Abstract:
Hydrophobic interaction chromatography with Phenyl Sepharose 6 Fast Flow resulted in 7-fold purification of xylanaseactivity from Melanocarpus albomyces with over 60% recovery of theactivity. The purified preparation consisted of two major isoenzymes out of seven present in the organism. Ethyleneglycol, used as the eluent, enhanced the thermal stability of the xylanase activity from a half-life of 2.2 h at 55 °C to almost 40 h.
