Zinc (Zn), lead (Pb) and cadmium (Cd) tolerance in populationsof seedlings ofPhragmites australisraised from seeds collectedfrom a mine site (Plombières, Belgium) contaminated withZn, Pb and Cd and three ‘clean’ sites (Felixstowe,UK; Wisbech, UK; and Mai Po, Hong Kong) were studied under glasshouseconditions. Small differences were found between the metal-contaminatedpopulation and the three ‘clean’ populations whenseedlings were grown in 1.0 µg  ml-1Zn and 10.0 µgml-1Pb treatment solutions. In general, however, different populationsof seedlings showed similar growth responses, metal uptake andindices of Zn, Pb and Cd tolerance when cultured in the samemetal-contaminated media for 89 d or in the same metal treatmentsolutions (ZnSO4:1.0 and 4.0 µg ml-1Zn; Pb(NO3)2: 10.0and 25.0 µg ml-1Pb; CdSO4: 0.5 and 1.0 µg ml-1Cd)for 3 weeks. There was insufficient evidence to support thehypothesis that the metal-contaminated population has evolvedto a Zn-, Pb- or Cd-tolerant ecotype but the results indicatedsome differentiation between the populations with that fromHong Kong being the least productive under the experimentalconditions used. The implications of the findings on selectionof provenances for use in constructed wetlands for wastewatertreatment are discussed. Secretin stimulates ductal secretion by activation of cAMP PKA CFTR Cl/HCO3 exchanger in cholangiocytes. We evaluated the expression of 2A-, 2B-, and 2C-adrenergic receptors in cholangiocytes and the effects of the selective 2-adrenergic agonist UK 14,304, on basal and secretin-stimulated ductal secretion. In normal rats, we evaluated the effect of UK 14,304 on bile and bicarbonate secretion. In bile duct-ligated (BDL) rats, we evaluated the effect of UK 14,304 on basal and secretin-stimulated 1) bile and bicarbonate secretion; 2) duct secretion in intrahepatic bile duct units (IBDU) in the absence or presence of 5-(N-ethyl-N-isopropyl)amiloride (EIPA), an inhibitor of the Na+/H+ exchanger isoform NHE3; and 3) cAMP levels, PKA activity, Cl efflux, and Cl/HCO3 exchanger activity in purified cholangiocytes. 2-Adrenergic receptors were expressed by all cholangiocytes in normal and BDL liver sections. UK 14,304 did not change bile and bicarbonate secretion of normal rats. In BDL rats, UK 14,304 inhibited secretin-stimulated 1) bile and bicarbonate secretion, 2) expansion of IBDU luminal spaces, and 3) cAMP levels, PKA activity, Cl efflux, and Cl/HCO3 exchanger activity in cholangiocytes. There was decreased lumen size after removal of secretin in IBDU pretreated with UK 14,304. In IBDU pretreated with EIPA, there was no significant decrease in luminal space after removal of secretin in either the absence or presence of UK 14,304. Shells from 14 populations of sphaeriid clams including Sphaeriumstriatinum, S. simile, Pisidium walkeri, Musculim partumeiumand M. iransversum were analyzed for organic carbon (µgCmg–1 shell), nitrogen (µg,N mg–1 shell) andCaCOs (%CaCO3 of total clam dry weight). Habitat waters wereassessed for total hardness (expressed as ppm CaCo3), ppm Ca,ppm Mg, conductivity (µmho) and suspended organic Carbon(µgCl–1). For all populations, shell organic C andN are positively correlated and there is an inverse relationshipbetween the amounts of shell CaCO3 and shell organic carbon.Trophic considerations give the best correlation with shelltype at the generic level of consideration since species ofMusculium are found at the opposite end of the trophic scale(eutrophic) from all other populations studied. For S. striatinum,the most extensively studied species, the amount of shell CaCO3is inversely related to water hardness. Effects of diffusion and upwelling on the formation of red tides   总被引:4,自引:0,他引:4  
In this paper, records on the timing and location of specificred tides monitored once or twice a week in Mikawa Bay, Japan,are related to horizontal and vertical mixing rates determinedfrom a numerical model. Horizontal (Kh) and vertical (Kz) diffusioncoefficients, and upwelling velocities, were estimated usinga box model analysis. In the wind-mixed period and in the upperlayer during the stratified period, Kh was estimated to be ofthe order of 102 m2 s–1. During the stratified period,Kz was estimated to be of the order of 10–5 m2 s–1.The upwelling velocity was calculated to be in the range 0.35–5.1m day–1 with an average of 1.5 m day–1. Comparisonbetween the literature values of the specific growth rate (µ)of the red tide-forming diatoms and calculated Kh values duringthe red tides show that diatoms which have a low µ cannotform red tides in a strongly diffusive environment, while specieshaving a high µ can form red tides even in a strong diffusiveenvironment. On the other hand, no clear relationship was foundbetween µ of the flagellate group and Kh, although theflagellate group formed red tides even in severe diffusive conditions.From the comparison between the literature values of sinkingrate and swimming speed and the physical parameters associatedwith vertical processes, it was concluded that flagellates willform red tides, even in severe diffusive conditions, by usingtheir swimming ability, while diatoms form red tides by theirhigh growth rates with the aid of vertical diffusion and theupwelling movement of water.  相似文献

Feeding on natural plankton populations and respiration of thesmall cyclopoid copepod Oithona similis were measured duringthe warm season in Buzzards Bay, Massachusetts, USA. AlthoughO.similis did not significantly ingest small autotrophic andheterotrophic flagellates (2–8 µn), this copepodactively fed on >10 µm particles, including autotrophic/heterotrophic(dino)flagel-lates and ciliates, with clearance rates of 0.03–0.38ml animal–1 h–1. The clearance rates increased withthe prey size. O.similis also fed on copepod nauplii (mainlycomposed of the N1 stage of Acartia tonsa with a clearance rateof 0.16 ml animal–1 h–1. Daily carbon ration fromthe combination of these food items averaged 148 ng C animal–1day–1 (41% of body C day–1), with ciliates and heterotrophicdino-flagellates being the main food source ({small tilde}69%of total carbon ration). Respiration rates were 020–0.23µl O2 animal–1 day–1. Assuming a respiratoryquotient of 0.8 and digestion efficiency of 0.7, the carbonrequirement for respiration was calculated to be 125–143ng C animal–1 day–1, close to the daily carbon rationestimated above. We conclude that predation on ciliates andheterotrophic dinoflagellates was important for O.similis tosustain its population in our study area during the warm season.  相似文献

The effect of different conditions of transfer (+;1 °C withwater, +20 °C with and without water) from the natural habitatand laboratory acclimation procedure (with and without bottomsediment) on the acid-base and electrolyte status of the freshwateruniomd mussel Anodonta anatina (L.) was studied. The shift inthe acid-base status of A. anatina during the transfer was smallerat 1°C than at 20°C. The pO2 content of the haemolymphdeclined significantly under all three transfer conditions.Neither the transfer nor the laboratory acclimation affectedhaemolymph [Na+]. However, during transfer at 1°C the haemolyraph[Ca2+] did not change, whereas [K+] increased up to three fold.Haemolymph [Ca2+] increased when mussels were transferred at20°C After a 17-day aquarium acclimation, the pO2 of all the musselskept in the sediment had returned to the field level and thetransfer seemed to have had no effect on the pCH of the haemolymph.Conversely, in the groups kept without sediment, the pO2 continuouslyremained above the field level. Whereas the [K+] of all themussels declined to the field level, the haemolymph [Ca2+] remainedelevated throughout the entire acclimation period of 17 days. Nodulated 1-1.5-year-old plants of Acacia littorea grown inminus nitrogen culture were each partnered with a single seedlingof the root hemiparasite Olax phyllanthi. Partitioning of fixedN between plant organs of the host and parasite was studiedfor the period 4–8 months after introducing the parasite.N fluxes through nodules of Acacia and xylem-tapping haustoriaof Olax were compared using measured xylem flows of fixed Nand anatomical information for the two organs. N2 fixation duringthe study interval (635 µg N g FW nodules–1 d–1)corresponded to a xylem loading flux of 0.20 µg N mm–2d–1 across the secretory membranes of the pencycle parenchymaof the nodule vascular strands. A much higher flux of N (4891µg mm–2 d–1) exited through xylem at the junctionof nodule and root. The corresponding flux of N from host xylemacross absorptive membranes of the endophyte parenchyma of Olaxhaustorium was 1.15 µg N mm–1 d–1, six timesthe loading flux in nodules. The exit flux from haustorium toparasite rootlet was 20.0 pg N mm–1 d–1, 200-foldless than that passing through xylem elements of the nodule.Fluxes of individual amino compounds in xylem of nodule andhaustorium were assessed on a molar and N basis. N flux valuesare related to data for transpiration and partitioning of Cand N of the association recorded in a companion paper. Conditions for measuring fluorescence induction curves (time-scalems) of in vivo chlorophyll a were studied using cultures ofDunaliella tertiolecta Butcher (Chlorophyceae) and of Thalassiosirapseudonana Hustedt (3H) (Bacillariophyceae), and samples ofnatural phytoplankton populations from the Grand Banks. Thearea above the fluorescence induction curve (ADCMU) and themaximum fluorescence intensity (Fmax) measured in the presenceof 3-(3,4-dichlorophenyl)-1, 1-dimethylurea (DCMU) were computedby microcomputer. Cells must be ‘conditioned’ or‘adapted’ prior to obtaining a fluorescence inductioncurve; dark-adaptation resulted in a lower ADCMU and Fmax thandid adaptation in far-red (720 nm) light, and was the conditioningmethod chosen. ADCMU and Fmax increased linearly with increasingirradiance up to 32.8 W m–2 the highest actinic irradianceavailable. Information on the light history of D. tertiolectawas obtained by following the time-course of change in ADCMUand in Fmax for cells exposed for 10 min to far-red or to bluelight. The rise-time of the fluorescence induction curve andvalues of Fmax were greater for samples of D. tertiolecta concentratedonto glass-fiber filters than for liquid samples, however, valuesof ADCMU for filtered and liquid samples were not significantlydifferent. Samples of Grand Banks phytoplankton collected ontoglass-fiber filters and frozen for 28 d exhibited a significantdecrease in Fmax and in ADCMU relative to the same freshly-filteredsamples. Diel patterns in the uptake of nitrogenous nutrients were observedin the coastal plume of the Chesapeake Bay system, but the specificpatterns varied with season. During the winter months, ratesof NH4+ and urea uptake were significantly higher at night thanduring the day, and rates of NO3 uptake were higher duringthe day. During the summer, rates of NH4+ and urea uptake weresignificantly higher at night only during half the studies conducted;during the remaining studies, there was either no significantdifference or rates of uptake of NH4+ were higher during theday. Rates of NO3 uptake during the summer months werealso higher during the day than at night. Seasonal differenceswere also apparent in the time of day at which maximum observeduptake rates of each nitrogen nutrient occurred. During thewinter-spring months, maximum observed rates of NO3 uptakeoccurred between first light and noon, whereas during the summermonths, maximum observed uptake rates of NO3 occurredboth morning and afternoon, and consistently 9–16 h afterthe maximum observed peak in the uptake of reduced nitrogen.We interpret these findings in terms of seasonal shifts in nitrogennutritional status of the assemblages, as well as species-specificdifferences in the effect of a given stimulus (e.g. a nitrogenpulse at the mouth of the Bay) to entrain an uptake response,and we suggest that the extent of this variability must be understoodbefore generalizations about the use of f-ratios as characteristicsof specific populations or water masses can be drawn.  相似文献

The primary leaves of kidney bean (Phaseolus vulgaris L.) openunder light and close in the dark by the deformation of thepulvinus resulting from diurnal distribution changes of K+,Cl, organic acid (or H+) and NO3. When Rb+ was added as a tracer of K+ to the seedlings throughtheir roots, it was transported to the pulvinus cells duringthe light period but not during the dark period. Transpirationoccurred vigorously in the light but almost stopped in the dark.We concluded that Rb+ absorbed by the roots was carried to thepulvinus by the transpiration stream. Spatial and seasonal egg production rates (Er) and egg hatchingsuccess in the copepods Calanus finmarchicus and Calanus helgolandicuswere measured in the North Sea from March to September. Foodavailability was monitored by chlorophyll and protist concentrationsand three size fractions of seston fatty acids. Seasonal andspatial distribution and production differed between the species.Calanus finmarchicus was found only offshore of the 50-m isobath,with decreasing Er (37–28 eggs female–1 day–1)from March to July. Calanus helgolandicus had two abundancepeaks, in spring and autumn, with a low in May during whichtime the highest Er were observed (38 eggs female–1 day–1).At other times, Er in C. helgolandicus remained lower than inC. finmarchicus (  相似文献

A 50-year-old Pinus sylvestris L. stand was exposed for 2 yearsto low concentrations of SO2 and NO2 in an open-air exposureexperiment in northern Sweden. The mean SO2 concentrations inthe centre of the exposed plot during the 1988 exposure from14 June to 25 September, and during the 1989 exposure from 6June to 30 September were 15 nl 1–1 and 12 nl 1–1,respectively. The corresponding values for NO2 were 15 nl 1–1and 10 nl 1–1, respectively. The concentration in thecontrol plot was never higher than a few ppb, and mostly below1 nl 1–1. Needles sampled from the SO2 and NO2-exposed area showed reducedactivities of glutathione reductase (GR; EC 1.6.4.2 [EC] ) and superoxidedismutase (SOD; EC 1.15.1.1 [EC] ) compared with controls. The GRactivity showed decreased levels in autumn and winter, whilethe exposure had ceased, and SOD showed decreased activity duringthe second summer of exposure. Neither membrane-bound nor water-solubleanti-oxidants such as -tocopherol, carotenoids or glutathionechanged due to the exposure. The sulphur/nitrogen ratio wasincreased in needles that were exposed to SO2 and NO2 implyinga changed nutrition balance. The results suggest that the capacityof SOD and GR in the ascorbate-glutathione pathway was reduceddue to the exposure to air pollutants. Ammonia excreted by mixed zooplankton populations over an annual(1972–1973) cycle in Narragansett Bay varied from 0.04to 3.21 µg at NH3-N dry wt–1 day–1, exclusiveof two exceptional rates measured one year apart: 11.74 and18.39 µg at NH3-N mg dry wt–1 day–1. Grossphytoplankton production integrated over the year (1972–1973)averaged 151 mg C m–3 day–1 for an 8 m water column;peaks of 332 and 905 mg C m–3 day–1 occurred duringthe winter-spring and summer blooms, respectively. Excretedammonia, integrated seasonally and annually, contributed only0.2% and 4.9% of the nitrogen required for observed gross productionduring the winter-spring and summer blooms, respectively, and4.4% annually. However, excreted ammonia may be an importantsource of the nitrogen required by Skeletonema costatum, thedominant diatom in Narragansett Bay, during the post-bloom periodwhen 186% of the nitrogen required for its net production wasmet by ammonia excretion. A combination of zooplankton ammoniaexcretion and benthic ammonia flux contributed 22% of the nitrogenrequired for the annual gross production (440 g C m–2)while 51% of the nitrogen required for the net production ofSkeletonema was accounted for by regenerated nitrogen. 1This research was supported by NSF grant GA 31319X awardedto Dr.T.J.Smayda.  相似文献

Wheat (Triticum aestivum L.) embryos form in dynamically-regulatedovular environments. Our objectives were to improve developmentof cultured immature wheat embryos by simulating, in vitro,abscisic acid (ABA) levels and O2 tensions as found in wheatovules during zygotic embryogenesis. We characterized from intactwheat kernels embryo respiration, embryo morphology and embryoand endosperm + ABA levels at 13, 19 and 25 d post-anthesis(DPA). Young (13 DPA) embryos were then excised and culturedin vitro, where they were exposed to 0·2 or 2·Ommol m–3 ±ABA and 2.·1, 2·5 or 7·4mol m–3 (6, 7 and 21%, respectively) gaseous O2. At 6and 12 d in culture, + ABA levels, embryo respiration and embryomorphology were characterized by treatment. Thirteen-day-oldembryos from two different plant populations differed by 17-foldin initial ABA content. However, this difference did not affectprecocious germination in vitro, nor did it affect the amountof exogenous ABA required to reduce precocious germination by40%. In this respect, embryos from both populations were equallysensitive to exogenous ABA. Cavity sap O2 levels (2·1to 2·5 mol m–3) were much more effective in preventingprecocious germination of cultured embryos than were cavitysap levels of ABA (0·2 to 2·0 mmol m–3).The combination of physiological levels of both ABA and O2 largelynormalized DW accumulation and embryo morphology without alteringendogenous + ABA levels. Upland grasslands are a major component of natural vegetationwithin the UK. Such grasslands support slow growing relativelystable plant communities. The response of native montane grassspecies to elevated atmospheric carbon dioxide concentrationshas received little attention to date. Of such studies, mosthave only focused on short-term (days to weeks) responses, oftenunder favourable controlled environment conditions. In thisstudy Agrostis caplllaris L.5, Festuca vivipara L. and Poa alpinaL. were grown under semi-natural conditions in outdoor open-topchambers at either ambient (340µmol mol–1) or elevated(680µmol mol–1) concentrations of atmospheric carbondioxide (CO2 for periods from 79 to 189 d, with a nutrient availabilitysimilar to that of montane Agrostis-Fescue grassland in Snowdonia,N. Wales. Whole plant dry weight was increased for A. capillarisand P. alpina, but decreased for F. vivipara, at elevated CO2.Major components of relative growth rate (RGR) contributingto this change at elevated CO2 were transient changes in specificleaf area (SLA) and leaf area ratio (LAR). Despite changes ingrowth rate at 680 µmol mol–1 CO2, partitioningof dry weight between shoot and root in plants of A. capillarisand P. alpina was unaltered. There was a significant decreasein shoot relative to root growth at elevated CO2 in F. viviparawhich also showed marked discoloration of the leaves and increasedsenescence of the foliage. Time-courses of 14CO2-fixation and of enzyme activities involvedin photorespiration and photosynthesis were determined duringthe life span of cotyledons from sunflower seedlings (Helianthusannuus L.). Glycolate formation in vivo was estimated from theresults of combined labelling and inhibitor experiments. NADPH-glyceraldehyde-3-phosphatedehydrogenase, NADPH-glyoxylate reductase and chlorophyll werewell correlated with the time-course of 14CO2-fixation (photosynthesis).There was, however, a considerable discrepancy between the developmentalsequence of photosynthesis and that of both ribulose-l,5-bisphosphatecarboxylase and glycolate oxidase. Furthermore, time-coursesof glycolate oxidase activity in vitro and of glycolate formationin vivo differed significantly. Therefore, the use of glycolateoxidase as a marker for the activity of photorespiration ingreening sunflower cotyledons may be questionable. Results from14CO2-labelling experiments with cotyledons treated with theglycolate oxidase inhibitor 2-hydroxy butynoic acid suggestthat glycolate formation relative to CO2-fixation is reducedin senescent cotyledons. Conditions and maintenance of growth were chosen so that plantsof Clusia minor L. were obtained which showed the C3- and CAM-modes of CO2-exchange, respectively. C. minor is known to accumulateconsiderable amounts of citric acid in addition to malic acidduring the dark-phase of CAM. 14CO2-pulse-chase experiments were performed with these plants.Patterns of labelling during the pulse and redistribution oflabel during the chase in the C3-mode were as expected for C3-photosynthesis.Pulse-labelling in the CAM-mode during the last hour of thelight period, during the first part of the dark period and duringthe last hour of the dark period always led to an almost exclusiveincorporation of label into malate. Redistribution of labelfrom malate after the pulse at the end of the dark period duringthe chase in the subsequent light period followed the patternexpected for light-dependent reassimilation of CO2 remobilizedfrom malate in CAM during the light period. During the chasesin the dark period, label was transferred from l4C-malate tocitrate. This suggests that during accumulation of citric acidin the dark period of CAM in C. minor, citrate is synthesizedin the mitochondria from malate or oxaloacetate after formationof malate via phosphoenolpyruvate carboxylase. The experiment also showed that no labelled compounds are exportedfrom leaves in the CAM-mode during the dark period. In plantsof the C3-mode the roots proved to be strong sinks. Effects of light flux density (LFD) during growth and uptakeassay on induction of transport system and kinetics of transport were studied using the Azolla pinnata-Anabaena azollae association (Azolla). Theinduction and uptake kinetics of the transport system were determined using an automated system that measuredthe NO3 concentration in the growth medium as a function oftime, using an on-line high performance liquid chromatograph(HPLC) with a UV-VIS detector. Full induction of the transport system required about 1.5 to 2.0 h and occurred without any apparent lag phase,regardless of the LFD provided. The level of induction of transport of Azolla grown at 600 µmol m–2s–1 LFD was higher than for that grown at 100 µmolm–2 s–1. Similarly, 600 µmol m–1 s–1LFD during the assay resulted in a higher level of inductionthan did 100 umol m–2 s–1. An increase in the LFDeither during the growth or the assay period increased the uptake rate; however, an increase in LFD duringthe latter period had greater effect. Azolla grown and assayedat 600 umol m–2 s–1 had the highest uptake rate. The uptake rate at 50 cm3 m–3ambient CO2 concentration was initially higher than at 305 cm3m–3, but the uptake rate decreased rapidly with time andeventually dropped below that at 305 cm3 m–3 CO2. Thesedata suggest that the energy required for transport in Azolla may bypass the photosynthetic CO2 fixationand carbon-cycling. The specific respiration rates of nodulated root systems, ofnodules and of roots were determined during active nitrogenfixation in soya bean, navy bean, pea, lucerne, red clover andwhite clover, by measurements on whole plants before and afterthe removal of nodule populations. Similar measurements weremade on comparable populations of the six legumes, lacking nodulesbut receiving abundant nitrate-nitrogen, to determine the specificrespiration of their roots. All plants were grown in a controlled-environmentclimate which fostered rapid growth. The specific respiration rates of nodulated root systems ofthe three grain and three forage legumes during a 7–14-dayperiod of vegetative growth varied between 10 and 17 mg CO2g–1 (dry weight) h–1. This mean value consistedof two components: a specific root respiration rate of 6–9mg CO2 g–1 h–1 and a specific nodule respirationrate of 22–46 mg CO2 g–1 h–1. Nodule respirationaccounted for 42–70 per cent of nodulated root respiration;nodule weight accounted for 12–40 per cent of nodulatedroot weight. The specific respiration rates of roots lackingnodules and utilizing nitrate nitrogen were generally 20–30per cent greater than the equivalent rates of roots from nodulatedplants. Hydrosulfite-reduced FMN served as an electron donor for nitratereductase purified from broad bean leaves. FMN was successfullyreplaced with BV. The flavine nucleotide nitrate reductase hadits pH optima at about 7.8 with phosphate buffer and at about7.4 with Tris-HCl buffer. The Km's for nitrate and FMN were3.7 ? 10–4 M and 3.7 ? 10–5 M, respectively. NADH2: nitrate reductase activity was completely inhibited by0.1 mM p-CMB, whereas FMNH2: nitrate reductase activity wasnot. Inhibited activity was restored by the addition of cysteine.A sulfhydryl enzyme is involved in the NADH2: nitrate reductasesystem but not in the FMNH2 : nitrate reductase system. NADH2and FMNH2 probably feed electrons into the electron transportchain at different sites. The nitrate reductase preparationhad an NADH2-specific diaphorase activity which was almost completelyinhibited by 0.1 mM p-CMB. The NADH2-specific diaphorase mayform the sulfhydryl enzyme which mediates electron transferbetween NADH2 and nitrate. (Received May 6, 1969; )  相似文献
