Several genetically stable mutants blocked in nikkomycin biosynthesis were obtained after the slightly germinated spores of Streptomyces ansochromogenes, a nikkomycin producer, were treated with ultra violet radiation. One of the mutants is the same in morpholotical differentiation as the wild type strain and is designated as NBB19. A DMA library was constructed using plasmid plJ702 as cloning vector, NBB19 as cloning recipient. A 6 kb DNA fragment which can genetically complement NBB19 was cloned when screening the library for antifungal activity. Sequence analysis showed that the 3 kb Bgl II-Sal I fragment contains one complete ORF (ORF1) and one partial ORF (ORF2). ORF1 is designated as sanA. sanA is 1 365 bp, encoding a protein consisting of 454 amino acid residues. Database searching indicated that sanA is homologous to the hypothetical methyltransferase in Pyrococcus horikoshli with 25% identities and 41% positives. A 111-bp DNA fragment related to nikkomycin biosynthesis of Streptomyces ansochromogenes 7100 was obtained with the method of reverse genetics. Then, a 2.2-kb DNA fragment was cloned from the DNA library of S. ansochromogenes 7100 by using the 111-bp fragment as a probe. Sequence analysis showed that the fragment contains one complete open reading frame (ORF) that encodes a 219-amino acid (aa) protein, and this gene was designated sanF (GenBank Accession No. AF223971). The function of the sanF gene was studied by a strategy of gene disruption, and the resulting sanF mutants lost the ability to synthesize biologically active nikkomycin, indicating that sanF is essential for nikkomycin biosynthesis. A partial DNA library ofStreptomyces ansochromogenes 7100 was constructed by using plasmid pl J702 as vector and white mutant W19 as recipient. About 3 000 clones were obtained, two of which gave rise to the grey phenotype as wild type 7100. The plasmids were isolated from two transformants. The result indicated that the 5.2 kb and 5.8 kb DNA fragments were inserted into plJ702. The resulting recombinant plasmids were designated as pNL-1 and pNL-2 respectively. The 1.25 kbPstl I -Apa I DNA fragment from pNL-1 was recognized as its complementarity to W19 strain. The nucleotide sequence of the 3.0 kbPst I DNA fragment including 1.25 kb was determined and analyzed. The result indicated that this DNA fragment contains one complete open reading frame (ORF1) which encodes a protein with 295 amino acid residues, and this gene was designated assawB. The deduced protein has 81% amino acid identities in comparison with that encoded bywhiH inStreptomyces coelicolor. The function ofsawB gene was studied by using strategy of gene disruption, and the resultingsawB mutant failed to form spores and produced loosely coiled aerial hyphal. The result showed thatsawB is closely related to hyphal coiling and sporulation in S.ansochromogenes, and also indicated that thesawB can complementwhiH mutant (C119) to restore the grey phenotype ofStreptomyces coelicolor J 1501 (wild type).  相似文献

以尼可霉素生物合成相关的基因片段为探针,从圈卷产色链霉菌cosmid基因文库中筛选到1个大约7.5kb的DNA片段,交DNA片段克隆到载体pBluescripM13-的KpnⅠ位点,得到了重组质粒pNL2200.对pNL2200中外源DNA片段进行了一系列的亚克隆及部分核苷酸序列分析。结果表明,2.3kb的SalⅠ-BamHⅠDNA片段中含有1个完整的开放阅读框,起始密码子为271位的GTG,终止密码子为1954位的TGA,该基因的大小为1686bp,编码1个大小为561个氨基酸的蛋白质产物。利用blastx程序的蛋白质数据库中进行同源比较,结果揭示此基因产物与腺苷酸形成酶超家族的连接酶有44%的一致性,此外,该基因的破坏导致圈卷产色链霉菌尼可霉素生物合成能力的丧失,证明它是尼可霉素生物合成所必需的,命名为其为sanJ。  相似文献

距链霉菌发育分化控制启动子 P T H4 直接控制的下游基因 pro X 间隔24 个碱基处存在一个部分开放阅读框 ( O R F) , 根据序列分析推测为丝氨酸蛋白酶的一部分。以此部分 D N A 序列为探针, 在构建的圈卷产色链霉菌7100 的 D N A 文库中克隆到一个与链霉菌发育和分化有关的新基因, 称之为sa w D。序列测定及分析结果表明, 在1320bp 的 D N A 序列中有一个完整的开放阅读框 ( O R F) , 翻译起始位点为210 位碱基处的 G T G, 终止密码子 T G A 位于序列的999 位碱基处。在距翻译起始位点 G T G 上游4 个碱基间隔处有典型的核糖体结合位点区域 G A G G G A。在计算机蛋白文库中进行了同源性比较研究, 结果表明263个氨基酸的蛋白产物与 Caulobacter crescentus 的依赖于 A T P 的丝氨酸蛋白酶有447 % 的同源性, 其中存在功能活性区的丝氨酸保守位点 ( G P S A G) 。基因功能研究表明, saw D 在圈卷产色链霉菌发育分化中与气生菌丝分隔和色素的合成有关。该基因被阻断或破坏后, 使野生型圈卷产色链霉菌的分化停止在气生菌丝阶段, 不能形成具有灰色色素的孢子, 而出现白色  相似文献

采用常规转化方法用来自天蓝色链霉菌J1 5 0 1的质粒pUC1 1 6 9(pMT6 6 0∷Tn45 5 6∷vph)多次转化尼可霉素产生菌圈卷产色链霉菌野生型 71 0 0的原生质体 ,均未得到转化子。采用限制性热衰减法于 5 0℃ ,3 0min溶菌制备 71 0 0的原生质体 ,获得了转化子 ,但转化频率极低 ,只有 0 4个转化子 μgDNA。用来自 71 0 0的pUC1 1 6 9再转化不含pUC1 1 6 9的 71 0 0原生质体 ,转化频率提高 1 0 3 ~ 1 0 4 倍。于 3 9℃ ,MM Vio条件下培养携带有pUC1 1 6 9的 71 0 0孢子 ,Tn45 6 0发生转座 ,筛选到 40 6 8个转座菌落 ,并从中得到 8株尼可霉素阻断突变株 ;对这 8株突变株的总DNA进行Southern杂交分析表明 ,Tn45 6 0至少在 4个不同的位点插入到 71 0 0的染色体上。用实验室已获得的与尼可霉素生物合成有关的 3 0kbDNA片段为探针和经不同酶切的 8株突变株的总DNA进行Southern杂交 ,结果表明 ,除阻断突变株Nik5有杂交信号且杂交信号大小均同野生型…  相似文献

A 2-kb SmaI DNA fragment was cloned from the cosmid library of Streptomyces ansochromogenes. This DNA fragment contains a complete open reading frame which is 1275 bp in length, designated sanS (GenBank accession no. AF322179). The deduced SanS protein consists of 424 amino acids and belongs to a superfamily of enzymes with an unusual ATP-grasp fold. The disruption and complementation of sanS indicated that sanS is essential for nikkomycin biosynthesis in Streptomyces ansochromogenes. The sanS gene was subcloned into expression vector pET23b and overexpressed in E. coli BL21 (DE3). The protein was then purified and showed ATPase activity.  相似文献

A 2.8-kb BamHI fragment was cloned from the cosmid library of Streptomyces ansochromogenes by using the 1.35-kb BamHI-ApaI fragment of sanO involved in nikkomycin biosynthesis as a probe. Sequence analysis showed that the BamHI fragment contains an open reading frame with 1191 bp, which was designated sanQ. In search of databases, the deduced product of sanQ gene has 56% similarity to the cytochrome P450. sanQ gene was inactivated by insertion of a kanamycin resistance gene. The resulting disruptants failed to produce nikkomycin X, but nikkomycin Z was at the same level as the wild type, indicating that sanQ is essential for the biosynthesis of nikkomycin X. 通过反向遗传学方法克隆到圈卷产色链霉菌尼可霉素生物合成基因簇中约7.0kb的DNA片段。该片段除含有尼可霉素生物合成基因sanF外,对sanF上游约22kb的BglⅡDNA片段进行序列测定及分析表明,还含有两个完整的开放阅读框(ORF)。ORF1由1233个核苷酸组成,ORF2由195个核苷酸组成,它们分别编码由410个氨基酸残基和64个氨基酸残基组成的蛋白质,依次命名为sanH和sanI。蛋白序列数据库比较结果表明,SanH和SanI与浅灰链霉菌(\%Streptomyces griseolus)\%中共转录的细胞色素P450(cytochrome P450)和铁氧还蛋白(ferredoxin)有较高的同源性,一致性分别为46%和56%,相似性分别为62%和70%。基因功能研究表明,sanH基因的破坏虽不影响圈卷产色链霉菌产生的尼可霉素的生物活性,但该基因可能参与了尼可霉素羟基化反应的生物合成。  相似文献

A 4.6 kb DNA fragment was cloned from the DNA library ofStreptomyces ansochromogenes using a partial DNA fragment located in the downstream of promoter-PTH4 as probe. The experiments revealed that this DNA fragment consists ofsaw D gene and a 1.4 kbPvu II fragment which can accelerate mycelium formation ofS. ansochromogenes. The nucleotide sequence of 1.4 kb DNA fragment was determined and analysed; the result indicated that the fragment contains one complete open reading frame (ORF) which encodes a protein with 213 amino acids, and this gene was designated assamfR. The deduced protein has 36% amino acid identities and 52% amino acid similarities in comparison with that encoded byhppR gene, which is involved in the regulation of catabolism for 3-(3-hydroxyphenyl) propionate (3HPP) inRhodococcus globerulus. The function ofsamfR gene was studied using strategy of gene disruption, and the resultingsamfR mutant failed to form aerial hyphae and spores, its development and differentiation stopped at the stage of substrate mycelium in contrast with wild type strain. We report the cloning ofhermit, a member of thehAT family of transposable elements from the genome of the Australian sheep blowfly,Lucilia cuprina. Hermit is 2716 bp long and is 49% homologous to the autonomoushobo element,HFL1, at the nucleic acid level.Hermit has 15 bp terminal inverted repeats that share 10 bp with the terminal inverted repeats ofHFL1. Conceptual translation reveals a 583 residue open reading frame (ORF) that is 64% similar and 42% identical to theHFL1 ORF. However, the sequence of thehermit element contains two frameshifts within the putative ORF, indication thathermit is an inactive element. Analysis ofL. cuprina strains from within and outside Australia suggested thathermit is present as a single copy in all the genomes analysed.  相似文献

Mutants resistant to nikkomycin, an inhibitor of chitin biosynthesis, were isolated after exposure of wild-type spores of the fungus Phycomyces blakesleeanus to N-methyl-N-nitro-N-nitrosoguanidine. Genetic analysis revealed that nikkomycin resistance was due to mutations in a single gene, chsA. Mutants and wild type grew equally well in the absence of nikkomycin. In contrast to the wild type, whose spore germination and mycelial growth were inhibited by 5 M nikkomycin, chsA mutants grew reasonably well in the presence of 50 M nikkomycin. Chitin synthesis in vivo was much less affected by the drug in the mutants than in the wild type. Resistance was not due to impaired uptake or detoxification of the drug. Analysis of the kinetics of chitin synthesis in vitro showed that the mutants had a decreased Ka for the allosteric activator, N-acetylglucosamine, and gross alterations in nikkomycin inhibition kinetics. These results indicate that chsA is the structural gene for chitin synthetase, or at least for the polypeptide that bears the catalytic and allosteric sites.  相似文献

The mating type locus of the oomycete,Phytophthora infestans, is embedded in a region of DNA that displays distorted and non-Mendelian segregation. By using DNA probes linked to the mating type locus to genetically and physically characterize that region, a large zone of chromosomal heteromorphism was detected. LocusS1 was shown to represent a tandemly repeated array of DNA that was typically present in a hemizygous state in A1 isolates while being absent from A2 isolates. The analysis of the parents and progeny of seven crosses indicated that the tandem array was linked in cis to the A1-determining allele of the mating type locus. A worldwide survey of genotypically diverse field isolates ofP. infestans indicated thatS1 was present in each of 48 isolates of the A1 mating type that were tested, but was absent in 46 of 47 A2 strains. Physical analysis ofS1 indicated that the tandemly repeated DNA sequence spanned about 300 kb and had evolved from a 1.35-kb monomer. Internal deletions occurred withinS1 during sexual propagation. This and other mutations apparently contributed to a high degree of polymorphism within theS1 array.  相似文献

NIN1 is an essential gene for growth of the yeastSaccharomyces cerevisiae and was recently found to encode a component of the regulatory subunit of the 26S proteasome. Thenin1-1 mutant is temperature sensitive and its main defect is in G1/S progression and G2/M progression at non-permissive temperatures. One of the two multicopy suppressors ofnin1-1, SUN2 (SUppressor of Nin1-1), was found to encode a protein of 523 amino acids whose sequence is similar to those ofDrosophila melanogaster diphenol oxidase A2 and the mouse mast-cell Tum transplantation antigen, P91A. The C-terminal half of Sun2p was found to be functional as Sun2p at 25° C, 30° C, and 34° C but not at 37° C. The open reading frame (ORF) of theDrosophila diphenol oxidase A2 gene (Dox-A2) was obtained from a lambda phage cDNA library using the polymerase chain reaction technique. TheDox-A2 ORF driven by theTDH3 promoter complemented the phenotype of a strain deleted forsun2. ThisDox-A2-dependent strain was temperature sensitive and accumulated dumb-bell-shaped cells, with an undivided nucleus at the isthmus, after temperature upshift. This morphology is similar to that ofnin1-1 cells kept at a restrictive temperature. These results suggest thatSUN2 is a functional counterpart ofDox-A2 and that these genes play a pivotal role in the cell cycle in each organism.  相似文献

An iron-regulated gene, pbsC, required for siderophore production in fluorescent Pseudomonas sp. strain M114 has been identified. A kanamycin-resistance cassette was inserted at specific restriction sites within a 7 kb genomic fragment of M114 DNA and by marker exchange two siderophore-negative mutants, designated M1 and M2, were isolated. The nucleotide sequence of approximately 4 kb of the region flanking the insertion sites was determined and a large open reading frame (ORF) extending for 2409 by was identified. This gene was designated pbsC (pseudobactin synthesis C) and its putative protein product termed PbsC. PbsC was found to be homologous to a family of enzymes involved in the biosynthesis of secondary metabolites, including EntF of Escherichia coli. These enzymes are believed to act via ATP-dependent binding of AMP to their substrate. Several areas of high sequence homology between these proteins and PbsC were observed, including a conserved AMP-binding domain. The expression of pbsC is iron-regulated as revealed when a DNA fragment containing the upstream region was cloned in a promoter probe vector and conjugated into the wild-type strain, M114. The nucleotide sequence upstream of the putative translational start site contains a region homologous to previously defined –16 to –25 sequences of iron-regulated genes but did not contain an iron-box consensus sequence. It was noted that inactivation of the pbsC gene also affected other iron-regulated phenotypes of Pseudomonas M114.  相似文献

Summary Three cellulose-negative (Cel-) mutants of Acetobacter xylinum strain ATCC 23768 were complemented by a cloned 2.8 kb DNA fragment from the wild type. Biochemical analysis of the mutants showed that they were deficient in the enzyme uridine 5-diphosphoglucose (UDPG) pyrophosphorylase. The analysis also showed that the mutants could synthesize (1-4)-glucan in vitro from UDPG, but not in vivo from glucose. This result was expected, since UDPG is known to be the precursor for cellulose synthesis in A. xylinum. In order to analyze the function of the cloned gene in more detail, its biological activity in Escherichia coli was studied. These experiments showed that the cloned fragment could be used to complement an E. coli mutant deficient in the structural gene for UDPG pyrophosphorylase. It is therefore clear that the cloned fragment must contain this gene from A. xylinum. This is to our knowledge the first example of the cloning of a gene with a known function in cellulose biosynthesis from any organism, and we suggest the gene be designated celA.  相似文献
