| 摘 要： | 为了获得高纯度的破伤风毒素,用疏水层析和离子交换层析纯化破伤风毒素。破伤风毒素培养滤液经Phenyl Sepharose疏水层析除去大部分杂质,再经DEAE Sephadex离子交换层析进一步纯化。经两步层析纯化后,毒素纯度达到2000Lf/mg PN以上,回收率为52%~73%。用此方法,连续纯化五批毒素,均获得高纯度的破伤风毒素。试验证明破伤风毒素经疏水层析和离子交换层析可得到有效纯化。

| Abstract: | To obtain high purity of tetanus toxin,purification of tetanus toxin was carried out by chromatography procedures.First most of impurity in the tetanus culture medium can be eliminated with HIC on Phenyl Sepharose,then tetanus toxin can be further purified by the ion-exchange chromatography(IEX) on DEAE Sephadex.Throuhg HIC and IEX purification,the purity of tetanus toxin exceeded 2000Lf/mg PN and yield of toxin reached 52%~73%.We obtained a high purity of tetanus toxin in 5 consecutive batches by using the combined chromatography procedures. |
