Germination percentage of Dactylorhiza majalis (Rchb.f.) Hunt & Summerh. (Orchidaceae) was increased by illuminating surface-sterilized, rinsed and incubated seeds with white light in 16 h photoperiods. Optimal exposures (10–14 days) raised germination from 40 to 75%. Longer light treatments resulted in reduced germination percentage and smaller seedlings. Only about 2% of the seeds could actually germinate in photoperiods; the germination of the rest was delayed by initial light and required about 14 days in constant darkness. Interruption of this darkness period with two consecutive photoperiods increased the germination percentage when the interruption occurred before day 8, but not when occurring later than that.  相似文献

Growth of the heterotrophic seedlings of Dactylorhiza majalis (Rchb.f.) Hunt & Summerh. (Orchidaceae) in vitro with a symbiotic fungus ( Rhizoctonia sp.) is density-dependent, even at densities that are not high enough to exploit the growth medium fully. Competition was two-sided; increasing density among seedlings did not increase size inequality between them. The slowly growing and smaller seedlings are normally more likely to die than the bigger ones, but mortality was not increased by higher density within the studied range. Growth depression by neighbouring seedlings is independent of their physical distance but varies with the total number of seedlings sharing the same culture dish and the same mycelium.
Therefore, if the growth-promoting effects of different fungi or treatments are to be compared in future work, the density of planting in the culture dishes should be comparable.  相似文献

Concerns about declining populations of terrestrial orchids make it important to identify the environmental factors crucial to seedling recruitment. This study shows that seedlings of Tipularia discolor (cranefly orchid) primarily occur on decomposing wood. Extensive searches of decomposing logs and stumps in mature and successional forests revealed seedlings at 24 sites, of which 15 could be identified as originating from seven different deciduous trees and one conifer. Seeds were planted in natural habitats to test the hypothesis that germination requires decomposing wood. In one experiment, seeds were placed into soil at sites where adult plants were abundant; no germination resulted. In a second experiment, germination of seeds sown in ambient soil was compared with sowings in plots amended with decomposing wood collected from a stump where spontaneous seedlings grew. Germination was much more frequent in plots amended with decomposing wood. We conclude that germination of T. discolor is stimulated in substrates that contain decomposing wood; judging from the occurrence of spontaneous seedlings, wood from at variety of tree species offer a suitable substrate.  相似文献

BACKGROUND AND AIMS: The labellar 'hairs' of some Cymbidium spp. are said to be thin-walled and to contain 'plasma', oil and sugars and it has long been speculated that they may function as food-hairs. However, the present authors' preliminary studies showed that certain atypical papillae may have a different role and, by reflecting light, function as a speculum. The purpose of the paper is to test this hypothesis. METHODS: Light microscopy, scanning electron microscopy, transmission electron microscopy, histochemistry and ultraviolet photography were used to investigate the structure, food content and light-reflecting properties of these papillae. KEY RESULTS AND CONCLUSIONS: The labellum of Cymbidium lowianum (Rchb.f.) Rchb.f. is densely clothed with obconical to conical papillae with wide bases and pointed tips. However, on either side of the median axis of the lip occur silvery patches comprising papillae with truncated tips and it is thought that these reflect light and thereby attract insect pollinators. Similar patches are also found in Cymbidium devonianum Paxton, and in both species, they are set against a reddish background, which, since bees cannot perceive this colour, probably appears dark to the insect thus enhancing the visual impact of the light-reflecting patches. In Cymbidium tigrinum Parish ex Hook. and Cymbidium mastersii Griff. ex Lindl., however, the labellum is mainly white and no light-reflecting patches were observed. Instead, unlike C. lowianum and C. devonianum, these species are highly fragrant and the attraction of insects probably depends to a greater extent on olfactory cues. In C. lowianum both types of papillae contain protein, starch and lipid bodies but only protein is seemingly present at elevated concentrations. However, lipoidal material also occurs upon the surface of the labellum and it is possible that this may be gathered by insects as reported for C. iridifolium A. Cunn (syn. C. madidum Lindl.). The labellar papillae of C. lowianum, thus, have the potential to function as food-hairs, although direct evidence for this is lacking.  相似文献

Williams ex Hodge.Nevertheless, it is predicted that such nectaries are likelyto be very diverse and the purpose of this paper is to comparethe nectary and the process of nectar secretion in Hexisea imbricata(Lindl.) Rchb.f. with that of Maxillaria coccinea so as to beginto characterize the nectaries of presumed ornithophilous Neotropicalorchids. • Methods Light microscopy, transmission electronmicroscopyand histochemistry were used to examine the histology and chemicalcomposition of nectary tissue and the process of nectar secretionin H. imbricata. • Key Results and Conclusions The nectary of H. imbricatahas a vascular supply, is bound by a single-layered epidermiswith few stomata and comprises two or three layers of subepidermalsecretory cells beneath which lie several layers of palisade-likeparenchymatous cells, some of which contain raphides or mucilage.The secretory cells are collenchymatous and their walls havenumerous pits with associated plasmodesmata. They contain thefull complement of organelles characteristic of secretory cellsas well as intravacuolar protein bodies but some of the secretoryepidermal cells, following secretion, collapse and their anticlinalwalls seem to fold. Nectar secretion is thought to be granulocrineand, following starch depletion, lipid droplets collect withinthe plastids. The nectar accumulates beneath the cuticle whichsubsequently forms swellings. Finally, nectar collects in thesaccate nectary spur formed by the fusion of the margins ofthe labellum and the base of the column-foot. Thus, althoughthe nectary of H. imbricata and M. coccinea have many featuresin common, they nevertheless display a number of important differences.  相似文献

Background and Aims

Floral secretions are common in Bulbophyllum Thouars, and the labella of a number of Asian species are said to produce secretions rich in lipids that act as food rewards for insect pollinators. Although some of these reports are based on simple histochemical tests, a much greater number are anecdotal and, hitherto, neither the ultrastructure of the labellum nor the secretory process has been investigated in detail. Furthermore, sophisticated histochemical approaches have generally not been applied. Here, both the labellar structure and the secretory process are investigated for four species of Asian Bulbophyllum sect. Racemosae Benth. & Hook. f., namely Bulbophyllum careyanum (Hook.) Spreng., B. morphologorum Kraenzl., B. orientale Seidenf. and B. wangkaense Seidenf., and compared with those of unequivocal lipid-secreting orchids.

Methods

Labellar, secretory tissue was investigated using light microscopy, scanning electron microscopy, transmission electron microscopy and histochemistry.

Key Results

The adaxial median longitudinal groove of the labellum contained secretory tissue comprising palisade-like epidermal cells, similar to those of certain lipid-secreting Oncidiinae Benth. However, these cells and their secretions gave positive results mainly for protein and mucilage, and their organelle complement was consistent with that of cells involved in protein and mucilage synthesis. Sub-cuticular accumulation of secretion resulted in cuticular distension and blistering. The sub-epidermal layer of isodiametric parenchyma contained starch and, like the epidermal cells, ultrastructure consistent with mucilage synthesis. Lipids were mainly confined to the cuticle, and hardly any intracellular lipid droplets were observed.

Conclusions

It is proposed that mucilage is produced by dictyosomes present in the palisade-like epidermal cells. Mucilage precursors may also be produced by these same organelles in sub-epidermal cells and are thought to pass along the symplast via plasmodesmata into the adjoining palisade-like secretory cells, which contain abundant arrays of rough endoplasmic reticulum. Here, they become chemically modified and form a protein-rich, mucilaginous secretion that, following vesicle-mediated transport across the cytoplasm, traverses the cell wall and accumulates in blisters formed from the distended cuticle. Rupture of these blisters releases the secretion onto the labellar surface. However, in certain species, there is some evidence that the secretion may traverse the cuticle via cuticular pores, and micro-channels may permit the passage of fragrance. Hydrolysis of sub-epidermal starch probably generates the carbohydrate and, together with mitochondria, much of the energy required for the secretory process. This anatomical organization resembles that found in certain lipid-secreting, Neotropical species of Bulbophyllum and Oncidiinae, but since the chemical composition of their secretions is different, and these taxa occur on a separate continent and have different insect pollinators, parallelism of floral anatomy is likely.  相似文献

Summary An efficient seed germination system was developed for an endemic and endangered orchid of the prairies of Bogotá, Colombia. The effects of three culture media [Murashige and Skoog (MS) salts (1962); Knudson C (KC) salts (1946); and Hydro-Coljap? salts], activated charcoal [0 and 0.5% (w/v)], α-naphthaleneacetic acid (NAA; 0.0, 2.68 and 5.37 μM), and four light regimes (white light, darkness, red light, and far-red light) on asymbiotic germination of Odontoglossum gloriosum, were studied. The best germination percentage and germination time was obtained on agar-solidified medium supplemented with Hydro-Coljap? salts and 2.68 μM NAA under red light with a 16-h photoperiod. The addition of activated charoal did not stimulate either germination or the development of O. gloriosum seedlings. This study describes a single medium-based protocol able to achieve more than 330 000 seedlings within 40wk, starting from a single capsule. This protocol is sufficient for both large-scale propagation and in vitro conservation of this threatened orchid.  相似文献

Vanilla claviculata (W.Wright) Sw. and K barbellata Rchb.f. grow sympatrically in the southwestern part of herto Rico. At three localities in the range of the two species, individuals with flowers that appeared Merent from previously known species were found. To test whether these were hybrids between V. claviculata and V. barbellata , 11 morphological floral characters were recorded at seven populations and allozyme profiles of the three taxa compared. Principal component analysis on the morphological characters gave three distinct groups with minor overlaps. The first component axis reflects variation in overall size of the flowers, while the second component reflects variation in the size of the distal aperture of the labellum. Allozyme data revealed that the putative hybrids have a significant surplus of heterozygotes ( F IS = - 0.65) as expected in a F 1 generation. Both data sets support the hypothesis that the individuals are hybrids, which is further augmented by interspecific pollination experiments between the putative parental species. This is the first case of natural hybridization reported in the genus Vanilla .  相似文献

Detailed ecological, morphological and molecular analyses were performed in mixed populations of diploid and autotetraploid Dactylorhiza maculata s.l. in Scandinavia. Comparisons were made with pure populations of either diploid ssp. fuchsii or tetraploid ssp. maculata. It was shown that mixed populations are the result of secondary contact between ssp. fuchsii and ssp. maculata. No patterns of recent and local autopolyploidization were found. Morphology and nuclear DNA markers (internal transcribed spacers of nuclear ribosomal DNA) showed that diploids and tetraploids from mixed populations have similar levels of differentiation to diploids and tetraploids from pure populations. Vegetation analyses, as well as analyses of environmental variables, revealed that diploid and tetraploid individuals in mixed populations are ecologically well differentiated on a microhabitat level. Diploids and tetraploids in pure populations have wider ecological amplitudes than they do in mixed populations. Triploid hybrids grew in intermediate microhabitats between diploids and tetraploids in the mixed populations. Plastid DNA markers indicated that both diploids and tetraploids may act as the maternal parent. Based on morphology and nuclear markers triploids are more similar to tetraploids than to diploids. There were indications of introgressive gene flow between ploidy levels. Plastid markers indicated that gene flow from diploid to tetraploid level is most common, but nuclear markers suggested that gene flow in opposite direction also may occur. Similar patterns of differentiation and gene flow appeared in localities that represented contrasting biogeographic regions. Disturbance and topography may explain why hybridization was slightly more common and the differentiation patterns somewhat less clear in the Scandinavian mountains than in the coastal lowland. BACKGROUND AND AIMS: Many orchid flowers have glands called elaiophores and these reward pollinating insects with oil. In contrast to other reward-producing structures such as nectaries, the anatomy of the elaiophore and the process of oil secretion have not been extensively studied. In this paper, elaiophore structure is described for two members of Oncidiinae, Oncidium trulliferum Lindl. and Ornithophora radicans (Rchb.f.) Garay & Pabst. METHODS: Elaiophores of both species were examined using light microscopy, scanning electron microscopy and transmission electron microscopy. KEY RESULTS AND CONCLUSIONS: In flowers of Oncidium trulliferum and Ornithophora radicans, oil is secreted by morphologically distinct elaiophores associated with the labellar callus. However, in O. trulliferum, elaiophores also occur on the lateral lobes of the labellum. In both these species, the epithelial elaiophores are composed of a single layer of palisade-like epidermal cells and a distinct subepithelial layer. Secretory elaiophore cells may contain numerous, starchless plastids, mitochondria and smooth endoplasmic reticulum profiles. In O. trulliferum, the cytoplasm contains myelin-like figures but these are absent from O. radicans. In the former species, cavities occur in the cell wall and these presumably facilitate the passage of oil onto the elaiophore surface. In O. radicans, the accumulation of oil between the outer tangential wall and the cuticle causes the latter to become distended. Since it is probable that the full discharge of oil from the elaiophores of O. radicans occurs only when the cuticle is ruptured by a visiting insect, this may contribute towards pollinator specificity. The structure of the elaiophore in these species resembles both that found in previously investigated species of Oncidiinae and that of certain members of the Malpighiaceae.  相似文献

Fatty acid analysis, a common method for the identification of bacteria, was modified and applied to characterize isolates of Fusarium oxysporum f. sp. vasinfectum. After evaluating the fatty acid profiles by means of cluster analysis and three-dimensional plotting of the main fatty acids, the isolates were classified into five groups. Identical patterns were obtained for isolates of races 3 and 5 and for isolates of races 2 and 6. Isolates with so far unknown race determinations were arranged into their corresponding fatty acid groups.  相似文献

The hypothesis of gene flow between species with large differences in chromosome numbers has rarely been tested in the wild, mainly because species of different ploidy are commonly assumed to be reproductively isolated from each other because of instantaneous and strong postzygotic barriers. In this study, a broad‐scale survey of molecular variation was carried out between two orchid species with different ploidy levels: Epidendrum fulgens (2n = 2x = 24 chromosomes) and Epidendrum puniceoluteum (2n = 4x = 52 chromosomes). To test the strength of their reproductive barriers, we investigated the distribution of genetic variation in sympatric and allopatric populations of these two species and conducted crossing experiments. Nuclear and plastid microsatellite loci were used to genotype 463 individuals from eight populations across the geographical range of both species along the Brazilian coastal plain. All six sympatric populations analysed presented hybrid zones, indicating that hybridization between E. fulgens and E. puniceoluteum is a common phenomenon. Bayesian assignment analysis detected the presence of F1 and F2 individuals and also signs of introgression, demonstrating a high potential for interspecific gene flow. Introgression occurs preferentially from E. fulgens to E. puniceoluteum. Pure parental individuals of both species display strong genotype–habitat associations, indicating that environment‐dependent selection could be acting in all hybrid zones. This study suggests that hybridization and introgression are evolutionary processes playing a role in the diversification of Epidendrum and indicates the importance of investigations of hybrid zones in understanding reproductive barriers and speciation processes in Neotropical orchid species.  相似文献

Human pancreatic cells of the Capan-1 cell line differentiate in culture. During the exponential growth phase, the cells are undifferentiated, only becoming differentiated during the stationary phase. The formation of domes in this phase is related to the exchange of water and electrolytes. The present study was designed to characterize the localization and expression of alkaline phosphatases (AP) in Capan-1 cells during growth in culture. Biochemical, cytoenzymatic and immunocytochemical methods were employed combined with light and electron microscopic examination. AP essentially of the placental type were expressed progressively during the exponential growth phase, and were seen to be distributed over the surface of the Capan-1 cells. In the stationary phase, the AP became localized on the surface of microvilli. The precipitates of the enzyme reaction highlighted regular four-bodied structures. Biochemical assays showed a progressive increase in activity of this enzyme in cells during both the exponential and stationary growth phases. However, in the stationary phase between days 7 and 8, there was a fall in enzyme activity, with a corresponding increase in this activity in the culture medium. Cytological examination indicated that this fall could be accounted for by loss of AP-positive membranes by vesiculization of apical microvilli and release of microvesicles into the culture medium. Immunoblots showed that Capan-1 cells expressed two types of AP, a placental type (70 kDa) and to a lesser extent a liver type (80 kDa). Expression of the placental type was attributed to a neoplastic derepression of the coding gene, while the liver type was assumed to be a normal gene expression of human duct cells. The placental type AP might thus serve as a marker of transformation, and the liver type as a marker of differentiation.  相似文献

We investigated the effect of genotype, seed maturity and culture medium on the in vitro germination and development of protocorms and plantlets from seeds of 13 different Ophrys species (O. apifera, O. attica, O. cornuta, O. delfinensis, O. ferrum-equinum, O. lutea, O. mammosa, O. speculum, O. spruneri, O. umbilicata, O. argolica, O. irricolor and O. tenthredinifera) collected in Greece, some of which are endemic to this country. Mature seeds (10 months after collection) and immature seeds (2 months after anthesis) were cultured in a coconut milk-enriched or a pineapple-enriched medium (CEM or PEM, respectively). The highest percentage of callogenesis (96%) was observed in immature seeds of O. delphinensis in the CEM, while the highest percentage of protocorm formation (52%) was observed in mature seeds of O. spuneri in the CEM. Protocorm formation was significantly lower in immature seeds than in mature seeds in both culture media. Eventually almost all of the transferred protocorms developed to plantlets, which later formed minitubers. PEM appeared to be the most suitable for the development of minitubers from plantlets. All of the factors investigated—as well as their interactions—significantly affected callogenesis and protocorm formation.
