvae generated 0.48±0.03 µl mg–1 h–1 CO2 at 18 °C, and 3.97±0.50 µl mg–1 h–1 CO2 at 38 °C. The temperature-dependent respiration rate was fitted to a logistic curve. We found that the inflection point of this curve (32.5 °C) is below the normal brood nest temperature (33–36 °C). The average Q10 was 3.13, which is higher than in freshly emerged resting honeybees but similar to adult bees. This strong temperature dependency enables the bees to speed up brood development by achieving high temperatures. The intrinsic rate of natural increase(rm) is useful to estimate the populationgrowth potential of insects and mites, whichmay help predict the outcome of pest-naturalenemy interactions. This study was conductedto determine how 12 constant temperatureregimes between 10–38 °C (± 0.5 °C) may differentially affect therm of the McDaniel spider mite, Tetranychus mcdanieli McGregor (Acarina: Tetranychidea), a common pest of raspberry, andits coccinellid predator, Stethoruspunctillum Weise (Coleoptera: Coccinellidae). Tetranychus mcdanieli survived tomaturity in the 14–36 °C range, comparedto the 14–32 °C range for S.punctillum. Survival above 24 °Cremained high for the spider mite, butdecreased markedly for the coccinellid. Tetranychus mcdanieli's range forreproduction was similar to its survival range,but S. punctillum failed to reproduce at14 °C and reproduced only poorlyat 16 °C. Offspring production peakedat 24 °C for both T. mcdanieli(average 152 eggs per female), and S.punctillum (280 eggs per female). At alltemperatures suitable for reproduction, femalelongevity of the coccinellid was greater thanthe spider mite, which was characterized byearlier/faster reproduction than thecoccinellid. As temperature increased, therm followed a typical asymmetricaldome-shape pattern, with maximum values of0.196 d–1 and 0.385 d–1at 30 °C and 34 °C, for S.punctillum and T. mcdanieli,respectively. For each species, therm-temperature relation was successfullymodelled using a curvilinear regressionequation previously shown to predictdevelopment rate. In both species, thedevelopment rate response to temperature has amajor influence on the temperature-rmrelationship. In the 16–32 °C rangesuitable for population growth of both species,the rm of T. mcdanieliwas 1.9 (30 °C) to 8 (16 °C) times greaterthan S. punctillum. These growthpotential ratios are consistently in favor ofthe prey, suggesting a limitation of thecoccinellid with respect to its capacity totrack T. mcdanieli populations. However,under short season conditions, the inferiorreproductive dynamics of S. punctillum'svs. spider mite prey should not have aprevailing influence in determining impact, andmay be compensated by high voracity incombination with a strong aggregativeresponse.  相似文献

Scenedesmus protuberans Fritsch was grown in light-limited continuous cultures with a light-dark cycle, at temperatures of 20° and 28° C. At 20° irradiances of 12 and 38 W m–2 were used, at 28° 38 W m–2.The relationships between growth rate and light uptake rate were of diphasic linear character. Summary In Antarctica, we investigated the energy consumption of Adélie (Pygoscelis adeliae), Gentoo (P. papua) and Chinstrap (P. antarctica) penguins while resting in the water (8.4 W-kg–1) and swimming underwater at various speeds, using a 21m long canal filled with sea-water at 4°C in conjunction with respirometry. The birds swam at will and consumed 15.7, 16.1 and 10 W·kg–1 at the speed where cost of transport was minimal (2.1, 2.3 and 2.5 m·s–1 in Adélie, Gentoo and Chinstrap penguins, respectively). Thermal conductance in pygoscelid penguins was 3.3 W·°C–1. m–2 and energy expenditure (Pi, W·kg–1) while resting in the water is given by Pj = -0.3 ta+9.6, where ta is water temperature in °C. During the breeding season, pygoscelid penguins spend 25–40% of their daily energy expenditure while foraging at sea. The importance of accurate estimates of at-sea activity and energy consumption is discussed.  相似文献

Summary Radiosodium efflux from barnacle muscle fibers is a function of pH e , the threshold pH e for stimulation of Na efflux into HCO 3 -artificial sea water (ASW) being 6.8 and the fixed thresholdpCO2 (in an open CO2 system) being approximately 30 mm Hg. Acidification of ASW containing non-HCO 3 buffer is without effect on the Na efflux. The Na efflux following stimulation by reducing the pH of 10mM HCO 3 -ASW from 7.8 to 6.3 is reduced by 17.3% as the result of microinjecting 100mM EGTA, and increased by 32.6% as the result of microinjecting 0.5M ATP. The Na efflux into K-free HCO 3 -ASW is markedly stimulated by external acidification. Ouabain-poisoned fibers are more responsive to a low pH e than unpoisoned fibers. Applying the 2-14C-DMO technique, it is found that fibers bathed in 10mM HCO 3 -ASW at pH 7.8 have an internal pH of 7.09±0.106 (mean±SD), whereas fibers bathed in 25mM TRIS-ASW at pH 7.8 have a pH i of 7.28±0.112. The relationship between pH i and pH e as external pH is varied by adding H+ is linear. Measurements of the resting membrane potential indicate that external acidification in the presence of HCO 3 as buffer is accompanied by a fall inE m , the threshold pH e being 7.3 both at 24 and 0°C. This sensitivity amounts to 8.2 mV per pH unit (at 24°C) over a wide range of pH e . Membrane resistance following external acidification remains unchanged. Microinjection of the protein inhibitor of Walsh before external acidification fails to stop depolarization from occurring. Cooling to 0°C also fails to abolish depolarization following acidification. Whereas external ouabain and ethacrynic acid reduceE m in the absence or presence of acidification, DPH hyperpolarizes the membrane or arrests depolarization both at 24 and 0°C. This effect of DPH at 0°C is seen in the absence or presence of acidification. It is suggested that depolarization following acidification of a HCO 3 -containing medium is due to activation of a Cl-and/or HCO 3 -pump and that ouabain and ethacrynic acid reducesE m by abolishing uncoupled Na transport.  相似文献
