彭泽鲫两个雌核发育克隆的染色体组型分析   总被引:10,自引:0,他引:10  
采用PHA和秋水仙素体内注射法直接制作肾细胞染色体标本,对彭泽鲫种群内两个不同雌核发育克隆的亲本进行染色体数目及组型分析。结果表明,彭泽鲫种群内的两个不同克隆存在染色体数目及组型差异,其中克隆H包含6条超数染色体在内的染色体众数是156,150条基本染色体的组型公式为:42M 36SM 39ST 33T,NF=228;克隆L包含12条超数染色体在内的染色体众数是162,150条基本染色体的组型公式为:36M 45SM 33ST 36T,NF=231。两个不同雌核发育克隆的发现及其染色体的差异说明彭泽鲫种群内同样存在着类似银鲫种群内的遗传多样性。  相似文献

Karyotypic and cytogenetic characteristics of catfish Harttia carvalhoi (Paraíba do Sul River basin, S?o Paulo State, Brazil) were investigated using differential staining techniques (C-banding, Ag-staining) and fluorescent in situ hybridization (FISH) with 18S and 5S rDNA probes. The diploid chromosome number of females was 2n = 52 and their karyotype was composed of nine pairs of metacentric, nine pairs of submetacentric, four pairs of subtelocentric and four pairs of acrocentric chromosomes. The diploid chromosome number of males was invariably 2n = 53 and their karyotype consisted of one large unpaired metacentric, eight pairs of metacentric, nine pairs of submetacentric, four pairs of subtelocentric, four pairs of acrocentric plus two middle-sized acrocentric chromosomes. The differences between female and male karyotypes indicated the presence of a sex chromosome system of XX/XY1Y2 type, where the X is the largest metacentric and Y1 and Y2 are the two additional middle-sized acrocentric chromosomes of the male karyotype. The major rDNA sites as revealed by FISH with an 18S rDNA probe were located in the pericentromeric region of the largest pair of acrocentric chromosomes. FISH with a 5S rDNA probe revealed two sites: an interstitial site located in the largest pair of acrocentric chromosomes, and a pericentromeric site in a smaller metacentric pair of chromosomes. Translocations or centric fusions in the ancestral 2n = 54 karyotype is hypothesized for the origin of such multiple sex chromosome systems where females are fixed translocation homozygotes whereas males are fixed translocation heterozygotes. The available cytogenetic data for representatives of the genus Harttia examined so far indicate large kayotype diversity.  相似文献

A karyometric analysis of the chromosome set of the marine turbellariansMonocelis fusca, M. lineata andParotoplana macrostyla has been carried out. The karyotype of the twoMonocelis species investigated (2n=6) is formed by three pairs of small and similarly sized chromosomes: InM. fusca, chromosome 1 is metacentric, chromosome 2 acrocentric and chromosome 3 is subtelocentric.M. lineata also presents one pair of metacentric chromosomes (chromosome 2), while chromosomes 1 and 3 are submetacentric.P. macrostyla (2n=12) reveals two pairs of large metacentric and four pairs of small chromosomes, three of which are metacentric, whereas the last is subtelocentric.  相似文献

Mitotic chromosomes of Heterobilharzia americana from Louisiana are described from parasite material that was dissected from snails, air-dried on slides, and stained with conventional Giemsa and C-band methods. As in other schistosomes, the female is the heterogametic sex. This Louisiana strain, however, differs from a Texas strain and other schistosome species in that the male and female have different diploid numbers of chromosomes (male, 20; female, 19), and the strain has a ZZ male/ZWA female sex-determining mechanism. The chromosomes of the male resemble those of the Texas strain in number and morphology with the Z chromosomes being metacentric and the largest elements in the karyotype. The others form a series decreasing in size to very small number 10's. Chromosomes 2,3, and 4 are subtelocentric; 5 is subtelocentric to acrocentric and is satellited; 6 is submetacentric to subtelocentric; 7 is submetacentric; 8 is subtelocentric to submetacentric; 9 is metacentric to submetacentric; and 10 is metacentric. The female complement differs from the male of this strain in having only 1 normal chromosome 5. The other number 5 and most of the original W apparently have fused tandemly to form the WA chromosome (a "neo-W").  相似文献

A karyological study was carried out in order to compared the chromosome numbers, chromosome morphologies and karyotypes of the oriental liver fluke, Clonorchis sinensis (Trematoda: Opisthorchiidae), collected from Korea and China. Chromosome preparations were made by means of air-drying method. The chromosome number was 2n = 56 in both Korean and Chinese flukes, and chromosomes were divided into two groups based on this size; consisting of 8 pairs of large and 20 pairs of small chromosomes. However, the karyotypes showed some differences between Korean and Chinese flukes. The karyotype of liver flukes from Korea consisted of three metacentric pairs, one meta-/submetacentric pair, 16 submetacentric pairs and eight subtelocentric pairs of chromosomes. On the other hand, liver flukes from China consisted of two metacentric pairs, two meta-/submetacentric pairs, 16 submetacentric pairs and eight subtelocentric pairs of chromosomes.  相似文献

The karyotype and major ribosomal sites as revealed using silver staining of Anatolian leuciscine cyprinid fish Acanthobrama marmid were studied. The diploid chromosome number was invariably 2n = 50. Karyotype consisted of eight pairs of metacentric, 13 pairs of submetacentric and four pairs of subtelocentric to acrocentric chromosomes. The largest chromosome pair of the complement was subtelo-to acrocentric characteristically, which is a characteristic cytotaxonomic marker for representatives of the cyprinid lineage Leuciscinae. The nucleolar organizer regions (NORs) were detected in the telomeres of two pairs of medium sized submeta-to subtelocentric chromosomes. No heteromorphic sex chromosomes were found. The karyotype pattern of A. marmid is nearly identical to that found in most other representatives of the Eurasian leuciscine cyprinids, while the multiple NOR phenotype appears to be more derived as opposed to a uniform one, ubiquitous in this group.  相似文献

The razor clam Solen marginatus has a diploid chromosome number of 38. The karyotype consists of one metacentric/submetacentric, three submetacentric/metacentric, five submetacentric, one submetacentric/subtelocentric, one subtelocentric/submetacentric, six subtelocentric and two telocentric chromosome pairs. Staining with chromomycin A3 revealed bright positive bands subcentromerically in the long arms of one medium-sized subtelocentric pair, while DAPI staining showed uniform fluorescence in all chromosomes of the complement. Fluorescence in situ hybridization using an 18S-5.8S-28S rDNA probe locates these loci at the subcentromeric region of one subtelocentric pair and at the subtelomeric region of another subtelocentric pair.  相似文献

Cytogenetic analyses of Bryconamericus aff. iheringii specimens from the upper Paraná River basin (State of Paraná, Brazil) are provided. They had 2n = 52 chromosomes and two cytotypes with variations in their karyotypic formulae: cytotype I with 12 metacentric, 18 submetacentric, 8 subtelocentric and 14 acrocentric chromosomes with a fundamental number (FN) of 90; cytotype II with 8 metacentric, 28 submetacentric, 6 subtelocentric and 10 acrocentric chromosomes with a fundamental number (FN) of 94. Differences in C- and G-band patterns between the cytotypes, distinguishing marker chromosomes for each karyotype, were reported. The R-band pattern by 5-bromodeoxyuridine incorporation was obtained in chromosomes of the cytotype II sample. In some metaphases, the second pair of submetacentric chromosomes is distinctive: its short arm is heterochromatic (positive C-band), corresponding to a late replication region. In the same cytotype, a G- and R-band size heteromorphism w as recorded in the long arm of pair 9 (submetacentric). These methodologies revealed an actual karyotypic differentiation in the B. aff. iheringii population analyzed. Morphometrical comparative analyses and a discussion of evolutionary aspects of chromosome diversification in species of this genus are provided as well.  相似文献

A new karyotype for blind mole rats was recorded in Tunceli province in Eastern Turkey. The karyotype contained 44 chromosomes, including 13 biarmed pairs, 7 acrocentric pairs, and one heteromorphic pair with a submetacentric and an acrocentric homologue in the autosomal complement (FNa=69). The X chromosome was submetacentric and the Y chromosome medium-sized subtelocentric (FN=73). Distinct dark centromeric C-bands were observed on most of the biarmed and three pairs of the acrocentric autosomes. The NORs were detected on short arms of three subtelocentric pairs and one acrocentric pair of autosomes. The diploid number of chromosomes and the karyotype characteristics observed are obviously unique among hitherto studied populations of blind mole rats and the complement can be evaluated as a new chromosome race of Nannospalax xanthodon. The distribution ranges of individual chromosome races of the species recorded in Eastern Anatolia are revised and possible interracial hybridization is discussed in respect of the finding of a new race.  相似文献

The karyotype of the snow sculpin Myoxocephalus brandti, 2n = 44, NF = 46, from Peter the Great Bay was studied. Two-armed chromosomes were presented by one pair of metacentric chromosomes of medium size; one-armed chromosomes included two pairs of large subtelocentric chromosomes and a pair of large acrocentric chromosomes. Ag-NOR-staining in the telomere vicinity revealed nucleolus-organizing regions in one metacentric chromosome and in one medium size acrocentric chromosome in one of the fishes, in two homological small acrocentric chromosomes in three fishes, and in one acrocentric chromosome of average size in six fishes. No difference between the male and female karyotypes and any type of variability was revealed. The karyotypes of the snow sculpin M. brandti and the frog sculpin M. stelleri were compared. Six species/subspecies of Asian high-mountain voles, genus Alticola, were studied cytogenetically via conventional staining and C- and G-banding. The karyotypes are very similar. The standard karyotype, as in A. strelzovi strelzovi, consists of 56 chromosomes. These are split into 25 acrocentric pairs, one large subtelocentric pair, one small metacentric pair, a large acrocentric X chromosome, and a small Y chromosome, which varies in shape. Constitutive heterochromatin is almost entirely restricted to small centromeric regions. A small submetacentric pair of autosomes in both subspecies of A. semicanus and a medium-sized Y chromosome in A. argentatus severtzovi are of importance in systematics. The data suggest that A. barakshin, A. semicanus, and A. argentatus are separate species.  相似文献

The karyotype of one female Brachyteles arachinoides (E. Geoffroy, 1806) was studied. The specimen exhibited 62 chromosomes, which could be arranged in three clearly distinguishable groups: the first one including 5 pairs of subtelocentric chromosomes, the second one including 8 pairs of metacentric and submetacentric chromosomes and the third one including 18 pairs of acrocentric chromosomes. Chromosomes of C. glaucum from Baltic (Gdansk Bay) and Mediterranean (Thau lagoon, Sète) populations were studied using karyometric analysis and silver-staining. The karyotype of the Gdansk population consists of three metacentric, ten submetacentric and six subtelocentric chromosome pairs. The karyotype of the Thau population shows four metacentric, nine submetacentric and six subtelocentric pairs. Nucleolus organizer regions were found terminally on the second largest submetacentric chromosome pair of the Gdansk population and on the second largest metacentric chromosome pair of the Thau population. This suggests a cytotaxomomic difference between these two geographically isolated populations. Striking differences exist between C. glaucum and C. edule karyotypes, probably resulting from a long divergence time.  相似文献

Karyotypes of three species of the subfamily Clupeinae collected from northern Japan were analyzed by in vitro methods and their cellular DNA contents were measured using an integrating microdensitometer.Sardinella zunasi andSardinops melanostictus show very similar karyotypes: 2n = 48, consisting of acrocentric or subtelocentric chromosomes with a gradual decrease in chromosome size, but with differences in cellular DNA of 2.32 and 2.69pg/cell respectively.Clupea pallasii differs from the aforementioned species in karyotype: 2n = 52, consisting of 6 metacentric or submetacentric chromosomes and 46 acrocentric or subtelocentric chromosomes, with a cellular DNA content of 1.96 pg/cell. The results showed two different modes in karyological evolution within the subfamily Clupeinae, i.e. an increase of cellular DNA content without apparent change in karyotype, as shown bySardinella zunasi andSardinops melanostictus, and less change in cellular DNA content but with marked change in karyotype, as shown byClupea pallasii.  相似文献

Individuals of two populations of the fish Characidium cf. fasciatum were cytogenetically studied and showed a basic diploid number of 50 chromosomes. Some fishes were found to have 51 to 54 chromosomes due to the presence of one to four small subtelocentric/acrocentric supernumerary chromosomes. When analyzed by conventional Giemsa staining, male and female specimens of C. cf. fasciatum from the Quinta stream and Pardo River presented the same basic karyotypic macro- and microstructure, consisting of 32 metacentric and 18 submetacentric chromosomes. Ag-NORs were terminally located on the long arms of two submetacentric chromosome pairs. Constitutive heterochromatin was identified by C-banding as small pericentromeric blocks in the majority of the chromosomes, and B-chromosomes were found to be heterochromatic. The occurrence of one totally heterochromatic submetacentric chromosome restricted to females and considered as an unusual feature in fish karyotypes led to the identification of a ZZ/ZW sex-chromosome system. It has been proposed that the genera Zebrina and Setcreasea of the family Commelinaceae should be united and reunited, respectively, with the genus Tradescantia, mainly based on morphological studies. In the present study, karyotypes and Giemsa C-banding patterns in the root-tip cells of three Zebrina and two Setcreasea clones were analyzed, and were compared with those of a triploid Tradescantia clone. Z. pendula and Z. purpusii (both 2n = 24) were found to have similar karyotypes (4 M + 6 ST + 14 T; M = meta-, ST = subtelo-, T = telocentric chromosomes), while Z. pendula cv Quadricolor (2n = 23) had a unique karyotype (6 M + 5 ST + 11 T + 1 SA; SA = short acrocentric chromosome). The only clear difference between Z. pendula and Z. purpusii was that one and two subtelocentric chromosomes, respectively, had satellites at the short arms. Two clones of S. purpurea (2n = 24) had karyotypes (8 M + 8 M' + 8 SM; M' = nearly meta-, SM = submetacentric chromosomes) similar to each other. T. ohiensis (2n = 18) had a symmetric karyotype (9 M + 9 SM) consisting of larger chromosomes than S. purpurea. Many clear Giemsa C-bands were detected, in addition to centromeric bands in all chromosomes of all clones. Z. pendula and Z. purpusii commonly had single clear interstitial bands in eight telocentric chromosomes each, but they also had unique telomeric and other interstitial bands, respectively. Z. pendula cv Quadricolor had a unique banding pattern, i.e., satellite bands in the unique short chromosome, telomeric bands at the long arms of all metacentric chromosomes, and single interstitial bands in six telocentric chromosomes. Two clones of S. purpurea had telomeric bands at many chromosome arms and satellite bands in two nearly metacentric and one submetacentric chromosomes, but some differences were found between them. On the other hand, all the chromosomes of T. ohiensis had telomeric bands at both arms, and three submetacentric chromosomes had satellite bands. These result prove structural differentiation of chromosomes occurred among the clones, especially in Zebrina, and show that S. purpurea is relatively close to T. ohiensis, while Zebrina is obviously distant from the other two genera. Therefore, there remains a question cytologically at least for uniting Zebrina with Tradescantia.  相似文献

采用常规压片法对砂燕麦、西班牙燕麦和短燕麦3个二倍体燕麦种进行了核型研究。结果表明:砂燕麦染色体核型公式为2n=2x=14=10m+4sm(2SAT),具近中部和中部着丝点染色体,第4对染色体组的短臂上有1对随体,核不对称系数为68.17%;西班牙燕麦染色体核型公式为2n=2x=14=10m+4sm(2SAT),具近中部和中部着丝点染色体,第7对染色体短臂上有1对随体,核不对称系数为59.31%;短燕麦染色体核型公式为2n=2x=14=6m+4sm+4st(2SAT),具近端部、近中部和中部着丝点染色体,第6对染色体组的短臂上有1对随体,核不对称系数为63.91%。虽然3个燕麦种的核型均为2A,但它们的染色体形态有明显不同,比较认为砂燕麦相对进化,短燕麦次之,西班牙燕麦较原始。本研究对燕麦种质资源的核型分析及进化地位研究具有参考价值。  相似文献

Karyotypes of Diplomystes composensis and Diplomystes nahuelbutaensis were the same diploid number (n= 56).The chromosome formula for D. composensis was 16 metacentric + 24 submetacentric + 8 subtelocentric + 8 telocentric chromosomes and for D. nahuelbutaensis was 14 metacentric + 26 submetacentric + 8 subtelocentric +8 telocentric chromosomes. In contrast, the differences in the chromosomal C-banding patterns between these species was large. For instance, chromosome pairs 5,6, and 7 of D. nahuelbutaensis showed heterochromatic centromeres and pairs 23, 24, 27, and 28 were entirely heterochromotic. Diplomystes composensis showed conspicuous C-banded blocks in pairs 7, 24, and 25 (chromosome pair 7 had one heterochromatic arm, chromosome pair 24 was entirely heterochromatic, and chromosome pair 25 had heterochromatin close to centromere). Comparison with other ostariophysan karyotypes (e.g. gymnotiforms, characiforms, and cypriniforms), does not allow any conclusions about the ploesiomorphic catfish condition, because the karyotypes of the outgroups are too variable. A synapomorphy shared by characiforms, gymnotiforms, and diplomystid catfishes is the presence of more metacentric to submetacentric than substelocentric to telocentric chromosomes. Cypriniforms are more primitive because they have more subtelocentric to telocentric than metacentric to submetacentric chromosomes.  相似文献

The karyotype of a subspecies of the golden loach,Sabanejewia aurata balcanica from eastern Slovakia was studied by conventional Giemsa staining, Ag-NOR staining, and C-banding. The diploid chromosome number was 2n = 50. The karyotype comprised 2 pairs of metacentric, 6 pairs of submetacentric and 17 pairs of subtelocentric to acrocentric chromosomes. Both metacentric pairs and 2 large subtelocentric pairs had massive pericentromeric blocks, while all other elements had only weak blocks of heterochromatin. The NORs were localized on the short arms of one middle-sized subtelocentric pair. The karyotype ofS. a. balcanica differs from that ofS. aurata kubanica, suggesting chromosomal polymorphism of this widely distributed, polytypic cobitid species. The polymorphic karyotypes of the golden loach may thus demonstrate transient stages, linking primitive and advanced cobitid karyotypes.  相似文献

对伞形科前胡属(PeucedanumL.)2个种以及羌活属(NotopterygiumH.Boiss.)1个种3个居群的染色体数目和核型进行了研究。研究表明,它们的染色体数目均为2n=22,核型公式可分别表示为长前胡:2n=2x=22=22 m(1 SAT),属1A型;松潘前胡:2n=2x=22=20 m 2 sm,属2A型;宽叶羌活的3个居群分别是:马边大风顶居群1为2n=2x=22=6 m 12 sm 4 st,属2A型;马边大风顶居群2为2n=2x=22=12 m 4 sm 6 st,属2B型;屏山老君山居群为2n=2x=22=4 m 14 sm 4 st,属2A型。其中长前胡和松潘前胡的染色体数目和核型为首次报道。  相似文献
