Ward, D. A. and Drake, B. G. 1987. Photoinhibition under atmosphericO2, the activation state of RuBP carboxylase and the contentof photosynthetic intermediates in soybean and wheat.—J.exp. Bot. 38: 1937–1948. Associations between photosynthesis, the activation state ofRuBP carboxylase and the contents of photosynthetic intermediateswere compared in soybean and wheat leaves before and after exposureto photoinhibitory treatments in the presence of atmosphericO2. Exposing attached leaves to a supra-saturating irradiance(3 800 µmol quanta m– 2 s–1) for 2 h in CO2-freeair decreased carboxylation efficiency and the light-saturatedphotosynthetic rate in air by approximately 50%. Exposure tothe photoinhibitory treatment for periods in excess of 2 h didnot cause a further decrease of photosynthesis in soybean. Althoughphotosynthesis was reduced, the initial and total (fully-activated)activities of ribulose 1,5-bisphosphate carboxylase (RuBPCase)in leaf extracts were unaltered in each species by the photoinhibitorytreatment. This was true for leaves sampled under both air andat a rate-limiting intercellular CO2 partial pressure (Ci) of75 µPa Pa–1. The contents of ribulose l,5-bisphosphate(RuBP) and 3-phosphoglyceric acid (3-PGA) were reduced by thephotoinhibitory treatment in soybean leaves sampled in air andat a rate-limiting Ci, although the RuBP/3-PGA ratio was unaffected.The relative reduction of RuBP content in soybean leaves atrate-limiting C1 was similar to the corresponding reductionof carboxylation efficiency. For wheat,the relative reductionof RuBP content at rate-limiting Ci (–19%) caused by thephotoinhibitory treatment was considerably less than the correspondingdecrease of carboxylation efficiency (–49%).The RuBP/3-PGAratio of wheat was also increased significantly by the photoinhibitorytreatment The significance of these observations to the regulationof CO2-limited photosynthesis in leaves experiencing photoinhibitionunder atmospheric oxygen is discussed. Consideration is alsogiven to the previous contention that contemporary measurementsof initial activity in crude extracts may provide a spuriousindication of the amount of the enzyme-CO2-Mg2 + form of RuBPcarboxylase present in the leaf. Lehnherr, B., Mächler, F. and Nösberger, J. 1985.Influence of temperature on the ratio of ribulose bisphosphatecarboxylase to oxygenase activities and on the ratio of photosynthesisto photorespiration of leaves.—J. exp. Bot. 36: 1117–1125. Rates of net and gross photosynthesis of intact white cloverleaves were measured by infrared gas analysis and by short termuptake of 14CO2 respectively. Ribulose bisphosphate carboxylaseoxygenase (RuBPCO) was purified from young leaves and kineticproperties investigated in combined and separate assays. Theratio of carboxylase to oxygenase activities was compared withthe ratio of photosynthesis to photorespiration at various temperaturesand CO2 concentrations. The ratio of photosynthesis to photorespiration at 30 Pa p(CO2)was consistent with the ratio of carboxylase activity to oxygenaseactivity when each was measured above 20 °C. However, theratio of photosynthesis to photorespiration increased with decreasingtemperature, whereas the ratio of carboxylase to oxygenase activitywas independent of temperature. This resulted in a disagreementbetween the measurements on the purified enzyme and intact leafat low temperature. No disagreement between enzyme and leafat low temperature occurred, when the ratio of photosynthesisto photorespiration was determined at increased CO2 concentrations. The results suggest an effect of low temperature and low CO2concentration on the ratio of photosynthesis to photorespirationindependent of the enzyme. Discrepancies betweenmeasurements made in vitro and in vivo are attributed to theeffect of a CO2 concentrating system in the leaf cells. Interferencefrom dark respiration is discussed. The rate of net photosynthesis (P) of whole plant stands oftomato (Lycopersicon esculentum Mill.), cucumber (Cucumis sativusL.) and sweet pepper (Capsicum annuum L.) was measured in sixlong-term experiments in large greenhouses under normal operatingconditions and CO2-concentrations between 200 and 1200 µmolmol-1. The objective was to quantify the responses to lightand carbon dioxide and to obtain data sets for testing simulationmodels. The method of measuring canopy photosynthesis involvedan accurate estimation of the greenhouse CO2 balance, usingnitrous oxide (N2O) as tracer gas to determine, on-line, theexchange rate between greenhouse and outside air. The estimatedrelative error in the observed P was about ± 10%, exceptthat higher relative errors could occur under particular conditions. A regression equation relating P to the photosynthetically activeradiation, the CO2 concentration and the leaf area index explained83-91% of the variance. The main canopy photosynthesis characteristicscalculated with the fitted regression equations were: canopyPmax 5-9 g m-2 h-1 CO2 uptake; ratio Pmax/LAI 1·5-3 gm-2 h-1; light compensation point 32-86 µmol s-1 m-2;light use efficiency (quantum yield) at low light 0·06-0·10µmol µmol-1 and CO2 compensation point 18-54 µmolmol-1. The effects of elevated carbon dioxide (CO2 and ozone (O3) onsoybean (Glycine max (L.) Merr.] photosynthesis and photorespiration-relatedparameters were determined periodically during the growing seasonby measurements of gas exchange, photorespiratory enzyme activitiesand amino acid levels. Plants were treated in open-top fieldchambers from emergence to harvest maturity with seasonal meanconcentrations of either 364 or 726 µmol mol–1 CO2in combination with either 19 or 73 nmol mol–1 O3 (12h daily averages). On average at growth CO2 concentrations,net photosynthesis (A) increased 56% and photorespiration decreased36% in terminal mainstem leaves with CO2 enrichment. Net photosynthesisand photorespiration were suppressed 30% and 41%, respectively,by elevated O3 during late reproductive growth in the ambientCO2 treatment, but not in the elevated CO2 treatment. The ratioof photorespiration to A at growth CO2 was decreased 61% byelevated CO2 There was no statistically significant effect ofelevated O3 on the ratio of photorespiration to A. Activitiesof glycolate oxidase, hydroxypyruvate reductase and catalasewere decreased 10–25% by elevated CO2 and by 46–66%by elevated O3 at late reproductive growth. The treatments hadno significant effect on total amino acid or glycine levels,although serine concentration was lower in the elevated CO2and O3 treatments at several sampling dates. The inhibitoryeffects of elevated O3 on photorespiration-related parameterswere generally commensurate with the O3-induced decline in A.The results suggest that elevated CO2 could promote productivityboth through increased photoassimilation and suppressed photorespiration. Exposure to atmospheric conditions which promote photorespirationstrongly inhibits photosynthesis in a mutant of Arabidopsislacking mitochondrial serine transhydroxymethylase activity,and glycine accumulates as a stable end-product of photorespiratorycarbon and nitrogen flow. By providing exogenous serine andammonia to leaves of the mutant, wild-type photosynthesis ratescan be temporarily maintained in the absence of photorespiratoryCO2 evolution. In these circumstances, the rate of glycine accumulationprovides a direct measure of photorespiratory flux which isnot complicated by the efflux and refixation of photorespiredCO2, the dilution of radioactive label by endogenous metabolicpools, or non-specific effects of metabolic inhibitors. At thestandard atmospheric concentration of CO2, the rate of glycineaccumulation in the mutant was proportional to the oxygen concentration,amounting to 53% of the rate of gross CO2-fixation at 21% O2.At normal levels of O2, glycine accumulation was maximal atabout 475 µl CO21–1 and was reduced at higher orlower CO2 concentrations, being almost abolished at 3000µ1CO21–1. These observations are discussed in the contextof a model of photorespiration based on the properties of ribulose1, 5-bisphosphate carboxylase/oxygenase, and in relation tothe results of previous attempts to measure photorespiration.Preliminary evidence from 14CO2-labelling experiments whichsuggests a non-photorespiratory pathway of serine synthesisis also presented. The rate of light-dependent ammonia accumulation in L-methioninesulphoximine (MSO: glutamine synthetase inhibitor)-treated wheat(Triticum aestivum L. cv. Maris Huntsman) primary leaf sectionsincreased with mesophyll cell maturity. Ammonia production inthe more mature sections (beyond 2.0 cm from the basal meristem)was inhibited by elevated CO2 concentrations and by incubationwith 10 mol m–3 pyrid-2-yl hydroxymethane sulphonate (HPMS).In contrast, the low levels of ammonia which accumulated inthe immature sections (0 to 2.0 cm from the base) were unaffectedby such treatments. This indicates that the ammonia producedin mature wheat leaf sections is of photorespiratory originand that the capacity of this pathway increases with mesophyllcell and chloroplast development. Rates of CO2-dependent oxygenevolution by leaf sections (under saturating CO2) increasedin parallel with ammonia production. Levels of endogenous nitratewere relatively high and increased from 5.15 mol x 10–13mesophyll cell–1 in meristematic cells to 6.6 mol x 10–12mesophyll cell–1 in mature tissue. There was no significantchange in leaf nitrate level during 30 min light incubationof the wheat leaf sections, indicating that the majority ofthe nitrate was metabolically inactive and stored in the vacuole.Activities of key enzymes of photorespiration (glutamine synthetase,glycollate oxidase), nitrogen metabolism (nitrate reductase,glutamate dehydrogenase, glutamine synthetase) and mitochondrialrespiration (cytochrome oxidase), showed specific and distinctpatterns of development during leaf growth. Chloroplast glutaminesynthetase (GS2) and peroxisomal glycollate oxidase developedin apparent synchrony with the major increase in activity occurringin regions beyond4.0 cm from the leaf base, i.e. where photorespirationwas developing. Cytosolic glutamine synthetase (GS1) and nitratereductase (in vivo) activities were identical throughout leafgrowth, reaching maximum rates at 4.0 cm from the base and thenremaining constant. Activities of the mitochondrial enzymesglutamate dehydrogenase (GDH) and cytochrome oxidase were highin meristematic cells and increased in parallel, attaining amaximum towards the leaf tip. This indicated a respiratory,as opposed to a photorespiratory, role for GDH in wheat leafmetabolism. A study was made of the incorporation of 14C by intact leavesof Coffea arabica (cultivars Mundo Novo, Catuai, 1130–13,and H 6586–2) and Coffea canephora (cultivar Guarini)supplied with gas mixtures containing 14CO2 under controlledconditions. Samples of the leaves were combusted and the 14Cin the CO2 produced measured using a liquid scintillation counter.The results were used to estimate photosynthetic rates. Theeffects of changing the partial pressures of O2 and CO2 on thephotosynthetic rate were studied and estimates made of the CO2compensation point and photorespiration. The data obtained show differences between the mean net photosyntheticrates of the C. arabica cultivars (6·14 mg CO2 dm–2h–1) and the mean rate for the C. canephora cultivar (3·96mg CO2 dm–2 h–1). The cultivar of the latter speciesphotorespired more rapidly than the cultivar Catuai of C. arabica.Rates of photosynthesis in coffee measured using the 14CO2 methodwere similar to rates obtained by others using an infrared gasanalyser. The 14CO2 method proved to be reliable for photosyntheticmeasurements and the apparatus is suitable for use in fieldconditions.  相似文献

The carboxylase activity of ribulose-1,5-bisphosphate carboxylase/oxygenaseand of phosphoenolpyruvate carboxylase, and the light saturatedrate of net photosynthesis were measured in the developing 5thleaf of tomato plants. Values for light saturated net photosynthesiswere also calculated from the measured carboxylase activitiesand estimates of internal CO2 and oxygen concentrations. Thecalculated rate using the activity of ribulose bisphosphatecarboxylase alone for net CO2 assimilation in 300 mm3 dm–3CO2 was greater than the measured rate at 80% and full expansionbut less than the measured rate in younger leaves. When theactivities of both the carboxylases were taken into accountbetter agreement was evident for young leaves but the rate wasfurther overestimated for older leaves The calculated rate forphotosynthesis in 1200 mm3 dm–3 CO2, assuming saturationof ribulose bisphosphate carboxylase with RuBP, was an overestimatefor young leaves but was close to the observed values for leavesnear full expansion. The effects of elevated atmospheric CO2 concentrations on theecophysiological responses (gas exchange, chlorophyll a fluorescence,Rubisco activity, leaf area development) as well as on the growthand biomass production of two poplar clones (i.e. Populus trichocarpax P. deltoides clone Beaupré and P. x euramericana cloneRobusta) were examined under open top chamber conditions. Theelevated CO2 treatment (ambient + 350 µmol mol-1) stimulatedabove-ground biomass of clones Robusta and Beaupré afterthe first growing season by 55 and 38%, respectively. This increasedbiomass production under elevated CO2 was associated with asignificant increase in plant height, the latter being the resultof enhanced internode elongation rather than an increased productionof leaves or internodes. Both an increased leaf area index (LAI)and a stimulated net photosynthesis per unit leaf contributedto a significantly higher stem biomass per unit leaf area, andthus to the increased above-ground biomass production underthe elevated CO2 concentrations in both clones. The larger LAIwas caused by a larger individual leaf size and leaf growthrate; the number of leaves was not altered by the elevated CO2treatment. The higher net leaf photosynthesis was the resultof an increase in the photochemical (maximal chlorophyll fluorescenceFm and photochemical efficiency Fv/Fm) as well as in the biochemical(increased Rubisco activity) process capacities. Single, seed-grown plants of ryegrass (Lolium perenne L. cv.Melle) were grown for 49 d from the early seedling stage ingrowth cabinets at a day/night temperature of 20/15 C, witha 12 h photoperiod, and a CO2 concentration of either 340 or680µI 1–1 CO2. Following complete acclimation tothe environmental regimes, leaf and whole plant CO2 effluxesand influxes were measured using infra-red gas analysis techniques.Elevated CO2 increased rates of photosynthesis of young, fullyexpanded leaves by 35–46% and of whole plants by morethan 50%. For both leaves and whole plants acclimation to 680µI–1 CO2 reduced rates of photosynthesis in bothCO2 regimes, compared with plants acclimated to 340µll–1. There was no significant effect of CO2 regime onrespiration rates of either leaves or whole plants, althoughleaves developed in elevated CO2 exhibited generally lower ratesthan those developed in 340µI I–1 CO2. Initially the seedling plants in elevated CO2 grew faster thantheir counterparts in 340µI I–1 CO2, but this effectquickly petered out and final plant weights differed by onlyc. 10%. Since the total area of expanded and unexpanded laminaewas unaffected by CO2 regime, specific leaf area was persistently13–40% lower in elevated CO2 while, similarly, root/shootratio was also reduced throughout the experiment. Elevated CO2reduced tissue nitrogen contents of expanded leaves, but hadno effect on the nitrogen contents of unexpanded leaves, sheathsor roots. The lack of a pronounced effect of elevated CO2 on plant growthwas primarily due to the fact that CO2 concentration did notinfluence tiller (branch) numbers. In the absence of an effecton tiller numbers, any possible weight increment was restrictedto the c. 2.5 leaves of each tiller. The reason for the lackof an effect on tillering is not known. CO2 exchange characteristics of detached mature and senescentflag leaves and of bracts in some Poaceae and Cyperaceae species,respectively, were studied using a closed IR system. Senescentleaves, 30 to 45 days after flowering, showed lower rates ofapparent photosynthesis and dark respiration, and higher CO2compensation points (CCP) than those measured at the floweringstage. In senescent C4-Poaceae, the increase of CCP was small(from 4.8 to 10.1 ppm on the average) with little influenceof temperature, and the photorespiration level, 0.4 mg CO2/dm2/hr,was as low as that in mature leaf, indicating the presence ofnormal C4-characteristics. On the other hand, a C4-Cyperaceae,Cyperus microria Steud., showed extensive increases of CCP (from9 to 41 ppm) and photorespiration (from 0.8 to 2.1 mg CO2/dm2/hr)with senescence. Mistletoes usually have slower rates of photosynthesis thantheir hosts. This study examines CO2assimilation, chlorophyllfluorescence and the chlorophyll content of temperate host–parasitepairs (nine hosts parasitized by Ileostylus micranthus and Carpodetusserratus parasitized by Tupeia antarctica). The hosts of I.micranthus had higher mean annual CO2assimilation (3.59 ±0.41 µmol m-2 s-1) than I. micranthus(2.42 ± 0.20µmol m-2 s-1), and C. serratus(2.41 ± 0.43 µmolm-2 s-1) showed higher CO2assimilation than T. antarctica(0.67± 0.64 µmol m-2 s-1). Hosts saturated at significantlyhigher electron transport rates (ETR) and light levels thanmistletoes. The positive relationship between CO2assimilationand electron transport suggests that the lower CO2assimilationrates in mistletoes are a consequence of lower electron transportrates. When photosynthetic rates, ETR and chlorophyll a /b ratioswere adjusted for photosynthetically active radiation, hostsdid not have significantly higher CO2assimilation (3.21 ±0.37 µmol m-2 s-1) than mistletoes (2.54 ± 0.41µmol m-2 s-1), but still had significantly higher ETRand chlorophyll a / b ratios. The electron transport rates,saturating light and chlorophyll a / b ratios of sun leavesfrom mistletoes were similar to host shade leaves. These responsesindicate that in comparison with their hosts, mistletoe leaveshave the photosynthetic characteristics of the leaves of shadeplants. Rates of net photosynthesis of the flag leaves of 15 genotypesof wheat and related species were measured throughout theirlife, using intact leaves on plants grown in the field. At thestage when rates were maximal, they were in general highestfor the diploid species, intermediate for the tetraploidspeciesand lowest for Triticum aestivum (means of 38, 32 and 28 mgCO2 dm–2 h–1 respectively). Rates were stronglynegatively correlated with leaf area, leaf width and the meanplan area per mesophyll cell and positvely correlated with stomatalfrequency and number of veins per mm of leaf width. The differencesamong species in these attributes were mainly related to ploidylevel. It was not possible to determine the relative importanceof each anatomical feature, though the changes in stomatal frequencyhad only slight effects on stomatal conductance and the observeddifferences in rates of photosynthesis were much greater thanwould be expected from those in stomatal conductance alone. There was genetic variation in rates of light dependent oxygenevolution of isolated protoplasts and intact chloroplasts butno difference attributable to ploidy. The mean rate, 91 µmolO2 mg–1 chlorophyll h–1, equivalent to 3.9 mg CO2mg-1chlorophyll h-1 was considerably less than the rate of photosynthesisin comparable intact leaves, which was 7.2 mg CO2 mg–1chlorophyll h–1. The total above-ground dry matter yields were least for thewild diploids T. urartu and T. thauodar and the wild tetraploidT. dicoccoides, but the other wild diploids produced as muchdry matter as the hexaploids. The prospects of exploiting differences in photosynthetic ratein the breeding of higher yielding varieties are discussed. Dark respiration in attached and detached mature leaves of thefield bean (Vicia faba L.) was studied whilst leaves experiencedup to 60 h of darkness. The results showed: (1) the initialrespiration rate to vary according to the irradiance duringthe previous photoperiod; (2) the dark respiration rate (perunit area) of attached leaves to be essentially constant duringa normal 12 h night although there was a rapid loss in leafd. wt during this time; (3) after 12 h, the respiration rateof attached leaves decayed to an asymptotic value at about 36h; (4) the respiration rate of leaves detached at the end ofthe photoperiod and maintained in the dark on deionised water,decayed only after 36 h of darkness; (5) there was no differencebetween the respiration rate of attached and detached leavesduring the normal 12 h night. It is concluded that the dark respiration of attached fieldbean leaves is intially related to the synthesis and translocationof sucrose in addition to maintenance. After about 36 h, whenthe rate of CO2 efflux is more or less steady, the CO2 effluxreflects the intensity of maintenance processes only. The maintenancerespiration rate (determined after 60 h in the dark) rangedfrom 062 to 151 mg CO2 (g d. wt)–1 h–1 but wasrelatively unaffected by several applied treatments. The rates of net photosynthesis by closed canopies of tomatoplants were measured at three CO2 concentrations and three humiditiesover a range of natural light flux densities. The data havebeen analysed using a model of canopy photosynthesis which allowsfor variation in leaf area index and other leaf and canopy characteristics.The model also deals explicitly with the effects of CO2 concentration,leaf conductance, and photorespiration on the leaf photochemicalefficiency, . The leaves were found to have a photochemicalefficiency in the absence of photorespiration, m, of 12?6 ?10–9 kg (CO2) J–1. At a CO2 concentration of 0?73 ? 10–3 kg m–3 (400vpm) the leaf photochemical efficiency, , and canopy light utilizationefficiency, c, were 18 per cent greater at a vapour pressuredeficit of 0?5 kPa than at 1?0 kPa. At a CO2 concentration of2?2 ? 10–3 kg m–3 (1200 vpm) they were only 5 percent greater.  相似文献

Time-courses of 14CO2-fixation and of enzyme activities involvedin photorespiration and photosynthesis were determined duringthe life span of cotyledons from sunflower seedlings (Helianthusannuus L.). Glycolate formation in vivo was estimated from theresults of combined labelling and inhibitor experiments. NADPH-glyceraldehyde-3-phosphatedehydrogenase, NADPH-glyoxylate reductase and chlorophyll werewell correlated with the time-course of 14CO2-fixation (photosynthesis).There was, however, a considerable discrepancy between the developmentalsequence of photosynthesis and that of both ribulose-l,5-bisphosphatecarboxylase and glycolate oxidase. Furthermore, time-coursesof glycolate oxidase activity in vitro and of glycolate formationin vivo differed significantly. Therefore, the use of glycolateoxidase as a marker for the activity of photorespiration ingreening sunflower cotyledons may be questionable. Results from14CO2-labelling experiments with cotyledons treated with theglycolate oxidase inhibitor 2-hydroxy butynoic acid suggestthat glycolate formation relative to CO2-fixation is reducedin senescent cotyledons. 14CO2 assimilation was studied with diploid, tetraploid, hexaploidspecies of the genera Triticum and their wild relatives Aegilops.Attached mature leaves of 3–4 weekold plants were allowedto undergo photosynthesis under air at ambient temperature.The pattern of distribution of 14C was notably similar in Triticumand Aegilops species whatever the level of ploidy. Sucrose wasthe sink for photosynthetic carbon. 14C for sucrose synthesis was supplied either through the glycolatepathway by glycolate, the product of the photorespiration orby the Calvin cycle intermediates exported into the cytoplasm.Depending on the species, the glycolate pathway provided 40to 75%of the sucrose 14C. The higher labeling of sucrose was associated with the greaterparticipation of the glycolate pathway in the wild diploid (DD)A. squarrosa and in the cultivated hexaploid (AABBDD) T. aestivum.The results suggest that the expression of the male D genomeis dominant over the female AB genome in T. aestivum. In T. aestivum under ambient conditions lowering (low temperature)or hindering (1% O2 ) photorespiration, sucrose labeling decreased,but serine and glycine labeling was favoured. We propose thatin wheat leaves, the role of photorespiration is to drain artof the carbon exported from the chloroplast as glycolate, towardssucrose synthesis. (Received March 16, 1979; )  相似文献
