This paper concerns tolerance to 50–200 mM NaCl of submerged rice (Oryza sativa cv. Amaroo) during germination and the first 138–186 h of development in aerated solution. Rice was able to germinate and the seedlings even tolerated exposure to 200 mM NaCl, albeit with severe growth restrictions. After return to 0.3 mM NaCl, growth increased, indicating that even at 200 mM NaCl there was no irreparable injury. Osmotic adjustment was achieved by using Na+ and Cl as the major osmotica. At 200 mM NaCl commenced at sowing, the shoot Na+ and Cl concentrations between 50–110 h were about 210 and 260 mM, respectively, i.e. above the external concentration. Thus, there was a high tissue tolerance to NaCl. The internal concentrations declined subsequently, concurrent with a decline in growth. At 50–200 mM NaCl, the contributions from ions to πsap were 81–92% in roots and 62–74% in shoots. The assessed turgor pressures at 200 mM NaCl were 0.33 MPa in shoots and 0.15 MPa in roots, compared to 0.62 and 0.43 MPa at 0.3 mM NaCl. In the General Discussion section, we compare the different responses of submerged seedlings to the responses of transpiring rice plants, reported in the literature, and suggest that the submerged system is useful to evaluate effects of NaCl on turgor pressure and particularly to establish whether there are specific effects of Na+ and Cl in tissues.  相似文献

Grafting onto salt‐tolerant pumpkin rootstock can increase cucumber salt tolerance. Previous studies have suggested that this can be attributed to pumpkin roots with higher capacity to limit the transport of Na+ to the shoot than cucumber roots. However, the mechanism remains unclear. This study investigated the transport of Na+ in salt‐tolerant pumpkin and salt‐sensitive cucumber plants under high (200 mM) or moderate (90 mM) NaCl stress. Scanning ion‐selective electrode technique showed that pumpkin roots exhibited a higher capacity to extrude Na+, and a correspondingly increased H+ influx under 200 or 90 mM NaCl stress. The 200 mM NaCl induced Na+/H+ exchange in the root was inhibited by amiloride (a Na+/H+ antiporter inhibitor) or vanadate [a plasma membrane (PM) H+‐ATPase inhibitor], indicating that Na+ exclusion in salt stressed pumpkin and cucumber roots was the result of an active Na+/H+ antiporter across the PM, and the Na+/H+ antiporter system in salt stressed pumpkin roots was sufficient to exclude Na+. X‐ray microanalysis showed higher Na+ in the cortex, but lower Na+ in the stele of pumpkin roots than that in cucumber roots under 90 mM NaCl stress, suggesting that the highly vacuolated root cortical cells of pumpkin roots could sequester more Na+, limit the radial transport of Na+ to the stele and thus restrict the transport of Na+ to the shoot. These results provide direct evidence for pumpkin roots with higher capacity to limit the transport of Na+ to the shoot than cucumber roots.  相似文献

Summary The effects of increasing concentrations of NaCl and CaCl2 on quince (Cydonia oblonga Mill. BA 29 clone) somatic embryogenesis and adventitious root regeneration were investigated. Leaves collected from in vitro-grown shoots were used as explants and induced for 2d in liquid Murashige and Skoog medium containing 11.3 μM 2,4-dichlorophenoxyacetic acid. Explants were then cultured on semisolid Murashige and Skoog medium enriched with 4.7 μM kinetin and 0.5 μM naphthaleneacetic acid under red light for 25 d and under white light for another 25 d. Two experiments were performed: in the first, NaCl was used at 0,25, 50, 100, and 200 mM in factorial combination with CaCl2 at 3, 9, and 27 mM; in the second, NaCl was applied at 0, 5, 10, 20, 40, and 80 mM in combination with CaCl2 at 0.3, 1.0, and 3.0 mM. Quince leaves revealed the capacity to regenerate somatic embryos and/or adventitious roots. Quantitative and qualitative regeneration from leaves was affected by NaCl treatments: increasing NaCl concentrations, in combination with CaCl2 at 1 mM, led to an increase in the proportion of leaves producing somatic embryos only, and to a decrease of both leaves regenerating roots only and leaves simultaneously producing somatic embryos and adventitious roots. This suggests a beneficial effect of salt stress on the embryogenic process. The regeneration response decreased with increasing salt concentrations and was almost totally inhibited above 50 mM NaCl and 9 mM CaCl2. The presence of CaCl2 in the culture medium apparently mitigated the effects of salt stress, but only when NaCl was applied at 40 mM. NaCl at 5 mM, in the presence of 0.3 or 1 mM CaCl2, was favorable both to somatic embryo and root production. No value of the ratio Na+/Ca2+ was found to be optimal for the regeneration processes.  相似文献

Na+ transport across the tonoplast and its accumulation in the vacuoles is of crucial importance for plant adaptation to salinity. Mild and severe salt stress increased both ATP- and PPi-dependent H+ transport in tonoplast vesicles from sunflower seedling roots, suggesting the possibility that a Na+/H+ antiport system could be operating in such vesicles under salt conditions (E. Ballesteros et al. 1996. Physiol. Plant. 97: 259–268). During a mild salt stress, Na+ was mainly accumulated in the roots. Under a more severe salt treatment, Na+ was equally distributed in shoots and roots. In contrast to what was observed with Na+, all the salt treatments reduced the shoot K+ content. Dissipation by Na+ of the H+ gradient generated by the tonoplast H+-ATPase, monitored as fluorescence quenching of acridine orange, was used to measure Na+/H+ exchange across tonoplast-enriched vesicles isolated by sucrose gradient centrifugation from sunflower (Helianthus annuus L.) roots treated for 3 days with different NaCl regimes. Salt treatments induced a Na+/H+ exchange activity, which displayed saturation kinetics for Na+ added to the assay medium. This activity was partially inhibited by 125 μM amiloride, a competitive inhibitor of Na+/H+ antiports. No Na+/H+ exchange was detected in vesicles from control roots. The activity was specific for Na+. since K+ added to the assay medium slightly dissipated H+ gradients and displayed non-saturating kinetics for all salt treatments. Apparent Km for Na+/H+ exchange in tonoplast vesicles from 150 mM NaCl-treated roots was lower than that of 75 mM NaCl-treated roots, Vmax remaining unchanged. The results suggest that the existence of a specific Na+/H+ exchange activity in tonoplast-enriched vesicle fractions, induced by salt stress, could represent an adaptative response in sunflower plants, moderately tolerant to salinity.  相似文献

Above-canopy sprinkler irrigation with saline water favours the absorption of salts by wetted leaves and this can cause a yield reduction additional to that which occurs in salt-affected soils. Outdoor pot experiments with both sprinkler and drip irrigation systems were conducted to determine foliar ion accumulation and performance of maize and barley plants exposed to four treatments: nonsaline control (C), salt applied only to the soil (S), salt applied only to the foliage (F) and salt applied to both the soil and to the foliage (F+S). The EC of the saline solution employed for maize in 1993 was 4.2 dS m–1 (30 mM NaCl and 2.8 mM CaCl2) and for barley in 1994, 9.6 dS m–1 (47 mM NaCl and 23.5 mM CaCl2). The soil surface of all pots was covered so that in the F treatment the soil was not salinized by the saline sprinkling and drip irrigation supplied nutrients in either fresh (treatments C and F) or saline water (treatments S and F+S).Saline sprinkling increased leaf sap Na+ concentrations much more than did soil salinity, especially in maize, even though the saline sprinkling was given only two or three times per week for 30 min, whereas the roots of plants grown in saline soil were continuously exposed to salinity. By contrast, leaf sap Cl concentrations were increased similarly by saline sprinkling and soil salinity in maize, and more by saline sprinkling than saline soil in barley. It is concluded that barley leaves, and to a greater extent maize leaves, lack the ability to selectively exclude Na+ when sprinkler irrigated with saline water. Moreover, maize leaves selectively absorbed Na+ over Cl whereas barley leaves showed no selectivity. When foliar and root absorption processes were operating together (F+S treatment) maize and barley leaves accumulated 11–14% less Na+ and Cl than the sum of individual absorption processes (treatment F plus treatment S) indicating a slight interaction between the absorption processes. Vegetative biomass at maturity and cumulative plant water use were significantly reduced by saline sprinkling. In maize, reductions in biomass and plant water use relative to the control were of similar magnitude for plants exposed only to saline sprinkling, or only to soil salinity; whereas in barley, saline sprinkling was more detrimental than was soil salinity. We suggest that crops that are salt tolerant because they possess root systems which efficiently restrict Na+ and Cl transport to the shoot, may not exhibit the same tolerance in sprinkler systems which wet the foliage with saline water. ei]T J Flowers  相似文献

The rise of sap in mangroves has puzzled plant physiologists for many decades. The current consensus is that negative pressures in the xylem exist which are sufficiently high to exceed the osmotic pressure of seawater (2.5 MPa). This implies that the radial reflection coefficients of the mangrove roots are equal to unity. However, direct pressure probe measurements in xylem vessels of the roots and stems of mangrove (Rhizophora mangle) grown in the laboratory or in the field yielded below-atmospheric, positive (absolute) pressure values. Slightly negative pressure values were recorded only occasionally. Xylem pressure did not change significantly when the plants were transferred from tap water to solutions containing up to 1700 mOsmol kg?1 NaCl. This indicates that the radial reflection coefficient of the roots for salt, and therefore the effective osmotic pressure of the external solution, was essentially zero as already reported for other halophytes. The low values of xylem tension measured with the xylem pressure probe were consistent with previously published data obtained using the vacuum/leafy twig technique. Values of xylem tension determined with these two methods were nearly two orders of magnitude smaller than those estimated for mangrove using the pressure chamber technique (?3 to ?6MPa). Xylem pressure probe measurements and staining experiments with alcian blue and other dyes gave strong evidence that the xylem vessels contained viscous, mucilage- and/or protein-related compounds. Production of these compounds resulting from wound or other artifactual reactions was excluded. The very low sap flow rates of about 20–50 cm h?1 measured in these mangrove plants were consistent with the presence of high molecular weight polymeric substances in the xylem sap. The presence of viscous substances in the xylem sap of mangroves has the following implications for traditional xylem pressure measurement techniques, development of xylem tension, and longdistance water transport: (1) high external balancing pressures in the pressure chamber are needed to force xylem sap to the cut surface of the twig; (2) stable tensions much larger than 0.1 MPa can be developed only occasionally because viscous solutions provide nucleation sites for gas bubble formation; (3) the frequent presence of small gas bubbles in viscous solutions allows water transport by interfacial, gravity-independent streaming at gas/water interfaces and (4) the increased density of viscous solutions creates (gravity-dependent) convectional flows. Density-driven convectional flows and interfacial streaming, but also the very low radial reflection coefficient of the roots to NaCl are apparently the means by which R. mangle maintains water transport to its leaves despite the high salinity of the environment.  相似文献

The aim of this study was to determine the range of NaCl concentrations in the nutrient solution that allow Suaeda altissima (L.) Pall., a salt-accumulating halophyte, to maintain the upward gradient of water potential in the “medium-root-leaf” system. We evaluated the contribution of Na+ ions in the formation of water potential gradient and demonstrated that Na+ loading into the xylem is involved in this process. Plants were grown in water culture at NaCl concentrations ranging from zero to 1 M. The water potential of leaf and root cells was measured with the method of isopiestic thermocouple psychrometry. When NaCl concentration in the growth medium was raised in the range of 0–500 mM (the medium water potential was lowered accordingly), the root and leaf cells of S. altissima decreased their water potential, thus promoting the maintenance of the upward water potential gradient in the medium-root-leaf system. Growing S. altissima at NaCl concentrations f 750 mM and 1 M disordered water homeostasis and abolished the upward gradient of water potential between roots and leaves. At NaCl concentrations of 0–250 mM, the detached roots of S. altissima were capable of producing the xylem exudate. The concentration of Na+ in the exudate was 1.3 to 1.6 times higher than in the nutrient medium; the exudate pH was acidic and was lowered from 5.5 to 4.5 with the rise in the salt concentration. The results indicate that the long-distance Na+ transport and, especially, the mechanism of Na+ loading into the xylem play a substantial role in the formation of water potential gradient in S. altissima. Reed plants (Phragmites australis Trinius) grow not only in fresh and brackish water areas but also in arid and high salinity regions. Reed plants obtained from a riverside (Utsunomiya) were damaged by 257 mM NaCl, whereas desert plants (Nanpi) were not. When the plants were grown under salt stress, the shoots of the Utsunomiya plants contained high levels of sodium and low levels of potassium, whereas the upper part of the Nanpi plants contained low levels of sodium and high levels of potassium. One month salt stress did not affect potassium contents in either Utsunomiya or Nanpi plants, but it did dramatically increase sodium contents only in the Utsunomiya plants. The ratio of K+ to Na+ was maintained at a high level in the upper parts of the Nanpi plants, whereas the ratio markedly decreased in the Utsunomiya plants in the presence of NaCl. Accumulation of Na+ in the roots and Na+ efflux from the roots were greater in the Nanpi plants than in the Utsunomiya plants. These results suggest that the salt tolerance mechanisms of Nanpi reed plants include an improved ability to take up K+ to prevent an influx of Na+ and an improved ability to exclude Na+ from the roots.  相似文献

To explore the mechanisms of 5‐aminolevulinic acid (ALA)‐improved plant salt tolerance, strawberries (Fragaria × ananassa Duch. cv. ‘Benihoppe’) were treated with 10 mg l?1 ALA under 100 mmol l?1 NaCl stress. We found that the amount of Na+ increased in the roots but decreased in the leaves. Laser scanning confocal microscopy (LSCM) observations showed that ALA‐induced roots had more Na+ accumulation than NaCl alone. Measurement of the xylem sap revealed that ALA repressed Na+ concentrations to a large extent. The electron microprobe X‐ray assay also confirmed ALA‐induced Na+ retention in roots. qRT‐PCR showed that ALA upregulated the gene expressions of SOS1 (encoding a plasma membrane Na+/H+ antiporter), NHX1 (encoding a vacuolar Na+/H+ antiporter) and HKT1 (encoding a protein of high‐affinity K+ uptake), which are associated with Na+ exclusion in the roots, Na+ sequestration in vacuoles and Na+ unloading from the xylem vessels to the parenchyma cells, respectively. Furthermore, we found that ALA treatment reduced the H2O2 content in the leaves but increased it in the roots. The exogenous H2O2 promoted plant growth, increased root Na+ retention and stimulated the gene expressions of NHX1, SOS1 and HKT1. Diphenyleneiodonium (DPI), an inhibitor of H2O2 generation, suppressed the effects of ALA or H2O2 on Na+ retention, gene expressions and salt tolerance. Therefore, we propose that ALA induces H2O2 accumulation in roots, which mediates Na+ transporter gene expression and more Na+ retention in roots, thereby improving plant salt tolerance.  相似文献

  • In the present work, the influence of inoculation with the symbiotic fungus, Piriformospora indica, on gas exchange, water potential, osmolyte content, Na/K ratio and chlorophyll fluorescence of tomato plants under three salinity levels (0, 50, 100 and 150 mm NaCl) and three time periods (5, 10 and 15 days after exposure to salt) was investigated.
  • Results indicate that P. indica inoculation improved growth parameters of tomato under salinity stress. This symbiotic fungus significantly increased photosynthetic pigment content under salinity, and more proline and glycine betaine accumulated in inoculated roots than in non‐inoculated roots. P. indica further significantly improved K+ content and reduced Na+ level under salinity treatment. After inoculation with the endophytic fungus, leaf physiological parameters, such as water potential, net photosynthesis, stomatal conductance and transpiration, were all higher under the salt concentrations and durations compared with controls without P. indica. With increasing salt level and salt treatment duration, values of F0 and qP increased but Fm, Fv/Fm, F′v/F′m and NPQ declined in the controls, while inoculation with P. indica improved these values.
  • The results indicate that the negative effects of NaCl on tomato plants were alleviated after P. indica inoculation, probably by improving physiological parameters such as water status and photosynthesis.

Summary The effects of salinity in the root medium, time, and relative humidity on the salt secretion of Glaux maritima were investigated. Both in the greenhouse and in the field increasing salinity stimulated sodium and chloride secretion, whereas the essential elements potassium, calcium, and magnesium remained at low secretion levels, which might be interpreted as efficient mineral economy. The low secretion level of potassium is remarkable, because growing on a nutrient solution containing 6 mM potassium, the concentration of the plant sap increases to 150 mM K+ and the secreted quantity amounts to only 2 m mol l-1 plant sap 24 h-1.Attempts were made to establish the secretion rate. The maximum secretion rate calculated may be 80 pEq NaCl cm-2 s-1, but for long periods (days) the secretion rate will be lower. Measurement of salt secretion unavoidably leads to removal of secreted salt. Salt was removed by rinsing with distilled water, which artificially accelerates the secretion process or parts of it by diffusion of salt from the cuticle cavity or secretory cells. At increasing salinities the amount of secreted ions showed a fivefold increase, whereas the osmotic potential of the plant sap was raised only twofold, indicating the importance of secretion as a rapid regulation mechanism with regard to the salt economy.  相似文献

Solution culture-grown, six-month old jack pine (Pinus banksiana Lamb.) seedlings were treated with naphthenic acids (NAs) (150 mg l–1) and sodium chloride (45 mM NaCl) which were applied together or separately to roots for four weeks. NAs aggravated the effects of NaCl in inhibiting stomatal conductance (g s) and root hydraulic conductance (Kr). Naphthenic acids did not affect needle and root electrolyte leakage in the absence of NaCl. However, in plants treated with NaCl, NAs further increased electrolyte leakage from needles and NaCl induced electrolyte leakage from needles, but not from roots. Both NaCl and NAs treatments resulted in a reduction in root respiration. The measured Na+ and Cl concentrations in the shoots for combined NaCl + NAs treatments were lower than in NaCl-only treatments. These decreases were correlated with a reduction in water conductance. The accumulation of Na+ and Cl in shoots was accompanied by an increased in needle electrolyte leakage. However, greater concentrations of Cl compared with Na+ were present in shoots and in the xylem sap suggesting that roots had relatively lower capacity for Cl storage compared with Na+.  相似文献

Summary The composition of the vacuolar sap of Chara vulgaris growing in a brackish water lake was estimated weekly over 2 years (1984–1985). The ionic concentrations of the main cations Na+, K+, Ca2+, and Mg2+ and the anion Cl- varied depending on cell age, developmental state, and season. The average of all measurements (in mM) was Na+: 35, K+: 106, Ca2+: 7, Mg2+: 23, Cl-: 101, SO 2- 4 : 20, and PO 3- 4 : 5. At the onset of growth in May/June the ionic content was lower compared to the mean value for the year, steadily increasing until it reached its maximum above the annual mean in winter. During the period of fructification (sexual reproduction: formation of antheridia and oogonia), when up to 100 mM sucrose was accumulated in the vacuolar sap, ionic content was lowest. This resulted in a fairly constant osmotic potential throughout the year. Mg2+ and Ca2+ concentrations were correlated with the physiological age of the cells.  相似文献

Reaumuria vermiculata (L.), a perennial dwarf shrub in the family of Tamaricaceae, is a salt-secreting xero-halophyte found widely in arid areas of Tunisia. In the present study, physiological attributes of R. vermiculata were investigated under salt stress. Four-month-old plants were subjected to various salinity levels (0, 100, 200, 300, 400 or 600 mM NaCl) for 30 days under greenhouse conditions. Results showed that plants grew optimally when treated with standard nutrient solution without NaCl supply. However, increasing osmolality of nutrient solutions caused a significant reduction in biomass production and relative growth rate. This reduction was more pronounced in roots than in shoots. In addition, this species was able to maintain its shoot water content at 30% of the control even when subjected to the highest salt level, whereas root water content seemed to be unaffected by salt. Shoot water potential declined significantly as osmotic potential of watering solutions was lowered and the more negative values were reached at 600 mM NaCl (−3.4 MPa). Concentrations of Na+ and Cl in the shoots of R. vermiculata were markedly increased with increasing osmolality of nutrient solutions, whereas concentration of K+ was not affected by NaCl supply. Salt excretion is an efficient mechanism of Na+ exclusion from the shoots of this species exhibiting high K+/Na+ selectivity ratio over a wide range of NaCl salinity. Proline accumulation in shoots was significantly increased with increase in salt level and may play a role in osmoregulation.  相似文献

The relative importance of endogenous abscisic acid (ABA), as well as Na+ and Cl in NaCl-induced responses related to growth in roots of rice seedlings were investigated. The increase in ammonium, proline and H2O2 levels, and cell wall peroxidase (POD) activity has been shown to be related to NaCl-inhibited root growth of rice seedlings. Increasing concentrations of NaCl from 50 to 150 mM progressively decreased root growth and increased both Na+ and Cl. Treatment with NaCl in the presence of 4,4-diisothiocyano-2,2-disulfonic acid (DIDS, a nonpermeating amino-reactive disulfonic acid known to inhibit the uptake of Cl) had less Cl level in roots than that in the absence of DIDS, but did not affect the levels of Na+, and responses related to growth in roots. Treatment with 50 mM Na-gluconate (the anion of which is not permeable to membrane) had similar Na+ level in roots as that with 100 mM NaCl. It was found that treatment with 50 mM Na-gluconate effected growth reduction and growth-related responses in roots in the same way as 100 mM NaCl. All these results suggest that Cl is not required for NaCl-induced responses in root of rice seedlings. Endogenous ABA level showed no increase in roots of rice seedlings exposed to 150 mM NaCl. It is unlikely that ABA is associated with NaCl-inhibited root growth of rice seedlings.  相似文献

Summary Callus of the mangrove plant, Sonneratia alba J. Smith, established from pistils of flower buds were cultured on solid Murashige and Skoog medium supplemented with 0 to 500 mM NaCl. Maximum growth was observed with 50 mM NaCl, and net growth of callus occurred for concentrations up to 200 mM NaCl. At 500 mM NaCl, growth of callus was completely inhibited, although a part of the tissue was still alive after 30 d. Cellular levels of Na+ and Cl were greatly increased by the treatment with NaCl. Uptake of K+ was also enhanced and was accompanied by increasing levels of Na+ and Cl so that the Na+/K+ ratio was almost constant (4.1–4.2) in callus grown with 50–200 mM NaCl. Levels of Mg2+ and Ca2+ were not changed significantly with 50–200 mM NaCl, whereas levels of free NH 4 + , NO 3 and SO 4 2− ions, which are convertible to organic compounds, were lowest in callus grown with 50 mM NaCl. The rate of conversion of 15NH 4 + into macromolecules during 30 d culture with 0–100 mM NaCl did not vary greatly, but 200 mM NaCl reduced the biosynthesis of macromolecules from this ion. The highest rate of conversion of 15NO 3 into macromolecules was observed at 50 mM NaCl. Identification of compatible solutes with NMR-spectroscopy indicated that mannitol is the compatible solute for intact plants of Sonneratia alba, but no accumulation of mannitol was found in calluses, not even in those grown at high concentrations of NaCl.  相似文献

Caulogenesis was induced on in vitro-grown leaves treated for 2d in liquid Murashige and Skoog (MS) medium with 11.3 μM 2,4-dichlorophenoxyacetic acid and cultured on MS gelled medium supplemented with 4.5 μM thidiazuron and 0.5 μM naphthaleneacetic acid. Three experiments were performed: in the first, we compared the effects of NaCl at 0, 25, 50, 100, and 200 mM in factorial combination with 3, 9, and 27 mM CaCl2. In the second, NaCl was tested at 0, 5, 10, 20, 40, and 80 mM with CaCl2 at 0.3, 1.0, and 3.0 mM. The third experiment was carried out with the same experimental design as the second one but replacing NaCl with Na2SO4. Shoot regeneration was evaluated after 50 d of culturing: 25 in darkness and 25 in white light. In the first experiment, shoot regeneration was very poor and was observed only at the lower salt concentrations. In the second experiment, the percentages of caulogenic leaves were much higher, but decreased with increasing NaCl concentration. The more pronounced negative effect of the highest NaCl concentrations appeared to be partly mitigated by CaCl2 at 1 and 3 mM. The presence of 3 mM CaCl2, in the experiment with Na2SO4, appeared to be even more effective in reducing the adverse effect of sodium stress on caulogenesis. This result was attributed to the lower Cl concentration in the growth medium, which resulted from replacing NaCl with Na2SO4. NaCl applied at low concentrations (5 and 10 mM) in combination with 3 mM CaCl2 exerted a favorable effect on adventitious shoot regeneration. As regards the Na+ and Ca2+ interaction, when the Na+/Ca2+ ratio was below roughly 35 and 20, with NaCl and Na2SO4, respectively, at least 60% of leaves showed regenerating capacity, but optimal values of this ratio were not derived.  相似文献

以当年生圆柏幼苗为实验材料,采用温室调控盆栽土培法研究了不同浓度NaCl(0、100、200、300mmol·L-1)胁迫21d对其生长情况及不同器官(根、茎、叶)中K~+、Na~+、Ca~(2+)和Mg~(2+)的吸收和分配的影响,以探讨圆柏幼苗对盐环境的生长适应性及耐盐机制。结果表明:(1)随着NaCl胁迫浓度的增加,圆柏幼苗生长,包括株高、地径、相对生长量以及生物量的积累均呈下降趋势,而其根冠比却增加。(2)在各浓度NaCl胁迫处理下,圆柏幼苗根、茎、叶中Na~+含量较对照均显著增加,而且叶中Na~+含量显著高于茎和根,叶中Na~+含量是根中的5倍。(3)随着NaCl胁迫浓度的升高,圆柏幼苗各器官中K~+、Ca~(2+)和Mg~(2+)含量以及K~+/Na~+、Ca~(2+)/Na~+及Mg~(2+)/Na~+比值均呈下降趋势。(4)在NaCl胁迫条件下,圆柏幼苗根系离子吸收选择性系数SK,Na、SCa,Na、SMg,Na显著提高,茎、叶离子转运选择性系数SCa,Na、SMg,Na则逐渐降低,叶中离子转运选择性系数SK,Na则随着NaCl胁迫浓度的升高显著降低,大量Na~+进入地上部,减缓了盐胁迫对根系的伤害。研究认为,圆柏幼苗的盐适应机制主要是通过根系的补偿生长效应及茎、叶对Na~+的聚积作用来实现的,同时也与根对K~+、Ca~(2+)、Mg~(2+)的选择性运输能力增强和茎、叶稳定的K~+、Ca~(2+)、Mg~(2+)的选择性运输能力有关。  相似文献

Glucose‐6‐phosphate dehydrogenase (G6PDH) is important for the activation of plant resistance to environmental stresses, and ion homeostasis is the physiological foundation for living cells. In this study, we investigated G6PDH roles in modulating ion homeostasis under salt stress in Carex moorcroftii callus. G6PDH activity increased to its maximum in 100 mM NaCl treatment and decreased with further increased NaCl concentrations. K+/Na+ ratio in 100 mM NaCl treatment did not exhibit significant difference compared with the control; however, in 300 mM NaCl treatment, it decreased. Low‐concentration NaCl (100 mM) stimulated plasma membrane (PM) H+‐ATPase and NADPH oxidase activities as well as Na+/H+ antiporter protein expression, whereas high‐concentration NaCl (300 mM) decreased their activity and expression. When G6PDH activity and expression were reduced by glycerol treatments, PM H+‐ATPase and NADPH oxidase activities, Na+/H+ antiporter protein level and K+/Na+ ratio dramatically decreased. Simultaneously, NaCl‐induced hydrogen peroxide (H2O2) accumulation was abolished. Exogenous application of H2O2 increased G6PDH, PM H+‐ATPase and NADPH oxidase activities, Na+/H+ antiporter protein expression and K+/Na+ ratio in the control and glycerol treatments. Diphenylene iodonium (DPI), the NADPH oxidase inhibitor, which counteracted NaCl‐induced H2O2 accumulation, decreased G6PDH, PM H+‐ATPase and NADPH oxidase activities, Na+/H+ antiporter protein level and K+/Na+ ratio. Western blot result showed that G6PDH expression was stimulated by NaCl and H2O2, and blocked by DPI. Taken together, G6PDH is involved in H2O2 accumulation under salt stress. H2O2, as a signal, upregulated PM H+‐ATPase activity and Na+/H+ antiporter protein level, which subsequently resulted in the enhanced K+/Na+ ratio.
